Abstract:Fe3O4@C nanocapsules were synthesized via a sacrificial-template method by coating SiO2 nanospheres with an Fe3O4@C double-shell structure, followed by dissolving SiO2 nanospheres through ammonia water under hydrothermal conditions. The nanocapsules show a loading capacity as high as 1300 mg g-1 for doxorubicin (DOX), and the DOX loaded on the surface of the carbon shells displays pH-sensitive release behavior. Drug release experiments were carried out at pH 7.4, 6.2 and 5.0. It was found that the drug release rate at pH 6.2 was about two times as fast as that at pH 7.4, and even faster at pH 5.0. A MTT assay was used to test the cytotoxicity of the DOX, nanocapsules and DOX-nanocapsules, which indicated the low cytotoxicity of the nanocapsules towards cells. The DOX-nanocapsules can be taken up by cancer cells through endocytosis, releasing DOX into the cytoplasm, which was observed by both transmission electron microscopy and confocal microscopy. In vitro magnetic resonance imaging (MRI) experiments validated the potential use of these nanocapsules as MRI contrast agents.
Magnetic Fe3O4@ZIF-8 nanoparticles as a drug release vehicle: pH-sensitive release of norfloxacin and its antibacterial activity

Abstract:Metal-organic frameworks (MOFs) have a high specific surface area and inherent biodegradability due to their unique structure and composition. As well, owing to the properties of nanomaterials and especially their magnetic features, Fe3O4 nanoparticles and MOFs composite materials have great potential in the design and application of drug release. The present work: firstly, investigated norfloxacin loading in magnetic metal organic framework (Fe3O4@ZIF-8); and secondly, studied the release of norfloxacin and its antibacterial activity. Results showed the release efficiencies reached 97 % at 310 K after 84 h (pH 7.4). Drug release behavior was tested at various pH levels and it was found that Fe3O4@ZIF-8 has pH-sensitive properties. Furthermore, the release model calculation illustrated that the release process fitted well to the Bhaskar model. The magnetic properties of Fe3O4@ZIF-8 confirmed that the composite has potential application for a targeted drug delivery system. The mechanism of pH-responsive norfloxacin release was combined with diffusion, ion exchange and electrostatic repulsion. Furthermore, the antibacterial activities of Fe3O4@ZIF-8 and NOR-Fe3O4@ZIF-8 were tested against Escherichia coli. Results showed that Fe3O4@ZIF-8 had good biocompatibility while NOR-Fe3O4@ZIF-8 can deter or inhibit the actions of microorganisms.
Polymer Grafted-Magnetic Halloysite Nanotube for Controlled and Sustained Release of Cationic Drug

Abstract:In this research, novel polymer grafted-magnetic halloysite nanotubes with norfloxacin loaded (NOR-MHNTs) and controlled-release, was achieved by surface-initiated precipitation polymerization. The magnetic halloysite nanotubes exhibited better adsorption of NOR (72.10mgg−1) compared with the pristine HNTs (30.80mgg−1). Various parameters influencing the drug adsorption of the MHNTs for NOR were studied. Polymer grafted NOR-MHNTs has been designed using flexible docking in computer simulation to choose optimal monomers. NOR-MHNTs/poly (methacrylic acid or acrylamide-co-ethylene glycol dimethacrylate) nanocomposite were synthesized using NOR-MHNTs, methacrylic acid (MAA) or acrylamide (AM), ethylene glycol dimethacrylate (EGDMA) and AIBN as nanotemplate, monomers, cross linker and initiator, respectively. The magnetic nanocomposites were characterized by FTIR, TEM, XRD and VSM. The magnetic nanocomposites show superparamagnetic property and fast magnetic response (12.09emug−1). The copolymerization of monomers and cross linker led to a better sustained release of norfloxacin (>60h) due to the strong interaction formed between monomers and this cationic drug. The cumulative release rate of NOR is closely related to the cross linker amount. In conclusion, combining the advantages of the high adsorption capacity and magnetic proprieties of this biocompatible clay nanotube and the advantages of polymer shell in the enhancement of controlled-sustained release of cationic drug, a novel formulation for the sustained-controlled release of bioactive agents is developed and may have considerable potential application in targeting drug delivery system.

