Abstract:Objective To investigate the role and possible mechanism of caveolin-1 (CAV1) in the forming of cholesterol gallstone in mice fed with lithogenic diet.Methods Cholesterol gallstone susceptible C57BL/6 mice were study objects.The mice of control group (n=6) and experiment group (n=6) were fed with normal diet and lithogenic diet for four weeks respectively.The condition of cholesterol gallstone forming,changes of serum lipid and bile composition were measured,and the expressions of CAV1 and scavenger receptor classB member Ⅰ (SR-BⅠ) at mRNA and protein level in the liver and gallbladder were detected by realtime-polymerase chain reaction and Western blot,respectively.The t test was performed for mean comparsion between the two groups.Results The incidence rate of gallstone in experimental group was 100％ after fed with lithogenic diet for four weeks,the lipid level significantly increased,and the proportion of cholesterol in bile raised and bile salt decreased.Compared with those of control group,the expressions of CAV1 at mRNA and protein level in the liver and gallbladder tissues siginificantly decreased (in liver tissue,mRNA 0.53 ± 0.13 vs 1.00 ± 0.32,t =3.330,protein level 0.39 ± 0.07vs 0.92±0.06,t=10.280; in gallbladder tissue,mRNA 0.40±0.22 vs 1.00±0.22,t=3.823,protein level 1.04±0.07 vs 1.34 ± 0.04,t =6.367,all P＜0.01).There was no significant difference in the relative expression of SR-BⅠ at mRNA and protein level in the liver and gallbladder tissues between the mice of experiment group and control group.Conclusion The changes of CAV 1 expression at mRNA and protein level in liver and gallbladder tissues may affect lipids metabolism and cholesterol transportation in liver and gallbladder tissues of experiment mice,which might play an important role in the formation of cholesterol gallstone.
Laparoscopic Hepatectomy for Hepatolithiasis: a Feasibility and Safety Study in 29 Patients

Abstract:Background: Hepatolithiasis is a prevalent disease in Southeast Asia. Heaptectomy was considered the best treatment for majority of cases. Laparoscopic hepatectomy is a new procedure for liver lesions that uses a minimal invasive approach. The aim of this study was to evaluate the feasibility and safety of laparoscopic hepatectomy for hepatolithiasis by comparing it with open hepatectomy.Methods: From November 2002 to March 2006 a total of 30 consecutive patients underwent laparoscopic hepatectomy for hepatolithiasis in Sir Run Run Shaw Hosptial. Twenty-nine were included in this study (a converted case was excluded) and called the laparoscopic hepatectomy group (LH). During the same period 22 patients with hepatiolithiasis who met the inclusion criteria for laparoscopic hepatectomy were selected for open hepatectomy and called the open group (OH). All operations were performed by the authors. There was no significant difference in preoperative data between the two groups. Data were statistically compared.Results: Compared with open hepatectomy, those who underwent laparoscopic hepatectomy had a shorter postoperative hospital stay and fasting time, a lower postoperative serum aminotransferase level, and a higher postoperative serum albumin level. Stone clearance rate (intermediate rate, 89.7% vs. 86.4%; final rate, 100% vs. 96.5%), stone recurrence rate (0% vs. 4.5%), operating time, and intraoperative blood loss were similar for the two groups. Six complications occurred, two (6.8%) in LH and four (18.2%) in OH. There was no perioperative mortality in either group.Conclusion: Laparoscopic hepatectomy for hepatolithiasis is feasible and safe in selected patients.
Construction of mouse model of liver implanted tumor after hepatectomy

