Abstract:We thank Metcalf et al. for the innovative education technology they described in their paper ‘Effectiveness of an online curriculum for medical students on genetics, genetic testing and counseling’,[1] which may serve as our online learning model in education practice. Online education can provide far greater flexibility and convenience than traditional classes.[2,3] There has been a recent growth in the availability of medical curricula for students and the public, as well as a growth in medical continuing education programs, on the internet.[2,3] Students appreciate innovative educational experiences that can be accessed whenever and wherever is most convenient to the student. They are also calling for more individualized learning formats that take into account the pace of learning, motivation, and learning profile of each student. Meanwhile, use of mobile interactive media, such as smart phones, among students is increasing rapidly owing to the portability and instant accessibility of modern devices.[4] The convenience of mobile internet education enables people to learn anytime and anywhere; the breaking down of the limits of time and space that it represents can be applied to medical education.[5,6] Biochemistry and molecular biology are important parts of the basic medical curricula and examination subjects covered by the Graduate Candidate Test and National Medical Licensing Examination in China.[7] With the increasing insights into molecular mechanisms of life sciences, the American Society for Biochemistry and Molecular Biology recommends that Biochemistry and Molecular Biology programs develop curricula based on concepts, content, topics, and expected student outcomes, rather than traditional course delineations. However, teaching and learning these curricula are difficult because of the immensity and rapid evolution of the contents. Hence, there is an urgent need for pedagogical methods that enhance learning.[8] To address this need, and taking advantage of the recent progress in internet technology, we developed a mobile phone-based online biochemistry and molecular biology curriculum for medical education in China. As of May of 2017, there were some 938 million users of WeChat, developed by Tencent Inc., the most popular Chinese social media in China.[9] Apart from providing a free chat application, WeChat’s large-scale network platform hosts a vast amount of user-generated data, including texts, voice calls, videos, and images. Enabled by its immediate communication and source sharing, WeChat offers a convenient medium for delivery of education. We developed and made available a Biochemistry and Molecular Biology WeChat ‘classroom’, which includes online classes, examinations, and recent biomedical progress tracking. The online class function delivers content that is in sync with brick-and-mortar classroom teaching. The examinations are aimed at preparing students for the Graduate Candidate Test and National Medical Licensing Examination. And the recent progress tracking provides students access to information about the newest scientific and technical research advances before such information is published in textbooks. The online class program delivers PowerPoint presentations with voice-over explanations in Mandarin Chinese. Students can review high-resolution images within the PowerPoint presentation on their mobile phones. They can also preview, review, and discuss class materials online easily. The main curricula are consistent with medical major requirements, including clinical medical specialty content, but are also available for students with other majors. The program is gaining student enrolment steadily owing to the curricula being free with open sharing and encompassing interactive communication functionality. Since its introduction on 28 May 2015, 110,000 page views have been accessed. Student feedback indicates that this platform stimulates students’ learning interests and enthusiasm, while also enhancing selfdirected learning. All teachers and students are welcome to join the Biochemistry and Molecular Biology WeChat class, whose WeChat public code is BioChem_SZU. The quick response (QR) code for accessing the curriculum in shown in Figure 1 and the instructions for enrolling are provided in the Figure 1 legend. We intend to extend the WeChat class enrolment internationally via English-language offerings and to provide access to the class via other social media forums, such as Facebook and Twitter. Characterization of a novel functional porcine CD3+CD4lowCD8α+CD8β+ T-helper/memory lymphocyte subset in the respiratory tract lymphoid tissues of swine influenza A virus vaccinated pigs

