Abstract:All organisms experience fundamental conflicts between divergent metabolic processes. In plants, a pivotal conflict occurs between allocation to growth, which accelerates resource acquisition, and to defense, which protects existing tissue against herbivory. Trade-offs between growth and defense traits are not universally observed, and a central prediction of plant evolutionary ecology is that context-dependence of these trade-offs contributes to the maintenance of intraspecific variation in defense [Züst and Agrawal, Annu. Rev. Plant Biol., 68, 513-534 (2017)]. This prediction has rarely been tested, however, and the evolutionary consequences of growth-defense trade-offs in different environments are poorly understood, especially in long-lived species [Cipollini et al., Annual Plant Reviews (Wiley, 2014), pp. 263-307]. Here we show that intraspecific trait trade-offs, even when fixed across divergent environments, interact with competition to drive natural selection of tree genotypes corresponding to their growth-defense phenotypes. Our results show that a functional trait trade-off, when coupled with environmental variation, causes real-time divergence in the genetic architecture of tree populations in an experimental setting. Specifically, competitive selection for faster growth resulted in dominance by fast-growing tree genotypes that were poorly defended against natural enemies. This outcome is a signature example of eco-evolutionary dynamics: Competitive interactions affected microevolutionary trajectories on a timescale relevant to subsequent ecological interactions [Brunner et al., Funct. Ecol. 33, 7-12 (2019)]. Eco-evolutionary drivers of tree growth and defense are thus critical to stand-level trait variation, which structures communities and ecosystems over expansive spatiotemporal scales.

Abstract:Plants respond to rapid environmental change in ways that depend on both their genetic identity and their phenotypic plasticity, impacting their survival as well as associated ecosystems. However, genetic and environmental effects on phenotype are difficult to quantify across large spatial scales and through time. Leaf hyperspectral reflectance offers a potentially robust approach to map these effects from local to landscape levels. Using a handheld field spectrometer, we analyzed leaf‐level hyperspectral reflectance of the foundation tree species Populus fremontii in wild populations and in three 6‐year‐old experimental common gardens spanning a steep climatic gradient. First, we show that genetic variation among populations and among clonal genotypes is detectable with leaf spectra, using both multivariate and univariate approaches. Spectra predicted population identity with 100% accuracy among trees in the wild, 87%–98% accuracy within a common garden, and 86% accuracy across different environments. Multiple spectral indices of plant health had significant heritability, with genotype accounting for 10%–23% of spectral variation within populations and 14%–48% of the variation across all populations. Second, we found gene by environment interactions leading to population‐specific shifts in the spectral phenotype across common garden environments. Spectral indices indicate that genetically divergent populations made unique adjustments to their chlorophyll and water content in response to the same environmental stresses, so that detecting genetic identity is critical to predicting tree response to change. Third, spectral indicators of greenness and photosynthetic efficiency decreased when populations were transferred to growing environments with higher mean annual maximum temperatures relative to home conditions. This result suggests altered physiological strategies further from the conditions to which plants are locally adapted. Transfers to cooler environments had fewer negative effects, demonstrating that plant spectra show directionality in plant performance adjustments. Thus, leaf reflectance data can detect both local adaptation and plastic shifts in plant physiology, informing strategic restoration and conservation decisions by enabling high resolution tracking of genetic and phenotypic changes in response to climate change.

Abstract:Climate change represents an existential threat to many forest ecosystems because tree populations are often adapted to local climate means and variability. If tree populations cannot migrate or adapt, they risk becoming increasingly maladapted with climate change. This emerging mismatch underscores the need for climate adaptive management techniques, such as assisted migration of tree species, to help mitigate climate change impacts on forest ecosystems. Although biotic and abiotic factors are known to constrain tree establishment success, the extent to which they may determine the success of assisted migration plantings is poorly understood. Thus, defining the extent to which trees affect—and in turn are affected by local soil environments and microbial communities (i.e., plant-soil feedbacks; hereafter PSFs) remains important for guiding effective climate-adaptive forest management. Our objectives were to synthesize the current state of knowledge about the direction and magnitude of PSF effects on temperate tree species of eastern North America, and to identify key hypotheses important for guiding future research. To accomplish these goals, we conducted a meta-analysis of 26 peer-reviewed publications that addressed our criteria. Our compiled database included 61 tree species and was composed primarily of short-term greenhouse experiments that studied PSF effects by manipulating the soil biota in three ways: (1) soil was previously inoculated by a conspecific or heterospecific tree species (i.e., home vs. away), (2) soil was live or sterilized, or (3) soil was untreated or treated with fungicide. We found that PSF had significant effects on tree growth, with the direction and magnitude of PSF strongly dependent on tree mycorrhizal guild. Arbuscular mycorrhizal tree species grown in live or home soils grew 13–33% less than those in sterile or away soils, while ectomycorrhizal tree species grew 11–44% more in live or home than sterile or away soils. PSF effects were associated with several plant functional traits, including specific leaf area, tissue nitrogen, and specific root length. We provide suggestions on incorporating PSFs into assisted migration trials and outline key knowledge gaps for future research. Our synthesis of context-dependent effects of PSFs on tree performance will help inform management decisions involving assisted migration.

