Abstract:The photosynthetic oxygen-evolving center (OEC) is a unique Mn4 CaO5 cluster that serves as a blueprint to develop superior water-splitting catalysts for the generation of solar fuels in artificial photosynthesis. It is a great challenge and long-standing issue to reveal the synthesizing mechanism of this Mn4 CaO5 cluster in both natural and artificial photosynthesis. Herein, efforts were made to reveal the synthesizing mechanism of an artificial Mn4 CaO4 cluster, a close mimic of the OEC. Four key intermediates were successfully isolated and structurally characterized for the first time. It was demonstrated that the Mn4 CaO4 cluster could be formed through a reaction between a thermodynamically stable Mn3 CaO4 cluster and an unusual four-coordinated MnIII ion, followed by stabilization process through binding an organic base (e.g., pyridine) on the "dangling" Mn ion. These findings shed new light on the synthesizing mechanism of the OEC and rational design of new artificial water-splitting catalysts.

Abstract:The photosynthetic oxygen‐evolving center (OEC) is a unique Mn4CaO5‐cluster that catalyzes water splitting into electrons, protons, and dioxygen. Precisely structural and functional mimicking of the OEC is a long‐standing challenge and pressingly needed for understanding the structure‐function relationship and catalytic mechanism of O‐O bond formation. Herein we report two simple and robust artificial Mn3Ce2O5‐complexes that display a remarkable structural similarity to the OEC in regarding of the ten‐atom core (five metal ions and five oxygen bridges) and the alkyl carboxylate peripheral ligands. This Mn3Ce2O5‐cluster can catalyze the water‐splitting reaction on the surface of ITO electrode. These results clearly show that cerium can structurally and functionally replace both calcium and manganese in the cluster. Mass spectroscopic measurements demonstrate that the oxide bridges in the cluster are exchangeable and can be rapidly replaced by the isotopic oxygen of H218O in acetonitrile solution, which supports that the oxide bridge(s) may serve as the active site for the formation of O‐O bond during the water‐splitting reaction. These results would contribute to our understanding of the structure‐reactivity relationship of both natural and artificial clusters and shed new light on the development of efficient water‐splitting catalysts in artificial photosynthesis.

Abstract:The understanding of the structure-function relationship of the oxygen-evolving center (OEC), a Mn4Ca-cluster, in photosystem II is impeded mainly due to the complexity of the protein environment and lack of rational chemical models as a reference. In this study, two novel Mn4-oxido complexes have been synthesized and characterized, in which the peripheral ligands of the [MnIII4] core are provided by eight μ2-carboxylate groups and two neutral terminal ligands (pyridine or isoquinoline). This type of peripheral ligation is very similar to the Mn4Ca-oxide model complexes recently reported to mimic the OEC. The new Mn4-oxide complex can catalyze the oxygen-evolving reaction in the presence of ButOOH as an oxidant. The structure and redox properties comparison of the Mn4-oxido and Mn4Ca-oxido complexes provide important clues to understanding the functional role of Ca in the OEC in natural photosynthesis, and develop more efficient artificial catalysts for the water-splitting reaction in the future.

Abstract:The oxygen-evolving center (OEC) in photosynthesis is a unique biological Mn4CaO5 cluster catalyzing the water-splitting reaction. A great current challenge is to achieve a robust and precise mimic of the OEC in the laboratory. Herein, we report synthetic Mn4XO4 clusters (X = calcium, yttrium, gadolinium) that closely resemble the OEC with regard to the main metal-oxide core and peripheral ligands, as well as the oxidation states of the four Mn ions and the redox potential of the cluster. We demonstrate that rare-earth elements can structurally replace the calcium in neutral Mn4XO4 clusters. All three Mn4XO4 clusters with different redox-inactive metal ions display essentially the same redox properties, challenging the conventional view that the Lewis acidity of the redox-inactive metal ions could modulate the redox potential of the heteronuclear-oxide clusters. Abstract:Photosynthetic water oxidation is catalyzed by a manganese-calcium oxide cluster, which experiences five "S-states" during a light-driven reaction cycle. The unique "distorted chair"-like geometry of the Mn4CaO5(6) cluster shows structural flexibility that has been frequently proposed to involve "open" and "closed"-cubane forms from the S1 to S3 states. The isomers are interconvertible in the S1 and S2 states, while in the S3 state, the open-cubane structure is observed to dominate inThermosynechococcus elongatus (cyanobacteria) samples. In this work, using density functional theory calculations, we go beyond the S3+Yz state to the S3nYz• → S4+Yz step, and report for the first time that the reversible isomerism, which is suppressed in the S3+Yz state, is fully recovered in the ensuing S3nYz• state due to the proton release from a manganese-bound water ligand. The altered coordination strength of the manganese-ligand facilitates formation of the closed-cubane form, in a dynamic equilibrium with the open-cubane form. This tautomerism immediately preceding dioxygen formation may constitute the rate limiting step for O2 formation, and exert a significant influence on the water oxidation mechanism in photosystem II.

