Abstract:<p>Zinc oxide nanoparticles (ZnO-NPs) has been demonstrated to positively regulate plant tolerance to multiple environmental stresses. However, till date little information has been gained regarding the role of ZnO-NPs in the salt stress regulation in plants. Hence, the objective of our study was to investigate the role of ZnO-NPs in the regulation of salt tolerance in tomato (<em>Lycopersicon esculentum</em> Mill.). In this regard, the tomato plants were subjected to salt stress by using NaCl (150 mM) at the time of transplantation [15 days after sowing (DAS)]. Foliar application of ZnO-NPs at different levels viz., 10, 50 and 100 mg/L in the presence/absence of NaCl (150 mM) was carried out at 25 DAS and sampling was done at 35 DAS. Results of our study revealed that foliar spray of ZnO-NPs significantly increased shoot length (SL) and root length (RL), biomass, leaf area, chlorophyll content and photosynthetic attributes of tomato plants in the presence/absence of salt stress. Besides, the application of ZnO-NPs mitigates the negative impacts of salt stress on tomato growth, and enhanced protein content and antioxidative enzyme activity such as peroxidase (POX), superoxide dismutase (SOD) and catalase (CAT) under salt stress. In conclusion, the ZnO-NPs plays an important role in the alleviation of NaCl toxicity in tomato plants. Hence, the ZnO-NPs can be used to boost the growth performance and mitigate the adverse effects caused by NaCl in cereals.</p>

Abstract:Salinity stress affects plant growth, development, nutrient uptake, and yield. Applications of micronutrients, specifically zinc (Zn), can mitigate the harmful consequences of salt stress. During the winter season of 2022, an experiment was conducted in the net house of BINA substation Satkhira, Bangladesh, to examine the impact of different Zn concentrations (5 and 10 kg ha-1) on tomato (Lycopersicon esculentum L.) growth, yield, fruit quality, and nutrient acquisition abilities under different salt stress (SS) conditions (SS0.5%, SS1.0%, and SS1.5% NaCl). The result of the study showed that different stress conditions lowered the plant height, the number of branches per plant, flower clusters, and fruits per plant, plant yield, vitamin C, protein and lycopene contents, and the acquisition of different nutrients, i.e., nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), zinc (Zn) and iron (Fe). The application of 10 kg Zn ha-1 (Zn10) increased all previously mentioned parameters in both saline and usual conditions. On the other hand, a decrease in the amount of Na in fruit was observed when Zn application was increased from 5 to 10 kg ha-1. Plant Na/K ratios were consequently lowest at the highest Zn concentration. Therefore, the findings indicate that Zn application improves tomato growth, yield, fruit quality, and nutrient acquisition traits by mitigating the negative impacts of saline environments.

Abstract:Salinity reduces crop yields and quality, causing global economic losses. Zinc oxide nanoparticles (ZnO-NPs) improve plant physiological and metabolic processes and abiotic stress resistance. This study examined the effects of foliar ZnO-NPs at 75 and 150 mg/L on tomato Kecskeméti 549 plants to alleviate salt stress caused by 150 mM NaCl. The precipitation procedure produced ZnO-NPs that were characterized using UV-VIS, TEM, STEM, DLS, EDAX, Zeta potential, and FTIR. The study assessed TPCs, TFCs, total hydrolyzable sugars, total free amino acids, protein, proline, H2O2, and MDA along with plant height, stem width, leaf area, and SPAD values. The polyphenolic burden was also measured by HPLC. With salt stress, plant growth and chlorophyll content decreased significantly. The growth and development of tomato plants changed by applying the ZnO-NPs. Dosages of ZnO-NPs had a significant effect across treatments. ZnO-NPs also increased chlorophyll, reduced stress markers, and released phenolic chemicals and proteins in the leaves of tomatoes. ZnO-NPs reduce salt stress by promoting the uptake of minerals. ZnO-NPs had beneficial effects on tomato plants when subjected to salt stress, making them an alternate technique to boost resilience in saline soils or low-quality irrigation water. This study examined how foliar application of chemically synthesized ZnO-NPs to the leaves affected biochemistry, morphology, and phenolic compound synthesis with and without NaCl.

