Abstract:Proper disposal and utilization of dead pig carcasses are problems of public concern. The combination of hydrothermal pretreatment (HTP) and anaerobic digestion is a promising method to treat these wastes, provided that digestion inhibition is reduced. For this reason, the aim of this work was to investigate the optimal HTP temperature (140–180°C) for biogas production during anaerobic digestion of dead pigs in batch systems. In addition, the effects of hydrochar addition (6 g/L) on anaerobic digestion of pork products after HTP in continuous stirred tank reactors (CSTR) were determined. According to the results, 90% of lipids and 10% of proteins present in the pork were decomposed by HTP. In addition, the highest chemical oxygen demand (COD) concentration in liquid products (LP) reached 192.6 g/L, and it was obtained after 170°C HTP. The biogas potential from the solid residue (SR) and LP was up to 478 mL/g-VS and 398 mL/g-COD, respectively. A temperature of 170°C was suitable for pork HTP, which promoted the practical biogas yield because of the synergistic effect between proteins and lipids. Ammonia inhibition was reduced by the addition of hydrochar to the CSTR during co-digestion of SR and LP, maximum ammonia concentration tolerated by methanogens increased from 2.68 to 3.38 g/L. This improved total biogas yield and degradation rate of substrates, reaching values of 28.62 and 36.06%, respectively. The acetate content in volatile fatty acids (VFA) may be used as an index that reflects the degree of methanogenesis of the system. The results of the present work may also provide guidance for the digestion of feedstock with high protein and lipid content.

Abstract:To investigate the effects of heat treatment on the microstructure and digestive behaviors of pork, meat samples were subjected to a 100 °C water bath for 26 min. The inner, medium, and outer layers were assigned and analyzed according to the temperature gradient. Compared to the raw samples, significant changes were observed in the microscopic structure of pork. As the temperature increased, the myofibrillar structure of pork underwent increasingly severe damage and the moisture content decreased significantly (P < 0.05). Moreover, differential peptides were identified in digested products of the inner, middle, and outer layers of cooked pork, which are mainly derived from the structural proteins of pork. The outcomes of molecular docking indicated that a greater number of hydrogen bonds were formed between myosin and the digestive enzyme in the inner layer, rather than other parts, contributing to the transformation of digestive behaviors.

Abstract:Heat treatment affects the structural properties of meat proteins, which in turn leads to changes in their sensitivity to digestive enzymes, further affecting the nutritional value of meat and meat products. The mechanism of changes in the structure and digestive properties of myosin under different heating conditions were studied. An increase in heating temperature led to the exposure of internal groups to a polar environment, but to a decrease in the sturdy alpha-helix structure of myosin (p < 0.05). The results of tryptophan fluorescence verified that the tertiary structure of the protein seemed to be unfolded at 70 degrees C. Higher protein denaturation after overheating, as proven by the sulfhydryl contents and turbidity, caused irregular aggregate generation. The excessive heating mode of treatment at 100 degrees C for 30 min caused myosin to exhibit a lower degree of pepsin digestion, which increased the Michaelis constant (Km value) of pepsin during the digestion, but induced the production of new peptides with longer peptide sequences. This study elucidates the effects of cooking temperature on the conformation of myosin and the change in digestibility of pepsin treatment during heating.

Abstract:Abstract Tibetan pigs inhabit the plateau regions and engage in extensive exercise. Their meat is firm and less palatable, and appropriate cooking conditions can improve the flavor of the meat. This study aimed to explore the impact of cooking temperatures and times on the cooking loss rate, moisture content, tenderness, and color of Tibetan pork to elucidate their effects on the eating quality of this meat. The results indicated a negative correlation between temperature and moisture content, which decreased from 72% to 65% as the temperature increased. Meanwhile, the oxidation of myofibrillar proteins substantially increased with cooking temperature and time, resulting in a gradual augmentation of the cooking loss and shear force of the pork. The migration of bound water and the release of free water from the pork were faster after treatment at 80 °C compared with 50 °C. The results of infrared spectroscopy suggested that prolonged cooking led to the conversion of α-helix into β-sheet in the secondary structure of myofibrillar proteins. The microstructures observed through scanning electron microscopy exhibited a gradual degradation of connective tissues in the muscles at high temperatures, and the fiber structure of the muscles became more condensed and granular, blurring the textural appearance of the muscles. These results indicated that prolonged cooking time at a low temperature (steaming for 6 h at 50°C ) was conducive to the optimal tenderness of Tibetan pork. This study provides a theoretical basis for improving the meat quality of Tibetan pork products and developing more rational processing methods.

