Abstract:Wheat gluten (WG) and potato protein (PP) were modified to a basic pH by NaOH to impact macromolecular and structural properties. Films were processed by compression molding (at 130 and 150 °C) of WG, PP, their chemically modified versions (MWG, MPP) and of their blends in different ratios to study the impact of chemical modification on structure, processing and tensile properties. The modification changed the molecular and secondary structure of both protein powders, through unfolding and re-polymerization, resulting in less cross-linked proteins. The β-sheet formation due to NaOH modification increased for WG and decreased for PP. Processing resulted in cross-linking of the proteins, shown by a decrease in extractability; to a higher degree for WG than for PP, despite higher β-sheet content in PP. Compression molding of MPP resulted in an increase in protein cross-linking and improved maximum stress and extensibility as compared to PP at 130 °C. The highest degree of cross-linking with improved maximum stress and extensibility was found for WG/MPP blends compared to WG/PP and MWG/MPP at 130 °C. To conclude, chemical modification of PP changed the protein structures produced under harsh industrial conditions and made the protein more reactive and attractive for use in bio-based materials processing, no such positive gains were seen for WG.

Abstract:Yoghurt style gels are a promising way to increase the consumption of plant derived proteins. However, reaching texture properties similar to those commonly known from milk yoghurts while incorporating large amounts of plant-derived proteins, is a challenge that needs to be addressed to meet consumers expectations. Therefore, this study aims to investigate the influence of pH conditions (pH 6.0 to pH 8.0) during pre-treatment on the rheological properties of fermentation induced pea protein gels with a protein content of 10%. Results showed a strong correlation between the pH value during pre-treatment and the protein solubility after pH readjustment to pH 8. Solubility was highest if pea protein was pre-treated at pH 8.0 and lowest if it was pre-treated at pH 6.0. Since only soluble aggregates are believed to participate in network formation, networks formed by pea protein pre-treated at pH 6 were coarser, than those formed by pea protein pre-treated at pH 8 owing to a lower degree of crosslinking caused by less available protein. Coarser networks and higher proportions of insoluble particles increased loss of water and lowered the storage modulus G' as well as the ability of the networks to recover after intense shearing. In particular pre-treatment at pH values below 7.0 led to gels with storage moduli of the same magnitude as those measured in commercial milk derived yoghurts. Adjusting the pH value during pre-treatment of pea protein can therefore be considered a promising approach for the customisation of texture properties while maintaining a constantly high protein content.

Abstract:Faba bean is a potential ingredient due to its high protein yield and its possible cultivation in colder climate regions. In this study, meat analogues made from faba bean protein isolate (FPI) and concentrate (FPC) blends were produced using high moisture extrusion. The aim of this study was to investigate the effect of the FPI content (FPIc), feed water content (FWC), and temperature of the long cooling die (LT) during extrusion on the mechanical and physicochemical properties as well as on the structure of the meat analogues. Increased FPIc resulted in higher values in hardness, gumminess, chewiness, and cutting strengths as well as in darker colour and decreased water absorption capacity. The effect of increased FWC on these properties was weaker and the opposite. Images from microtomography revealed that higher FPIc led to a less organised fibrous structure. In conclusion, fibrous structures can be achieved by utilising a mixture of faba bean protein ingredients, and a higher FPC content seemed to promote fibre formation in the meat analogue.

Abstract:The objective of this study was to investigate the impact of pH-shifting and oil inclusion on the textural properties of plant protein-based fermentation-induced gels resembling dairy yogurt in consistency. Gels were formed by pH12-treated or native mung bean protein (MBP) with or without emulsified coconut oil (3% w/v) and transglutaminase (0.1% w/w protein) during fermentation at 43 °C for 8 h. Quinoa flour hydrolysate was used to modulate the MBP gel network. Creep-recovery and viscoelasticity (Gʹ/Gʹʹ) tests showed that gels prepared with pH12 treated MBP were less deformable and stiffer than native MBP gels. The pH12 treated MBP gels also exhibited superior hardness and water-holding capacity over the native MBP gels. Consistently, the pH12 gels displayed more compact and denser network structures, and oil emulsion droplets as well as transglutaminase further contributed to such packing effect. 1H-LF-NMR confirmed less mobility of bulk water in pH12 gels compared with native gels.