Abstract:The pig is emerging as a physiologically relevant biomedical large animal model. Delineating the functional roles of porcine adaptive T-lymphocyte subsets in health and disease is of critical significance, which facilitates mechanistic understanding of antigen-specific immune memory responses. We identified a novel T-helper/memory lymphocyte subset in pigs and performed phenotypic and functional characterization of these cells under steady state and following vaccination and infection with swine influenza A virus (SwIAV). A novel subset of CD3+CD4lowCD8α+CD8β+ memory T-helper cells was identified in the blood of healthy adult pigs under homeostatic conditions. To understand the possible functional role/s of these cells, we characterized the antigen-specific T cell memory responses by multi-color flow cytometry in pigs vaccinated with a whole inactivated SwIAV vaccine, formulated with a phytoglycogen nanoparticle/STING agonist (ADU-S100) adjuvant (NanoS100-SwIAV). As a control, a commercial SwIAV vaccine was included in a heterologous challenge infection trial. The frequencies of antigen-specific IL-17A and IFNγ secreting CD3+CD4lowCD8α+CD8β+ memory T-helper cells were significantly increased in the lung draining tracheobronchial lymph nodes (TBLN) of intradermal, intramuscular and intranasal inoculated NanoS100-SwIAV vaccine and commercial vaccine administered animals. While the frequencies of antigen-specific, IFNγ secreting CD3+CD4lowCD8α+CD8β+ memory T-helper cells were significantly enhanced in the blood of intranasal and intramuscular vaccinates. These observations suggest that the CD3+CD4lowCD8α+CD8β+ T-helper/memory cells in pigs may have a protective and/or regulatory role/s in immune responses against SwIAV infection. These observations highlight the heterogeneity and plasticity of porcine CD4+ T-helper/memory cells in response to respiratory viral infection in pigs. Comprehensive systems immunology studies are needed to further decipher the cellular lineages and functional role/s of this porcine T helper/memory cell subset.

Abstract:Regulatory T cells (Tregs) expressing TCRαβ play a critical role in the maintenance of the immune system homeostasis. Tregs express the cell surface markers CD4 and CD25 as well as the transcription factor Foxp3. Foxp3+CD4+CD25+TCRαβ+ Tregs can be generated from mouse and human CD4+CD25− T cells in vitro via TGF-β induction. As growing evidences suggest that γδ T cells also have immunoregulatory function, we have attempted to identify and characterize Foxp3+ cells in mouse and human γδ T cells. We found that freshly isolated mouse splenic γδ T cells did not express Foxp3. When mouse splenocytes were stimulated with anti-TCRγδ in the presence of TGF-β, a population of Foxp3+ γδ T cells appeared, in most of which expressed CD25 as well. Compared with CD25− γδ T cells, TGF-β induced CD25+ γδ T cells not only expressed Foxp3, but also had increased TGF-β and GITR expression. Furthermore, the TGF-β induced γδ T cells mediated a potent immunosuppressive effect on anti-CD3 stimulated T cell activation and proliferation. In contrast, although a small fraction of human peripheral blood and tumor infiltrating γδ T cells expressed Foxp3, similar culture condition with anti-TCRγδ plus TGF-β failed to generate functional human Foxp3+ γδ T cells. In conclusion, our results suggest that mouse splenic Foxp3+ γδ T cells with suppressive function can be induced by TCR and TGF-β costimulation, whereas functional human Foxp3+ γδ T cells in peripheral blood could not be generated under the same condition.
Longitudinal shortening accounts for the majority of right ventricular contraction and improves after pulmonary vasodilator therapy in normal subjects and patients with pulmonary arterial hypertension.

