Abstract:A novel strategy for discovering and describing organizational goals and goal conflicts is presented. The procedure represents an integration of two alternative approaches that have been presumed incompatible: (a) a rank ordering of organizational goals on the basis of their relative importance, and (b) an active dormant or sequential view of organizational goals. Advantages and disadvantages associated with each of these approaches are assessed, and the manner and value of combining the two strategies into a single procedure are demonstrated. Utilities of this integrated procedure for planning, policy setting, and implementation are discussed, and it is concluded that adoption of the procedure would help administrators manage and stimulate more productive human services agencies.
Auditory processing in the zebra finch midbrain: single unit responses and effect of rearing experience

Abstract:In birds the auditory system plays a key role in providing the sensory input used to discriminate between conspecific and heterospecific vocal signals. In those species that are known to learn their vocalizations, for example, songbirds, it is generally considered that this ability arises and is manifest in the forebrain, although there is no a priori reason why brainstem components of the auditory system could not also play an important part. To test this assumption, we used groups of normal reared and cross-fostered zebra finches that had previously been shown in behavioural experiments to reduce their preference for conspecific songs subsequent to cross fostering experience with Bengalese finches, a related species with a distinctly different song. The question we asked, therefore, is whether this experiential change also changes the bias in favour of conspecific song displayed by auditory midbrain units of normally raised zebra finches. By recording the responses of single units in MLd to a variety of zebra finch and Bengalese finch songs in both normally reared and cross-fostered zebra finches, we provide a positive answer to this question. That is, the difference in response to conspecific and heterospecific songs seen in normal reared zebra finches is reduced following cross-fostering. In birds the virtual absence of mammalian-like cortical projections upon auditory brainstem nuclei argues against the interpretation that MLd units change, as observed in the present experiments, as a result of top-down influences on sensory processing. Instead, it appears that MLd units can be influenced significantly by sensory inputs arising directly from a change in auditory experience during development.

Abstract:Rare disruptions of the transcription factor FoxP1 are implicated in a human neurodevelopmental disorder characterised by autism and/or intellectual disability with prominent problems in speech and language abilities. Avian orthologues of this transcription factor are evolutionarily conserved and highly expressed in specific regions of songbird brains, including areas associated with vocal production learning and auditory perception. Here, we investigated possible contributions of FoxP1 to song discrimination and auditory perception in juvenile and adult female zebra finches. They received lentiviral knockdowns of FoxP1 in one of two brain areas involved in auditory stimulus processing, HVC (proper name) or CMM (caudomedial mesopallium). Ninety-six females, distributed over different experimental and control groups were trained to discriminate between two stimulus songs in an operant Go/Nogo paradigm and subsequently tested with an array of stimuli. This made it possible to assess how well they recognised and categorised altered versions of training stimuli and whether localised FoxP1 knockdowns affected the role of different features during discrimination and categorisation of song. Although FoxP1 expression was significantly reduced by the knockdowns, neither discrimination of the stimulus songs nor categorisation of songs modified in pitch, sequential order of syllables or by reversed playback were affected. Subsequently, we analysed the full dataset to assess the impact of the different stimulus manipulations for cue weighing in song discrimination. Our findings show that zebra finches rely on multiple parameters for song discrimination, but with relatively more prominent roles for spectral parameters and syllable sequencing as cues for song discrimination.

Abstract:A well replicated result in humans is that performance, whether good or bad, is consistent across a wide variety of cognitive tasks. Factor analysis extracts one factor that can account for approximately half of the variance in performance. This factor is termed g and almost all cognitive tasks positively load onto this factor. While some neurobiological correlates of g have been identified in humans, causal experiments are only feasible in animals. When mice and some avian species are assessed with cognitive test batteries, performance positively correlates, and the first component extracted has similar properties to g . There are some limitations to the species tested thus far, including comparability in the cognitive domains assessed. The pigeon is an ideal subject to overcome these issues since pigeons, humans, and other primates are frequently given similar tasks and many neural correlates of performance have been identified in the pigeon. We created a test battery that assessed different domains, including associative learning, memory, cognitive flexibility, and reaction time. When all tasks were included, there was evidence for a two-component structure that was influenced by subjects' age. When the reaction time task was excluded, there was a g-like component. The implications for these results when constructing future test batteries and comparing across species are discussed.

