Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.

Abstract:Polymer-drug conjugates are widely investigated to enhance the selectivity of therapeutic drugs to cancer cells, as well as increase circulation lifetime and solubility of poorly soluble drugs. In order to direct these structures selectively to cancer cells, targeting agents are often conjugated to the nanoparticle surface as a strategy to limit drug accumulation in non-cancerous cells and therefore reduce systemic toxicity. Here, we report a simple procedure to generate biodegradable polycarbonate graft copolymer nanoparticles that allows for highly efficient conjugation and intracellular release of S-(+)-camptothecin, a topoisomerase I inhibitor widely used in cancer therapy. The drug-polymer conjugate showed strong efficacy in inhibiting cell proliferation across a range of cancer cell lines over non-cancerous phenotypes, as a consequence of the increased intracellular accumulation and subsequent drug release specifically in cancer cells. The enhanced drug delivery towards cancer cells in vitro demonstrates the potential of this platform for selective treatments without the addition of targeting ligands.

Abstract:Three kinds of amphiphilic copolymer, that is, poly(ε-caprolactone)-SS-poly(ethylene glycol) (PCL-SS-PEG), poly(ε-caprolactone)-polyethylenimine (PCL-PEI), and poly(ε-caprolactone)-polyethylenimine-folate (PCL-PEI-Fol) were synthesized and self-assembled into surface engineered hybrid nanoparticles (NPs). Morphological studies elucidated the stable, spherical, and uniform sandwich structure of the NPs. PCL-PEI and PCL-SS-PEG segments have introduced pH and reduction responsive characteristics in these NPs, while PCL-PEI-FA copolymers could provide specific targeting capability to cancer cells. The stimuli responsive capabilities of these NPs were carried out. Negative-to-positive charge reversible property, in response to the pH change, was investigated by zeta potential and nuclear magnetic resonance (NMR) measurements. The structure cleavage, due to redox gradient, was studied by dynamic light scattering (DLS) and transmission electron microscopy (TEM). These NPs showed controlled degradation, better drug release, less toxicity, and effective uptake in MCF-7 breast cancer cells. These multifunctional NPs showed promising potential in the treatment of cancer.

Abstract:Abstract Gemcitabine, a nucleoside antimetabolite, is a widely known chemotherapeutic agent being used as a first line treatment alone or in combination with other chemo drugs for different types of cancer. However, due to its extremely short half-life in the body caused by enzymatic degradation and poor pharmacokinetics, the I.V. dosage of administration is quite high i.e., >1000 mg/m2 for metastatic breast cancer treatment. Moreover, gemcitabine in combination with anthracyclines imparts synergy but free anthracyclines are reported to be associated with long term cardiac toxicity. Nanoparticle-based drug delivery systems have provided a befitted solution for improving the poor pharmacokinetic properties and systemic toxicity caused by the chemo drugs because of their ability to improve bioavailability of the drugs at the desired site as well as overcoming multi-drug resistance (MDR).In the present work, PLA-based biodegradable block copolymeric nanoparticles have been employed to co-load Gemcitabine and Pirarubicin for improved anti-cancer activity. Gemcitabine and Pirarubicin have been chemically conjugated to the amphiphilic block copolymer mPEG PLA via a pH responsive Schiff’s Base linkage using a linker molecule Levulinic acid. The prepared PIRA/GEM co-loaded nanoparticles exhibited an optimum size, zeta potential and polydispersity index i.e., 187 ± 3.5 nm, -10 ± 2.0 and < 0.1 PDI respectively. Further, in the drug release experiments, these Nps displayed sustained and higher drug release in lysosomal pH (pH 5.0) than in the physiological buffer environment (pH 7.4) attributed to the pH sensitive bonding. The co-loaded Nps were prepared in multiple ratios (Pira: Gem - 1:1, 1:3, and 3:1) and investigated for in-vitro cellular inhibition studies in comparison to singly loaded drug Nps and corresponding free drug formulations in various breast cancer cell lines. Additionally, apoptotic studies revealed significant enhancement in synergistic activity in the case of PIRA/GEM co-loaded Nps in comparison to conventional free drug formulations. In conclusion, this nano-formulation demonstrated excellent anti-cancer activity against breast cancer cells therefore it may be used as a potential candidate for therapeutic use. Abstract:In this study, a new class of amphiphilic methoxy-poly(ethylene glycol) grafted polyphosphazene with glycine ethyl ester side groups (PPP-g-PEG/GlyEt) was synthesized and characterized. An anti-cancer agent doxorubicin (DOX) was encapsulated into polymeric micelles derived from those copolymers, which exhibited considerably strong impact on micelle morphology: turned the rod-like and spherical drug free micelles into spheres and vesicles respectively. The in vitro release behavior of those drug-loaded micelles exhibits a sustained release manner and is affected by drug content. Cytotoxicity assay against adriamycin-resistant human breast cancer MCF-7 cell line showed that drug-loaded micelles based on PPP-g-PEG/GlyEt micelles can effectively suppress cell proliferation and the cytotoxicity was both time and concentration related, an enhanced cytotoxicity was observed either with increasing drug concentration or with prolonged incubation time. Moreover, flow cytometry results revealed a particle size dependency in cellular uptake of drug-loaded micelles. These findings suggest that the present copolymers can encapsulate water insoluble anti-cancer agents and contribute to improve drug sensitivity of adriamycin-resistant cell line.

