Abstract:Objective: To explore the protective effect of coenzyme Q9 against H2O2-induced oxidative damage in human vascular endothelial cells.Methods: An HUVEC(human umbilical vein endothelial cells) model of H2O2-induced oxidative damage was created by MTT assay and used to study the preventive effect and its mechanism of coenzyme Q9 against H2O2-induced oxidative damage.Results: Coenzyme Q9 at 100 μg/mL could exert preventive and repairing effects against H2O2-induced oxidative damage in HUVEC cells.The mechanism was partially due to that fact that the generation of ROS and MDA was inhibited by coenzyme Q9.Meanwhile,preventive treatment with coenzyme Q9 could effectively protect SOD against H2O2-induced damage but had no repairing effect.Conclusion: Coenzyme Q9 has protective and repairing effects against oxidative damage in human vascular endothelial cells.

Abstract:Background Diabetic retinopathy is a major complication of dysregulated hyperglycemia. Retinal vascular endothelial cell dysfunction is an early event in the pathogenesis of diabetic retinopathy. Studies showed that hyperglycemia-induced excess proliferation of retinal vascular endothelial cells can be abrogated by docosahexaenoic acid (DHA, 22:6 ω-3) and eicosapentaenoic acid (EPA, 20:5 ω-3). The influence of dietary omega-3 PUFA on brain zinc metabolism has been previously implied. Zn 2+ is essential for the activity of Δ 6 desaturase as a co-factor that, in turn, converts essential fatty acids to their respective long chain metabolites. Whether essential fatty acids (EFAs) α-linolenic acid and linoleic acid have similar beneficial effect remains poorly understood. Methods RF/6A cells were treated with different concentrations of high glucose, α-linolenic acid and linoleic acid and Zn 2+ . The alterations in mitochondrial succinate dehydrogenase enzyme activity, cell membrane fluidity, reactive oxygen species generation, SOD enzyme and vascular endothelial growth factor (VEGF) secretion were evaluated. Results Studies showed that hyperglycemia-induced excess proliferation of retinal vascular endothelial cells can be abrogated by both linoleic acid (LA) and α-linolenic acid (ALA), while the saturated fatty acid, palmitic acid was ineffective. A dose–response study with ALA showed that the activity of the mitochondrial succinate dehydrogenase enzyme was suppressed at all concentrations of glucose tested to a significant degree. High glucose enhanced fluorescence polarization and microviscocity reverted to normal by treatment with Zn 2+ and ALA. ALA was more potent that Zn 2+ . Increased level of high glucose caused slightly increased ROS generation that correlated with corresponding decrease in SOD activity. ALA suppressed ROS generation to a significant degree in a dose dependent fashion and raised SOD activity significantly. ALA suppressed high-glucose-induced VEGF secretion by RF/6A cells. Conclusions These results suggest that EFAs such as ALA and LA may have beneficial action in the prevention of high glucose-induced cellular damage.
Buyang Huanwu Decoction Attenuates H2O2-induced Apoptosis by Inhibiting Reactive Oxygen Species-Mediated Mitochondrial Dysfunction Pathway in Human Umbilical Vein Endothelial Cells

Abstract:Background: Apoptosis of endothelial cells caused by reactive oxygen species plays an important role in ischemia/reperfusion injury after cerebral infarction. Buyang Huanwu Decoction (BYHWD) has been used to treat stroke and stroke-induced disability, however, the mechanism for this treatment remains unknown. In this study, we investigated whether BYHWD can protect human umbilical vein endothelial cells (HUVECs) from H2O2-induced apoptosis and explored the underlying mechanisms.Methods: To investigate the effect of BYHWD on the apoptosis of HUVECs, we established a H2O2-induced oxidative stress model and detected apoptosis by Hoechst 33342 and propidium iodide staining. JC-1 and DCFH-DA assays, western blotting and electron microscopy were used to examine the mechanism of BYHWD on apoptosis.Results: Pretreatment with BYHWD significantly inhibited H2O2-induced apoptosis and protein caspase-3 expression in a concentration-dependent manner. In addition, BYHWD reduced reactive oxygen species production and promoted endogenous antioxidant defenses. Furthermore, loss of mitochondrial membrane potential and structural disruption of mitochondria were both rescued by BYHWD.Conclusions: BYHWD protects HUVECs from H2O2-induced apoptosis by inhibiting oxidative stress damage and mitochondrial dysfunction. These findings indicate that BYHWD is a promising treatment for cerebral ischemia diseases.
[Resveratrol Protects Against Hydrogen Peroxide Induced Injury in Human Umbilical Vein Endothelial Cells].