Abstract:The properties of nanomaterials and their potential applications have been given considerable attention by researchers in various fields, especially agricultural biotechnology. However, not much has been done to evaluate the role or effect of zinc oxide nanoparticles (ZnO-NP) in regulating physiological and biochemical processes in response to salt-induced stress. For this purpose, some callus growth traits, plant regeneration rate, mineral element (sodium, potassium, phosphorous and nitrogen) contents and changes in the activity of superoxide dismutase (SOD) and glutathione peroxidase (GPX) in tissues of five tomato cultivars were investigated in a callus culture exposed to elevated concentrations of salt (3.0 and 6.0 g L-1NaCl), and in the presence of zinc oxide nanoparticles (15 and 30 mg L-1). The relative callus growth rate was inhibited by 3.0 g L-1 NaCl; this was increased dramatically at 6.0 g L-1. Increasing exposure to NaCl was associated with a significantly higher sodium content and SOD and GPX activities. Zinc oxide nanoparticles mitigated the effects of NaCl, and in this application of lower concentrations (15 mg L-1) was more effective than a higher concentration (30 mg L-1). This finding indicates that zinc oxide nanoparticles should be investigated further as a potential anti-stress agent in crop production. Different tomato cultivars showed different degrees of tolerance to salinity in the presence of ZnO-NP. The cultivars Edkawy, followed by Sandpoint, were less affected by salt stress than the cultivar Anna Aasa.

Abstract:Plants antioxidative system is the first line of defense against oxidative stress caused secondarily by toxic ions under salinity. Plants with pre-activated antioxidative system can better adapt to salinity and can result in higher growth and yield. The current experiment was conducted to assess the adaptation of two tomato genotypes (Riogrande and Green Gold) with pre-activated antioxidative enzymes against salt stress. Tomato seedlings were exposed to mild stress (Ni: 0, 15 and 30 mg L −1 ) for three weeks to activate the antioxidative enzymes. The seedlings with pre-activated antioxidative enzymes were then grown under severe stress in hydroponics (0, 75 and 150 mM NaCl) and soil (control, 7.5 and 15 dS m −1 ) to check the adaptation, growth and yield. The results showed that Ni toxicity significantly enhanced activities of antioxidant enzymes (SOD, CAT, APX and POX) in both the genotypes and reduced growth with higher values in genotype Riogrande than Green Gold. The seedlings with pre-activated antioxidant enzymes showed better growth, low Na + and high K + uptake and maintained higher antioxidative enzymes activity than non-treated seedlings after four weeks of salt stress treatment in hydroponics. Similarly, the results in soil salinity treatment of the Ni pretreated seedlings showed higher yield characteristics (fruit yield per plant, average fruit weight and fruit diameter) than non-treated seedlings. However, Ni pretreatment had nonsignificant effect on tomato fruit quality characteristics like fruit dry matter percentage, total soluble solids, fruit juice pH and titratable acidity. The genotype Riogrande showed better growth, yield and fruit quality than Green Gold due to higher activity of antioxidant enzymes and better ion homeostasis as a result of Ni pretreatment. The results suggest that pre-activation antioxidant enzymes by Ni treatment proved to be an effective strategy to attenuate salt stress for better growth and yield of tomato plants.

Abstract:The interaction between zinc and cadmium was studied in tomato leaves (Solanum lycopersicum), in a hydroponic medium. Ten-day-old seedlings were treated with 25 μM CdCl2 associated to different concentrations of ZnCl2 (10, 50, 100, and 150 μM). We first observed that seven days after the metallic exposure Zn supply clearly reduced Cd concentration in leaves and simultaneously increased Zn uptake. Moreover, the addition of 10 μM ZnCl2 relieved the leaves growth inhibition observed under Cd stress. At the same concentration (10 μM), Zn repaired the Cd damages on the membrane lipid content and fatty acids composition. In fact, Zn supplementation at low level (10 μM) restored and enhanced the quantities of the total lipid (TL), galactolipids (GL), phospholipids (PL) and neutral lipids (NL) that were reduced by 25 μM CdCl2. There was also a substantial amelioration in total lipid biosynthesis that was noticeably altered with Cd treatment. In 10 μM Zn-Cd treated leaves the [1-14C]acetate incorporation in total lipids as well as in all lipid classes was similar to that of control leaves. Likewise, levels of tri-unsaturated fatty acids that dropped in total lipids of Cd treated leaves showed an increase after supplementation of 10 μM Zn. Moreover, the beneficial effect of the same dose of Zn (10 μM) on Cd toxicity was confirmed by a significant decrease in the oxidative stress state which was marked by a reduction in thiobarbituric acid-reacting substances (TBARS) levels and a reduction in lipoxygenase (LOX, E.C.1.11.1.7) activity that was significantly enhanced by Cd treatment. These results suggest that 10 μM Zn supplementation proved to be inhibitory for Cd-induced oxidative stress and beneficial for the plant’s survival. However, higher Zn concentrations (50, 100 and 150 μM) and Cd are synergistic in their effect on plant growth parameters and lipid content.

