Abstract:Objective To explore the effect and the most sensitive period of low-iron intervention in pregnant rats on serum lipids and serum glucose of its offspring in adulthood.Methods Six week-age Wistar female rats were divided into two groups randomly.The rats of experimental group were fed with low-iron diet and the rats in control group were fed with iron-sufficient diet.The rats were mated with healthy male rats after four weeks and the iron intervention were carried out untill the end of lactation period.Iron-sufficient diet were then given to all rats for ten weeks(iron balancing period).After that control group and experimental group were divided into two groups and fed with low-or sufficient-iron diet,respectively,and all the rats were killed to detect serum triglyceride(TG),cholesterol(TC),blood glucose,free fatty acids(FFA),high density lipoprotein(HDL) and leptin after sixteen weeks.Results Compared with the control group,the levels of serum cholesterol,triglyceride,glucose,weight of heart and liver of the offsprings with their mothers fed with low-level iron diet during pregnancy were elevated significantly.Besides,hydropic degeneration in hepatocytes,cytoplasm puffing and disappearance of hepatic sinusoid were observed in the rats.Conclusion Maternal iron deficiency during pregnancy will result in the increased levels of serum triglyceride,cholesteroland,blood glucose and pathologic changes of liver tissue and hepatocytes in its thirty-week-old offsprings.

Abstract:Injuries to the developing brain due to hypoxia–ischemia (HI) are common causes of neurological disabilities in preterm babies. HI, with oxygen deprivation to the brain or reduced cerebral blood perfusion due to birth asphyxia, often leads to severe brain damage and sequelae. Injury mechanisms include glutamate excitotoxicity, oxidative stress, blood–brain barrier dysfunction, and exacerbated inflammation. Nutritional intervention is emerging as a therapeutic alternative to prevent and rescue brain from HI injury. Lactoferrin (Lf) is an iron-binding protein present in saliva, tears, and breast milk, which has been shown to have antioxidant, anti-inflammatory and anti-apoptotic properties when administered to mothers as a dietary supplement during pregnancy and/or lactation in preclinical studies of developmental brain injuries. However, despite Lf’s promising neuroprotective effects, there is no established dose. Here, we tested three different doses of dietary maternal Lf supplementation using the postnatal day 3 HI model and evaluated the acute neurochemical damage profile using 1H Magnetic Resonance Spectroscopy (MRS) and long-term microstructure alterations using advanced diffusion imaging (DTI/NODDI) allied to protein expression and histological analysis. Pregnant Wistar rats were fed either control diet or bovine Lf supplemented chow at 0.1, 1, or 10 g/kg/body weight concentration from the last day of pregnancy (embryonic day 21–E21) to weaning. At postnatal day 3 (P3), pups from both sexes had their right common carotid artery permanently occluded and were exposed to 6% oxygen for 30 min. Sham rats had the incision but neither surgery nor hypoxia episode. At P4, MRS was performed on a 9.4 T scanner to obtain the neurochemical profile in the cortex. At P4 and P25, histological analysis and protein expression were assessed in the cortex and hippocampus. Brain volumes and ex vivo microstructural analysis using DTI/NODDI parameters were performed at P25. Acute metabolic disturbance induced in cortical tissue by HIP3 was reversed with all three doses of Lf. However, data obtained from MRS show that Lf neuroprotective effects were modulated by the dose. Through western blotting analysis, we observed that HI pups supplemented with Lf at 0.1 and 1 g/kg were able to counteract glutamatergic excitotoxicity and prevent metabolic failure. When 10 g/kg was administered, we observed reduced brain volumes, increased astrogliosis, and hypomyelination, pointing to detrimental effects of high Lf dose. In conclusion, Lf supplementation attenuates, in a dose-dependent manner, the acute and long-term cerebral injury caused by HI. Lf reached its optimal effects at a dose of 1 g/kg, which pinpoints the need to better understand effects of Lf, the pathways involved and possible harmful effects. These new data reinforce our knowledge regarding neuroprotection in developmental brain injury using Lf through lactation and provide new insights into lactoferrin’s neuroprotection capacities and limitation for immature brains.