Abstract:BACKGROUND The right ventricle has a unique contraction pattern, with a greater portion of the shortening occurring in the longitudinal plane. However, the relative contributions of longitudinal and transverse shortening to overall right ventricular (RV) function have not been quantified. We sought to quantify the proportions of longitudinal and transverse shortening to RV function in normal subjects and in patients with pulmonary arterial hypertension (PAH) at baseline and following PAH-specific therapy. METHODS The normal cohort comprised 90 subjects with normal clinical echocardiograms, whereas the PAH cohort included 36 patients, of whom 25 had echocardiograms before and after initiation of PAH-specific therapy. Assessment of RV function included tricuspid annular plane systolic excursion, RV fractional area change (RVFAC), and relative change in RV area in longitudinal and transverse planes. RESULTS Longitudinal fractional area change (LFAC) accounted for the majority of total RVFAC (77% ± 14%) in normal subjects. Among patients with PAH, longitudinal shortening still represented the majority of RVFAC, even though it was less than in normal subjects (63% ± 18%, P < .0001). Following PAH therapy, overall RV function improved (RVFAC, 30% ± 13% to 36% ± 9%; P = .026), solely because of an increase in longitudinal area change. As a result, the proportion of longitudinal shortening increased (LFAC, 58% ± 18% to 69% ± 17%; P = .002), whereas transverse shortening fell (transverse fractional area change, 42% ± 18% vs 31% ± 17%; P = .002). CONCLUSIONS Longitudinal shortening accounts for the majority of RV contraction in normal subjects and patients with PAH, although less so in PAH. Improved RV function following pulmonary vasodilator therapy occurs solely from improvements in longitudinal contraction, suggesting that longitudinal shortening may represent the afterload-responsive element of RV functional recovery.

Abstract:Many aminoacyl-tRNA synthetase and amino acid biosynthesis genes in Gram-positive bacteria are regulated by an unusual transcription termination control system, designated the T box system. This mechanism operates by controlling the activity of an intrinsic terminator located in the leader region of the mRNA, upstream of the start of the coding sequence; interaction of the leader RNA with the cognate uncharged tRNA stabilizes the formation of an antiterminator conformation of the leader RNA, which prevents formation of the competing terminator stem±loop, thereby driving readthrough transcription (Grundy and Henkin, 1993, Cell 74: 475±482; Henkin, 1994, Mol Microbiol 13: 381±387). The T box system can be recognized on the basis of a complex set of primary sequence and secondary structural elements conserved in the leader regions of the target genes, the most prominent of which is the T box itself, a 14 bp highly conserved sequence. There are 19 transcriptional units with these features in the Bacillus subtilis genome, and many others in a variety of Gram-positive organisms. To date, no example of a T box gene has been reported in a Gram-negative organism. This raises interesting questions about the evolutionary origin of this regulatory mechanism. We have now uncovered an example of an apparent T box gene in a very unusual Gram-negative organism, Symbiobacterium thermophilum. S. thermophilum was isolated as a producer of a thermostable tryptophanase (Suzuki et al. 1988, J Gen Microbiol 134: 2353±2362). A surprising observation was that the organism could be grown only in co-culture with a speci®c strain of Bacillus, which appeared to be related to B. stearothermophilus and B. coagulans. Neither cell-free extracts nor killed cells of the Bacillus strain could support independent growth of S. thermophilum. Co-culture was optimal under low aeration at 608C; under these conditions, the Bacillus strain grew ®rst, then began to lyse in late exponential phase, concomitant with growth of S. thermophilum. While the Bacillus strain could be grown in pure culture, S. thermophilum appears to be an obligate symbiont, but the requirements of the symbiosis are mysterious. The two strains could be separated by differential centrifugation, permitting isolation of S. thermophilum DNA and cloning of the tryptophanase genes (Hirahara et al. 1992, Appl Environ Microbiol 58: 2633±2642). Recent data deposited in GenBank (accession number AB010832) by K. Ueda and T. Beppu include additional sequence information covering the region downstream of the tryptophanase genes. This region includes a coding sequence with high similarity to leucyl-tRNA synthetases, and we found that the region upstream of the coding sequence includes the elements characteristic of members of the T box family. The putative S. thermophilum leuS leader region is shown in Fig. 1. This leader region contains all of the primary sequence and secondary structural features normally conserved in T box leaders (Rollins et al. 1997, Mol Microbiol 25: 411±421), including an appropriately placed CUC speci®er sequence, corresponding to the leucine codon used as the regulatory signal in the B. subtilis leuS and ilv-leu genes. The S. thermophilum leuS leader also contains a putative intrinsic transcriptional terminator, with the capacity to fold into an alternative antiterminator structure, as found in other T box genes. The terminator precedes the translation initiation region, and the leader region is preceded by a standard housekeeping-type promoter (TTGACT±17 bp spacer±GATAAT). Although the function of these putative regulatory elements has not yet been demonstrated, it appears likely that S. thermophilum leuS is in fact a member of the T box family, the ®rst to be uncovered in a Gram-negative organism. As S. thermophilum lives in such close proximity to a Bacillus strain and its lifestyle includes immersion in Bacillus cell lysate, it is possible that the leuS gene may have been acquired by horizontal gene transfer. The leuS coding region, although incomplete, exhibits highest similarity to leuS from Synechocystis (64% amino acid identity), followed by Aquifex (57%), Haemophilus (56%), Escherichia coli (54%), Rickettsia (53%), Borrelia (51%), human mitochondria (50%), Chlamydia (51%) and B. subtilis (47%). The relatively low similarity to the Bacillus gene suggests that the leuS coding region was unlikely to have been acquired recently from the Bacillus strain with which S. thermophilum lives. The G C content of the leuS coding region is 65%, which matches that of S. thermophilum, while the GC content of the Bacillus strain is 53% (Suzuki et al. 1988, J Gen Microbiol 134: 2353±2362). It remains possible that the regulatory region alone was, in fact, passed from the Bacillus strain to the S. thermophilum genome. Investigation of activation-induced markers (AIM) in porcine T cells by flow cytometry