Abstract:ObjectiveTo explore the construction of mouse model of liver implanted tumor after hepatectomy by implantation of liver tumor tissue.MethodsH22 hepatocellular carcinoma (HCC) cells of mice were implanted to the scapula of 2 BALB/c (nu/nu) nude mice and tumors developed in 2 weeks. Eighteen nude mice were divided into three groups according to random number table method: non-hepatectomy group (group A), 30% hepatectomy group (group B) and 70% hepatectomy group (group C) with 6 mice in each group. The hepatectomy model of nude mice was established. The left lateral lobe and middle lobe were dissected for the nude mice in group A. The tissues of left lateral lobe were ligated and resected in group B. And the tissues of left lateralobe and middle lobe were ligated and resected in group C. The tumor tissues incised from the scapula were implanted to the livers of nude mice in three groups. The tumor formation rate and the volume of implanted tumors of three groups were compared. The tumor formation rate was compared using Fisher's exact probability test, and the volume of implanted tumors was compared using Kruskal-Wallis test and Bonferroni method.ResultsThe tumor formation rate of group A, B and C after 2 weeks was respectively 6/6, 6/6 and 5/6, and no signiifcant difference was observed (P=1.000). The median volume of implanted tumors in group A, B and C was respectively 19(6-33), 22(8-44) and 150(58-186) mm3. The median volume of implanted tumors in group C was signiifcantly larger than those in group A and B (Z=9.03, 7.12;P<0.0167). The implanted tumors in three groups were all conifrmed as HCC and with different degrees of necrosis and inlfammatory cell inifltration by histopathological examination.ConclusionsImplantation of liver tumor tissue can be used to construct an ideal mouse model of liver implanted tumor after major hepatectomy with the advantages of short experimental period, high tumor formation rate and good stability.
Effects of Ursodeoxycholic Acid on Intrahepatic Cholestasis in Rats

Abstract:Objective To investigate the effects of treatment with ursodeoxycholic acid (UDCA) on intrahepatic cholestasis in rats, and to explore its mechanism.Method Rats suffering from intrahepatic cholestasis were treated with UDCA. Their serum alanine transaminase (ALT), alkaline phosphatase (ALP), total bilirubin (TBIL), direct bilirubin (DBIL), γ-glutamyl transpeptidase (γ-GT), total cholesterol (TCH), bile flow, total bile acid excretion, total Na+ and TCH of bile were measured before and after treatment. In addition, the changes of liver tissue under microscrope were observed and recorded.Results Compared with the control group, serum ALT, ALP, TBIL, DBIL, γ-GT and TCH of rats in the treatment group decreased, while bile flow, total bile acid excretion, total Na+ and TCH decreased significantly. Degeneration of hepatocytes, infiltration of inflamed cells and proliferation of small bile ducts in the treatment group were improved under microscope.Conclusion UDCA may have therapeutic effects on cholestatic hepatitis. The mechanism may involve in its hydrophilicity, choleretic effect and immune modulation.
A novel in vivo model of ureteral fibrosis induced by calcium oxalate beads in C57BL/6J mice

Abstract:The global incidence of ureteroliths in humans is increasing, particularly posing a problem in developed countries. The most common stone type is calcium oxalate, which is associated with a high recurrence rate. In veterinary medicine, stones are the most common cause of ureteral obstruction in cats, accounting for 72–87% of cases. In cats, stones cause irreversible ureteral damage, necessitating stone treatment as well as ureteral therapy. However, the mechanisms underlying the ureteral damage caused by stones remain unclear. Therefore, this study aimed to create a mouse model suitable for studying the ureteral fibrosis caused by oxalate stones by artificially embedding calcium oxalate in the ureter. Pathological tissue analysis was used to compare normal ureters without ligation and ureters with sham or oxalate bead implantation. The ureters of the sham and oxalate bead groups showed granulation tissue formation, transitional epithelium exfoliation, and densely packed connective tissue in the proprietary and muscle layer regions. Particularly in the oxalate bead group, infiltration of degenerated neutrophils, presence of foreign body giant cells, and hyperplasia of the transitional epithelium were observed. The proportion of fibrosis was higher in the oxalate group than in the sham group. Overall, this mouse model created using oxalate bead implantation has the potential to efficiently induce ureteral obstruction. This mouse model is expected to be used for elucidating the molecular mechanisms of ureteral fibrosis and evaluating therapeutic drugs in future.

[Establishment Nephrolithiasis Rat Model Induced by Nanobacteria and Analysis of Stone Formation].