Effects of dietary supplementation of bovine lactoferrin on growth performance, immune function and intestinal health in weaning piglets

Abstract:Weaning is a crucial period in the pig's life cycle, which is frequently followed by gastrointestinal (GI) infections, diarrhea and even death. This study focused on the impact of bovine lactoferrin (bLF) supplementation on the intestinal health of weaning piglets. Weaning piglets (Duroc × Landrace × Yorkshire, 23 days) were randomly allocated into four groups, which included negative control group (CON): basic diet; positive control group (ANT): basic diet + 20 mg/kg flavomycin + 100 mg/kg aureomycin; treatment group bLF-A: basic diet + 1 g/kg bLF; treatment group bLF-B: basic diet + 3 g/kg bLF. The result showed that dietary supplementation of bLF can improve growth performance and reduce diarrhea, which exhibits dose-dependency (P < 0.05). Compared with CON group, supplementation with bLF significantly improved immunity, and increased villus height and ratio of villus height/crypt depth at the small intestinal mucosa (P < 0.05). The mRNA expression of claudin-1, occludin and ZO-1 was greatly increased in the ileum of bLF group on days 7 and 14 (P < 0.05). Furthermore, the supplementation of bLF increased the abundance of Lactobacillus and Bifidobacterium and decreased the abundance of Escherichia coli in the cecum on day 7 (P < 0.05). The dietary supplementation of bLF enhanced the growth performance, reduced diarrhea rate in weaning piglets by improving intestinal immunity, morphology and barrier function, balancing intestinal microbiota. And bLF can be a promising feed additive in relieving stress situation of weaning piglets.

Abstract:The experiment investigated the effect of dietary bovine lactoferrin (bLF) on growth performance and antioxidant systems of piglets. Duroc–Landrace–Yorkshire crossbred female piglets (n=120, 35 days of age, live-weight 9.70±0.71kg) were fed a diet containing 0, 1250 or 2500mg/kg bLF for 30 days. After completion of the feeding experiment, eight animals from each treatment were randomly selected to determine malondialdehyde (MDA) and total antioxidant capacity (TAOC), copper–zinc–superoxide dismutase (CuZnSOD), glutathione peroxidase (GPx), catalase (CAT) activities in serum and longissimus muscle, and CuZnSOD, GPx and CAT mRNA levels in longissimus muscle. Results showed that supplementation with bLF improved average daily gain (ADG), average daily feed intake (ADFI) and reduced diarrhea ratios of piglets with the linear and quadratic polynomial contrasts being significant (P<0.01). The highest weight gain and the best feed conversion occurred at 2500mg/kg bLF. Serum and longissimus muscle CuZnSOD and GPx, and serum TAOC linearly and quadraticly increased (P<0.01), serum CAT and longissimus muscle CAT and TAOC linearly increased (P<0.01), whereas MDA concentrations in serum and longissimus muscle linearly decreased (P<0.01) as dietary bLF supplementation increased. Increasing dietary bLF levels improved (P<0.01) mRNA expression of CuZnSOD and CAT in longissimus muscle of piglets. The results indicated that exogenous antioxidant bLF was effective in improving growth performance and enhancing antioxidant enzymes activity and mRNA levels of piglets.

Abstract:Introduction/Objectives. To investigate whether oral administration of lactoferrin attenuates intestinal injury induced by perinatal hypoxia and hypothermia. Methods. Newborn Wistar rat pups were exposed to perinatal asphyxia, followed by global hypothermia. The pups were distributed into two groups: lactoferrin group (LG) ? pups that received lactoferrin orally for seven days (20 mg/day), control group (CG) ? pups that received normal saline orally during first seven days of life. After seven days macroscopic examination of the bowels and pathohistological analyses of tissue samples have been performed. Results. The incidence of macroscopic injuries was significantly lower in LG group compared to CG. The incidence of pathological findings, as well as the values of injury scores used to assess the intensity and extent of intestinal injury at microscopic level were significantly lower in LG group. Conclusions. Lactoferrin attenuates perinatal hypoxia/hypothermia-induced intestinal injury in newborn rats.

