Abstract:Due to the widespread utilization of antiepileptic drugs for seizure control and patient well-being, it is crucial to determine their presence in biological and water samples. This is essential for optimizing therapy, comprehending drug behavior, identifying drug interactions, monitoring environmental contamination, and safeguarding public health and ecology. Hence, a novel method that combines dispersive micro solid phase extraction with HPLC-DAD was developed to simultaneously determine the presence of Lacosamide, Levetiracetam, and Phenytoin. To achieve this, a new sorbent was synthesized using the hydrothermal and sol–gel methods, which consisted of bimetallic MIL-100 (Fe, Ni) on MIL-100 (Mn), SiO 2 , and Fe 3 O 4 nanoparticles. We compared the extraction capabilities of several sorbents in order to select the most effective one. The optimal ratio of components for the chosen sorbent was determined through the simplex lattice design to enhance its efficiency. The optimized values for SiO 2 , Fe 3 O 4 , and MIL-100 (Fe, Ni) on MIL-100 (Mn) were found to be 0.1, 0.34, and 0.56, respectively. The parameters that affected the dispersive micro solid phase extraction method were optimized using both experimental design and one factor at a time. Under the optimal conditions, the linear range for the detection of Lacosamide, Levetiracetam, and Phenytoin was determined to be 0.1–363.0, 0.3–372.0, and 0.4–435.0 ng mL −1 , respectively, with low LODs below 0.13 ng mL −1 . Additionally, a preconcentration factor of 492.6, 487.1, and 469.5 was achieved for measuring Lacosamide, Levetiracetam, and Phenytoin, respectively. The intra-day and inter-day relative standard deviations (RSDs) ranged from 3.8 % to 4.6 % and 4.2 % to 5.2 %, respectively. The recoveries and RSDs for analyzing real water, human urine, and serum samples were found to be between 89.6–97.0 % and 4.53–6.44 %, respectively.

Abstract:Highly selective molecularly imprinted polymers (MIPs) for solid-phase extraction and determination of furosemide in human plasma have been designed and prepared. In order to study the intermolecular interactions in the pre-polymerization mixture and to find a suitable functional monomer in MIP preparation, a computational approach was developed. It was based on the comparison of the binding energy of the complexes between the template and functional monomers. Having confirmed the results of computational method, three MIPs were synthesized with different functional monomers, i.e. acrylamide (AAM), 4-vinylpiridine (4-VP) and acrylonitrile (ACN), and then evaluated using Langmuir-Freundlich (LF) isotherm. Using the MIP prepared by AAM as functional monomer, a molecularly imprinted solid-phase extraction procedure followed by high performance liquid chromatography with ultraviolet detector (MISPE-HPLC-UV) was developed for selective extraction and determination of furosemide in human plasma. For the proposed MISPE-HPLC-UV method, the linearity between responses (peak area) and concentration was found over the range of 75-3500 ng mL(-1) with a linear regression coefficient (R(2)) of 0.997. The limit of detection (LOD) and quantification (LOQ) in plasma were 12.9 and 43.3 ng mL(-1), respectively.

Abstract:A simple and efficient bioanalytical method for simultaneous determination of phenobarbital (PB), phenytoin (PHT), carbamazepine (CBZ), and its active metabolite carbamazepine 10,11-epoxide (CBZE) in human plasma using online solid phase extraction (SPE)-liquid chromatography (LC) coupled with high resolution mass spectrum (HRMS) under targeted MS/MS (t-MS(2)) analysis mode has been developed. The procedure integrated an automated sample clean-up of human plasma by Oasis®HLB SPE cartridge, a separation by ZORBAX SB-C18 analysis column, and a quantification by Q-Exactive Hybrid Quadrupole-Orbitrap. The total running time was 13min. The lower limit of quantification (LLOQ) of PB, PHT, CBZ, and CBZE were 0.008, 0.008, 0.0016 and 0.0016μgmL(-1) respectively and the linearities were in the range of 0.008-2.500, 0.008-2.500, 0.0016-0.500 and 0.0016-0.500μgmL(-1) respectively. The mean recovery was between 91.82% and 108.27% and the matrix effect was between 93.29% and 102.09%. The relative standard deviations of interday and intraday were less than 6.41%. The method has been successfully applied in therapeutic drug monitoring (TDM) of four Chinese epilepsy patients. This fully automated, simple, sensitive and reliable online-SPE-LC-HRMS/MS method serves well for TDM of PB, PHT, CBZ and CBZE at clinics for either single or combination treatment.