Song Processing in the Zebra Finch Auditory Forebrain Reflects Asymmetric Sensitivity to Temporal and Spectral Structure

Abstract:Despite being commonly referenced throughout neuroscientific research on songbirds, reports of hemispheric specialization in the processing of song remain controversial. The notion of such asymmetries in songbirds is further complicated by evidence that both cerebral hemispheres in humans may be specialized for different aspects of speech perception. Some studies suggest that the auditory neural substrates in the left and right hemispheres of humans process temporal and spectral elements within speech sounds, respectively. To determine whether songbirds process their conspecific songs in such a complementary, bilateral manner, we performed functional magnetic resonance imaging (fMRI) on 15 isoflurane anesthetized adult male zebra finches (<i>Taeniopygia guttata</i>) while presenting them with (1) non-manipulated, (2) spectrally-filtered (reduced spectral structure), and (3) temporally-filtered (reduced temporal structure) conspecific song. Our results revealed sensitivity of both primary (Field L) and secondary (caudomedial nidopallium, NCM) auditory regions to changes in spectral and temporal structure of song. On the one hand, temporally-filtered song elicited a bilateral decrease in neural responses compared to the other stimulus types. On the other hand, spectrally filtered song elicited significantly greater responses in left Field L and NCM than temporally filtered or non-manipulated song while concurrently reducing the response relative to non-manipulated song in the right auditory forebrain. The latter hemispheric difference in sensitivity to manipulations of spectral structure in song, suggests that there is an asymmetry in spectral and temporal domain processing in the zebra finch auditory forebrain bearing some resemblance to what has been observed in human auditory cortex.

Abstract:Since ecology influences the expression of cognitive traits, intra-specific variation in ecological demands can drive differences in cognition. This is often the case, for instance, when sexes face different ecological challenges. However, so far, most studies have focused on few cognitive domains (i.e., spatial cognition), which limits our understanding of the evolution of sexually dimorphic cognition in animals. Endangered Southern ground-hornbills ( Bucorvus leadbeateri ), for example, show sex-specific ecological differences in age at dispersal, where females disperse from their natal group earlier than males. Based on this potential sex-specific source of selection, females and males may differ in their capacity to behave flexibly. Here, we used the reversal-learning paradigm in ten Southern ground-hornbills in two conditions: spatial and colour. During the pre-test (learning phase), regardless the sex, all subjects were faster at associating the food reward with spatial rather than with colour cues. Similarly, during the test (reversal-learning phase), both sexes learned the new association quicker with spatial cues. There were no sex differences in learning or reversal learning during both experimental phases. This possibility, however, requires further observation and experimentation. We hope our study will provide the impetus to assess further the cognitive capacities of this still overlooked species.

Social reinforcement guides operant behaviour and auditory learning in a songbird

Abstract:Motivation to seek social interactions is inherent to all social species. For instance, even with risk of disease transmission in a recent pandemic, humans sought out frequent in-person social interactions. In other social animals, socialization can be prioritized even over water or food consumption. Zebra finches, Taeniopygia guttata, are highly gregarious songbirds widely used in behavioural and physiological research. Songbirds, like humans, are vocal learners during development, which rely on intense auditory learning. Aside from supporting song learning, auditory learning further supports individual identification, mate choice and outcome associations in songbirds. To study auditory learning in a laboratory setting, studies often employ operant paradigms with food restriction and reinforcement and require complete social isolation, which can result in stress and other unintended physiological consequences for social species. Thus, in this work, we designed an operant behavioural method leveraging the sociality of zebra finches for goal-directed behaviours. Our approach relies on visual social reinforcement, without depriving the animals of food or social contact. Using this task, we found that visual social reinforcement was a strong motivational drive for operant behaviour. Motivation was sensitive to familiarity towards the stimulus animal and higher when engaging with a familiar versus a novel individual. We further show that this tool can be used to assess auditory discrimination learning using either songs or synthetic pure tones as stimuli. As birds gained experience in the task, they developed a strategy to maximize reward acquisition in spite of receiving more punishment, i.e. liberal response bias. Our operant paradigm provides an alternative to tasks using food reinforcement and could be applied to a variety of highly social species, such as rodents and nonhuman primates.
Human-like brain hemispheric dominance in birdsong learning

