Abstract:Biogenic amines (BAs) are a class of bioactive organics produced during the fermentation of soy sauce. A high concentration of BAs may bring about serious physiological and toxicological effects on the human body. In this study, we reported an optimized process to produce soy sauce with lower BA concentration and found the contents of putrescine, cadaverine and histamine increased with the increase of fermentation temperature but decreased with the increase of NaCl concentration. The final content of total BAs with improved fermentation was 105.56 +/- 0.13 mg/L, which was reduced by 89.11% compared to traditional brewing. Besides, the pilot production test was performed to verify the optimized conditions and physicochemical indexes were measured to better understand the change principle of the chemical compounds. Taken together, we present an effective process to inhibit the formation of BAs while ensuring that characteristic nutrients are not lost.
Exploring the core microorganisms and mechanisms responsible for biogenic amines production during soy sauce fermentation

Abstract:Hazardous levels of biogenic amines (BA) are frequently detected in soy sauce. However, the microorganisms and mechanisms responsible for BA formation in soy sauce are not well understood. In this study, we investigated the dynamic changes and metabolic pathways of BAs during soy sauce fermentation via metabolome and metagenomic analyses. Spermidine and putrescine were the main BAs detected during koji making, while putrescine, tyramine, phenylethylamine, and histamine were dominant during moromi fermentation. PCA analysis revealed that the soy sauce fermentation process could be divided into five representative stages based on the dynamics of BAs. Metagenomic analysis uncovered that a total of 15 genera were mainly involved in BAs formation, particularly, histamine was principally produced by Klebsiella, and Bacillus was the major tyramine-synthesizing bacteria. Correlation analysis showed that pH increase inhibited the spermidine production during koji making, whereas elevated NaCl concentration facilitated the phenylethylamine formation during moromi fermentation. Co-occurrence network analysis unveiled the associations between the BA-producing microorganisms and other genera. Overall, these results have improved our understanding of the mechanism of BA formation during soy sauce fermentation and guided the development of fermentation agents capable of degrading BA in industrial manufacture soy sauce production.

Abstract:Soy sauce contains a series of biogenic amines (BAs) which is a kind of bioactive organics relating to food quality and safety. High concentration of BAs may lead to remarkable physiological and toxicological influences on human bodies, including hypotension, dizziness, and headaches. Here, we systematically compared the levels of ten main BAs among 53 Chinese commercial soy sauces using an improved high-performance liquid chromatography (HPLC) method. The results showed that the brands and production regions were both important factors accounting for the BAs' content. The contents of Cad, Spm, Try, Phe, His, and Tyr in dark soy sauces were higher than those in light soy sauces. His and Phe in dark soy sauces were 3.7 and 1.84 times higher than in light samples, respectively. Besides, it was surprising that the content of BAs in soy sauces hugely varied from place to place. This work comprehensively compared the content of BAs in soy sauces, showing the relation between soy sauce processes and BAs, offering abundant information for further research on BAs control.
Metagenomic and metaproteomic analyses of microbial amino acid metabolism during Cantonese soy sauce fermentation

Abstract:Cantonese soy sauce is an important type of traditional Chinese brewed soy sauce that was developed in southern China, mainly in Guangdong. Due to the long fermentation period and complex microbiota in Cantonese soy sauce, there are few reports on the microbial metaproteomics of Cantonese soy sauce. In this study, integrative metagenomic and metaproteomic analyzes were used to identify the changes in the dominant microbiota and amino acid synthesis-related enzymes and metabolism during Cantonese soy sauce fermentation. Metagenomic analysis revealed that Tetragenococcus halophilus, Weissella confusa, Weissella paramesenteroides, Enterobacter hormaechei, and Aspergillus oryzae were the dominant microbiota. Using the Top 15 dominant microbiota identified by metagenomics as the database, LTQ Orbitrap Velos Pro ETD mass spectrometry was used to obtain metaproteomic information about the microbes in the soy sauce, and the results indicated that the active enzymes involved in the metabolism of amino acids were secreted by microorganisms such as A. oryzae, T. halophilus, and Zygosaccharomyces rouxii. During the Cantonese soy sauce fermentation process. Among them, early fermentation (0-15d) was dominated by A. oryzae and T. halophilus, mid-term fermentation (60-90d) was dominated by Z. rouxii, A. oryzae, and T. halophilus, and late fermentation (90-120d) was dominated by A. oryzae, Z. rouxii, and T. halophilus. Kyoto Encyclopedia of Genes and Genomes analysis revealed that the main enzymes involved in the metabolism of umami amino acids were aspartate aminotransferase, citrate synthase, aconitase, and isocitrate dehydrogenase, which were produced by Z. rouxii and A. oryzae during early fermentation (0-15 d) and the middle fermentation stage (60-90 d). This study constructed a regulatory network of enzymes potentially involved in the metabolism of flavor amino acids, which provided a theoretical basis for studying the amino acid metabolism of Cantonese soy sauce.