Abstract:Objective:To establish a method for simultaneous determination of three consitituents in AEC Tablet containing as- pirin,ethenzamide and caffeine.Method:Inertsil-ODS column is used.The mobile phase is composed of methanol -0.1% di- ethylamine-acetic acid(40:56:4)with flow rate of 1.0mL/min.The detection wavelength is 280nm.Results:The linear re- sponse range were 120-480μg/mL for aspirin(r=0.9998),40-160μg/mL for ethenzamide(r=0.9998),20-80μg/mL for caf- feine(r=0.9997).The mean recoveries of aspirin,ethenzamide,caffeine were 100.3%,100.7%,99.7% respectively.Conclu- sion:The method is simple,quick and suitable for determination of these three constituents in compound preparation simulta- neously.
Rapid high-performance liquid chromatographic determination with fluorescence detection of furosemide in human body fluids and its confirmation by gas chromatography-mass spectrometry

Abstract:Furosemide (FD: Lasix) is a loop diuretic which strongly increases both urine flow and electrolyte urinary excretion. Healthy volunteers were administered 40 mg orally (dissolved in water) and concentrations of FD were determined in serum and urine for up to 6 h for eight subjects, who absorbed water at a rate of 400 ml/h. Quantification was performed by HPLC with fluorescence detection (excitation at 233 nm, emission at 389 nm) with a limit of detection of 5 ng/ml for a 300-microliters sample. The elution of FD was completed within 4 min using a gradient of acetonitrile concentration rising from 30 to 50% in 0.08 M phosphoric acid. The delay to the peak serum concentration ranged from 60 to 120 min. FD was still easily measurable in the sera from all subjects 6 h after administration. In urine, the excretion rates reached their maximum between 1 and 3 h. The total amount of FD excreted in the urine averaged 11.2 mg (range 7.6-14.0 mg), with a mean urine volume of 3024 ml (range 2620-3596 ml). Moreover, the urine density was lower than 1.010 (recommended as an upper limit in doping analysis to screen diuretics) only for 2 h. An additional volunteer was administered 40 mg of FD and his urine was collected over a longer period. FD was still detectable 48 h after intake. Gas chromatography-mass spectrometry with different types of ionization was used to confirm the occurrence of FD after permethylation of the extract. Negative-ion chemical ionization, with ammonia as reactant gas, was found to be the most sensitive method of detection.
Salting-out assisted liquid-liquid extraction coupled with hydrophilic interaction chromatography for the determination of biguanides in biological and environmental samples

Abstract:A new salting-out assisted liquid-liquid extraction (SALLE) sample preparation method for the determination of the polar anti-diabetic biguanide drugs (metformin, buformin and phenformin) in blood plasma, urine and lake water samples were developed. The SALLE was performed by mixing samples (plasma (0.2mL), urine or lake water (1.0mL)) with acetonitrile (0.4mL for plasma, 0.5mL for urine or lake water), sodium hydroxide powder was then added for the phase separation. The effects of type of salting-out reagent, type of extraction solvent, volumes of acetonitrile and sample, amount of sodium hydroxide, vortexing and centrifugation times on the extraction efficiency were investigated. The upper layer, containing the biguanides, was directly injected into a HPLC unit using ZIC-HILIC column (150mm×2.1mm×3.5μm) and was detected at 236nm. The method was validated and calibration curves were linear with r2>0.99 over the range of 20-2000μgL-1 for plasma and 5-2000μgL-1 for urine and lake water samples. The limits of detection were in the range (3.8-5.6)μgL-1, (0.8-1.5)μgL-1 and (0.3-0.8)μgL-1 for plasma, urine and lake water, respectively. The accuracies in the three matrices were within 87.3-103%, 87.4-109%, 82.2-109% of the nominal concentration for metformin, buformin and phenformin, respectively. The relative standard deviation for inter- and intra -day precision were in the range of 1.0-17% for all analytes in the three matrices.
Eco-friendly solvent bar microextraction based on a natural deep eutectic solvent and multivariate optimization for simultaneous determination of spironolactone and canrenone in urine and plasma samples