Abstract:OBJECTIVE:To investigate the effect of Resveratrol on Hydrogen peroxide-treated human umbilical vein endothelial cells (HUVEC).METHODS:Oxidative stress in HUVEC was induced by hydrogen peroxide (H2O2). The cells were divided in to 5 groups: A. Control group; B. H2O2 group; C. Res + H2O2 group; D. Res + H2O2 + LY294002 group; E. H2O2 + LY294002 group. The cell viability were measured by cck-8 assay; Malondialdehyde (MDA) and Superoxide dismutase (SOD) were determined by using commercially available kits; Reactive oxygen species (ROS)were detect by flow cytometer; the expression of Nuclear factor-E2-related factor(Nrf2), Protein kinase B(Akt), Mammalian target of rapamycin (mTOR) were determined by Western blot.RESULTS:Compared with control, resveratrol increased the cells viability (36.14 ± 2.20) to (72.05 ± 2.46) (P = 0.004), decreased MDA formation (299.38 ± 12.42) to (159.17 ± 7.30)(P = 0.006), elevated SOD activity(35.61 ± 3.26) to (81.37 ± 4.55) (P = 0.007), inhibited ROS production (177.86 ± 8.98) to (133.96 ± 4.17) (P = 0.003). These changes were accompany with the activation of p-Nrf2 (198.57 ± 6.89) to (371.10 ± 8.41) (P = 0.005), p-Akt (102.93 ± 3.29) to (221.62 ± 6.51) (P = 0.004), p-mTOR (93.26 ± 4.12) to (172.14 ± 5.74) (P = 0.008). However these protective effect of resveratrol were abolished by LY294002.CONCLUSION:Resvertrol could protects HUVEC cells from H2O2-induced oxidative stress, in which the PI3K-Akt-mTOR pathway may be involved.
Protective Effects of Diosgenin in the Hyperlipidemic Rat Model and in Human Vascular Endothelial Cells Against Hydrogen Peroxide-Induced Apoptosis.

Abstract:Hyperlipidemia is a major cause of atherosclerosis and atherosclerosis-associated conditions in cardiovascular diseases. Oxidative stress, as a main risk factor causes vascular endothelial cell apoptosis, which is implicated in the pathogenesis of cardiovascular disorders. Diosgenin, an aglycone of steroidal saponins, has been reported to exert anti-proliferative and proapoptotic actions on cancer cells widely. In this study, we propose that diosgenin can protect the hyperlipidemic rats and prevent endothelial apoptosis under oxidative stress. We investigated the hypolipidemic and antioxidative effects of diosgenin on rats fed with high cholesterol and high fat diet for 6 weeks. Serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), glutathione peroxidase (GSH-PX), nitric oxide synthase (NOS), hepatic malondialdehyde (MDA), lipoprotein lipase (LPL), hepaticlipase (HL) and superoxide dismutase (SOD) activities were evaluated. Then we explored the effects and mechanism of diosgenin against hydrogen peroxide-induced apoptosis of human vein endothelium cells (HUVECs). Intracellular reactive oxygen species (ROS), glutathione (GSH), nitric oxide (NO), DNA fragment formation and mitochondrial membrane potentials (ΔΨm) were determined. Diosgenin treatment increased LPL, HL, SOD, GSH-PX and NOS activities, thus attenuated oxygen free radicals, decreased MDA, TC, TG and LDL-C levels in hyperlipidemic rats. Diosgenin pretreatment significantly attenuated H2O2-induced apoptosis in HUVECs, intracellular ROS, GSH depletion, DNA fragment formation, and restored NO, ΔΨm. These results suggested that diosgenin is a very useful compound to control hyperlipidemia by both improving the lipid profile and modulating oxidative stress and prevent H2O2-induced apoptosis of HUVECs, in partly through regulating mitochondrial dysfunction pathway.