Abstract:Salinity is an edaphic stress that dramatically restricts worldwide crop production. Nanomaterials and plant growth-promoting bacteria (PGPB) are currently used to alleviate the negative effects of various stresses on plant growth and development. This study investigates the protective effects of different levels of zinc oxide nanoparticles (ZnO-NPs) (0, 20, and 40 mg L−1) and PGPBs (no bacteria, Bacillus subtilis, Lactobacillus casei, Bacillus pumilus) on DNA damage and cytosine methylation changes in the tomato (Solanum lycopersicum L. ‘Linda’) seedlings under salinity stress (250 mM NaCl). Coupled Restriction Enzyme Digestion-Random Amplification (CRED-RA) and Randomly Amplified Polymorphic DNA (RAPD) approaches were used to analyze changes in cytosine methylation and to determine how genotoxic effects influence genomic stability. Salinity stress increased the polymorphism rate assessed by RAPD, while PGPB and ZnO-NPs reduced the adverse effects of salinity stress. Genomic template stability was increased by the PGPBs and ZnO-NPs application; this increase was significant when Lactobacillus casei and 40 mg L−1 of ZnO-NPs were used.A decreased level of DNA methylation was observed in all treatments. Taken together, the use of PGPB and ZnO-NPs had a general positive effect under salinity stress reducing genetic impairment in tomato seedlings.
Mitigation of sodium chloride toxicity in Solanum lycopersicum L. by supplementation of jasmonic acid and nitric oxide

Abstract:We investigated the effects of exogenous application of jasmonic acid (JA) and nitric oxide (NO) on growth, antioxidant metabolism, physio-biochemical attributes and metabolite accumulation, in tomato (Solanum lycopersicum L.) plants exposed to salt stress. Treating the plants with NaCl (200 mM) resulted in considerable growth inhibition in terms of biomass, relative water content, and chlorophyll content, all of which were significantly improved upon application of JA and NO under both normal and NaCl-stress treatments. Salt treatment particularly 200 mM NaCl caused an apparent increase in electrolyte leakage, lipid peroxidation, and hydrogen peroxide production, which were reduced by exogenous application of JA and NO. Salt treatment triggered the induction of antioxidant system by enhancing the activities of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR). Application of JA and NO separately as well as in combination caused a significant improvement in activities of SOD, CAT, APX, and GR activities. JA and NO either applied individually or in combination boosted the flavonoid, proline and glycine betaine synthesis under NaCl treatments. In conclusion, the exogenous application of JA and NO protected tomato plants from NaCl-induced damage by up-regulating the antioxidant metabolism, osmolyte synthesis, and metabolite accumulation.