Insights from Ca2+→Sr2+ substitution on the mechanism of O-O bond formation in photosystem II

Abstract:In recent years, there has been a steady interest in unraveling the intricate mechanistic details of water oxidation mechanism in photosynthesis. Despite the substantial progress made over several decades, a comprehensive understanding of the precise kinetics underlying O-O bond formation and subsequent evolution remains elusive. However, it is well-established that the oxygen evolving complex (OEC), specifically the CaMn4O5 cluster, plays a crucial role in O-O bond formation, undergoing a series of four oxidative events as it progresses through the S-states of the Kok cycle. To gain further insights into the OEC, researchers have explored the substitution of the Ca2+ cofactor with strontium (Sr), the sole atomic replacement capable of retaining oxygen-evolving activity. Empirical investigations utilizing spectroscopic techniques such as XAS, XRD, EPR, FTIR, and XANES have been conducted to probe the structural consequences of Ca2+→Sr2+ substitution. In parallel, the development of DFT and QM/MM computational models has explored different oxidation and protonation states, as well as variations in ligand coordination at the catalytic center involving amino acid residues. In this review, we critically evaluate and integrate these computational and spectroscopic approaches, focusing on the structural and mechanistic implications of Ca2+→Sr2+ substitution in PS II. We contribute DFT modelling and simulate EXAFS Fourier transforms of Sr-substituted OEC, analyzing promising structures of the S3 state. Through the combination of computational modeling and spectroscopic investigations, valuable insights have been gained, developing a deeper understanding of the photosynthetic process.

Abstract:The low activation barrier for O‐O coupling in the closed‐cubane Oxygen‐Evolving Centre (OEC) of Photosystem II (PSII) requires water coordination with the Mn4 'dangler' ion in the Mn(V)‐oxo fragment. This coordination transforms the Mn(V)‐oxo into a more reactive Mn4(IV)‐oxyl species, enhancing O‐O coupling. This study explains the mechanism behind this and indicates that in the most stable form of the OEC, the Mn4 fragment adopts a trigonal bipyramidal geometry but needs to transition to a square pyramidal form to be activated for O‐O coupling. This transition stabilizes the Mn4 dxy orbital, enabling electron transfer from the oxo ligand to the dxy orbital, converting the oxo ligand into an oxyl species. The role of the water is to coordinate with the square pyramidal structure, reducing the energy gap between the oxo and oxyl forms, thereby lowering the activation energy for O‐O coupling. This mechanism applies not only to the OEC system but also to other Mn(V)‐based catalysts. For other catalysts, ligands like OH‐ stabilize the Mn(IV)‐oxyl species better than water, improving catalyst activation for reactions like C‐H bond activation. This study is the first to explain the Mn(V)‐oxo to Mn(IV)‐oxyl conversion, providing new foundation for Mn‐based catalyst design.

Abstract:Inspecting S states in photosynthesis Oxygenic photosynthesis uses a Mn 4 CaO 5 cluster in the oxygen-evolving complex to extract electrons from water and produce dioxygen. Visualizing each of the chemical states in this process, S 0 to S 4 , and assigning chemical identities and mechanisms on the basis of structures has been a challenge addressed recently by work at x-ray free-electron lasers. Suga et al. used serial crystallography at cryogenic temperatures to trap and determine the structures of several stable states during photosystem II water oxidation (see the Perspective by Britt and Marchiori). Changes around the water cluster already happen in the S 2 state and set the stage for water insertion that occurs during transition to the S 3 state. A short 1.9-angstrom distance between the two oxygen atoms in the S 3 state is consistent with theoretical studies supporting an oxyl/oxo mechanism for oxygen-oxygen coupling. Abstract:In Nature, the four-electron reduction of O2 is catalyzed at preorganized multimetallic active sites. These complex active sites often feature low-coordinate, redox-active metal centers precisely positioned to facilitate rapid O2 activation processes that obviate the generation of toxic, partially-reduced oxygen species. Very few biomimetic constructs simultaneously recapitu-late the complexity and reactivity of these biological cofactors. Herein, we report solid-state O2 activation at a triiron(II) active site templated by phosphinimide ligands. Insight into the structure of the O2 reduction intermediates was obtained via in crystallo O2 dosing experiments in conjunction with spectroscopic, structural, magnetic, and computational studies. These data support the in situ formation of an Fe2IIIFeIV-dioxo intermediate upon O2 exposure that participates in oxygen atom and hydrogen atom transfer reactivity with exogenous substrates to furnish a stable FeIIFe2III-oxo species. Combined, these stud-ies provide an extraordinary level of detail into the dynamics of bond forming and breaking processes operative at complex multimetallic active sites.

