Abstract:Metal-organic frameworks are a class of new and promising anti-cancer materials. MOFs with adjustable pore size, large specific surface area, diverse structure, and excellent chemical and physical properties make them a class of effective protection carriers for anti-cancer substances. This review is centered on the core point of "anti-cancer" and discusses MOFs' research progress in anti-cancer therapies. Firstly, we provided readers with the different types of MOFs, their preparation strategies and the resulting structures. Then, different MOF composites and their biological applications were systematically presented. The specificity of biomolecules endows MOFs with broader anti-cancer applications, while MOFs can protect the drugs and biomolecules to make the best of a challenging situation. Finally, we elucidated a comprehensive overview of the biological applications of MOFs, including research hotspots as drug delivery and biomolecule carriers. Besides, we looked forward to the future developments of MOFs in the field of anti-cancer therapies. As a class of novel materials, the anti-cancer applications of MOFs are extended through the combination of different materials and different methods to improve their efficacy.

Abstract:The nanotechnology is an interdisciplinary field that has become a hot topic in cancer therapy. Metal-organic frameworks (MOFs) are porous materials and hybrid composites consisted of organic linkers and metal cations. Despite the wide application of MOFs in other fields, the potential of MOFs for purpose of cancer therapy has been revealed by the recent studies. High surface area and porosity, significant drug loading and encapsulation efficiency are among the benefits of using MOFs in drug delivery. MOFs can deliver genes/drugs with selective targeting of tumor cells that can be achieved through functionalization with ligands. The photosensitizers and photo-responsive nanostructures including carbon dots and gold nanoparticles can be loaded in/on MOFs to cause phototherapy-mediated tumor ablation. The immunogenic cell death induction and increased infiltration of cytotoxic CD 8+ and CD 4+ T cells can be accelerated by MOF platforms in providing immunotherapy of tumor cells. The stimuli-responsive MOF platforms responsive to pH, redox, enzyme and ion can accelerate release of therapeutics in tumor site. Moreover, MOF nanocomposites can be modified ligands and green polymers to improve their selectivity and biocompatibility for cancer therapy. The application of MOFs for the detection of cancer-related biomarkers can participate in the early diagnosis of patients.

Abstract:Metal-organic frameworks (MOFs) have proven to be very effective carriers for drug delivery in various biological applications. In recent years, the development of hybrid nanostructures has made significant progress, including developing an innovative MOF-loaded nanocomposite with a highly porous structure and low toxicity that can be used to fabricate core-shell nanocomposites by combining complementary materials. This review study discusses using MOF materials in cancer treatment, imaging, and antibacterial effects, focusing on oral cancer cells. For patients with oral cancer, we offer a regular program for accurately designing and producing various anticancer and antibacterial agents to achieve maximum effectiveness and the lowest side effects. Also, we want to ensure that the anticancer agent works optimally and has as few side effects as possible before it is tested in vitro and in vivo. It is also essential that new anticancer drugs for cancer treatment are tested for efficacy and safety before they go into further research.

Abstract:Effective penetration into cells, or binding to cell membranes is an essential property of an effective nanoparticle drug delivery system (DDS). Nanoparticles are generally internalized through active transport mechanisms such as apoptosis, and cargo can be released directly into the cytoplasm. A metal–organic framework (MOF) is a network structure consisting of metal clusters connected by organic linkers with high porosity; MOFs provide a desirable combination of structural features that can be adjusted with large cargo payloads, along with Cu, Co, and Zn-MOFs, which have the chemical stability required for water-soluble use. Bioactive MOFs containing copper, cobalt, and zinc were prepared by modifying previous methods as therapeutic drugs. Their structures were characterized via PXRD, single-crystal crystallographic analysis, and FT-IR. The degradability of MOFs was measured in media such as deionized water or DPBS by PXRD, SEM, and ICP-MS. Furthermore, we investigated the anticancer activity of MOFs against the cell lines SKOV3, U87MG, and LN229, as well as their biocompatibility with normal fibroblast cells. The results show that a nanoporous 3D Cu-MOF could potentially be a promising candidate for chemoprevention and chemotherapy.