Abstract:Simultaneous measurement of spironolactone and canrenone in urine and plasma provides valuable insight into renal function, and therapeutic efficacy and can be utilized to identify potential health risks and ensure patient safety throughout treatment. By adopting greener methods to analyze these compounds, significant reductions in the environmental impact of such studies can be achieved. For this purpose, a sensitive and eco-friendly solvent bar microextraction method using natural deep eutectic solvent (NDE) followed by high-performance liquid chromatography-diode array detection (HPLC–DAD) was developed to determine spironolactone and canrenone in urine and plasma samples. The extraction solvents were synthesized using NDE-based terpenoids containing menthol and camphor in various ratios. The extraction efficiency percentage (EE%) of both drugs was measured using response surface methodology (RSM) based on central composite design (CCD), and 29 extraction tests were conducted to determine the optimum conditions. Although all parameters were found to be significant, the extraction and elution times were critical for isolating the target analytes. Under optimized conditions, the linear dynamic ranges for spironolactone (SPI)/canrenone (CAN) were 11.7–10 4 /13.1–10 4 μg L −1 and 21.7–10 4 /24.6–10 4 μg L −1 in urine and plasma samples, respectively with R 2 ≥ 0.993. The ranges of intra-/interprecision (relative standard deviation (RSD) %, n = 5) were 1.31–9.17%/ 2.4–11% with extraction recovery ≥ 88.6% for both drugs. The comparison findings with previously published methods confirmed that the developed NDE-solvent bar microextraction (SBME)-HPLC–DAD method for spironolactone and canrenone analysis displayed confident sensitivity, feasible operation, and simple analysis. Furthermore, the method's applicability and effectiveness were proven by successfully analyzing spironolactone and its metabolite canrenone in patients' urine and plasma samples.

Abstract:A four-part and sustainable nanocomposite composed of carbon quantum dots modified with zeolite imidazolate framework-90, polyvinyl pyrrolidone and magnetite (CQDs/ZIF-90/PVP/Fe3O4) was fabricated and applied in ultrasound-assisted magnetic dispersive micro-solid-phase extraction (US-A-MDMSPE). US-A-MDMSPE was followed by high-performance liquid chromatography with ultraviolet detection (HPLC-UV) for extraction, enrichment and simultaneous low-level monitoring of paracetamol (PCM) and etodolac (EDL) in human plasma. To increase the extraction yield and improve the sensitivity, nanohybrid arrays of metal-organic frameworks and conductive polymers were immobilized on the surface of CQDs followed by magnetization. The nano-extractor was characterized via Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), field-emission scanning electron microscopy (FE-SEM) and energy-dispersive X-ray spectroscopy (EDX). The recent method provided limits of detection (LODs) of 0.16 and 0.09 ng mL-1 for PCM and EDL, respectively. The calibration curve for PCM and EDL was linear in the range of 0.7-2000 ng mL-1 and 0.5-1200 ng mL-1 with a regression (r 2) value between 0.993 and 0.998, respectively. Acceptable precisions including intra-assay (≤6.9%) and inter-assay (≤8.3%) accuracies and notable accuracy (≤8.5%) were achieved to demonstrate the applicability of this method for the evaluation of the pharmacokinetic data of target drugs in human plasma.
Simultaneous determination of ethamsylate, tramadol and lidocaine in human urine by capillary electrophoresis with electrochemiluminescence detection.

