Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.

Abstract:Chitosan is a promising naturally derived polysaccharide to be used in hydrogel forms for pharmaceutical and biomedical applications. The multifunctional chitosan-based hydrogels have attractive properties such as the ability to encapsulate, carry, and release the drug, biocompatibility, biodegradability, and non-immunogenicity. In this review, the advanced functions of the chitosan-based hydrogels are summarized, with emphasis on fabrications and resultant properties reported in literature from the recent decade. The recent progress in the applications of drug delivery, tissue engineering, disease treatments, and biosensors are reviewed. Current challenges and future development direction of the chitosan-based hydrogels for pharmaceutical and biomedical applications are prospected.

Abstract:THE observations about to be described form a further part of a series of investigations into the metabolic activities over given periods. They differ from those already publishe%l in that the observation time is much longer-24 hours. Working with short periods, one of us (E. C. D.() demonstrated that there were more or less definite relations between the alveolar and blood C02 tension, gastro-intestinal secretion (2), and certain urinary constituents(3). These relations were altered by the effect of meals. The results of the previous investigations can be summarised briefly as follows: 1. During a period of starvation, the alveolar C02 tension remains more or less constant, whilst the urinary pH and A + N value do not show any definite change. 2. After a meal, the alveolar C02 tensions show the characteristic rise followed by a fall. The urinary changes were found to vary according to the meal. Thus the breakfast was followed by a definite alkaline tide, as shown by a rise in the pH and a fall in the A + N value. This alkaline tide was usually of short duration, and was followed by a prolonged acid tide, as shown by a fall in the urinary pH and a rise in the A + N values-both of which obscured any alkaline effect of the next meal. It was felt that these investigations were incomplete, since the observation times were never longer than 8 hours, and ouly the pH and A + N values were determined. The present investigation was undertaken, therefore, in order to clear up the difficulties of the previous observations. Variations in the composition of urine throughout 24 hours have been studied by many observers. Thus Bence Jones(4) was perhaps the first to notice the alkaline tide following a meal. Abstract:As a chitin- and glucosamine-derived polysaccharide, chitosan is biodegradable, biocompatible, and non-toxic. Among the latest high-value engineering materials, chitosan hydrogels own outstanding features such as biodegradability, biocompatibility, non-toxicity, swelling reversibility, flexible adaption to external triggers, and the capability of drug loading. Recently, applying chitosan hydrogels to facilitate human biological processes like inflammation has become an interesting topic demonstrated by many publications about their potential in wound dressing, tissue engineering, and treatment of cancer, Parkinson's disease, and gastric ulcer. This paper reviews the most recent procedures for synthesizing chitosan hydrogels as a smart anti-inflammatory drug delivery system. The characteristics of chitosan hydrogel, including pH sensitivity, temperature sensitivity, electric sensitivity, and magnetic strength, for flexible anti-inflammation, are all discussed to provide a comprehensive overview of anti-inflammatory chitosan hydrogel.

Abstract:Chitosan (CS) has become a focal point of extensive research in the pharmaceutical industry due to its remarkable biodegradability, biocompatibility and sustainability. Chitosan hydrogels (CS HGs) are characterized by their viscoelasticity, flexibility and softness. The polar surfaces exhibit properties that mitigate interfacial tension between the hydrogel and body fluids. The inherent compatibility of CS HGs with body tissues and fluids positions them as outstanding polymers for delivering therapeutic proteins, peptides, DNA, siRNA, and vaccines. Designed to release drugs through mechanisms such as swelling-based diffusion, bioerosion, and responsiveness to stimuli, CS HGs offer a versatile platform for drug delivery. CS HGs play pivotal roles in serving purposes such as prolonging the duration of preprogrammed drug delivery, enabling stimuli-responsive smart delivery to target sites, protecting encapsulated drugs within the mesh network from adverse environments, and facilitating mucoadhesion and penetration through cell membranes. This review comprehensively outlines various novel preparation methods of CS HGs, delving into the parameters influencing drug delivery system design, providing a rationale for CS HG utilization in drug delivery, and presenting diverse applications across the pharmaceutical landscape. In synthesizing these facets, the review seeks to contribute to a nuanced understanding of the multifaceted role that CS HGs play in advancing drug delivery methodologies.

Abstract:Chitosan, derived from the deacetylation of chitin, is an abundant natural biopolymer on earth. Chitosan and its derivatives have become promising biological materials because of their unique molecular structure and excellent biological activities. The reactive functional groups of chitosan such as the amino and hydroxyl groups play a crucial role in facilitating the synthesis of three-dimensional hydrogel. Chitosan-based hydrogels have been widely used in medical, pharmaceutical, and environmental fields for years. Nowadays, chitosan-based hydrogels have been found in a wide range of applications in the food industry such as food sensors, dye adsorbents and nutrient carriers. In this review, recently developed methods for the preparation of chitosan-based hydrogels were given, and the biological activities of chitosan-based hydrogels were systematically introduced. Additionally, the recent progress in food sensors, packaging, dye adsorbents, and nutrient carriers was discussed. Finally, the challenges and prospects for the future development of chitosan-based hydrogels were discussed.

