Abstract:The gene expression patterns of antioxidative enzymes in cucumber (Cucumis sativus L.) fruits at four different maturity stages, immature (3-8 d after anthesis (DAA), mature (9-16 DAA), breaker (17-22 DAA), and yellow (35-40 DAA), were determined before and after cold storage at 2°C for 9 d and after subsequent rewarming at 20°C for 2 d. The electrolyte leakage and malondialdehyde content in cucumber fruits were increased after cold storage and subsequent rewarming. Increased expressions of peroxidase, ascorbate peroxidase (APX), and monodehydroascorbate reductase after cold storage played an important role in cucumber fruits to cope with chilling injury. The elevated cyt-superoxide dismutase, catalase, APX and dehydroascorbate reductase after subsequent rewarming in cucumber fruits facilitated the recovery from chilling stress. The highest expression levels of all the seven antioxidative enzyme genes in yellow fruits might be responsible for the enhanced chilling tolerance. Cucumber fruits at earlier developmental stages was more susceptible to chilling stress than those at later stages. The relative higher gene expressions of antioxidative enzymes genes at earlier developmental stages may be the responses to the sever oxidative stress caused by chilling injury.

Abstract:BACKGROUND: The aim of the present study was to reveal the effect of fruit maturity on the chilling tolerance of cucumber (Cucumis sativus L.) fruit and the oxidative and antioxidative mechanisms involved. Chinese mini-cucumber (cv. Hangcui-1) fruits were harvested at four developmental stages: Immature (38 days after anthesis (DAA)), Mature (916 DAA), Breaker (1722 DAA) and Yellow (3540 DAA). All fruits were stored at 2 degrees C for 9 days and rewarmed at 20 degrees C for 2 days. RESULTS: The chilling injury index declined with advancing fruit maturity. High superoxide anion radical production rate and hydrogen peroxide content were observed in Immature fruits after cold storage and rewarming. Under chilling stress, superoxide dismutase showed an early response. Fruits at earlier maturity stages exhibited higher catalase, ascorbate peroxidase and monodehydroascorbate reductase activities and glutathione content as well as its redox state, and lower peroxidase, dehydroascorbate reductase and glutathione reductase activities and ascorbate content as well as its redox state. CONCLUSION: Fruits at the earlier developmental stage are more susceptible to chilling injury, which is related to increased oxidative stress. High peroxidase activity and ascorbate content and maintenance of the latter's redox state appear critical to the chilling tolerance of cucumber fruits at later developmental stages. Quantitative Trait Loci Mapping and Candidate Gene Analysis of Low Temperature Tolerance in Cucumber Seedlings

Abstract:Cucumber (Cucumis sativus L.) is an economically important vegetable crop worldwide, but it is sensitive to low temperatures. Cucumber seedlings exposed to long-term low temperature stress (LT), i.e., below 20°C during the day, and 8°C at night, exhibit leaf yellowing, accelerated senescence, and reduced yield, therefore posing a threat to cucumber production. Studying the underlying mechanisms involved in LT tolerance in cucumber seedlings, and developing germplasm with improved LT-tolerance could provide fundamental solutions to the problem. In this study, an F2 population was generated from two parental lines, "CG104" (LT-tolerant inbred line) and "CG37" (LT-sensitive inbred line), to identify loci that are responsible for LT tolerance in cucumber seedlings. Replicated phenotypic analysis of the F2-derived F3 family using a low-temperature injury index (LTII) suggested that the LT tolerance of cucumber seedlings is controlled by multiple genes. A genetic map of 990.8 cM was constructed, with an average interval between markers of 5.2 cM. One quantitative trait loci (QTL) named qLTT5.1 on chromosome 5, and two QTLs named qLTT6.1 and qLTT6.2 on chromosome 6 were detected. Among them, qLTT6.2 accounted for 26.8 and 24.1% of the phenotypic variation in two different experiments. Single-nucleotide polymorphism (SNP) variations within the region of qLTT6.2 were analyzed using two contrasting in silico bulks generated from the cucumber core germplasm. Result showed that 214 SNPs were distributed within the 42-kb interval, containing three candidate genes. Real-time quantitative reverse transcription PCR and sequence analysis suggested that two genes Csa6G445210, an auxin response factor, and Csa6G445230, an ethylene-responsive transmembrane protein, might be candidate genes responsible for LT tolerance in cucumber seedlings. This study furthers the understanding of the molecular mechanism underlying LT tolerance in cucumber seedlings, and provides new markers for molecular breeding.