Abstract:Ethamsylate, tramadol and lidocaine, partly excreted by the kidney, are generally used as hemostatic, analgesic and local anesthetic in surgery. We developed a simple and sensitive method for their simultaneous monitoring in human urine based on CE coupled with electrochemiluminescence, detection by end-column mode. Under optimized conditions the proposed method yielded linear ranges from 5.0 x 10(-8) to 5.0 x 10(-5), 1.0 X 10(-7) to 1.0 X 10(-4) and 1.0 X 10(-7) to 1.0 X 10(-4) M with LODs of 8.0 x 10(-9) M (36 amol), 1.6 x 10(-8) M (72 amol) and 1.0 x 10(-8) M (45 amol) (S/N = 3) for ethamsylate, tramadol and lidocaine, respectively. The RSD for their simultaneous detection at 1.0 X 10(-6) M was 2.1, 2.8 and 3.2% (n = 7), respectively. For practical application an extraction step with ethyl acetate at pH 11 was performed to eliminate the influence of the sample ionic strength. The recoveries of ethamsylate, tramadol and lidocaine at different levels in human urine were between 87 and 95%. This method was used for simultaneous detection of ethamsylate, tramadol and lidocaine in clinic urine samples from two medicated patients. It was valuable in clinical and biochemical laboratories for monitoring these drugs for various purposes.

Abstract:In the present study, a combination of two useful extraction techniques including fiber solid phase microextraction based on the new synthesized sorbent and the porous hollow fiber liquid phase microextraction is reported. The selective anion-exchangeable Zn-Al-layered double hydroxide as a fiber was electrodeposited on the surface of pencil graphite substrate through simple and cost-effective method. The prepared fiber was characterized and put into the lumen of the porous hollow fiber, which was filled by alkaline solution. The obtained optimum condition for the method was as follows: donor phase HCl concentration of 0.10 mM, acceptor phase NaOH concentration of 7.60 mM and extraction time of 16 min. It is necessary to adjust the concentration of the donor phase in order to extract acidic analytes from an aqueous sample into an organic phase. The method was used for the quantification of three antibiotics including amoxicillin, ciprofloxacin and cefixime. The extracted antibiotics were analyzed by high performance liquid chromatography-ultraviolet detection (HPLC-UV). In the optimal conditions, the linearity of the proposed method was in the range of 0.50-500.00 μg L-1 for selected analytes in water and urine matrices. All calibration curves showed determination of coefficient higher than 0.9958. The limits of detection and limits of quantification were calculated to be between 0.08 and 0.16 μg L-1 and 0.28-0.56 μg L-1, respectively. In order to define the precision of the method, the inter-day and intra-day relative standard deviations were determined between 3.25 and 5.73% in the three selected concentrations. The enrichment factors (EFs) were between 46.20 and 55.81 for selected drugs. Moreover, the calculated absolute recoveries were between 69.30 and 83.71%. The relative recoveries were obtained in the range of 93.00-105.00%. The proposed method was also employed for analysis of various urine samples containing the selected drugs. The obtained recoveries indicated that the method was useful and applicable in complicated biological samples.
Analysis of basic drugs in biological samples using dynamic single-interface hollow fiber liquid-phase microextraction combined with fast electromembrane extraction

Abstract:In this study, a combination of single-interface hollow fiber liquid-phase microextraction and electromembrane extraction was applied for the extraction of some basic drugs from biological samples. The extraction process was followed by the HPLC-UV instrument. In this dynamic system, 1-octanol was impregnated into the pores of hollow fiber wall as an acceptor phase, the sample solution (pH = 12) was pumped into the lumen of hollow fiber by a syringe pump, the extraction efficiency was improved by filling and emptying (25 times) of sample solution (step 1). The extracted analytes in the organic acceptor phase were back-extracted by a fast electromembrane extraction procedure (step 2). Effective parameters on the extraction efficiency of both methods were investigated and optimized. Under optimized conditions (organic acceptor phase: 1-octanol, sample solution volume: 1500 µL, flow rate: 2.0 mL min-1, the number of filling and emptying cycles: 25, voltage: 100 V, back-extraction time: 2 min, and aqueous acceptor phase: 100 mM HCl), the proposed method provided good linearity with determination coefficients ranging from 0.992 to 0.996 over a concentration range of 2.5-1000 ng mL-1. Extraction recoveries were obtained in the range of 65.1 and 88.8%, which resulted in preconcentration factors in the range of 32.6-44.4. The limits of detection were found to be within the range of 0.12-0.36 ng mL-1, while the corresponding repeatability ranged from 3.7 to 9.3% (n = 3). Finally, the optimized method was applied for the quantification of propranolol, diltiazem and, lidocaine in urine and, plasma samples with relative recoveries ranged between 94.1 and 105.4%, indicating the reliability of the method.