Effect of dietary lactoferrin on the immune functions and serum iron level of weanling piglets.

Abstract:A total of 90 weanling female pigs (Duroc x Landrace x Yorkshire) were used in a 30-d growth experiment to investigate the effect of lactoferrin (LF) on growth performance, immune function, and serum iron concentrations. The pigs were allocated on the basis of BW and litter to 3 dietary treatments in a randomized complete block design. The dietary treatments were: control group (basal diet), antibiotics group (basal diet + 20 mg/kg of flavomycin + 110 mg/kg of aureomycin), and LF group (basal diet + 1.0 g/kg of LF). There were 3 replicate pens per treatment, and pigs were grouped with 10 pigs per pen. Six pigs, randomly selected from each treatment (2 pigs/pen), were slaughtered for serum and spleen samples on d 15 and 30. Supplementation with LF improved the phytohemagglutinin (PHA)-stimulated peripheral lymphocyte proliferation by 36% (P < 0.01), increased concanavalin A (ConA)- and PHA-induced spleen lymphocyte proliferation by 332% (P < 0.01) and 258% (P < 0.01), enhanced serum IgG by 20% (P < 0.05), IgA by 13% (P < 0.05), IgM by 15% (P < 0.05), complement 4 (C-4) by 29% (P < 0.05), IL-2 by 12% (P < 0.01), and serum iron values by 22% (P < 0.05) on d 15 compared with the control. Lactoferrin supplementation increased PHA-stimulated lymphocyte proliferation (P < 0.01), serum IgG by 16% (P < 0.05), IgA by 17% (P < 0.05), C-4 by 11% (P < 0.05), IL-2 by 14% (P < 0.05), and serum iron values by 23% (P < 0.01), and decreased the diarrhea ratio (P < 0.05) relative to the control on d 30. Compared with the controls, supplementation with antibiotic increased ConA- and PHA-induced spleen lymphocyte proliferation (P < 0.05) on d 15, decreased the diarrhea ratio (P < 0.05), and increased the PHA-induced spleen lymphocyte proliferation (P < 0.05) and serum iron values (P < 0.01) on d 30. These results support the possible use LF as an immunostimulant to improve immune functions and strengthen host defenses and would seem to be a good method for defending weanling piglets from infections and weanling stress.
Effect of Dynamic Regulation of Viscosity and Nutritional Ingredients of Milk-Substitutes on Artificial Rearing of Germfree Rats

Abstract:A program was established for artificial rearing(AR) of germfree rat pups according to the law of rat pups' suck.Six kinds of milk-substitutes(LWF) for pups at different ages were prepared.Their major nutrient compositions and viscosity were determined.Under axenic conditions,30 cesarean-derived pups were reared artificially by suck feeding until weaning,while 30 pups were reared by foster mothers as controls.The body weight gain and the early neonatal body development were observed every day.Survival rates were calculated.Fertility was tested when 8 pairs of AR rats were over 100 days old.Except for LWF-6,other LWFs were comparable to rat's milk in the concentrations of protein,fat,carbohydrate,the major minerals,gross energy and pH.From LWF-1 to LWF-5, the contents of protein,fat,carbohydrate,energy and viscosity increased gradually.Although the AR rats grew less well than the mother-reared(MR) pups(at age of 2 days,P0.05;at age of 3 days,P0.01),development of these pups was normal.Survival rate of AR rats was 73.3%,that of MR was 100% when weaning.All the mated rats had normal fertility.It is indicated that dynamic regulation of viscosity and concentrations of the macronutrients of LWF may be suit for artificial rearing of germfree rats with high survival rate.
Highly Selective and Sensitive Luminescence Turn-On-Based Sensing of Al3+ Ions in Aqueous Medium Using a MOF with Free Functional Sites.

