Abstract:The coupled analysis of corticomuscular function based on physiological electrical signals can identify differences in causal relationships between electroencephalogram (EEG) and surface electromyogram (sEMG) in different motor states. The existing methods are mainly devoted to the analysis in the same frequency band, while ignoring the cross-band coupling, which plays an active role in motion control. Considering the inherent multiscale characteristics of physiological signals, a method combining Ordinal Partition Transition Networks (OPTNs) and Multivariate Variational Modal Decomposition (MVMD) was proposed in this paper. The EEG and sEMG were firstly decomposed on a time-frequency scale using MVMD, and then the coupling strength was calculated by the OPTNs to construct a corticomuscular coupling network, which was analyzed with complex network parameters. Experimental data were obtained from a self-acquired dataset consisting of EEG and sEMG of 16 healthy subjects at different sizes of constant grip force. The results showed that the method was superior in representing changes in the causal link among multichannel signals characterized by different frequency bands and grip strength patterns. Complex information transfer between the cerebral cortex and the corresponding muscle groups during constant grip force output from the human upper limb. Furthermore, the sEMG of the flexor digitorum superficialis (FDS) in the low frequency band is the hub in the effective information transmission between the cortex and the muscle, while the importance of each frequency component in this transmission network becomes more dispersed as the grip strength grows, and the increase in coupling strength and node status is mainly in the γ band (30~60Hz). This study provides new ideas for deconstructing the mechanisms of neural control of muscle movements.

Abstract:Hand movement in humans is verified as asymmetries and lateralization, and two hemispheres make some distinct but complementary contributions in the control of hand movement. However, little research has been done on whether the information transfer of the motor system is different between left and right hand movement. Considering the importance of functional corticomuscular coupling (FCMC) between the motor cortex and contralateral muscle in movement assessment, this study aimed to explore the differences between left and right hand by investigating the interaction between muscle and brain activity. Here, we applied the transfer spectral entropy (TSE) algorithm to quantize the connection between electroencephalogram (EEG) over the brain scalp and electromyogram (EMG) from extensor digitorum (ED) and flexor digitorum superficialis (FDS) muscles recorded simultaneously during a gripping task. Eight healthy subjects were enrolled in this study. Results showed that left hand yielded narrower and lower beta synchronization compared to the right. Further analysis indicated coupling strength in EEG-EMG(FDS) combination was higher at beta band than that in EEG-EMG(ED) combination, and exhibited distinct differences between descending (EEG to EMG direction) and ascending (EMG to EEG direction) direction. This study presents the distinctions of beta-range FCMC between left and right hand, and confirms the importance of beta synchronization in understanding the mechanism of motor stability control. The cortex-muscle FCMC might be used as an evaluation approach to explore the difference between left and right movement system.

Abstract:Objective: Functional coupling between the motor cortex and muscle activity is commonly detected and quantified by cortico-muscular coherence (CMC) or Granger causality (GC) analysis, which are applicable only to linear couplings and are not sufficiently sensitive: some healthy subjects show no significant CMC and GC, and yet have good motor skills. The objective of this work is to develop measures of functional cortico-muscular coupling that have improved sensitivity and are capable of detecting both linear and non-linear interactions. Methods: A multiscale wavelet transfer entropy (TE) methodology is proposed. The methodology relies on a dyadic stationary wavelet transform to decompose electroencephalogram (EEG) and electromyogram (EMG) signals into functional bands of neural oscillations. Then, it applies TE analysis based on a range of embedding delay vectors to detect and quantify intra- and cross-frequency band cortico-muscular coupling at different time scales. Results: Our experiments with neurophysiological signals substantiate the potential of the developed methodologies for detecting and quantifying information flow between EEG and EMG signals for subjects with and without significant CMC or GC, including non-linear cross-frequency interactions, and interactions across different temporal scales. The obtained results are in agreement with the underlying sensorimotor neurophysiology. Conclusion: These findings suggest that the concept of multiscale wavelet TE provides a comprehensive framework for analysing cortex-muscle interactions. Significance: The proposed methodologies will enable developing novel insights into movement control and neurophysiological processes more generally.

