Abstract:Every year, seafood waste produced globally contains about 10 million tons of wasted crab, shrimp and lobster shells, which are rich in chitin resources. The exploitation and utilization of chitin resources are of great significance to environmental protection, economic development and sustainable development. Lytic polysaccharide monooxygenases (LPMOs) can catalyze polysaccharides by oxidative breakage of glycosidic bonds and have catalytic activity for chitin and cellulose, so they play an important role in the transformation of refractory polysaccharides into biomass. Although there have been many studies related to LPMOs, the research related to lytic chitin monooxygenases (LCMs) is still very limited. The specific catalytic mechanism of LCMs has not been fully elucidated, which poses a challenge to their application in industrial biomass conversion. This review introduces the present situation of resource development and utilization in chitin, the origin and classification of different LCMs families, the structural characteristics of LCMs and the relationship between structure and function. The research results related to activity detection, screening, preparation and transformation of LCMs were summarized and discussed. Finally, the synergistic effect of LCMs and chitin enzyme on biomass degradation was reviewed, and the existing problems and future research directions were pointed out. This is the first review focusing on Chitin-Active LPMOs in recent years, intending to provide a reference for applying chitin degradation enzymes system in the industry.

Abstract:Cellulose, the main structural component of plant cell walls, is the most abundant renewable resource in nature, but it is extremely difficult to be degraded. Cellulosome is a multienzyme complex that can efficiently degrade cellulose and the degradation products can be used by some anaerobic microorganisms to produce ethanol. Recent research status in anaerobic degradation of cellulose for ethanol production by cellulosome-producing bacteria was reviewed in this paper. The latest achievements and research development in cellulosomes’ structure and function, designer cellulosomes, metabolic engineering and co-culture of cellulosome producing microorganisms were also summarized. It is expected that the development of new cellulose degrading microorganisms and relative technology will further cut the cost of cellulosic ethanol in the near future and make it more competitive with gasoline.

Abstract:Lignocellulosic biorefinery has long been recognized as a potential strategy for sustainable production of biofuels, materials and chemicals. These productions in many cases rely on pretreatment and enzymatic hydrolysis of lignocellulosic carbohydrates, whereas the low degradation efficiency and high enzyme dosage are challenging bottlenecks of economically industrial production. The complex recalcitrant nature of lignocellulose requires a well-matched enzyme cocktail to achieve complete and efficient degradation. Numerous studies have investigated the relationship between biomass components, structural properties and enzymes synergistic action in the past decades. This article aims to present a comprehensive review on this subject by introducing recent advances in substrate modification techniques, underlying mechanisms of enzyme synergism and governing factors in structure-activity interaction. In addition, newly developed modellings and multi-omic analysis techniques for preparing commercial enzyme preparations are introduced, revealing possible avenues for achieving economic viability of lignocellulose bioconversion.

Abstract:Lignocellulosic biomass represents perhaps the most abundant renewable resource with a potential to replace fossil‐based feedstock for sustainable energy, chemical and materials production. Among the three chief lignocellulosic biomass components (i.e. cellulose, hemicellulose and lignin), lignin is a macromolecule with an aromatic skeleton with a variety of functional groups (e.g. hydroxyl, methoxy, carbonyl, double bond) and carries a higher energy density. The unique structure makes lignin an intriguing substrate for energy, chemicals and materials productions. However, the high molecular weight and complex macromolecular structure have made lignin a challenging substrate to be transformed by many conversion methods. Microbial enzyme degradation and modification of lignin have been subjected to a significant amount research in the last a few decades. Yet so far little success has been demonstrated to merit the use of enzymatic technology for lignin transformation at a commercial scale. This paper provides an updated review of the development of lignin degrading/modifying enzymes with an emphasis on identifying the key barriers and challenges toward practical applications of microbial enzymes for lignin valorization with a hope to generate new insights and direction that can overcome these challenges.