Abstract:Abstract Nanoparticles based drug delivery systems have provided a befitted solution for the poor pharmacokinetic properties and systemic toxicity caused by the chemo drugs because of their ability to improve bioavailability of the drugs at the desired site and thereby decreasing extracellular toxicity. The present work investigates the single or dual chemo drugs loaded PLA-based biodegradable polymeric nanoparticles for improved anti-tumor efficacy as compared to free drug formulations. For the study, an amphiphilic block copolymer; mPEG PLA has been synthesized and characterized using Gel permeation chromatography (GPC) for its molecular weight and poly dispersity index as well as with Nuclear magnetic resonance (NMR) and Fourier-transform Infrared (FT-IR) spectroscopies. Thereafter, two different chemo drugs Pirarubicin (PIRA, THP analogue of Doxorubicin) and Gemcitabine (GEM) have been chemically conjugated individually to mPEG PLA block copolymer via a linker molecule, Levulinic acid to give an acid labile polymer-drug conjugate and the % drug conjugation efficiency has been calculated using High performance liquid chromatography (HPLC). The synthesized polymer-drug conjugates have been employed to prepare PIRA/GEM single or dual-loaded nanoparticles via nanoprecipitation technique and the physiochemical properties including size, zeta potential, and stability were analyzed using Dynamic Light Scattering (DLS) and High-resolution transmission electron microscopy (HR-TEM). To carry out toxicity assessment of the mPEG PLA nanoparticles, both cytocompatibility and hemocompatibility were checked using MTT assay and Hemolysis assay respectively. Moreover, the cellular internalization studies of dye-loaded mPEG PLA nanoparticles were done using Confocal microscopy on SUM-149, breast cancer cells. Besides, the drug release kinetic studies of the free PIRA/GEM from the PIRA/GEM single or dual-loaded nanoparticles were carried out in different pH environments; pH 7.4 and pH 5.0. The in-vitro cell proliferation inhibition studies of PIRA/GEM or dual-loaded nanoparticles were carried out on different human and murine breast cancer cell lines including MCF-7, MDA-MB-468, SUM 149, MDA-MB-231 and 4T1 and half-maximal inhibitory concentrations (IC50 values) were compared to that of respective free drug formulations. The overall results suggested that the prepared sub-nano sized PIRA/GEM single or dual-loaded particles are highly stable, uniformly spherical in size and biocompatible in nature. They were found to possess high cellular uptake, sustained drug release rate and synergistic efficacy and potential against both human and murine breast cancer cells in-vitro conditions. Abstract:Platinum (II) demonstrates potent anticancer activity, but it is hampered by drawbacks such as poor solubility, facile decomposition, bone marrow suppression, and nephrotoxicity, which severely restrict its clinical use. To overcome these limitations, polymer prodrugs were developed by conjugation of platinum drugs onto polymers. The protective shield provided by polymers enables drugs to evade enzymatic degradation and immune system attacks in the physiological environment, achieving sustained release and long-term efficacy. In this study, we synthesized novel vinyl- or alkynyl-substituted cysteine N -carboxyanhydride (NCA) monomers, followed by the preparation of amphiphilic poly(ethylene glycol)-poly(amino-acid) copolymers through the copolymerization of PEG with the NCAs. Next, 1,2-didentate dicarboxylic groups were introduced into the side chains of the block copolymer by employing the "Thiol-ene" or "Thiol-yne" reactions. These carboxylic acid groups were then chelated with (1R,2R)-1,2-diaminocyclohexane platinum (II)(DACH-Pt) and simultaneously assembled into nanoparticles. The morphology and particle size were characterized using transmission electron microscopy (TEM) and dynamic light scattering (DLS). Remarkably, the chelation of platinum (II) led to a decrease in micellar size. In vitro drug release experiments demonstrated a significantly faster release rate in the intracellular environment (pH 5.0) compared to physiological conditions (pH 7.4). MTT assays confirmed the excellent biocompatibility of the polymer. Moreover, concentration-dependent inhibition of cancer cell growth was observed with poly(amino acid)-platinum (II) chelate micelles. Overall, these findings suggest that self-assembled nanoparticles of poly(amino acid)-platinum (II) chelates hold immense potential for drug delivery applications.

