Abstract:Immune protection against new and recurrent infections relies on long-term maintenance of a highly diversified T-cell repertoire. Transcription factors cooperate to enforce T-cell metabolic quiescence and maintenance. However, less is known about the post-transcriptional networks that preserve peripheral naïve T cells. Here we describe the RNA binding proteins TIA1 and TIAL1 as key promoters of CD4 and CD8 T cell quiescence. T cells deficient in TIA1 and TIAL1 undergo uncontrolled cell proliferation in the absence of cognate antigens, leading this to a premature T-cell activation, exhaustion and death. Mechanistically, TIA1 and TIAL1 control the expression of master regulatory transcription factors, FOXP1, LEF1 and TCF1, that restrain homeostatic T-cell proliferation. In summary, our study highlights a previously unrecognised dependency on post-transcriptional gene regulation by TIA1 and TIAL1 for implementing the quiescent transcriptional programs for long survival of T cells.

Abstract:Many aspects remain elusive of the mechanisms governing T cell quiescence. Here we show that E protein activity helps to establish a quiescent program in naïve T cells. Decreased E protein activity, as the consequence of enforced expression of an Id1 transgene, led to the accumulation of CD4+CD44hi T cells. The naïve CD4+ T cells from this transgenic strain mounted a vigorous proliferative response upon TCR stimulation, as a result of direct inhibition of E protein activity. Transcriptome analyses demonstrated that Id1-tg naïve CD4+ T cells exhibited a transcriptional profile characteristic of activated CD4+ T cells, with particular enrichment in the gene set related to PI3K-AKT signaling. Western blot analysis confirmed low but constitutive activation of this pathway. Moreover, the Id1-tg CD4+ T cells displayed enhanced formation of TCR microcluster. Taken together, these data support that downregulation of E protein activity facilitates the exit of naïve T cells from quiescence.

Abstract:Regulation of autoreactive CD4 T cells is essential to maintain self-tolerance and prevent autoimmune disease. Although CD8 T regulatory (Treg) cells that recognize self-peptides restricted by Qa-1 (HLA-E in humans) inhibit autoreactive CD4 cells and attenuate experimental autoimmune encephalomyelitis (EAE), the mechanism of this interaction is unclear. We generated Qa-1 mutant knock-in mice that impair Qa-1 binding to the T cell receptor (TCR) and CD94/NKG2A receptors. Analysis of these mice showed that TCR-dependent recognition of Qa-1-peptide complexes on target CD4 cells is essential for suppression by CD8 Treg cells. Further analysis revealed that genetic disruption of the Qa-1-CD94/NKG2A interaction unleashes robust CD8 Treg cell activity that completely abolishes development of EAE.
Quiescence of Memory CD8(+) T Cells Is Mediated by Regulatory T Cells through Inhibitory Receptor CTLA-4

Abstract:Immune memory cells are poised to rapidly expand and elaborate effector functions upon reinfection yet exist in a functionally quiescent state. The paradigm is that memory T cells remain inactive due to lack of T cell receptor (TCR) stimuli. Here, we report that regulatory T (Treg) cells orchestrate memory T cell quiescence by suppressing effector and proliferation programs through inhibitory receptor, cytotoxic-T-lymphocyte-associated protein-4 (CTLA-4). Loss of Treg cells resulted in activation of genome-wide transcriptional programs characteristic of effector T cells and drove transitioning as well as established memory CD8(+) T cells toward terminally differentiated KLRG-1(hi)IL-7Rα(lo)GzmB(hi) phenotype, with compromised metabolic fitness, longevity, polyfunctionality, and protective efficacy. CTLA-4 functionally replaced Treg cells in trans to rescue memory T cell defects and restore homeostasis. These studies present the CTLA-4-CD28-CD80/CD86 axis as a potential target to accelerate vaccine-induced immunity and improve T cell memory quality in current cancer immunotherapies proposing transient Treg cell ablation.