Abstract:Neuroprotective effects of dehydroepiandrosterone (DHEA) have been shown to be associated with its antioxidant properties, but the mechanisms remain unknown. Considering that the thioredoxin (Trx) system, an important cellular redox modulation system, changes under oxidative stress and could exert protective effects, the relationship between the antioxidant effects of DHEA and the Trx system regulation was explored. Using MTT assay and morphological observation, the effects of DHEA in the model of H2O2-induced oxidative stress in SH-SY5Y cells were analyzed, then RT-PCR and Western blot assay were used to detect the alteration in mRNA and protein level of Trx. The results showed that a pre-treatment of DHEA (10-100 nM) protected cells against the toxicity induced by H2O2 in a dose-dependent manner, which could be confirmed in morphological observation by phase-contract microscope. In addition, Trx mRNA transcription was inhibited by H2O2 (300 microM), which could be reversed by the pre-administration of DHEA in various concentrations (0.1100 nM). Western blot assay confirmed that protein level of Trx could be elevated by the pre-treatment of DHEA (10100 nM) with the exposure of H2O2. Taken together, these data suggest that DHEA may be useful in treating age-related neurodegenerative diseases based on its up-regulating effects on an antioxidant and neuroprotective protein thioredoxin, a substrate in the Trx redox system.
Antitumor Effects and the Potential Mechanism of 10-HDA against SU-DHL-2 Cells

Abstract:10-hydroxy-2-decenoic acid (10-HDA), which is a unique bioactive fatty acid of royal jelly synthesized by nurse bees for larvae and adult queen bees, is recognized for its dual utility in medicinal and nutritional applications. Previous research has indicated that 10-HDA exerts antitumor effects on numerous tumor cell lines, including colon cancer cells, A549 human lung cancer cells, and human hepatoma cells. The present study extends this inquiry to lymphoma, specifically evaluating the impact of 10-HDA on the SU-DHL-2 cell line. Our findings revealed dose-dependent suppression of SU-DHL-2 cell survival, with an IC50 of 496.8 μg/mL at a density of 3 × 106 cells/well after 24 h. For normal liver LO2 cells and human fibroblasts (HSFs), the IC50 values were approximately 1000 μg/mL and over 1000 μg/mL, respectively. The results of label-free proteomics revealed 147 upregulated and 347 downregulated differentially expressed proteins that were significantly enriched in the complement and coagulation cascades pathway (adjusted p-value = 0.012), including the differentially expressed proteins prothrombin, plasminogen, plasminogen, carboxypeptidase B2, fibrinogen beta chain, fibrinogen gamma chain, and coagulation factor V. The top three hub proteins, ribosomal protein L5, tumor protein p53, and ribosomal protein L24, were identified via protein–protein interaction (PPI) analysis. This result showed that the complement and coagulation cascade pathways might play a key role in the antitumor process of 10-HDA, suggesting a potential therapeutic avenue for lymphoma treatment. However, the specificity of the effect of 10-HDA on SU-DHL-2 cells warrants further investigation.

Abstract:Royal jelly (RJ) is known as a functional food for its diverse health-beneficial properties and complicated chemical compositions. Trans-10-hydroxy-2-decenoic acid (10-HDA) is the exclusive lipid component in RJ. In the present study, the in vitro anti-inflammatory effect of 10-HDA in LTA (Lipoteichoic acid from Staphylococcus aureus) induced RAW 264.7 macrophages are evaluated. The results showed that 10-HDA had potent, dose-dependent inhibitory effects on the release of major inflammatory mediators and NO. Several key inflammatory genes, including IL-1β, IL-6, MCP-1 and COX-2 have also been suppressed by 10-HDA. Furthermore, the effects of 10-HDA on LTA-induced pulmonary damage were also examined in mice. It was found that the administration of 10-HDA (100 mg/kg) can provide protective effects by attenuating lung histopathological changes and modulating the secretion of LTA-stimulated inflammatory cytokines in mice, such as IL-10, MCP-1 and TNF-α. Conclusively, the results reveal the potent anti-inflammatory properties of 10-HDA and provide biological information for the future application.