Abstract:Polymeric nanocapsules hold considerable applications in cancer drug delivery, but the synthesis of well-defined nanocapsules with a tunable drug release property remains a significant challenge in fabrication. Herein, we demonstrate a supramolecular complexation strategy to assemble small molecular platinum (Pt) compounds into well-defined nanocapsules with high drug loading, acidity-sensitivity, and tunable Pt releasing profile. The design utilizes poly(ethylene glycol)-dendritic polylysine-G4/amides to complex with Pt compounds, forming stable nanocapsules with diameters approximately ∼20 nm and membrane thickness around several nanometers. The stability, drug content, and release profiles are tunable by tailoring the dendritic structure. The designated polymer-Pt nanocapsules, PEG-G4/MSA-Pt, showed sustained blood retention, preferential tumor accumulation, enhanced cellular uptake, lysosomal drug release, and nuclear delivery capability. PEG-G4/MSA-Pt showed enhanced antitumor efficacy compared to free cisplatin and other nanocapsules, which stopped the progression of both A549 cell xenografts and patient-derived xenografts (PDXs) of hepatocellular carcinoma on a mice tumor model. Thus, we believe this strategy is promising for developing Pt-based nanomedicine for cancer drug delivery.

Abstract:Poly (ethylene glycol) methyl ether-block-poly (Ɛ-caprolactone) copolymers are useful biomedical materials owing to their amphiphilic nature, biodegradability, biocompatibility and also due to their semi-crystalline form having a low glass transition temperature. Due to their slow drug release profile, PEG-PCL copolymers are excellent candidates for sustaineddelivery applications over a period of time of more than a year. This unique property has provoked their relevance specially in the area of anticancer drug delivery applications in the form of various nanoaggregates like microspheres, nanogels, nanospheres, polymersomes, micelles, etc. A large variety of anticancer drugs/bioactives have been encapsulated in PEG-PCL copolymers for developing effective anticancer drug delivery systems and also for producing controlled drug release profiles. In recent times, PEG-PCL copolymers based nanoparticles have shown tremendous advancements as anticancer drug delivery vehicles owing to their higher drug loading capacities of the hydrophobic drugs, enhanced bioavailability, keeping away from being overpowered by way of phagocytes, decreased blasted discharge, and also in increasing the scattering time of drug within the blood stream during their systemic inoculation. These nano-formulated drug delivery systems can be used for the delivery of anticancer drugs at a specific site in a targeted approach. The developed nano-formulations possess promising potential in the treatment of cancer with improved anticancer efficacy and reduced toxicity invivo. In the current review, our main focus is to highlight the development and synthesis of different types of PEG-PCL copolymers (both conventional and greener approaches) along with the physico-chemical characterization techniques used primarily for the PEG-PCL co-polymeric systems. We have also summarized the uses of PEG-PCL co-polymeric nano formulations with various anticancer drugs as drug delivery platforms in the area of anticancer chemotherapy.

