Abstract:Many diseases involve either the formation of new blood vessels (e.g., tumor angiogenesis) or the damage of existing ones (e.g., diabetic retinopathy) at the microcirculation level. Optical-resolution photoacoustic microscopy (OR-PAM), capable of imaging microvessels in 3D in vivo down to individual capillaries using endogenous contrast, has the potential to reveal microvascular information critical to the diagnosis and staging of microcirculation-related diseases. In this study, we have developed a dedicated microvascular quantification (MQ) algorithm for OR-PAM to automatically quantify multiple microvascular morphological parameters in parallel, including the vessel diameter distribution, the microvessel density, the vascular tortuosity, and the fractal dimension. The algorithm has been tested on in vivo OR-PAM images of a healthy mouse, demonstrating high accuracy for microvascular segmentation and quantification. The developed MQ algorithm for OR-PAM may greatly facilitate quantitative imaging of tumor angiogenesis and many other microcirculation related diseases in vivo.

Abstract:Optical-resolution photoacoustic microscopy (OR-PAM) has been shown to be an excellent tool for high-resolution imaging of microvasculature, and quantitative analysis of the microvasculature can provide valuable information for the early diagnosis and treatment of various vascular-related diseases. In order to address the characteristics of weak signals, discontinuity and small diameters in photoacoustic micro-vascular images, we propose a method adaptive to the microvascular segmentation in photoacoustic images, including Hessian matrix enhancement and the morphological connection operators. The accuracy of our vascular segmentation method is quantitatively evaluated by the multiple criteria. To obtain more precise and continuous microvascular skeletons, an improved skeleton extraction framework based on the multistencil fast marching (MSFM) method is developed. We carried out in vivo OR-PAM microvascular imaging in mouse ears and subcutaneous hepatoma tumor model to verify the correctness and superiority of our proposed method. Compared with the previous methods, our proposed method can extract the microvascular network more completely, continuously and accurately, and provide an effective solution for the quantitative analysis of photoacoustic microvascular images with many small branches.
Synthetic multi-focus optical-resolution photoacoustic microscope for large volumetric imaging

Here, we developed a synthetic multi-focus optical-resolution photoacoustic microscope using multi-scale weighted gradient-based fusion. Based on the saliency of the image structure, a gradient-based multi-focus image fusion method is used, and a multi-scale method is used to determine the gradient weights. We pay special attention to a dual-scale scheme, which effectively solves the fusion problem caused by anisotropic blur and registration error. First, the structure-based large-scale focus measurement method is used to reduce the effect of anisotropic blur and registration error on the detection of the focus area, and then the gradient weights near the edge wave are used by applying the small-scale focus measure. Simulation was performed to test the performance of our method, different focused images were used to verify the feasibility of the method. Performance of our method was analyzed by calculating Entropy, Mean Square Error (MSE) and Edge strength. The result of simulation shown that this method can extend the depth of field of PAM two times without the sacrifice of lateral resolution. And the in vivo imaging of the zebra fish further demonstrates the feasibility of our method.

Abstract:Optical-resolution photoacoustic microscopy (OR-PAM) is used for in vivo imaging of a variety of albino and pigmented eyes taking advantages of requiring no exogenous dye, performing high-resolution imaging, and achieving morphologic and functional imaging at the same time. However, to accurately diagnose the ophthalmic disease in the OR-PAM images, vascular enhancement algorithms are necessary for extracting vessels and quantifying them correctly. Vascular enhancement algorithms developed for other imaging technologies, are not suitable to be used for OR-PAM, because of the underlying differences in the physics of the formation of images. In this study, a new vascular enhancement algorithm called photoacoustic imaging vasculature enhancement filter (PAIVEF) is proposed, which not only enhances vasculature including micro-vessels signals, suppresses noise signals effectively, but also achieves highly sensitive and accurate enhancement of the vasculature within a large depth range in and out of the system's depth of focus (DOF). Using the PAIVEF, the morphologic and functional 3D images of the whole rat's ocular anterior vasculature segment was displayed simultaneously for a depth range of ∼0.6 mm, which was ∼7 times of the system's DOF. This study paves the way for the application of OR-PAM technology in ophthalmic disease research.