Abstract:Resources play an important role in shaping the evolution of plant defensive strategies. How resource availability influences overall phytochemical diversity within species, and how this in‐turn affects herbivore damage is not well understood. Using Monarda fistulosa, a species that produces diverse terpene compounds and distinct chemotypes (i.e. thymol or carvacrol), we addressed three questions: 1) how do phytochemical components (i.e. total concentration, Shannon diversity, and chemotypes) vary between distinct populations from two large, widely separated regions that differ in climate and productivity? 2) How do these components influence attack by different herbivore species? 3) What are the growth costs of higher phytochemical levels in different chemotypes? Seeds were collected from 12 populations spread across two regions that differ in precipitation‐driven productivity, six from a low‐productivity environment in Montana and six from a high‐productivity environment in Wisconsin, and grown in a common garden in Wisconsin. We analyzed terpene concentrations, quantified the number of herbivores and damage, and measured plant above‐ground biomass. Plants from Montana exhibited higher terpene concentrations but lower diversity, whereas those from Wisconsin displayed lower terpene concentrations but higher diversity. Plants from Wisconsin received more damage from a specialist seed predator and leaf‐chewing herbivores, although abundance of a specialist aphid was lower, compared to plants from Montana. Total terpene concentration emerged as the primary predictor of herbivore damage, although some differences were observed between region of origin and with terpene richness. Herbivores exhibited mixed responses to phytochemistry; some herbivores were negatively affected, while a specialized leaf galler responded positively. Costs of producing defenses were evident by negative correlations between plant biomass and total terpene concentration (but not chemical richness or Shannon diversity). Our study revealed that regional differences in productivity can influence the evolution of phytochemical concentration and diversity, which, in turn, affects plant damage and incurs growth‐related costs.

Abstract:In long‐lived tree populations, when environmental change outpaces rates of evolutionary adaptation, plasticity in traits related to stress tolerance, dormancy, and dispersal may be vital for preventing extinction. While a population's genetic background partly determines its ability to adapt to a changing environment, so too do the many types of epigenetic modifications that occur within and among populations, which vary on timescales orders of magnitude faster than the emergence of new beneficial alleles. Consequently, phenotypic plasticity driven by epigenetic modification may be especially critical for sessile, long‐lived organisms such as trees that must rely on this plasticity to keep pace with rapid anthropogenic environmental change. While studies have reported large effects of DNA methylation, histone modification, and non‐coding RNAs on the expression of stress‐tolerance genes and resulting phenotypic responses, little is known about the role of these effects in non‐model plants and particularly in trees. Here, we review new findings in plant epigenetics with particular relevance to the ability of trees to adapt to or escape stressors associated with rapid climate change. Such findings include specific epigenetic influences over drought, heat, and salinity tolerance, as well as dormancy and dispersal traits. We also highlight promising findings concerning transgenerational inheritance of an epigenetic 'stress memory' in plants. As epigenetic information is becoming increasingly easy to obtain, we close by outlining ways in which ecologists can use epigenetic information better to inform population management and forecasting efforts. Understanding the molecular mechanisms behind phenotypic plasticity and stress memory in tree species offers a promising path towards a mechanistic understanding of trees' responses to climate change.

