Abstract:OBJECTIVES: The aim of this study was to investigate the response of the growth hormone (GH) in rat anterior pituitary to intermittent hypoxia (IH) and its modulation by hypothalamic somatostatin (SS). SETTING AND DESIGN: To observe the hypoxic response, rats were exposed to simulated altitude hypoxia (2km or 5km) in a hypobaric chamber for various days (4h/d); to clarify SS-involvement, rats were pretreated with SS antagonist (cysteamine, CSH, 200mg/kg/d, s.c.) then exposed to IH (5km) for 2d. The GH mRNA and immunostaining GH in pituitary as well as immunostaining SS in median eminence (ME) of hypothalamus were assayed by RT-PCR and immunohistochemistry, respectively. RESULTS: IH of 5 km altitude (IH5) markedly suppressed the body weight gain (BWG) of rats from Id to 10d, and it was returned to control level henceforth, while no significant influence was showed in the group of IH of 2 km altitude (IH2). IH5 for 2 and 5d significantly decreased GH mRNA expression in the pituitary. The pituitary immunostaining GH was remarkably increased in groups of IH2 for 5, 10, and 15 d, and in groups of IH5 for 2, 5, and 10d. Immunostaining SS in ME was significantly reduced in group of IH2 for 5d, and in groups of IH5 for 2d and 5d. Pretreatments (s.c.) with SS antagonist (CSH) significantly reversed IH5-caused increase of immunostaining GH and reduction of mRNA level in pituitary. CONCLUSIONS: IH may cause a short-term and recoverable suppression of GH release, and reduce GH mRNA expression in anterior pituitary, which may depend on the intensity and duration of the hypoxia. This suppression may be due to a modulation of hypoxia-activated SS.
A Randomized Controlled Phase 3 Study on the Efficacy and Safety of Recombinant Human Growth Hormone in Children with Idiopathic Short Stature

Abstract:BackgroundTo evaluate the safety and efficacy of daily somatropin (Jintropin®), a recombinant human growth hormone, in prepubertal children with ISS in China.MethodsThis study was a multicenter, randomized, controlled, open-label, phase 3 study. All subjects were randomized 3:1 to daily somatropin 0.05 mg/kg/day or no treatment for 52 weeks. A total of 481 subjects with a mean baseline age of 5.8 years were enrolled in the study. The primary endpoint was change in (△) height standard deviation score (HT-SDS) for chronological age (CA). Secondary endpoints included △height from baseline; △bone age (BA)/CA; △height velocity (HV) and △insulin-like growth factor 1 (IGF-1 SDS).Results△HT-SDS at week 52 was 1.04 ± 0.31 in the treatment group and 0.20 ± 0.33 in the control group (P < 0.001). At week 52, statistical significance was observed in the treatment group compared with control for △height (10.19 ± 1.47 cm vs. 5.85 ± 1.80 cm; P < 0.001), △BA/CA (0.04 ± 0.09 vs. 0.004 ± 0.01; P < 0.001), △HV (5.17 ± 3.70 cm/year vs. 0.75 ± 4.34 cm/year; P < 0.001), and △IGF-1 SDS (2.31 ± 1.20 vs. 0.22 ± 0.98; P < 0.001). The frequencies of treatment-emergent adverse events (TEAEs) were similar for the treatment and the control groups (89.8% vs. 82.4%); most TEAEs were mild to moderate in severity and 23 AEs were considered study-drug related.ConclusionsDaily subcutaneous administration of somatropin at 0.05 mg/kg/day for 52 weeks demonstrated improvement in growth outcomes and was well tolerated with a favorable safety profile.Trial RegistrationClinicalTrials.gov (identifier: NCT03635580). Abstract:Background: The insulin-like growth factor (IGF) system plays a vital role in regulating gonadotropin-releasing hormone (GnRH), whether the IGF2 can act on the GnRH neurons during the pubertal period is unclear. Methods: Central precocious puberty (CPP) rats were induced by danazol, and when the rats met the first diestrus, they were euthanized and tissues were collected. GT1-7 cells were cultured and treated with 0, 1, 10 ng/mL IGF2 for 4 hours and the changes in GnRH were measured. Mice were injected intracerebroventricularly with IGF2 (15 ng/g, 5 μL) or with the same dose of phosphate buffered saline (PBS), after eight hours, they were euthanized and tissues collected. Results: CPP rats had increased expression of IGF2 and GnRH mRNA and their respective proteins in the preoptic area (POA) of the hypothalamus. Treatment of GT1-7 cells with 10 ng/mL of IGF2 increased GnRH mRNA and protein expression, and GnRH concentration in the culture medium. Injection of IGF2 protein into the lateral ventricle of mice increased the expression of GnRH mRNA and protein in the POA. Conclusions: IGF2 may upregulate the synthesis of GnRH during the pubertal period, and may also take part in the pathology of CPP.
Follicle stimulating hormone receptor expression in human pancreas and effects on insulin secretion: A translational study