Abstract:The objective of this study was to investigate the effects of ohmic (OH) and water bath (WB) cooking on shear parameters, protein degradation and ultrastructure changes of porcine longissimus dorsi muscle at the same endpoint temperatures (EPTs; range, 20–100 °C). The cooking loss and Warner–Bratzler shear force of the OH-cooked meat were significantly lower (P < 0.05) while protein solubility, pH and endothermic transition temperature were higher than those obtained by WB cooking at the same EPTs (range, 20–80 °C). Sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis showed that during OH cooking, the meat had slightly fainter protein bands than that of WB-cooked ones. Less obvious shrinkage of the sarcomere and loss in the structure of Z discs were detected especially in OH-cooked meat at EPTs (100 °C). Strong correlations among pH, cooking loss, Warner–Bratzler shear force, sarcoplasmic protein solubility, Tmax2, and Tmax3 were observed in meat following OH cooking.

Abstract:The author presents part II of his review on the treatment of refractory myeloma. Treatment with large doses of melphalane, 140 mg/m2, was associated with a high death rate and therefore it is used nowadays only in combination with autologous transplantation or treatment with leucocytic growth factors (GM-CSF and G-CSF). Medium doses of melphalane (25 mg/m2 to 70 mg/m2) are tolerated better and are one of the possible approaches. Another possible therapeutic procedure is whole-body irradiation. The advantage of extensive irradiation is rapid regression of pain. Interferon-alpha achieves a therapeutic response in some 20% of refractory patients. Finally the author presents some data on transplantation of bone marrow in patients with multiple myeloma.

Abstract:This study aimed to evaluate the effect of Sous-vide (SV) cooking on the quality, flavor and digestion characteristics of braised pork. Traditional (TD) sample had the highest fat content and malondialdehyde (MDA) value, but the lowest protein content and total sulfhydryl (SH) content (P < 0.05). The SH content in SV samples decreased with the heating time, but MDA content increased (P < 0.05). In addition, alpha-helix content of TD samples was higher but beta-sheet content was lower than SV samples. LC-MS/MS indicated that SV samples at 65 degrees C for 8 h had potential bioactive and antioxidant peptides. GC-MS mainly identified 395 volatile components and SV samples at 70 degrees C for 8 h and 75 degrees C for 8 h had similar flavor compounds to TD samples. Thus, Sous-vide cooking produced better quality and digestion characteristics of braised pork. These findings give a new insight into the associations of processing methods with meat quality.

Abstract:The aim of this study was to determine the influence of cutting methods on the fiber structure and quality attributes of the cooked pork loin muscle. The culinary portions were cut perpendicularly (D, diagonal) or along the muscle fibers (P, parallel). The samples were heated to 72 °C in water bath (85 °C or 95 °C), steam (100 °C), or oven (125 °C or 150 °C). The P samples had a significantly smaller cooking loss (2-4.5%), greater water-holding capacity (3-5%), and lower shear force (0.35-2.2 N) than D samples except for the 100P, whose cooking loss was higher (by 1.9%) than the 100D. Heat-induced structural changes in the muscle, sarcomere shortening, and destruction of the perimysium and endomysium were affected by the cutting methods and cooking conditions. The findings justify the advisability of cutting individual culinary portions from pork loin muscle in parallel to the muscle fibers in order to increase raw meat efficiency.