Abstract:Activation-induced markers (AIMs) are frequently analyzed to identify re-activated human memory T cells. However, in pigs the analysis of AIMs is still not very common. Based on available antibodies, we designed a multi-color flow cytometry panel comprising pig-specific or cross-reactive antibodies against CD25, CD69, CD40L (CD154), and ICOS (CD278) combined with lineage/surface markers against CD3, CD4, and CD8α. In addition, we included an antibody against tumor necrosis factor alpha (TNF-α), to study the correlation of AIM expression with the production of this abundant T cell cytokine. The panel was tested on peripheral blood mononuclear cells (PBMCs) stimulated with phorbol 12-myristate 13-acetate (PMA)/ionomycin, Staphylococcus enterotoxin B (SEB) or PBMCs from African swine fever virus (ASFV) convalescent pigs, restimulated with homologous virus. PMA/ionomycin resulted in a massive increase of CD25/CD69 co-expressing T cells of which only a subset produced TNF-α, whereas CD40L expression was largely associated with TNF-α production. SEB stimulation triggered substantially less AIM expression than PMA/ionomycin but also here CD25/CD69 expressing T cells were identified which did not produce TNF-α. In addition, CD40L-single positive and CD25+CD69+CD40L+TNF-α- T cells were identified. In ASFV restimulated T cells TNF-α production was associated with a substantial proportion of AIM expressing T cells but also here ASFV-reactive CD25+CD69+TNF-α- T cells were identified. Within CD8α+ CD4 T cells, several CD25/CD40L/CD69/ICOS defined phenotypes expanded significantly after ASFV restimulation. Hence, the combination of AIMs tested will allow the identification of primed T cells beyond the commonly used cytokine panels, improving capabilities to identify the full breadth of antigen-specific T cells in pigs.
Identification of porcine ileal innate lymphoid cells using single-cell RNA sequencing

Abstract:Abstract Innate lymphoid cells (ILCs) are integral in maintaining intestinal defense and homeostasis, but intestinal ILCs have not been identified in pigs. Pigs are an important global food source and human biomedical model, thus identifying and characterizing porcine intestinal ILCs can have broad implications. Single-cell RNA sequencing of porcine ileal lymphocytes yielded 31,983 cells distributed across 54 clusters. Five clusters were closely related to T cell clusters yet lacked expression of the pan-T cell marker, CD3E. One cluster had high expression of genes related to type 3 immunity, including IL22, RORC, and CXCL8, indicating identification of putative ILC3s. The other four clusters expressed genes encoding for NK receptors (KLRK1, KLRC1, KLRD1) and effector molecules (CCL5, PRF1), yet they lacked expression of genes traditionally expressed by porcine NK cells (CD8A, KLRB1, HCST). Thus, we putatively identified the four clusters as ILC1s. Profiles of putative porcine ILC clusters were further compared to transcriptomic signatures of murine intestinal ILC subsets. Results indicated a relative enrichment of murine ILC3 gene signatures in the porcine ILC3 cluster, while relative enrichment of murine ILC1 gene signatures was observed for the other four clusters of porcine ILC1s. We further utilized single-cell gene expression profiles to develop a gating strategy to identify porcine intestinal ILCs as CD45+CD2+CD3ɛ−CD79α− lymphocytes at the protein level by flow cytometry. Altogether, the data allowed us to create the first transcriptomic descriptions of porcine intestinal ILCs and develop approaches to identify ILCs at the protein level, which will be applicable for future research.