Abstract:It is unclear how quiescence is enforced in naive T cells, but activation by foreign antigens and self-antigens is allowed, despite the presence of inhibitory signals. We showed that active transforming growth factor β (TGF-β) signaling was present in naive T cells, and T cell receptor (TCR) engagement reduced TGF-β signaling during T cell activation by downregulating TGF-β type 1 receptor (TβRI) through activation of caspase recruitment domain-containing protein 11 (CARD11) and nuclear factor κB (NF-κB). TGF-β prevented TCR-mediated TβRI downregulation, but this was abrogated by interleukin-6 (IL-6). Mitigation of TCR-mediated TβRI downregulation through overexpression of TβRI in naive and activated T cells rendered T cells less responsive and suppressed autoimmunity. Naive T cells in autoimmune patients exhibited reduced TβRI expression and increased TCR-driven proliferation compared to healthy subjects. Thus, TCR-mediated regulation of TβRI-TGF-β signaling acts as a crucial criterion to determine T cell quiescence and activation.

Abstract:A major monoaminergic innervation in infant rat neocortex, predominantly in layer IV, has been demonstrated by ultrastructural and biochemical studies after the administration of exogenous catecholamine precursors and congeners. One-third of all cortical synapses have an uptake-storage mechanism for catecholamines. In newborn cortex, the storage capacity for catecholamines is tenfold greater than the endogenous levels, and the uptake-storage mechanism matures earlier than the ability to synthesize neurotransmitter.

Abstract:There is increasing evidence that the immune response can be inhibited by several T cell subsets, including NK T cells, CD25+CD4+ T cells, and a subpopulation of CD8+ T cells. Animal model studies of multiple sclerosis have suggested an important role for suppressor CD8+ T cells in protection against disease recurrence and exacerbation. The molecular lynchpin of CD8+ suppressive activity is the murine MHC molecule Qa-1, termed HLA-E in humans. Here we summarize findings from work on Qa-1 that have begun to delineate suppressor CD8+ T cells and their mechanisms of action in the context of self tolerance and autoimmune disease.

Abstract:Under steady‐state conditions, the pool size of peripheral CD8+ T cells is maintained through turnover and survival. Beyond TCR and IL‐7R signals, the underlying mechanisms are less well understood. In the present study, we found a significant reduction of CD8+ T cell proportion in spleens but not in thymi of mice with T cell‐specific deletion of Mediator Subunit 1 (Med1). A competitive transfer of wild‐type (WT) and Med1‐deficient CD8+ T cells reproduced the phenotype in the same recipients and confirmed intrinsic role of Med1. Furthermore, we observed a comparable degree of migration and proliferation but a significant increase of cell death in Med1‐deficient CD8+ T cells compared with WT counterparts. Finally, Med1‐deficient CD8+ T cells exhibited a decreased expression of interleukin‐7 receptor α (IL‐7Rα), down‐regulation of phosphorylated‐STAT5 (pSTAT5) and Bim up‐regulation. Collectively, our study reveals a novel role of Med1 in the maintenance of CD8+ T cells through IL‐7Rα/STAT5 pathway‐mediated cell survival.

Abstract:The ability of natural killer (NK) cells to recognize and lyse activated T cells in vitro may allow these cells to participate in regulating adaptive immune response through inhibition of clonal expansion of antigen-activated T cells. This interaction may be tightly constrained by an interaction between inhibitory NKG2A receptors on NK cells and Qa-1-Qdm ligands on activated T cells (Qa-1-NKG2A interaction). We tested the hypothesis that Qa-1-NKG2A interaction protects activated T cells from NK lysis and is essential for clonal expansion and development of immunologic memory. We found that Qa-1-deficient CD4 cells fail to undergo antigen-induced or homeostatic expansion and develop autoimmune EAE disease due to increased susceptibility to NK cells lysis. Re-expression of Qa-1-Qdm on autoreactive CD4 cells rescues expansion of self-reactive T cell clones. These findings were confirmed using Qa-1 knock-in mice which express an amino acid exchange mutation (R72A) that eliminates Qa-1 binding to NKG2A but spares binding to the TCR. Dissection of the NK cell population revealed that the NKG2A+ but not NKG2A– subset of NK cells mediate lysis of Qa-1-deficient CD4 cells. Blockage of the Qa-1-NKG2A interaction that constrains NK lysis of activated T cells resulted in potent NK-dependent elimination of activated autoreactive T cells and amelioration of autoimmune EAE. These findings extend the functional reach of the NK system to include regulation of adaptive T cell responses and demonstrate that antibody-mediated blockade of NKG2A-Qdm interactions as a new clinical approach to eliminate antigen-activated T cells in the context of autoimmune disease or transplantation.