Abstract:Follicle-stimulating hormone (FSH) is traditionally known for its role in reproduction, but recent studies suggest it may also influence metabolic processes. This study aimed to examine FSH receptor (FSHR) expression in human pancreatic islets and the direct effects of FSH on insulin secretion, as well as explore FSH's metabolic role during puberty, focusing on enhanced insulin secretion during this critical period. FSHR gene and protein expression were detected in isolated human pancreatic islets and co-localized with insulin-producing beta-cells. Additionally, FSH at prepubertal (0.1 IU/L) and pubertal concentrations (10 IU/L) significantly enhanced glucose-stimulated insulin secretion (GSIS) and increased intracellular cAMP concentrations in intact human pancreatic islets. In children with obesity from the Beta-JUDO cohort (n=608), plasma FSH levels were positively associated with several insulin secretion indices, particularly in pubertal children. These findings suggest that FSH has significant metabolic roles beyond reproduction, involving insulin secretion and potentially contributing to puberty-related hyperinsulinemia and insulin resistance.

Changes in the concentration of follicle-stimulating hormone in plasma during development in the guinea-pig

Abstract:The concentration of FSH in the plasma of guinea-pigs from day 50 of gestation to day 45 of postnatal life was assayed by a radioimmunological procedure utilizing a cross-reaction with the NIAMDD S6 antiserum to rat FSH. At 68 days of gestation the mean plasma FSH concentration of female foetuses was greater than that of the males, although the concentrations in the two sexes were similar on day 50 of gestation. Maternal levels remained consistently low throughout gestation. Postnatally there were no marked changes in plasma FSH levels through to maturity, although a transient rise in the male occurred over the first 3 days after birth. Gonadectomy on days 0, 5, 10, 15, 25 or 35 of postnatal life, or when adult, resulted in a significant increase in plasma levels of FSH within 10 days. The rise in plasma FSH concentration was greater in males than in females at all ages, although, a larger increase was observed in females spayed at 5 or 10 days of age than at other times.

Abstract:The effect of castration and of administration of charcoal-treated porcine follicular fluid (pFF) containing inhibin-like activity on plasma concentration of gonadotropic hormones was studied in neonatal pigs. Plasma follicle-stimulating hormone (FSH) concentration averaged 25.1 +/- 1.5 ng/ml (mean +/- SEM) in 1-wk-old females and gradually declined to 20.2 +/- 0.7 ng/ml 6 wk later. Ovariectomy did not significantly influence plasma FSH concentration. In males, concentration averaged 8.0 +/- 0.7 ng/ml before castration but rose significantly within 2 days after castration. Injection of luteinizing hormone-releasing hormone (LHRH) did not influence plasma FSH concentrations in intact males, but did in females and in 7-wk-old males castrated at 1 wk. Plasma luteinizing hormone (LH) concentrations in 1-wk-old females (2.2 +/- 0.4 ng/ml) gradually declined and were not influenced by castration. Concentrations of plasma LH in 1-wk-old male piglets (2.8 +/- 0.7 ng/ml) were not significantly influenced by castration within 2 days but were significantly higher 6 wk later. LHRH induced a significant rise in plasma LH concentrations in all animals. Injection of pFF resulted in a decline of plasma FSH concentrations in intact and castrated males and in intact females, but did not influence plasma LH concentrations. These data demonstrate a sex-specific difference in the control of plasma FSH, but not in plasma LH concentration in the neonatal pig. Plasma FSH concentrations, but not plasma LH concentrations, are suppressed by testicular hormones in 1-wk-old piglets. Plasma FSH concentrations can be suppressed in both neonatal male and female pigs by injections of pFF.