Abstract:The changes in the concentration of glucagon and insulin in arterial plasma which occur in response to splanchnic nerve stimulation have been investigated in adrenalectomized dogs, cats and sheep. 2. In dogs, stimulation of both splanchnic nerves at a low frequency (2‐0 c/s) for 10 min produced a small but statistically significant increase in plasma glucagon concentration and appeared to inhibit the release of insulin. Stimulation at a higher frequency (10‐0 c/s) produced a much greater increase in plasma glucagon concentration, which was normally accompanied by a rise in plasma glucose concentration. 3. Qualitatively similar changes in plasma glucagon and insulin concentration were observed in both sheep and cats in response to adrenergic stimulation. 4. Intramesenteric infusions of glucagon at a dose of 5‐0 ng kg‐1 min‐1 in dogs produced a comparable rise in plasma glucagon concentration to that elicited by splanchnic nerve stimulation at high frequency (10‐0 c/s) and invariably caused a rise in plasma glucose concentration. 5. In dogs given exogenous glucose, release of glucagon in response to splanchnic nerve stimulation was unaffected by induced hyperglycaemia. Secretion of insulin was partially inhibited by stimulation at 2‐0 c/s and completely suppressed at higher frequency (10‐0c/s). 6. It is concluded that stimulation of the sympathetic innervation to the pancreatic islets, at frequencies within thephysiological range, stimulates the release of glucagon and inhibits that of insulin in each of these species.
Reduced formation of biogenic amines in low-salt Zhacai via fermentation under CO2-modified atmosphere

Abstract:Reducing sodium salt content in traditional fermented vegetables and developing low-salt fermented products have attracted increasing attention.However, low-salt fermented vegetables are prone to accumulate toxic biogenic amines (BAs) caused by the undesirable metabolism of spoilage microorganisms. This study aimed to investigate the impact of a CO2-modified atmosphere (MA) approach to the fermentation of low-salt Zhacai and the accumulation of BAs. The results show CO2-MA effectively suppressed the production of excessive BAs in low-salt Zhacai, as evidenced by a decrease in the total BA content from 63.66 to 161.41 mg/ kg under natural air conditions to 1.88-24.76 mg/ kg under CO2-MA. Overall, the mechanism of hindering BA formation was closely related to the change in the microbial community and the downregulation of BA-producing enzymes. Lactic acid bacteria, including Lactiplantibacillus plantarum, Weissella spp., and Pediococcus spp., were enriched under CO2-MA, whereas amine-producing microorganisms (e.g., Halomonas spp., Psychrobacter spp., Corynebacterium spp., and Levilactobacillus brevis) were greatly inhibited. Moreover, metagenomic analysis revealed that genes encoding amino acid decarboxylase, amine deiminase, and amine synthase were downregulated, which could be the fundamental reason for BA reduction. This study provides an alternative method for reducing BA production in fermented food.
Systematic analysis of key fermentation parameters influencing biogenic amines production in spontaneous fermentation of soy sauce

Abstract:Biogenic amines (BAs), a class of toxic nitrogen-containing compounds, are commonly found in high-protein fermented foods. This study investigated whether, what and how the fermentation parameters influence the BAs accumulation in spontaneous soy sauce fermentation. The results showed that BAs accumulation in soy sauce fermentation throughout the year showed significant seasonal differences, which were closely related to temperature and pH value. Furthermore, different microbial communities were found in high- and low-BAs groups, indicating that seasonal fermentation factors affect BAs accumulation by influencing the microbial population. Following bioinformatic analysis, single-microorganism culture analysis, and simulated fermentation, it was found that lower temperature or higher NaCl concentration can reduce BAs accumulation, most likely by suppressing the proliferation of BAs-producing bacteria. This is a comprehensive study of the impact of fermentation parameters on BAs production, and will serve as a foundation for future research on BAs management.