Protective effect of enzymatic hydrolysates from seahorse (Hippocampus abdominalis) against H2O2-mediated human umbilical vein endothelial cell injury

Abstract:Oxidative stress-mediated endothelial dysfunction and LDL oxidation have been implicated in the pathogenesis of atherosclerosis. Thus, the protection of the endothelial cells against oxidative stress-mediated injury and the inhibition of LDL oxidation by the use of antioxidants are a good strategy against atherosclerosis development. Here, we investigated the protective effect and the inhibition of LDL oxidation of seahorse H. abdominalia hydrolysates by Alcalase (SHAH). SHAH showed higher antioxidant activities by measuring DPPH, ABTS+, and ORAC assays than the other hydrolysates. SHAH reduced the formation of thiobarbituric acid reactive substance in Cu2+-induced LDL oxidation. In human umbilical vein endothelial cell (HUVEC), SHAH ameliorated H2O2-mediated HUVEC injury through the restoration of antioxidant enzyme activities and glutathione. In addition, SHAH inhibited HUVEC apoptosis through the down-regulation of caspase-3 and p53 and the increase bcl-2/bax ratio. These results suggested that seahorse H. abdominalia could be developed as potential agents for atherosclerosis.
7, 8-Dihydroxyflavone Protects an Endothelial Cell Line from H2O2 Damage

Abstract:7, 8-dihydroxyflavone (7, 8-DHF), a selective agonist for TrkB receptors, has been well studied for its neurotrophic functions. However, its roles outside the neural tissues have scarcely been studied as yet. In this study, we investigated the protecting roles of 7, 8-DHF in EA.hy926 cells, a human umbilic vein endothelial cell line which was exposed to hydrogen peroxide (H2O2). We found that 7, 8-DHF significantly protected the cells from being damaged by H2O2 through suppression of apoptosis, attenuation of inflammatory factor releasing and inhibition of reactive oxygen species generation. The potent biological effects of 7, 8-DHF were probably executed via its binding to TrkB receptors because the receptor specific antagonist ANA-12 significantly blocked its protecting effects. The protecting roles of 7, 8-DHF in EA.hy926 cells suggest that it will be a promising compound to be developed into a health product that definitely benefits endothelial functions and prevents cardiovascular diseases.

Trans-10-hydroxy-2-decenoic Acid Alleviates LPS-induced Blood-Brain Barrier Dysfunction by Activating the AMPK/PI3K/AKT Pathway.

Abstract:We previously reported that trans-10-hydroxy-2-decenoic acid (10-HDA), the exclusive lipid component of royal jelly (RJ), alleviates Lipopolysaccharide (LPS)-induced neuroinflammation both in vivo and in vitro. However, whether 10-HDA can protect against LPS-induced blood-brain barrier (BBB) damage is largely unexplored. In this study, we first observed that 10-HDA decreased BBB permeability in LPS-stimulated C57BL/6 mice by Evan's blue (EB) dye. Immunostaining and Western blot results showed that 10-HDA alleviated BBB dysfunction by inhibiting the degradation of tight junction proteins (occludin, claudin-5 and ZO-1). In LPS-stimulated human brain microvascular endothelial cells (HBMECs), 10-HDA decreased the expression of chemokines (CCL-2 and CCL-3), adhesion molecules (ICAM-1 and VCAM-1), reactive oxygen species, matrix metalloproteinases (MMP-2 and MMP-9) and increased the expression of tight junction proteins. Interestingly, LC-MS/MS analysis showed that 10-HDA pretreatment upregulated the expression of mitochondria-associated proteins, which may reflect the mechanism underlying the regulatory effect of 10-HDA on reactive oxygen species. We further illustrated that 10-HDA promoted the activation of the AMPK pathway and the downstream PI3K/AKT pathway. Compound C (an AMPK inhibitor) and LY294002 (a PI3K inhibitor) markedly reversed the alleviating effect of 10-HDA on the expression of tight junction proteins, indicating that 10-HDA inhibited LPS-induced BBB dysfunction by triggering the activation of the AMPK/PI3K/AKT pathway. Collectively, these data reveal that 10-HDA may be an interesting candidate for clinical evaluation in the treatment of diseases related to BBB damage.