Abstract:High-moisture extrusion is a promising thermomechanical technology extensively employed in manufacturing fibrous meat analogues from plant-based proteins, garnering considerable research attention. However, polysaccharide-based extrusion has been rarely explored. The present study investigates the effects of varying extruder barrel temperatures (130 °C–200 °C) on the texture and structure of curdlan extrudates, and highlights the formation mechanism. Results showed that the single chain of curdlan aggregates to form triple-helix chains upon extrusion, consequently enhancing the crystallinity, particularly at 170 °C. The hardness, chewiness, and mechanical properties improved with increasing barrel temperature. Moreover, barrel temperatures affected the macrostructure, the extrudates maintained intact morphologies except at 160 °C due to the melting of curdlan gel as confirmed by the differential scanning calorimetry thermogram. Microstructural analysis revealed that curdlan extrudates transited through three phases: original gel (130 °C, 140 °C, and 150 °C), transition state (160 °C), and regenerated gel (170 °C, 180 °C, 190 °C, and 200 °C). The steady state of regenerated gel (170 °C) exhibited higher crystallinity and smaller fractal dimension, resulting in a more compact and crosslinked gel network. This study elucidates the structure transition of curdlan gel at extremely high temperatures, offering valuable technical insights for developing theories and methods with respect to polysaccharide-based extrusion that may find applications in food-related fields.

Abstract:The effects of cysteine addition (0%, 0.05%, 0.10%, 0.15%, 0.20%, 0.25%) on the physicochemical properties of plant-based extrudates by high-moisture extrusion were investigated. The texturization degree, rheological properties, hardness, springiness and chewiness of the extrudates significantly improved with the addition of cysteine (<0.15%). Analysis of the microstructure showed that the addition of cysteine (<0.15%) improved the formation of the fiber structure in the extrudates. Cysteine ( 0.15%) had a negative impact on the formation of fiber structure. Analysis of secondary structure suggested that the ordered β-sheet structures gradually converted to the disordered β-turn and random coil structures with the addition of cysteine (>0.15%). In addition, analysis of odor properties with the addition of cysteine using an electronic nose showed the difference in volatile components of the extrudates mainly existed in halides, hydrocarbons, sulfides and amines. On the whole, the addition of cysteine could improve the quality of plant-based extrudates.

Abstract:As a simple and convenient technology to fabricate micron-to-nanoscale fibers with controllable structure, electrostatic spinning has produced fiber films with many natural advantages, including a large specific surface area and high porosity. Maize zein, as a major storage protein in corn, showed high hydrophobicity and has been successfully applied as a promising carrier for encapsulation and controlled release in the pharmaceutical and food areas. Proteins exhibit different physical and chemical properties at different pH values, and it is worth investigating whether this change in physical and chemical properties affects the properties of electrospun fiber films. We studied the pH effects on zein solution rheology, fiber morphology, and film properties. Rotational rheometers were used to test the rheology of the solutions and establish a correlation between solution concentration and fiber morphology. The critical concentrations calculated by the cross-equation fitting model were 17.6%, 20.1%, 20.1%, 17.1%, and 19.5% (w/v) for pH 4, 5, 6, 7, and 8, respectively. The secondary structure of zein changed with the variation in solution pH. Furthermore, we analyzed the physical properties of the zein films. The contact angles of the fiber membranes prepared with different pH spinning solutions were all above 100, while zein films formed by solvent evaporation showed hydrophilic properties. The results indicated that the rheological properties of zein solutions and the surface properties of the film were affected by the pH value. This study showed that zein solutions can be stabilized to form electrospun fibers at a variety of pH levels and offered new opportunities to further enhance the encapsulation activity of zein films for bioactive materials.

Abstract:Pea flour was extruded at 50, 70, and 90 °C. The in vitro digestibility and characteristics of native and extruded pea flour were investigated. The in vitro starch digestibility (IVSD) and in vitro protein digestibility (IVPD) of the extruded pea flour were higher than those of the native pea flour and increased with increasing extrusion temperature from 50 to 90 °C. The rapidly digestible starch increased to 28.34% at 90 °C, the highest slowly digestible starch (SDS) content was 22.70% at 50 °C, and resistant starch content decreased to 4.71% at 90 °C. The IVPD increased from 80.94% relative to the native pea flour to 90.21% at 90 °C. Improved swelling power enabled the extruded pea flour to exhibit better performance and higher breakdown viscosity and lower setback viscosity than the native pea flour demonstrated that extrusion reduced the thermal stability and retrogradation tendency. Increasing extrusion temperatures greatly reduced the relative crystallinity (based on X-ray diffraction analysis) from 32.69% relative to the native pea flour to 9.76% at 90 °C. Extrusion treatment also reduced β-sheet content (based on Fourier transform infrared spectroscopy analysis) from 36.40% relative to the native pea flour to 31.79% at 90 °C. IVPD and IVSD improved, and the SDS content increased at 50 °C and 70 °C, thereby indicating that extruded pea flour can be applied to healthy food products.