Abstract:In recent years, increasing interest has been accorded to polyester-based polymer microstructures, driven by their promising potential as advanced drug delivery systems. This study presents the preparation and characterization of new polymeric microparticles based on poly(ethylene brassylate-co-squaric acid) loaded with norfloxacin, a broad-spectrum antibiotic. Polymacrolactone was synthesised in mild conditions through the emulsion polymerization of bio-based and renewable monomers, ethylene brassylate, and squaric acid. The microparticles were obtained using the precipitation technique and subsequently subjected to comprehensive characterization. The impact of the copolymer/drug ratio on various properties of the new system was systematically evaluated, confirming the structure of the copolymer and the encapsulation of norfloxacin. The microspheres are approximately spherical and predominantly homogeneously distributed. The average hydrodynamic diameter of the microparticles falls between 400 and 2000 nm, a decrease that is observed with the increase in norfloxacin content. All samples showed good encapsulation efficiency and drug loading capacity, with the highest values obtained for microparticles synthesised using an equal ratio of copolymer and drug. In vitro drug release results disclose that norfloxacin molecules are released in a sustained biphasic manner for up to 24 h. Antimicrobial activity was also studied, with samples showing very good activity against E. coli and moderate activity against S. aureus and E. faecalis. In addition, HDFA human fibroblast cell cultures demonstrated the cytocompatibility of the microparticles.

Abstract:Magnetic nitrogen-doped porous carbon (MNPC) has been prepared via self-catalytic pyrolysis of bimetallic metal-organic frameworks (MOFs). The as-obtained MNPC showed favorable features for antibiotics adsorption such as high specific surface area (871 m2 g−1), high pore volume (0.75 cm3 g−1), porous structure, good graphitization degree, and rich N-doping. Moreover, the MNPC has magnetic properties due to the Co species, which is embedded with a high dispersion, so the absorbent can be easily separated. Based on the above excellent characteristics, the MNPC was used as the absorbent for norfloxacin (NOR) removal. The experimental maximum NOR adsorption capacity of MNPC was 55.12 mg g−1 at 298.15 K and a pH of 6.0 with an initial NOR concentration of 50 mg L−1. The data analysis of the kinetics revealed that the experimental data of NOR uptakes versus time agreed with the pseudo-second order model. The isotherm data analysis revealed the favorable application of the Freundlich model. Based on the adsorption results over a wide range of conditions, the dominant adsorption mechanisms were found to be pore-filling, electrostatic interaction, and the H-bond.
Long-term evolution of cytophagic histiocytic panniculitis.

Abstract:BACKGROUND Cytophagic histiocytic panniculitis is a disorder manifested by multiple subcutaneous nodules, characterized by lobular panniculitis, and occasionally accompanied by severe systemic manifestations, which require an aggressive medical approach. Several case reports with a good response to immunosuppressive therapy in the acute phase can be found in the literature. OBJECTIVE AND CONCLUSION Nevertheless, the long-term evolution of this entity has not been described yet, nor have precise recommendations regarding the most adequate posology and duration of therapy. We report the result of switching from a long-term cyclosporine therapy to tacrolimus in a patient with a severe form of cytophagic histiocytic panniculitis.
Magnetic Mesoporous Silica Spheres for Drug Targeting and Controlled Release