Abstract:A sensitive, reproducible, robust, high-throughput ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the simultaneous quantification of fexofenadine and olmesartan in human serum. Samples (50 μL) undergo protein precipitation prior to UPLC-MS/MS analysis. The analytes were separated using an Acquity BEH C18 column (2.1 mm × 50 mm, 1.7 μm) at a flow rate of 0.5 mL/min using a gradient elution with a total run time of 4 min. The analytes were detected in positive ion mode and selected reaction monitoring (SRM) was used for quantitation. The standard curve concentration range was 1.0-500.0 ng/mL for both analytes and each analyte showed excellent linearity with correlation coefficients (R 2 > 0.99). The intra- and inter-day accuracy and precision were ±15% for each analyte, and excellent recovery was demonstrated (93% to 98%) for both analytes. The method is well suited for high-throughput quantitative determination of fexofenadine and olmesartan simultaneously and was successfully applied to an in vivo pharmacokinetic and transporter phenotyping study in humans.

Abstract:Ultrasonic–assisted dispersive micro solid phase extraction (UA-DMSPE) is proposed as a fast and easy technique for the extraction and preconcentration of methadone and tramadol from serum samples. Different sorbents including carbon nanotubes, oxidized carbon nanotubes, and TiO 2 nanoparticles were compared to extract methadone and tramadol. The best performance was obtained using oxidized carbon nanotubes due to the strong affinity between the drugs and carbon nanotube adsorbents. Final analysis of drugs performed by using gas chromatography–mass spectrometric detection. Different parameters affecting the extraction efficiency, such as the sample volume, amount of adsorbent, desorption solvent type and volume, centrifugation time, and speed were investigated and optimized. The striking features of this technique are correlated to its speed and the small volumes of sample (about 1 mL), desorption solvent (about 50 μL), and adsorbent (about 0.001 g) for analysis of drugs, and finally, milder centrifugation conditions relative to the previously reported adsorbent. The optimal parameters were achieved as follows: pH value was set at 9, the sample volume was adjusted to 1200 μL, the amount of adsorbent used was 1 mg, the extraction time was set at 5 minutes, and the volume of the desorption solvent was adjusted to 50 μL. The limits of detections (0.5 and 0.8 ng mL −1 ) and quantifications (1.5 and 2.5 ng mL −1 ) were obtained for methadone and tramadol, respectively. The developed method also showed good repeatability, relative standard deviation (RSD) of 9.49% and 7.47% (n = 5), for the spiked aqueous solution at the concentration level of 10, 50, and 100 ng mL −1 for analytes, and linearity, R≥0.9809. The results showed that UA-DMSPE is a quick, relatively inexpensive, and environmentally friendly alternative technique for the extraction of opiate drugs from serum samples.

Abstract:Amphotericin B (AMB), fluconazole (FZ), and fluorocytosine (FC) are recommended for HIV-associated cryptococcal meningitis (CM) patients as preferred antibiotics. Automated sample clean-up was performed on the human plasma and CSF samples with stop-flow heart-cutting two dimensional (2D) separation using a online-dual-SPE system, allowing retention and accumulation of AMB, FZ, and carbamazepine (CBZ, Internal standard (IS)) by the Oasis®HLB cartridge, and retention and accumulation of FC and 5-methylcytosine hydrochloride (MC, IS) by the HyperSep Hypercarb cartridge respectively. Followed by LC elution, quantification by Q-Exactive Hybrid Quadrupole-Orbitrap with targeted-selected ion monitoring (t-SIM) mode was applied to simultaneously determine the concentrations of AMB, FZ and FC. The bioanalysis was achieved in a total running time of 7min. The method was fully validated according to FDA guidelines. The lowest limit of quantification (LLOQ) was 0.04, 0.04, and 0.40μgmL-1 for AMB, FZ, and FC, respectively. AMB, FZ, and FC levels were linear in the ranges of 0.04-2.00μgmL-1, 0.04-2.00μgmL-1 and 0.40-20.00μgmL-1, respectively. The method showed good performance for human plasma and CSF samples with linearity (R2>0.99), intra-day and inter-day precision (relative standard deviation, RSD<4.32% and <4.06%, respectively), recovery (89.93-93.28% and 90.09-93.58%, respectively) and matrix effect (96.35-103.78% and 92.32-101.48%, respectively). The validated method was successfully applied in real samples of Chinese patients. Overall, our results indicate that this fully automated, sensitive, and reliable online-dual-SPE-LC-HRMS method is effective for therapeutic drug monitoring (TDM) of AMB, FZ, and FC levels.