Abstract:Due to increasing global climate change problems to biota, salinity has been recognized as a realistic hazard critically affects the sustain agri-food production and crop quality in many regions of the world. Nanotechnology as an innovative approach can effectively improve plant performance under risky conditions such as salinity. Taking in consideration the ameliorative role of nanoparticles such as calcium nanoparticles (Ca-NPs) in enhancing plant growth and tolerance against various abiotic stresses, the present study was undertaken to illuminate the powerful effect and the underlying mechanism of soil-applied Ca-NPs (20 mM) in preventing salt damage at saline conditions (NaCl at 50, 100 and 200 mM) in tomato. Data revealed that NaCl drastically imposed the morphological parameters, primary and secondary metabolism, photosynthesis pigment content, hydrogen peroxide and lipid peroxidation levels, antioxidant enzyme activities, mineral contents, and protein patterns. In contrast, the supplementation of exogenous Ca-NPs modified salinity toxicity effects by improving the survival, growth parameters, anabolic (soluble ions and osmolytes) and defense mechanisms (enzymatic and nonenzymatic antioxidants). Interestingly, under lethal salinity level (200 mM), Ca-NPs was capable of suppressing the excessive damage effect of salinity by up-regulating the performance of the plants when these plants were completely dead in the absence of Ca-NPs. The descriptive cluster analysis separated treatments and characteristics into 3 to elucidate negative and positive correlations. Moreover, Ca-NPs was more efficient than CaCl2 in eliciting salt tolerance under all investigated NaCl levels. Therefore, all these findings together conclude that Ca-NPs have a positive role in motivating resilience strategies in tomato plants toward salt stress via lessening the ROS overproduction, stimulating enzymatic antioxidants, promoting osmolytes accumulation, and renovating protein profile under mild and severe salinity levels.

Abstract:Salinity stress (SS) is a challenging abiotic stress that limits crop growth and productivity. Sustainable and cost effective methods are needed to improve crop production and decrease the deleterious impacts of SS. Zinc (Zn) nano-particles (NPs) have emerged as an important approach to regulating plant tolerance against SS. However, the mechanisms of SS tolerance mediated by Zn-NPs are not fully explained. Thus, this study was performed to explore the role of Zn-NPs (seed priming and foliar spray) in reducing the deleterious impacts of SS on wheat plants. The study comprised different SS levels: control, 6 and 12 dS m −1 , and different Zn-NPs treatments: control, seed priming (40 ppm), foliar spray (20 ppm), and their combination. Salinity stress markedly reduced plant growth, biomass, and grain yield. This was associated with enhanced electrolyte leakage (EL), malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ), sodium (Na), chloride (Cl) accumulation, reduced photosynthetic pigments, relative water contents (RWC), photosynthetic rate (Pn), transpiration rate (Tr), stomata conductance (Gs), water use efficiency (WUE), free amino acids (FAA), total soluble protein (TSP), indole acetic acid (IAA), gibberellic acid (GA), and nutrients (Ca, Mg, K, N, and P). However, the application of Zn-NPs significantly improved the yield of the wheat crop, which was associated with reduced abscisic acid (ABA), MDA, H 2 O 2 concentration, and EL, owing to improved antioxidant activities, and an increase in RWC, Pn, Tr, WUE, and the accumulation of osmoregulating compounds (proline, soluble sugars, TSP, and FAA) and hormones (GA and IAA). Furthermore, Zn-NPs contrasted the salinity-induced uptake of toxic ions (Na and Cl) and increased the uptake of Ca, K, Mg, N, and P. Additionally, Zn-NPs application substantially increased the wheat grain Zn bio-fortification. Our results support previous findings on the role of Zn-NPs in wheat growth, yield, and grain Zn bio-fortification, demonstrating that beneficial effects are obtained under normal as well as adverse conditions, thanks to improved physiological activity and the accumulation of useful compounds. This sets the premise for general use of Zn-NPs in wheat, to which aim more experimental evidence is intensively being sought. Further studies are needed at the genomic, transcriptomic, proteomic, and metabolomic level to better acknowledge the mechanisms of general physiological enhancement observed with Zn-NPs application.

The Endophytic Strain ZS-3 Enhances Salt Tolerance in Arabidopsis thaliana by Regulating Photosynthesis, Osmotic Stress, and Ion Homeostasis and Inducing Systemic Tolerance