Abstract:In this research, we examined the toxicity of Ag-graphene oxide (GO) nanocomposites against both the Gram-negative bacterium Escherichia coli and Glioblastoma cancer cells (U87MG). Our findings reveal that Ag-GO possesses bactericidal properties, with a minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of 160 μg/mL. The antibacterial efficacy of Ag-GO is contingent on contact time and concentration, making it a potential candidate for integration into materials designed to combat microbial infections. The bactericidal effect of Ag-GO can be attributed to the release of silver ions and the physical damage inflicted by the sharp edges of GO sheets. Furthermore, our study demonstrates that Ag-GO exhibits anticancer activity against U87MG cells, with an IC 50 value of 270 μg/mL. The mechanism underlying the anticancer activity of Ag-GO likely involves cell membrane disruption and apoptosis induction. These findings signify the promising medical and biological applications of Ag-graphene oxide nanocomposites.

Abstract:Nanomaterials, with unique physical, chemical, and biocompatible properties, have attracted significant attention as an emerging active platform in cancer diagnosis and treatment. Amongst them, metal–organic framework (MOF) nanostructures are particularly promising as a nanomedicine due to their exceptional surface functionalities, adsorption properties, and organo-inorganic hybrid characteristics. Furthermore, when bioactive substances are integrated into the structure of MOFs, these materials can be used as anti-tumor agents with superior performance compared to traditional nanomaterials. In this review, we highlight the most recent advances in MOFs-based materials for tumor therapy, including their application in cancer treatment and the underlying mechanisms.

Abstract:Metal-organic frameworks (MOFs) demonstrate excellent intrinsic characteristics for a variety of applications, including energy storage, catalysis, optics, sensory responses, and biomedical sciences. MOFs are being synthesized using various methods, and now efforts are mainly focused on designing a series of novel MOF composites with multifunctional attributes and physicochemical structures. Furthermore, current investigations are directed toward deploying the morphological and size properties to open new application windows. Cancer has turned into a hazard to human life, killing millions of people every year. Early-stage cancer detection is a challenging task, and most cancer deaths occur due to late-stage diagnosis because people do not get regular cancer screenings. Extensive research has been conducted on theranostic nanoplatforms that integrate several imaging technologies and therapy techniques for the detection and treatment of cancer. However, MOFs have been demonstrated to be effective as nanoplatforms for a wide range of biomedical applications due to their unique features, low cytotoxicity, high porosity, biodegradability due to labile metal–ligand bond, and tunable framework. MOF is an advanced material in the fight against cancer with significant importance. This review unlocks new routes to intricate MOFs in cancer diagnosis, drug delivery, bioimaging and highlights the development of these mechanisms for theranostic nanoplatforms. Furthermore, the future potential and significant obstacles of MOFs for cancer theranostics, bioimaging, and drug delivery are also demonstrated in this review.
Antitumor efficiency and photostability of newly green synthesized silver/graphene oxide nanocomposite on different cancer cell lines

Abstract:It is crucial to enhance new compounds for the treatment of most malignancies, and graphene oxide/silver nanocomposite (GO/Ag NC) has been paying attention to biomedical applications such as malignancies. In this work, green synthesized Ag@Cht NPs were successfully produced using chitosan for reduction and stabilization and added on GO sheet forming novel GO/Ag NC. Then, the produced anticancer nanomaterials GO, Ag@Cht NPs, and GO/Ag NC were assessed for their cytotoxicity against four distinct cancer cell lines: H460, HCT116, MDA-MB-468, and FaDu cells, at varying concentrations, using SRP assay after 48 h. The prepared nanomaterials were characterized by TEM, UV–Vis spectrophotometry, FTIRs, Raman spectroscopy and XRD. TEM images showed a regular spread of Ag@Cht NPs on the GO sheets with an average particle size of 15 nm. UV–Vis spectrophotometry shows two main characteristic peaks for GO/Ag NC, one close to 230 nm corresponds to GO, while the other peak at 425 nm due to Ag@Cht decorating the GO surface was blue shifted by few nanometers from 427 nm for Ag@Cht. Results of the laser irradiation by DPSS (diode-pumped solid state) confirm the photothermal stability of the prepared nanocomposite as there is no change in surface plasmon resonance (SPR) with varying exposure time. FTIRs measurements indicate that Ag ions interact with a hydroxyl group. This interaction shifts the O–H wavenumber and decreases the bond stretching intensity. In addition, Ag@Cht NPs and Ag/GO NC showed enhanced activity against cancerous cells. Results showed that GO, Ag@Cht NPs, and GO/Ag NC at (200, 400, and 600 μg/ml) had an impact on all evaluated cell lines. In MDA-MB-468, HCT116, H460, and FaDu cells, Ag@Cht NPs had the most effect across all employed cell lines, with IC50 values of 5.5, 9, 6, and 7.75 μg/ml, respectively. In conclusion, the produced novel nanocomposite may be an effective way to treat different cell lines, and future work is to use the prepared nanomaterials as anticancer drug delivery in photothermal chemotherapy combination treatment.