Constitutive and Induced Defenses in Long-lived Pines Do Not Trade Off but Are Influenced by Climate

Abstract:Plants resist herbivores and pathogens by using constitutive (baseline) and inducible (change in defense after an attack) defenses. Inducibility has long been predicted to trade off with constitutive defense, reflecting the economic use of resources. However, empirical evidence for such tradeoffs is variable, and we still lack understanding about when and where defense trade-offs occur. We tested for tradeoffs between constitutive and induced defenses in natural populations of three species of long-lived pines ( Pinus balfouriana, P. flexilis, P. longaeva ) that differ greatly in constitutive defense and resistance to mountain pine beetle (MPB, Dendroctonus ponderosae ). We also assessed how climate influenced constitutive and inducible defenses. At seven high-elevation sites in the western U.S., we simulated MPB attack to induce defenses and measured concentrations of terpene-based phloem defenses on days 0, 15, and 30. Constitutive and induced defenses did not trade off among or within species. Simulated MPB attack induced large increases in defense concentrations in all species independent of constitutive levels. MPB and its symbiotic fungi typically kill trees and thus could be selective forces maintaining strong inducibility within and among species. The contrasting constitutive concentrations in these species could be driven by the adaptation for specializing in harsh, high-elevation environments (e.g., P. balfouriana and P. longaeva ) or by competition (e.g., P. flexilis ), though these hypotheses have not been empirically examined. Climate influenced defenses, with the greatest concentrations of constitutive and induced defenses occurring at the coldest and driest sites. The interactions between climate and defenses have implications for these species under climate change.

Conservation prioritization based on past cascading climatic effects on genetic diversity and population size dynamics: Insights from a temperate tree species

Abstract:The biogeographic history affects adaptive evolution by altering the pattern of genetic variation and provides the background upon which other evolutionary processes are operating. Assessment of the consequence of such eco‐evolutionary interactions is crucial to population's genetic diversity maintenance. Yet, considering how biogeographic and multiple evolutionary processes interact in complex environments to alter genetic diversity in conservation planning remains unresolved. The Pacific Northwest. Herein I document the impact of geographic landscapes and past climatic fluctuations on the evolutionary processes that drive population divergence of Populus trichocarpa by assembling genomic, climatic and species occurrence data. Based on inferences of demographic history of the species and tests for niche divergence, results show that the British Columbia (BC)‐North and BC‐South populations are ancestral and subject to more recent bottlenecks than the Oregon population. Consistently, ecological niche modelling illustrates that compared to the Last Glacial Maximum (~21 Ka), the present niche suitability of the BC‐North and ‐South populations is overall marginally inferior to that of the Oregon population. However, genomic analysis demonstrates that the Oregon harbours the lowest genetic diversity, possibly due to an extended bottleneck experienced by the population. Model prediction indicates that in the 2050s, the impact of climate change is minor compared to the niche suitability of the species as a whole, but climate change will likely result in considerable genetic shifts over two generations especially in the Oregon and BC‐South populations, which will attenuate along a northward gradient. This study underpins the long‐lasting impact of demographic histories and natural selection on genetic diversity, rather than highlighting the consequence of recent demographic events (e.g. bottleneck in the Holocene epoch; ~12 Ka) potentially cascading to eroding genetic diversity and yielding conservation urgency.

Abstract:One of the best known ecological consequences of climate change is the advancement of spring phenology. Yet, we lack insights into how changes in climate interact with intraspecific genetic variation in shaping spring and autumn phenology, and how such changes in phenology will translate into seasonal dynamics of tree‐associated organisms. To elucidate the impact of warming and tree genotype on spring and autumn phenology, as well as the consequences for the population dynamics of a fungal pathogen Erysiphe alphitoides and plant‐feeding insect Tuberculatus annulatus, we conducted an active field heating experiment using grafts of five oak genotypes Quercus robur. We found that experimental warming generally advanced oak bud burst in spring and delayed leaf senescence in autumn, while additional variation was explained by tree genotype and warming‐by‐genotype interactions. Warming or tree genotype did not affect disease levels at the beginning of the season, but shaped both disease levels and aphid density during the latter part of the season. Overall, our findings demonstrate that elevated temperature and genetic variation affect spring and autumn phenology, as well as the seasonal dynamics of higher trophic levels. Such effects may be either direct (i.e. temperature affecting tree phenology and attack independently) or indirect (as due to climate‐induced changes in plant traits or the synchrony between trees and their attackers). To achieve a predictive understanding of the ecological responses and potential evolutionary changes of natural food webs in response to climate warming, we should merge the frameworks of global warming and community genetics.This article is protected by copyright. Genetically based variation in fitness and carbon assimilation among bur oak populations

