Abstract:Primary aromatic amines (PAAs) may be found in food contact materials and can potentially migrate from packaging material into the packed foodstuffs. Certain members of this substance class are of particular concern due to their toxicological properties. Legislators and authorities require a risk assessment regarding the transfer of PAAs into food. The evaluation of PAAs' migration into food relies on migration or extraction tests using aqueous and acidic food simulants or solvents. However, PAAs exhibit stability issues in acidic media, which could lead to an underestimation of migration. Therefore, a systematic stability study was conducted for 28 PAAs in 3% acetic acid and water at different storage conditions. Standard solutions of 2 and 10 ng/ml of the amines in these media were prepared and stored under the following conditions: 2 h at 40 °C, 70 °C, and 100 °C, and 10 days at − 20 °C, 4 °C, 20 °C, 40° C, and 60 °C. The test solutions were analyzed using liquid chromatography coupled to mass spectrometry, and recovery was calculated against freshly prepared standard solutions. It was found that several PAAs undergo a loss of recovery under the investigated conditions; 7 of the substances were identified to be the most unstable: 3,3ʹ-dimethoxybenzidine, 3,3ʹ-dimethylbenzidine, 6-methoxy- m -toluidine, 4-methoxy- m -phenylendiamine, 2,6-toluenediamine, benzidine and 2,4-toluenediamine. It was concluded that both time and storage temperature play a pivotal role in the stability of PAAs. The experiments also demonstrated that PAAs exhibit greater stability in water than in 3% acetic acid.
A liquid chromatography-tandem mass spectrometry method for the analysis of primary aromatic amines in human urine

Abstract:Aromatic amines are widely used in personal care products and human exposure to this class of chemicals is widespread. Bioanalytical methods to determine trace levels of aromatic amines in human urine are scarce. In this study, a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to determine 39 primary aromatic amines (AAs) along with nicotine and cotinine in human urine. Chromatographic separation of the 41 analytes was achieved on an Ultra Biphenyl (100 mm × 2.1 mm, 5 µm) column. Mass spectrometry was operated in electrospray ionization positive ion multiple reaction monitoring (MRM) mode. The method exhibited excellent linear dynamic range (0.1-50 ng/mL) with correlation coefficients (r) > 0.999 for all analytes. Urine samples (2 mL) were hydrolyzed using 10 M NaOH at 95 °C for 15 h and target analytes were extracted using methyl-tert-butyl ether (MTBE). Addition of 15 µL of 0.25 M HCl to the sample extracts improved the recoveries of several target analytes. The method was validated through the analysis of fortified quality control (QC) samples and a certified standard reference material (SRM). Relative recoveries (%) of target analytes fortified in QC samples were in the range of 75-114% for 37 of the 41 analytes while the other analytes exhibited lower recoveries (16-74%). The limits of detection (LOD) and limits of quantification (LOQ) of target analytes were in the range of 0.025-0.20 ng/mL and 0.1-1.0 ng/mL, respectively. Intra-day and inter-day precision of the method assessed through the analysis of fortified urine QC samples at three different concentrations were < 11.7% and < 15.9% (measured as RSD), respectively. The method was applied in the analysis of urine samples from the general population and known smokers; aniline, para-anisidine, para-toluidine, ortho/meta-toluidine, 3-chloroaniline, 4-chloroaniline, 3,4-dichloroaniline, and 4,4'-methylenedianiline were found in all smoker's urine at sum concentrations ranging from 0.04 to 9.16 ng/mL.
Determination of 14 nitrosamines at nanogram per liter levels in drinking water.