Abstract:Soil salinity is one of the main factors limiting agricultural development worldwide and has an adverse effect on plant growth and yield. To date, plant growth-promoting rhizobacteria (PGPR) are considered to be one of the most promising eco-friendly strategies for improving saline soils. The bacterium Bacillus megaterium ZS-3 is an excellent PGPR strain that induces growth promotion as well as biotic stress resistance and tolerance to abiotic stress in a broad range of host plants. In this study, the potential mechanisms of protection against salinity stress by B. megaterium ZS-3 in Arabidopsis thaliana were explored. Regulation by ZS-3 improved growth in A . thaliana under severe saline conditions. The results showed that ZS-3 treatment significantly increased the biomass, chlorophyll content and carotenoid content of A . thaliana . Compared to the control, the leaf area and total fresh weight of plants inoculated with ZS-3 increased by 245% and 271%, respectively; the chlorophyll a, chlorophyll b, and carotenoid contents increased by 335%, 146%, and 372%, respectively, under salt stress. Physiological and biochemical tests showed that ZS-3 regulated the content of osmotic substances in plants under salt stress. Compared to the control, the soluble sugar content of the ZS-3-treated group was significantly increased by 288%, while the proline content was significantly reduced by 41.43%. Quantification of Na + and K + contents showed that ZS-3 treatment significantly reduced Na + accumulation and increased the K + /Na + ratio in plants. ZS-3 also isolated Na + in vesicles by upregulating NHX1 and AVP1 expression while limiting Na + uptake by downregulating HKT1 , which protected against Na + toxicity. Higher levels of peroxidase and catalase activity and reduced glutathione were detected in plants inoculated with ZS-3 compared to those in uninoculated plants. In addition, it was revealed that ZS-3 activates salicylic acid ( NPR1 and PR1 ) and jasmonic acid/ethylene ( AOS , LOX2 , PDF1.2 , and ERF1 ) signaling pathways to induce systemic tolerance, thereby inducing salt tolerance in plants. In conclusion, the results of this study indicate that ZS-3 has the potential to act as an environmentally friendly salt tolerance inducer that can promote plant growth in salt-stressed environments.

Abstract:Seed priming with nanoparticles (NPs) has gained considerable scientific attention; especially in regulating seed metabolism and signaling pathways, to establish seedling with improved germination in stress condition. Hence, the present study was conducted to understand the effect of zinc oxide nanoparticles (ZnO-NPs) as a seed priming agent on physio-biochemical and seed quality parameters of tomato ( Solanum lycopersicum L.) under various salinity stress level (50 and 100 mM NaCl). Results showed that significant decrease was recorded in seed quality, physiological and biochemical parameters in unprimed seeds under 50 and 100 mM NaCl salinity stress condition, respectively as compared to control (without NaCl condition). However, ZnO NP @ 750 ppm for 6 h duration was found to notably increase the germination (4.8 and 12.6%), seed vigour index I (42.6 and 69%), II (43.1 and 68.1%), photosynthetic pigments (28.9 and 33.1%), TSS (27.0 and 26.5%), soluble protein (14.2 and 15.1%), phenol (40.4 and 34.1%), proline (59.0 and 62.1%) and antioxidant potential, however, the mean germination time (12.6 and 14.1%) was found to decrease under 50 and 100 mM NaCl salinity stress conditions, respectively, as compared to unprimed seeds. The genotype, QA001 performed significantly better than QA002 genotype under both control and salinity stress condition. Overall, seed priming with ZnO NP significantly enhanced chlorophyll content, enzymatic and non-enzymatic antioxidative defense mechanism and osmotic adjustment, which in turn reduced ROS accumulation and lipid peroxidation in cell membrane in tomato at the early seedling stage.

Abstract:ABSTRACT One of the most deleterious abiotic stresses is the salinity stress, which causes inhibition of growth and development. Therefore, this investigation was conducted to evaluate the effects of sodium nitroprusside (SNP) and silicic acid (Si) on biochemical response of Solanum lycopersicum (cv. Isabella) under different salinity levels during vegetative stage. For this purpose, the seedlings were subjected to different salt stress levels (0, 25, 50, 100, and 150 mM) and supplemented with optimized concentration of silicon (Si) (0, and 2.5 mM of H4SiO4) and sodium nitroprusside (SNP) (0, and 100 μM) to assess variations in enzyme activity and biochemical properties of tomato plants during vegetative growth. Salt stress inhibited the chlorophyll and carotenoid contents of tomato plants. The antioxidant enzyme activities such as catalase (CAT) and superoxide dismutase (SOD) as well as the levels of osmolytes (proline, glycine betaine), malondialdehyde (MDA), and hydrogen peroxide (H2O2) increased in tomato plants due to high salinity. Furthermore, the exogenous use of SNP and Si to alleviate the effect of salinity on the plants increased the antioxidant enzyme activities and osmolyte levels compared to NaCl-treated plants. In addition, in the plants under salt stress, supplemented with SNP and Si, the contents of MDA and H2O2 decreased. Therefore, the exogenous use of Si and SNP led to protecting a tomato plant against oxidative damage induced by salt stress by stimulating synthesis of antioxidant enzyme. The findings indicated that, with the improvement in antioxidative defense system, pigment syntheses, and osmolyte accumulation, SNP and Si had the ability to alleviate adverse impact of high salinity on tomato plants.