Abstract:The mouse protein Qa-1 (HLA-E in humans) is essential for immunological protection and immune regulation. Although Qa-1 has been linked to CD8 T cell–dependent suppression, the physiological relevance of this observation is unclear. We generated mice deficient in Qa-1 to develop an understanding of this process. Qa-1-deficient mice develop exaggerated secondary CD4 responses to foreign and self peptides. Enhanced responses to proteolipid protein self peptide were associated with resistance of Qa-1-deficient CD4 T cells to Qa-1-restricted CD8 T suppressor activity and increased susceptibility to experimental autoimmune encephalomyelitis. These findings delineate a Qa-1-dependent T cell–T cell inhibitory interaction that prevents the pathogenic expansion of autoreactive CD4 T cell populations and consequent autoimmune disease.
The LncRNA Snhg1 is Essential for Memory CD8 T Cell Establishment

Abstract:Abstract The efficient induction and long-term persistence of pathogen-specific memory CD8 T cells are pivotal to rapidly curb the reinfection. Recent evidence indicates that lncRNAs possess higher lineage and stage specificity than mRNAs in T cell development, differentiation and function. However, the pivotal lncRNAs regulating CD8 T cell differentiation remain largely unclear. Through profiling of CD8 T cell lineages in this study, the lncRNA Snhg1 with the conserved on-off-on expression pattern in naïve-effector-memory CD8 T cells from mouse to human, is found to be required for memory CD8 T cell generation. Further, Snhg1 is found interacting with the highly conserved vesicle trafficking protein Vps13D to facilitate IL-7Rα membrane location specifically. IL-7 has been reported to sustain memory survival through STAT5-BCL2 axis, but whether IL-7 can directly promote memory differentiation remain cloudy. In this research, Snhg1/Vps13D depletion impair memory formation through IL-7-STAT3-TCF1-Blimp1 axis. Whether TCF-1 is induced by canonical Wnt-β-Catenin signaling in memory CD8 generation remain controversial. This work finds gene Tcf7 is regulated by direct binding of p-STAT3 in CD8 stimulated with IL-7. Furthermore, TCF-1 is found promoting memory with blocking effector mainly through impeding Prdm1 expression directly to balance Blimp1-BCL6 and Blimp1-ID3 axis. Thus, this study proposes a model for lncRNA as critical factor that couples membrane trafficking of cytokine receptor with transcriptional induction to promote memory CD8 T cell establishment.
Manipulation of Qa‐1‐restricted CD8 Suppressor Cell Activity in Experimental Autoimmune Encephalomyelitis

Abstract:Peripheral regulation of CD4 T cell responses by other cell types is essential for the maintenance of self-tolerance and prevention of autoimmune disease. Our previous studies have revealed a role for Qa-1-restricted CD8 suppressor cell activity in controlling EAE development. Here we have extended our understanding of this process by creating two Qa-1 mutant knock-in mouse strains according to its ability to bind the two different receptors: the T-cell receptor (TCR) and the CD94/NKG2A receptors. The Qa-1 D227K mutant carries a point mutation at its CD8 binding region which prevents Qa1/CD8 interaction, where as Qa1 R72A mutant cannot bind CD94/NKG2A receptors. Studies from Qa1 D227K mice provided direct molecular evidence that Qa-1 mediated suppression is linked to its ability to stimulate CD8+ T cells through TCR engagement, which is dependent on the binding to the CD8 co-receptor. More importantly, by using Qa1 R72A mutant CD4 T cells, we found out that CD8 suppressive activity is inhibited by the Qa-1-CD94/NKG2A interaction between target CD4 T cells and CD8 suppressor cells. Disruption of this interaction by the R72A Qa1 mutant leads to enhanced CD8 suppressive activity, which in turn, diminishes the development of EAE induced by pathogenic CD4 T cells. Finally, we showed that blockade of the Qa1-NKG2A interaction by antibody can mimic the effect of the R72A genetic mutant and can be used to treat EAE.