New warped LPC-Based Feature for Fast and robust speech/Music Discrimination

Abstract:ABSTRACT Automatic discrimination of speech and music is an im-portant tool in many multimedia applications. The pa-per presents a low complexity but effective approach forspeech/music discrimination, which exploits only one sim-ple feature, called Warped LPC-based Spectral Centroid(WLPC-SC). A three-component Gaussian Mixture Model(GMM) classiﬁer is used because it showed a slightly bet-ter performance than other Statistical Pattern Recognition(SPR) classiﬁers. Comparison between WLPC-SC andthe timbral features proposed in [11] is performed, aimingto assess the good discriminatory power of the proposedfeature. Experimental results reveal that our speech/musicdiscriminator is robust and fast, making it suitable for real-time multimedia applications. 1. INTRODUCTION Automatic discrimination between speech and music hasbecome a research topic of interest in the last few years.Several approaches have been described in the recent lit-erature for different applications [1][2] [3][4][5]. Eachof these uses different features and pattern classiﬁcationtechniques and describes results on different material.Saunders [1] proposed a real-time speech/music dis-criminator, which was used to automatically monitor theaudio content of FM audio channels. Four statistical fea-tures on the zero-crossing rate and one energy-related fea-ture were extracted, a multivariate-Gaussian classiﬁer wasapplied, which resulted in an accuracy of 98%.In Automatic Speech Recognition (ASR) of broadcastnews, it’s desirable to disable the input to the speech rec-ognizer during the non-speech portion of the audio stream.Scheirer and Slaney [2] developed a speech/music dis-crimination system for ASR of audio sound tracks. Thir-teen features to characterize distinct properties of speechand music, and three classiﬁcation schemes (MAP Gaus-sian, GMM and k-NN classiﬁers) were exploited, result-ing in an accuracy of over 90%.Another application that can beneﬁt from distinguish-ing speech from music is low bit-rate audio coding. De-signing an universal coder to reproduce well both speechand music is the best approach. However, it is not a trivialproblem. An alternative approach is to design a multi-mode coder that can accommodate different signals. Theappropriate module is selected using the output of a speech-music classiﬁer [6] [7].Automatic discrimination of speech and music is animportant tool in many multimedia applications. KhaledEl-Maleh et al. [3] combined the line spectral frequen-cies and zero-crossings-based features for frame-level nar-rowband speech/music discrimination. The classiﬁcationsystem operates using only a frame delay of 20 ms, mak-ing it suitable for real-time multimedia applications. Anemerging multimedia application is content-based index-ing and retrieval of audiovisual data. Audio content analy-sis is an important task for such application [8]. Minami etal. [9] proposed an audio-based approach to video index-ing, where a speech/music detector is used to help users tobrowse a video database.Comparative view of the value of different types of fea-tures in speech music discrimination is provided in [10],where four types of features (amplitudes, cepstra, pitchand zero-crossings) are compared for discriminating speechand music signals. Experimental results showed cepstraand delta cepstra bring the best performance. Mel Fre-quencies Spectral or Cepstral Coefﬁcients (MFSC or MFCC)are very often used features for audio classiﬁcation tasks,providing quite good results. In [4], MFSC’s ﬁrst orderstatistics are combined with neural networks to form aspeech music classiﬁer that is able to generalize from alittle amount of learning data. MFCC are a compact rep-resentation of the spectrum of an audio signal taking intoaccount the nonlinear human perception of pitch, as de-scribed by the mel scale. They are one of the most usedfeatures in speech recognition and have recently proposedin musical genre classiﬁcation of audio signals [11][12].Unlike the previous works, speech/music discrimina-tion approaches based on only one type of features arepresented in [13] and [5], which result in fast and robustclassiﬁcation systems. The approach in [13] takes psy-choacoustic knowledge into account in that it uses the lowfrequency modulation amplitudes over 20 critical bands toform a good discriminator for the task, while the approachin [5] exploits a new energy-related feature, called mod-iﬁed low energy ratio, that improves the results obtainedwith the classical low energy ratio.In this paper, we present our contribution to the de-sign of a robust speech/music discrimination system. Heterogeneity of pituitary folliculo-stellate cells: implications for interleukin-6 production and accessory function in vitro