Abstract:Traditional cancer treatments include surgery, radiation, and chemotherapy. According to medical sources, chemotherapy is still the primary method for curing or treating cancer today and has been a major contributor to the recent decline in cancer mortality. Nanocomposites based on polymers and metal nanoparticles have recently received the attention of researchers. In the current study, a nanocomposite was fabricated based on carboxymethyl cellulose and silver nanoparticles (CMC-AgNPs) and their antibacterial, antifungal, and anticancer activities were evaluated. The antibacterial results revealed that CMC-AgNPs have promising antibacterial activity against Gram-negative (Klebsiella oxytoca and Escherichia coli) and Gram-positive bacteria (Bacillus cereus and Staphylococcus aureus). Moreover, CMC-AgNPs exhibited antifungal activity against filamentous fungi such as Aspergillus fumigatus, A. niger, and A. terreus. Concerning the HepG2 hepatocellular cancer cell line, the lowest IC50 values (7.9 ± 0.41 μg/mL) were recorded for CMC-AgNPs, suggesting a strong cytotoxic effect on liver cancer cells. As a result, our findings suggest that the antitumor effect of these CMC-Ag nanoparticles is due to the induction of apoptosis and necrosis in hepatic cancer cells via increased caspase-8 and -9 activities and diminished levels of VEGFR-2. In conclusion, CMC-AgNPs exhibited antibacterial, antifungal, and anticancer activities, which can be used in the pharmaceutical and medical fields.

Abstract:Metal-organic frameworks (MOFs) have been broadly applied in biomedical and other fields. MOFs have high porosity, a large comparative area, and good biostability and have attracted significant attention, especially in cancer therapies. This paper presents the latest applications of MOFs in chemodynamic therapy (CDT), sonodynamic therapy (SDT), photodynamic therapy (PDT), photothermal therapy (PTT), immunotherapy (IT), and combination therapy for breast cancer. A combination therapy is the combination of two different treatment modalities, such as CDT and PDT combination therapy, and is considered more effective than separate therapies. Herein, we have also discussed the advantages and disadvantages of combination therapy in the treatment of breast cancer. This paper aims to illustrate the potential of MOFs in new cancer therapeutic approaches, discuss their potential advantages, and provide some reflections on the latest research results.

Abstract:<p>Nanoscale metal‐organic frameworks (NMOFs) have attracted increasing attention for biomedical applications due to their large specific surface area, good biocompatibility, adjustable structures, and diverse functions. By choosing appropriate metal ions and ligands, NMOFs can be synthesized and regulated to assist the diagnosis and treatment of cancer, acting as imaging agents, drug carriers, and cancer therapeutic agents. This review summarizes the recent advances of NMOFs in synthesis, biocompatibility, imaging, and applications in cancer therapies. Among these, the term "biocompatibility" is used to outline their various biological characteristics, and it is mainly discussed from the aspects of size and surface properties of NMOFs. The imaging section mainly emphasizes the application advantages of NMOFs as imaging agents in magnetic resonance, computed tomography, and fluorescence imaging. The applications of NMOFs in four cancer therapies, including phototherapy, radiotherapy, microwave therapy, and ultrasonic therapy, are addressed, especially for thermal and dynamic therapy. Finally, the prospects and challenges of NMOFs in imaging and cancer therapies are also discussed.</p>

Abstract:Silver nanoparticles (AgNPs) are a potential class of nanomaterial for antibiosis and chemotherapeutic effects against human carcinoma cells. However, the DNA-damaging ability of free AgNPs pose the critical issues in their biomedical applications. Herein, we demonstrated a facile method to capture Ag+ ions and reduce them into active AgNPs within Zr-based metal-organic frameworks (MOFs) of UiO-66 with a mild reductant of DMF (AgNPs@UiO-66(DMF)). The average diameters of UiO-66 carriers and AgNPs were facilely controlled to be 140 and 10 nm, respectively. The obtained UiO-66 nanocarriers exhibited excellent biocompatibility and could be effectively endocytosed by cancer cells. Additionally, the AgNPs@UiO-66(DMF) could rapidly release Ag+ ions and efficiently inhibit the growth of cancer cells. The half maximal inhibitory concentration (IC50) values of the encapsulated AgNPs were calculated to be 2.7 and 2.45 μg mL−1 for SMMC-7721 and HeLa cells, respectively, which were much lower than those of free AgNPs in the reported works. Therefore, the developed AgNPs@UiO-66(DMF) not only maintained the therapeutic effect against cancer cells but also reduced the dosage of free AgNPs in chemotherapy treatment.Graphical abstractA mild reduction process was developed for the fabrication of AgNPs@UiO-66, which exhibited efficient induction of apoptosis in cancer cells.