Abstract:Cold stress detrimentally influences fruit development, leading to a substantial yield reduction in many fruit-bearing vegetables. Cucumber, a vegetable of subtropical origin, is especially sensitive to cold. Cold-inducible parthenocarpy (CIP) promises fruit yield under cold conditions. Previously, we identified a CIP line EC5 in cucumber, which showed strong parthenocarpy and sustained fruit growth under cold conditions (16°C day/10°C night). However, the candidate gene and genetic mechanism underlying CIP in cucumber remain unknown. In this study, both BSA-seq and conventional QTL mapping strategies were employed on F 2 populations to delve into the genetic control of CIP. A single QTL, CIP5.1 , was consistently mapped across two winter seasons in 2021 and 2022. Fine mapping delimited the CIP locus into a 38.3 kb region on chromosome 5, harboring 8 candidate genes. Among these candidates, CsAGL11 ( CsaV3_5G040370 ) was identified, exhibiting multiple deletions/insertions in the promoter and 5'UTR region. The CsAGL11 gene encodes a MADS-box transcription factor protein, which is homologous to the genes previously recognized as negative regulators in ovule and fruit development of Arabidopsis and tomato. Correspondingly, cold treatment resulted in decreased expression of CsAGL11 during the early developmental stage of the fruit in EC5. A promoter activity assay confirmed promoter polymorphisms leading to weak transcriptional activation of CsAGL11 under cold conditions. This study deepens our understanding of the genetic characteristics of CIP and elucidates the potential role of the CsAGL11 gene in developing cucumber cultivars with enhanced fruiting under cold conditions.

GWAS reveals novel loci and identifies a pentatricopeptide repeat-containing protein (CsPPR) that improves low temperature germination in cucumber

Abstract:Low temperatures (LTs) negatively affect the percentage and rate of cucumber ( Cucumis sativus L.) seed germination, which has deleterious effects on yield. Here, a genome-wide association study (GWAS) was used to identify the genetic loci underlying low temperature germination (LTG) in 151 cucumber accessions that represented seven diverse ecotypes. Over two years, phenotypic data for LTG i.e., relative germination rate (RGR), relative germination energy (RGE), relative germination index (RGI) and relative radical length (RRL), were collected in two environments, and 17 of the 151 accessions were found to be highly cold tolerant using cluster analysis. A total of 1,522,847 significantly associated single-nucleotide polymorphism (SNP) were identified, and seven loci associated with LTG, on four chromosomes, were detected: gLTG1.1 , gLTG1.2 , gLTG1.3 , gLTG4.1 , gLTG5.1 , gLTG5.2 , and gLTG6.1 after resequencing of the accessions. Of the seven loci, three, i.e., gLTG1.2 , gLTG4.1 , and gLTG5.2 , showed strong signals that were consistent over two years using the four germination indices, and are thus strong and stable for LTG. Eight candidate genes associated with abiotic stress were identified, and three of them were potentially causal to LTG: CsaV3_1G044080 (a pentatricopeptide repeat-containing protein) for gLTG1.2 , CsaV3_4G013480 (a RING-type E3 ubiquitin transferase) for gLTG4.1 , and CsaV3_5G029350 (a serine/threonine-protein kinase) for gLTG5.2. The function for CsPPR ( CsaV3_1G044080 ) in regulating LTG was confirmed, as Arabidopsis lines ectopically expressing CsPPR showed higher germination and survival rates at 4°C compared to the wild-type, which preliminarily illustrates that CsPPR negatively regulates cucumber cold tolerance at the germination stage. This study will provide insights into cucumber LT-tolerance mechanisms and further promote cucumber breeding development.