Abstract:The combined flow injection solid-phase extraction capillary zone electrophoresis (FI-SPE-CZE) system described previously was refined and modified for determination of traces of pseudoephedrine in human plasma samples for pharmacokinetic studies following oral administration. The plasma sample was deproteinized off-line by precipitation with trichloroacetic acid. The supernatant plasma was adjusted to pH 4.2 with acetate buffer and 2 ml of plasma sample was loaded on a C18 micro-column in the FI system. After washing the column with water to remove residual salt, the sample was eluted with a solution of 40% 62.5 mmol l−1 acetate buffer (pH 3.5) and 60% acetonitrile (v/v), and by means of a FI zone-sampling procedure, the most concentrated 45-μl zone of the eluate (preceded by a water zone) was introduced via a carrier and running buffer of 200 mmol l−1 acetic acid-sodium acetate (pH 4.4) containing 0.1% poly(ethylene glycol)-6000 (PEG-6000) into an electrokinetic split-flow sample introduction interface for separation by CZE. The requirements of sensitivity and selectivity for pharmacokinetic studies were met by on-line preconcentration, modification of the sample matrix, increasing the sample volume, and exploiting field-amplified electrostacking effects. A detection limit of 12 μg l−1 (3σ) was achieved at a sample throughput of 10 h−1, and with cimetidine as internal standard, a relative standard deviation of 1.2% (n = 10) at 0.5 μg l−1 was obtained. Recoveries were in the range 95–98% and 92–101% for inter-day and intra-day studies, respectively.

Abstract:Optimization of extraction and chromatographic conditions is essential for the simultaneous analysis of drugs having different physico-chemical properties, especially for in vivo applications. The present work describes concurrent estimation of metformin (MET) and three sodium-glucose co-transporter 2 (SGLT-2) inhibitors namely canagliflozin (CANA), dapagliflozin (DAPA) and empagliflozin (EMPA) in human plasma by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Sample clean-up was optimized using ion-pair solid-phase extraction with sodium lauryl sulphate on Strata-X extraction cartridges. Consistent recoveries were obtained by critically optimizing parameters such as washing and elution solvents during extraction. The extraction recovery ranged from 79 to 88% for all the analytes. Chromatographic separation were accomplished on a Cyano (150 × 4.6 mm, 5 μm) column within 5.0 min using acetonitrile and 10 mM ammonium formate buffer (75:25, v/v) as the mobile phase. The resolution factors between MET-EMPA, MET-DAPA, MET-CANA, DAPA-EMPA and CANA-DAPA were 4.92, 5.85, 7.13, 1.01 and 1.44, respectively. Calibration curves were linear in the concentration range of 2.00-2000, 3.00-3000, 0.20-200 and 1.50-1500 ng/mL for MET, CANA, DAPA and EMPA, respectively. Mass spectrometric analysis was performed using a polarity switching approach to achieve high sensitivity in multiple reaction monitoring mode. The method was validated using current regulatory guidelines and applied to study the pharmacokinetics of fixed-dose formulations of MET and SGLT-2 inhibitors in healthy subjects.