Abstract:Ongoing climate change will negatively impact tree populations unless they are able to acclimate to the changes in their local environment. Effective planning for climate adaptation management requires an understanding of the current state of local adaptation and physiological performance to assess whether populations are at risk of local extinction, determine if assisted migration is appropriate, and to select appropriate seed sources if intervention is needed. We established a new reciprocal transplant experiment (ACE, Adaptation to Climate and Environment) across a latitudinal gradient in North America to examine the impacts of warming on three bur oak (Quercus macrocarpa) populations across much of the species range. We established common gardens in Minnesota, Illinois, and Oklahoma with seedlings grown from seeds collected within 50 km of each of those locations from a total of sixty maternal families . We aimed to 1) assess local adaptation in each of the populations using survival and size as fitness metrics, and 2) evaluate physiological responses to different environments along the latitudinal gradient. We found that northern populations are maladapted to hotter climates as evidenced by their low survival, growth, and photosynthetic rates in the warmest common garden. The southernmost population had the highest survival rate, growth rate, and fitness of the three populations in the southernmost garden, providing evidence for local adaptation to the warmest site. However, conditions in the middle garden resulted in the highest fitness and best physiological performance for all populations. Growth and survival were correlated in the middle garden but were decoupled in the northern and southern gardens. This decoupling is likely due to stress associated with the relatively extreme climates in the gardens at the ends of the gradient that led to greater resource allocation to survival than growth. Our results suggest that southern seed sources may perform well in warmer conditions in the north brought on by climate change, which has important implications for managers assisting broadly ranged tree species in adapting to climate change.

Abstract:Background and aims: Invasive species usually demonstrate remarkable adaptability across diverse environments, successfully inhabiting a wide variety of regions. This adaptability often links to genetic differentiation and phenotypic plasticity, leading to latitudinal trends in phenotypic traits. In this study, we collected seeds of invasive plant Phytolacca americana from different latitudes and planted them in homogeneous gardens to investigate the latitudinal variation of P. americana phenotypic traits and to evaluate the effects of herbivory and heavy metals on plant growth, defence, and reproductive characteristics. Methods: P. americana seeds from different latitudes were planted in a homogeneous garden. For the experimental treatment, the seeds were divided into four groups: a heavy metal treatment group and its corresponding control group, and a cover treatment group with its corresponding control group. After the fruits matured, their growth, reproduction, and defence indicators were measured. Key results: Significant latitudinal trends were observed in P. americana's growth and defence characteristics, including changes in branch number, underground biomass, total biomass, and leaf tannin content. Compared to previous field surveys on P. americana, our study found that the latitude trends in growth structure and defence traits were consistent. But the latitudinal trend of reproductive structure is different. Moreover, heavy metals and herbivory substantially influenced the plant's growth, reproduction, and defence mechanisms, further shaping its latitudinal patterns. Conclusions: The observed phenotypic variations in P. americana across latitudes can be largely attributed to the synergistic effects of phenotypic plasticity and genetic variation. At a broader geographical scale, adaptations to heavy metal stress and herbivory pressure among different P. americana populations involve distinct trade-offs related to growth, reproduction, and defence strategies.
Decoupling of height growth and drought or pest resistance tradeoffs is revealed through multiple common-garden experiments of lodgepole pine

Abstract:Abstract The environment could alter growth and resistance tradeoffs in plants by affecting the ratio of resource allocation to various competing traits. Yet, how and why functional tradeoffs change over time and space is poorly understood particularly in long-lived conifer species. By establishing four common-garden test sites for five lodgepole pine populations in western Canada, combined with genomic sequencing, we revealed the decoupling pattern and genetic underpinnings of tradeoffs between height growth, drought resistance based on δ13C and dendrochronology, and metrics of pest resistance based on pest suitability ratings. Height and δ13C correlation displayed a gradient change in magnitude and/or direction along warm-to-cold test sites. All cold test sites across populations showed a positive height and δ13C relationship. However, we did not observe such a clinal correlation pattern between height or δ13C and pest suitability. Further, we found that the study populations exhibiting functional tradeoffs or synergies to various degrees in test sites were driven by non-adaptive evolutionary processes rather than adaptive evolution or plasticity. Finally, we found positive genetic relationships between height and drought or pest resistance metrics and probed five loci showing potential genetic tradeoffs between northernmost and the other populations. Our findings have implications for deciphering the ecological, evolutionary, and genetic bases of the decoupling of functional tradeoffs due to environmental change.