Abstract:The second and most often utilized natural polymer is chitosan (CS), a naturally existing amino polysaccharide that is produced by deacetylating chitin. Numerous applications have been the subject of in-depth investigation due to its non-hazardous, biologically compatible, and biodegradable qualities. Chitosan's characteristics, such as mucoadhesion, improved permeability, controlled release of drugs, in situ gelation process, and antibacterial activity, depend on its amino (-NH2) and hydroxyl groups (-OH). This study examines the latest findings in chitosan research, including its characteristics, derivatives, preliminary research, toxic effects, pharmaceutical kinetics and chitosan nanoparticles (CS-NPs) based for non-parenteral delivery of drugs. Chitosan and its derivatives have a wide range of physical and chemical properties that make them highly promising for use in the medicinal and pharmaceutical industries. The characteristics and biological activities of chitosan and its derivative-based nanomaterials for the delivery of drugs, therapeutic gene transfer, delivery of vaccine, engineering tissues, evaluations, and other applications in medicine are highlighted in detail in the current review. Together with the techniques for binding medications to nanoparticles, the application of the nanoparticles was also dictated by their physical properties that were classified and specified. The most recent research investigations on delivery of drugs chitosan nanoparticle-based medication delivery methods applied topically, through the skin, and through the eyes were considered.

Abstract:Chitosan is one of the natural cationic polymers with unique properties such as non-toxicity, biodegradability, biocompatibility, environmentally friendly that has various biomedical applications. Chitosan-based hydrogels are hydrophilic polymer biomaterials with a three-dimensional cross-linked network structure. The methods of chitosan hydrogels fabricating are categorized into chemically and physically cross-linking between polymer chains. These hydrogels are employed for clinical applications because of their biological and physicochemical features. The most important applications of chitosan hydrogels have been considered in this review. Chitosan hydrogels are applicable in drug delivery due to their ability to control releasing of drugs through pH-responsive and temperature-responsive release approaches and the network structure that can carry drugs. Also, chitosan hydrogels are an excellent choice for wound healing because of their features such as antibacterial, providing moisture and heat in the wound area, biocompatibility, and so forth. Furthermore, swelling capacity, porosity, and mechanical properties make these hydrogels a good candidate as a scaffold for tissue formation. The other applications of chitosan hydrogels in biosensors, protein purification, and dye materials separation from wastewater have been considered in this review.Graphical abstract

Abstract:Polymers are used in drug delivery devices for drug encapsulation and release. Natural polymers often have advantages such as biocompatibility, biodegradability and biologically recognizable moieties which support cellular activities, compared to synthetic polymers. Chitosan, a naturally occurring polysaccharide obtained from chitin, has recently drawn attention because chitosan is non-toxic, biocompatible, biodegradable, stable and sterilizable. The additional qualities of chitosan are the release rate of drug, easy modification, cross-linking ability with other polymers, antimicrobial properties, gel forming ability, bioadhesion, immunostimulation, activation of macrophages and gas permeability. Here, I review applications of chitosan in drug delivery. The properties of chitin and chitosan, and the mechanisms of drug release are also presented.

Abstract:Chitosan, as a unique positively charged polysaccharide, has been one of the most popular biopolymers for development of drug delivery systems for various applications, due to its promising properties, including high biocompatibility, excellent biodegradability, low toxicity, as well as abundant availability and low production cost. Since last decade, increasing attention has been attracted by delivery systems fabricated from natural biopolymer-based polyelectrolyte complexes (PEC), formed by electrostatic interactions between two oppositely charged biopolymers. In order to tailor specific applications of chitosan-based PEC drug delivery systems, various forms have been developed in recent years, including nanoparticles, microparticles, beads, tablets, gels, as well as films and membranes. The present review focuses on the recent advances in drug delivery applications of chitosan-based PEC with other natural polysaccharides, including alginate, hyaluronic acid, pectin, carrageenan, xanthan gum, gellan gum, gum arabic, and carboxymethyl cellulose, etc. The fabrication techniques, characterizations, as well as in vitro and in vivo evaluations of each PEC delivery system are discussed in detail.