Abstract:The focus of this research is to design a bioengineered drug delivery vehicle that is efficient in anti-cancer drug delivery in a controlled manner. The experimental work focuses on constructing a methotrexate-loaded nano lipid polymer system (MTX-NLPHS) that can transport methotrexate (MTX) in MCF-7 cell lines in a controlled manner through endocytosis via phosphatidylcholine. In this experiment, MTX is embedded with polylactic-co-glycolic acid (PLGA) in phosphatidylcholine, which acts as a liposomal framework for regulated drug delivery. Scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and dynamic light scattering (DLS) were utilized to characterize the developed nanohybrid system. The particle size and encapsulation efficiency of the MTX-NLPHS were found to be 198 ± 8.44 nm and 86.48 ± 0.31 %, respectively, which is suitable for biological applications. The polydispersity index (PDI) and zeta potential of the final system were found to be 0.134 ± 0.048 and -28 ± 3.50 mV, respectively. The lower value of PDI showed the homogenous nature of the particle size, whereas higher negative zeta potential prevented the system from agglomeration. An in vitro release kinetics was conducted to see the release pattern of the system, which took 250 h for 100% drug release This kind of system may carry the drug for a long time in the circulatory system and prevent the drug discharge. Other cell culture assays such as 3-(4, 5-dimethyl thiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) and reactive oxygen species (ROS) monitoring were used to see the effect of inducers on the cellular system. MTT assay showed cell toxicity of MTX-NLPHS reduced at the lower concentration of the MTX, however, toxicity increased at the higher concentration of the MTX as compared to free MTX. ROS monitoring c revealed more scavenging of ROS using MTX-NLPHS as compared to free MTX. Confocal microscopy suggested the MTX-NLPHS induced more nuclear elongation with cell shrinkage comparatively.
In Vivo Evaluation of Cisplatin-Loaded PEG-PCL Block Copolymeric Nanoparticles for Anticancer Drug Delivery

Abstract:Background Cisplatin has proved itself an effective therapeutic approach in a variety of solid tumors. However, its therapeutic concentration was limited since the presence of variant inevitable side effects. Nanoparticles (NPs) developed from amphiphilic copolymers have drawn great attention for prolonging drug residency in blood circulation, increasing tumor accumulation, reducing serum protein adherence and preventing uptake by the reticuloendothelial system (RES). Previously, we synthesized polyethylene glycol (PEG)- polycaprolactone (PCL) NPs as anticancer drug and had confirmed its efficacy in vitro. To comprehensively study the influences of cisplatin-loaded PEG-PCL copolymeric NPs, we enlarge the amount of murine models and applied more physical tests to offer a broader view of the impact that cisplatin-loaded PEG-PCL copolymeric NPs brought about. Methods We observed the physical alteration of hepatoma-bearing mice and the tumors after administration of cisplatin-loaded PEG-PCL nanoparticles (NPs). Cell uptake and apoptotic rates were measured through fluorescence microscopy and flowcytometry. Murine blood was collected and examined. Positron emission tomography/computed tomography (PET/CT) was utilized for the detection of tumor metabolic status. Results The biopsy and the measurement of tumor volume and murine weights have confirmed the therapeutic efficacy of cisplatin-loaded NPs and was positively related to the dosage of cisplatin. Fluorescence microscope images indicated a better cellular uptake rate of cisplatin-loaded NPs with intracellularly-aggregated, heterogeneous rhodamine B observed in LoVo cells. A decline of apoptotic rates in cisplatin-loaded NPs was measured. Blood tests exhibited less side effects in cisplatin-loaded NPs with higher level of white blood cells and lower aspartate transaminase (AST). PET/CT images revealed less intensity of FDG uptake in murine received higher dosage of cisplatin-loaded NPs. Conclusions Nanoparticles prepared from PEG-PCL/PCL hybrids could be a promising drug carrier for intratumoral delivery with greater efficacy and less side effects. This study provides a theoretical basis for possible clinical application. Abstract:Combination therapy using chemically distinct drugs has appeared as one of the promising strategies to improve anticancer treatment efficiency. In the present investigation, poly-(lactic-co-glycolic) acid (PLGA) nanoparticles electrostatically conjugated with polyethylenimine (PEI)-based co-delivery system for epirubicin and paclitaxel (PLGA-PEI-EPI-PTX NPs) has been developed. The PLGA-PEI-EPI-PTX NPs exhibited a monodispersed size distribution with an average size of 240.93 ± 12.70 nm as measured through DLS and 70.8–145 nm using AFM. The zeta potential of 41.95 ± 0.65 mV from −17.45 ± 2.15 mV further confirmed the colloidal stability and PEI modification on PLGA nanoparticles. Encapsulation and loading efficiency along with in vitro release of drug for nanoparticles were done spectrophotometrically. The FTIR analysis of PLGA-PEI-EPI-PTX NPs revealed the involvement of amide moiety between polymer PLGA and PEI. The effect of nanoparticles on the cell migration was also corroborated through wound healing assay. The MTT assay demonstrated that PLGA-PEI-EPI-PTX NPs exhibited considerable anticancer potential as compared to the naïve drugs. Further, p53 protein expression analysed through western blot showed enhanced expression. This study suggests that combination therapy using PLGA-PEI-EPI-PTX NPs represent a potential approach and could offer clinical benefits in the future for lung cancer patients.