Abstract:Magnetically functionalized mesoporous silica spheres with different size (average diameter, A.D.) from 150 nm to 2 mu m and pore size distribution were synthesized by generating magnetic FexOy nanoparticles onto the mesoporous silica hosts using the sol-gel method. The X-ray diffraction (XRD), field emission scanning electron microscope (FESEM), N-2 adsorption/desorption results show that these composites conserved regular sphere morphology and ordered mesoporous structure after the formation of FexOy nanoparticles. XRD and X-ray photoelectron spectroscopy (XPS) analysis confirmed that the FexOy generated in these mesoporous silica hosts is mainly composed of gamma-Fe2O3. Magnetic measurements reveal that these composites with different gamma-Fe2O3 loading amounts possess super-paramagnetic properties at 300 K, and the saturation magnetization increases with increasing Fe ratio loaded. The ibuprofen-containing spheres were then coated with biodegradable poly(DL-lactide-co-glycolide) (PLGA) by the S/O/W single-emulsion method. The PLGA hybrid magnetic spheres realize a reduced initial burst with a sustained release stage lasting for 20 days. These composites with or without the PLGA can be used as potential vectors for drug targeting and controlled-release systems.
Fe3O4-SiO2 Mesoporous Core/Shell Nanoparticles for Magnetic Field-Induced Ibuprofen-Controlled Release

Abstract:Hybrid magnetic nanoparticles made up of an iron oxide, Fe3O4, core and a mesoporous SiO2 shell with high magnetization and a large surface area were proposed as an efficient drug delivery platform. The core/shell structure was synthesized by two seed-mediated growth steps combining solvothermal and sol-gel approaches and using organic molecules as a porous scaffolding template. The system presents a mean particle diameter of 30(5) nm (9 nm magnetic core diameter and 10 nm silica shell thickness) with superparamagnetic behavior, saturation magnetization of 32 emu/g, and a significant AC magnetic-field-induced heating response (SAR = 63 W/gFe3O4, measured at an amplitude of 400 Oe and a frequency of 307 kHz). Using ibuprofen as a model drug, the specific surface area (231 m2/g) of the porous structure exhibits a high molecule loading capacity (10 wt %), and controlled drug release efficiency (67%) can be achieved using the external AC magnetic field for short time periods (5 min), showing faster and higher drug desorption compared to that of similar stimulus-responsive iron oxide-based nanocarriers. In addition, it is demonstrated that the magnetic field-induced drug release shows higher efficiency compared to that of the sustained release at fixed temperatures (47 and 53% for 37 and 42 °C, respectively), considering that the maximum temperature reached during the exposure to the magnetic field is well below (31 °C). Therefore, it can be hypothesized that short periods of exposure to the oscillating field induce much greater heating within the nanoparticles than in the external solution.
Synthesis of Magnetic Nanocomposites Based on Imidazole Zeolite-8 Framework Doped with Silver Nanoparticles for Effective Removal of Norfloxacin from Effluents

Abstract:In this study, an operative technique was presented for the synthesis of the magnetically separable γ-Fe 2 O 3 @SiO 2 @ZIF8@Ag photocatalyst. The synthesized nanostructures were identified using various structural analyses, including XRD, EDX/SEM, FTIR, bandgap, and VSM. Their ability to remove norfloxacin (NOR) was then examined by studying the effects of various parameters, including photocatalyst dose, solution pH, initial NOR concentration, and reaction time. The results showed that the catalyst had the best performance, with an efficiency of 100% under UV light and 96.2%unnder visible light, at a catalyst dose of 0.4 g/L and a reaction time of 45 min. Stability tests also showed that the synthesized photocatalyst maintained its proper performance after five cycles, and its efficiency was reduced by only 4.5%. Also, a comparison between the adsorption and the photocatalytic process showed that the adsorption process removed only 42% of NOR after 60 min, whereas the photocatalytic process, under both visible and UV light irradiation, was able to eliminate 100% of NOR in the same time period. The results showed that the degradation kinetics follow the first-order kinetic model. The reaction rate constants using UV and visible lamps were 0.082 and 0.056 min − 1 , respectively, which indicates the degradation rate for UV light is 1.46 times higher compared to visible light. Also, the half-life times for the process with UV and visible light were 8.4 and 12.3 min, respectively. The average oxidation state (AOS) and carbon oxidation state (COS) of the process increased over time, indicating good degradation of NOR and conversion of non-biodegradable wastewater into biodegradable wastewater. Reactive oxygen species (ROS) assays showed that hydroxyl radicals and holes have the main role in the degradation process. Therefore, the proposed photocatalysts can be considered suitable, cost-effective, and reusable for the treatment of hospital wastewater.