Abstract:Lignin, a rigid polymer composed of phenolic subunits with high molecular weight and complex structure, ranks behind only cellulose in the contribution to the biomass of plants. Therefore, lignin can be used as a new environmentally friendly resource for the industrial production of a variety of polymers, dyes and adhesives. Since laccase was found to be able to degrade lignin, increasing attention had been paid to the valorization of lignin. Research has mainly focused on the identification of lignin-degrading enzymes, which play a key role in lignin biodegradation, and the potential application of lignin degradation products. In this review, we describe the source, catalytic specificity and enzyme reaction mechanism of the four classes of the lignin-degrading enzymes so far discovered. In addition, the major pathways of lignin biodegradation and the applications of the degradative products are also discussed. Lignin-degrading bacteria or enzymes can be used in combination with chemical pretreatment for the production of value-added chemicals from lignin, providing a promising strategy for lignin valorization.

Abstract:The field of enzymatic degradation of lignocellulose is actively growing and the recent updates of the last few years indicate that there is still much to learn. The growing number of protein sequences with unknown function in microbial genomes indicates that there is still much to learn on the mechanisms of lignocellulose degradation. In this review, a summary of the progress in the field is presented, including recent discoveries on the nature of the structural polysaccharides, new technologies for the discovery and functional annotation of gene sequences including omics technologies, and the novel lignocellulose-acting enzymes described. Novel enzymatic activities and enzyme families as well as accessory enzymes and their synergistic relationships regarding biomass breakdown are described. Moreover, it is shown that all the valuable knowledge of the enzymatic decomposition of plant biomass polymers can be employed towards the decomposition and upgrading of synthetic polymers, such as plastics.
One-step Solvent-free Mechanochemical Oxidation of Lignocellulosic Biomass to Cellulose Nanospheres by a Fe-complex

Abstract:The most abundant biopolymer in nature, lignocellulosic biomass, is often disregarded as agricultural waste, contributing to environmental pollution. Several reports have demonstrated their utilization by generating nanocellulosic materials using multistep harsh treatments involving H2SO4 and NaOH. However, these processes do not reduce the waste generated and release toxic gases into the environment. To address these pressing issues and effectively utilize our bioresources, we herein demonstrate a single-step, solvent-free method to oxidatively degrade lignocellulosic biomass such as corncob waste into completely water-dispersible carboxylate-functionalized cellulose nanospheres (CNS). This process uses an environmental-friendly catalyst (Et4N)2[Fe-(Ph,Me)bTAML] and sodium percarbonate as the oxidant. Our catalyst acts as a combined mimic of LPMO and lignin peroxidase, effectively breaking down the biomass without generating any toxic by-products. We confirmed the reaction mechanism using smaller repeating units like glucose and cellobiose, demonstrating a pathway similar to LPMO and unlike TEMPO-mediated oxidation. Finally, we extended our idea of using the CNS, due to its enhanced surface activity, as an O/W Pickering emulsion stabilizer and used it as a sustainable alternative to commercial detergents.

Abstract:Abstract Lignin, the most abundant renewable aromatic compound in nature, is an excellent feedstock for value-added bioproducts manufacturing; while the intrinsic heterogeneity and recalcitrance of which hindered the efficient lignin biorefinery and utilization. Compared with chemical processing, bioprocessing with microbial and enzymatic catalysis is a clean and efficient method for lignin depolymerization and conversion. Generally, lignin bioprocessing involves lignin decomposition to lignin-based aromatics via extracellular microbial enzymes and further converted to value-added bioproducts through microbial metabolism. In the review, the most recent advances in degradation and conversion of lignin to value-added bioproducts catalyzed by microbes and enzymes were summarized. The lignin-degrading microorganisms of white-rot fungi, brown-rot fungi, soft-rot fungi, and bacteria under aerobic and anaerobic conditions were comparatively analyzed. The catalytic metabolism of the microbial lignin-degrading enzymes of laccase, lignin peroxidase, manganese peroxidase, biphenyl bond cleavage enzyme, versatile peroxidase, and β-etherize was discussed. The microbial metabolic process of H-lignin, G-lignin, S-lignin based derivatives, protocatechuic acid, and catechol was reviewed. Lignin was depolymerized to lignin-derived aromatic compounds by the secreted enzymes of fungi and bacteria, and the aromatics were converted to value-added compounds through microbial catalysis and metabolic engineering. The review also proposes new insights for future work to overcome the recalcitrance of lignin and convert it to value-added bioproducts by microbial and enzymatic catalysis.