Abstract:A new metal–organic framework [Co(OBA)(DATZ)0.5(H2O)] {OBA = 4,4′-oxybis(benzoic acid) and DATZ = 3,5-diamino-1,2,4-triazole}, 1, was synthesized by hydrothermal reaction. Single-crystal X-ray data of 1 confirmed two-dimensional rhombus grid network topology with a free nitrogen site of triazole ring and two amine groups of each DATZ. Photoluminescence study of 1 in aqueous medium shows blue emission at 407 nm upon excitation at 283 nm. This emissive property was used for the sensing of Al3+ ions in aqueous medium through very high luminescence turn-on (6.3-fold) along with the blue shifting (∼24 nm) of the emission peak. However, luminescence studies in the presence of other common metal ions such as Mg2+, Zn2+, Ni2+, Co2+, Mn2+, K+, Na+, Ca2+, Cd2+, Hg2+, Cu2+, Fe2+, Fe3+, and Cr3+ in aqueous medium shows luminescence quenching in varying extent. Interestingly, the luminescence turn-on-based selectivity of Al3+ ions in aqueous medium was achieved even in the presence of the highest quenchable metal ion, Fe3+. Furthermore, very high sensitivity was observed in the case of Al3+ ions with a limit of detection of Al3+ of 57.5 ppb, which is significantly lower than the higher limit of U.S. Environmental Protection Agency recommendation of Al3+ ion for drinking water (200 ppb).

Abstract:<b>Introduction:</b> Intrauterine Growth Restriction (IUGR) refers to an impaired development of the fetus and hence results in adverse neurodevelopmental and psychiatric consequences later in life. Lactoferrin (Lf) is a glycoprotein present in milk that has already shown neuroprotective effects through its anti-inflammatory and antioxidant properties on impaired developing brains. The aim of this study was to characterize a rat model of IUGR and assess the neuroprotective effect of a nutritional supplementation with bovine Lf during pregnancy and lactation on this model. <b>Methods:</b> A model of 50% gestational caloric restriction (CR) was used. Three groups were designed, and pregnant rats had either <i>ad libitum</i> access to food (control group, CTL) or 50% of the controls' intake (restricted group, IUGR). The diet was isocaloric and supplemented with bovine Lf for the caloric restricted dams (restricted-Lf, IUGR_Lf). At postnatal day 7 and 21, advanced <i>ex-vivo</i> diffusion MRI techniques at 9.4T were used to investigate brain cortical and white matter microstructure. Further, genes and proteins involved in structure (synaptophysin, MBP), microglia (Iba-1), metabolism (MCT2, βCaMKII) and apoptosis (Bcl-2) were analyzed in the cortex and striatum. In the cortex, the number of parvalbumin immunoreactive interneurons and their perineuronal nets were quantified. Behavioral tests were performed at P31. <b>Results:</b> Effects of the CR were significant in the cortex and striatum with reduction of synaptophysin (marker of synaptogenesis) at P7 and MBP (marker of myelin) at P21 in the cortex. Indeed, MCT2 (energy metabolism), Bcl-2 (anti-apoptotic protein) and βCaMKII (synapse activity) expressions were reduced in IUGR groups at P7. In the striatum NG2 (marker of oligodendrocyte precursor cells) and Bcl-2 at P7 as well as βCaMKII at P21 were decreased following IUGR and restored by Lf. Cortical microstructure was impaired following CR with partial effect of Lf. Lf prevented oxidative stress induced parvalbumin interneurons impairments whereas striatum and external capsule showed alterations in microstructure depicted by diffusion MRI, which were also partially reversed by Lf. <b>Discussion and Conclusion:</b> The model of 50% caloric restriction induced mild impairment partially reversed by nutritional intervention using Lf during pregnancy and lactation.