Abstract:Introduction: The development and migration of T cells in the thymus and peripheral tissues are crucial for maintaining adaptive immunity in mammals. However, the regulatory mechanisms underlying T cell development and thymocyte identity formation in pigs remain largely underexplored. Method: Here, by integrating bulk and single-cell RNA-sequencing data, we investigated regulatory signatures of porcine thymus and lymph node T cells. Results: The comparison of T cell subpopulations derived from porcine thymus and lymph nodes revealed that their transcriptomic differences were influenced more by tissue origin than by T cell phenotypes, and that lymph node cells exhibited greater transcriptional diversity than thymocytes. Through weighted gene co-expression network analysis (WGCNA), we identified the key modules and candidate hub genes regulating the heterogeneity of T cell subpopulations. Further, we integrated the porcine thymocyte dataset with peripheral blood mononuclear cell (PBMC) dataset to systematically compare transcriptomic differences between T cell types from different tissues. Based on single-cell datasets, we further identified the key transcription factors (TFs) responsible for maintaining porcine thymocyte identity and unveiled that these TFs coordinately regulated the entire T cell development process. Finally, we performed GWAS of cell type-specific differentially expressed genes (DEGs) and 30 complex traits, and found that the DEGs in thymus-related and peripheral blood-related cell types, especially CD4_SP cluster and CD8-related cluster, were significantly associated with pig productive and reproductive traits. Discussion: Our findings provide an insight into T cell development and lay a foundation for further exploring the porcine immune system and genetic mechanisms underlying complex traits in pigs.

Gene expression profiles of CD4/CD8 double-positive T cells in porcine peripheral blood

Abstract:A characteristic subset of T cells, known as double positive T cells (DPTC) and expressing both cluster of differentiation 4 (CD4) and CD8, is observed in porcine peripheral blood. Previous studies suggested that DPTC might be memory cells. However, detailed phenotypes and functions of DPTC are yet to be fully elucidated and thus, the relatedness of DPTC with memory phenotypes remains unclear. In this study, DPTC gene expression profiles in peripheral blood were analyzed by DNA microarray in Experiment 1 and compared with those of CD4 single positive T cells (4SPTC) and CD8 single positive T cells (8SPTC). Expressions of IFNG, CCL5, NCK2, CCR2 and ITGB1 were higher than that of 4SPTC and 8SPTC. In contrast, expressions of CCR7 and SELL were lower than that of 4SPTC and 8SPTC. These results suggested that DPTC were either effector T cells or effector memory T cells (TEM ). Next, to determine whether DPTC were effector T cells or TEM , differences in the response of DPTC and 8SPTC against immunized/primed antigens were compared (Experiment 2). While DPTC showed quick elevation of IL2 and CD25 gene expressions against in vitro stimulation of primed/immunized antigens, 8SPTC did not. These results suggest that at least some DPTC likely belong to TEM .
Intestinal single-cell atlas reveals novel lymphocytes in pigs with similarities to human cells