Abstract:The population of folliculo-stellate (FS) cells of the rat anterior pituitary has been shown to be ultrastructurally and immunohistochemically heterogeneous. Based on the overlap of ultrastructural characteristics, the localization in the anterior pituitary and the co-expression within the same cel of the S-100 protein (a marker for FS cells) and MHC-class II determinants (an immune marker) we concluded that a partial overlap exists between the population of FS cells and the monocyte-derived dendritic cells (DC). In this report we describe that interleukin-6 (IL-6) immunoreactivity was found in situ in stellate cells of the rat, mouse and human anterior pituitary at a very low density (< 1% of the cells); the topography was reminiscent of the distribution of FS cells. In the present study we also analyse three different pituitary cell separation methods, in order to study the functional heterogeneity of the FS cells in vitro, and to verify whether functionally distinct subpopulations exist within the FS cell group. Production of bioactive IL-6 was measured in conditioned media of rat anterior pituitary cells separated by (i) bovine serum albumin (BSA) gradient sedimentation at 1 g, (ii) Nycodenz gradient and (iii) a magnetic cell separation (MACS) technique. Production of bioactive IL-6 by cell cultures of 1 to 4 days was correlated with the proportional number of S100 immunoreactive and S100 producing cells, but was not correlated with the proportional number of MHC-class II expressing (OX6-positive) dendritic cells (DC). The distribution pattern of OX6-positive DC was found to partly overlap with the distribution pattern of S100-positive cells in the BSA gradient. Co-sedimentation of S100-positive FS cells and MHC-class II-expressing DC was not restricted to the top fractions of the BSA gradient, but was also found in the low density Nycodenz fraction. MACS separation of the rat anterior pituitary cells resulted into a population enriched in OX6 and OX62 positive DC and a population devoid of such cells, while S100+ cells were equally divided into these two subpopulations. Although there was a significantly decreased production of IL-6 as compared to that of an original pituitary cell population, both MACS separated populations were equal in IL-6 production. The diminution in IL-6 production in both populations may be the result of an impediment of paracrine communication due to the MACS separation into these two populations. Our data also show that a subpopulation of FS cells was capable of stimulating T cell proliferation in vitro. Concomitantly with the distribution pattern of S100- and OX6-immunoreactive cells in the BSA and Nycodenz gradient fractions, we found a similar pattern of stimulation of T cell proliferation. Unlike the IL-6 production pattern, the T cell stimulating capacity was present in the MHC-class II-enriched cell population but absent in the MHC-class II-depleted cell population. These findings-together with earlier in situ histochemical data-suggest that there is an OX6+ S100- subpopulation of FS cells in the anterior pituitary that in itself is capable of stimulating T cell proliferation in vitro, and acts as lymphoid DC. There is also an S100+ OX6- population that is unable to stimulate T cell proliferation in vitro. Both populations are able to produce IL-6, but probably need stimuli from other subpopulations of pituitary cells (or exogeneous stimuli) to produce maximal amounts of IL-6.
A new role for follicle-stimulating hormone in the regulation of calcium flux in Sertoli cells: inhibition of Na+/Ca++ exchange

Abstract:Elucidation of mechanisms regulating intracellular calcium levels in steroidogenic tissues is important for understanding control of cellular function. We have previously described FSH receptor-mediated flux of 45Ca++ into cultured rat Sertoli cells and receptor-enriched proteoliposomes via voltage-sensitive and voltage-independent calcium channels. In the present study, we report heretofore unrecognized inhibitory effects of FSH on Na+/Ca++ exchange in these two systems. An outwardly directed Na+ gradient, developed by preincubating Sertoli cell monolayers in buffer made hypertonic with NaCl, resulted in uptake of 45Ca++ that was unaffected by calcium channel blocking agents, ruthenium red or methoxyverapamil, but was enhanced by ouabain, a specific inhibitor of Na+/K(+)-ATPase. Sodium-dependent 45Ca++ flux into Sertoli cells was inhibited in a concentration-related manner by increased extracellular Na+ (up to 135 mM). FSH consistently and reproducibly (28.9 +/- 3.8%, 10 separate assays) reduced sodium-dependent 45Ca++ influx in the absence or presence of ouabain. A lesser effect on Na+/Ca++ exchange was seen when Li+ replaced Na+ in the preincubation buffer, and a marked reduction occurred when Sertoli cells were incubated in buffer containing KCl, presumably due to membrane depolarization. FSH-sensitive Na+/45Ca++ exchange was also observed when using FSH receptor-enriched proteoliposomes. Our earlier calcium channel studies indicated that FSH affects Ca++ entry into Sertoli cells via a receptor-mediated process. The results reported here demonstrate that the interaction of FSH with its receptor is associated with changes in Na+/Ca++ exchange as well, and suggest that this activity may also be involved in regulating intracellular free Ca++ levels in the Sertoli cell.