Lignocellulose Degradation in Bacteria and Fungi for Biomass Conversion

Abstract:Lignocellulose biomass is one of the most abundant resources for sustainable biofuels. However, scaling up the biomass-to-biofuels conversion process for widespread usage is still pending. Bottlenecks during the process of enzymatic hydrolysis are the high cost of enzymes and the labor-intensive need for substrate-dependent enzyme mixtures. Current research efforts are therefore targeted at searching for or engineering lignocellulolytic enzymes of high efficiency. One way is to engineer multi-enzyme complexes that mimic the bacterial cellulosomal system, known to increase degradation efficiency up to 50-fold when compared to freely-secreted enzymes. However, these designer cellulosomes are instable and less efficient than wild type cellulosomes. Fungi cellulosomes discovered in recent years have significant differences from bacterial counterparts and hold great potential for industrial applications, both as designer cellulosomes and as additions to the enzymatic repertoire. Up to date, they are only found in a few anaerobic fungi. In this review, we extensively compared the degradation mechanisms in bacteria and fungi, and highlighted the essential gaps in applying these mechanisms in industrial applications. To better understand cellulosomes in microorganisms, we examined their sequences in 66,252 bacterial species and 823 fungal species and identified several bacterial species that are potentially cellulosome-producing. These findings act as a valuable resource in the biomass community for further proteomic and genetic sequence analysis. We also collated the current strategies of bioengineering lignocellulose degradation to suggest concepts that could be favorable for industrial usage.

Abstract:The conversion of lignocellulosic biomass (LB) to sugar is an intricate process which is the costliest part of the biomass conversion process. Even though acid/enzyme catalysts are usually being used for LB hydrolysis, enzyme immobilization has been recognized as a potential strategy nowadays. The use of nanobiocatalysts increases hydrolytic efficiency and enzyme stability. Furthermore, biocatalyst/enzyme immobilization on magnetic nanoparticles enables easy recovery and reuse of enzymes. Hence, the exploitation of nanobiocatalysts for LB to biofuel conversion will aid in developing a lucrative and sustainable approach. With this perspective, the effects of nanobiocatalysts on LB to biofuel production were reviewed here. Several traits, such as switching the chemical processes using nanomaterials, enzyme immobilization on nanoparticles for higher reaction rates, recycling ability and toxicity effects on microbial cells, were highlighted in this review. Current developments and viability of nanobiocatalysts as a promising option for enhanced LB conversion into the biofuel process were also emphasized. Mostly, this would help in emerging eco-friendly, proficient, and cost-effective biofuel technology.

Abstract:Microbial degradation of lignocellulosic biomass (LCB) is of great human interest as it results in the production of numerous value-added products. 2G ethanol is an important lignocellulosics-based product that offers a long-term biotechnological solution to the depleting crude oil reserves without competing with food resources. The efficient degradation of LCB requires synergistic action of an array of microbial enzymes that include modular and non-modular glycosyl hydrolases (endoglucanase, exoglucanase, β-glucosidase, endoxylanase, β-xylosidase, α-arabinofuranosidase, α-glucuronidase, α-galactosidases and β-mannosidases), carbohydrate esterases (CE proteins) and other auxiliary enzymes (LPMOs, CDH and laccases). The genomics and proteomics studies have suggested a variety of culturable and non-culturable lignocellulolytic microorganisms inhabiting diverse ecological niches such as decaying plant materials, soil, compost piles, rumens, forest waste piles, wood processing plants, methanogenic sludge and surface of seashore. Among the rich microbial diversity, fungi are known for their ability to produce copious amounts of these lignocellulolytic enzymes. The wild-type fungal strains (with low specific activities) are subjected to several strain improvement strategies employing cyclic mutagenesis, recombinant technologies and other molecular techniques targeting regulatory elements to enhance their enzyme titres, specific activities and catalytic/hydrolytic efficiencies. The cellulase-/hemicellulase-rich preparations produced by growing developed strains on inexpensive agro-residues as carbon sources (under SSF and/or SmF) find potential applications in biorefineries, paper and pulp industry, animal feed, food and beverages industry and textile and detergent industry, making global business of USD 800 million per year.