Abstract:<p>Microbial colonization on material surfaces is ubiquitous. Biofilms derived from surface‐colonized microbes pose serious problems to the society from both an economical perspective and a health concern. Incorporation of antimicrobial nanocompounds within or on the surface of materials, or by coatings, to prevent microbial adhesion or kill the microorganisms after their attachment to biofilms, represents an important strategy in an increasingly challenging field. Over the last decade, many studies have been devoted to preparing meta‐based nanomaterials that possess antibacterial, antiviral, and antifungal activities to combat pathogen‐related diseases. Herein, an overview on the state‐of‐the‐art antimicrobial nanosized metal‐based compounds is provided, including metal and metal oxide nanoparticles as well as transition metal nanosheets. The antimicrobial mechanism of these nanostructures and their biomedical applications such as catheters, implants, medical delivery systems, tissue engineering, and dentistry are discussed. Their properties as well as potential caveats such as cytotoxicity, diminishing efficacy, and induction of antimicrobial resistance of materials incorporating these nanostructures are reviewed to provide a backdrop for future research.</p>

Abstract:Metal-organic frameworks (MOFs) have received extensive attention in tumor therapy because of their advantages, including large specific surface area, regular pore size, adjustable shape, and facial functionalization. MOFs are porous materials formed by the coordination bonding of metal clusters and organic ligands. This review summarized the most recent advancements in tumor treatment based on nMOFs. First, we discuss the classification of MOFs, which primarily include the series of Isoreticular MOF (IRMOF), Zeolitic imidazolate framework (ZIF), Coordination Pillared-Layer (CPL), Materials of Institute Lavoisier (MIL), Porous Coordination Network (PCN), University of Oslo (UiO) and Biological metal-organic frameworks (BioMOFs). Then, we discuss the use of nMOFs to anticancer therapy, including drug delivery strategies, photodynamic therapy (PDT), photothermal therapy (PTT) and combination therapy. Finally, the obstacles and opportunities in nMOFs are discussed.

Abstract:Research on biocompatible metal-organic frameworks (MOFs) has recently attracted much attention from researchers as drug delivery systems. In this study, for the first time, Cu MOFs grown on graphene oxide capped with magnetic hydroxyapatite (MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 ), and then the surface of the resulting nanocomposites was modified with the l -lysine amino acid (Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 ) to prepare a controlled and pH-sensitive nanocomposite for targeted delivery of doxorubicin (DOX) to MDA-MB-231 breast cancer cells and MCF-10A normal cells. Different techniques confirmed the successful preparation of the designed nanocomposites. The values of encapsulation efficiencies (EE%) for MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 was found to be 79.3 % and 96.5 %, respectively. The in vitro DOX release results showed that the Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 has a more controlled release than that of MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 due to the slow diffusion of the drug through the Lys shell. In addition, both nanocomposites showed a pH-response release behavior. The release mechanism of DOX by the designed nanocomposites also revealed that the Korsmeyer-Peppas model is the best model to describe the release of the drug. Furthermore, in vitro cytotoxicity investigation confirmed that the prepared MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 nanocomposite had good biocompatibility against MDA-MB-231 and MCF-10A cells. Whereas, DOX-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 and DOX-Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 against MDA-MB-231 cells increased the cytotoxicity as a result of the targeting and controlled delivery of the DOX to the cancer cells. The in vitro antioxidant activity and hemolysis experiments revealed that the designed Lys-MOF Cu@GO/HAp-SiO 2 -Fe 3 O 4 have excellent antioxidant activity and good haemocompatibility, respectively, as a suitable nanocomposites for drug applications. Therefore, it is suggested that the prepared nanocomposites can be good for biological applications.