Evolutionary genomics predicts adaptive genetic and plastic gene expression responses to climate change in a key alpine forest tree species

Abstract:Despite widespread biodiversity loss, our understanding of how species and populations will respond to accelerated climate change remains limited. In this study, we predict the evolutionary responses of Populus lasiocarpa, a key alpine forest tree species primarily found in the mountainous regions of a global biodiversity hotspot, to climate change. We accomplish this by generating and integrating a new reference genome for P. lasiocarpa, re-sequencing data for 200 samples, and gene expression profiles for leaf and root tissue following exposure to heat and waterlogging. Analyses of the re-sequencing data indicate that demographic dynamics, divergent selection, and long-term balancing selection have shaped and maintained genetic variation within and between populations over historical timescales. In examining genomic signatures of contemporary climate adaptation, we found that haplotype blocks, characterized by inversion polymorphisms that suppress recombination, play a crucial role in clustering environmentally adaptive variations. Comparison of evolved and plastic gene expression show that genes with expression plasticity generally align with evolved responses, highlighting the adaptive role of plasticity. Lastly, we incorporated local adaptation, migration, genetic load, and plasticity responses into our predictions of population-level climate change risks. Our findings reveal that western populations, primarily distributed in the Hengduan Mountains-a region known for its environmental heterogeneity and significant biodiversity-are the most vulnerable to climate change and should be prioritized for conservation and management. Overall, our study advances understanding of the relative roles of long-term natural selection, local environmental adaptation, and immediate plasticity responses in driving evolutionary adaptation to climate change in keystone species.

Abstract:Although hybridization in plants has been recognized as an important pathway in plant speciation, it may also affect the ecology and evolution of associated communities. Cottonwood species (Populus angustifolia and P. fremontii) and their naturally occurring hybrids are known to support different plant, animal, and microbial communities, but no studies have examined community structure within the context of phylogenetic history. Using a community composed of 199 arthropod species, we tested for differences in arthropod phylogenetic patterns within and among hybrid and parental tree types in a common garden. Three major patterns emerged. (1) Phylogenetic diversity (PD) was significantly different between arthropod communities on hybrids and Fremont cottonwood when pooled by tree type. (2) Mean phylogenetic distance (MPD) and net relatedness index (NRI) indicated that communities on hybrid trees were significantly more phylogenetically overdispersed than communities on either parental tree type. (3) Community distance (Dpw) indicated that communities on hybrids were significantly different than parental species. Our results show that arthropod communities on parental and hybrid cottonwoods exhibit significantly different patterns of phylogenetic structure. This suggests that arthropod community assembly is driven, in part, by plant-arthropod interactions at the level of cottonwood tree type. We discuss potential hypotheses to explain the effect of plant genetic dissimilarity on arthropod phylogenetic community structure, including the role of competition and environmental filtering. Our findings suggest that cottonwood species and their hybrids function as evolutionarily significant units (ESUs) that affect the assembly and composition of associated arthropod communities and deserve high priority for conservation.

Abstract:A central tenet of plant defense theory is that adaptation to the abiotic environment sets the template for defense strategies, imposing a trade-off between plant growth and defense. Yet this trade-off, commonly found among species occupying divergent resource environments, may not occur across populations of single species. We hypothesized that more favorable climates and higher levels of herbivory would lead to increases in growth and defense across plant populations. We evaluated whether plant growth and defense traits covaried across 18 populations of showy milkweed (Asclepias speciosa) inhabiting an east-west climate gradient spanning 25° of longitude. A suite of traits impacting defense (e.g., latex, cardenolides), growth (e.g., size), or both (e.g., specific leaf area [SLA], trichomes) were measured in natural populations and in a common garden, allowing us to evaluate plastic and genetically based variation in these traits. In natural populations, herbivore pressure increased toward warmer sites with longer growing seasons. Growth and defense traits showed strong clinal patterns and were positively correlated. In a common garden, clines with climatic origin were recapitulated only for defense traits. Correlations between growth and defense traits were also weaker and more negative in the common garden than in the natural populations. Thus, our data suggest that climatically favorable sites likely facilitate the evolution of greater defense at minimal costs to growth, likely because of increased resource acquisition.