Abstract:OBJECTIVE:To establish nephrolithiasis rat model induced by nanobacteria and to determine the relationship of nanobacteria with nephrolithiasis.METHODS:Thirty adult male SD rats were randomized into 3 groups (n=10 each): group A, given an intravenous injection of nanobacteria; group B, given an intravenous injection of nanobacteria and taken tetracycline; group C, as normal control. Eight weeks later, all rats were sacrificed and the kidneys were examined for pathology and the 24-h urine N-acetyl-beta-D-aminoglycoside enzyme (NAG) and lactate dehydrogenase (LDH) were detected.RESULTS:Histopathological studies revealed nanobacteria induced renal tubular crystallization, which was significantly different among the 3 groups (P=0.033). The crystallization was higher in group A than that in group B. The urinary NAG and LDH excretion were significantly different among the 3 groups (P<0.001).CONCLUSION:Nanobacteria may be involved in the pathogenesis of nephrolithiasis. Tetracycline may inhibit stone formation.
A Study of Minimally Invasive Treatment for Hepatolithiasis

Abstract:ObjectiveTo investigate the feasibility and therapeutic effect of minimally invasive treatment for hepatolithiasis. MethodsAn investigation was conducted in 15 patients with hepatolithiasis who underwent total laparoscopic treatment from May 2006 to November 2012. The procedures included simple laparoscopic hepatectomy, simple laparoscopic bile duct exploration, and Oddi sphincter-preserved subcutaneous tunnel and hepatocholangioplasty with the use of gallbladder or free jejunum. ResultsAll cases completed the operation, with no perioperative mortality, and postoperative residual stones were found in 26% of all cases (4/15). The follow-up rate was 100%; the mean follow-up was 29 months (range, 2-80 months). There was one case of postoperative anastomotic stenosis, and no recurrence of hepatolithiasis was seen. ConclusionMinimally invasive treatment for hepatolithiasis can be achieved if the operative indications are fully understood, the procedures are carefully performed, and the choledochoscope and subcutaneous tunnel are properly used.
A Modified Rat Model for Infection-Induced Stone Formation

Abstract:OBJECTIVE:To employ a newly modified rat model for infection-induced bladder stone formation.METHODS:24 adult male Sprague Dawley rats were divided into 3 groups, model group (n=12), sham operation group (n=8) and control group (n=4). The surgical procedures were performed aseptically under anesthesia (25% Ultane 1.0 g/kg). The bladder in model group was exposed through a short lower midline abdominal incision, the puncture needle (G18) with guideline was inserted aseptically into bladder, a metal wire, which have been contaminated by the Proteus mirabilis, was put into the puncture canal, then implanted into the bladder lumen through the guideline. In the sham operated group the puncture needle (G18) with guideline was inserted into bladder without metal wire implanted into the bladder. There was no any operation in control group. The rats were sacrificed by excessive anesthesia at 21 days post challenge. The bladder were removed aseptically and inspected for evidence of urolithiasis.RESULTS:On Day 2 after surgery, two rats died in model group, no rats died in other groups. Twenty-one days after surgery, all of rats in model group developed various-sized bladder stones. There was no stone formation in sham operation group and control group. All stones were verified by infrared spectroscopy and optical crystallography. These stone were struvite stone.CONCLUSION:This model has a less trauma, faster recovery and excellent stone formation so that it may be used for the study of infection stone.