Computational insights into the O2-evolving complex of photosystem II

Abstract:Mechanistic investigations of the water-splitting reaction of the oxygen-evolving complex (OEC) of photosystem II (PSII) are fundamentally informed by structural studies. Many physical techniques have provided important insights into the OEC structure and function, including X-ray diffraction (XRD) and extended X-ray absorption fine structure (EXAFS) spectroscopy as well as mass spectrometry (MS), electron paramagnetic resonance (EPR) spectroscopy, and Fourier transform infrared spectroscopy applied in conjunction with mutagenesis studies. However, experimental studies have yet to yield consensus as to the exact configuration of the catalytic metal cluster and its ligation scheme. Computational modeling studies, including density functional (DFT) theory combined with quantum mechanics/molecular mechanics (QM/MM) hybrid methods for explicitly including the influence of the surrounding protein, have proposed chemically satisfactory models of the fully ligated OEC within PSII that are maximally consistent with experimental results. The inorganic core of these models is similar to the crystallographic model upon which they were based, but comprises important modifications due to structural refinement, hydration, and proteinaceous ligation which improve agreement with a wide range of experimental data. The computational models are useful for rationalizing spectroscopic and crystallographic results and for building a complete structure-based mechanism of water-splitting in PSII as described by the intermediate oxidation states of the OEC. This review summarizes these recent advances in QM/MM modeling of PSII within the context of recent experimental studies.
Uncovering quantum characteristics of incipient evolutions at the photosynthetic oxygen evolving complex

Abstract:Water oxidation of photosynthesis at the oxygen evolving complex (OEC) is driven by the polarization field induced by the photoelectric hole. By highlighting the role of the polarization field in reshaping the spin and orbit potentials, we reveal in this work the characteristics and underlying mechanism in the relatively simpler OEC evolutions within the states S0 - S2 prior to the water oxidation. The characteristic shifts of the density of states (DOS) of the electron donor Mn atom are observed in the vicinity of the Fermi surface to occur with the spin flips of Mn atoms and the change of the Mn oxidation states during the electron transfer. Notably, the spin flips of Mn atoms point to the resulting spin configuration of the next states. It is found that the electron transfer tend to stabilize the catalyst OEC itself, whereas the proton transfer pushes the evolution forward by preparing a new electron donor, demonstrating the proton-coupled electron transfer. Meanwhile, it shows that the Mn-O bonds around the candidate Mn atom of the electron donor undergo characteristic changes in the bond lengths during the electron transfer. These concomitant phenomena uncovered in first-principle calculations characterize the essential equilibrium of the OEC between the state evolution and stability that forms a ground of the dynamic OEC cycles. In particular, the characteristic undulation of the DOS around the Fermi level occurring at the proton-coupled electron transfer can be used to reveal crucial processes in a wide range of realistic systems.