Abstract:Here, we show that residual contaminants in purified single-wall carbon nanotube bundles may be responsible for the reported sensitivity of the electronic and transport properties to oxygen. Removal of these contaminants makes the electronic spectra insensitive to O(2), CO, H(2)O, and N(2), while a strong sensitivity to NO(2), SO(2), and NH(3) is observed, confirming the possible application of single-wall nanotubes as powerful sensors capable of measuring environmentally significant levels of toxic gases.
Dynamics of Bacterial Composition and Association with Quality Formation and Biogenic Amines Accumulation during Fish Sauce Spontaneous Fermentation

Abstract:High-throughput sequencing and high-pressure liquid chromatography (HPLC) methods were used to investigate the influences of microbial dynamics on the quality and biogenic amine (BA) content during fish sauce fermentation. The homogeneity of total viable bacteria and lactic acid bacteria in fish sauce becomes higher as fermentation progresses. Tetragenococcus was the key genus of fish sauce fermentation. Carnobacterium (38.43%) and Lentibacillus (41.01%) were the dominant genera in the samples fermented for 3 months and 18 months, respectively. These three bacterial genera were significantly related to the physicochemical characteristics and characteristic flavors of the sauces. Tetragenococcus was significantly positively correlated with nitrogen oxides, the main characteristic flavor components in fish sauce. The BA content in fish sauce fermentation increased from 106.88 to 376.03 mg/kg, and the content of histamine reached 115.30 mg/kg at the end of fermentation, indicating that fish sauce has health risks. About 66.67% of Lentibacillus isolates were able to produce a large amount of BA, suggesting that Lentibacillus was the key genus for BA accumulation in fish sauce fermentation. Research on reducing the content of BA in fish sauce by intervening with regard to the fermentation temperature showed that a safe fish sauce product could be obtained at the fermentation temperature of about 25°C. These results help us to understand the contribution of microbial community composition to fish sauce fermentation and provide a basis for improving the quality and safety of fermented fish sauce. IMPORTANCE Traditional fermentation of fish sauce is mainly carried out by complex microbial communities from raw anchovies and processing environments. However, it is still unclear how the environmental microbiota influences the quality and the safety of fish sauce products. Therefore, this study comprehensively explained the influence of microorganisms on the quality and safety of fish sauce during the fermentation process in terms of physicochemical characters, flavors, and BA. Additionally, the accumulation of BA in fish sauce fermentation was controlled by intervening in the fermentation temperature. This finding contributes to a deeper understanding of the role of environmental microbiota during fermentation and provides data support for improving the safety of fish sauce.
Brewing-related genes annotation of Bacillus velezensis CS1.10S isolated from traditional moromi and its effects on promoting soy sauce fermentation

Abstract:The abundance of salt-tolerant microorganisms in moromi contributes greatly to the flavor of soy sauce, and Bacillus velezensis CS1.10S, isolated from the traditional Chinese sun-brewed moromi, is one of them. In order to understand the intrinsic mechanism of the fermentation properties of this bacterium, 4211 genes obtained from the sequencing of its genome were annotated, and it was found that genes encoding carbohydrate metabolism and transport accounted for 7.43%. Genes encoding amino acid metabolism and transport accounted for 9.66%, with a total of 284 genes related to amino acid synthesis, and 13 metabolic pathways covering 19 amino acids, which showed that CS1.10S had good carbon and nitrogen metabolism and amino acid production ability. KEGG also annotated three kinds of salt tolerance-related genes, which explained the good performance of CS1.10S in tolerating salt up to 160 g/L. A comparative study of the quality indexes of fermented soy sauce supplemented with CS1.10S revealed that the amino acid nitrogen content of soy sauce supplemented with CS1.10S on the 10th day of fermentation was 7.51% higher than that of the control group, and its total free amino acid content also increased by 42.38%. The contents of reducing sugars and volatile flavor compounds also increased, but the total acid did not change much, indicating that CS1.10S was beneficial to the taste and aroma of soy sauce. The annotation of CS1.10S with brewing-related genes and the corresponding evidence from fermentation tests indicated that CS1.10S has the potential to improve the flavor of soy sauce.