Abstract:The aim of the study was to evaluate the effect of marinating (3 or 6 days) in kefir (KE), yogurt (YO) and buttermilk (BM) and sous-vide cooking (SV) at 60 or 80 °C on changes in the protein profile of pork in relation to its sensory quality. In the marinated raw meat, an increased share of some fractions of myofibrillar and cytoskeletal proteins and calpains were found. The greatest degradation of proteins, regardless of time, was caused by marinating in YO and KE and cooking SV at 80 °C. The lowest processing losses were in samples marinated in KE and YO and cooked SV at 60 °C, with marinating time having no significant effect. The odor, flavor, tenderness and juiciness of meat marinated in BM was better than in KE and YO. Meat marinated and cooked SV at 60 °C was rated better by the panelists. Changes in proteins significantly affect the formation of meat texture, tenderness and juiciness, which confirms the correlations. This is also reflected in the sensory evaluation. During the process of marinating and cooking meat, protein degradation should be taken into account, which can be a good tool for shaping the sensory quality of cooked pork.

Abstract:Estrogen interacts with N‐methyl‐D‐aspartate (NMDA) receptors to regulate multiple aspects of morphological and functional plasticity. In hippocampus, estrogen increases both dendritic spine density and synapse number, and NMDA antagonists block these effects. Thus, estrogen‐mediated hippocampal plasticity may be of particular importance in the context of age‐related changes in endocrine status and cognitive performance. NR1 levels per synapse are increased in CA1 by estrogen in aged rats but not young rats, although no information is available on estrogen‐induced synaptic alterations in other NMDA receptor subunits that might impact function. Therefore, the present study was designed to investigate the effect of estrogen on the synaptic and subsynaptic distributions of the NMDA receptor subunits, NR2A and NR2B in CA1 pyramidal cells, within the context of aging. Our results demonstrated that the overall synaptic levels of NR2A and NR2B are similar in young and aged female rats, regardless of estrogen treatment. However, in the aged CA1, estrogen restores NR2B levels back to young levels in the lateral portions of the active synaptic zone. Thus, estrogen may impact the mobility of NMDA receptors across the synapse and, in the process, restore a more youthful synaptic profile. These findings have important implications for the mechanism of estrogen‐induced alterations in NMDA receptor‐mediated processes, particularly in the context of aging. J. Comp. In vitro digestion mimicking conditions in adults and elderly reveals digestive characteristics of pork from different cooking ways

Abstract:This study aimed to investigate the impact of three cooking ways (sous vide (SV), frying (FR) and roasting (RO)) on pork protein digestion characteristics under conditions simulating healthy adult (control, C) and elderly individuals with achlorhydria (EA). Changes in degree of hydrolysis (DH), SDS-PAGE profiles, zeta potential, particle size and secondary structure during digestion were evaluated. Our results revealed the EA condition markedly affected the protein digestion process of pork with different cooking ways. The DH values of SV (25.62%), FR (21.38%) and RO (19.40%) under the EA condition were significantly lower than those of under the control condition (38.32%, 33.00% and 30.86%, respectively). Moreover, differences were also observed among three cooking ways under the EA condition. For a given cooking way, the differences between control and EA conditions gradually diminished from the gastric to the intestinal phase. Under a certain digestion condition, SV maintained the highest degree of digestion throughout the process, particularly under the EA condition. Therefore, we conclude that pork cooked by sous vide is more recommendable for the elderly considering protein digestibility.

Abstract:The relationship between water-protein interactions and heat-induced protein denaturation in low temperature long time (LTLT) treated pork Longissimus dorsi was investigated by combining low-field NMR T₂ relaxometry with DSC measurements and measures of shrinkage of porcine Longissimus dorsi heated to 53 °C, 55 °C, 57 °C and 59 °C for either 3 or 20 h. Water within the myofibrils, measured by NMR T₂₁ relaxation times, was affected by both temperature and holding time during LTLT treatment between 53 °C and 59 °C. The changes in NMR T₂₁ relaxation times were associated with decreased fiber diameter and increased cooking loss, revealing a relationship between transverse shrinkage, water-protein interactions and cooking loss. DSC measurements revealed a concomitant decrease in ΔH(68 °C), which suggests impact of collagen denaturation on the retention of water within the meat during LTLT treatment. Furthermore, a decrease in ΔH(75 °C) suggested that prolonged cooking (20 h) resulted in actin denaturation leading to decreased T₂₁ relaxation times and higher cooking loss.