Abstract:Lymphocytes can heavily influence intestinal health, but resolving intestinal lymphocyte function is challenging as the intestine contains a vastly heterogeneous mixture of cells. Pigs are an advantageous biomedical model, but deeper understanding of intestinal lymphocytes is warranted to improve model utility. Twenty-six cell types were identified in the porcine ileum by single-cell RNA sequencing and further compared with cells in human and murine ileum. Though general consensus of cell subsets across species was revealed, some porcine-specific lymphocyte subsets were identified. Differential tissue dissection and in situ analyses conferred spatial context, revealing similar locations of lymphocyte subsets in Peyer's patches and epithelium in pig-to-human comparisons. Like humans, activated and effector lymphocytes were abundant in the ileum but not periphery of pigs, suggesting tissue-specific and/or activation-associated gene expression. Gene signatures for peripheral and ileal innate lymphoid cells newly discovered in pigs were defined and highlighted similarities to human innate lymphoid cells. Overall, we reveal novel lymphocyte subsets in pigs and highlight utility of pigs for intestinal research applications.

Abstract:Since 1921 and until recently, insulin by injection has been the only treatment for patients with diabetes mellitus type 1. After pancreas transplantation, which became possible in 1977, the next logical step to cure patients with diabetes mellitus type 1 is the transplantation of the islets of Langerhans. In the last few years, the results of islet transplantation are markedly improved thanks to developments in the isolation technique and better immunosuppressive protocols. Ongoing problems in islet transplantation are allo-immunity, auto-immunity and the growing shortage of donor pancreases. Alternatives to pancreas donation, be it post-mortem or from a living donor, could be: sources for islets are xenotransplantation with the aid of pig islets and beta-cell neogenesis from embryonic stem cells or pancreatic duct cells.

Abstract:Transcutaneous delivery of vaccines to specific skin dendritic cells (DC) subsets is foreseen as a promising strategy to induce strong and specific types of immune responses such as tolerance, cytotoxicity or humoral immunity. Because of striking histological similarities between human and pig skin, pig is recognized as the most suitable model to study the cutaneous delivery of medicine. Therefore improving the knowledge on swine skin DC subsets would be highly valuable to the skin vaccine field. In this study, we showed that pig skin DC comprise the classical epidermal langerhans cells (LC) and dermal DC (DDC) that could be divided in 3 subsets according to their phenotypes: (1) the CD163(neg)/CD172a(neg), (2) the CD163(high)CD172a(pos) and (3) the CD163(low)CD172a(pos) DDC. These subtypes have the capacity to migrate from skin to lymph node since we detected them in pseudo-afferent lymph. Extensive phenotyping with a set of markers suggested that the CD163(high) DDC resemble the antibody response-inducing human skin DC/macrophages whereas the CD163(neg)CD172(low) DDC share properties with the CD8(+) T cell response-inducing murine skin CD103(pos) DC. This work, by showing similarities between human, mouse and swine skin DC, establishes pig as a model of choice for the development of transcutaneous immunisation strategies targeting DC.

Single-Cell Transcriptional Analysis of Lamina Propria Lymphocytes in the Jejunum Reveals Innate Lymphoid Cell-like Cells in Pigs

Abstract:Pigs are the most suitable model to study various therapeutic strategies and drugs for human beings, although knowledge about cell type-specific transcriptomes and heterogeneity is poorly available. Through single-cell RNA sequencing and flow cytometry analysis of the types in the jejunum of pigs, we found that innate lymphoid cells (ILCs) existed in the lamina propria lymphocytes (LPLs) of the jejunum. Then, through flow sorting of live/dead-lineage (Lin)-CD45+ cells and single-cell RNA sequencing, we found that ILCs in the porcine jejunum were mainly ILC3s, with a small number of NK cells, ILC1s, and ILC2s. ILCs coexpressed IL-7Rα, ID2, and other genes and differentially expressed RORC, GATA3, and other genes but did not express the CD3 gene. ILC3s can be divided into four subgroups, and genes such as CXCL8, CXCL2, IL-22, IL-17, and NCR2 are differentially expressed. To further detect and identify ILC3s, we verified the classification of ILCs in the porcine jejunum subgroup and the expression of related hallmark genes at the protein level by flow cytometry. For systematically characterizing ILCs in the porcine intestines, we combined our pig ILC dataset with publicly available human and mice ILC data and identified that the human and pig ILCs shared more common features than did those mouse ILCs in gene signatures and cell states. Our results showed in detail for the first time (to our knowledge) the gene expression of porcine jejunal ILCs, the subtype classification of ILCs, and the markers of various ILCs, which provide a basis for an in-depth exploration of porcine intestinal mucosal immunity.