Abstract:Unlike nonhuman primates, songbirds learn to vocalize very much like human infants acquire spoken language. In humans, Broca’s area in the frontal lobe and Wernicke’s area in the temporal lobe are crucially involved in speech production and perception, respectively. Songbirds have analogous brain regions that show a similar neural dissociation between vocal production and auditory perception and memory. In both humans and songbirds, there is evidence for lateralization of neural responsiveness in these brain regions. Human infants already show left-sided dominance in their brain activation when exposed to speech. Moreover, a memory-specific left-sided dominance in Wernicke’s area for speech perception has been demonstrated in 2.5-mo-old babies. It is possible that auditory-vocal learning is associated with hemispheric dominance and that this association arose in songbirds and humans through convergent evolution. Therefore, we investigated whether there is similar song memory-related lateralization in the songbird brain. We exposed male zebra finches to tutor or unfamiliar song. We found left-sided dominance of neuronal activation in a Broca-like brain region (HVC, a letter-based name) of juvenile and adult zebra finch males, independent of the song stimulus presented. In addition, juvenile males showed left-sided dominance for tutor song but not for unfamiliar song in a Wernicke-like brain region (the caudomedial nidopallium). Thus, left-sided dominance in the caudomedial nidopallium was specific for the song-learning phase and was memory-related. These findings demonstrate a remarkable neural parallel between birdsong and human spoken language, and they have important consequences for our understanding of the evolution of auditory-vocal learning and its neural mechanisms.
Tracing the development of learned song preferences in the female zebra finch brain with functional magnetic resonance imaging

Abstract:In sexually dimorphic zebra finches (Taeniopygia guttata), only males learn to sing their father's song, whereas females learn to recognize the songs of their father or mate but cannot sing themselves. Memory of learned songs is behaviorally expressed in females by preferring familiar songs over unfamiliar ones. Auditory association regions such as the caudomedial mesopallium (CMM; or caudal mesopallium) have been shown to be key nodes in a network that supports preferences for learned songs in adult females. However, much less is known about how song preferences develop during the sensitive period of learning in juvenile female zebra finches. In this study, we used blood‐oxygen level‐dependent (BOLD) functional magnetic resonance imaging (fMRI) to trace the development of a memory‐based preference for the father's song in female zebra finches. Using BOLD fMRI, we found that only in adult female zebra finches with a preference for learned song over novel conspecific song, neural selectivity for the father's song was localized in the thalamus (dorsolateral nucleus of the medial thalamus; part of the anterior forebrain pathway, AFP) and in CMM. These brain regions also showed a selective response in juvenile female zebra finches, although activation was less prominent. These data reveal that neural responses in CMM, and perhaps also in the AFP, are shaped during development to support behavioral preferences for learned songs.

Comparative Cognition Research Demonstrates the Similarity between Humans and Other Animals

Abstract:The field of comparative cognition represents the interface between the cognitive behavior of humans and other animals. In some cases, research demonstrates that other animals are capable of showing similar cognitive processes. In other cases, when animals show behavior thought to be culturally determined in humans, it suggests that simpler processes may be involved. This review examines research primarily with pigeons (out of convenience because of their visual ability). I start with the concept of sameness and follow with the concept of stimulus equivalence, the building blocks of human language. This is followed by research on directed forgetting, the cognitive ability to maintain or forget information. A hallmark of cognition is transitive inference performance (if A < B, and B < C, the understanding that A < C), but the variety of species that show this ability suggests that there may be simpler accounts of this behavior. Similarly, experiments that demonstrate a form of cognitive dissonance in animals suggest that dissonance may not be necessary to explain this biased behavior. Furthermore, examples of sunk cost in pigeons suggests that the human need to continue working on a failing project may also have a biological basis. Finally, pigeons show a preference for a suboptimal choice that is similar to unskilled human gambling, a finding that may clarify why humans are so prone to engage in this typically losing activity.
Early Auditory Experience Modifies Neuronal Firing Properties in the Zebra Finch Auditory Cortex

Abstract:Songbirds learn to sing much as humans learn to speak. In zebra finches, one of the premier songbird models, males learn to sing for later courtship through a multistep learning process during the developmental period. They first listen to and memorize the song of a tutor (normally their father) during the sensory learning period. Then, in the subsequent sensory-motor learning phase (with large overlap), they match their vocalizations to the memorized tutor song <i>via</i> auditory feedback and develop their own unique songs, which they maintain throughout their lives. Previous studies have suggested that memories of tutor songs are shaped in the caudomedial nidopallium (NCM) of the brain, which is analogous to the mammalian higher auditory cortex. Isolation during development, which extends the sensory learning period in males, alters song preference in adult females, and NCM inactivation decreases song preference. However, the development of neurophysiological properties of neurons in this area and the effect of isolation on these neurons have not yet been explained. Here, we performed whole-cell patch-clamp recording on NCM neurons from juvenile zebra finches during the sensory learning period, 20, 40, or 60 days post-hatching (DPH) and examined their neurophysiological properties. In contrast to previous reports in adult NCM neurons, the majority of NCM neurons of juvenile zebra finches showed spontaneous firing with or without burst firing patterns, and the percentage of neurons that fired increased in the middle of the sensory learning period (40 DPH) and then decreased at the end (60 DPH) in both males and females. We further found that auditory isolation from tutor songs alters developmental changes in the proportions of firing neurons both in males and females, and also changes those of burst neurons differently between males that sing and females that do not. Taken together, these findings suggest that NCM neurons develop their neurophysiological properties depending on auditory experiences during the sensory song learning period, which underlies memory formation for song learning in males and song discrimination in females.

