Abstract:We report the first quantitative trait loci (QTL) mapped in an F8 recombinant inbred line (RIL) population of Lupinus angustifolius. Traits mapped were early vigour, days to flowering, height at maturity, and seed size. Twenty-two QTLs were found, located on 13 linkage groups, with alleles beneficial to the crop contributed by both parents. Early vigour was controlled by 8 QTLs on 7 linkage groups. Time to flowering was controlled by 10 QTLs and the height at maturity was found to be under the control of 4 QTLs. Seed size was linked to 2 QTLs. A region linked to the Ku gene that promotes early flowering by removal of the vernalisation requirement appeared to play a role in all 4 traits. The gene mollis controlling soft-seededness appeared to also be linked to early vigour, and iucundis controlling alkaloid production was linked to seed size. Five pairs of QTLs were found to be involved in epistasis, 2 of these having an effect on early vigour and another 3 influencing the time to opening of the first florets. Variation explained for each trait ranged from 28% for seed size, to 88% for days to flowering. We showed that it was possible to use these data to predict genotypes of superior progeny for these traits under Mediterranean conditions. QTL regions were compared on a second published linkage map and regions of conserved synteny with the model legume Medicago truncatula highlighted.

Abstract:BACKGROUND:Physiological and morphological traits of flag leaf play important roles in determining crop grain yield and biomass. In order to understand genetic basis controlling physiological and morphological traits of flag leaf, a double haploid (DH) population derived from the cross of Huaai 11 × Huadamai 6 was used to detect quantitative trait locus (QTL) underlying 7 physiological and 3 morphological traits at the pre-filling stage in year 2012 and 2013.RESULTS:Total of 38 QTLs distributed on chromosome 1H, 2H, 3H, 4H, 6H and 7H were detected, and explained 6.53% - 31.29% phenotypic variation. The QTLs flanked by marker Bmag829 and GBM1218 on chromosome 2H were associated with net photosynthetic rate (Pn), stomatal conductance (Gs), flag leaf area (LA), flag leaf length (FLL), flag leaf width (FLW), relative chlorophyll content (SPD) and leaf nitrogen concentration (LNC).CONCLUSION:Two QTL cluster regions associated with physiological and morphological traits, one each on the chromosome 2H and 7H, were observed. The two markers (Bmag829 and GBM1218) may be useful for marker assisted selection (MAS) in barley breeding.
Qtl Identification on Two Genetic Systems for Rapeseed Glucosinolate and Erucic Acid Contents over Two Seasons

Abstract:Glucosinolate and erucic acid are important plant compounds in rapeseed believed to have numerous functions in rapeseed-environment interactions. However, little is known about the QTL information related to the two different genetic systems including the embryo nuclear chromosomes and maternal plant nuclear chromosomes for glucosinolate content (GSLC) and erucic acid content (EAC) in rapeseed. Differences in QTL distribution between these two genetic systems, which control the performance of GSLC and EAC across different environmental conditions, were analyzed in the present study. A set of 202 DH populations derived from an elite hybrid cross of ‘Tapidor’ × ‘Ningyou7’ and their two backcross populations BC1F1 1 (DHs × Tapidor) and BC1F1 2 (DHs × Ningyou7) generated in two years were used as experimental materials for the study. A total of nine loci for GSLC and three loci for EAC with significant embryo additive main effects, embryo dominant main effects and/or maternal additive main effects, explaining 83.8 and 89.7 %, respectively, of their phenotypic variation, were identified. Although QTL × environment interaction effects were also detected in the present experiment, they played a minimum role in influencing the phenotypic variation. It was noted that qEAC-7-1 for EAC mapped on linkage group A7 was detected as the major QTL and could explain 68.32 % of the phenotypic variation for this trait. These results could be useful for the molecular maker-assisted breeding of GSLC and EAC quality traits based on the influence of two genetic systems.