Abstract:This study investigated effects of carboxymethylcellulose sodium (CMC) on the conformational evolution of pea protein during the high moisture extrusion process. The morphological observation showed that the addition of CMC facilitated the formation of fibrous structure of pea protein. In comparison with the pea protein in the melting zone and extrudate, the combination of CMC increased the denaturation enthalpy of pea protein by 2.09 % and 2.34 %. Compared with the material in the mixing zone, the degree of grafting between CMC and pea protein in the die was enhanced by 98.95 %. In general, the supplementation of CMC depressed the exposure of hydrophobic groups in the pea protein. In the extrusion barrel, the CMC increased the unfolding of protein molecular chains while it promoted the refolding of protein chains in the die. For the extrudate, the addition of CMC decreased the contents of α-helix and β-sheet of pea protein by 9.67 % and 6.93 % while the contents of β-turn and random coil were increased, leading to changes in the molecular weight distribution of protein molecules. In conclusion, these results provided new strategies toward producing the high-quality pea protein-based meat analogues by adding CMC.

Abstract:This study aimed to investigate the structural characteristics and physicochemical properties of mung bean protein (MBP) at different pH levels. Structures and physicochemical properties were evaluated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, circular dichroism, ultraviolet visible and fluorescence spectrophotometry, differential scanning calorimetry, etc. MBP contained six bands of 64, 62, 48, 32, 25 and 23 kDa, among which the 64, 48 and 32 kDa bands consisted of glycoproteins. The β-sheets and random coils of MBP had structures close to double α-helices and β-turns at pH 3.0, 7.0 and 9.0. MBP in an alkaline environment had higher solubility and free sulfhydryl contents, smaller particle sizes, and lower surface hydrophobicity. Rheological analysis showed that MBP at pH 3.0, 5.0, 7.0 and 9.0 exhibited shear-thinning pseudoplastic fluids. A higher shear stress was needed to initiate flow at pH 5.0 than at pH 3.0, 7.0 or 9.0. MBP (130 mg/mL) formed self-supporting gels at pH 7.0 and 9.0. MBP had good foaming capacity (125.00 %), emulsion activity index (117.05 m2/g) and emulsion stability index (20.86 min) in alkaline environment. These results provide fundamental information about MBP for use in neutral and alkaline environments as a functional food ingredient.

Abstract:Collagen is an important biopolymer widely used in food, cosmetics and biomedical applications. Understanding the effect of pH on the structure and properties of collagen is beneficial for its further processing and exploitation. In this study, greenfin horse-faced filefish skin collagen (GHSC) was prepared and identified as a type I collagen. We systematically investigated the effect of pH on the structural, functional and rheological properties of GHSC. Scanning electron microscopy showed that the collagen morphology changed from an ordered stacked sheet structure to a rough silk-like structure as pH increased. Gaussian-fitted Fourier infrared spectroscopy results of the collagen revealed that it unfolded with increasing pH. Moreover, the ordered structure was reduced, and random coils became the dominant conformation. Its β-sheet and random coil contents increased from 18.43 ± 0.08 and 33.62 ± 0.17 to 19.72 ± 0.02 and 39.53 ± 1.03%, respectively, with increasing pH. α-helices and β-turns decreased from 35.00 ± 0.26 and 12.95 ± 0.01 to 29.39 ± 0.92 and 11.36 ± 0.10%, respectively. The increase in β-sheets and random coils allowed the pI-treated collagen to exhibit maximum water contact angle. The emulsification and foaming properties decreased and then increased with increasing pH in a V-shape. The increased net surface charge and β-sheets in collagen benefited its emulsification and foaming properties. The rheological results showed that the protoprotein exhibited shear-thinning properties in all pH ranges. The collagen solutions showed liquid-like behaviour in low-pH (2, 4) solutions and solid-like behaviour in high-pH (6, 7.83 and 10) solutions. Moreover, the frequency-dependent properties of the storage modulus (G′) and loss modulus (G′′) of the collagen solutions weakened with increasing pH. Collagen has considerable frequency-dependent properties of G′ and G′′ at low pH (2, 4). Thus, the importance of collagen raw material preparation for subsequent processing was emphasised, which may provide new insights into applying collagen-based materials in food, biomaterials and tissue engineering.