Abstract:Low temperature (LT) stress threatens cucumber production globally, however, the molecular mechanisms underlying LT tolerance in cucumber remain largely unknown. Here, using a genome wide association study (GWAS), we found a naturally occurring SNP in the STAYGREEN (CsSGR) coding region at the gLTT5.1 locus associated with LT tolerance. Knockout mutants of CsSGR generated by CRISPR/Cas9 exhibit enhanced LT tolerance, in particularly, increased chlorophyll content and reduced ROS accumulation in response to LT. Moreover, the C‐repeat Binding Factor 1 (CsCBF1) transcription factor can directly activate the expression of CsSGR. We demonstrate that the LT‐sensitive haplotype CsSGRHapA, but not the LT‐tolerant haplotype CsSGRHapG could interact with NON‐YELLOW COLORING 1 (CsNYC1) to mediate chlorophyll degradation. Geographic distribution of the CsSGR haplotypes indicated that the CsSGR HapG was selected in cucumber accessions from high latitudes, potentially contributing to LT tolerance during cucumber cold‐adaptation in these regions. CsSGR mutants also showed enhanced tolerance to salinity, water‐deficit, and Pseudoperonospora cubensis, thus CsSGR is an elite target gene for breeding cucumber varieties with broad‐spectrum stress tolerance. Comprehensive analysis of cucumber C-repeat/dehydration-responsive element binding factor family genes and their potential roles in cold tolerance of cucumber

Abstract:Background: Cold stress is one of the main abiotic stresses limiting cucumber (Cucumis sativus L.) growth and production. C-repeat binding factor/Dehydration responsive element-binding 1 protein (CBF/DREB1), containing conserved APETALA2 (AP2) DNA binding domains and two characteristic sequences, are key signaling genes that can be rapidly induced and play vital roles in plant response to low temperature. However, the CBF family has not been systematically elucidated in cucumber, and the expression pattern of this family genes under cold stress remains unclear. Results: In this study, three CsCBF family genes were identified in cucumber genome and their protein conserved domain, protein physicochemical properties, gene structure and phylogenetic analysis were further comprehensively analyzed. Subcellular localization showed that all three CsCBFs were localized in the nucleus. Cis-element analysis of the promoters indicated that CsCBFs might be involved in plant hormone response and abiotic stress response. Expression analysis showed that the three CsCBFs could be significantly induced by cold stress, salt and ABA. The overexpression of CsCBFs in cucumber seedlings enhanced the tolerance to cold stress, and importantly, the transcript levels of CsCOR genes were significantly upregulated in 35S:CsCBFs transgenic plants after cold stress treatment. Biochemical analyses ascertained that CsCBFs directly activated CsCOR genes expression by binding to its promoter, thereby enhancing plant resistance to cold stress. Conclusion: This study provided a foundation for further research on the function of CsCBF genes in cold stress resistance and elucidating its mechanism.

Abstract:Introduction: Low ground temperature is a major factor limiting overwintering in cucumber cultivation facilities in northern alpine regions. Lower temperatures in the root zone directly affect the physiological function of the root system, which in turn affects the normal physiological activity of plants. However, the importance of the ground temperature in facilities has not attracted sufficient attention. Methods: Therefore, this study tested the cucumber variety Jinyou 35 under three root zone temperatures (room temperature, 20-22°C; suboptimal temperature, 13- 15°C; and low temperature, 8-10°C) to investigated possible cold resistance mechanisms in the root of cucumber seedlings through hormone, metabolomics, and transcriptomics analyses. Results and discussion: The results showed that cucumber roots were subjected to chilling stress at different temperatures. Hormone analysis indicated that auxin content was highest in the roots. Jasmonic acid and strigolactone participated in the low-temperature stress response. Auxin and jasmonate are key hormones that regulate the response of cucumber roots to low temperatures. Phenolic acid was the most abundant metabolite in cucumber roots under chilling stress. Additionally, triterpenes may play an important role in chilling resistance. Differentially expressed genes and metabolites were significantly enriched in benzoxazinoid biosynthesis in the room temperature vs. suboptimal temperature groups and the room temperature vs. low temperature groups. Most differentially expressed transcription factor genes in AP2/ERF were strongly induced in cucumber roots by both suboptimal and low-temperature stress conditions. These results provide guidance for the cultivation of cucumber in facilities.
Identification of QTLs Controlling Low‐temperature Tolerance During the Germination Stage in Cucumber (cucumis Sativus L.)