Abstract:Introduction Retinal thickness in primary open-angle glaucoma patients was determined to establish its correlation with advancement of glaucoma neuropathy. Material and methods One hundred ninety-four patients (371 eyes, age 30–65 years) were divided in 3 groups including 50 patients with confirmed primary open-angle glaucoma, 67 primary open-angle glaucoma-suspected patients and 77 healthy persons (control group). The retinal foveola, foveal, peri-foveal and posterior pole average thickness values were determined with an RTA analyzer. For comparison, linear cup-to-disc diameter ratio (C/D), nerve fiber index (NFI) and mean defect of the retina sensitivity (MD) were also determined. Results Statistically highly significant differences in the retinal thickness and glaucoma parameters between the individual groups were observed. In the peri-foveal region, the retinal thickness in glaucoma patients was only 173.0 ±11.4 µm while in the healthy patients 201.1 ±13.1 µm. In the posterior pole region, the thickness values were only 168.1 ±11.3 µm and 195.7 ±12.3 µm, respectively. A moderate correlation between retinal thickness in peri-foveal and posterior pole regions and the C/D, NFI and MD parameters was also established (Pearson coefficients below –0.351 or above 0.284). Conclusions The retinal thickness in the peri-foveal and posterior pole regions depends on the degree of glaucoma advancement. This original observation may be a basis for acceptance of this method as a quite new tool in glaucoma diagnosis.
Mapping of Quantitative Trait Loci for Oil Content in Cottonseed Kernel

Abstract:Oil content in cottonseed is a major quality trait which when improved through breeding could enhance the competitiveness of cottonseed oil among other vegetable oils. Cottonseed oil content is a quantitative trait controlled by genes in the tetraploid embryo and tetraploid maternal plant genomes, and the knowledge of quantitative trait loci (QTLs) and the genetic effects related to oil content in both genomes could facilitate the improvement in its quality and quantity. However, till date, QTL mapping and genetic analysis related to this trait in cotton have only been conducted in the tetraploid embryo genome. In the current experiment, an IF2 population of cottonseed kernels from the random crossing of 188 intraspecific recombinant inbred lines which were derived from the hybrid of two parents, HS46 and MARCABUCAG8US-1-88, were used to simultaneously locate QTLs for oil content in the embryo and maternal plant genomes. The four QTLs found to be associated with oil content in cottonseed were: qOC-18-1 on chromosome 18; qOC-LG-11 on linkage group 11; qOC-18-2 on chromosome 18; and qOC-22 on chromosome 22. At a high selection threshold of 0.05, there was strong evidence linking the QTLs above the oil content in cottonseed. Embryo additive and dominant effects from the tetraploid embryo genome, as well as maternal additive effects from the tetraploid maternal plant genome were found to be significant contributors to genetic variation in cottonseed oil content.

Abstract:Polygalacturonase (PG) is one of the cell wall hydrolytic enzymes involving in pectin degradation. A comparison of two highly conserved duplicated PG genes, namely, Brassica campestris Male Fertility 26a (BcMF26a) and BcMF26b, revealed the different features of their expression patterns and functions. We found that these two genes were orthologous genes of At4g33440, and they originated from a chromosomal segmental duplication. Although structurally similar, their regulatory and intron sequences largely diverged. QRT-PCR analysis showed that the expression level of BcMF26b was higher than that of BcMF26a in almost all the tested organs and tissues in Brassica campestris. Promoter activity analysis showed that, at reproductive development stages, BcMF26b promoter was active in tapetum, pollen grains, and pistils, whereas BcMF26a promoter was only active in pistils. In the subcellular localization experiment, BcMF26a and BcMF26b proteins could be localized to the cell wall. When the two genes were co-inhibited, pollen intine was formed abnormally and pollen tubes could not grow or stretch. Moreover, the knockout mutants of At4g33440 delayed the growth of pollen tubes. Therefore, BcMF26a/b can participate in the construction of pollen wall by modulating intine information and BcMF26b may play a major role in co-inhibiting transformed plants.
Quantitative trait loci associated with agronomic and fiber traits of upland cotton