Abstract:The structural, thermodynamic and functional properties of soy protein hydrolysate (SPH) modified by treatment at different pH values (3, 5, and 9 and pH 7 as control) followed by ultrasound treatment (240 W, 30 min) were investigated. The treatment of SPH at alkaline pH combined with ultrasound treatment resulted in a reduction in the particle size and turbidity, enhancement in the surface negative charge and disulfide bond (SS) content, and exposure of more surface sulfhydryl (SH) groups, resulting in increased surface hydrophobicity and fluorescence intensity compared to those of the samples treated at pH 3–7. In addition, the alkaline-treated samples were more structurally stable than those treated at other pH values, having higher denaturation temperatures and enthalpies; moreover, these samples had higher solubility and emulsifying and foaming capacities. In addition, ultrasound-assisted pH treatment altered the secondary and tertiary structures of SPH by altering the covalent and non-covalent interactions, although there was no effect on the molecular weight distribution of proteins. In conclusion, ultrasound-assisted pH treatment is an effective method to improve physicochemical properties of SPH for applications in the food industry.

Abstract:The molecular mechanism of plant protein texturization under extrusion conditions was unraveled at the secondary structure level by decoupling the effects of heating, cooling and shearing on protein secondary structure. Native pea protein isolate hydrated at 50% w/w in H2O and in D2O, to allow detailed resolution of protein secondary structure, was subjected to temperature cycling in a temperature-controlled ATR-FTIR and was texturized at the gram scale by microcompounding. Upon heating without shearing, native α-helices and intramolecular-β-sheets unfold to random domains, followed by the formation of intermolecular β-sheets, inducing aggregation. During cooling, the intermolecular β-sheets become increasingly ordered, and random domains partially fold into non-native β-structures. Combined heating and shearing results in more extensive β-sheets than heating alone. The resulting β-rich structures provide for an entangled network of protein chains and a cohesive protein matrix. The effect of shear on protein association/dissociation is controlled by the specific mechanical energy (SME), with the degree of intermolecular -sheet formation increasing with increasing SME values up to ∼1000 kJ/kg, followed by a gradual decrease with further increases of the SME. The detailed molecular insights in the mechanism of plant protein texturization allows for a more controlled design of novel food products, including matrices for use in meat analogues.

Abstract:Improving the functional properties of soybean protein isolate (SPI) is essential for expanding its applications in food-related fields, given its poor solubility and gelation. This study investigated the effect of alkaline pH-shifting and mild heating on the structure of SPI, as well as the gel properties after TGase cross-linking. Solubility of SPI increased with increasing alkalinity from pH 7.0 to 12.0 at 25 °C, accompanied by decreased particle size and increased surface hydrophobicity. At pH 12.0 and 50 °C, SPI showed the highest solubility, smallest particle size, and best surface hydrophobicity. Intrinsic fluorescence spectra revealed that the protein structure unfolded after combined treatment, exposing more hydrophobic groups. TGase cross-linking modified SPI exhibited significantly increased gel hardness and water holding power. Therefore, alkaline pH transfer combined with mild heating treatment has great potential for generating functional modified SPI and provides a simple and easy method for preparing SPI with diverse gel and structural properties.

Abstract:Plant proteins are constantly gaining attention as potential substitutes for dairy proteins, due to their suitable functionality and nutritional value. This study was designed to compare the structural and functional responses of different plant protein isolates (soy, pea, lentil, and chickpea) with two commonly used dairy protein (whey protein isolates and sodium caseinate) under different pH treatments (pH 3.0, 5.0, 7.0, and 9.0). The results showed that pH had a different alteration on the structural, surface properties and functional properties of plant and dairy proteins. Plant protein generally possessed a darker color, lower solubility, emulsifying properties, and foaming capacity, whereas their foaming stability and water holding capacity were higher than those of dairy proteins. Soy protein isolates were characterized by its comparable proportion of β-turn and random coils, zeta-potential, emulsifying (30.37 m2/g), and water-holding capacity (9.03 g/g) at alkaline conditions and chickpea protein isolates showed good oil-holding capacity (3.33 g/g at pH 9) among plant proteins. Further analysis confirmed that pH had a greater influence on the structural and functional properties of proteins as compared to protein sources, particularly at acidic conditions. Overall, this study might help processors select the appropriate plant protein as dairy alternatives for their target application in plant-based food products.