Abstract:Using the laser ablation method, the study explores the synthesis and characterisation of three nanocomposites: titanium dioxide (TiO2)/polyethylene glycol (PEG), silver (Ag)/titanium dioxide (TiO2)/polyethylene glycol (PEG) and silver (Ag)/titanium dioxide (TiO2)/chitosan (Ct's). These composites act as delivery systems for 5-Fluorouracil (5Fu), an anticancer medication. Various characterization techniques were utilised, such as Fourier transform infrared spectroscopy, UV-visible, X-ray diffraction analysis and transmission electron microscopy. The results show that the freshly produced composites have chemical interactions; the Ag-TiO2-PEG-5Fu composite has the highest crystallinity. Particle size distribution has an average size of 45 nm and a range of 10 to 100 nm. The UV-visible absorption spectrum shows a clear blue shift, which improves the possibility for medication delivery. Through cytotoxicity tests on the AMN3 cell line, the study illustrates the promise of nanocomposites in pharmaceutical applications, particularly in boosting the efficacy of cancer therapy. The co-administration of polymers with 5-FU augments their cytotoxic effect, indicating viable substitutes for the treatment of cancer. The AgTiO2 PEG 5-Fu composite, in particular, has the strongest cytotoxic effects, suggesting that it is a viable therapy option for cancer. PEGylation improves medication dispersion, which raises cytotoxicity slightly. Chitosan demonstrates inherent anti-tumorogenic properties at particular dosage ranges. Furthermore, decreasing cytotoxicity by doubling the content of silver (Ag) in the AgTiO2 PEG 5-FU composite may be attributed to Ag particle agglomeration. It's interesting to note that when compared to the AgTiO2 composite, the TiO2 composite shows noticeably more cytotoxicity. All things considered, the study highlights the potential of nanocomposites in drug delivery applications, especially when it comes to improving the administration of anticancer drugs. The combination of polymers, nanometallic materials and 5-FU offers bright futures for cancer treatment; positive results have been seen even in created composites that do not include 5-FU.

Abstract:Abstract The practical application of nanoparticles (NPs) as chemotherapeutic drug delivery systems is often hampered by issues such as poor circulation stability and targeting inefficiency. Here, we have utilized a simple approach to prepare biocompatible and biodegradable pH-responsive hybrid NPs that overcome these issues. The NPs consist of a drug-loaded polylactic-co-glycolic acid (PLGA) core covalently ‘wrapped’ with a crosslinked bovine serum albumin (BSA) shell designed to minimize interactions with serum proteins and macrophages that inhibit target recognition. The shell is functionalized with the acidity-triggered rational membrane (ATRAM) peptide to facilitate internalization specifically into cancer cells within the acidic tumor microenvironment. Following uptake, the unique intracellular conditions of cancer cells degrade the NPs, thereby releasing the chemotherapeutic cargo. The drug-loaded NPs showed potent anticancer activity in vitro and in vivo while exhibiting no toxicity to healthy tissue. Our results demonstrate that the ATRAM-BSA-PLGA NPs are a promising targeted cancer drug delivery platform.