Abstract:Identification of quantitative trait loci (QTLs) for agronomic and fiber traits in upland cotton (Gossypium hirsutum L.) and their allelic association with molecular markers would be useful in cotton breeding. We used the mixed model approach of Zhu and Weir (1998) to analyze for QTLs associated with 19 agronomic and fiber traits across 96 F 2-derived families from the cross of two cotton lines, MARCABUCAG8US-1-88 x'HS46' (female parent). In the mixed model, molecular markers are random variables and QTLs are fixed variables. Thus, with the mixed model analysis, the QTLs are not dependent upon a particular fixed set of markers being in the model. The model also provides estimates of additive and dominance genetic effects as well as the direction of the effects of alleles from both parents. The fiber andagronomic traits, except seed index and bloom rate, were measured in F 2-derived F5 families. We mapped 100 QTLs to 60 maximum likelihood positions in 24 linkage groups. Several QTLs influence more than one trait. The most frequent association of QTLs with multiple traits was for fiber traits related to maturity and fineness. A positive correlation among traits would be beneficial for marker-assisted selection in plant breeding as well as for cloning genes for transformation. For example, in linkage group 14 near markers C117C5RI and F26ERI, a QTL is located that affects micronaire, arealometer high pressure reading, weight fineness, and wall thickness. In linkage group 19, four closely linked QTLs, located in an 8 cMregion near marker C80F1RV, influence strength, fineness, and maturity of fiber. Maximum likelihood locations such as those obtained in this study do not necessarily represent physical distances, thus, a physical map of linkage groups is also needed. © The Cotton Foundation 1998.
Dissection of the E8 locus in two early maturing Canadian soybean populations

Abstract:Soybean [ Glycine max (L.) Merr.] is a short-day crop for which breeders want to expand the cultivation range to more northern agro-environments by introgressing alleles involved in early reproductive traits. To do so, we investigated quantitative trait loci (QTL) and expression quantitative trait loci (eQTL) regions comprised within the E8 locus, a large undeciphered region (~7.0 Mbp to 44.5 Mbp) associated with early maturity located on chromosome GM04. We used a combination of two mapping algorithms, (i) inclusive composite interval mapping (ICIM) and (ii) genome-wide composite interval mapping (GCIM), to identify major and minor regions in two soybean populations (QS15524 F2:F3 and QS15544 RIL ) having fixed E1 , E2 , E3 , and E4 alleles. Using this approach, we identified three main QTL regions with high logarithm of the odds (LODs), phenotypic variation explained (PVE), and additive effects for maturity and pod-filling within the E8 region: GM04:16,974,874-17,152,230 ( E8-r1 ); GM04:35,168,111-37,664,017 ( E8-r2 ); and GM04:41,808,599-42,376,237 ( E8-r3 ). Using a five-step variant analysis pipeline, we identified Protein far-red elongated hypocotyl 3 ( Glyma.04G124300 ; E8-r1 ), E1-like-a ( Glyma.04G156400 ; E8-r2 ), Light-harvesting chlorophyll-protein complex I subunit A4 ( Glyma.04G167900 ; E8-r3 ), and Cycling dof factor 3 ( Glyma.04G168300 ; E8-r3 ) as the most promising candidate genes for these regions. A combinatorial eQTL mapping approach identified significant regulatory interactions for 13 expression traits (e-traits), including Glyma.04G050200 ( Early flowering 3/E6 locus), with the E8-r3 region. Four other important QTL regions close to or encompassing major flowering genes were also detected on chromosomes GM07, GM08, and GM16. In GM07:5,256,305-5,404,971, a missense polymorphism was detected in the candidate gene Glyma.07G058200 ( Protein suppressor of PHYA-105 ). These findings demonstrate that the locus known as E8 is regulated by at least three distinct genomic regions, all of which comprise major flowering genes.