Abstract:A simple, easy to use and efficient method was described for simultaneous determination of ten cardiovascular drugs with a broad range of physicochemical properties in rat plasma via online solid phase extraction (online SPE) and high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Following a simple centrifugation step, a 10 mL aliquot of the plasma sample was injected directly onto the HPLC system. The LiChrospher (R) RP-18 ADS (25 mm x 4 mm, 25 mu m, Merck) cartridge was washed with 10 mM ammonium acetate buffer (pH 9.5) for 1 min, after which time the analytes were removed by back-flushing directly onto the analytical column (Acclaim 120 C18 column, 150 mm x 4.6 mm, 5 mu m) with gradient elution using acetonitrile-10 mM ammonium acetate buffer (pH 3.5) as mobile phase. The flow rate through both columns was 1 mL min(-1), and the analytes were quantified using a triple-quadrupole tandem mass spectrometer in multiple-reaction monitoring mode. Linear calibration curves were obtained over the range of 0.2-100 ng mL(-1), and the method limits ranged from 0.2 ng mL(-1) to 1 ng mL(-1) which is sensitive enough for clinical drug monitoring. The intra-and inter-day precisions were in the range of 0.20-2.32%, and the accuracies were between 93.33% and 114.60%. Excellent recoveries from plasma were achieved with a range from 83.52% to 107.38%. The procedure was easier to execute and required less sample handling than methods previously described in the literature. This easy to use and high-throughput method with direct injection of plasma samples for the analysis of multiple cardiovascular drugs may provide a practical solution for tailoring drug dosage in a rational manner to rapidly achieve optimal efficacy and safety of medication.

Abstract:Metformin (MET) and vildagliptin (VLD) are coformulated in tablets for the management of diabetes mellitus. The aim of this study is the development of a new fast ultraperformance liquid chromatography method with tandem mass detection (UPLC–MS/MS) for their simultaneous determination with 3-amino-1-adamantanol (starting compound for vildagliptin synthesis; VLI) in human plasma. Separation of MET, VLD, and VLI was performed on a 5 cm UPLC-C18 column using a mobile phase of 0.5% acetic acid in methanol and 0.02 M aqueous ammonium acetate (10:90, v/v). The injection volume was 10 µL and electrospray positive ionization was applied. Extraction from human plasma was carried out by acid precipitation of plasma proteins using pregabalin as an internal standard. The assay was validated according to ICH guidelines. The developed method is valid, fast, and simple and was successfully applied in pharmacokinetic studies in human volunteers.

Abstract:This study aimed to preconcentration of sulfamethoxazole (SMX) in water and biological samples. Ultrasound-assisted dispersive liquid-liquid microextraction (UA-DLLME) and ultrasound-assisted dispersive solid-phase microextraction (UA-DSPME) methods paired with spectrophotometry were applied to extraction and preconcentration of SMX. ZnFe 2 O 4 nanoparticles were prepared as adsorbent in UA-DSPME method by hydrothermal method. The scanning electron microscopy (SEM) technique showed that the adsorbent had symmetrical, bullet-shaped particles with uniform size. The results of the X-ray diffraction (XRD) showed the successful synthesis of the ZnFe 2 O 4 nanoparticles. Effective parameters in extraction, including ultrasonication time, disperser solvent volume, adsorbent amount, extraction solvent volume, eluent volume, and pH were investigated and optimized. The practical and optimal conditions of the process were determined by the central composite design (CCD). The optimal conditions were 0.024 g of adsorbent, 535 μL of disperser solvent volume, 7.5 min of ultrasonication time, 235 μL of eluent volume, pH of 5, and 185 μL of extraction solvent volume. Linear ranges and detection limits were 20–1,200 μg L −1 and 6 μg L −1 for UA-DSPME and 10–800 μg L −1 and 3 μg L −1 for UA-DLLME. Relative standard deviation (RSD) of less than 4% were obtained for UA-DSPME and UA-DLLME methods. The reusability showed that the ZnFe 2 O 4 adsorbent could extract SMX up to five cycles of adsorption/desorption without significant reduction in its efficiency. Also, interference studies showed that the presence of different cations and anions did not significantly interfere in the extraction of SMX. The outcomes of real-time samples analysis showed that the extraction of SMX for both methods was in the range of 92.44%–99.12%. The results showed the developed methods are simple, sensitive, and suitable for SMX preconcentration in environmental water and biological samples.