Abstract:A simple and effective method was developed for norfloxacin detection using an innovative magnetic molecularly imprinted polymer (MMIP) with the aid of computer simulation. The best two functional monomers are selected according to the docking results. Four kinds of MMIP were synthesized with different functional monomers (methylacrylic acid and acrylamide) and solvents (dimethylsulfoxide and ethanol). The zeta potential was introduced to explain the interaction mechanism between MMIP and norfloxacin when the pH system was optimized. The final MMIP (MMIP-1) possesses a high efficiency recognition for norfloxacin (163.07 mu g mg(-1)) because of the thin layer of MIP (30 nm). Transmission electron microscopy shows uniform morphology for MMIP-1 (460 nm) with a core-shell structure. Fourier transform infrared spectroscopy and vibrating sample magnetometry were used to study the chemical groups and magnetic properties of the materials. MMIP-1 possesses a high imprinting factor (greater than 3). A possible mechanism for the adsorption behavior was briefly explained with the Freundlich model and rate controlled steps. Simulated pharmaceutical wastewater was used for method validation and the method was then used to research using the method with real pharmaceutical wastewater. Because of its good properties, MMIP-1 proved to be an effective material when applied for norfloxacin extraction in complex pharmaceutical wastewater with detection using high performance chromatography.
Mesoporous Silica Nanoparticles Decorated with Polycationic Dendrimers for Infection Treatment

Abstract:This work aims to provide an effective and novel solution for the treatment of infection by using nanovehicles loaded with antibiotics capable of penetrating the bacterial wall, thus increasing the antimicrobial effectiveness. These nanosystems, named "nanoantibiotics", are composed of mesoporous silica nanoparticles (MSNs), which act as nanocarriers of an antimicrobial agent (levofloxacin, LEVO) localized inside the mesopores. To provide the nanosystem of bacterial membrane interaction capability, a polycationic dendrimer, concretely the poly(propyleneimine) dendrimer of third generation (G3), was covalently grafted to the external surface of the LEVO-loaded MSNs. After physicochemical characterization of this nanoantibiotic, the release kinetics of LEVO and the antimicrobial efficacy of each released dosage were evaluated. Besides, internalization studies of the MSNs functionalized with the G3 dendrimer were carried out, showing a high penetrability throughout Gram-negative bacterial membranes. This work evidences that the synergistic combination of polycationic dendrimers as bacterial membrane permeabilization agents with LEVO-loaded MSNs triggers an efficient antimicrobial effect on Gram-negative bacterial biofilm. These positive results open up very promising expectations for their potential application in new infection therapies.
Vortex-Mixing Microfluidic Fabrication of Micafungin-Loaded Magnetite–Salicylic Acid–Silica Nanocomposite with Sustained-Release Capacity

Abstract:Iron oxide nanoparticles were synthesized using a vortex microfluidic system and subsequently functionalized with a primary shell of salicylic acid, recognized for its ability to increase the stability and biocompatibility of coated materials. In the second stage, the vortex platform was placed in a magnetic field to facilitate the growth and development of a porous silica shell. The selected drug for this study was micafungin, an antifungal agent well regarded for its effectiveness in combating fungal infections and identified as a priority compound by the World Health Organization (WHO). The resulting nanocomposite system was characterized using various techniques, including Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), transmission electron microscopy (TEM), dynamic light scattering (DLS), Brunauer–Emmett–Teller (BET) analysis, UV-Vis spectroscopy, and Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS). The synthesis method produced nanoparticles with dimensions of 5–7 nm, highlighting the advantages of the chosen approach. A desorption profile was established using a continuous-flow, UV-Vis analysis system, indicating that the bioactive compound was released slowly; after two hours, approximately 50% of the loaded micafungin was detected in the release medium. Furthermore, the results obtained from the FT-ICR MS analysis provided molecular-level confirmation, thereby supporting the release mechanism of micafungin from the nanosystem.