Abstract:Naive T cells are metabolically quiescent. Here, we report an unexpected role of endoplasmic reticulum (ER) Ca2+ in preserving T cell metabolic quiescence. TMEM41B, an ER-resident membrane protein previously known for its crucial roles in autophagy, lipid scrabbling and viral infections, is identified as a novel type of concentration-dependent ER Ca2+ release channel. Ablation of TMEM41B induces ER Ca2+ overload, triggering the upregulation of IL-2 and IL-7 receptors in naive T cells. Consequently, this leads to increased basal signaling of the JAK-STAT, AKT-mTOR, and MAPK pathways, propelling TMEM41B-deficient naive T cells into a metabolically activated yet immunologically naive state. ER Ca2+ overload also downregulates CD5, a suppressor of TCR signaling, thereby reducing the activation threshold of TMEM41B-deficient T cells, resulting in attenuated tolerance and heightened T cell responses during infections. In summary, TMEM41B-mediated ER Ca2+ release is a pivotal determinant governing metabolic quiescence and responsiveness of naive T cells. ### Competing Interest Statement The authors have declared no competing interest.

Abstract:The life cycle of many cell types can hinge on the presence of death factors that can control programmed cell death. The Fas-mediated apoptotic pathway has been implicated in controlling apoptosis during spermatogenesis in a number of mammalian species. In the human, the presence of nuclear DNA damage in ejaculated spermatozoa has pointed to a possible role for apoptosis during spermatogenesis. The presence of other molecular markers of apoptosis has, however, not been shown. More importantly, differences in these markers have not been investigated in men with normal and abnormal sperm parameters. In this study we examine for the presence of the cell surface protein Fas in ejaculated human spermatozoa. Ejaculated spermatozoa (55 samples) were labeled with anti-human Fas antibody and the number of spermatozoa displaying Fas were counted using a fluorescence-activated cell sorter (FACS). In 30/31 (96.8%) normal males (>20 million sperm per milliliter), less than 10% of the spermatozoa were Fas positive. In contrast, 14/24 (58.3%) oligozoospermic samples (<20 million sperm per milliliter) contained more than 10% Fas-positive spermatozoa. Similar differences were observed in men whose spermatozoa had poor motility and morphology. These results indicate that apoptosis is a major mechanism in regulating spermatogenesis in the human and that there are clear differences in molecular markers of apoptosis between males with normal and abnormal sperm parameters. We propose that the presence of Fas-labeled spermatozoa in the ejaculate of these men is indicative of an "abortive apoptosis" having taken place, whereby the normal apoptotic mechanisms have misfunctioned, have been overridden, or have not been completed.
CD8+ T Cells Mediate Lethal Lung Pathology in the Absence of PD-L1 and Type I Interferon Signalling following LCMV Infection

Abstract:CD8+ T cells are critical to the adaptive immune response against viral pathogens. However, overwhelming antigen exposure can result in their exhaustion, characterised by reduced effector function, failure to clear virus, and the upregulation of inhibitory receptors, including programmed cell death 1 (PD-1). However, exhausted T cell responses can be "re-invigorated" by inhibiting PD-1 or the primary ligand of PD-1: PD-L1. Further, the absence of the type I interferon receptor IFNAR1 also results in T cell exhaustion and virus persistence in lymphocytic choriomeningitis virus Armstrong (LCMV-Arm)-infected mice. In this study, utilizing single- and double-knockout mice, we aimed to determine whether ablation of PD-1 could restore T cell functionality in the absence of IFNAR1 signalling in LCMV-Arm-infected mice. Surprisingly, this did not re-invigorate the T cell response and instead, it converted chronic LCMV-Arm infection into a lethal disease characterized by severe lung inflammation with an infiltration of neutrophils and T cells. Depletion of CD8+ T cells, but not neutrophils, rescued mice from lethal disease, demonstrating that IFNAR1 is required to prevent T cell exhaustion and virus persistence in LCMV-Arm infection, and in the absence of IFNAR1, PD-L1 is required for survival. This reveals an important interplay between IFNAR1 and PD-L1 with implications for therapeutics targeting these pathways.