Abstract:Lignocellulosic biomass is the earth's most abundant renewable feedstock alternative that comprises of cellulose, hemi-cellulose and lignin. The synergistic action of cellulolytic/xylanolytic enzymes produced by lignocellulolytic micro-organisms such as bacteria, algae and fungi are capable of robust cellulosic biomass deconstruction. Most of the microorganisms dwelling in extreme environmental habitats such as rumen environment, hot/cold springs, deep ocean trenches, acidic/alkaline pH environment have been considered as an attractive producers of hemi/cellulolytic lignocellulolytic and other biotechnological enzymes with enhanced bio-chemical properties essential for industrial bioconversion processes. However, the potential microbial sources of cellulolytic enzymes and the underlying mechanism to achieve this is not fully elucidated. In this review article, first we detail the composition of lignocellulosic biomass. Next, we describe the structure and functions of divergent hydrolytic enzymes (cellulolytic and xylanolytic enzymes) involved in cellulosic biomass degradation. Third, we analyze, outline and unveil the prospective source of microbes encoding exceptionally diverse set of biotechnologically relevant cellulolytic enzymes which are critical to answer the specific ecological question of by whom, where and how cellulosic biomass is degraded in the environment. Finally, this review article features the snapshot about the future developments and perspectives on microbial enzymes, high-throughput techniques and molecular tools that could be exploited to derive those enzymes from the potential sources.

Abstract:Along with long-term evolution, the plant cell wall generates lignocellulose and other anti-degradation barriers to confront hydrolysis by fungi. Lytic polysaccharide monooxygenase (LPMO) is a newly defined oxidase in lignocellulosic degradation systems that significantly fuels hydrolysis. LPMO accepts electrons from wide sources, such as cellobiose dehydrogenase (CDH), glucose-methanol-choline (GMC) oxidoreductases, and small phenols. In addition, the extracellular cometabolic network formed by cosubstrates improves the degradation efficiency, forming a stable and efficient lignocellulose degradation system. In recent years, using structural proteomics to explore the internal structure and the complex redox system of LPMOs has become a research hotspot. In this review, the diversity of LPMOs, catalytic domains, carbohydrate binding modules, direct electron transfer with CDH, cosubstrates, and degradation networks of LPMOs are explored, which can provide a systematic reference for the application of lignocellulosic degradation systems in industrial approaches.

Abstract:Lignin is an important commercially produced polymeric material. It is used extensively in both industrial and agricultural activities. Recently, it has drawn much attention from the scientific community. It is abundantly present in nature and has significant application in the production of biodegradable materials. Its wide usage includes drug delivery, polymers and several forms of emerging lignin nanoparticles. The synthesis of lignin nanoparticles is carried out in a controlled manner. The traditional manufacturing techniques are costly and often toxic and hazardous to the environment. This review article highlights simple, safe, climate-friendly and ecological approaches to the synthesis of lignin nanoparticles. The changeable, complex structure and recalcitrant nature of lignin makes it challenging to degrade. Researchers have discovered a small number of microorganisms that have developed enzymatic and non-enzymatic metabolic pathways to use lignin as a carbon source. These microbes show promising potential for the biodegradation of lignin. The degradation pathways of these microbes are also described, which makes the study of biological synthesis much easier. However, surface modification of lignin nanoparticles is something that is yet to be explored. This review elucidates the recent advances in the biodegradation of lignin in the ecological system. It includes the current approaches, methods for modification, new applications and research for the synthesis of lignin and lignin nanoparticles. Additionally, the intricacy of lignin’s structure, along with its chemical nature, is well-described. This article will help increase the understanding of the utilization of lignin as an economical and alternative-resource material. It will also aid in the minimization of solid waste arising from lignin.