Abstract:Utilization of by-products remains a critical technical challenge in the food industry. This work aimed to improve the gelation performance of insoluble soybean fiber (ISF) from okara based on structural modifications via pH-shifting during steam-cooking treatment. Okara was intrinsically compact and tightly aggregated. Its amorphous region was degraded by steam-cooking at pH 3 and inter-molecular H-bonds were reduced, resulting in increased content of cellulose from 37.86 wt% to 43.32 wt% and crystallinity degree from 7.79% to 40.10%. The compact structure of okara was dissociated into flexible fibers by steam-cooking at pH 11, allowing for higher porosity with a specific surface area reaching 1.4428 m 2 /g. However, there were no significant changes in microstructure except for a decreased particle size as pH approached neutral. The outstanding structure and adsorption capacity of ISF supported the higher gelatin performance. The hydrogels with a self-supporting and dense network structure were observed after continuous stirring from ISF steam-cooked at pH 3, 4, and 11, which was attributed to H-bonds between ISF , while okara and other ISF suspensions exhibited good fluidity. These findings provide a new method for ISF treated by steam-cooking to enable hydrogel formation for potential applications in reconstituted foods and low-calorie gel systems.

Effects of Phytase-Assisted Processing Method on Physicochemical and Functional Properties of Soy Protein Isolate

Abstract:Phytate is an important antinutritional factor in food products. In this study, a phytase-assisted processing method was used to produce low-phytate soybean protein isolate (SPI) samples, and their physicochemical and functional properties were examined. Hydrolysis condition at low temperature (room temperature) and pH 5.0 was better than that recommended by manufacturer (pH 5.0, 55 °C) at keeping the properties of SPI, so the former condition was selected to prepare SPI samples with phytate contents of 19.86-0.11 mg/g by prolonging hydrolysis time (0 (traditional method), 5, 10, 20, 40, and 60 min). Ash content (R(2) = 0.940), solubility (R(2) = 0.983), ζ-potential value (R(2) = 0.793), denaturation temperatures (β-conglycinin, R(2) = 0.941; glycinin, R(2) = 0.977), emulsifying activity index (R(2) = 0.983), foaming capacity (R(2) = 0.955), and trypsin inhibitor activity (R(2) = 0.821) of SPI were positively correlated with phytate content, whereas protein content (R(2) = 0.876), protein recovery (R(2) = 0.781), emulsifying stability index (R(2) = 0.953), foaming stability (R(2) = 0.919), gel hardness (R(2) = 0.893), and in vitro digestibility (R(2) = 0.969) were negatively correlated with phytate content. Simulated gastrointestinal digestion and subsequent dialysis showed that percentages of dialyzable Zn and Ca were increased with decreasing phytate content, whereas the amounts of dialyzable Zn and Ca revealed different behaviors: the former was increased and the latter was decreased. Circular dichroism spectra showed that secondary structure of SPI was changed by phytase. Compared with traditional processing method, the phytase-assisted processing method could produce SPI with lower phytate and higher protein contents, which had better in vitro digestibility and could be used to prepare gels with higher hardness by partially losing some other functional properties.

Abstract:Hazardous materials, i.e., chemicals that are toxic, corrosive, flammable, or explosive, are a ubiquitous aspect of modern life. They are manufactured throughout the United States, shipped by truck, train, barge, and pipeline, and stored at a wide variety of locations, including factories, military bases, and warehouses. Accidents involving hazardous materials present an added dimension of danger to emergency personnel arriving first at the scene, and have the potential to produce chemically contaminated patients who require special medical treatment. Personnel arriving first at the scene must understand how to evaluate the scene for fast and safe mitigation without endangering themselves. Chemically contaminated patients require prompt treatment, which, for optimal outcome, must begin at the scene. Although frequently the identification of the hazardous materials involved is not known initially, emergency personnel may safely provide medical care to the victims by understanding and following the principles of hazardous materials accidents and the pathophysiology of chemical injuries as presented in this paper.