Abstract:N-Nitrosamines, probable human carcinogens, are a group of disinfection byproducts under consideration for drinking water regulation. Currently, no method can determine trace levels of alkyl and tobacco-specific nitrosamines (TSNAs) of varying physical and chemical properties in water by a single analysis. To tackle this difficulty, we developed a single solid-phase extraction (SPE) method with high-performance liquid chromatographytandem mass spectrometry (HPLCMS/MS) for the determination of 14 nitrosamines of health concern with widely differing properties. We made a cartridge composed of a vinyl/divinylbenzene polymer that efficiently concentrated the 14 nitrosamines in 100 mL of water (in contrast to 500 mL in other methods). This single SPEHPLCMS/MS technique provided calculated method detection limits of 0.01-2.7 ng/L and recoveries of 5393% for the 14 nitrosamines. We have successfully demonstrated that this method can determine the presence or absence of the 14 nitrosamines in drinking water systems (eight were evaluated in Canada and the U.S.), with occurrence similar to that in other surveys. N-Nitrosodimethylamine (NDMA), N-nitrosodiphenylamine, and the TSNA 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol were identified and quantified in authentic drinking water. Formation potential (FP) tests demonstrated that NDMA and TSNA precursors were present in (1) water samples in which tobacco was leached and (2) wastewater-impacted drinking water. Our results showed that prechlorination or ozonation destroyed most of the nitrosamine precursors in water. Our new single method determination of alkylnitrosamines and TSNAs significantly reduced the time and resource demands of analysis and will enable other studies to more efficiently study precursor sources, formation mechanisms, and removal techniques. It will be useful for human exposure and health risk assessments of nitrosamines in drinking water.

Abstract:Methods for measuring hair dye ingredients in urine, shampoo-water mixture, and river water were developed using solid phase extraction (SPE) and liquid chromatography/tandem mass spectrometry (LC/MS/MS). Coloring of hair results in the deposition of aromatic amine constituents on the hair and scalp, a portion of which passes through the skin, accumulating in the human body or being excreted. Ingredients in hair dye have been implicated in many kinds of adverse effects in humans, for example, dermatitis, allergies, and cancer. For purposes of this study, we checked for aromatic amines routinely present in hair dyes, and selected three target chemicals that are the most frequently used, namely, 2-methyl-5-aminophenol, 2,5-diaminotoluene, and 4-aminophenol. These amines are extremely water soluble and highly susceptible to oxidation. In the case of river water analysis, fifty milliliters of the sample with added ascorbic acid, as the antioxidant, was passed through an ion exchange cartridge. In the analysis of shampoo-water mixture, ten milliliters of the sample diluted nineteen fold with 1% ascorbic acid was passed through the ion exchange cartridge. In the case of urine, the sample containing ascorbic acid was diluted with a mixture of methanol and 2-propanol followed by centrifugation, and the supernatant was filtered. Before concentrating the eluted samples under a stream of nitrogen, acetonitrile was added as a keeper solvent to the solution eluted from the SPE cartridge, and the temperature of the solution was maintained below 37 ℃ to prevent the evaporation and oxidation of the target chemicals. The recoveries of the target chemicals were 96.5 ~123.8% (RSD 3.8 ~12.8%) for river water, 75.8 ~129.0% (RSD 3.3 ~8.3%) for shampoo-water mixture, and 75.8 ~127% (RSD 7.7 ~14.8%) for urine.