Abstract:Cellulose is a very abundant polymer that is found in nature. Cellulose has been used as a raw material for production of biofuels for many years. However, there are multiple processing steps that are required so that cellulose can be used as a raw material for biofuel production. One of the most important steps is the breakdown of cellulose into intermediate sugars which can then be a viable substrate for biofuel production. Cellulases are enzymes which play a role in the catalysis of the breakdown of cellulose into glucose. Nanomaterials and micromaterials have been gaining a lot of attention over the past few years for its potential in immobilizing enzymes for industrial procedures. Immobilization of enzymes on these nanomaterials has been observed to be of great value due to the improvement in thermal stability, pH stability, regenerative capacity, increase in activity and the reusability of enzymes. Similarly, there have been multiple reports of cellulase enzymes being immobilized on various nanoparticles. The immobilization of these cellulase enzymes have resulted in very efficient processing and provide a great and economic solution for the processing of cellulose for biofuel production. Hence in this paper, we review and discuss the various advantages and disadvantages of enzymes on various available nanomaterials.

Abstract:For many industries, the committed step during the treatment of lignocellulosic materials is the elimination of lignin. This recalcitrant material is commonly removed by many physical/chemical procedures that produce hazardous effluents that poison the environment, and usually the treatment of these effluents are cost-expensive. An eco-friendly alternative to the use of traditional treatments is the degradation of lignocellulosic materials using microorganisms or enzymes, mainly by the use of lignin-degrading enzymes. They are classified as lignin-modifying enzymes (ligninolytic enzymes that catalyze an oxidative cleavage of lignin) and lignin-degrading auxiliary enzymes. The lignin-modifying enzymes include phenol oxidases (such as laccases) and heme-peroxidases (lignin-peroxidases, manganese-peroxidases, versatile-peroxidases and, dye-decolorizing peroxidases), both classes of enzymes are members of the oxidase super-family. These enzymes have biochemical features that point them as excellent tools for being used in several industrial processes. They show a great versatility and can oxidize a wide range of organic and inorganic substrates, such as vanillyl alcohol, catechol, acetosyringone, syringic acid, and guaiacol, several dyes, phenolic and non-phenolic lignin model compounds. Their prospective applications include pulp and paper making, bio-refinery (biofuel), bioremediation and, textile, pharmaceutical and dermatological industries, among others. In this review, we summarize some of their biochemical features and potential industrial applications.

Abstract:The bio-enzyme degradation technology is a promising approach to sustainably remove pollution in the water and laccase is one of the most widely used enzymes in this area. Nevertheless, the further industrial application of laccase is limited by low stability, short service, low reusability and high price. The immobilization technology can significantly improve the stability and reusability of enzymes and thus promoting their industrial applications. Nanocomposite materials have been developed and applied in the efficient immobilization of laccase due to their superior physical, chemical, and biological performance. This paper presents a comprehensive review of various nanocomposite immobilization methods for laccase and the consequent changes in enzymatic properties post-immobilization. Additionally, a comprehensive analysis is conducted on the factors that impact laccase immobilization and its water removal efficiency. Furthermore, this review examines the effectiveness of common contaminants' removal mechanisms while summarizing and discussing issues related to laccase immobilization on nanocomposite carriers. This review aims to provide valuable guidance for enhancing laccase immobilization efficiency and enzymatic water pollutant removal.