Abstract:Objective: This study aims to explore alterations in corticomuscular and cortical coupling during the rehabilitation of stroke patients. Methods: We initiated the analysis by employing variational modal decomposition (VMD) on electromyography (EMG) data, followed by the application of VDM-transfer entropy (VMD-TE) to quantify the coupling strength between electroencephalogram (EEG) and EMG signals. Subsequently, we constructed the VMD-TE connection matrix and analyzed the clustering coefficient and small-world attributes within the cortico-muscular functional network (CMFN). Finally, a random forest algorithm was employed to extract features from the VMD-TE connection matrix across different rehabilitation periods. Results: Beta waves in EEG were emerged as the key information carrier between the cortex and muscle, and the CMFN of patients with the beta frequency band has small-world characteristics. During rehabilitation, we observed a decrease in coupling between the initially affected motor cortex and muscle, accompanied by an increase in coupling between the frontal region and muscle. Conclusion: Our findings suggest potential neuro-remodeling in stroke patients after rehabilitation, with CFMN serving as a valuable metric for assessing cortico-muscular coupling.

Abstract:Background During isometric compensation of modulated low-level forces corticomuscular coherence (CMC) has been shown to occur in high-beta or gamma-range. The influence of the frequency of force modulation on CMC has up to now remained unexplored. We addressed this question by investigating CMC, motor performance, and cortical spectral power during a visuomotor task in which subjects had to compensate a modulated force of 8% of the maximum voluntary contraction exerted on their right index finger. The effect of three frequencies of force modulation (0.6, 1.0 and 1.6 Hz) was tested. EEG, EMG from first dorsal interosseus, hand flexor and extensor muscles, and finger position were recorded in eight right-handed women. Results Five subjects showed CMC in gamma- (28-45 Hz) and three in beta-range (15-30 Hz). Beta- and gamma-range CMC and cortical motor spectral power were not modulated by the various frequencies. However, a sharp bilateral CMC peak at 1.6 Hz was observed, but only in the five gamma-range CMC subjects. The performance error increased linearly with the frequency. Conclusions Our findings suggest that the frequency of force modulation has no effect on the beta- and gamma-range CMC during isometric compensation for modulated forces at 8% MVC. The beta- and gamma-range CMC may be related to interindividual differences and possibly to strategy differences.

Abstract:Cortical-muscular functional coupling reflects the interaction between the cerebral cortex and the muscle activities. Corticomuscular coherence (CMC) has been extensively revealed in sustained contractions of various upper- and lower-limb muscles during static and dynamic force outputs. However, it is not well-understood that the CMC modulation mechanisms, i.e., the relation between a cerebral hemisphere and dynamic motor controlling limbs at constant speeds, such as isokinetic movement. In this paper, we explore the CMC between upper arm flexors/extensors movement and motor cortex during isometric exercise and cyclically isokinetic movement. We also provide further insights of frequency-shift and the neural pathway mechanisms in isokinetic movement by evaluating the coherence between motor cortex and agonistic or antagonistic muscles. This study is the first to investigate the relationship between cortical-muscular functional connections in elbow flexion-extension movement with constant speeds. The result shows that gamma-range coherence for isokinetic movement is greatly increased compared with isometric exercise, and significant CMC is observed in the entire flexion-extension stage regardless the nature of muscles contraction, although dominant synchronization of cortical oscillation and muscular activity resonated in sustained contraction stage principally. Besides, the CMC for extensors and flexors are explicitly consistent in contraction stage during cyclically isokinetic elbow movement. It is concluded that cortical-muscular coherence can be dynamically modulated as well as selective by cognitive demands of the body, and the time-varying mechanisms of the synchronous motor oscillation exist in healthy individuals during dynamic movement.

Abstract:Corticomuscular coupling analysis, i.e., examining the relations between simultaneously recorded brain (e. g., electroencephalography-EEG) and muscle (e. g., electromyography-EMG) signals, is a useful tool for understanding aspects of human motor control. Traditionally, the most popular method to assess corticomuscular coupling has been the pairwise magnitude-squared coherence (MSC) between EEG and concomitant EMG recordings. In this paper, we propose assessing corticomuscular coupling by combining partial least squares (PLS) and independent component analysis (ICA), which addresses many of the limitations of MSC, such as difficulty in robustly assessing group inference and relying on the biologically implausible assumption of pairwise interaction between brain and muscle recordings. In the proposed framework, response relevance and statistical independence are jointly incorporated into a multiobjective optimization function to meaningfully combine the goals of PLS and ICA under the same mathematical umbrella. Simulations, performed under realistic assumptions, illustrated the utility of such an approach. The method was extended to address intersubject variability to robustly discover common corticomuscular coupling patterns across subjects. We then applied the proposed framework to concurrent EEG and EMG data collected in a Parkinson's disease (PD) study. The results from applying the proposed technique revealed temporal components in the EEG and EMG that were significantly correlated with one another. In addition to the expected motor areas, the corresponding spatial activation patterns demonstrated enhanced occipital connectivity in PD subjects, consistent with previous studies suggesting that PD subjects rely excessively on visual information to counteract the deficiency in being able to generate internal commands from their affected basal ganglia.