Abstract:The mammalian pituitary gland drives highly conserved physiological processes such as somatic cell growth, pubertal transformation, fertility, and metabolism by secreting a variety of hormones. Recently, single-cell transcriptomics techniques have been used in pituitary gland research. However, more studies have focused on adult pituitary gland tissues from different species or different sexes, and no research has yet resolved cellular differences in pituitary gland tissue before and after sexual maturation. Here, we identified a total of 15 cell clusters and constructed single-cell transcriptional profiles of rats before and after sexual maturation. Furthermore, focusing on the gonadotrope cluster, 106 genes were found to be differentially expressed before and after sexual maturation. It was verified that Spp1, which is specifically expressed in gonadotrope cells, could serve as a novel marker for this cell cluster and has a promotional effect on the synthesis and secretion of follicle-stimulating hormone. The results provide a new resource for further resolving the regulatory mechanism of pituitary gland development and pituitary hormone synthesis and secretion.

Corticotropin-Releasing Hormone: A Novel Stimulator of Somatolactin in Teleost Pituitary Cells

Abstract:Corticotropin-releasing hormone (CRH) is known for its crucial role in the stress response system, which could induce pituitary adrenocorticotropic hormone (ACTH) secretion to promote glucocorticoid release in the adrenal gland. However, little is known about other pituitary actions of CRH in teleosts. Somatolactin is a fish-specific hormone released from the neurointermediate lobe (NIL) of the posterior pituitary. A previous study has reported that ACTH was also located in the pituitary NIL region. Interestingly, our present study found that CRH could significantly induce two somatolactin isoforms’ (SLα and SLβ) secretion and synthesis in primary cultured grass carp pituitary cells. Pharmacological analysis further demonstrated that CRH-induced pituitary somatolactin expression was mediated by the AC/cAMP/PKA, PLC/IP3/PKC, and Ca2+/CaM/CaMK-II pathways. Finally, transcriptomic analysis showed that both SLα and SLβ should play an important role in the regulation of lipid metabolism in primary cultured hepatocytes. These results indicate that CRH is a novel stimulator of somatolactins in teleost pituitary cells, and somatolactins may participate in the stress response by regulating energy metabolism.

Immunocytochemical evidence for the presence of S-100 protein in insulin-containing cells of cultured fetal rat islets

Abstract:S-100 protein was long considered to be specific to glial and Schwann cells, but was subsequently proved to be present in various organs. In particular, S-100 proteinimmunoreactivity was demonstrated in the parathyroid gland, adenohypophysis and endocrine pancreas. In the present study cultured fetal rat islets were investigated in view of the possible presence of S-100 protein immunoreactivity in their cells. In the initial 5-day period, continuity between islets and ducts could be demonstrated, and the islets appeared to bud from the ducts. During this time, S-100 protein-immunoreactive cells were found in either the budding islets or ducts. The colocalization of S-100 protein and insulin was demonstrated immunocytochemically. In contrast, the newly formed islets from endocrine monolayers did not display S-100 protein immunoreactivity. After this initial period, numerous free-floating islets were observed, but only some of them contained S-100 protein immunoreactivity. S-100 protein-immunoreactive cells had the same distribution as those storing insulin, again suggesting the coexistence of the two peptides. The results suggest that S-100 protein might be involved in the regulation of islet function.
Diagnosis and management of congenital hypopituitarism in children

Abstract:Hypopituitarism (or pituitary deficiency) is a rare disease with an estimated prevalence of between 1/16,000 and 1/26,000 individuals, defined by insufficient production of one or several anterior pituitary hormones (growth hormone [GH], thyroid-stimulating hormone [TSH], adrenocorticotropic hormone [ACTH], luteinizing hormone [LH], follicle-stimulating hormone [FSH], prolactin), in association or not with diabetes insipidus (antidiuretic hormone [ADH] deficiency). While in adults hypopituitarism is mostly an acquired disease (tumors, irradiation), in children it is most often a congenital condition, due to abnormal pituitary development. Clinical symptoms vary considerably from isolated to combined deficiencies and between syndromic and non-syndromic forms. Early signs are non-specific but should not be overlooked. Diagnosis is based on a combination of clinical, laboratory (testing of all hormonal axes), imaging (brain magnetic resonance imaging [MRI] with thin slices centered on the hypothalamic-pituitary region), and genetic (next-generation sequencing of genes involved in pituitary development, array-based comparative genomic hybridization, and/or genomic analysis) findings. Early brain MRI is crucial in neonates or in cases of severe hormone deficiency for differential diagnosis and to inform syndrome workup. This article presents recommendations for hormone replacement therapy for each of the respective deficient axes. Lifelong follow-up with an endocrinologist is required, including in adulthood, with multidisciplinary management for patients with syndromic forms or comorbidities. Treatment objectives include alleviating symptoms, preventing comorbidities and acute complications, and optimal social and educational integration.