Abstract:Today, with the advances in technology and science, more advanced drug delivery formulations are required. One of these new systems is an intelligent hydrogel. These systems are affected by the environment or conditions that become a gel, stay in the circumstance for a certain period, and slowly release the drug. As an advantage, only a lower dose of the drug is required, and it provides less toxicity and minor damage to other tissues. Hydrogels are of different types, including temperature-sensitive, pH-sensitive, ion change-sensitive, and magnetic field-sensitive. In this study, we investigated a kind of temperature-sensitive smart hydrogel, which has a liquid form at room temperature and becomes gel with increasing temperature. Chitosan-β-glycerophosphate hydrogels have been researched and used in many studies. This study investigates the various factors that influence the gelation mechanism, such as gel formation rates, temperature, pH, time, and gel specificity. Hydrogels are used in many drug delivery systems and diseases, including nasal drug delivery, vaginal drug delivery, wound healing, peritoneal adhesion, ophthalmic drug delivery, tissue engineering, and peptide and protein delivery. Overall, the chitosan-β-glycerophosphate hydrogel is a suitable drug carrier for a wide range of drugs. It shows little toxicity to the body, is biodegradable, and is compatible with other organs. This system can be used in different conditions and different medication ways, such as oral, nasal, and injection.

Abstract:Chitosan is a biodegradable natural polymer with many advantages such as nontoxicity, biocompatibility, and biodegradability. It can be applied in many fields, especially in medicine. As a delivery carrier, it has great potential and cannot be compared with other polymers. Chitosan is extremely difficult to solubilize in water, but it can be solubilized in acidic solution. Its insolubility in water is a major limitation for its use in medical applications. Chitosan derivatives can be obtained by chemical modification using such techniques as acylation, alkylation, sulfation, hydroxylation, quaternization, esterification, graft copolymerization, and etherification. Modified chitosan has chemical properties superior to unmodified chitosan. For example, nanoparticles produced from chitosan derivatives can be used to deliver drugs due to their stability and biocompatibility. This review mainly focuses on the properties of chitosan, chitosan derivatives, and the origin of chitosan-based nanoparticles. In addition, applications of chitosan-based nanoparticles in drug delivery, vaccine delivery, antimicrobial applications, and callus and tissue regeneration are also presented. In summary, nanoparticles based on chitosan have great potential for research and development of new nano vaccines and nano drugs in the future.

Effect of Crosslinking Agents on Chitosan Hydrogel Carriers for Drug Loading and Release for Targeted Drug Delivery

Abstract:Numerous studies report on chitosan hydrogels in different forms, such as films, porous structures, nanoparticles, and microspheres, for biomedical applications; however, this study concentrates on their modifications with different crosslinking agents and observes their effects on drug loading and releasing capacities. Linear chitosan, along with chitosans crosslinked with two major crosslinkers, i.e., genipin and disulfide, are used to formulate three different hydrogel systems. The crosslinking process is heavily impacted by temperature and pH conditions. Three different drugs, i.e., thymoquinone, gefitinib, and erlotinib, are loaded to the hydrogels in de-ionized water solutions and released in phosphate-buffered solutions; thus, a total of nine combinations are studied and analyzed for their drug loading and releasing capabilities with ultraviolet–visible (UV–Vis) spectroscopy. This study finds that thymoquinone shows the lowest loading efficacy compared to the two other drugs in all three systems. Gefitinib shows stable loading and releasing regardless of crosslinking system, and the genipin-crosslinked system shows stable loading and releasing with all three drug molecules. These experimental results agree well with the findings of our previously published results conducted with molecular dynamics simulations.

Abstract:Smart stimuli-responsive materials can respond to different signals (pH, temperature, light, electricity, etc.), and they have become a hot research topic for drug delivery. As a polysaccharide polymer with excellent biocompatibility, chitosan can be obtained from diverse natural sources. Chitosan hydrogels with different stimuli-response capabilities are widely applied in the drug delivery field. This review highlights and discusses the research progress on chitosan hydrogels concerning their stimuli-responsive capabilities. The feature of various stimuli-responsive kinds of hydrogels is outlined, and their potential use of drug delivery is summarized. Furthermore, the questions and future development chances of stimuli-responsive chitosan hydrogels are analyzed by comparing the current published literature, and the directions for the intelligent development of chitosan hydrogels are discussed.

Abstract:Chitosan (CS), a biocompatible and biodegradable polymer produced from chitin, has attracted interest in drug delivery due to its unique properties and diverse applications. Over the past decade, chitosan-based devices have been developed for targeted and regulated drug delivery. This review discusses current advances in chitosan-based drug delivery, focusing on formulations, methods and applications. Films, hydrogels, microparticles and CS nanoparticles transport tiny molecules (drugs, or other chemical agents), proteins, peptides and nucleic acids, according to research. The mucoadhesive properties of CS improve the absorption of drugs on the mucous membranes of the gastrointestinal tract, respiratory tract and eyes. The study also highlights the theranostic potential of CH-based devices that combine therapeutic and diagnostic capabilities. The study also looks at the latest advances in customized and stimuli-responsive delivery of CH-based drugs. Despite tremendous progress, consistency of drug release and reduction in immunogenicity remain a concern. As research continues into innovative modifications, nanotechnologies and theranostic techniques to improve CH-based drug delivery systems, the future appears promising. This review examines current advances in chitosan-based drug delivery and their likely impact on personalised medicine and targeted therapy.