Abstract:The generation of effector CD8+ T cells (TEFF) requires activation of naive CD8+ T cells (TN) by dendritic cells (DCs) within lymphoid tissue. To date, it remains elusive how the duration of TN-DC interactions and integration of activation signals are controlled in vivo. Here, we report that lymphoid stroma-secreted ligands for CCR7 constrained interaction duration by gradually inducing CD8+ T cell release from DCs. At late time points of interactions, CCR7 ligands repositioned the F-actin-promoting factor DOCK2 away from the DC interface to enable CD8+ T cell detachment, proliferation onset and acquisition of cytotoxicity. Lack of CCR7 signaling, as during ex vivo activation or in chronically inflamed lymphoid tissue, caused sustained T cell-DC interactions, and generated dysfunctional TEFF with high expression of inhibitory receptors, impaired antimicrobial activity, and poor recall responses. In sum, our findings uncover that lymphoid stromal chemokines act as built-in "disruptors" of T cell-DC interactions for long-term preservation of TEFF functionality.

Abstract:This paper presents a common-path (CP) swept-source optical coherence tomography (SSOCT) system based on a special endoscopic probe design with an in-line internal reflection as the reference and a two-axis electrothermal microelectromechanical system mirror for image scanning. The rear surface of a gradient reflective index (GRIN) lens inside the probe is set as the reference reflection plane. The length of the GRIN lens is optimized to eliminate the artifacts in SSOCT images successfully. Doppler OCT is also demonstrated based on the CP endoscopic probe. The diameter of the probe is only 2.5 mm, so it can be easily inserted into the biopsy channel of traditional endoscopes to access human internal organs for in vivo diagnoses.

In the response to acute lymphocytic choriomeningitis virus infection in a mouse model, CD160 ablation resulted in the failure of the development of all three memory CD8+ T cell subsets (central, effective, and tissue-resident memory), concomitant with a skewed differentiation into short-lived effector T cells. Such memory-related defect was manifested by a diminished protection from viral rechallenge. Mechanistically, CD160 deficiency led to downregulation of 4-1BB in activated CD8+ T cells, which contributes to the impaired cell survival and decreased respiratory capacity. The nexus between CD160 and 4-1BB was substantiated by the observation that ectopic introduction of 4-1BB was able to largely complement the loss of CD160 in memory CD8+ T cell development. Collectively, our studies discovered that CD160, once thought to be a coinhibitor of T cell signaling, is an essential promoter of memory CD8+ T cell development via activation of the costimulatory molecule 4-1BB.

Abstract:OBJECTIVES The objective of this study was to evaluate the surgical times for removal of ovaries, and the frequency of intra-operative complications with two different instruments used for canine laparoscopic ovariectomy. METHODS A randomised prospective clinical study. Laparoscopic ovariectomy was performed under general anaesthesia on 10 healthy female dogs admitted for elective ovariectomy. Each ovary was randomly assigned to removal by use of either a SonoSurg™ ultrasonic surgical device (Olympus), or a LigaSure™ vessel-sealing system (Valleylab/Covidien). Surgical time for removal of each ovary was measured and the frequency of intra-operative complications was recorded. A follow-up was made by interview with the owners, 2·5 years after surgery. RESULTS The two techniques did not statistically differ with regard to surgical time, or frequency of complications. All owners were very satisfied with the procedure. CLINICAL RELEVANCE Both the SonoSurg™ and the LigaSure™ devices appear to be effective, safe and easy to use when performing laparoscopic ovariectomy in dogs.