Abstract:The performance of courtship signals provides information about the behavioural state and quality of the signaller, and females can use such information for social decision-making (e.g. mate choice). However, relatively little is known about the degree to which the perception of and preference for differences in motor performance are shaped by developmental experiences. Furthermore, the neural substrates that development could act upon to influence the processing of performance features remains largely unknown. In songbirds, females use song to identify males and select mates. Moreover, female songbirds are often sensitive to variation in male song performance. Consequently, we investigated how developmental exposure to adult male song affected behavioural and neural responses to song in a small, gregarious songbird, the zebra finch. Zebra finch males modulate their song performance when courting females, and previous work has shown that females prefer the high-performance, female-directed courtship song. However, unlike females allowed to hear and interact with an adult male during development, females reared without developmental song exposure did not demonstrate behavioural preferences for high-performance courtship songs. Additionally, auditory responses to courtship and non-courtship song were altered in adult females raised without developmental song exposure. These data highlight the critical role of developmental auditory experience in shaping the perception and processing of song performance.
Social and auditory experience shapes forebrain responsiveness in zebra finches before the sensitive period of vocal learning

Abstract:Early-life experiences with signals used in communication are instrumental in shaping an animal's social interactions. In songbirds, which use vocalizations for guiding social interactions and mate choice, recent studies show that sensory effects on development occur earlier than previously expected, even in embryos and nestlings. Here, we explored the neural dynamics underlying experience-dependent song categorization in young birds prior to the traditionally studied sensitive period of vocal learning that begins around 3 weeks post-hatch. We raised zebra finches either with their biological parents, cross-fostered by Bengalese finches beginning at embryonic day 9, or with only the non-singing mother from 2 days post-hatch. Then, 1-5 days after fledging, we conducted behavioral experiments and extracellular recordings in the auditory forebrain to test responses to zebra finch and Bengalese finch songs. Auditory forebrain neurons in cross-fostered and isolated birds showed increases in firing rate and decreases in responsiveness and selectivity. In cross-fostered birds, decreases in responsiveness and selectivity relative to white noise were specific to conspecific song stimuli, which paralleled behavioral attentiveness to conspecific songs in those same birds. This study shows that auditory and social experience can already impact song 'type' processing in the brains of nestlings, and that brain changes at this age can portend the effects of natal experience in adults.
Dynamic encoding of phonetic categories in zebra finch auditory forebrain

Abstract:Abstract Vocal communication requires the formation of acoustic categories to enable invariant representations of sounds despite superficial variations. Humans form acoustic categories for speech phonemes, enabling the listener to recognize words independent of speakers; animals can also discriminate speech phonemes. We investigated the neural mechanisms of this process using electrophysiological recordings from the zebra finch secondary auditory area, caudomedial nidopallium (NCM), during passive exposure to human speech stimuli consisting of two naturally spoken words produced by multiple speakers. Analysis of neural distance and decoding accuracy showed improvements in neural discrimination between word categories over the course of exposure, and this improved representation transferred to the same words by novel speakers. We conclude that NCM neurons formed generalized representations of word categories independent of speaker-specific variations that became more refined over the course of passive exposure. The discovery of this dynamic encoding process in NCM suggests a general processing mechanism for forming categorical representations of complex acoustic signals that humans share with other animals.

Categorical and semantic perception of the meaning of call-types in zebra finches

Abstract:Vocal communication in social animals involves the production and perception of various calls that ethologists categorize into call-types, based on their acoustical structure and the behavioral context of production. Whether animals perceive these categories and associate distinct meanings to them remains unknown. The zebra finch, a gregarious songbird, uses approximately 11 call-types to communicate hunger, danger, social conflict, and establish social contact and bonding. Using auditory discrimination tasks, we show that the birds discriminate and categorize all the call-types in their vocal repertoire. In addition, systematic errors were more frequent between call-types used in similar behavioral contexts than could be expected from their acoustic similarity. Thus, zebra finches organize their calls into categories and create a mental representation of the meaning of these sounds.