Abstract:The poly(l-glutamic acid)-cisplatin nanoformulations with detachable PEGylation are developed for cancer therapy.DePEGylation of nanoformulations triggered by tumor microenvironment exhibit enhanced tumor cell internalization.The dePEGylation strategy exhibits promising prospects for clinical application of nanomedicines.PEGylation has been widely applied to prolong the circulation times of nanomedicines via the steric shielding effect, which consequently improves the intratumoral accumulation. However, cell uptake of PEGylated nanoformulations is always blocked by the steric repulsion of PEG, which limits their therapeutic effect. To this end, we designed and prepared two kinds of poly(l-glutamic acid)-cisplatin (PLG-CDDP) nanoformulations with detachable PEG, which is responsive to specific tumor tissue microenvironments for prolonged circulation time and enhanced cell internalization. The extracellular pH (pHe)-responsive cleavage 2-propionic-3-methylmaleic anhydride (CDM)-derived amide bond and matrix metalloproteinases-2/9 (MMP-2/9)-sensitive degradable peptide PLGLAG were utilized to link PLG and PEG, yielding pHe-responsive PEG-pHe-PLG and MMP-sensitive PEG-MMP-PLG. The corresponding smart nanoformulations PEG-pHe-PLG-Pt and PEG-MMP-PLG-Pt were then prepared by the complexation of polypeptides and cisplatin (CDDP). The circulation half-lives of PEG-pHe-PLG-Pt and PEG-MMP-PLG-Pt were about 4.6 and 4.2 times higher than that of the control PLG-Pt, respectively. Upon reaching tumor tissue, PEG on the surface of nanomedicines was detached as triggered by pHe or MMP, which increased intratumoral CDDP retention, enhanced cell uptake, and improved antitumor efficacy toward a fatal high-grade serous ovarian cancer (HGSOC) mouse model, indicating the promising prospects for clinical application of detachable PEGylated nanoformulations.Download : Download high-res image (319KB)Download : Download full-size image

Abstract:In this study, we aim to synthesize self-assembled amphiphilic diblock poly(PEGA- b -HEA-PCL) copolymers through RAFT living polymerization, targeting the delivery of hydrophobic anticancer drugs. The synthesized self-assembled diblock copolymers polymeric micelles (PMs) comprising poly(ethylene glycol) methyl ether acrylate (PEGA), as a hydrophilic segment and 2-hydroxyethyl acrylate-polyhexanoate monomer (HEA-PCL) with different block lengths, as a hydrophobic segment. The chemical structures, compositions, and self-assembled behavior were identified through 1 H NMR spectroscopy. The thermal stability was assessed through TGA and DSC. Furthermore, DOX was encapsulated into all PMs. The drug-loaded PMs exhibited enhanced drug release profiles in acidic medium. Particle diameter was measured through DLS and TEM techniques. The cell viability of diblock polymers and selected DOX-loaded PMs were evaluated against non-cancerous (L929) and cancerous cells (SK-N-AS), respectively, through well-known MTT assay. Micellar aggregates with mean diameters of approximately 127.2–145.3 nm formed in aqueous solution. The diameters of PMs increased to 141.5–173.1 nm upon the incorporation of DOX. The drug loading content and encapsulation efficiency of PMs were approximately 8.09–18.84% and 30.43–54.07%, respectively. The MTT assay results indicated that all synthesized materials had minimal effects on the viability of L929 cells, while DOX-loaded materials inhibited the viability of neuroblastoma cells by 68.7%. The highest drug release was 89.20% at pH 7.4, while 83.45% at pH 5.0 for 40 h. These findings suggest that the synthesized amphiphilic PMs are promising candidates for drug delivery systems.

Abstract:A unique drug delivery system, in which silver nanoparticles (AgNPs) are covered with camptothecin (CPT)-based polymer prodrug, has been developed, and the polymer prodrug, in which the CPT is linked to the polymer side chains via an acid-labile β-thiopropionate bond, is prepared by RAFT polymerization. For poly(2-(2-hydroxyethoxy)ethyl methacrylate-co-methacryloyloxy-3-thiahexanoyl-camptothecin)@AgNPs [P(HEO2MA-co-MACPT)@AgNPs], the polymer thickness on the AgNP surface is around 5.9 nm (TGA method). In vitro tests in buffer solutions at pH = 7.4 reveal that fluorescence of the CPT in the hybrid nanoparticles is quenched due to the nanoparticle surface energy transfer (NSET) effect, but under acidic conditions, the CPT fluorescence is gradually recovered with gradual release of the CPT molecules from the hybrid nanoparticles through cleavage of the acid-labile bond. The NSET "on" and "off" is induced by the CPT-AgNP distance change. This unique property makes it possible to track the CPT delivery and release process from the hybrid nanoparticles in the living cells in a real-time manner. The internalization and intracellular releasing tests of the hybrid nanoparticles in the HeLa cells demonstrate that the lysosome containing the hybrid nanoparticles displays CPT blue fluorescence due to release of the CPT under acidic conditions, and the drug-releasing kinetics shows fluorescence increase of the released CPT with incubation time. The cytotoxicity of hybrid nanoparticles is dependent on activity of the acid-labile bond. Therefore, this is a potential efficient drug delivery system in cancer therapy and a useful approach to study the mechanism of release process in the cells.