Abstract:Wood‐rotting fungi are the sole organisms in nature able to degrade the lignin polymer making the polysaccharide components of lignocellulose fully accessible. This process has been investigated for decades as a model for biotechnological application in the pulp and paper industry, animal feeding, and ethanol production. In the current lignocellulose biorefinery concept, ligninolytic fungi and the oxidoreductases (laccases and peroxidases) secreted by these fungi constitute powerful biotechnological tools for the complete utilization of plant biomass. The evolution of molecular biology, which brings into play specifically designed biological systems and on‐demand enzymes, together with the technological advances in processing of plant biomass, smoothes the way for a sustainable conversion of renewable feedstocks to new added‐value products, with lower energy costs and less environmental impact. The present study reviews some of the main achievements attained by our group in the field of lignin biodegradation that have contributed to: (i) better understanding of the mechanisms by which fungi delignify the lignocellulosic materials; and (ii) assessing the applicability of these ligninolytic systems to increase the efficiency of some industrial processes and to develop new means for sustainable and environmentally sound production of chemicals, materials, and fuels. © 2014 Society of Chemical Industry and John Wiley & Sons, Ltd

Abstract:Converting biomass to produce renewable chemicals is one of the significant ways to realize the development of green chemistry and sustainable chemical industry. As the main component of lignocellulosic biomass, lignin is the most abundant natural polyphenol. Its unique phenolic properties and high carbon content make it potentially exploited. Biomass lignin depolymerization can be divided into thermochemical, chemical catalysis, electrocatalysis, and biological depolymerization methods. In this review, the catalytic reaction systems involved in different methods of lignin depolymerization were extensively described from various aspects, and the degradation products and reaction mechanisms were discussed and analyzed. The effects of novel green-deep eutectic solvents on biomass lignin depolymerization were reviewed in particular. Each method of biomass lignin depolymerization has its own advantages and disadvantages. The different sources of biomass lignin should be selected objectively according to its compositional structure, so as to achieve efficient biomass lignin depolymerization. Lignin-enzyme interaction: A roadblock for efficient enzymatic hydrolysis of lignocellulosics

Abstract:Efficiently producing second-generation biofuels from biomass is of strategic significance and meets sustainability targets, but it remains a long-term challenge due to the existence of biomass recalcitrance. Lignin contributes significantly to biomass recalcitrance by physically limiting the access of enzymes to carbohydrates, and this could be partially overcome by applying a pretreatment step to directly target lignin. However, lignin typically cannot be completely removed, and its structure is also significantly altered during the pretreatment. As a result, lignin residue in the pretreated materials still significantly hindered a complete conversion of carbohydrate to its monosugars by interacting with cellulase enzymes. The non-productive adsorption driven by hydrophobic, electrostatic, and/or hydrogen bonding interactions is widely considered as the major mechanism of action governing the unfavored lignin-enzyme interaction. One could argue this type of interaction between lignin residue and the activated enzymes is the major roadblock for efficient enzymatic hydrolysis of pretreated lignocellulosics. To alleviate the negative effects of lignin on enzyme performance, a deep understanding of lignin structural transformation upon different types of pretreatments as well as how and where does lignin bind to enzymes are prerequisites. In the last decade, the progress toward a fundamental understanding of lignin-enzyme interaction, structural characterization of lignin during pretreatment and/or conformation change of enzyme during hydrolysis is resulting in advances in the development of methodologies to mitigate the negative effect of lignin. Here in this review, the lignin structural transformation upon different types of pretreatments and the inhibition mechanism of lignin in the bioconversion of lignocellulose to bioethanol are summarized. Some technologies to minimize the adverse impact of lignin on the enzymatic hydrolysis, including chemical modification of lignin, adding blocking additives, and post-treatment to remove lignin were also introduced. The production of liquid biofuels from lignocellulosic biomass has shown great environmental benefits such as reducing greenhouse gas emissions and mitigate climate change. By addressing the root causes of lignin-enzyme interaction and how to retard this interaction, it is our hope that this comprehensive review will pave the way for significantly reducing the high cost associated with the enzymatic hydrolysis process, and ultimately achieving a cost-effective and sustainable biorefinery system.