Abstract:Bile contains four calcium anions which are readily precipitated by calcium: bilirubinate, "palmitate", carbonate and phosphate. Precipitation of insoluble calcium salts is common in calculous biliary disease. All animal models of calcium cholelithiasis probably share a common pathogenesis--an increased concentration of a calcium-sensitive anion. In calcium bilirubinate cholelithiasis, an increased concentration of bilirubinate occurs. In mice, this occurs in animals having a genetic hemolytic anemia; in rats, it may be induced by an essential fatty acid in combination with diet modification; in dogs, by a low-taurine, low-protein diet; and in prairie dogs, by ileal resection which probably increases the bilirubinate/bile acid ratio in bile. In the two models of calcium-bile acid cholelithiasis, an increased biliary concentration of a calcium-sensitive glycine conjugated bile acid is induced by administration of a suitable precursor. In the rabbit, gallstones composed mostly of calcium allodeoxycholyl glycine are induced by the feeding of 5 beta-cholestan-3 beta-ol. In the taurine-deficient rat, the feeding of lithocholate leads to formation of gallstones which contain a mixture of calcium lithocholyl glycine and 3 alpha, 6 beta-dihydroxy cholanoyl glycine. These animal models may be of value in testing agents designed to lower calcium activity in bile.

Abstract:Objective To build liver fibrosis models using uPA knock-out(uPA-/-) mice and observe the pathologic changes.Methods Adult male BL/6J WT mice and uPA knock-out(uPA-/-) mice were divided into four groups with 10 mice in each group: control groups(WT,uPA-/-) and liver fibrosis model groups(WT,uPA-/-).Mice were intraperitoneally injected 0.15 ml 10% CCl4(or olive oil as control) twice per week for 6 weeks.Animal body weight,liver weight,spleen weight and kidney weight were measured,and organ coefficients were calculated.General pathology of livers was observed at sacrifice,histopathology was performed using HE staining and Masson staining,and electron microscopic exam was performed to observe the ultrastructural changes.Results Liver weight and liver coefficient of uPA-/-model mice increased significantly compared with WT model mice(P0.05).The enlargement of the liver,pale gray color,and apparent granular nodules were observed in uPA-/-model mice.HE staining,Masson staining and electron microscopy all showed in consistency that liver cells were severely damaged,extensive collagen was deposited and false lobules were formed in uPA-/-model mice.Conclusion uPA-/-model mice shows more severe liver fibrosis compared with WT mice.uPA-/-mice are susceptible animal strain to establish liver fibrosis model.
Establishment of Rat Model of Ceftriaxone Associated Urolithiasis

Abstract:Objective To establish a rat model of ceftriaxone associated urolithiasis and investigate the possible mechanism of its formation.Methods Thirty-two male Wistar rats aged 8 weeks were randomly divided to four groups:group A served as control group;groups B,C and D received ceftriaxone sodium for 10 days with dosage of 100,150 and 200 mg/kg,respectively.The 24-h urine was gathered before intraperitoneal injection was conducted and before the end of experiment for the determination of oxalate and biochemical analysis.All rats were sacrificed with over-dose of carbon dioxide.The blood was collected for biochemical analysis.Kidneys were collected to observe the urinary stone formation and for Von Kossa staining.All the data were processed with the statistical software of SPSS 16.0.Results There were no visible calculi in these four groups,however,Von Kossa stains were positive in the three experimental groups,with the incidence of 50.0％ (4/8) in group B,62.5％ (5/8) in group C and 62.5％ (5/8) in group D.The incidence of massive calcium crystal deposits was 12.5％ (1/8) in group C and 12.5％ (1/8) in group D.No significant differences were observed in the incidences of calcium crystal deposits in groups with different dosages.Both the 24-h urinary excretion of calcium and uCa/Cr ratio in the 150 mg/kg group were increased markedly as compared with the control group.Conclusion Ceftriaxone could induce the formation of crystals in the kidney of rats.Ceftriaxone has the potential to significantly increase urinary excretion of calcium,which may be responsible for ceftriaxone associated urolithiasis.