Abstract:Effective antibody responses are essential to generate protective humoral immunity. Different inflammatory signals polarize T cells towards appropriate effector phenotypes during an infection or immunization. Th1 and Th2 cells have been associated with the polarization of humoral responses. However, T follicular helper cells (Tfh) have a unique ability to access the B cell follicle and support the germinal center (GC) responses by providing B cell help. We investigated the specialization of Tfh cells induced under type-1 and type-2 conditions. We first studied homogenous Tfh cell populations generated by adoptively transferred TCR-transgenic T cells in mice immunized with type-1 and type-2 adjuvants. Using a machine learning approach, we established a gene expression signature that discriminates Tfh cells polarized towards type-1 and type-2 response, defined as Tfh1 and Tfh2 cells. The distinct signatures of Tfh1 and Tfh2 cells were validated against datasets of Tfh cells induced following lymphocytic choriomeningitis virus (LCMV) or helminth infection. We generated single-cell and spatial transcriptomics datasets to dissect the heterogeneity of Tfh cells and their localization under the two immunizing conditions. Besides a distinct specialization of GC Tfh cells under the two immunizations and in different regions of the lymph nodes, we found a population of Gzmk + Tfh cells specific for type-1 conditions. In human individuals, we could equally identify CMV-specific Tfh cells that expressed Gzmk. Our results show that Tfh cells acquire a specialized function under distinct types of immune responses and with particular properties within the B cell follicle and the GC.

Abstract:Toll-like receptors (TLR) triggering of B cells are known to promote B cell expansion, differentiation of B cells into antibody-producing and memory cells, but the TLR responses of porcine B cells is poorly characterized. Therefore, this study investigated the response pattern of porcine B cell subsets to a large collection of TLR ligands and demonstrates that the TLR2 ligand Pam3Cys-SK4 and the TLR7/8 ligands gardiquimod and resiquimod are particularly efficient at inducing proliferation, CD25 and CCR7. This activation was also determined in B-cell subpopulations including a CD21+IgM+ subset, an IgG+ subset and two putative B1-like subsets, defined as CD21-IgMhighCD11R1+CD11c+CD14+ and CD21-IgMhigh CD11R1-CD11c+CD14- B cells. The latter two were larger and expressed higher levels of CD80/86 and spontaneous phospholipase C-γ2 phosphorylation. All porcine B-cell subsets were activated by TLR2, TLR7, and TLR9 ligands. Naïve and memory conventional B cells responded similar to TLR ligands. The CD11R1+ B1-like subset had the highest proliferative responses. While both B1-like subsets did not spontaneously secrete IgM, they were the only subsets to produce high level of TLR-induced IgM. Similar to polyclonal IgM responses, memory B cells were efficiently induced to produce specific antibodies by CpG oligodinucleotide, resiquimod, and to a weaker extend by Pam3Cys-SK4. Depletion of plasmacytoid dendritic cells (pDCs) enhanced TLR-induced antibodies. The same set of TLR ligands also induced CD40 on cDCs, pDCs, and monocytes with the exception of TLR4 ligand being unable to activate pDCs. Gardiquimod and resiquimod were particularly efficient at inducing CCR7 on pDCs. Porcine B cells expressed high levels of TLR7, but relatively little other TLR mRNA. Nevertheless, TLR2 on B cells was rapidly upregulated following stimulation, explaining the strong responses following stimulation. Subset-specific analysis of TLR expression demonstrated a comparable expression of TLR2, TLR7, and TLR9 in all B cell subsets, but TLR3 was restricted to B1-like cells, whereas TLR4 was only expressed on conventional B cells, although both at low levels. Altogether, our data describe porcine innate B1-like cells, and how different B cell subsets are involved in innate sensing.