Abstract:Introduction: Eucalyptus urophylla , E. tereticornis and their hybrids are the most important commercial forest tree species in South China where they are grown for pulpwood and solid wood production. Construction of a fine-scale genetic linkage map and detecting quantitative trait loci (QTL) for economically important traits linked to these end-uses will facilitate identification of the main candidate genes and elucidate the regulatory mechanisms. Method: A high-density consensus map (a total of 2754 SNPs with 1359.18 cM) was constructed using genotyping by sequencing (GBS) on clonal progenies of E. urophylla × tereticornis hybrids. QTL mapping of growth and wood property traits were conducted in three common garden experiments, resulting in a total of 108 QTLs. A total of 1052 candidate genes were screened by the efficient combination of QTL mapping and transcriptome analysis. Results: Only ten QTLs were found to be stable across two environments, and only one ( qSG10Stable mapped on chromosome 10, and associated with lignin syringyl-to-guaiacyl ratio) was stable across all three environments. Compared to other QTLs, qSG10Stable explained a very high level of phenotypic variation (18.4–23.6%), perhaps suggesting that QTLs with strong effects may be more stably inherited across multiple environments. Screened candidate genes were associated with some transcription factor families, such as TALE, which play an important role in the secondary growth of plant cell walls and the regulation of wood formation. Discussion: While QTLs such as qSG10Stable , found to be stable across three sites, appear to be comparatively uncommon, their identification is likely to be a key to practical QTL-based breeding. Further research involving clonally-replicated populations, deployed across multiple target planting sites, will be required to further elucidate QTL-by-environment interactions.

Abstract:Abstract Background Eucalyptus is a highly diverse genus of the Myrtaceae family and widely planted in the world for timber and pulp production. Tissue culture induced callus has become a common tool for Eucalyptus breeding, however, our knowledge about the genes related to the callus maturation and shoot regeneration is still poor. Results We set up an experiment to monitor the callus induction and callus development of two Eucalyptus species - E. camaldulensis (high embryogenic potential) and E. grandis x urophylla (low embryogenic potential). Then, we performed transcriptome sequencing for primary callus, mature callus, shoot regeneration stage callus and senescence callus. We identified 707 upregulated and 694 downregulated genes during the maturation process of the two Eucalyptus species and most of them were involved in the signaling pathways like plant hormone and MAPK. Next, we identified 135 and 142 genes that might play important roles during the callus development of E. camaldulensis and E. grandis x urophylla , respectively. Further, we found 15 DEGs shared by these two Eucalyptus species during the callus development, including Eucgr.D00640 (stem-specific protein TSJT1), Eucgr.B00171 (BTB/POZ and TAZ domain-containing protein 1), Eucgr.C00948 (zinc finger CCCH domain-containing protein 20), Eucgr.K01667 (stomatal closure-related actinbinding protein 3), Eucgr.C00663 (glutaredoxin-C10) and Eucgr.C00419 (UPF0481 protein At3g47200). Interestingly, the expression patterns of these genes displayed “N” shape in the samples. Further, we found 51 genes that were dysregulated during the callus development of E. camaldulensis but without changes in E. grandis x urophylla , such as Eucgr.B02127 (GRF1-interacting factor 1), Eucgr.C00947 (transcription factor MYB36), Eucgr.B02752 (laccase-7), Eucgr.B03985 (transcription factor MYB108), Eucgr.D00536 (GDSL esterase/lipase At5g45920) and Eucgr.B02347 (scarecrow-like protein 34). These 51 genes might be associated with the high propagation ability of Eucalyptus and 22 might be induced after the dedifferentiation. Last, we performed WGCNA to identify the co-expressed genes during the callus development of Eucalyptus and qRT-PCR experiment to validate the gene expression patterns. Conclusions This is the first time to globally study the gene profiles during the callus development of Eucalyptus. The results will improve our understanding of gene regulation and molecular mechanisms in the callus maturation and shoot regeneration.