Ultrasonic synthesis, characterization, DFT and molecular docking of a biocompatible Zn-based MOF as a potential antimicrobial, anti-inflammatory and antitumor agent

Abstract:Zinc metal-organic frameworks have emerged as promising candidates, demonstrating excellent biological properties stemming from the unique characteristics of MOFs and zinc. In this study, we employed a facile method to synthesize a zinc metal-organic framework [Zn(IP)(H2O)] using ultrasound irradiation, with the linker being isophthalic acid (IPA) (1,3-benzene dicarboxylic acid). The parent Zn-MOF and two Ag/Zn-MOF samples prepared via loading and encapsulation methods were comprehensively characterized using various techniques, including FT-IR, XRD, SEM, TEM, N2 adsorption-desorption isotherm, UV-vis spectroscopy and TGA. The parent Zn-MOF and two Ag/Zn-MOF samples exhibited a broad spectrum of antibacterial effects. Remarkably, genomic DNA of P. aeruginosa was effectively degraded by Zn-MOF, further supporting its potent antibacterial results. The free radical inhibition assay demonstrated a 71.0% inhibition under the influence of Zn-MOF. In vitro cytotoxicity activity of Zn-MOF against HepG-2 and Caco-2 cell lines revealed differential cytotoxic effects, with higher cytotoxicity against Caco-2 as explored from the IC50 values. This cytotoxicity was supported by the high binding affinity of Zn-MOF to CT-DNA. Importantly, the non-toxic property of Zn-MOF was confirmed through its lack of cytotoxic effects against normal lung cell (Wi-38). The anti-inflammatory treatment of Zn-MOF achieved 75.0% efficiency relative to the standard Ibuprofen drug. DFT and docking provided insights into the geometric stability of Zn-MOF and its interaction with active amino acids within selected proteins associated with the investigated diseases. Finally, the synthesized Zn-MOF shows promise for applications in cancer treatment, chemoprevention, and particularly antibacterial purposes.

Abstract:Applications of metal-organic framework (MOF) nanostructures in biomedicine have rapidly expanded over the past decade. MOFs are characterized by their high functionality, surface area, and porosity, providingan ideal platform forseveral biomedical applications. This study explores the potential of a novel therapeutic approach for liver cancer, incorporating Safranal, a bioactive molecule derived from saffron, within the iron-based Metal-Organic Framework (MOF), MIL-88B(Fe). The successfully loaded Safranal-MIL-88B(Fe) composite nanostructure was comprehensively characterized and tested for effectiveness against HepG2 liver cancer cells. Furthermore, we investigated its antibacterial efficacy against E. coli and Lactobacillus strains. The proposed mechanism of action of the Safranal-MIL-88B(Fe) nanostructures potentially shows the effect of the release of the ferric ions and Safranal in vitro against HepG2 cancer cells and both bacterial strains. Our findings show significant promise for Safranal-loaded MIL-88B(Fe) as a dual-purpose therapeutic agent, opening exciting avenues for future biomedical applications.

Abstract:Nanotechnology is a growing field, with many potential biomedical applications of nanomedicine for the treatment of different diseases, particularly cancer, on the horizon. Graphene oxide (GO) nanoparticles can act as carbon-based nanocarriers with advantages such as a large surface area, good mechanical strength, and the capacity for surface modification. These nanostructures have been extensively used in cancer therapy for drug and gene delivery, photothermal therapy, overcoming chemotherapy resistance, and for imaging procedures. In the current review, we focus on the biological functions of GO nanoparticles as regulators of apoptosis and autophagy, the two major forms of programmed cell death. GO nanoparticles can either induce or inhibit autophagy in cancer cells, depending on the conditions. By stimulating autophagy, GO nanocarriers can promote the sensitivity of cancer cells to chemotherapy. However, by impairing autophagy flux, GO nanoparticles can reduce cell survival and enhance inflammation. Similarly, GO nanomaterials can increase ROS production and induce DNA damage, thereby sensitizing cancer cells to apoptosis. In vitro and in vivo experiments have investigated whether GO nanomaterials show any toxicity in major body organs, such as the brain, liver, spleen, and heart. Molecular pathways, such as ATG, MAPK, JNK, and Akt, can be regulated by GO nanomaterials, leading to effects on autophagy and apoptosis. These topics are discussed in this review to shed some lights towards the biomedical potential of GO nanoparticles and their biocompatibility, paving the way for their future application in clinical trials.