Abstract:Pheomelanin is an important type of melanin distributed in the skin, eye and hair in the mammal, which is of great social, clinical and cosmetic significance. In this study, a new HPLC method with fluorescence detection is described originally for the sensitive determination of pheomelanin in biological materials. The pheomelanin polymer is decomposed into two specific degradation products, 3-amino-4-hydroxyphenylalanine (3-AHP) and 4-amino-3-hydroxyphenylalanine (4-AHP) with hydriodic acid. Then the two AHP isomers are derivatized using a fluorescent probe naphthalene-2,3-dicarboxaldehyde in the presence of cyanide. The resulting highly stable 2-substituted 1-cyanobenz[f] isoindole derivatives were separated on a 5NH(2)-MS aminopropyl packed HPLC column with binary isocratic elution profile and detected fluorimetrically. The assay shows high sensitivity of 0.11 nM (2.2 fmol per injection, the lowest reported) at signal-to-noise ratio of 3 for each AHPs, good accuracy and precision (RSDs < 3.1%), and linearity (range of 0.02-10 mu M, r > 0.995). The results obtained by using fluorescence detection have been compared with other detection systems (electrochemical and UV). The sensitivity can increase from 100 to respect electrochemical detection and 30000 times respect UV detection. The method has been used for the quantitative determination of pheomelanin in various biological samples, including cell cultures from five types of melanoma cell lines of human and rat origin, hair samples of various colors, melanoma tissue and the urines from human melanoma patients and healthy subjects. This original application of HPLC-fluorescence detection represents a powerful tool for investigating pheomelanin synthesis in vitro and in vivo under physiological and pathophysiological conditions. Abstract:The migration of primary aromatic amines (PAAs) from food contact materials raises significant public health concerns. In this study, the migration levels of 26 PAAs were analyzed in 242 nylon cooking utensils using ultra-performance liquid chromatography-tandem mass spectrometry. A total of 18 PAAs were detected, of which 14 were quantified, with 4,4'-diaminodiphenylmethane (4,4'-MDA) and aniline being the most prevalent ones. The compliance rates for nylon kitchenware were similar under both legislation of European Commission (76.9%) and Chinese legislation (77.9%). Probabilistic non-carcinogenic and carcinogenic risk assessment were conducted using Monte Carlo simulation, with read-across approach applied to fill the gap of toxicity data. The hazard quotients for 18 PAAs were calculated, revealing that 17 PAAs (excluding 4,4'-MDA) had acceptable hazard quotients (<1). Lifetime cancer risks for 17 PAAs were determined, with 15 PAAs (excluding benzidine and 4,4'-MDA) showing acceptable cancer risks (<10-4). The study suggests that the non-carcinogenic and carcinogenic health risks posed by PAAs migrating from FCMs can be effectively mitigated by promptly identifying non-compliant products and reducing exposure to high-risk PAAs such as 4,4'-MDA and benzidine. Enhancing the understanding of PAA hazard characterization and implementing measures to minimize health risks associated with PAA migration from FCMs is hence recommended.
Analysis of six aromatic amines in the mainstream smoke of tobacco products

Abstract:Aromatic amines are a class of carcinogenic compounds in tobacco smoke that are listed on the FDA list of harmful and potentially harmful constituents (HPHCs). A method using solid-phase microextraction-coupled to gas chromatography-triple quadrupole mass spectrometry (SPME headspace GC-MS/MS) was developed and validated for the quantitative determination of six aromatic amines, including 1-aminonaphthalene (1-AN), 2-aminonaphthalene (2-AN), 3-aminobiphenyl (3-ABP), 4-aminobiphenyl (4-ABP), o-toluidine (o-TOL), and o-anisidine (o-ANI), in the mainstream smoke of cigarettes, cigars, and heated tobacco products. The method developed here combines high sensitivity with simple sample preparation and has demonstrated satisfactory linearity for all six aromatic amines with correlation coefficients greater than 0.9994. The limits of detection range and the limits of quantitation range were 12-96 pg/mL and 41-320 pg/mL, respectively. Their recoveries and coefficients of variation (CV%) were 90-112% and 2.1-6.6%, respectively. The new SPME headspace GC/MS/MS method has been successfully applied to measure the contents of the six aromatic amines in the mainstream smoke of cigarettes, cigars, and heated tobacco products.
Conversion to isothiocyanates via dithiocarbamates for the determination of aromatic primary amines by headspace-solid phase microextraction and gas chromatography