However, there are certain limitations associated with the MSC, including the difficulty in robustly assessing group inference, only dealing with two types of datasets simultaneously and the biologically implausible assumption of pair-wise interactions. To overcome such limitations, in this paper, we propose assessing corticomuscular coupling by combining multiset canonical correlation analysis (M-CCA) and joint independent component analysis (jICA). Abstract:Corticomuscular coupling analysis based on multiple data sets such as electroencephalography (EEG) and electromyography (EMG) signals provides a useful tool for understanding human motor control systems. Two probably most popular methods are the pair-wise magnitude-squared coherence (MSC) between EEG and simultaneously-recorded EMG signals, and partial least square (PLS). Unfortunately, MSC and PLS generally deal with only two types of data sets at the same time, while we may need to analyze more than two types of data sets. Moreover, it is not straightforward to extend MSC to the group level for combining results across subjects. Also, PLS can have the information mixing problem since only the variations in one data set are used to predict the other data set. To address these concerns, we propose a joint multimodal analysis framework for corticomuscular coupling analysis. The proposed framework models multiple data spaces simultaneously in a multidirectional fashion. Furthermore, to address the inter-subject variability concern in real-world medical applications, we extend the proposed framework from the individual subject level to the group level to obtain common corticomuscular coupling patterns across subjects. We apply the proposed framework to concurrent EEG, EMG and behavior data collected in a Parkinson's disease (PD) study. The results reveal several highly correlated temporal patterns among the three types of signals and their corresponding spatial activation patterns. In PD subjects, there are enhanced connections between occipital region and other regions, which is consistent with the previous medical finding. The proposed framework is a promising technique for performing multi-subject and multi-modal data analysis.

Abstract:Objective. Corticomuscular coherence (CMC) is widely used to detect and quantify the coupling between motor cortex and effector muscles. It is promisingly used in human–machine interaction (HMI) supported rehabilitation training to promote the closed-loop motor control for stroke patients. However, suffering from weak coherence features and low accuracy in contingent neurofeedback, its application to HMI rehabilitation robots is currently limited. In this paper, we propose the concept of spatial–temporal CMC (STCMC), which is the coherence by refining CMC with delay compensation and spatial optimization. Approach. The proposed STCMC method measures the coherence between electroencephalogram (EEG) and electromyogram (EMG) in the multivariate spaces. Specifically, we combined delay compensation and spatial optimization to maximize the absolute value of the coherence. Then, we tested the reliability and effectiveness of STCMC on neurophysiological data of force tracking tasks. Main results. Compared with CMC, STCMC not only enhanced the coherence significantly between brain and muscle signals, but also produced higher classification accuracy. Further analysis showed that temporal and spatial parameters estimated by the STCMC reflected more detailed brain topographical patterns, which emphasized the different roles between the contralateral and ipsilateral hemisphere. Significance. This study integrates delay compensation and spatial optimization to give a new perspective for corticomuscular coupling analysis. It is also feasible to design robotic neurorehabilitation paradigms by the proposed method.

Abstract:Isometric compensation of predictably frequency-modulated low forces is associated with corticomuscular coherence (CMC) in beta and low gamma range. It remains unclear how the CMC is influenced by unpredictably modulated forces, which create a mismatch between expected and actual sensory feedback. We recorded electroencephalography from the contralateral hand motor area, electromyography (EMG), and the motor performance of 16 subjects during a visuomotor task in which they had to isometrically compensate target forces at 8% of the maximum voluntary contraction with their right index finger. The modulated forces were presented with predictable or unpredictable frequencies. We calculated the CMC, the cortical motor alpha-, beta-, and gamma-range spectral powers (SP), and the task-related desynchronization (TRD), as well as the EMG SP and the performance. We found that in the unpredictable condition the CMC was significantly lower and associated with lower cortical motor SP, stronger TRD, higher EMG SP, and worse performance. The findings suggest that due to the mismatch between predicted and actual sensory feedback leading to higher computational load and less stationary motor state, the unpredictable modulation of the force leads to a decrease in corticospinal synchrony, an increase in cortical and muscle activation, and a worse performance.