Abstract:Detrimental effects of salinity could be mitigated by exogenous zinc (Zn) application; however, the mechanisms underlying this amelioration are poorly understood. This study demonstrated the interaction between Zn and salinity by measuring plant biomass, photosynthetic performance, ion concentrations, ROS accumulation, antioxidant activity and electrophysiological parameters in barley ( Hordeum vulgare L.). Salinity stress (200 mM NaCl for 3 weeks) resulted in a massive reduction in plant biomass; however, both fresh and dry weight of shoots were increased by ~30% with adequate Zn supply. Zinc supplementation also maintained K + and Na + homeostasis and prevented H 2 O 2 toxicity under salinity stress. Furthermore, exposure to 10 mM H 2 O 2 resulted in massive K + efflux from root epidermal cells in both the elongation and mature root zones, and pre-treating roots with Zn reduced ROS-induced K + efflux from the roots by 3–4-fold. Similar results were observed for Ca 2+ . The observed effects may be causally related to more efficient regulation of cation-permeable non-selective channels involved in the transport and sequestration of Na + , K + and Ca 2+ in various cellular compartments and tissues. This study provides valuable insights into Zn protective functions in plants and encourages the use of Zn fertilisers in barley crops grown on salt-affected soils.

Abstract:Abstract Background Salinity is one of the most challenging abiotic stresses restricting the growth of plants. In vitro screening will increase the efficiency and speed of salinity tolerant genotypes identifications. The response of four tomato cultivars under salinity was analyzed in vitro to evaluate the seedlings growth, biochemical, and gene expression responses as well as the effect of nano zinc and iron on callus induction and plant regeneration. Results The results showed that an increase in salinity stress in the medium decreased the germination percentage, fresh and dry weight of shoot, root length, chlorophyll a, b and carotenoids content, K and Ca content, and on the other hand, Na content was increased. MDA content (‘Nora’, ‘PS-10’, ‘Peto’ and ‘Roma’: 1.71, 1.78, 1.66 and 2.16 folds, respectively), electrolyte leakage (‘PS-10’: 33.33%; ‘Roma’: 56.33%), were increased with salinity of 100 mM compared to control. Proline content was increased in 50 mM NaCl (10.8 fold). The most activity of antioxidant enzymes including CAT, SOD, APX, GPX, and GR was observed in the ‘PS-10’ cultivar, and the lowest activity of these enzymes was observed in ‘Roma’ under salinity stress. The AsA and GSH were decreased and DHA and GSSG were increased with the increased intensity of salinity. The relative expression of SOD , APX , and GR genes varied in different cultivars at different salinity concentrations. The most percentage of callus induction was observed with applying iron oxide nanoparticles, and the most regeneration rate was recorded using zinc oxide nanoparticles. Conclusion The results showed that salt-tolerant cultivars such as ‘PS-10’ with better osmotic adjustment, are suitable candidates for the future production and breeding programs. The use of nutrient nanoparticles under salinity stress for different tomato cultivars increased their performance.