Abstract:A novel and highly selective method has been developed for the determination of aromatic primary amines by their conversion to dithiocarbamates by reaction with carbon disulphide, and then to isothiocyanates, which are volatile, by heating in the presence of a heavy metal ion. Zinc(II) was selected owing to its low toxicity and optimum yield of isothiocyanates. The latter were sampled by headspace-solid phase microextraction (HS-SPME) on divinylbenzene-carboxen-polydimethylsiloxane fibre, 50/30 μm. The HS-SPME procedure was optimized to provide adequate limits of detection in the analysis of aromatic amines in their real samples by gas chromatography with mass spectrometry (GC-MS) or flame ionization detection (GC-FID). The method gave rectilinear calibration graph, correlation coefficient and limit of detection, respectively, over the range 0.08-100 μg L(-1), 0.9950-0.9990 and 25-240 ng L(-1) in gas chromatography-mass spectrometry, and 0.01-10 mg L(-1), 0.9910-0.9991 and 0.8-3.0 μg L(-1) in gas chromatography-flame ionization detection. At two different levels, 10 and 40 μg L(-1), the range of intra-day RSD was 3.7-8.5% (GC-MS) and 3.3-9.2% (GC-FID), respectively. The proposed method is simple and rapid, and has been applied to determine aromatic primary amines in the environmental waters, food samples of ice cream powder and soft drinks concentrate, and food colours. The intra-day RSD in the analysis of real samples by GC-MS was in the range 3.6-6.2%. The food/colour samples were found to contain elevated levels of aniline and 2-toluidine.

Abstract:A fully automated, rapid, and reliable method for simultaneous determination of six carcinogenic primary aromatic amines (AAs), including o-toluidine (o-TOL), 2, 6-dimethylaniline (2, 6-DMA), o-anisidine (o-ASD), 1-naphthylamine (1-ANP), 2-naphthylamine (2-ANP), and 4-aminobiphenyl (4-ABP), in mainstream cigarette smoke was established. The proposed method was based on two-dimensional online solid phase extraction combined with liquid chromatography tandem mass spectrometry (SPE/LC-MS/MS). The particulate phase of the mainstream cigarette smoke was collected on a Cambridge filter pad and pretreated via ultrasonic extraction with 2% formic acid (FA), while the gas phase was trapped by 2% FA without pretreatment for determination. The two-dimensional online SPE comprised of two cartridges with different absorption characteristics was applied for sample pretreatment. Analysis was performed by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) under multiple reaction monitoring mode. Each sample required about 0.5h for solid phase extraction and analysis. The limit of detections (LODs) for six AAs ranged from 0.04 to 0.58ng/cig and recoveries were within 84.5%-122.9%. The relative standard deviations of intra- and inter-day tests for 3R4F reference cigarette were less than 6% and 7%, respectively, while no more than 7% and 8% separately for a type of Virginia cigarette. The proposed method enabled minimum sample pretreatment, full automation, and high throughput with high selectivity, sensitivity, and accuracy. As a part of the validation procedure, fifteen brands of cigarettes were tested by the designed method.
Clinico-pathologic correlation of 385 lesions of marginal destructive periodontitis.

Abstract:THIS STUDY investigated the relationship between the degree of periodontal destruction present in advanced periodontal lesions and consistently associated infiltrates of lymphocytes and/or plasma cells. A total of 385 biopsy specimens were obtained from the depths of pockets during routine periodontal surgery. The selected lesions were in "established" or "advanced" stages and were classified according to pocket depth as Class I (4-6 mm) or Class II (7-10 mm). The infiltrates attached to the cementum, bone, or lining the pocket soft tissue wall were classified in five types according to the distribution of lymphocytes, plasmocytes, and neutrophils. Statistical evaluation of the findings demonstrates a strong, significant relationship (P less than 0.001) between the degree of destruction of supporting periodontal tissues (Class II) and the predominance of plasma cells in the infiltrate (Type a). A weaker significant correlation (P less than 0.02) was found between predominant lymphocytic populations (Type b) and Class I lesions. The assumption was made that local, specific, cellular infiltrates reflecting immunologic mechanisms are associated with the degree of periodontal destruction in humans.
Determination of Toxic Pyrrolizidine Alkaloids in Traditional Chinese Herbal Medicines by UPLC-MS/MS and Accompanying Risk Assessment for Human Health