Abstract:Superparamagnetic Iron Oxide Nanoparticles (SPIONs) have shown great potential for being utilized in Nanocarriers (NCs) applications throughout the Drug Delivery System (DDS). However, there are several obstacles to make a practical magnetic NCs, such as low dispersity and high toxicity in the biological systems, and also low surface area for drug loading. In this work, magnetic NCs have been synthesized through a facile three-step process, first SPIONs were synthesized by the co-precipitation method, then decorated via mesoporous silica and finally the calcinated NCs functionalized with NH2 by a simple process in the ethanol solvent. The structure and morphology of the as-synthesized NCs have been characterized by the usage of different analyzing methods such as XRD, FTIR, TEM, FE-SEM, and TGA. Also, the magnetic properties have been investigated by the means of VSM throughout each step of the procedure. Lastly, we have applied the technique of N 2 adsorption-desorption to observe the surface area, pore size, and volume. Besides optimal magnetization of final nanoparticles (30 emu/gr), the as-synthesized NCs claimed the high surface area and small diameter, at 371 m 2 .g -1 and 70 nm, respectively. Moreover, the functionalized NCs have demonstrated well dispersity over a day in the PBS solution. As a result, the as-prepared nanocarrier able to overcome drug delivery obstacles and used as a potential nanocarrier owing to its small diameter, high surface area/ pore volume, optimal magnetization, and well dispersity in the biological condition.

Abstract:This review examines the latest developments in nanoscopic antibiotic formulations used to treat infections caused by bacteria. A wide range of nanocarrier platforms are discussed, including polymer-based nanoparticles (NPs), lipid-based vesicles, mesoporous silica, and other inorganic materials. The antibiotic levofloxacin (LVF) is predominantly used as a model drug given its broad-spectrum activity. Studies in this regard have evaluated drug loading and encapsulation efficiency (EE) using analytical techniques such as FTIR, DLS, and TEM. In vitro release kinetics was characterized through dialysis and fluorescence-based assays. Zone of inhibition and viability studies provided insights into antibacterial efficacy. Some approaches incorporated stimuli-responsive polymers or targeting ligands to facilitate controlled or targeted drug release. Overall, the nanocarriers demonstrated potential for sustained antibiotic levels, reduced dosing, and improved treatment of biofilms and intracellular infections compared to free drug administration. The review offers a comprehensive analysis of this promising field with implications for combating antibiotic resistance.

Abstract:In recent decades, extensive research has been conducted on nano drug delivery systems for treating inflammation. Despite this, the lack of specificity in many carriers and materials has hindered their effectiveness in enhancing treatment efficiency. To address this challenge, among numerous candidate biomaterials, stimuli-responsive biopolymers have received significant interest for precisely delivering their payload to inflamed tissues. Herein, to overcome this deficiency, pH-responsive magnetic nanocarriers were fabricated. A cationic dextran was synthesized based on the oxidation of glucose ring and conjugation of spermine. Drug-loaded magnetic dextran-spermine nanoparticles (DS-NPs) were prepared by in situ drug loading via ionic gelation. Fabricated nanoparticles have smaller size (60–100 nm), with relatively narrow size distribution (pdI = 0.175–0.251), and a positive surface charge (+ 17.1 to + 29.0 mV). The in vitro release study in simulated normal and inflamed organ environments confirmed the pH sensitivity of the drug release with a slight burst effect in normal conditions and accelerated release in acidic pH (final release < 70% and < 50% of the total encapsulated drug for pH 7.4 and pH 5.0, respectively). Antibacterial study on Staphylococcus aureus showed no changes in the bactericidal activity of encapsulated vancomycin. Cytotoxicity studies on HUVEC cells confirmed that drug-free nanoparticles exhibited no toxicity, even up to 1 mg/mL. Furthermore, the cytotoxicity of encapsulated vancomycin against HUVEC cells was lower than that of free vancomycin in all incubation times (24 h, 48 h, 72 h).