Abstract:As an important branch of photoacoustic microscopy, optical-resolution photoacoustic microscopy suffers from limited depth of field due to the strongly focused laser beam. In this work, a 3D information fusion algorithm based on 3D stationary wavelet transform and joint weighted evaluation optimization is proposed to fuse multi-focus photoacoustic data to achieve large-volumetric and high-resolution 3D imaging. First, a three-dimensional stationary wavelet transform was performed on the multi-focus data to obtain eight wavelet coefficients. Differential evolution algorithm based on joint weighted evaluation was then employed to optimize the block size of division for each wavelet coefficient. Corresponding sub-coefficients of multi-focus 3D data were fused with the proposed fusion rule utilizing standard deviation for focus detection. Finally, photoacoustic microscopy with large depth of field can be achieved by applying the inverse stationary wavelet transform on the 8 fused sub-coefficients. The fusion result of multi-focus vertically tilted fiber shows that the depth of field of optical-resolution photoacoustic microscopy is doubled without sacrificing lateral resolution via the proposed method. Furthermore, the effectiveness of the proposed method was verified through the fusion results of multi-focus vessel data. Our work provides a feasible solution for achieving large-volumetric, high-resolution photoacoustic microscopy for further data analysis, processing and applications.

Abstract:Optical‐resolution photoacoustic microscopy (OR‐PAM) has been shown to be an excellent imaging modality for monitoring and study of tumor microvasculature. However, previous studies focused mainly on the normal tissues and did not quantify the tumor microvasculature. In this study, we present an in vivo OR‐PAM imaging of the melanomas and hepatoma implanted in the mouse ear. We quantify the vessel growth by extracting the skeletons of both dense and thin branches of the tumor microvasculature obtained by Hessian matrix enhancement followed by improved two‐step multistencils fast marching method. Compared with the previous methods of using OR‐PAM for normal tissues, our method was more effective in extracting the binary vascular network in the tumor images and in obtaining the complete and continuous microvascular skeleton maps. Our demonstration of using OR‐PAM in improving microvasculature of tumors and quantification of tumor growth would push deep this technology for the early diagnosis and treatment of cancers.

Abstract:SignificancePhotoacoustic imaging is an emerging imaging modality that combines the high contrast of optical imaging and the high penetration of acoustic imaging. However, the strong focusing of the laser beam in optical-resolution photoacoustic microscopy (OR-PAM) leads to a limited depth-of-field (DoF).AimHere, a volumetric photoacoustic information fusion method was proposed to achieve large volumetric photoacoustic imaging at low cost.ApproachFirst, the initial decision map was built through the focus detection based on the proposed three-dimensional Laplacian operator. Majority filter-based consistency verification and Gaussian filter-based map smoothing were then utilized to generate the final decision map for the construction of photoacoustic imaging with extended DoF.ResultsThe performance of the proposed method was tested to show that our method can expand the limited DoF by a factor of 1.7 without the sacrifice of lateral resolution. Four sets of multi-focus vessel data at different noise levels were fused to verify the effectiveness and robustness of the proposed method.ConclusionsThe proposed method can efficiently extend the DoF of OR-PAM under different noise levels.

Abstract:Optical-resolution photoacoustic microscopy (OR-PAM) technique is a noninvasive imaging technique that can be used to obtain high-resolution images. This technique does not require the use of exogenous contrast agents. The technique can be effectively used to realize endogenous and exogenous imaging of microvasculature. However, the depth of focus (DOF) realized using OR-PAM is low and inadequate to cover the entire 3D data for microvasculature imaging. A varying extent of signal-to-noise ratio (SNR) is recorded when varying depths of the 3D sample are studied. This significantly hindered the processes of image recognition, segmentation, and analysis. Herein, a deep learning-based OR-PAM technique to image and analyze 3D datasets is proposed. Endogenous and exogenous multi-organ imaging data are sequentially imaged and segmented, and excellent generalization ability is observed. Wide-field and ultradense exogenous 3D imaging of mouse brain vasculature located at different depths is obtained and segmented by OR-PAM for the first time. The results reveal that the proposed method can be used for effectively imaging the entire microvasculature. The method can be potentially used for imaging the microcirculation system in clinics.

Abstract:By considering the line pattern of acoustic-resolution photoacoustic microscopy (AR-PAM) vessel images, we develop modified algorithms for synthetic aperture focusing technique (SAFT) and deconvolution based on a directional approach to enhance images. The modified algorithms consist of Fourier accumulation SAFT (FA-SAFT) and directional model-based (D-MB) deconvolution. To evaluate the performance of our algorithms, we conduct a series of imaging experiments and apply our algorithms, and existing SAFT and deconvolution algorithms are also applied for side-by-side comparison. By imaging tungsten wire phantom, our algorithms enable full width at half maximum of 26 - 31 µm over depth of focus of 1.8 mm and minimum resolvable distance of 46 - 49 µm, besting existing SAFT and deconvolution algorithms. Imaging of leaf skeleton phantom and in vivo imaging of mouse blood vessels also prove that our algorithm is capable of providing high-resolution, high-signal-to-noise ratio, and good-fidelity results for complex structures and for in vivo applications, especially for the images with the line pattern. The proposed directional approach can not only be used in AR-PAM but also in other imaging modalities to deal with the line pattern, such as FA-SAFT for ultrasound imaging and D-MB deconvolution for optical coherence tomography angiography.