Abstract:P>Physiological breeding is an effective way to improve crop productivity. Complex genetics and strong environmental influences can hinder progress in chlorophyll improvement during conventional cotton breeding. In this study, quantitative trait loci (QTL) mapping was conducted to better understand the genetic mechanisms of leaf chlorophyll production. Based on data from three developmental stages (early square, full bloom, and early boll opening) of a cross between Gossypium hirsutum x Gossypium barbadense, 24 QTLs were identified in a population of 140 BC1 plants. QTLs controlling chlorophyll-a and -b and total chlorophyll content at the three developmental stages explained 6.11-64.49% of the phenotypic variation. One main effect QTL for chlorophyll-a (qCa-A12-2) was detected on chromosome A12 and expressed at all three developmental stages. Two QTLs on chromosome A06, including qCb-A6-1 for chlorophyll-b and qTC-A6-1 for total chlorophyll content were expressed at both stages 1 and 3. QTLs detected in this experiment are expected to be valuable to advance cotton-breeding programmes targeted at improvement in leaf chlorophyll composition and photosynthetic efficiency.
Genetic Model Identification and Major QTL Mapping for Petiole Thickness in Non-Heading Chinese Cabbage

Abstract:Petioles of non-heading Chinese cabbage are not only an important edible part but also a conduit for nutrient transport, holding significant agricultural and research value. In this study, we conducted a comprehensive genetic analysis of petiole-related traits using a segregating population. Modern quantitative genetic approaches were applied to investigate the genetic regulation of petiole thickness. The results indicated that petiole thickness is a quantitative trait, and the identified genetic model was consistent with two pairs of additive-dominant main genes and additive-dominant polygenes (2MG-AD). BSA-seq analysis identified a major effect of QTL controlling petiole thickness on chromosome A09: 42.08–45.09 Mb, spanning 3.01 Mb, designated as QTL-BrLH9. Utilizing InDel markers, the interval was narrowed down to 51 kb, encompassing 14 genes with annotations for 10 of them. Within the interval, four mutated genes were detected. Combined with gene annotation, protein sequence analysis, and homology alignment, it was found that BraA09g063520.3C's homologous gene SMXL6 in Arabidopsis (Arabidopsis thaliana (L.) Heynh) is an inhibitor of the coding and synthesis of the strigolactone pathway. Strigolactone (SLs) plays an important role in plant growth and development. The cloning results showed that multiple frameshift mutations and non-synonymous mutations occurred on the exon. The qPCR results showed that the expression of the gene was significantly different between the two parents at the adult stage, so it was speculated that it would lead to changes in petiole thickness. BraA09g063520.3C was predicted as the final candidate gene.

Quantitative Trait Loci Controlling Plant Architectural Traits in Cotton

Abstract:Cotton plant architecture is an important characteristic influencing the suitability of specific cotton varieties in cultivation, fiber yield and quality. However, complex multigenic relationships and substantial genotype–environment interaction underlie plant architecture, and will hinder the efficient improvement of these traits in conventional cotton breeding programs. An enhanced understanding of the molecular-genetic regulation of plant morphological developmental can aid in the modification of agronomically relevant traits. In this study, an interspecific Gossypium hirsutum and Gossypium barbadense BC1 population was used to identify QTL associated with plant architectural traits. Twenty-six single QTL were identified for seven plant architecture traits. The phenotypic variation explained by an individual QTL ranged from 9.56% to 44.57%. In addition, 11 epistatic QTL for fruit branch angle (FBA), plant height (PH), main-stem leaf size (MLS), and fruiting branch internode length (FBI) explained 2.28–15.34% of the phenotypic variation in these traits. The majority of the interactions (60%) occurred between markers linked to QTL influencing the same traits. The QTL detected in this study are expected to be valuable in future breeding programs to develop cultivars exhibiting desirable cotton architecture.