Abstract:The interaction between cortex and muscles under hand motor with different force states has not been quantitatively investigated yet, which to some extent places the optimized movement tasks design for brain-computer interface (BCI) applications in hand motor rehabilitation under uncertainty. Converging evidence has suggested that both the descending corticospinal pathway and ascending sensory feedback pathway are involved in the generation of corticomuscular coupling. The present study aimed to explore the corticomuscular coupling during hand motor task with enhancing force and steady-state force. Twenty healthy subjects performed precision grip with enhancing and static force using the right hand with visual feedback of exerted force. Mutual information and Granger causal connectivity were assessed between electroencephalography (EEG) over primary motor cortex and electromyography (EMG) recordings, and statistically analyzed. The results showed that the mutual information value was significantly larger for static force in the beta and alpha frequency band than enhancing force state. Furthermore, compared with enhancing force, the Granger causal connectivity of descending pathways from cortex to muscle was significantly larger for static force in the beta and high alpha frequency band (10-20 Hz), indicating the connection between the primary motor cortex and muscle was strengthened for static force. In summary, the hand grip with static force resulted in an increasing corticomuscular coupling from EEG over the primary motor cortex to EMG compared with enhancing force, implying more attention was required in the static force state. These results have important implications toward motor rehabilitation therapy design for the recovery of impaired hand motor functions.

Abstract:Surface electromyography (sEMG) can capture muscle activity and motor function, which is considered an effective tool for assessing local muscle fatigue. Due to the complex environment and individual differences, the collected sEMG signals contain lots of noising information. A novel method is devised to effectively solve the problem of sEMG denoising based on complementary ensemble empirical mode decomposition (CEEMD) and multi-scale entropy (MSEn). The method uses the concept of MSEn and analyzes the feature trend of MSEn of intrinsic mode functions (IMFs) obtained by CEEMD. The method adaptively determines the main components of IMFs according to the complexity of sEMG on the time scale, avoiding insufficient selection based on experience. Actual experiments verified the effectiveness of the method.
Altered Coupling between Motion-Related Activation and Resting-State Brain Activity in the Ipsilesional Sensorimotor Cortex after Cerebral Stroke.

Abstract:Functional connectivity maps using resting-state functional magnetic resonance imaging (rs-fMRI) can closely resemble task fMRI activation patterns, suggesting that resting-state brain activity may predict task-evoked activation or behavioral performance. However, this conclusion was mostly drawn upon a healthy population. It remains unclear whether the predictive ability of resting-state brain activity for task-evoked activation would change under different pathological conditions. This study investigated dynamic changes of coupling between patterns of resting-state functional connectivity (RSFC) and motion-related activation in different stages of cerebral stroke. Twenty stroke patients with hand motor function impairment were involved. rs-fMRI and hand motion-related fMRI data were acquired in the acute, subacute, and early chronic stages of cerebral stroke on a 3-T magnetic resonance (MR) scanner. Sixteen healthy participants were enrolled as controls. For each subject, an activation map of the affected hand was first created using general linear model analysis on task fMRI data, and then an RSFC map was determined by seeding at the peak region of hand motion activation during the intact hand task. We then measured the extent of coupling between the RSFC maps and motion-related activation maps. Dynamic changes of the coupling between the two fMRI maps were estimated using one-way repeated measures analysis of variance across the three stages. Moreover, imaging parameters were correlated with motor performances. Data analysis showed that there were different coupling patterns between motion-related activation and RSFC maps associating with the affected motor regions during the acute, subacute, and early chronic stages of stroke. Coupling strengths increased as the recovery from stroke progressed. Coupling strengths were correlated with hand motion performance in the acute stage, while coupling recovery was negatively correlated with the recovery outcome of hand motion performance in the early chronic stages. Couplings between RSFC and motion-related activation were dynamically changed with stroke progression, which suggested changes in the prediction of resting-state brain activity for task-evoked brain activity in different pathological states. The changes in coupling strength between these two types of brain activity implicate a reparative mechanism of brain injury and may represent a biomarker for predicting motor recovery in cerebral stroke.

Abstract:Functional coupling between the motor cortex and muscle activity is usually detected and characterized using the spectral method of corticomuscular coherence (CMC). This functional coupling occurs with a time delay, which, if not properly accounted for, may decrease the coherence and make the synchrony difficult to detect. In this paper, we introduce the concept of CMC with time lag (CMCTL), that is the coherence between segments of motor cortex electroencephalogram (EEG) and electromyography (EMG) signals displaced from a central observation point. This concept is motivated by the need to compensate for the unknown delay between coupled cortex and muscle processes. We demonstrate using simulated data that under certain conditions the time lag between EEG and EMG segments at points of local maxima of CMCTL corresponds to the average delay along the involved corticomuscular conduction pathways. Using neurophysiological data, we then show that CMCTL with appropriate time lag enhances the coherence between cortical and muscle signals, and that time lags which correspond to local maxima of CMCTL provide estimates of delays involved in corticomuscular coupling that are consistent with the underlying physiology.