Abstract:Objective To establish a mouse model of calcium oxalate urolithiasis.Methods experimental animals were C57BL/6 mice.All the mice accepted intraabdominal injection treatment,using glycolate 80 mg/kg or 120 mg/kg per day,glyoxylate 80 mg/kg or 120 mg/kg per day,sodium oxalate 140 mg/kg or 210 mg/kg per day.The survival rate and weight were measured.The stone formation were evaluated by Pizzolato' s staining.Results Glycolate 80 mg/kg per day and glyoxylate 80 mg/kg per day intraabdominal injection exerted minor influence on mice survive.Glyoxylate 80 mg/kg per day intraabdominal was better than glycolate.Glycolate intraabdominal injection couldn' t induce stone formation in kidney,while glyoxylate and sodium oxalate could form crystals in the mice kidneys.The oxalate crystals increased in the first 6 days and remained on day 9.Then the crystals decreased on day 9-15.Conclusion Glyoxylate 80 mg/kg per day intraabdominal injection is the best stone-formation reagent.The calcium oxalate crystals in mouse has a stone-forming phase and a stone-eliminating phase.
Untargeted and targeted metabolomics reveal bile acid profile changes in rats with ethylene glycol-induced calcium oxalate nephrolithiasis

Abstract:Calcium oxalate (CaOx) nephrolithiasis is a prevalent disorder linked to metabolism. Examining metabolic alterations could potentially give an initial understanding of the origins of CaOx nephrolithiasis. This study aims to determine gut metabolic biomarkers differentiating CaOx nephrolithiasis utilizing untargeted and targeted metabolomics. CaOx nephrolithiasis model rats were built by 1% ethylene glycol administration. Histologic staining and renal function measurement revealed the presence of crystals in the lumen of the renal tubules, the renal injury and interstitial fibrosis in CaOx rats, demonstrating that the models of CaOx were established successfully. Hematoxylin & eosin (H&E) staining showed that CaOx group had inflammation and damage in the ileal tissue. Immunofluorescence and PCR results displayed that the tight junction proteins, ZO-1 and Occludin levels were decreased in the ileal tissues of the CaOx group. The untargeted metabolomic analysis revealed that 269 gut metabolites were differentially expressed between the CaOx group and the control group. Meanwhile, bile secretion, the main metabolic pathway in CaOx nephrolithiasis, was identified. Following, five significant bile acid metabolites were selected utilizing the targeted bile acid metabolomics, including Hyodeoxycholic acid (HDCA), Glycohyodeoxycholic acid (GHDCA), Nor-Deoxycholic Acid, omega-muricholic acid, and Taurolithocholic acid. Among these metabolites, HDCA and GHDCA presented the highest predictive accuracy with AUC = 1 to distinguish the CaOx group from the control group. As a result of network pharmacology, target genes of HDCA and GHDCA in CaOx nephrolithiasis were enriched in oxidative stress and apoptosis pathways. Conclusively, our study provides insight into bile acids metabolic changes related to CaOx nephrolithiasis. Although alterations in biochemical pathways indicate a complex pathology in CaOx rats, bile acid changes may serve as biomarkers of CaOx nephrolithiasis.

Abstract:Objective. Develop an experimental model of liver cirrhosis and describe pathomorphological changes in organs and tissues of laboratory animals. Materials and methods. Liver cirrhosis was simulated on 11 sexually mature Wistar male white rats. The control group consisted of 12 healthy laboratory animals. Acute toxic liver injury resulting in cirrhosis was induced by intraperitoneal injection of 50% tetrachloromethane (CCl 4 ) solution on olive oil on the first day of the experiment in the dosage of 0.1 ml CCl 4 + 0.4 ml olive oil per 100 g of the animal body weight, on the second day of the experiment 0.3 ml CCl 4 + 0.2 ml olive oil per 100 g of the animal body weight. To synergize and potentiate the hepatotoxic effect of CCl 4 , animals had 10% ethanol solution freely available daily. The duration of the experiment was 65 days. Clinical and laboratory parameters were evaluated, histological evaluation of the preparations was performed. The obtained data were compared with the control group of animals. Results. Reproducibility of the model was 82% (9 animals). The indicator of lymphocyte blabbing index showed the development of liver cirrhosis in animals of the experimental group. Comprehensive morphological confirmation of the developed liver cirrhosis was obtained. Conclusion. The proposed methodology of liver cirrhosis formation correlates with pathomorphological changes in lymphocyte cell wall, changes in liver tissue and internal organs in human liver cirrhosis.