Abstract:Iron-fortified formulas and iron drops (both usually ferrous sulfate, FS) prevent early life iron deficiency, but may delay growth and adversely affect neurodevelopment by providing excess iron. We used a rat pup model to investigate iron status, growth, and development outcomes following daily iron supplementation (10 mg iron/kg body weight, representative of iron-fortified formula levels) with FS or an alternative, bioavailable form of iron, ferrous bis-glycinate chelate (FC). On postnatal day (PD) 2, sex-matched rat litters (n = 3 litters, 10 pups each) were randomly assigned to receive FS, FC, or vehicle control until PD 14. On PD 15, we evaluated systemic iron regulation and CNS mineral interactions and we interrogated iron loading outcomes in the hippocampus, in search of mechanisms by which iron may influence neurodevelopment. Body iron stores were elevated substantially in iron-supplemented pups. All pups gained weight normally, but brain size on PD 15 was dependent on iron source. This may have been associated with reduced hippocampal oxidative stress but was not associated with CNS mineral interactions, iron regulation, or myelination, as these were unchanged with iron supplementation. Additional studies are warranted to investigate iron form effects on neurodevelopment so that iron recommendations can be optimized for all infants.

Abstract:Lactoferrin, a glycoprotein derived from breastmilk, is recognized for its health benefits in infants and children; however, its protective effects when administered during gestation and lactation against offspring hypertension remain unclear. This study aimed to investigate whether maternal lactoferrin supplementation could prevent hypertension in offspring born to mothers with chronic kidney disease (CKD), with a focus on nitric oxide (NO), renin-angiotensin system (RAS) regulation, and alterations in gut microbiota and short-chain fatty acids (SCFAs). Prior to pregnancy, female rats were subjected to a 0.5% adenine diet for 3 weeks to induce CKD. During pregnancy and lactation, pregnant rats received one of four diets: normal chow, 0.5% adenine diet, 10% lactoferrin diet, or adenine diet supplemented with lactoferrin. Male offspring were euthanized at 12 weeks of age (n = 8 per group). Supplementation with lactoferrin during gestation and lactation prevented hypertension in adult offspring induced by a maternal adenine diet. The maternal adenine diet caused a decrease in the index of NO availability, which was restored by 67% with maternal LF supplementation. Additionally, LF was related to the regulation of the RAS, as evidenced by a reduced renal expression of renin and the angiotensin II type 1 receptor. Combined maternal adenine and LF diets altered beta diversity, shifted the offspring's gut microbiota, decreased propionate levels, and reduced the renal expression of SCFA receptors. The beneficial effects of lactoferrin are likely mediated through enhanced NO availability, rebalancing the RAS, and alterations in gut microbiota composition and SCFAs. Our findings suggest that maternal lactoferrin supplementation improves hypertension in offspring in a model of adenine-induced CKD, bringing us closer to potentially translating lactoferrin supplementation clinically for children born to mothers with CKD.

The pigs were allocated on the basis of weight and litter to three dietary treatments in a randomized complete block design. There were three replicate pens per treatment and pigs were grouped with 10 pigs per pen. The dietary treatments were: (1) basal diet, (2) basal diet+20mg/kg flavomycin+110mg/kg aureomycin, and (3) basal diet+1.0g/kg lactoferrin. Six randomly selected pigs from each treatment (two piglets per pen) were slaughtered for the intestinal microflora and intestinal morphology at the end of the experiment. Supplementation with LF improved growth performance, significantly increased the ADG by 34.0% (P<0.01) and ADFI by 17.0% (P<0.05), decreased the F/G by 12.8% (P<0.05) and the diarrhea by 66.2% (P<0.05). Reducing the total viable counts of Escherichia coli and Salmonella (P<0.05), enriched the colonic Lactobacillus and Bifidobacterium (P<0.05) in the small intestine. The villus height was increased (P<0.01) and crypt depth decreased (P<0.05) at the small intestinal mucosa as compared with the control. Supplemental antibiotic also increased ADG and ADFI (P<0.05), decreased diarrhea (P<0.05) and reduced the total viable counts of some pathogenic bacteria (P<0.05), but not significantly affected beneficial bacteria and intestinal morphology. The use of lactoferrin as an additive to improve growth performance, prevent diarrhea, and affect on the intestinal microflora and morphology of weanling pigs would be good a method of defending weaned pigs from infections and weanling stress.
Immunoregulation of bovine lactoferrin together with osteopontin promotes immune system development and maturation