Abstract:Hydroxyl Safflower Yellow A (HSYA) is the primary bioactive compound derived from Safflower, which has been scientifically proven to possess anti-inflammatory, anti-apoptotic, and ameliorative properties against mitochondrial damage during acute myocardial ischemia-reperfusion injury (MIRI); however, its effects during the recovery stage remain unknown. Angiogenesis plays a crucial role in the rehabilitation process. Aim of the study: The objective of this study was to investigate the long-term angiogenic effect of HSYA and its contribution to recovery after myocardial ischemia, as well as explore its underlying mechanism using non-targeted metabolomics and network pharmacology. Materials and methods: The MIRI model in rat was established by ligating the left anterior descending branch of the coronary artery. The effect of HSYA was assessed based on myocardial infarction volume and histopathology. Immunofluorescence staining was employed to evaluate angiogenesis, while ELISA was used to detect markers of myocardial injury. Additionally, a rat myocardial microvascular endothelial cell (CMECs) injury model was established using oxygen-glucose deprivation/reoxygenation (OGD/R), followed by scratch assays, migration assays, and tube formation experiments to assess angiogenesis. Western blot analysis was conducted to validate the underlying mechanism. Results: Our findings provide compelling evidence for the therapeutic efficacy of HSYA in reducing myocardial infarction size, facilitating cardiac functional recovery, and reversing pathological alterations within the heart. Furthermore, we elucidate that HSYA exerts its effects on promoting migration and generation of myocardial microvascular endothelial cells through activation of the HIF-1α-VEGFA-Notch1 signaling pathway. Conclusion: These results underscore how HSYA enhances cardiac function via angiogenesis promotion and activation of the aforementioned signaling cascade.

Abstract:Vascular aging is an important factor leading to cardiovascular diseases such as hypertension and atherosclerosis. Hyperlipidemia or fat accumulation may play an important role in vascular aging and cardiovascular disease. Isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate (IDHP) has biological activity and can exert cardiovascular protection, which may be related to ferroptosis. However, the exact mechanism remains undefined. We hypothesized that IDHP may have a protective effect on blood vessels by regulating vascular aging caused by hyperlipidemia or vascular wall fat accumulation. The aim of this study is to investigate the protective effect and mechanism of IDHP on palmitic acid-induced human umbilical vein endothelial cells (HUVEC) based on senescence and ferroptosis. We found that IDHP could delay vascular aging, reduce the degree of ferrous ion accumulation and lipid peroxidation, and protect vascular cells from injury. These effects may be achieved by attenuating excessive reactive oxygen species (ROS) and ferroptosis signaling pathways generated in vascular endothelial cells. In short, our study identified IDHP as one of the antioxidant agents to slow down lipotoxicity-induced vascular senescence through the ROS/ferroptosis pathway. IDHP has new medicinal value and provides a new therapeutic idea for delaying vascular aging in patients with dyslipidemia.

Protective Effects and Mechanisms of Oleanolic Acid on Damaged Human Umbilical Vein Endothelial Cells

Abstract:Objective:To study the protective effects and mechanisms of oleanic acid on damaged human umbilical vein endothelial cells(HUVECs) induced by hydrogen peroxide(H2O2).Methods:HUVECs were divided into control group,H2O2 group(200 %mol/L H2O2 treatment),H2O2 of 200 %mol/L combined with OA of 0.25 %mol/L treatment group,H2O2 of 200 %mol/L combined with OA of 0.50 %mol/L treatment group,H2O2 of 200 %mol/L combined with OA of 1.00 %mol/L treatment group,H2O2 of 200 %mol/L combined with vitamin C of 1.50 mmol/L treatment group.Cell viability was determined by MTT method.Lactate dehydrogenase(LDH) and nitric oxide(NO) content in the supernatant were determined by biochemical method.The activity of malondialdehyde(MDA),glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) in the cell lysate was determined by biochemical method.Intracellular reactive oxygen species(ROS) and cell apoptosis were determined by flow cytometry.Results:OA restored the H2O2-injuried cell viability,reduced the H2O2-induced LDH release,and reduced H2O2-induced MDA formation,recovered H2O2-injuried GSH-Px and SOD activity,removed H2O2-induced ROS,reduced the H2O2-induced apoptosis,and enhanced the NO production in HUVECs.Conclusion:OA has a protective effect on HUVECs,which is related to initiating the antioxidant defense,getting rid of ROS,inhibiting H2O2-induced apoptosis,and improving NO production in HUVECs.
Protective effects of hydroxysafflor yellow an on high oxidized low density lipoprotein induced human coronary artery endothelial cells injuries