Abstract:The problems of health and development faced by contemporary adolescents require that our communities have greater access to effective, adequately scaled, and sustained youth-serving programs. These 3 dimensions of youth programs are discussed, and the role of evaluations is specified, especially those aimed at program improvement and community empowerment, in facilitating efforts to promote the positive development of young people. These areas of development include competence, confidence, character, connection, and caring/compassion-the "five C's" of positive youth development. Ten features of effective positive youth development programs are described, and the importance of engaging policymakers in order to broadly disseminate and sustain effective programs is stressed.

Abstract:The pituitary gland, often called the "master gland", orchestrates multiple effector hormonal organs and other glands by secreting various tropic hormones, which play a significant role in a myriad of physiological processes including skeletal modeling and remodeling, fat and glucose metabolism, and cognitive and psychological processes. The findings of the expression of receptors for each pituitary hormone and the hormone itself in skeleton, fat and immune cells suggested that their role is much broader than the traditional or classic role. Follicle-stimulating hormone (FSH), once believed to regulate gonadal function – gonadal development and maturation at puberty and gamete production during the fertile phase – is also found to involve in fat and bone metabolism as well as cognition, which provides us a better understanding of complex physiology. This emerging understanding of the non-reproductive role of FSH opens potential therapeutic opportunity to address detrimental health burden during and after menopause, namely osteoporosis, obesity and dementia. In this Review, we outline the current understanding of crosstalk between the pituitary, bone, adipose tissue and brain through FSH. The pre-clinical evidence from genetic and pharmacologic intervention in rodent models, and human data from population-based observation, genetic studies, and a small number of studies with interventional nature support an independent skeletal, lipogenic and cognitive effect of FSH and more.

Abstract:BACKGROUND: Growth hormone-secreting pituitary adenomas (somatotroph adenoma) predominantly express somatostatin receptors (SSTRs) subtypes 2 and 5. Higher SSTR2 expression on somatotroph adenomas results in a better response to somatostatin analogues (SSAs), which preferentially bind, but also downregulate, SSTR2. The effect of the combined treatment with SSAs and the GH receptor antagonist pegvisomant (PEGV) on SSTR expression in somatotroph adenomas is currently unknown.AIM OF THE STUDY: To assess SSTR2 and SSTR5 expression in three groups of somatotroph adenomas: drug-naive, treated with long-acting (LA) SSA monotherapy, or LA-SSA/PEGV combination therapy before surgery. Additionally, we evaluated the required PEGV dose to achieve insulin-like growth factor I (IGF-I) normalization in relation to the SSTR expression.MATERIALS AND METHODS: At our Pituitary Center Rotterdam, we selected acromegalic patients who underwent transsphenoidal neurosurgery. All patients were eventually treated with LA-SSA/PEGV combination therapy during their medical history. SSTR2 and SSTR5 expression in somatotroph adenoma tissues was determined using immunohistochemistry.RESULTS: Out of 39 somatotroph adenoma tissue samples, 23 were drug-naive, 9 received pretreatment with LA-SSA and 7 LA-SSA/PEGV combined treatment. SSTR2 expression was significantly higher in treatment-naive compared to combined treatment somatotroph adenomas (p = 0.048), while SSTR5 expression did not differ. Noteworthy, SSTR2 expression in naive somatotroph adenoma tissues was inversely correlated with the required PEGV dose to achieve IGF-I normalization during postsurgical medical treatment (ρ = -0.538, p = 0.024).CONCLUSION: In our specific cohort, the SSTR2 expression was lower in patients pretreated with LA-SSA/PEGV compared to the drug-naive acromegalic patients. Additionally, the SSTR2 expression in treatment-naive somatotroph adenoma tissues was inversely correlated with the required PEGV dose to achieve IGF-I normalization.