Abstract:The immune system of infants is partly weak and immature, and supplementation of infant formula can be of vital importance to boost the development of the immune system. Lactoferrin (LF) and osteopontin (OPN) are essential proteins in human milk with immunoregulation function. An increasing number of studies indicate that proteins have interactions with each other in milk, and our previous study found that a ratio of LF : OPN at 1 : 5 (w/w, denoted as LOP) had a synergistic effect on intestinal barrier protection. It remains unknown whether LOP can also exert a stronger effect on immunoregulation. Hence, we used an in vitro model of LPS-induced macrophage inflammation and in vivo models of LPS-induced intestinal inflammation and early life development. We showed that LOP increased the secretion of the granulocyte-macrophage colony-stimulating factor (132%), stem cell factor (167%) and interleukin-3 (176%) in bone marrow cells, as well as thymosin (155%) and interleukin-10 (161%) in the thymus, more than LF or OPN alone during development, and inhibited changes in immune cells and cytokines during the LPS challenge. In addition, analysis of the components of digested proteins in vitro revealed that differentially expressed peptides may provide immunoregulation. Lastly, LOP increased the abundance of Rikenellaceae, Muribaculum, Faecalibaculum, and Elisenbergiella in the cecum content. These results imply that LOP is a potential immunomodifier for infants and offers a new theoretical basis for infant formula innovation.
Milk Fat Globule Membrane Supplementation in Formula-fed Rat Pups Improves Reflex Development and May Alter Brain Lipid Composition

Abstract:Human milk contains nutritional, immunoprotective and developmental components that support optimal infant growth and development. The milk fat globule membrane (MFGM) is one unique component, comprised of a tri-layer of polar lipids, glycolipids, and proteins, that may be important for brain development. MFGM is not present in most infant formulas. We tested the effects of bovine MFGM supplementation on reflex development and on brain lipid and metabolite composition in rats using the “pup in a cup” model. From postnatal d5 to d18, rats received either formula supplemented with MFGM or a standard formula without MFGM; a group of mother-reared animals was used as reference/control condition. Body and brain weights did not differ between groups. MFGM supplementation reduced the gap in maturation age between mother-reared and standard formula-fed groups for the ear and eyelid twitch, negative geotaxis and cliff avoidance reflexes. Statistically significant differences in brain phospholipid and metabolite composition were found at d13 and/or d18 between mother-reared and standard formula-fed groups, including a higher phosphatidylcholine:phosphatidylethanolamine ratio, and higher phosphatidylserine, glycerol-3 phosphate, and glutamine in mother-reared compared to formula-fed pups. Adding MFGM to formula narrowed these differences. Our study demonstrates that addition of bovine MFGM to formula promotes reflex development and alters brain phospholipid and metabolite composition. Changes in brain lipid metabolism and their potential functional implications for neurodevelopment need to be further investigated in future studies.