Abstract:Poor seed germination of cucumber at suboptimal temperatures is a great concern for growers wishing to take advantage of the early market. The development of low-temperature-tolerant varieties would be aided by understanding the inheritance of the trait and mapping its locus. In this study, a set of 140 recombinant inbred lines (RILs) derived from a cross between 65G (low-temperature-tolerant) and 02245 (low-temperature-sensitive) was used to identify the QTLs linked with low-temperature tolerance. A linkage map was developed using 135 simple sequence repeat (SSR) markers and five insertion-deletion (Indel) markers, and three QTLs were identified, qLTG1.1, qLTG2.1 and qLTG4.1. qLTG1.1, the major one for germination rate, germination energy and germination index, explained more than 50% of the observed phenotypic variability. The major QTL for radicle length, qLTG4.1, explained 13.8% of the phenotypic variability. The results showed that qLTG1.1 and qLTG4.1 play an important role in low-temperature tolerance during seed germination of cucumber and provide a basis for further fine mapping to determine the molecular mechanism for this trait.
Cucumber (cucumis Sativus L.) Over-Expressing Cold-Induced Transcriptome Regulator ICE1 Exhibits Changed Morphological Characters and Enhances Chilling Tolerance

Abstract:Cold is an environmental factor that limits the growing season of cucumber and adversely affects fruit quality and productivity. ICE1 is a positive regulator of CBF3 and has a critical role in cold stress (Chinnusamy et al., 2003). To test the function and potential use of cold-induced transcriptome regulator ICE1 (inducer of CBF expression) in improving the chill tolerance of cucumbers, the Arabidopsis ICE1 gene driven by super promoter was introduced into cucumbers by Agrobacterium-mediated transformation. Transgenic plants were obtained, and were confirmed by PCR and southern blot, then the expression of ICE1 was tested by RT-PCR. Transgenic plants exhibited dwarf phenotypes, and shorter internodes, with higher nodes in the first female flowers and a lower female flower rate of every node than wild type plants. Under cold stress, the chilling index of four transgenic lines (IF3, IF6, IF7, and IF10) was decreased significantly, indicating their improved chilling tolerance. Over-expression of ICE1 in cucumbers could induce the expression of cold-responsive genes, further activating the accumulation of soluble sugars and free proline, and inhibiting malondialdehyde (MDA) accumulation. This leads to electrolyte leakage and regulation of osmotic potential to osmotic stress under cold stress.

Abstract:Cucumber is an essential vegetable crop throughout the world. Cucumber development is vital for accomplishing both quality and productivity requirements. Meanwhile, numerous factors have resulted in substantial cucumber losses. However, the calreticulin domain-encoding genes (CDEGs) in cucumber were not well-characterized and had little function. In the genome-wide association study (GWAS), we recognized and characterized the CDEGs in Cucumis sativus (cucumber). Through a comprehensive study of C. sativus, our research has unveiled the presence of three unique genes, denoted as CsCRTb, CsCRT3, and CsCNX1, unevenly distributed on three chromosomes in the genome of C. sativus. In accordance to the phylogenetic investigation, these genes may be categorized into three subfamilies. Based on the resemblance with AtCDE genes, we reorganized the all CsCDE genes in accordance with international nomenclature. The expression analysis and cis-acting components revealed that each of CsCDE gene promoter region enclosed number of cis-elements connected with hormone and stress response. According to subcellular localization studies demonstrated that, they were found in deferent locations of the cell such as endoplasmic reticulum, plasma membrane, golgi apparatus, and vacuole, according to subcellular localization studies. Chromosomal distribution analysis and synteny analysis demonstrated the probability of segmental or tandem duplications within the cucumber CDEG gene family. Additionally, miRNAs displayed diverse modes of action, including mRNA cleavage and translational inhibition. We used the RNA seq data to analyze the expression of CDEG genes in response to cold stress and also improved cold tolerance, which was brought on by treating cucumber plants to an exogenous chitosan oligosaccharide spray. Our investigation revealed that these genes responded to this stress in a variety of ways, demonstrating that they may adapt quickly to environmental changes in cucumber plants. This study provides a base for further understanding in reference to CDE gene family and reveals that genes play significant functions in cucumber stress responses.