Abstract:Understanding how trees prioritize carbon gain at the cost of drought vulnerability under severe drought conditions is crucial for predicting which genetic groups and individuals will be resilient to future climate conditions. In this study, we investigated variations in growth, tree-ring anatomy as well as carbon and oxygen isotope ratios to assess the sensitivity and the xylem formation process in response to an episode of severe drought in 29 mature white spruce ( Picea glauca [Moench] Voss) families grown in a common garden trial. During the drought episode, the majority of families displayed decreased growth and exhibited either sustained or increased intrinsic water-use efficiency (iWUE), which was largely influenced by reduced stomatal conductance as revealed by the dual carbon‐oxygen isotope approach. Different water-use strategies were detected within white spruce populations in response to drought conditions. Our results revealed intraspecific variation in the prevailing physiological mechanisms underlying drought response within and among populations of Picea glauca . The presence of different genetic groups reflecting diverse water-use strategies within this largely-distributed conifer is likely to lessen the negative effects of drought and decrease the overall forest ecosystems' sensitivity to it.

Abstract:Summary Extreme droughts can have long‐lasting effects on forest community dynamics and species interactions. Yet, our understanding of how drought legacy modulates ecological relationships is just unfolding. We tested the hypothesis that leaf chemistry and herbivory show long‐term responses to premature defoliation caused by an extreme drought event in European beech (Fagus sylvatica L.). For two consecutive years after the extreme European summer drought in 2018, we collected leaves from the upper and lower canopy of adjacently growing drought‐stressed and unstressed trees. Leaf chemistry was analyzed and leaf damage by different herbivore‐feeding guilds was quantified. We found that drought had lasting impacts on leaf nutrients and on specialized metabolomic profiles. However, drought did not affect the primary metabolome. Drought‐related phytochemical changes affected damage of leaf‐chewing herbivores whereas damage caused by other herbivore‐feeding guilds was largely unaffected. Drought legacy effects on phytochemistry and herbivory were often weaker than between‐year or between‐canopy strata variability. Our findings suggest that a single extreme drought event bears the potential to long‐lastingly affect tree–herbivore interactions. Drought legacy effects likely become more important in modulating tree–herbivore interactions since drought frequency and severity are projected to globally increase in the coming decades.

Abstract:To examine whether deciduous and evergreen tree species differ in their performance and plastic responses to elevated atmospheric CO2 and temperature, we investigated growth, leaf area ratio (LAR), specific root length (SRL), nitrogen uptake efficiency (NUpE) and nitrogen use efficiency (NUE) of Populus yunnanensis and Abies faxoniana grown in environment-controlled chambers. Our results showed that temperature stimulated growth more than did CO2 in both species. The magnitude of temperature and CO2 effects varied between the species, as greater stimulation was detected in A. faxoniana than in P. yunnanensis. Greater responses of A. faxoniana were associated with higher LAR and higher NUE. However, NUpE did not differ between species and could not explain this advantage. On the other hand, the studied species did not differ in their mean overall plasticity (calculated for each species by averaging the indices of plasticity obtained for each of the 17 variables), which, however, was achieved in different ways. Across CO2 and temperature treatments, A. faxoniana exhibited greater plasticity in stomatal conductance, LAR, SRL and NUE, whereas P. yunnanensis exhibited greater plasticity in net photosynthesis rate, leaf respiration and root/shoot ratio. Our results suggested that plasticity of key traits, such as LAR, SRL and NUE, may have important implications for the superiority of A. faxoniana in the response to climate change. Our results highlight the importance of exploring the functional significance of traits rather than the extent of plasticity as potential determinants of interspecific differences in responses to climate change.