Abstract:Nitric oxide (NO) as a chemosensitizer has attracted a lot of attention in anti-multidrug resistance (MDR) tumor therapy. In this study, a three-segment amphiphilic copolymer was synthesized through click reaction by azido and alkyne groups. The PEI part with abundant secondary amines was utilized to load NO gas molecules, the hydrophobic PLLA core to encapsulate the hydrophobic drug paclitaxel (PTX), and the mPEG shell to provide a hydrophilic interface and lead the copolymer to self-assemble into micelles in the water phase for biological applications. This multifunctional mPEG-PEI-PLLA-PTX-NO nanostructure is able to release NO spontaneously under physiological conditions and enhance the cytotoxicity for synergistic treatment of PTX and NO, demonstrating the potential to treat MDR tumors.

Abstract:A series of amphiphilic pH-responsive poly (ethylene glycol) methyl ether-b-(poly lactic acid-co-poly (β-amino esters)) (MPEG-b-(PLA-co-PAE)) block copolymers with different PLA/PAE ratios were designed and synthesized via a Michael-type step polymerization. The molecular structures of the copolymers were confirmed with (1)H NMR and gel permeation chromatography (GPC). These amphiphilic copolymers were shown to self-assemble into core/shell micelles in aqueous solution at low concentrations, and their critical micelle concentrations (CMC) in water were 1.2-9.5 mg/L. The pH-responsive PAE segment was insoluble at pH 7.4, but it became positively charged and soluble via protonation of amino groups at pH lower than 6.5. The average particle size and zeta potential of micelles increased from 180 nm and 15 mV to 220 nm and 40 mV, respectively, when the pH decreased from 7.4 to 5.0. Doxorubicin (DOX) was loaded into the core of these micelles with a high drug loading of 18%. The in vitro DOX release from the micelles was significantly accelerated when solution pH decreased from 7.4 to 5.0. DOX release in the first 10 h appeared to follow Fickian diffusion mechanism. Toxicity test showed that the copolymers had low toxicity whereas the DOX-loaded micelles remained high cytotoxicity for HepG2 cells. The results indicate the pH-sensitive MPEG-b-(PLA-co-PAE) micelle may be a potential hydrophobic drug delivery carrier for cancer targeting therapy with sustained release.

Abstract:The systemic toxicity, reduced cellular internalization, and uncontrollable intracellular drug release of smart nanoparticles (NPs) still need to be overcome for effective cancer therapy. Herein, a series of stepwise dual pH and redox responsive cross-linked polypeptides based on poly(L-lysine-co-N,Nbis(acryloyl)cystamine-co-g-glutamic acid) (PLBG), were prepared for enhanced cellular uptake and effective cancer therapy. The prepared cross-linked PLBG nanoparticles (PLBG-NPs) exhibit negatively charged surfaces under physiological conditions, and the surface charge of the PLBG-NPs was observed to switch from negative to positive in a slightly acidic tumor extracellular environment at a pH level of similar to 6.5. Finally, the endosome escape of the PLBG-NPs was facilitated via a "proton-sponge" effect via protonation of the polymer chain in the endo/lysosome environment of the tumor cell at a pH level of similar to 5.0. Moreover, rapid drug release was triggered by a high concentration of reducing glutathione (GSH) in tumor cells via destruction of the disulfide linkages in the PLBG-NPs. Cytotoxicity assays demonstrated that no noticeable cytotoxicity was observed for the PLBG-NPs. However, DOX-loaded PLBG-NPs (PLBG-NPs-DOX) demonstrated remarkable tumor cell killing effects. In HeLa tumor-bearing mice, PLBG-NPs-DOX showed sustained tumor accumulation and enhanced inhibitory effects on tumor growth and angiogenesis. In conclusion, it is suggested that the as-designed novel charge-conversion PLBG-NPs with stepwise pH-responsivity and biodegradability could be used as a potential drug carrier for effective cancer therapy with less systemic toxicity.