Abstract:The experiment was conducted to evaluate effect of dietary bovine lactoferrin (bLF) on lipid peroxidation and activities as well as mRNA levels of antioxidant enzymes of piglets. Duroc×Landrace×Yorkshire crossbred female piglets (n=120, 35 days of age, liveweight 9.70±0.71 kg) were fed a diet containing 0, 1250, or 2500 mg/kg bLF for 30 days. After completion of the feeding experiment, twelve female piglets with 4 animals in each treatment were randomly selected to determine malondialdehyde (MDA) and total antioxidant capacity (TAOC) levelsczinc-superoxide dismutase (CuZnSOD), glutathione perioxidase (GPx), catalase (CAT) activities in serum and liver, and CuZnSOD, GPx and CAT mRNA levels in liver. Results showed that piglets treated with 2500 mg/kg bLF significantly increased (P<0.05) TAOC levels, the activities of GPx, CuZnSOD and CAT, and mRNA levels of CuZnSOD, GPx and CAT, and decreased (P<0.05) the contents of MDA as compared with control. Supplementation with 1250 mg/kg bLF also increased (P<0.05) the activities of CuZnSOD, GPx and CAT and mRNA levels of GPx and CAT, and decreased (P<0.05) the contents of MDA as compared with control, but the effect was not better than that of dietary addition of 2500 mg/kg bLF (P<0.05). The study indicated that addition of bLF improved the antioxidant function of piglets by up-regulation of mRNA levels and activities of certain antioxidant enzymes associated with free radicals metabolism.

Abstract:Immunonutrition, which focuses on specific nutrients in breast milk and post-weaning diets, plays a crucial role in supporting infants' immune system development. This study explored the impact of maternal supplementation with Bifidobacterium breve M-16V and a combination of short-chain galacto-oligosaccharide (scGOS) and long-chain fructo-oligosaccharide (lcFOS) from pregnancy through lactation, extending into the early childhood of the offspring. The synbiotic supplementation's effects were examined at both mucosal and systemic levels. While the supplementation did not influence their overall growth, water intake, or food consumption, a trophic effect was observed in the small intestine, enhancing its weight, length, width, and microscopic structures. A gene expression analysis indicated a reduction in FcRn and Blimp1 and an increase in Zo1 and Tlr9, suggesting enhanced maturation and barrier function. Intestinal immunoglobulin (Ig) A levels remained unaffected, while cecal IgA levels decreased. The synbiotic supplementation led to an increased abundance of total bacteria and Ig-coated bacteria in the cecum. The abundance of Bifidobacterium increased in both the intestine and cecum. Short-chain fatty acid production decreased in the intestine but increased in the cecum due to the synbiotic supplementation. Systemically, the Ig profiles remained unaffected. In conclusion, maternal synbiotic supplementation during gestation, lactation, and early life is established as a new strategy to improve the maturation and functionality of the gastrointestinal barrier. Additionally, it participates in the microbiota colonization of the gut, leading to a healthier composition.

Abstract:Lactoferrin is an immuno-modulatory nutrient in human milk that may be neuroprotective. In 36 infants born <32 weeks' gestation, we sampled human milk at 14 and 28 days of chronologic age and measured lactoferrin by electrochemiluminescence multiplex immunoassay. Using 3T quantitative brain magnetic resonance imaging scans obtained at term equivalent, we estimated total and regional brain volumes. We compared outcomes between infants exposed to low (bottom tertile, range 0.06–0.13 mg/mL) vs. high (top tertile, range 0.22–0.35 mg/mL) lactoferrin using median regression in models adjusted for gestational age, birth weight z-score, sex, and postmenstrual age.

Abstract:Nutritional insults early in life, such as during the suckling phase, are associated with phenotypic alterations and promote adverse permanent effects that impair the capacity to maintain energy balance in adulthood. This study aimed to evaluate the long-term effects of a low-protein (LP) diet during lactation on the metabolism and antioxidant systems of adult female rat offspring. Dams were fed a low-protein diet (4% protein) during the first two weeks of lactation or a normal-protein (NP) diet (20% protein) during the entire lactation period. The female offspring received a standard diet throughout the experiment. At 90 days of age, female LP offspring exhibited decreased body weight, feeding efficiency, and fat pad stores. The adult LP female offspring displayed brown adipose tissue hyperplasia without alterations in glucose homeostasis. The LP diet decreased liver triglyceride content and improved the antioxidant system compared to the NP group. The LP diet during the suckling phase promotes a lean phenotype and improves the hepatocyte antioxidant system in adult female offspring. Thus, the LP diet may play an important role in homeostasis and the prevention of metabolic damage.