Abstract:Pyrrolizidine alkaloids (PAs) are a class of natural toxins with hepatotoxicity, genotoxicity and carcinogenicity. They are endogenous and adulterated toxic components widely found in food and herbal products. In this study, a sensitive and efficient ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was used to detect the PAs in 386 kinds of Chinese herbal medicines recorded in the Chinese Pharmacopoeia (2020). The estimated daily intake (EDI) of 0.007 μg/kg body weight (bw)/day was adopted as the safety baseline. The margin of exposure (MOE) approach was applied to evaluate the chronic exposure risk for the genotoxic and carcinogenic potential of PAs. Results showed that PAs was detected in 271 out of 386 samples with a content of 0.1–25,567.4 μg/kg, and there were 20 samples with EDI values above the baseline, 0.007 μg/kg bw/day. Beyond that, the MOE values for 10 out of 271 positive samples were below 10,000. Considering the actual situation, Haber’s rule was used to assume two weeks exposure every year during lifetime, and still the MOE values for four out of 271 positive samples were under 10,000, indicating these products may have potential health risk. The developed method was successfully applied to detect the PAs-containing Chinese herbal medicines. This study provides convincing data that can support risk management actions in China and a meaningful reference for the rational and safe use of Chinese herbal medicines.

Urinary and fecal excretion of aromatic amines in pet dogs and cats from the United States

Abstract:Several primary aromatic amines (AAs) are known or suspected carcinogens. Despite this, the exposure of pet animals to this class of chemicals is unknown. In this study, we investigated the occurrence of 30 AAs and two tobacco chemical markers (nicotine and cotinine) in 63 pet urine (42 dog and 21 cat) and 77 pet feces (37 dog and 40 cat) samples collected from the Albany area of New York State. Eight of the 30 AAs (∑8AAs) were found in > 38% of dog and cat urine samples, at median concentrations of 7.99 (range: 0.42-52.3 ng/mL) and 31.4 (2.63-75.9) ng/mL, respectively. Nine of the 30 AAs (∑9AAs) were found in > 73% of dog and cat feces samples, at median concentrations of 278 (range: 61.7-613 ng/g) and 240 (55.4-645) ng/g dry wt, respectively. Among the 30 AAs, 2,6-dimethylaniline (2,6-DMA) accounted for the highest median concentrations in both urine and fecal samples. Median concentrations of nicotine and cotinine were below 0.92 ng/mL in urine and below 3.86 ng/g in feces of both dogs and cats. No significant relationship was found between AA concentrations and pet age or gender. The lack of significant Spearman's rank correlation between the concentrations of AA and nicotine in pet urine/feces suggested that sources other than tobacco smoke contributed to AA exposure in pets. Furthermore, the calculated fecal excretion rates of AAs were higher than the intake rates (estimated through reverse dosimetry), which indicates that cats and dogs are exposed to AA precursors such as azo dyes. Concentrations in urine and feces reflected exposure to direct and indirect exposure sources, respectively, of AAs.
Study of P-Phenylenediamine (PPD) Concentrations after Hair Dye Mixing: A Call for Safety Reassessment