Abstract:Previously, we described a large Dutch family with hereditary vascular retinopathy (HVR), Raynaud's phenomenon and migraine. A locus for HVR was mapped on chromosome 3p21.1-p21.3, but the gene has not yet been identified. The fact that all three disorders share a vascular aetiology prompted us to study whether the HVR haplotype also contributed to Raynaud's phenomenon and migraine in this family. Whereas the parent-child transmission disequilibrium test (TDT) did not reach significance, the sibling TDT revealed that the HVR haplotype harbours a susceptibility factor for Raynaud's phenomenon and migraine. Identification of the HVR gene will improve the understanding of the pathophysiology of HVR, Raynaud's phenomenon and migraine.

Abstract:Near-infrared fluorescence imaging in the second window has emerged as a valuable tool for the non-invasive and real-time assessment of vascular information in skin flaps. Enhancing flap images to provide more accurate flap vascularization information is critical for predicting flap viability. To address the limitations of existing methods in enhancing vessel images, we propose a novel and adaptive technique for enhancing flap microvessel images. Multiple strategies can be employed to effectively enhance the visualization of small-scale vessels. Firstly, the proposed method leverages the multiscale rolling guided filter to acquire the base layer and detail layers at different scales. Furthermore, correlation coefficients are utilized to weigh and fuse the detail layers effectively. To suppress noise amplification while enhancing vascular structures, an improved adaptive gamma correction method based on local visual saliency is introduced. Meanwhile, the bilateral gamma correction is used to enhance the base layer. Finally, the enhanced base layer and detail layer are fused using the weighted fusion strategy. We conducted experiments on skin flap vessel images, retinal fundus images, finger vein images, and low-light images. Our method achieved excellent results in metrics such as NIQE, AMBE, and WPSNR, demonstrating significant advantages in preserving the structural integrity and brightness consistency of the images. The obtained results validate the potential of this method in enhancing vascular images, indicating promising prospects in the field of medicine.
Multi-Focus Image Fusion for Full-Field Optical Angiography

Abstract:Full-field optical angiography (FFOA) has considerable potential for clinical applications in the prevention and diagnosis of various diseases. However, owing to the limited depth of focus attainable using optical lenses, only information about blood flow in the plane within the depth of field can be acquired using existing FFOA imaging techniques, resulting in partially unclear images. To produce fully focused FFOA images, an FFOA image fusion method based on the nonsubsampled contourlet transform and contrast spatial frequency is proposed. Firstly, an imaging system is constructed, and the FFOA images are acquired by intensity-fluctuation modulation effect. Secondly, we decompose the source images into low-pass and bandpass images by performing nonsubsampled contourlet transform. A sparse representation-based rule is introduced to fuse the lowpass images to effectively retain the useful energy information. Meanwhile, a contrast spatial frequency rule is proposed to fuse bandpass images, which considers the neighborhood correlation and gradient relationships of pixels. Finally, the fully focused image is produced by reconstruction. The proposed method significantly expands the range of focus of optical angiography and can be effectively extended to public multi-focused datasets. Experimental results confirm that the proposed method outperformed some state-of-the-art methods in both qualitative and quantitative evaluations.
Viewing Individual Cells and Ambient Microvasculature Using Two Molecular Contrasts

Abstract:To view the individual cells and ambient microvasculature simultaneously will be helpful to study tumor angiogenesis and microenvironments. To achieve this, two molecular contrast mechanisms were exploited simultaneously by integrating two imaging modalities, confocal fluorescence microscopy (CFM) and photoacoustic microscopy (PAM). These share the same scanning optical path and laser source. The induced photoacoustic (PA) signal was detected by a highly sensitive needle hydrophone; while the back-traveling fluorescent photons emitted from the same sample were collected by an avalanche photodetector. Experiments on ex vivo rat bladders were conducted. The CFM image depicted the shape and size of the individual cells successfully. Besides large polygonal umbrella cells, some intracellular components can also be discerned. With the CFM image presenting morphologic cellular information in the bladder wall, the PAM image provides the complementary information, based on the endogenous optical absorption contrast, of the microvascular distribution inside the bladder wall, from large vessels to capillaries. Such multimodal imaging provides the opportunity to realize both histological assay and characterization of microvasculature using one imaging setup. This approach offers the possibility of comprehensive diagnosis of cancer in vivo.