Abstract:The somatomedins, also referred to as insulin-like growth factors, are a family of peptides whose serum concentrations are regulated principally by growth hormone and nutrient status. Evidence is now emerging that most or all of the biologic effects of the somatomedins can be attributed to two peptides, somatomedin-C/insulin-like growth factor I (Sm-C/IGF-I) and IGF-II (Van Wyk 1984). Sm-C/IGF-I has a molecular weight of 7649 and a pI of 8.0–8.7. It contains 70 amino acid residues in a single chain with 3 disulfide bridges. It is highly growth hormone dependent, and has potent growth-promoting activity in many in vitro systems (Zapf et al. 1984). IGF-II is also a single-chain peptide, which has a more nearly neutral pI and a molecular weight of 7471. IGF-II is less growth hormone dependent and appears to have less growth-promoting activity than Sm-C/IGF-I. Both Sm-C/IGF-I and IGF-II possess nearly 50% homology with proinsulin in regions of the molecule that correspond to the A and B chains of insulin. As in humans, two forms of somatomedin have been isolated from rat plasma. One of these, multiplication-stimulating activity (MSA) is the rat homolog of human IGF-II, differing from human IGF-II by only five amino acid residues (Marquardt et al. 1981). Although not certain, it is expected that other species will be found to have two somatomedins, similar to those in humans and rats. The term somatomedin-A has been applied to a substance that now appears to be a deamidated form of Sm-C/IGF-I, and the peptide referred to as somatomedin-B proved to be a fragment of a plasma-spreading factor contaminated with epidermal growth factor (EGF) (Heldin et al. 1981; Barnes et al. 1984).

Abstract:Introduction The catecholaminergic component of the brain-pituitary-gonadal axis, which mediates the influence of external and internal stimuli on the central nervous system and gonad development in vertebrates, is largely unexplored in Chondrichthyes. We considered Scyliorhinus canicula (L., 1758) females as a model for this vertebrate's class, to assess the involvement of the catecholaminergic system of the brain in its reproduction. Along the S. canicula reproductive cycle, we characterized and evaluated differences in somata morphometry and the number of putative catecholaminergic neurons in two brain nuclei: the periventricular preoptic nucleus, hypothesized to be a positive control for ovarian development, and the suprachiasmatic nucleus, examined as a negative control. Materials and methods 16 S. canicula wild females were sampled and grouped in maturity stages (immature, maturing, mature, and mature egg-laying). The ovary was histologically processed for the qualitative description of maturity stages. Anti-tyrosine hydroxylase immunofluorescence was performed on the diencephalic brain sections. The immunoreactive somata were investigated for morphometry and counted using the optical fractionator method, throughout the confocal microscopy. Results and discussions Qualitative and quantitative research confirmed two separate populations of immunoreactive neurons. The modifications detected in the preoptic nucleus revealed that somata were more numerous, significantly smaller in size, and more excitable during the maturing phase but decreased, becoming slightly bigger and less excitable in the egg-laying stage. This may indicate that the catecholaminergic preoptic nucleus is involved in the control of reproduction, regulating both the onset of puberty and the imminent spawning. In contrast, somata in the suprachiasmatic nucleus grew in size and underwent turnover in morphometry, increasing the total number from the immature-virgin to maturing stage, with similar values in the more advanced maturity stages. These changes were not linked to a reproductive role. These findings provide new valuable information on Chondrichthyes, suggesting the existence of an additional brain system implicated in the integration of internal and environmental cues for reproduction.

Identification of somatostatin-14 and -28 in rat pancreatic juice by a new HPLC method

Abstract:Recently we could demonstrate that in rats with pancreatic hypertrophy, somatostatin is secreted in higher concentrations into the pancreatic juice than into the portal vein blood. For measurement of juice somatostatin and to characterize the molecular forms, we established a new reverse-phase HPLC method, which we describe herein. This HPLC method, using a linear gradient system consisting of 0.2% heptafluorbutyric acid in 10 mM sodium acetate and acetonitrile, showed a stable recovery rate of about 85%. Applying the pure juice to this gradient system, we detect somatostatin-14 to be the major form of immunoreactive somatostatin (IRSS) in the pancreatic juice of the rat (5% of total IRSS). The remaining 35% were found to be somatostatin-28. The role of somatostatin in pancreatic juice is not known. It raises the hypothesis that it possibly interacts with the influences intraluminal intestinal growth factors. This study supports the assumption for the existence of an insuloacinar portal system to regulate exocrine pancreatic functions by islet hormones.