Abstract:Understanding the pulmonary adaptive immune system of pigs is important as respiratory pathogens present a major challenge for swine producers and pigs are increasingly used to model human pulmonary diseases. Single-cell RNA sequencing (scRNAseq) has accelerated the characterization of cellular phenotypes in the pig respiratory tract under both healthy and diseased conditions. However, combining scRNAseq with recovery of paired T cell receptor (TCR) α and β chains as well as B cell receptor (BCR) heavy and light chains to interrogate their repertoires has not to our knowledge been demonstrated for pigs. Here, we developed primers to enrich porcine TCR α and β chains along with BCR κ and β light chains and IgM, IgA, and IgG heavy chains that are compatible with the 10x Genomics VDJ sequencing protocol. Using these pig-specific assays, we sequenced the T and B cell receptors of cryopreserved lung cells from CD1D-expressing and -deficient pigs after one or two infections with influenza A virus (IAV) to examine whether natural killer T (NKT) cells alter pulmonary TCR and BCR repertoire selection. We also performed paired single-cell RNA and receptor sequencing of FACS-sorted T cells longitudinally sampled from the lungs of IAV-vaccinated and -infected pigs to track clonal expansion in response to IAV exposure. All pigs presented highly diverse repertoires. Pigs re-exposed to influenza antigens from either vaccination or infection exhibited higher numbers of expanded CD4 and CD8 T cell clonotypes with activated phenotypes, suggesting potential IAV reactive T cell populations. Our results demonstrate the utility of high throughput single-cell TCR and BCR sequencing in pigs.

Abstract:Swine skin is one of the best structural models for human skin, widely used to probe drug transcutaneous passage and to test new skin vaccination devices. However, little is known about its composition in immune cells, and among them dendritic cells (DC), that are essential in the initiation of the immune response. After a first seminal work describing four different DC subpopulations in pig skin, we hereafter deepen the characterization of these cells, showing the similarities between swine DC subsets and their human counterparts. Using comparative transcriptomic study, classical phenotyping as well as in vivo and in vitro functional studies, we show that swine CD163(pos) dermal DC (DDC) are transcriptomically similar to the human CD14(pos) DDC. CD163(pos) DDC are recruited in inflamed skin, they migrate in inflamed lymph but they are not attracted toward CCL21, and they modestly activate allogeneic CD8 T cells. We also show that CD163(low) DDC are transcriptomically similar to the human CD1a(pos) DDC. CD163(low) DDC migrate toward CCL21, they activate allogeneic CD8 and CD4 T cells and, like their potential human lung counterpart, they skew CD4 T cells toward a Th17 profile. We thus conclude that swine skin is a relevant model for human skin vaccination.

Abstract:Human LSm14A has recently been found as a processing body-associated sensor of intracellular viral nucleic acids and triggers signaling for type I IFN expression. Here porcine LSm14A (pLSm14A) was cloned from the PK-15 cells. The pLSm14A ORF is 1392bp in length, encoding 463 amino acids. The putative pLSm14A contains a Sm-like domain and two arginine–glycine–glycine (RGG) boxes. The pLSm14A has high identity at the amino acid level to those of bovine, human and mouse (93.5–97.4%) and is transcribed in different tissues of pigs. In HEK293 or Marc-145 cells, pLSm14A was localized in the cytosol as P-body-like dots. Expression of pLSm14A in HEK293 or Marc-145 cells enhanced activities of IFN-β and NF-κB promoters, induced IFN-β transcription, and potentiated poly(I:C)-induced IFN-β promoter activation, indicating that pLSm14A is a potential signal molecule in the IFN-β pathway of pigs. We also found that pLSm14A-induced IFN-β promoter activity was down-regulated by porcine reproductive and respiratory syndrome virus infection in Marc-145 cells. Since pLSm14A is constitutively expressed in virtually all tissues, more research is needed to explore its role in initial phase of viral infections of pigs and its relationship with RIG-I in sensing PAMPs for type I IFN induction.