Abstract:Para-phenylenediamine (PPD) is a chemical that is widely used in hair dyes. Multiple safety and regulatory agencies have categorized PPD as a potent sensitizer. In addition, PPD has carcinogenicity and genotoxicity attributes and, consequently, it is regulated at a maximal concentration of 2.0%. The aim of this study was to test whether the limit for PPD is surplus, and hence whether the consumer may be exposed to unnecessarily PPD levels. Experimentally, the analysis of PPD was performed using HPLC, where method validation and an inter-laboratory comparison test (ILC) were conducted to evaluate method performance. Thirty-three commercial products were analyzed, and five products were chosen to study the unconsumed PPD. Successfully, the implemented method confirmed its suitability and validity for the determination of PPD. For ILC results, PPD levels were 0.97 ± 0.04% and 0.92 ± 0.02%, quantified by our laboratory and an accredited laboratory, respectively. For all products, the initial concentration (T0) of PPD was lower than the regulatory limit. After 45 min, the content of PPD significantly reduced compared to T0. One product showed unconsumed PPD to be as high as 96% following the recommended dyeing time. In conclusion, the existence of high levels of unreacted PPD increases the likelihood of allergic events and elevates the risk of PPD-related chemicals. Collaborative efforts between industries, regulatory bodies, and health-related decision makers are deemed necessary to establish safe concentrations for PPD.

A molecularly imprinted polymers (MIPs) solid phase extraction (SPE) cartridge was applied to purify urine samples and no derivatization reaction was involved. Each analytes used respective stable isotope internal standards, which could well compensate matrix effect. Lower limit of detections (LODs) for four AAs were obtained and in the range of 1.5–5ngL−1. Recovery ranged from 87.7±4.5% to 111.3±6.4% and precision were less than 9.9%. The method was applied to analyze urine samples of 40 smokers and 10 nonsmokers. The 24h urinary excretion amounts of total AAs were higher for smokers compared with nonsmokers. What's more, 1-NA, 3-ABP and 4-ABP excretion amounts showed significant differences (p<0.05) between smokers and nonsmokers.
Determination of primary aromatic amines from cooking utensils by capillary electrophoresis-tandem mass spectrometry

Abstract:This paper describes a fast, sensitive, environment-friendly method for the determination of 19 primary aromatic amines (PAAs) in cooking utensils by capillary zone electrophoresis coupled with tandem mass spectrometry. The best electrophoretic separation of PAAs was obtained in 0.1 mol l-1 formic acid (pH 2.4) as the background electrolyte, fused silica capillary (67 cm) with a run time below 6 min. The proposed method presented a linear calibration with correlation coefficients higher than 0.99 and reproducibility in a range of 1-25%. Limits of detection were in the range of 0.2-1.3 μg kg-1 and recoveries were in a range of 85-120% for all the PAAs. The validated method was employed to determine PAAs on 36 samples of cooking utensils using acetic simulant. The results showed that 4,4'-diaminodiphenylmethane and aniline being the most frequently found PAAs in these samples and 28% of cooking utensils were not compliant.

Abstract:This review examines the environmental occurrence and fate of aromatic amines (AAs), a group of environmental contaminants with possible carcinogenic and mutagenic effects. AAs are known to be partially responsible for the genotoxic traits of industrial wastewater (WW), and AA antioxidants are acutely toxic to some aquatic organisms. Still, there are gaps in the available data on sources, occurrence, transport, and fate in domestic WW and indoor environments, which complicate the prevention of adverse effects in aquatic ecosystems. We review key domestic sources of these compounds, including cigarette smoke and grilled protein-rich foods, and their presence indoors and in aquatic matrices. This provides a basis to evaluate the importance of nonindustrial sources to the overall environmental burden of AAs. Appropriate sampling techniques for AAs are described, including copper-phthalocyanine trisulfonate materials, XAD resins in solid-phase extraction, and solid-phase microextraction methods, which can offer insights into AA sources, transport, and fate. Further discussion is provided on potential progress in the research of AAs and their behavior in an aim to support the development of a more comprehensive understanding of their effects and potential environmental risks.