Abstract:Background Drought, fire, and insects are increasing mortality of pine species throughout the northern temperate zone as climate change progresses. Tree survival may be enhanced by forest diversity, with growth rates often higher in mixed stands, but whether tree defenses are likewise aided remains in question. We tested how forest diversity-productivity patterns relate to growth and defense over three centuries of climate change, competition, wildfire, and bark beetle attack. We used detailed census data from a fully mapped 25.6-ha forest dynamics plot in California, USA to conduct a spatially explicit, dendroecological assessment of large-diameter Pinus lambertiana survival following fire reintroduction. Our structural equation models investigated direct and indirect pathways by which growth, defense, and forest composition together mediated pine resistance and resilience. Results In the historical era of frequent, mixed-severity fire (pre-1900), trees that were ultimately resistant or susceptible to the post-fire bark beetle epidemic all showed similar growth and defenses, as measured by axial resin duct traits. During the era of fire exclusion (1901–2012), however, susceptible trees had slower growth. Following fire re-entry in 2013, both growth and defense declined precipitously for susceptible trees, resulting in fatal bark beetle attack. Spatial analysis showed that monodominant crowding by shade-tolerant competitors contributed to the long-term stress that prevented susceptible trees from recuperating defenses quickly following fire re-entry. For beetle-resistant trees, however, we found positive feedbacks between diversity, growth, and survival: trees in species-rich communities had higher growth rates pre-fire, which promoted a rapid recuperation of defenses following fire that helped trees resist bark beetle attack. Overall, this associational resistance outweighed associational susceptibility (+8.6% vs. −6.4% change in individual tree survival odds), suggesting a relaxation effect that ultimately allowed 58% of large pines to survive. Conclusions Though climate change threatens forest biodiversity, biodiversity is key to forest climate adaptation in return. Our findings demonstrate centennial-scale feedbacks by which forest diversity increases pine resistance and resilience to climate-amplified disturbances. The spatially explicit, dendroecological framework provides new insights into diversity-productivity theory, while also informing climate-adaptive forest management by identifying thresholds of tree density and richness that maximize large pine survival.

Abstract:Climate change (CC) conditions projected for many temperate areas of the world, expressed by way of excessive temperatures and low water availability, will impact forest health directly by means of abiotic stress but also by predisposing trees to pathogenic attack. However, we do not yet know how such environmental conditions alter the physiology and metabolism of trees to render them more susceptible to pathogens. To explore these mechanisms, we conditioned 3-year-old Austrian pine saplings to a simulated CC environment (combined drought and elevated temperatures), followed by pathogenic inoculation with two sister fungal species characterized by contrasting aggressiveness, Diplodia sapinea (aggressive) and D. scrobiculata (less aggressive). Lesion lengths resulting from infection were measured after 3 weeks to determine phenotypes, while dual transcriptomics analysis was conducted on tissues collected from the margins of developing lesions on separate branches 72 h post inoculation. As expected, climate change conditions enhanced host susceptibility to the less aggressive pathogen, D. scrobiculata , to a level that was not statistically different from the more aggressive D. sapinea . Under controlled climate conditions, D. sapinea induced suppression of critical pathways associated with host nitrogen and carbon metabolism, while enhancing its own carbon assimilation. This was accompanied by suppression of host defense-associated pathways. In contrast, D. scrobiculata infection induced host nitrogen and fatty acid metabolism as well as host defense response. The CC treatment, on the other hand, was associated with suppression of critical host carbon and nitrogen metabolic pathways, alongside defense associated pathways, in response to either pathogen. We propose a new working model integrating concurrent host and pathogen responses, connecting the weakened host phenotype under CC treatment with specific metabolic compartments. Our results contribute to a richer understanding of the mechanisms underlying the oft-observed increased susceptibility to fungal infection in trees under conditions of low water availability and open new areas of investigation to further integrate our knowledge in this critical aspect of tree physiology and ecology.
The adaptive potential of Populus balsamifera L. to phenology requirements in a warmer global climate

Abstract:The manner in which organisms adapt to climate change informs a broader understanding of the evolution of biodiversity as well as conservation and mitigation plans. We apply common garden and association mapping approaches to quantify genetic variance and identify loci affecting bud flush and bud set, traits that define a tree's season for height growth, in the boreal forest tree Populus balsamifera L. (balsam poplar). Using data from 478 genotypes grown in each of two common gardens, one near the southern edge and another near the northern edge of P. balsamifera's range, we found that broad-sense heritability for bud flush and bud set was generally high (H(2) > 0.5 in most cases), suggesting that abundant genetic variation exists for phenological response to changes in the length of the growing season. To identify the molecular genetic basis of this variation, we genotyped trees for 346 candidate single nucleotide polymorphisms (SNPs) from 27 candidate genes for the CO/FT pathway in poplar. Mixed-model analyses of variance identified SNPs in 10 genes to be associated with variation in either bud flush or bud set. Multiple SNPs within FRIGIDA were associated with bud flush, whereas multiple SNPs in LEAFY and GIGANTEA 5 were associated with bud set. Although there was strong population structure in stem phenology, the geographic distribution of multilocus association SNP genotypes was widespread except at the most northern populations, indicating that geographic regions may harbour sufficient diversity in functional genes to facilitate adaption to future climatic conditions in many sites.