Role of important physiological traits and development of heat tolerance index in a large set of diverse cucumber germplasm

Abstract:Cucumber is an important vegetable crop that suffers from significant yield loss because of sub-optimal temperatures during the growing season. High temperature affects the plant's health and reduces the quality and quantity of the final harvest. Huge diversity in terms of different economically important traits, including wide temperature adaptation, is recorded in indigenous cucumber germplasm because of its Indian origin. It is necessary to identify the key traits and genotypes with the contrasting response from a large set of germplasm associated with heat stress response for understanding the physio-biochemical and molecular network associated with heat tolerance. A set of 123 germplasm was evaluated in a growth chamber with temperature stress treatment (40 °C/35 °C) for two subsequent seasons. Besides, 10 selected genotypes based on their response in the seedling stage were grown under natural field conditions with high temperatures to validate the physio-biochemical response in the seedling stage and yield parameters in the reproductive stage. Among the different parameters, slow degradation of chlorophyll, higher anti-oxidant enzyme activity, higher membrane stability index, and higher canopy temperature depression were identified as key traits explaining the heat stress response in cucumbers. Besides, the photosynthetic activities of the tolerant genotypes at the reproductive stage were also higher under field conditions, resulting in higher economic yield. Heat tolerance index was developed for 123 genotypes for seven physiological traits recorded in the present study. The optimised screening technique in the seedling stage and their validation for yield response under natural field facilitated the evaluation of a large number of genotypes for use in breeding for heat stress tolerance in cucumbers. Besides, the identified germplasm, WBC-13, DGC-103 and DARL-106 with effective heat stress tolerance will be instrumental in understanding the molecular basis of heat tolerance and designing climate-smart cucumber cultivars.

Abstract:Cucumber (Cucumis sativus L.) is an important economic vegetable crop worldwide that is susceptible to various common pathogens, including powdery mildew (PM), downy mildew (DM), and Fusarium wilt (FM). In cucumber breeding programs, identifying disease resistance and related molecular markers is generally a top priority. PM, DM, and FW are the major diseases of cucumber in China that cause severe yield losses and the genetic-based cucumber resistance against these diseases has been developed over the last decade. Still, the molecular mechanisms of cucumber disease resistance remain unclear. In this review, we summarize recent findings on the inheritance, molecular markers, and quantitative trait locus mapping of cucumber PM, DM, and FM resistance. In addition, several candidate genes, such as PM, DM, and FM resistance genes, with or without functional verification are reviewed. The data help to reveal the molecular mechanisms of cucumber disease resistance and provide exciting new opportunities for further resistance breeding.
CsBPC2 is essential for cucumber survival under cold stress