Abstract:Chitosan is one of the most used polysaccharides in the design of drug delivery strategies for administration of either biomacromolecules or low molecular weight drugs. For these purposes, it is frequently used as matrix forming material in both nano and micron-sized particles. In addition to its interesting physicochemical and biopharmaceutical properties, which include high mucoadhesion and a great capacity to produce drug delivery systems, ensuring the biocompatibility of the drug delivery vehicles is a highly relevant issue. Nevertheless, this subject is not addressed as frequently as desired and even though the application of chitosan carriers has been widely explored, the demonstration of systems biocompatibility is still in its infancy. In this review, addressing the biocompatibility of chitosan carriers with application in drug delivery is discussed and the methods used in vitro and in vivo, exploring the effect of different variables, are described. We further provide a discussion on the pros and cons of used methodologies, as well as on the difficulties arising from the absence of standardization of procedures.

Abstract:Controlled drug release systems have the characteristics of eliminating or reducing side effects and producing a therapeutic concentration of the drug that is stable in the body. The synthesis of hydrogels from natural polymers allows innovation in new materials that promote more effective, selective and safe therapies, in comparison with hydrogel systems based on synthetic polymers where their main limitation may be their biocompatibility and degradation. For this reason, in this work the synthesis of hydrogels of chitosan crosslinked with glutamic acid is proposed as the basis of new trends in smart materials for their potential use in the controlled release of drugs, the hydrogels were prepared from an amidation reaction between the amino groups of chitosan and the carboxyl groups of L-glutamic acid, using N-(3-Dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride (EDC) as activator of the carboxyl groups. The hydrogels obtained were characterized by SEM, FTIR, TGA techniques, swelling kinetics and antimicrobial activity. They exhibited excellent swelling capacity and good performance when exposed to different pH and temperature conditions. The bacterial inhibition percentages demonstrated the antimicrobial activity of chitosan hydrogels and the results obtained potentially favored their use as reservoirs for controlled drug release.

Abstract:In recent decades, drug delivery systems (DDSs) based on nanotechnology have been attracting substantial interest in the pharmaceutical field, especially those developed based on natural polymers such as chitosan, cellulose, starch, collagen, gelatin, alginate and elastin. Nanomaterials based on chitosan (CS) or chitosan derivatives are broadly investigated as promising nanocarriers due to their biodegradability, good biocompatibility, non-toxicity, low immunogenicity, great versatility and beneficial biological effects. CS, either alone or as composites, are suitable substrates in the fabrication of different types of products like hydrogels, membranes, beads, porous foams, nanoparticles, in-situ gel, microparticles, sponges and nanofibers/scaffolds. Currently, the CS based nanocarriers are intensely studied as controlled and targeted drug release systems for different drugs (anti-inflammatory, antibiotic, anticancer etc.) as well as for proteins/peptides, growth factors, vaccines, small DNA (DNAs) and short interfering RNA (siRNA). This review targets the latest biomedical approaches for CS based nanocarriers such as nanoparticles (NPs) nanofibers (NFs), nanogels (NGs) and chitosan coated liposomes (LPs) and their potential applications for medical and pharmaceutical fields. The advantages and challenges of reviewed CS based nanocarriers for different routes of administration (oral, transmucosal, pulmonary and transdermal) with reference to classical formulations are also emphasized.
Progress in preparation and properties of chitosan-based hydrogels

Abstract:Chitosan is a kind of natural polysaccharide biomass with the second highest content in nature after cellulose, which has good biological properties such as biocompatibility, biodegradability, hemostasis, mucosal adsorption, non-toxicity, and antibacterial properties. Therefore, hydrogels prepared from chitosan have the advantages of good hydrophilicity, unique three-dimensional network structure, and good biocompatibility, so they have received extensive attention and research in environmental testing, adsorption, medical materials, and catalytic supports. Compared with traditional polymer hydrogels, biomass chitosan-based hydrogels have advantages such as low toxicity, excellent biocompatibility, outstanding processability, and low cost. This paper reviews the preparation of various chitosan-based hydrogels using chitosan as raw material and their applications in the fields of medical materials, environmental detection, catalytic carriers, and adsorption. Some views and prospects are put forward for the future research and development of chitosan-based hydrogels, and it is believed that chitosan-based hydrogels will be able to obtain more valuable applications.