Abstract:At the initiation of lactation, Ca homeostatic mechanisms have to react to a tremendous increase in demand for Ca. Mobilization of Ca from bone and increased absorption from the gastrointestinal tract are required to re-establish homeostasis. It has been shown that dietary anions play an important role in the prevention of milk fever by mobilizing Ca from bone and by increasing Ca absorption in the GI tract. The purpose of the present study was to investigate the influence of different Ca contents in diets supplemented with anionic salts on bone metabolism of dairy cows. Twenty-four holstein cows (housed inside, second to fourth lactation) without a milk fever history were divided into four groups (A, B, C, D). Each group was fed a different diet which was given from day 263 of gestation till the day of parturition. Group A and B received a low calcium diet (4 g/kg DM) whereas group C and D received a high Ca diet (8 g/kg DM). In addition group B and D received anionic salts. The DCAD was calculated with the formula: DCAD (mEq/kg DM)=(0.2 Ca2++0.16 Mg2++Na++K+)-(Cl-+0.6 S2-+0.65 P3-). Blood and urine samples were collected on days 256, 270 and 277 of gestation, on the day of parturition as well as the following 5 days and on days 9, 14 and 19 after parturition. Serum Ca, P, Mg, ICTP, OC, VITD, PTH and urinary pH were analysed. The bone resorption marker ICTP showed a significant increase after parturition in all the groups. On the contrary, the bone formation marker OC decreased after parturition in all the groups. The VITD concentrations in group D and the urinary pH in group B were significantly lower compared to the other groups (p<0.05). The Ca concentrations tended to be higher in group B around parturition than in all the other groups. No significant influence of the four different diets on all the other parameters could be shown. In conclusion, this data showed that the addition of anions and the different Ca contents had no significant influence on bone resorption and bone formation markers. This may be because of the fact that the dietary cation-anion balance was not low enough (DCAD-group A: 181 mEq/kg DM, group B: -48 mEq/kg DM, group C: 210 mEq/kg DM and group D: 28 mEq/kg DM) to induce a metabolic acidosis with all its positive effects on calcium metabolism.

Abstract:Water oxidation at the oxygen-evolving complex (OEC) of the photosystem II is catalyzed by the core cluster CaMn4O5 , which has been projected to experience five intermediate states Si in the Kok's cycle since the 1970s. However, the detailed dynamics of state evolutions still remains unclear, albeit with the general fact that the process is initiated by the transfer of photoelectrons with the steady electron donors of the water molecules. Based on density-functional simulations, we find that the spin flips of manganese atoms between the consecutive states in the electric polarization field can be used as a marker to uncover the intricate dynamic evolutions and the underlying dynamics. The dynamic electron and proton transfers and water insertion and dissociation are traced to reveal the evolution pathways of S0–S3 with commensurate spin flips towards the exact spin configuration of the next state. In particular, the various water insertions and dissociations at coordination sites of the S2 open and closed cubane isomers are predicted with constraints on the necessary spin flips. Abstract:From atomic level to understand the cluster-size-dependant behavior of dye-sensitized photocatalysts is very important and helpful to design new photocatalytic materials. Although the relationship between the photocatalytic behaviors and particles' size/shape has been widely investigated by theoretical scientists, the experimental evidences are much less. In this manuscript, we successfully synthesized three new ruthenium dye-sensitized polyoxometalates (POM-n, n relate to different size clusters) with different-sized POM clusters. Under visible-light illumination, all three complexes show the stable O2 evolution with the efficient order POM-3 > POM-2 > POM-1. This cluster-size-dependent catalytic behavior could be explained by the different numbers of M = Ot (terminal oxygen) bonds in each individual cluster because it is well-known that Mo = Ot groups are the catalytically active sites for photooxidation reaction. The proposed mechanism of water oxidation for the dye-sensitized POMs is radical reaction process. This research could open up new perspectives for developing new POM-based WOCs.
Dynamic evolution of S$_0$-S$_3$ at the oxygen evolving complex with spin markers under photoelectric polarization

Abstract:Water oxidation at the oxygen evolving complex (OEC) of the photosystem II is catalyzed by the core cluster CaMn$_4$O$_5$ which was projected to experience five intermediate states S$\rm_i$ in the Kok's cycle since 1970's. However, the detailed dynamics of state evolutions still remains unclear, albeit with the general fact that the process is initiated by the transfer of photoelectrons with the steady electron donors of the water molecules. Based on density functional simulations, we find that the spin flips of Manganese atoms between the consecutive states in the electric polarization field can be used as a marker to uncover the intricate dynamic evolutions and the underlying dynamics. The dynamic electron and proton transfers and water insertion and dissociation are traced to reveal the evolution pathways of S$_0$-S$_3$ with commensurate spin flips towards the exact spin configuration of the next state. In particular, the various water insertions and dissociations at coordination sites of the S$_2$ open and closed cubane isomers are predicted with constraints on the necessary spin flips. Our study paves a way to uncover the animated OEC evolutions with the spin flips that await for more experimental verifications and lays a solid ground for revealing the mechanism of dioxygen generation via the pending S$_4$ state.