Abstract:Cotton (Gossypium spp.) is an important source of edible oil and protein meals. Complex genetics and strong environmental effects hinder progress in seed quality trait breeding in this species. The use of molecular markers can improve an understanding of the genetic factors conditioning seed quality traits, and is expected to assist in selection of superior genotypes. This study was conducted to identify quantitative trail loci (QTL) associated with seed physical and nutrient traits in cotton. To achieve this objective, a population of 140 BC1S1 lines developed from a cross between'TM-1' and 'Hai7124' was evaluated in 2003 and 2004. A linkage map consisting of 918 markers from this population was used to identify QTL using QTLNetwork-2.0 software. Eleven single QTL were identified for kernel percentage, kernel oil percentage, kernel protein percentage and seven amino acids (Asp, Ser, Gly, Ile, Leu, Phe and Arg). Phenotypic variation explained by each individual QTL ranged from 10.89 to 46.28%. Two epistatic QTL for Cys and Leu were detected, explaining 9.55 and 4.43% of the phenotypic variation. These QTL detected for seed quality traits in cotton are expected to be useful for further breeding programmes targeting development of cotton with improved nutrient quality.

Abstract:BackgroundTerpenoids are abundant in the foliage of Eucalyptus, providing the characteristic smell as well as being valuable economically and influencing ecological interactions. Quantitative and qualitative inter- and intra- specific variation of terpenes is common in eucalypts.ResultsThe genome sequences of Eucalyptus grandis and E. globulus were mined for terpene synthase genes (TPS) and compared to other plant species. We investigated the relative expression of TPS in seven plant tissues and functionally characterized five TPS genes from E. grandis. Compared to other sequenced plant genomes, Eucalyptus grandis has the largest number of putative functional TPS genes of any sequenced plant. We discovered 113 and 106 putative functional TPS genes in E. grandis and E. globulus, respectively. All but one TPS from E. grandis were expressed in at least one of seven plant tissues examined. Genomic clusters of up to 20 genes were identified. Many TPS are expressed in tissues other than leaves which invites a re-evaluation of the function of terpenes in Eucalyptus.ConclusionsOur data indicate that terpenes in Eucalyptus may play a wider role in biotic and abiotic interactions than previously thought. Tissue specific expression is common and the possibility of stress induction needs further investigation. Phylogenetic comparison of the two investigated Eucalyptus species gives insight about recent evolution of different clades within the TPS gene family. While the majority of TPS genes occur in orthologous pairs some clades show evidence of recent gene duplication, as well as loss of function.

Abstract:<h3>Background</h3><p>Some broccoli (<em>Brassica oleracea</em> L. <em>italic</em>) accessions have purple sepals and cold weather would deepen the purple color, while the sepals of other broccoli lines are always green even in cold winter. The related locus or gene is still unknown. In this study, a high-density genetic map was constructed based on specific locus amplified fragment (SLAF) sequencing in a doubled-haploid segregation population with 127 individuals. And mapping of the purple sepal trait in flower heads based on phenotypic data collected during three seasons was performed.</p><h3>Results</h3><p>A genetic map was constructed, which contained 6694 SLAF markers with an average sequencing depth of 81.37-fold in the maternal line, 84-fold in the paternal line, and 15.76-fold in each individual population studied. In all of the annual data recorded, three quantitative trait loci (QTLs) were identified that were all distributed within the linkage group (LG) 1. Among them, a major locus, <em>qPH.C01–2</em>, located at 36.393 cM LG1, was consistently detected in all analysis. Besides this locus, another two minor loci, <em>qPH.C01–4</em> and <em>qPH.C01–5</em>, were identified near <em>qPH.C01–2</em>, based on the phenotypic data from spring of 2018.</p><h3>Conclusion</h3><p>The purple sepal trait could be controlled by a major single locus and two minor loci The genetic map and location of the purple sepal trait of flower heads provide an important foundation for mapping other compound traits and the identification of the genes related to purple sepal trait in broccoli.</p>