Abstract:The aim of this study was to investigate the injury of human coronary artery endothelial cells (HCAECs) induced by high oxidized low density lipoprotein (ox-LDL) and to examine the protective effect of hydroxysafflor yellow A (HSYA) on HCAECs injury. It was found that in the high ox-LDL group the content of NO, the expression of endothelial nitric oxide synthase (eNOS) mRNA and protein were decreased. The expression of LDH, lectin-like low-density lipoprotein receptor 1 (LOX-1) mRNA and LOX-1 protein were up-regulated. The number of apoptotic cells were significantly increased after deal with high ox-LDL compared with the control group. Furthermore, in the high ox-LDL+HSYA group, the cell survival rate, the release of NO and the expression of eNOS mRNA and eNOS protein were increased. The secretion of LDH and the expression of LOX-1 at the level of mRNA and protein were down-regulated. The number of apoptosis were significantly reduced after combined with HSYA compared with the high ox-LDL group. Therefore, high ox-LDL had an obvious damage to HCAECs. The HSYA inhibited the high ox-LDL-induced HCAECs injury and protected and promoted the cell repaired, possibly by up-regulating the eNOS gene and protein expression, increasing NO release, inhibiting LDH release and down-regulating LOX-1 mRNA and protein expression to achieve the effect. The treatment and secondary prevention of coronary heart disease, as well as the recovery of patients undergoing postoperative percutaneous coronary intervention (PCI), will benefit to a large extent.

Abstract:Hippophae rhamnoides L. has been widely used in research and application for almost two decades. While significant progress was achieved in the examination of its fruits and seeds, the exploration and utilization of its by-products have received relatively less attention. This study aims to address this research gap by investigating the effects and underlying mechanisms of sea buckthorn seed residues both in vitro and in vivo. The primary objective of this study is to assess the potential of the hydroalcoholic extract from sea buckthorn seed residues (HYD-SBSR) to prevent cell apoptosis and mitigate oxidative stress damage. To achieve this, an H2O2-induced B16F10 cell model and a D-galactose-induced mouse model were used. The H2O2-induced oxidative stress model using B16F10 cells was utilized to evaluate the cellular protective and reparative effects of HYD-SBSR. The results demonstrated the cytoprotective effects of HYD-SBSR, as evidenced by reduced apoptosis rates and enhanced resistance to oxidative stress alongside moderate cell repair properties. Furthermore, this study investigated the impact of HYD-SBSR on antioxidant enzymes and peroxides in mice to elucidate its reparative potential in vivo. The findings revealed that HYD-SBSR exhibited remarkable antioxidant performance, particularly at low concentrations, significantly enhancing antioxidant capacity under oxidative stress conditions. To delve into the mechanisms underlying HYD-SBSR, a comprehensive proteomics analysis was conducted to identify differentially expressed proteins (DEPs). Additionally, a Gene Ontology (GO) analysis and an Encyclopedia of Genes and Genomes (KEGG) pathway cluster analysis were performed to elucidate the functional roles of these DEPs. The outcomes highlighted crucial mechanistic pathways associated with HYD-SBSR, including the PPAR signaling pathway, fat digestion and absorption, glycerophospholipid metabolism, and cholesterol metabolism. The research findings indicated that HYD-SBSR, as a health food supplement, exhibits favorable effects by promoting healthy lipid metabolism, contributing to the sustainable and environmentally friendly production of sea buckthorn and paving the way for future investigations and applications in the field of nutraceutical and pharmaceutical research.