Abstract:Polycyclic aromatic hydrocarbons (PAHs), a class of harmful and persistent organic contaminant, are widely distributed in the environment and eventually accumulated in water and food. Also, they are formed in different varieties and varying amounts during processing of food depending on the food composition, cooking method and processing condition. According to the International Agency for Research on Cancer (IARC), various PAHs are classified under Group 1 to 3 category, with Group 1 designated as carcinogenic to humans, Group 2A as probable carcinogen, Group 2B as possible carcinogen and Group 3 as noncarcinogenic. Therefore, it is imperative to develop rapid and highly sensitive analytical methods for determination of PAHs in food and water. This article aims to overview the recent advances of various chromatographic methods as well as electrochemical and SERS-based optical sensing methods for analysis of PAHs in food and water. Initially, several conventional sample preparation methods along with the advanced extraction for isolation of PAHs were summarized, followed by reviewing various gas chromatographic methods coupled with various detection techniques for PAHs analysis in various food products including meat/meat products, seafood, oil, milk/milk products, baby foods, honey, vegetable, cocoa products, tea/coffee, juice, rice, flour, noodle and cake. In addition, high performance liquid chromatographic methods coupled with fluorescence, diode array or mass/tandem mass detection techniques as well as an emerging supercritical fluid chromatographic technique employed for determination of PAHs in different food and water matrices were also overviewed. Finally, various electrochemical sensors and SERS-based optical sensors developed recently for onsite detection of PAHs were tabulated and discussed. Thus, this review article can provide a research update on chromatography and sensor-based analytical methods for PAH analysis as well as enable elucidation of research gaps for future studies.
[GC-MS Analysis of Tris(1-aziridinyl)phosphine Oxide Flame Retardants in Textile Products]

Abstract:Tris(1-aziridinyl)phosphine oxide (APO) used as flame retardant in textile products, such as curtains, carpets, and sleeping clothes, is prohibited in Japan under the "Act on the Control of Household Products Containing Harmful Substances." This study developed a GC-MS-based method to quantify APO more accurately and safely than the current official method. The APO in textile products was extracted with methanol, the extract was replaced with acetone instead of hexane as previously reported, and purified by florisil cartridge column. This cleanup method was instead of the harmful and carcinogenic dichloromethane used for open column to purify the sample in the official method, giving consideration to health of analysts. For accurate and sensitive quantification, deuterated compound, APO-d12, was used as a surrogate standard. The calibration curve displayed linearity within the 0.01-2.0 µg/mL range for APO. The detection limit for APO was 0.008 µg/g with S/N=5, which was 50 times more sensitive than the current detection limit of 0.4 µg/g, enabling the analysis of sufficiently low concentrations. The recoveries in non-treatment cloth and flame-retardant textiles were 73.5-126.6% and relative standard deviations were 3.3-24.6% when 2 µg APO was added to 0.5 g of samples, confirming that it can be analyzed satisfactorily. Thus, the developed method is applicable to textile products of various materials.

Abstract:Natural and synthetic dyes are added to different food commodities to enhance their appearance and acceptance by consumers. Acute and chronic exposure owing to the consumption of non-permitted dyes may lead to health concerns such as allergic reactions, eczema, and asthma. 4-(Dimethylamino)azobenzene (4-DMAAB) is a non-permitted dye that has been reported in adulterated mustard oil. Consumption of 4-DMAAB poses severe risks due to its mutagenic and carcinogenic properties. Several sensitive methods such as FT-NIR, FT-MIR and SERS are available for the detection of 4-DMAAB. Here, a spectrophotometric method was developed for the detection of 4-DMAAB. The developed method was translated to a point-of-test paper-based, chromogenic strip which showed a detection limit of 0.025 mM for 4-DMAAB. Also, an electrochemical sensor was developed by electro-depositing the test solution on a screen-printed electrode. The electrochemical sensor showed an LOD of 0.027 ± 0.008 mM with recovery in the range of 91–107% of 4-DMAAB. Oil samples collected from the market were processed by liquid–liquid extraction and the content of 4-DMAAB was assessed. The developed point-of-use sensors for the detection of 4-DMAAB have potential for use by the consumers, food industry and regulatory agencies for on-site analysis and assuring the quality of edible oils.