Mechanism underlying the tenderness evolution of stir-fried pork slices with heating rate revealed by infrared thermal imaging assistance

Abstract:This study aimed to explore the mechanism of cooking intensity on the tenderness of stir-fried pork slices from the perspective of the changes in temperature distribution. Infrared thermal imaging was used to monitor the distribution of temperature. Results showed that the high-level heat (HH) treatment could improve tenderness. When the center temperature increased to 100 °C, the shear force of samples from the low-level heat (LH) group increased by around 3-fold, and HH reduced this upward trend. This result was mainly attributed to the shorter heating time undergone by the HH-treated samples compared to the LH treatment, which resulted in less structural shrinkage and faster passing through the protein denaturation interval of the samples. These changes alleviated temperature fluctuations caused by water loss. This explanation could be confirmed by the results of T2 relaxation time and Fourier transform-infrared spectroscopy (FT-IR). However, the LH treatment caused a slower rise in oil temperature due to more moisture migration, which required the samples to undergo longer thermal denaturation, leading to a deterioration in tenderness. Moreover, histological analysis revealed that the greater integrity of endomysium in the HH group inhibited water loss and oil absorption, which contributed to obtain low-fat meat products with higher tenderness. This study provides support for the industrialization of traditional pork cuisines using oil as the heating medium.

Abstract:The effects of boiling, high-press steaming, roasting and microwaving on lipid oxidation and fatty acid profiles of traditional thoroughbred pork and lean pork were evaluated. All the cooking methods increased the peroxide value(POV) and 2-thiobarbituric acid(TBA) indexes, and statistical analysis displayed significant(P<0.05) differences,being the highest in roasted pork, followed by microwaved, boiled and roasted cooking. The results of GC revealed that the cooking methodshad considerable effects on the fattyacid compositions of traditional thoroughbred pork and lean pork(P<0.05). Thecontent of saturated fatty acids(SFA)decreased significantly after cooking(P<0.05), particularly after boiling,respectively: from 9.14 g/(100 g) to 4.85 g/(100 g) in lean pork and from 9.65 g/(100 g) to 3.49 g/(100 g) in traditional thoroughbred pork. Cooking treatments led to an decrease in unsaturated fatty acid, but increasedthe ratio of unsaturated fatty acids and saturated fatty acids(UFA/SFA). Studies have shown that cooking can not only improve the flavor of pork, but alsoincreasethe nutritional value.

Abstract:We explored whether ultrasound treatment affected digestibility of myofibrillar protein (MP) isolated from modified atmosphere packed (MAP, 70% N2 and 30% CO2) pork. MP digestibility under pepsin and pancreatin treatments decreased significantly with storage time. Ultrasound treatment increased the digestibility and produced a greater number of smaller peptides. However, the total peptide count and unique peptide counts were significantly reduced. Moreover, active sulfhydryl, total sulfhydryl, particle size, turbidity and surface hydrophobicity of MP increased with time, while protein solubility and ATPase activity decreased. Compared with the untreated samples, ultrasound treatment increased protein solubility, surface hydrophobicity, and active sulfhydryl content, but decreased total sulfhydryl content (except 10 d), particle sizes, turbidity and ATPase activity. Ultrasound treatment resulted in a decline in β-turn and α-helix contents. Therefore, ultrasound is conducive to the digestion. Additionally, structural and functional properties of protein in MAP were less stable than those in vacuum-packed pork reported before.

Abstract:A DIA-based quantitative proteomic strategy was used to investigate the effects of different cooking procedures (steaming and boiling) on pork meat quality. Results showed that steamed meats had higher redness, cohesion, springiness, but lower lightness, yellowness, shear force, hardness, chewiness and cooking loss than boiled meats. In total of 1608 proteins were identified and 103 proteins exhibited significant difference (fold change 1.5, P < 0.05). These DAPs mainly involved in protein structure, metabolic enzyme, protein turnover and oxidation stress. ALDOC, PVALB, PPP1R14C, AMPD1, CRYAB and SOD1 were validated as potential indicators of color variations in cooked meat. CFL1, COL1A1, COL3A1, RTN4, NRAP, NT5C3A, and SOD1 might be potential biomarker for texture changes of cooked meats. Moreover, these validated proteins exhibited significant (P < 0.05) correlation with cooking loss and could be serve as candidate predictors for cooking loss changes of meats in different cooking procedures.
Consumption of a garlic clove a day could be beneficial in preventing thrombosis.