Abstract:Phosphatidylinositol 3-kinase (PI3-kinase) is a novel intracellular transducer involved in a wide range of cancer-associated signaling pathways, which comprises various isoforms and splice variants with distinct biologic activities and clinical implications. Especially, the class Ia PI3-kinase 110 kD catalytic subunit alpha (PIK3CA) is the most important isoform in tumorigenesis and possibly, tumor angiogenesis. Several strategies have been developed to block PI3-kinase for cancer therapy; however, the approach to target specific PI3-kinase isoform has not been explored to date. In the present study, we show that RNA interference (RNAi) through small interfering (siRNA) sequences targeting PIK3CA has potential applications in isoform-specific "knock-down" of PI3-kinase. This strategy provides a novel tool to study the function of various PI3-kinase isoforms and may contribute to isoform-specific targeting of PI3-kinase in human cancer.

Abstract:AIMS:To investigate the cytoprotective effects of two sesquiterpene aminoquinones isolated from the marine sponge Dysidea fragilis, Dysidaminone H (DA8) and 3'-methylamino-avarone (DA14), we examined their effects against hydrogen peroxide (H2O2)-induced oxidative injury in human keratinocyte cell line and elucidated the underlying mechanisms. MAIN METHODS:Cell viability was detected using a CCK-8 assay kit. Intracellular reactive oxygen species (ROS) production was measured by fluorescence of 2, 7-Dichlorodi-hydrofluorescein diacetate (DCFH-DA). Messenger RNA and protein expression were measured by real-time quantitative PCR and western blotting analysis. Immunocytochemistry was performed to determine the intracellular location of nuclear factorerythroid 2 p45 related factor 2 (Nrf2). The antioxidant response element (ARE)-luciferase reporter gene assay and RNA interference were used to establish the role of ARE and Nrf2. KEY FINDINGS:DA8 and DA14 (DAs) resisted H2O2induced decline of cell viability by inhibiting the accumulation of ROS. Meanwhile, DAs increased HO-1 expression and ARE activity and induced Nrf2 expression, as well as the accumulation of Nrf2 in the cell nucleus. However, silencing of Nrf2 abolished DAs-induced HO-1 expression and ARE luciferase activation. In addition, DAs induced the phosphorylation of both cyclic AMP-activated protein kinase-α (AMPKα) and extracellular signal-regulated kinase (ERK), while specific inhibitors of AMPKα and ERK abrogated HO1 upregulation and Nrf2 activation. SIGNIFICANCE:DAs provided cytoprotective effects against H2O2-induced cytotoxicity by activation of the Nrf2/ARE/HO-1 pathway via phosphorylation of AMPKα and ERK. The findings suggested that DA8 and DA14 might be the candidate therapeutic agents for skin diseases caused by oxidative injury.

Abstract:Impaired vasodilation is one of the characterizations of hypertension. The effects of the omega-3 fatty acids on blood pressure and vascular function have been examined in many randomized controlled trials. In this study, we investigated the effects of omega-3 fatty acids, particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), on DNA damage, which has crucial effects on vascular endothelial cell function. We examined the effects of EPA and DHA on H2O2-induced DNA damage response in human aortic endothelial cells (HAECs). DNA double-strand breaks (DSBs) were detected by immunofluorescence staining of the phosphorylated form of the histone H2AX (gamma-H2AX). EPA and DHA significantly reduced accumulation of DSBs and phosphorylation of ATM, a serine/threonine kinase recruited and activated upon DSBs, in HAECs. Since EPA and DHA attenuated DNA damage without enhancement of DNA repair response, the effects of EPA and DHA on oxidative stress, a DNA damage inducer, were examined. CM-H2DCFDA measurement showed that EPA and DHA significantly reduced intracellular reactive oxygen species. In addition, mRNA levels of antioxidant molecules, such as heme oxygenase-1 and thioredoxin reductase 1, were significantly elevated by EPA and DHA. Silencing the gene encoding nuclear factor erythroid 2-related factor 2 (Nrf2) abrogated these increases. Furthermore, EPA and DHA reduced H2O2-induced senescence-associated beta-galactosidase activity, which was revoked by Nrf2 silencing. Our results suggest that EPA and DHA attenuate oxidative stress-induced DNA damage and cell senescence in vascular endothelial cells partly via antioxidant response mediated by Nrf-2. Therefore EPA and DHA may play a role in the prevention of impairment in vasodilation by their genome-protective properties.