Abstract:Tomato is affected by various biotic and abiotic stresses, especially salinity, which drastically hinders the growth and yield of tomato. Calcium (Ca) is a vital macronutrient which plays physiological and biochemical roles in plants. Hence, we studied the protective roles of Ca against salinity stress in tomato. There were eight treatments comprising control (nutrient solution), 5 mM Ca, 10 mM Ca, 15 mM Ca, 12 dS m-1 NaCl, 12 dS m-1 NaCl + 5 mM Ca, 12 dS m-1 NaCl + 10 mM Ca and 12 dS m-1 NaCl + 15 mM Ca, and two tomato varieties: BARI tomato-2 and Binatomato-5. Salinity significantly decreased the plant-growth and yield attributes, relative water content (RWC), photosynthetic pigments (SPAD value) and the uptake of K, Ca and Mg in leaves and roots. Salinity-induced oxidative stress was present in the form of increased Na+ ion concentration, hydrogen peroxide (H2O2) content and lipid peroxidation (MDA). Ca application reduced oxidative stress through the boosting of antioxidant enzymatic activity. Exogenous Ca application enhanced proline and glycine betaine content and reduced Na+ uptake, which resulted in the inhibition of ionic toxicity and osmotic stress, respectively. Hence, Ca application significantly increased the growth and yield attributes, RWC, SPAD value, and uptake of K, Ca and Mg. Calcium application also had a significant effect on the fruit quality of tomato and the highest total soluble solid, total sugar, reducing sugar, β-carotene, vitamin C and juice pH were found for the combined application of NaCl and Ca. Therefore, application of Ca reversed the salt-induced changes through increasing osmoprotectants, activation of antioxidants enzymes, and by optimizing mineral nutrient status.

Abstract:Salinity is among the most important abiotic stresses, which negatively affect growth, nutrient uptake and yield of crop plants. Application of different micronutrients, particularly zinc (Zn) have the potential to ameliorate the negative impacts of salinity stress. However, the role of Zn in improving salinity tolerance of basil ( Ocimum basilicum L.) is poorly understood. This study evaluated the impact of different Zn levels (0, 5 and 10 mg kg -1 ) on growth and nutrient acquisition traits of basil under different salinity levels (0, 0.5, 1.0 and 1.5% NaCl). Data relating to biomass production, chlorophyll index, sodium (Na), potassium (K) uptake, K/Na ratio, Zn, copper (Cu), manganese (Mn) and iron (Fe) uptake were recorded. Increasing salinity level reduced biomass production, chlorophyll index and nutrient uptake traits (except for Na and Fe accumulation) of basil. Zinc application (10 mg kg -1 ) improved biomass production, chlorophyll index and nutrient acquisition traits under normal as well as saline conditions. The reduction in chlorophyll index and biomass production was higher under 0 and 5 mg kg -1 than 10 mg kg -1 Zn application. The K concentration decreased under increasing salinity; however, Zn application improved K uptake under normal as well as saline conditions. Different growth and nutrient acquisition traits had negative correlations with Na accumulation; however, no positive correlation was recorded among growth and nutrient uptake traits. The results revealed that Zn application could improve the salinity tolerance of basil. However, actual biochemical and genetic mechanisms involved in Zn-induced salinity tolerance warrant further investigation.

Abstract:Saline-alkaline stress inhibits plant growth and reduces yield. Abscisic acid (ABA) is an important plant hormone in response to plant stress. However, the role of ABA under saline-alkaline stress is poorly understood. Therefore, the mechanisms of ABA accumulation and resistance improvement in tomato seedlings were studied under saline-alkaline stress. We investigated whether ABA accumulation improved the saline-alkaline stress resistance ability of tomato. Here, wild-type (Solanum lycopersicum cv. Ailsa Craig) and ABA-deficient mutant (notabilis) seedlings were used to determine the membrane lipid peroxidation, osmotic substance and chlorophyll contents. ABA synthesis and signal transduction changes and ABA roles regulating the antioxidation in tomato seedlings subject to saline-alkaline stress were further explored. Results showed that ABA synthesis and signal transduction were induced by saline-alkaline stress. Under saline-alkaline stress, tomato seedlings had decreased relative water content, increased relative electrical conductivity and malondialdehyde content, and these changes were alleviated by exogenous ABA treatment. Exogenous ABA alleviated the degradation of chlorophyll in the leaves of tomato seedlings caused by saline-alkaline stress, further promoted the accumulation of proline and soluble sugar, reduced the content of ROS and improved the ability of the antioxidant enzyme system. Moreover, notabilis appeared to be sensitive to saline-alkaline stress. Overall, ABA is involved in the resistance of tomato seedlings to saline-alkaline stress, and exogenous ABA improves the saline-alkaline tolerance of tomato seedlings.