Abstract:Objective. A body movement involves the complicated information exchange between the central and peripheral systems, which is characterized by the dynamical coupling patterns between the multiple brain areas and multiple muscle units. How the central and peripheral nerves coordinate multiple internal brain regions and muscle groups is very important when accomplishing the action. Approach. In this study, we extend the adaptive directed transfer function to construct the time-varying networks between multiple corticomuscular regions and divide the movement duration into different stages by the time-varying corticomuscular network patterns. Main results. The inter dynamical corticomuscular network demonstrated the different interaction patterns between the central and peripheral systems during the different hand movement stages. The muscles transmit bottom-up movement information in the preparation stage, but the brain issues top-down control commands and dominates in the execution stage, and finally, the brain's dominant advantage gradually weakens in the relaxation stage. When classifying the different movement stages based on time-varying corticomuscular network indicators, an average accuracy above 74% could be reliably achieved. Significance. The findings of this study help deepen our knowledge of central-peripheral nerve pathways and coordination mechanisms, and also provide opportunities for monitoring and regulating movement disorders.

Abstract:Introduction: Upper limb rehabilitation assessment plays a pivotal role in the recovery process of stroke patients. The current clinical assessment tools often rely on subjective judgments of healthcare professionals. Some existing research studies have utilized physiological signals for quantitative assessments. However, most studies used single index to assess the motor functions of upper limb. The fusion of surface electromyography (sEMG) and functional near-infrared spectroscopy (fNIRS) presents an innovative approach, offering simultaneous insights into the central and peripheral nervous systems. Methods: We concurrently collected sEMG signals and brain hemodynamic signals during bilateral elbow flexion in 15 stroke patients with subacute and chronic stages and 15 healthy control subjects. The sEMG signals were analyzed to obtain muscle synergy based indexes including synergy stability index ( SSI ), closeness of individual vector ( C V ) and closeness of time profile ( C T ). The fNIRS signals were calculated to extract laterality index ( LI ). Results: The primary findings were that C V , SSI and LI in posterior motor cortex (PMC) and primary motor cortex (M1) on the affected hemisphere of stroke patients were significantly lower than those in the control group ( p < 0.05). Moreover, C V , SSI and LI in PMC were also significantly different between affected and unaffected upper limb movements ( p < 0.05). Furthermore, a linear regression model was used to predict the value of the Fugl-Meyer score of upper limb (FMul) ( R 2 = 0.860, p < 0.001). Discussion: This study established a linear regression model using force, C V , and LI features to predict FMul scale values, which suggests that the combination of force, sEMG and fNIRS hold promise as a novel method for assessing stroke rehabilitation.

Abstract:Functional corticomuscular coupling (FCMC) probes multi-level information communication in the sensorimotor system. The canonical Coherence (caCOH) method has been leveraged to measure the FCMC between two multivariate signals within the single scale. In this paper, we propose the concept of multiscale canonical Coherence (MS-caCOH) to disentangle complex multi-layer information and extract coupling features in multivariate spaces from multiple scales. Then, we verified the reliability and effectiveness of MS-caCOH on two types of data sets, i.e., a synthetic multivariate data set and a real-world multivariate data set. Our simulation results showed that compared with caCOH, MS-caCOH enhanced coupling detection and achieved lower pattern recovery errors at multiple frequency scales. Further analysis on experimental data demonstrated that the proposed MS-caCOH method could also capture detailed multiscale spatial-frequency characteristics. This study leverages the multiscale analysis framework and multivariate method to give a new insight into corticomuscular coupling analysis.

Abstract:Summary: The present study was aimed at investigating the corticomuscular synchronization in beta- (15-30 Hz) and gamma-range (30-45 Hz) during isometric compensation of low-level forces. It is still unknown to what extent the synchronization processes in these frequency ranges can coexist or influence each other when the static component only is modulated in a dynamic stimulation pattern. We investigated the corticomuscular coherence (CMC), as well as the cortical spectral power (SP) during a visuomotor task, where 8%, 16% and 24% of the maximal voluntary contraction (MVC) were used. Seven healthy right-handed female subjects compensated isometrically the different dynamic forces with their right index finger. EEG was recorded from 52 scalp positions and belly-tendon bipolar EMG from the first dorsal interosseus muscle (FDI). Under the three conditions investigated, the beta- and gamma-range CMC existed in parallel. They behaved in a different manner: while the beta-range coherence increased linearly during higher force application, the gamma-range CMC was not significantly modulated by the force levels. Our results suggest that although gamma-range CMC is functionally associated to the isometric compensation of dynamic forces, broad beta-range CMC can fulfill functions of motor control simultaneously different when low-level forces are applied.