Abstract:In the present study, a novel pH-sensitive nanocarrier was prepared by grafting chitosan/polyvinylpyrrolidone (CS/PVP) on the surface of single-walled carbon nanotubes (SWCNTs). Levofloxacin (LVX), an anti-bacterial model drug, was loaded onto the resulting nanocomposite. The as-prepared nanocomposite was characterized using field emission scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy (EDX), thermogravimetric analysis (TGA), Fourier-transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD) techniques. The adsorption procedure was investigated under different sorption conditions, such as solution pH, adsorbent dosage, initial drug concentration, contact time, and temperature. The experimental data were analyzed using both non-linear and linear forms of kinetic and isotherm models. Based on the sum of squares errors and coefficient of determination values, the non-linear forms of the pseudo-2nd-order kinetic model and Langmuir isotherm model provided the best fit to the experimental data. Adsorption thermodynamic showed an exothermic and spontaneous nature of the drug sorption on the surface of the nanoadsorbent. In-vitro drug release tests were studied in simulated gastric fluid (SGF; pH = 1.2) and intestinal fluid (SIF; pH = 7.4) at 37 °C. The pH-sensitive nanocarrier indicated sustained drug release over 36 h. Nearly 99.76% of the drug was released in simulated intestinal fluid at pH = 7.4 in 36 h and 22.72% was released in simulated gastric fluid at pH = 1.2 in 30 min. The drug release profiles were well-fitted by the Korsmeyer-Peppas kinetic model, and the release mechanism of the nanocarrier was related to non-Fickian transport. Furthermore, the antimicrobial efficacy of the fabricated nanomaterials was evaluated against Staphylococcus aureus (Gram-positive). The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the nanoparticles were subsequently quantified.

Abstract:Mesoporous silica nanoparticles (MSNs) are promising drug nanocarriers for infection treatment. Many investigations have focused on evaluating the capacity of MSNs to encapsulate antibiotics and release them in a controlled fashion. However, little attention has been paid to determine the antibiotic doses released from these nanosystems that are effective against biofilm during the entire release time. Herein, we report a systematic and quantitative study of the direct effect of the antibiotic-cargo released from MSNs on Gram-positive and Gram-negative bacterial biofilms. Levofloxacin (LVX), gentamicin (GM) and rifampin (RIF) were separately loaded into pure-silica and amino-modified MSNs. This accounts for the versatility of these nanosystems since they were able to load and release different antibiotic molecules of diverse chemical nature. Biological activity curves of the released antibiotic were determined for both bacterial strains, which allowed to calculate the active doses that are effective against bacterial biofilms. Furthermore, in vitro biocompatibility assays on osteoblast-like cells were carried out at different periods of times. Albeit a slight decrease in cell viability was observed at the very initial stage, due to the initial burst antibiotic release, the biocompatibility of these nanosystems is evidenced since a recovery of cell viability was achieved after 72 h of assay. Biological activity curves for GM released from MSNs exhibited sustained patterns and antibiotic doses in the 2-6 μg/mL range up to 100 h, which were not enough to eradicate biofilm. In the case of LVX and RIF first-order kinetics featuring an initial burst effect followed by a sustained release above the MIC up to 96 h were observed. Such doses reduced by 99.9% bacterial biofilm and remained active up to 72 h with no emergence of bacterial resistance. This pioneering research opens up promising expectations in the design of personalized MSNs-based nanotherapies to treat chronic bone infection.
Enhancing the Cellular Uptake and Antibacterial Activity of Rifampicin through Encapsulation in Mesoporous Silica Nanoparticles