Abstract:Cold stress affects the growth and development of cucumbers. Whether the BPC2 transcription factor participates in cold tolerance and its regulatory mechanism in plants have not been reported. Here, we used wild-type (WT) cucumber seedlings and two mutant Csbpc2 lines as materials. The underlying mechanisms were studied by determining the phenotype, physiological and biochemical indicators, and transcriptome after cold stress. The results showed that CsBPC2 knockout reduced cucumber cold tolerance by increasing the chilling injury index, relative electrical conductivity and malondialdehyde (MDA) content and decreasing antioxidant enzyme activity. We then conducted RNA sequencing (RNA-seq) to explore transcript-level changes in Csbpc2 mutants. A large number of differentially expressed genes (1032) were identified and found to be unique in Csbpc2 mutants. However, only 489 down-regulated genes related to the synthesis and transport of amino acids and vitamins were found to be enriched through GO analysis. Moreover, both RNA-seq and qPT-PCR techniques revealed that CsBPC2 knockout also decreased the expression of some key cold-responsive genes, such as CsICE1 , CsCOR413IM2 , CsBZR1 and CsBZR2 . These results strongly suggested that CsBPC2 knockout not only affected cold function genes but also decreased the levels of some key metabolites under cold stress. In conclusion, this study reveals for the first time that CsBPC2 is essential for cold tolerance in cucumber and provides a reference for research on the biological function of BPC2 in other plants.

Abstract:The worldwide cultivated Cucurbita pepo L. is one of the most diverse species in the plant kingdom. In this study, chilling tolerance over a wide range of cultivars was characterized to discover the allelic variants to improving the postharvest quality of the immature fruit during cold storage. For this purpose, fruits from 126 accessions of worldwide origin have been evaluated for weight loss and chilling injury after 3, 7 and 14 days of cold storage, classifying them into tolerant, partially tolerant, and sensitive accessions. To verify this classification, antioxidant capacity and lipid peroxidation (MDA) of contrasting accessions (tolerant vs. sensitive) were assessed. The antioxidant capacity significantly decreased during cold storage in the sensitive accessions, while it was maintained in tolerant accessions. Additionally, the sensitive accessions presented a higher accumulation of MDA during this period. Finally, a GWAS analysis using GBS data available in CuGenDBv2, combined with weight loss percentage data, led to the identification of a candidate QTL located on chromosome 17 that regulates postharvest cold tolerance in zucchini. The region contains four SNPs whose alternative alleles were significantly associated with lower weight loss percentage and chilling injury indices during cold storage. Two SNPs are located in the 3' UTR region of the gene CpERS1, a gene involved in ethylene perception. The other two SNPs generate missense mutations in the coding region of a Pectin methyl esterase inhibitor gene (CpPMI). The role of this QTL and these variants in chilling tolerance is discussed.

Abstract:Soil salinization is one of the main problems faced by modern agricultural production, especially cucumber production. This study screened the salt–alkali-tolerant cultivar ‘D1909’ and the salt–alkali-sensitive cultivar ‘D1604’ from 32 different cucumber ecological types on the basis of morphological indicators and salt–alkali indices combined with relevant physiological and biochemical indices. By performing a transcriptome metabolome analysis, the key gene CsSRG1, which is responsive to salt–alkali stress in cucumber, was screened, and its function was verified. The role of CsSRG1 in reducing salt–alkali stress in cucumber was clarified, and the mechanism of salt–alkali tolerance in cucumber was preliminarily explored. This study provides germplasm resources for cucumber salt–alkali tolerance breeding and a theoretical basis for the effective use of saline alkali soil to achieve high quality and high yield in other crops.

Abstract:Cold stress is a major factor influencing the geographical distribution of soybean growth and causes immense losses in productivity. Understanding the molecular mechanisms that the soybean has undergone to survive cold temperatures will have immense value in improving soybean cold tolerance. This review focuses on the molecular mechanisms involved in soybean response to cold. We summarized the recent studies on soybean cold-tolerant quantitative trait loci (QTLs), transcription factors, associated cold-regulated (COR) genes, and the regulatory pathways in response to cold stress. Cold-tolerant QTLs were found to be overlapped with the genomic region of maturity loci of E1, E3, E4, pubescence color locus of T, stem growth habit gene locus of Dt1, and leaf shape locus of Ln, indicating that pleiotropic loci may control multiple traits, including cold tolerance. The C-repeat responsive element binding factors (CBFs) are evolutionarily conserved across species. The expression of most GmDREB1s was upregulated by cold stress and overexpression of GmDREB1B;1 in soybean protoplast, and transgenic Arabidopsis plants can increase the expression of genes with the DRE core motif in their promoter regions under cold stress. Other soybean cold-responsive regulators, such as GmMYBJ1, GmNEK1, GmZF1, GmbZIP, GmTCF1a, SCOF-1 and so on, enhance cold tolerance by regulating the expression of COR genes in transgenic Arabidopsis. CBF-dependent and CBF-independent pathways are cross-talking and work together to activate cold stress gene expression. Even though it requires further dissection for precise understanding, the function of soybean cold-responsive transcription factors and associated COR genes studied in Arabidopsis shed light on the molecular mechanism of cold responses in soybeans and other crops. Furthermore, the findings may also provide practical applications for breeding cold-tolerant soybean varieties in high-latitude and high-altitude regions.