Abstract:Retinal vascular disease has always been a common concern in the medical field. By performing blood vessel enhancement, fundus images help clinicians diagnosing with workload reduction and time-saving. Due to size varies of blood vessels and inhomogeneous intensities of fundus images, retinal vessel enhancement is still an open problem. In this paper, we propose a Multi-scale Space-Intensity domain Fusion Adaptive filtering model (MSIFA) to enhance retinal vessels from fundus images. Retinal vessels are roughly estimated by a proposed vessel indicating function. The indicating function is then used to define a local intensity adaptive Bilateral filtering model with spatial correlations being also considered. Finally, an improved response function is proposed to describe degrees of any a given pixel belonging to vascular regions based on eigenvalues of the Hessian matrix from images being filtered by the Bilateral filtering model. The proposed model has been evaluated with the Frangi filter and 14 representative models on STARE and DRIVE image repositories. Experimental results show advantages of the proposed models on both images repositories in terms of 6 evaluation metrics.

Performance of Medical Image Fusion in High-level Analysis Tasks: A Mutual Enhancement Framework for Unaligned PAT and MRI Image Fusion

Abstract:Photoacoustic tomography (PAT) offers optical contrast, whereas magnetic resonance imaging (MRI) excels in imaging soft tissue and organ anatomy. The fusion of PAT with MRI holds promising application prospects due to their complementary advantages. Existing image fusion have made considerable progress in pre-registered images, yet spatial deformations are difficult to avoid in medical imaging scenarios. More importantly, current algorithms focus on visual quality and statistical metrics, thus overlooking the requirements of high-level tasks. To address these challenges, we proposes a unsupervised fusion model, termed PAMRFuse+, which integrates image generation and registration. Specifically, a cross-modal style transfer network is introduced to simplify cross-modal registration to single-modal registration. Subsequently, a multi-level registration network is employed to predict displacement vector fields. Furthermore, a dual-branch feature decomposition fusion network is proposed to address the challenges of cross-modal feature modeling and decomposition by integrating modality-specific and modality-shared features. PAMRFuse+ achieves satisfactory results in registering and fusing unaligned PAT-MRI datasets. Moreover, for the first time, we evaluate the performance of medical image fusion with contour segmentation and multi-organ instance segmentation. Extensive experimental demonstrations reveal the advantages of PAMRFuse+ in improving the performance of medical image analysis tasks.

Abstract:Due to the limitations on the depth of field of high-resolution fluorescence microscope, it is difficult to obtain an image with all objects in focus. The existing image fusion methods suffer from blocking effects or out-of-focus fluorescence. The proposed multi-focus image fusion method based on local maximum luminosity, intensity variance and the information filling method can reconstruct the all-in-focus image. Moreover, the depth of tissue's surface can be estimated to reconstruct the 3D surface model.
HSD-PAM: High Speed Super Resolution Deep Penetration Photoacoustic Microscopy Imaging Boosted by Dual Branch Fusion Network

Abstract:Photoacoustic microscopy (PAM) is a novel implementation of photoacoustic imaging (PAI) for visualizing the 3D bio-structure, which is realized by raster scanning of the tissue. However, as three involved critical imaging parameters, imaging speed, lateral resolution, and penetration depth have mutual effect to one the other. The improvement of one parameter results in the degradation of other two parameters, which constrains the overall performance of the PAM system. Here, we propose to break these limitations by hardware and software co-design. Starting with low lateral resolution, low sampling rate AR-PAM imaging which possesses the deep penetration capability, we aim to enhance the lateral resolution and up sampling the images, so that high speed, super resolution, and deep penetration for the PAM system (HSD-PAM) can be achieved. Data-driven based algorithm is a promising approach to solve this issue, thereby a dedicated novel dual branch fusion network is proposed, which includes a high resolution branch and a high speed branch. Since the availability of switchable AR-OR-PAM imaging system, the corresponding low resolution, undersample AR-PAM and high resolution, full sampled OR-PAM image pairs are utilized for training the network. Extensive simulation and in vivo experiments have been conducted to validate the trained model, enhancement results have proved the proposed algorithm achieved the best perceptual and quantitative image quality. As a result, the imaging speed is increased 16 times and the imaging lateral resolution is improved 5 times, while the deep penetration merit of AR-PAM modality is still reserved.