Abstract:AbstractNonalcoholic steatohepatitis (NASH) is a major health problem since it often leads to hepatocellular carcinoma. However, the underlying mechanisms of NASH development and subsequent fibrosis have yet to be clarified. We compared comprehensive lipidomic profiles between mice with high fat diet (HFD)-induced steatosis and STAM mice with NASH and subsequent fibrosis. The STAM mouse is a model that demonstrates NASH progression resembling the disease in humans: STAM mice manifest NASH at 8 weeks, which progresses to fibrosis at 12 weeks and finally develop hepatocellular carcinoma. Overall, 250 lipid molecules were detected in the liver using liquid chromatography-mass spectrometry. We found that STAM mice with NASH presented a significantly higher abundance of sphingolipids and lower levels of triacylglycerols than the HFD-fed control mice. The abundance of certain fatty acids in phospholipid side chains was also significantly different between STAM and control mice, although global levels of phosphatidylcholines and phosphatidylethanolamines were comparable. Finally, increase in levels of acylcarnitines and some diacylglycerols was observed in STAM mice toward the fibrosis stage, but not in age-matched control mice. Our study provides insights into the lipid status of the steatotic, NASH and fibrotic liver that would help elucidate the molecular pathophysiology of NASH progression.

Abstract:The biliary extra-drainage model in rat orthotopic liver transplantation provides a novel, simple, and reliable method for dynamic collection of bile. The operation success rate was satisfactory. This model was successful and could be applied in various experimental studies.

Establishment of Novel Mouse Model of Dietary NASH Rapidly Progressing into Liver Cirrhosis and Tumors

Abstract:Non-alcoholic steatohepatitis (NASH), which is the most severe manifestation of non-alcoholic fatty liver disease (NAFLD), has been recognized as a major hepatocellular carcinoma (HCC) catalyst. However, the molecular mechanism of NASH-liver fibrosis-HCC sequence remains unclear and a specific and effective treatment for NASH has not yet been established. The progress in this field depends on the availability of reliable preclinical models which show the steady progression to NASH, liver cirrhosis, and HCC. However, most of the NASH mouse models that have been described to date develop NASH generally for more than 24 weeks and there is an uncertainty of HCC development. To overcome such shortcomings of experimental NASH studies, we established a novel NASH-HCC mouse model with very high reproducibility, generality, and convenience. We treated male C57BL/6J mice with a newly developed choline-deficient and methionine-restricted high-fat diet, named OYC-NASH2 diet, for 60 weeks. Treatment of OYC-NASH2 diet for 3 weeks revealed marked steatosis, lobular inflammation, and fibrosis, histologically diagnosed as NASH. Liver cirrhosis was observed in all mice with 48-week treatment. Liver nodules emerged at 12 weeks of the treatment, > 2 mm diameter liver tumors developed in all mice at 24 weeks of the treatment and HCC appeared after 36-week treatment. In conclusion, our rapidly progressive and highly reproducible NASH-liver cirrhosis-HCC model is helpful for preclinical development and research on the pathogenesis of human NAFLD-NASH-HCC. Our mouse model would be useful for the development of novel chemicals for NASH-HCC-targeted therapies.

Abstract:Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence.By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient-like metastatic model of human HCC in nude mice (LCI-D20) and a low metastatic model of human HCC in nude mice (LCI-D35) have been established.All mice with transplanted LCI-D20 tumors exhibited e x t r e m e l y h i g h m e t a s t a t i c a b i l i t y i n c l u d i n g spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors.PAI-1 increased gradually following tumor progression in LCI-D20 model, and correlated with tumor size and AFP level.Phasic expression of tissue intercellular adhesion molecule-1 in this model was also observed.Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor.Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature.This LCI-D20 model has been widely used for the studies on intervention of metastasis, including anti-angiogenesis, antisense approach, metallopro teinase inhibitor, differentiation inducer, etc.It is concluded that the establishment of metastatic human HCC model in nude mice and human HCC cell line with metastatic potential will provide important models for the in vivo and in vitro study of HCC invasiveness, angiogenesis as well as intervention of HCC recurrence.