Abstract:Soy sauce contains several nitrogen-containing compounds, most of which are proteins and their decomposition products during the fermentation process. These compounds include potentially harmful ammonium salts. In this study, the effects of changing the koji making and fermentation conditions of soy sauce on the ammonium salt and amino acid nitrogen content in soy sauce are researched. The results indicate that mixed microbial koji making can significantly influence the ammonium salt content in soy sauce, with the increase in the amount of microbial inoculated, the ammonium salt content exhibited an overall upward trend, and increase the total amount of free amino acids. Besides, the ammonium salt content and brine concentration was a significant negative correlation between the two. Moreover, as the temperature of koji making increases, the ammonium salt content first decreases and then increases, and the content of ammonium salt gradually decreased with the increasing fermentation temperature. This study demonstrates that changes in soy sauce brewing conditions will influence the formation of ammonium salts and amino acid nitrogen in soy sauce, providing a foundation for further optimization of the soy sauce brewing process.
Effect of different halophilic microbial fermentation patterns on the volatile compound profiles and sensory properties of soy sauce moromi

Abstract:The effect of inoculated or co-cultured halophilic lactic bacteria (Tetragenococcus halophilus) and yeasts (Zygosaccharomyces rouxii and Candida versatilis) on the quality of soy sauce moromi was investigated. Results showed no significant change in the major physicochemical properties of soy sauce moromi such as amino nitrogen, total acid, and reducing sugar at the end of fermentation, whereas the composition of free amino acids slightly changed with different fermentation patterns. In the sample inoculated with T. halophilus, the concentration of proline and tryptophan decreased by 18.83 and 50.87 % compared with the blank sample, whereas tyrosine and histidine increased by 32.41 and 27.04 %. In the sample inoculated with Z. rouxii and C. versatilis, the concentration of bitter-tasting lysine, tyrosine, and phenylalanine increased by more than 20 %, and the concentration of tryptophan decreased by 56.71 %. The proportion of amino acids with sweet taste increased by 0.8 % through the co-culture of the three strains. Meanwhile, the total amount of volatile compounds increased by 117.66, 23.28, and 216.88 % in the T. halophilus inoculated sample, two yeast strain inoculated sample, and three strain inoculated sample, respectively. Increased odour activity values of the nine compounds that mainly contributed to the flavour of moromi ranged from 1 to 7.87 times in the three strain inoculated sample. These findings revealed that the co-culture of the three strains during moromi fermentation was beneficial to the improvement of flavour of soy sauce moromi.

Abstract:In this study, the influence and relationship between the microbial structure composition at different spatial locations during soy sauce fermentation and the quality of soy sauce were investigated. Within 3-7 days of fermentation, the abundance of Chromohalobacter in the surface and upper layers of moromi was initially high but subsequently decreased. In contrast, Tetragenococcus exhibited low abundance on the surface of moromi at the beginning of fermentation but emerged as the absolute dominant bacteria by the end of the fermentation process. Throughout the fermentation period of 3-42 days, Staphylococcus and Bacillus were the predominant bacterial genera observed at the bottom of the moromi. In addition, Halanaerobium was the dominant bacterial genus in the crude soy sauce layer throughout the fermentation process. Tetragenococcus and Zygosaccharomyces were strongly positively correlated with total acid and ammonia nitrogen. Bacillus and Staphylococcus were significantly negatively correlated with the salt content.
Revealing salt concentration for microbial balance and metabolite enrichment in secondary fortified fermented soy sauce: A multi-omics perspective