Effect of cold stress on photosynthetic physiological characteristics and molecular mechanism analysis in cold-resistant cotton (ZM36) seedlings

Abstract:Low temperature and cold damage seriously hinder the growth, development, and morphogenesis of cotton seedlings. However, the response mechanism of cotton seedlings under cold stress still lacks research. In this study, transcriptome sequencing, gas exchange parameters, and rapid chlorophyll fluorescence parameters were analyzed in leaves of cold-tolerant upland cotton variety "ZM36" under different temperature stress [25°C (T25, CK), 15°C (T15), 10°C (T10), and 4°C (T4)]. The results showed that the net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), PSII potential maximum photochemical efficiency (Fv/Fm), and performance index (PIabs) of cotton leaves significantly decreased, and the intercellular CO 2 concentration (Ci) and Fo/Fm significantly increased under cold stress. The transcriptome sequencing analysis showed that a total of 13,183 DEGs were involved in the response of cotton seedlings at each temperature point (T25, T15, T10, and T4), mainly involving five metabolic pathways—the phosphatidylinositol signaling system, photosynthesis, photosynthesis antenna protein, carbon fixation in photosynthetic organisms, and carotenoid synthesis. The 1,119 transcription factors were discovered among all the DEGs. These transcription factors involve 59 families, of which 15.8% of genes in the NAC family are upregulated. Through network regulatory analysis, the five candidate genes GhUVR8 ( GH_A05G3668 ), GhPLATZ ( GH_A09G2161 ), GhFAD4-1 ( GH_A01G0758 ), GhNFYA1 ( GH_A02G1336 ), and GhFAD4-2 ( GH_D01G0766 ) were identified in response to cold stress. Furthermore, suppressing the expression level of GhPLATZ by virus-induced gene silencing led to the reduction of low temperature resistance, implying GhPLATZ as a positive regulator of low temperature tolerance. The findings of the study revealed a piece of the complex response mechanism of the cold-tolerant variety "ZM36" to different cold stresses and excavated key candidate genes for low temperature response, which provided support for accelerating the selection and breeding of cotton varieties with low temperature tolerance.

Preliminary Studies on Identification Method for Cucumber Tolerance to Low Temperature

Abstract:In this study, methods for identifying cucumber germplasm tolerance to low temperature at germination and seedling stages were compared systematically. First, the cold tolerance of 15 materials wereidentified under 13 ℃ and 15 ℃(CK:26 ℃), respectively using 6 indexes including germination percentage,germination power, germination index, radicle length, fresh weight and dry weight of seed buds. The results showed that 13℃ was suitable for screening materials with strong tolerance to low temperature, while 15 ℃ was suitable for evaluating cold tolerance of cucumber germplasm resources at budding stage. In addition, different treatments with stress duration under low temperature at the seedling stage were analyzed and compared taking injury index and recovery index as evaluation indexes. Treating cucumber seedlings under 6 ℃ constant low temperature, we found that 3-5 days were the suitable treatment of stressed time. While under 15℃ /5℃ variable temperature conditions, we found the suitable treatment of stressed time was 9-15 days. Taking recovery index as evaluation index to identify seedling tolerance to low temperature, we found that the days under stress condition could be 1-2 days shorter than that taking injury index as evaluation index.