Abstract:An urgent demand exists for the development of novel delivery systems that efficiently transport antibacterial agents across cellular membranes for the eradication of intracellular pathogens. In this study, the clinically relevant poorly water-soluble antibiotic, rifampicin, was confined within mesoporous silica nanoparticles (MSN) to investigate their ability to serve as an efficacious nanocarrier system against small colony variants of Staphylococcus aureus (SCV S. aureus) hosted within Caco-2 cells. The surface chemistry and particle size of MSN were varied through modifications during synthesis, where 40 nm particles with high silanol group densities promoted enhanced cellular uptake. Extensive biophysical analysis was performed, using quartz crystal microbalance with dissipation (QCM-D) and total internal reflection fluorescence (TIRF) microscopy, to elucidate the mechanism of MSN adsorption onto semi-native supported lipid bilayers (snSLB) and, thus, uncover potential cellular uptake mechanisms of MSN into Caco-2 cells. Such studies revealed that MSN with reduced silanol group densities were prone to greater particle aggregation on snSLB, which was expected to restrict endocytosis. MSN adsorption and uptake into Caco-2 cells correlated well with antibacterial efficacy against SCV S. aureus, with 40 nm hydrophilic particles triggering a ~2.5-log greater reduction in colony forming units, compared to the pure rifampicin. Thus, this study provides evidence for the potential to design silica nanocarrier systems with controlled surface chemistries that can be used to re-sensitise intracellular bacteria to antibiotics by delivering them to the site of infection.

Mesoporous Silica MCM-41 as a Carriers Material for Nystatine Drug in Delivery System

Abstract:In the present study, MCM-41 was synthesis as a carrier for poorly drugs soluble in water, by the sol-gel technique. Textural and chemical characterizations of MCM-41 were carried out by X-ray diffraction (XRD), Fourier transform infrared (FTIR), scanning electron microscope (SEM), and thermal gravimetric analysis (TGA). The experimental results were analyzed mesoporous carriers MCM-41. With maximum drug loading efficiency in MCM-41 determined to be 90.74%. The NYS released was prudently studied in simulated body fluid (SBF) pH 7.4 and the results proved that the release of NYS from MCM-41 was (87.79%) after 18 hr. The data of NYS released was found to be submitted a Weibull model with a correlation coefficient of (0.995). The Historical data experimental design facilitated the formulation and optimization of sustained discover the optimal formulation to loading drug, combine process variables, mixture components and categorical factors in one design.

Abstract:The modified sol-gel approach to the synthesis of well-structured magnetic mesoporous silica nanoparticles (M-MSNs) was described, which comprised magnetic nanoparticles resided within the mesoporous nanoparticles. A diversity of surface modification was subjected to systematic investigation using organic silanes, eventually resulting in the decoration with the carboxyl (-COOH), methyl phosphonate (-PO(3)(-)), amino (-NH(2)) and phenyl (-Ph) groups on the surface of M-MSNs. The careful characterizations demonstrated that the modified M-MSNs displayed the specifically charged surfaces and differently porous characters, yet without showing any influence on the shape and size. To exploit their potential in cancer treatment, we extensively studied the drug loading capacity and sustained release behaviour of the modified M-MSNs for representative drugs. The hydrophilically modified M-MSNs with -COOH and -PO(3)(-) were beneficial for loading the water-soluble doxorubicin hydrochloride (DOX) through electrostatic attraction. The results demonstrated that M-MSNs-PO(3)(-) achieved a higher loading content and M-MSNs-COOH presented a distinct pH-responsible release behavior. On the other hand, M-MSNs-Ph displayed a controlled release rate in a short term via the weakened hydrogen bonding interaction. The cytotoxicity of modified M-MSNs to normal cells and macrophage uptake indicated that the modified M-MSNs were suitable as drug carriers. These mesoporous nanoparticles were non-toxicity to HeLa cells, while the drug-loaded nanoparticles apparently led to the unambiguous cytotoxicity as a result of the sustained release of drugs. These results have an important implication that the modified M-MSNs are promising as platforms for storing the hydrophilic or hydrophobic anticancer drugs for tumour therapy.