Abstract:This study examined the impact of varying salt concentrations on microbiota, physicochemical properties, and metabolites in a secondary fortified fermentation process using multi-omics techniques. It aimed to determine the influence of salt stress on microbiota shifts and metabolic activities. The findings demonstrated that moderate salt reduction (MS) was found to enhance moromi's flavor and quality, while mitigating the negative effects of excessive low salt (LS). MS samples had 1.22, 1.13, and 2.92 times more amino acid nitrogen (AAN), non-volatiles, and volatiles, respectively, than high salt (HS) samples. In contrast, lactic acid and biogenic amines in LS samples were 1.56 g/100 g and 4115.11 mg/kg, respectively, decreasing to 0.15 g/100 g and 176.76 mg/kg in MS samples. Additionally, the contents of ethanol and small peptides increased in MS due to the growth of specific functional microorganisms such as Staphylococcus gallinarum, Weissella confusa, and Zygosaccharomyces rouxii, while food-borne pathogens were inhibited. Network analysis revealed that the core microbial interactions were enhanced in MS samples, promoting a balanced fermentation environment. Redundancy analysis (RDA) and correlation analyses underscored that the physicochemical properties significantly impacted bacterial community structure and the correlations between key microbes and flavor compounds. These findings provided a theoretical foundation for developing innovative reduced-salt fermentation techniques, contributing to the sustainable production of high-quality soy sauce.

Abstract:ObjectivesTo reduce the amount of citrulline produced by arginine-consuming bacteria in the moromi mash during soy sauce production.ResultsBacillus amyloliquefaciens JY06, a salt-tolerant strain with high arginine consumption ability and low citrulline accumulation capacity, was isolated from moromi mash. The concentration of citrulline was decreased from 26.8 to 5.1 mM and ethyl carbamate in soy sauce, after sterilization, decreased from 97 to 17 μg kg−1 when B. amyloliquefaciens JY06 was added during fermentation. The aroma of the sauce was improved by increasing the ester content.ConclusionsB. amyloliquefaciens JY06 is a beneficial bacterium that can be used in soy sauce fermentation to eliminate ethyl carbonate and enhance the flavor of the sauce.

Abstract:<p>Soy sauce is a traditional fermented food in Asia. In this study, we determined the contents of biogenic amines (BAs) in commercial high-salt liquid-state fermented (HLF) soy sauce. Using high performance liquid chromatography (HPLC), we evaluated the effects of salinity on the synthesis of BAs during soy sauce fermentation. According to the results, various grades of HLF soy sauce demonstrated significant differences (p < 0.05) in the contents of BAs. The major BAs in laboratory-made soy sauce include spermidine (SPD), histamine (HIS), and tyramine (TYR). After fermentation, the contents of SPD, HIS, and TYR in the 20% salinity group were significantly (p < 0.05) reduced than those in the 16% and 18% salinity groups, suggesting that the salinity might be a crucial factor for reducing the biogenic amines during HLF soy sauce fermentation. We also found a positive correlation between the contents of amino nitrogen and biogenic amines.</p>

Abstract:Soy sauce, as a traditional seasoning, is widely favoured by Chinese and other Asian people for its unique colour, smell, and taste. In this study, a salt-tolerance Saccharomyces cerevisiae strain HF-130 was obtained via three rounds of ARTP (Atmospheric and Room Temperature Plasma) mutagenesis and high-salt based screening. The ethanol production of mutant HF-130 was increased by 98.8% in very high gravity fermentation. Furthermore, ATF1 gene was overexpressed in strain HF-130, generating ester-producing strain HF-130-ATF1. The ethyl acetate concentration of strain HF-130-ATF1 was increased by 130% compared to the strain HF-130. Finally, the soy sauce fermentation performance of Torulopsis globosa and HF-130-ATF1 was compared with T. globosa, HF-130, HF-130-ATF1, and Torulopsis and HF-130. Results showed ethyl acetate and isoamyl acetate concentrations in co-fermentation of T. globosa and HF-130-ATF1 were increased by 2.8-fold and 3.3-fold, respectively. In addition, the concentrations of ethyl propionate, ethyl caprylate, phenylethyl acetate, ethyl caprate, isobutyl acetate, isoamyl alcohol, phenylethyl alcohol, and phenylacetaldehyde were also improved. Notably, other three important flavour components, trimethylsilyl decyl ester, 2-methylbutanol, and octanoic acid were also detected in the co-fermentation of T. globosa and HF-130-ATF1, but not detected in the control strain T. globosa. This work is of great significance for improving the traditional soy sauce fermentation mode, and thus improving the flavour formation of soy sauce.