Abstract:Pregnancy and lactation are window period during which the interventions on mothers bring beneficial effects to newborns. This study is aiming to investigate the effects of maternal supplementation of a human milk derived Lactiplantibacillus plantarum WLPL04-36e during pregnancy and lactation on the physiology, immunity and gut microbiota of dams and their offspring. We found that after maternal supplementation, L. plantarum WLPL04-36e could be detected in the intestine and extraintestinal tissues (liver, spleen, kidney, mammary gland, MLN and brain) of dams, as well as in the intestine of their offspring. The maternal supplementation of L. plantarum WLPL04-36e could significantly increase the body weights of dams and their offspring during the middle to late lactation period, elevate the serum levels of IL-4, IL-6, IL-10 of dams and IL-6 level of offspring, and increase the proportion of spleen CD4+ T lymphocytes of offspring. Moreover, L. plantarum WLPL04-36e supplementation could increase the alpha diversity of milk microbiota during early and middle lactation period, and elevated the Bacteroides abundance in intestine of offspring at week 2 and week 3 after birth. These results suggest that maternal supplementation of human milk derived L. plantarum can regulate the immunity and intestinal microbiota composition of offspring and play positive roles in the growth of offspring.

Abstract:<h3> Aims</h3><p>We developed a strategy for localized delivery of the LFCA (lactoferricin‐lactoferrampin), which is actively synthesized in situ by <i>Lactococcus lactis</i> (pAMJ399‐LFCA/LLMG1363), and explored the possibility of using the pAMJ399‐LFCA/LLMG1363 as an alternative additive diet to antibiotics. </p><h3> Methods and Results</h3><p>The antimicrobial activities of the LFCA derived from pAMJ399‐LFCA/LLMG1363 was tested <i>in vitro</i>. The results showed that LFCA had an inhibitory effect on <i>Staphylococcus aureus</i>,<i> Escherichia coli,</i> and <i>Salmonella enteritidis</i>. Then the pAMJ399‐LFCA/LLMG1363 was used as an additive diet to piglets. Our data demonstrated that oral administration of pAMJ399‐LFCA/LLMG1363 significantly improve the average daily gain (ADG), feed‐to‐gain ratio, intestinal mucosal integrity and decreased the serum endotoxin and D‐lactic acid levels. The mRNA expression levels of intestinal tight junction proteins (including Occludin, Claudin‐1, and ZO‐1) were significantly up‐regulated by pAMJ399‐LFCA/LLMG1363 administration. The serum immunoglobulin G (IgG) levels, intestinal secretory immunoglobulin A (sIgA) levels, IL‐2, IL‐10 and TGF‐<i>β</i> levels were significantly increased by pAMJ399‐LFCA/LLMG1363. Furthermore, our data revealed that oral administration of pAMJ399‐LFCA/LLMG1363 significantly increased the number of general <i>Lactobacillus</i>, and decreased the total viable <i>Escherichia coli</i> counts in the ileum and cecum contents. </p><h3> Conclusions</h3><p>We developed a novel pAMJ399‐LFCA/LLMG1363 secreting LFCA, which had probiotic effects on the growth performance, intestinal morphology, intestinal barrier function, and immunological indices of weaned piglets.</p><h3> Significance and Impact of the Study</h3><p>pAMJ399‐LFCA/LLMG1363, with probiotic effects on the health of weaned piglets, may be a promising feed additive for weaned piglets.</p><p>This article is protected by copyright.