Abstract:Oilseed rape (Brassica napus) is an allotetraploid with two subgenomes descended from a common ancestor. Accordingly, its genome contains syntenic regions with many duplicate genes, some of which may have retained their original functions, whereas others may have diverged. Here, we mapped quantitative trait loci (QTL) for stem rot resistance (SRR), a disease caused by the fungus Sclerotinia sclerotiorum, and flowering time (FT) in a recombinant inbred line population. The population was genotyped using B. napus 60K single nucleotide polymorphism arrays and phenotyped in six (FT) and nine (SSR) experimental conditions or environments. In total, we detected 30 SRR QTL and 22 FT QTL and show that some of the major QTL associated with these two traits were co-localized, suggesting a genetic linkage between them. Two SRR QTL on chromosome A2 and two on chromosome C2 were shown to be syntenic, suggesting the functional conservation of these regions. We used the syntenic properties of the genomic regions to exclude genes for selection candidates responsible for QTL-associated traits. For example, 152 of the 185 genes could be excluded from a syntenic A2-C2 region. These findings will help to elucidate polyploid genomics in future studies, in addition to providing useful information for B. napus breeding programs.

Abstract:Gibberellins (GAs) are the key regulators controlling plant growth, wood production and the stress responses in perennial woody plants. The role of GA in regulating the above-mentioned processes in Eucalyptus remain largely unclear. There is still a lack of systematic identification and functional characterization of GA-related genes in Eucalyptus. In this study, a total of 59,948 expressed genes were identified from the major vegetative tissues of the E. grandis × E. urophylla using transcriptome sequencing. Then, the key gene families in each step of GA biosynthesis, degradation and signaling were investigated and compared with those of Arabidopsis, rice, and Populus. The expression profile generated using Real-time quantitative PCR showed that most of these genes exhibited diverse expression patterns in different vegetative organs and in response to abiotic stresses. Furthermore, we selectively overexpressed EguGA20ox1, EguGA20ox2 and EguGA2ox1 in both Arabidopsis and Eucalyptus via Agrobacterium tumefaciens or A. rhizogenes-mediated transformation. Though both Arabidopsis EguGA20ox1- and EguGA20ox2-overexpressing (OE) lines exhibited better vegetative growth performance, they were more sensitive to abiotic stress, unlike EguGA2ox1-OE plants, which exhibited enhanced stress resistance. Moreover, overexpression of EguGA20ox in Eucalyptus roots caused significantly accelerated hairy root initiation and elongation and improved root xylem differentiation. Our study provided a comprehensive and systematic study of the genes of the GA metabolism and signaling and identified the role of GA20ox and GA2ox in regulating plant growth, stress tolerance, and xylem development in Eucalyptus; this could benefit molecular breeding for obtaining high-yield and stress-resistant Eucalyptus cultivars.
Genetic Analysis of an F2 Population Derived from the Cotton Landrace Hopi Identified Novel Loci for Boll Glanding

Abstract:Landraces are an important reservoir of genetic variation that can expand the narrow genetic base of cultivated cotton. In this study, quantitative trait loci (QTL) analysis was conducted using an F2 population developed from crosses between the landrace Hopi and inbred TM-1. A high-density genetic map spanning 2253.11 and 1932.21 cM for the A and D sub-genomes, respectively, with an average marker interval of 1.14 cM, was generated using the CottonSNP63K array. The linkage map showed a strong co-linearity with the physical map of cotton. A total of 21 QTLs were identified, controlling plant height (1), bract type (1), boll number (1), stem color (2), boll pitting (2), fuzz fiber development (2), boll shape (3), boll point (4), and boll glanding (5). In silico analysis of the novel QTLs for boll glanding identified a total of 13 candidate genes. Analysis of tissue-specific expression of the candidate genes suggests roles for the transcription factors bHLH1, MYB2, and ZF1 in gland formation. Comparative sequencing of open reading frames identified early stop codons in all three transcription factors in Hopi. Functional validation of these genes offers avenues to reduce glanding and, consequently, lower gossypol levels in cottonseeds without compromising the defense mechanisms of the plant against biotic stresses.