Effect of Lactobacillus plantarum or Enterococcus faecalis as co-inoculants with Aspergillus oryzae in koji making on the physicochemical properties of soy sauce

Abstract:Soy sauce fermentation is largely mediated by bacteria, including several non-salt-tolerant bacterial species. The present study aimed to compare the use of Aspergillus oryzae alone or in co-inoculation with Lactobacillus plantarum or Enterococcus faecalis on the resulting enzymatic activity, content of phenolic compounds, free amino acids, organic acids, and antioxidant capacity of soy sauce. Co-inoculation significantly increased acid protease, β-glucosidase, and pectinase activities during koji making. Additionally, the contents of total phenols, total flavonoids, organic acids, amino acids, isoflavones, as well as the antioxidant capacity were higher during moromi fermentation using co-inoculants compared to those obtained when A. oryzae was used as single inoculant. Overall, co-inoculation of A. oryzae with L. plantarum or E. faecalis during koji making was shown to effectively promote the production of active substances in soy sauce. PRACTICAL APPLICATION: In this study, Aspergillus oryzae in mixed inoculation with Lactobacillus plantarum or Enterococcus faecalis could be effectively used in koji making for soy sauce. The use of co-inoculants in koji making effectively promoted the production of active substances in soy sauce, hence it is improving health benefits, whilst diversifying the use of raw materials and the application value of soybean meal.
Effect of Sequential Inoculation of Tetragenococcus halophilus and Wickerhamomyces anomalus on the Flavour Formation of Early-Stage Moromi Fermented at a Lower Temperature

Abstract:Microbial inoculation in moromi fermentation has a great influence on the physicochemical and flavour properties of soy sauces. This work investigated the effect of inoculating Tetragenococcus halophilus and Wickerhamomyces anomalus on the flavour formation of early-stage moromi (30 days) fermented at a lower temperature (22 °C) by determining their physicochemical and aroma changes. The results showed that single yeast or LAB inoculation increased the production of amino nitrogen, lactic acid and acetic acid, as well as free amino acids and key flavour components. Particularly, the sequential inoculation of T. halophilus and W. anomalus produced more free amino acids and aromatic compounds, and there might be synergistic effects between these two strains. More characteristic soy sauce flavour compounds, such as benzaldehyde, HEMF, guaiacol and methyl maltol were detected in the sequentially inoculated moromi, and this sample showed higher scores in savoury, roasted and caramel intensities. These results confirmed that sequential inoculation of T. halophilus and W. anomalus could be a choice for the future production of moromi with good flavour and quality under a lower temperature.
Deciphering microbial community dynamics along the fermentation course of soy sauce under different temperatures using metagenomic analysis

Abstract:Fermented soy sauce consists of microorganisms that exert beneficial effects. However, the microbial community dynamics during the fermentation course is poorly characterized. Soy sauce production is classified into the stages of mash fermentation with koji (S0), brine addition (S1), microbial transformation (S2), flavor creation (S3), and fermentation completion (S4). In this study, microbial succession was investigated across stages at different temperatures using metagenomics analyses. During mash fermentation, Aspergillus dominated the fungal microbiota in all stages, while the bacterial composition was dominated by Bacillus at room temperature and by a diverse composition of enriched lactic acid bacteria (LAB) at a controlled temperature. Compared with a stable fungal composition, bacterial dynamics were mostly attributable to fluctuations of LAB, which break down carbohydrates into lactic acid. After adding brine, increased levels of Enterococcus and decreased levels of Lactococcus from S1 to S4 may reflect differences in salinity tolerance. Staphylococcus, as a fermentation starter at S0, stayed predominant throughout fermentation and hydrolyzed soybean proteins. Meanwhile, Rhizopus and Penicillium may improve the flavor. The acidification of soy sauce was likely attributable to production of organic acids by Bacillus and LAB under room temperature and controlled temperature conditions, respectively. Metagenomic analysis revealed that microbial succession was associated with the fermentation efficiency and flavor enhancement. Controlled temperature nurture more LAB than uncontrolled temperatures and may ensure the production of lactic acid for the development of soy sauce flavor.