Abstract:In cervical spinal cord transected and bilaterally vagotomized dogs, intrathecal (i.t.) injection of clonidine decreased the resting heart rate and mean arterial pressure (MAP) while isoprenaline increased the heart rate. The clonidine induced bradycardia and hypotension were antagonized by piperoxan pretreatment. Similarly, the isoprenaline induced tachycardia was antagonized by pretreatment (i.t.) with atenolol. Intrathecal atenolol per se decreased the heart rate and MAP while piperoxan had no effect. The post-coronary artery ligation cardiac arrhythmia, in the cervical spinal cord transected and bilaterally vagotomized dogs, was inhibited by clonidine, phenylephrine and methoxamine and facilitated by isoprenaline. beta-Adrenoreceptor antagonists propranolol, atenolol and acebutolol, given intrathecally, inhibited while piperoxan facilitated the cardiac arrhythmia. The clonidine and isoprenaline induced changes in the arrhythmia were antagonized by i.t. pretreatment with piperoxan and atenolol respectively. It appears that the alpha-adrenoreceptors are inhibitory while beta-adrenoreceptors are facilitatory for spinal control of heart rate and blood pressure and for post-coronary artery ligation cardiac arrhythmia.
Hemispheric dominance in HVC is experience-dependent in juvenile male zebra finches

Abstract:Juvenile male zebra finches (Taeniopygia guttata) must be exposed to an adult tutor during a sensitive period to develop normal adult song. The pre-motor nucleus HVC (acronym used as a proper name), plays a critical role in song learning and production (cf. Broca's area in humans). In the human brain, left-side hemispheric dominance in some language regions is positively correlated with proficiency in linguistic skills. However, it is unclear whether this pattern depends upon language learning, develops with normal maturation of the brain, or is the result of pre-existing functional asymmetries. In juvenile zebra finches, even though both left and right HVC contribute to song production, baseline molecular activity in HVC is left-dominant. To test if HVC exhibits hemispheric dominance prior to song learning, we raised juvenile males in isolation from adult song and measured neuronal activity in the left and right HVC upon first exposure to an auditory stimulus. Activity in the HVC was measured using the immediate early gene (IEG) zenk (acronym for zif-268, egr-1, NGFI-a, and krox-24) as a marker for neuronal activity. We found that neuronal activity in the HVC of juvenile male zebra finches is not lateralized when raised in the absence of adult song, while normally-reared juvenile birds are left-dominant. These findings show that there is no pre-existing asymmetry in the HVC prior to song exposure, suggesting that lateralization of the song system depends on learning through early exposure to adult song and subsequent song-imitation practice.

For animals that rely on cognition for survival, variation in cognitive ability may also affect social structure. Individuals with worse cognitive abilities could benefit from preferentially associating with conspecifics with better cognition, from being more gregarious, or both, allowing them access to resources. Climatic conditions influence resource availability, which may also affect the relative benefits of assortment or gregariousness, resulting in spatiotemporal variation of social patterns. Using 5 years of data, we investigated assortment by spatial cognitive ability and associations between sociality and spatial cognition in food-caching mountain chickadees, Poecile gambeli , inhabiting harsher environments at higher elevations and milder environments at lower elevations. Elevation environments differ in overwinter severity and in the harshest winter, high elevation chickadees with better spatial learning and memory abilities were less gregarious and showed lower social differentiation (or choosiness) than those with worse spatial learning and memory abilities but did not assort based on these traits. In two other seasons, including the second harshest winter, high elevation chickadees that were less cognitively flexible showed more social differentiation than those that were more cognitively flexible. In these two seasons alone, chickadees negatively assorted by spatial learning and memory performance. Chickadees from low elevations only demonstrated negative assortment in the second harshest winter. All other results were nonsignificant, suggesting that overall, individual spatial cognitive abilities or those of their associates have little influence on social preferences.

Abstract:Male zebra finches, Taeniopygia guttata, acquire their song during a sensitive period for auditory-vocal learning by imitating conspecific birds. Laboratory studies have shown that the sensitive period for song acquisition covers a developmental phase lasting from 25 to 65days post hatch (dph); formation of auditory memory primarily occurs between 25 and 35dph. The duration of the sensitive period is, however, dependent upon model availability. If a tutor is not available early in development, birds will learn from an adult male introduced to their cage even after they reach 65dph. Birds who are exposed to a second tutor as late as 63dph can successfully adjust their song 'template' to learn a new song model. However, if second-tutor song exposure occurs after 65dph, learning of a new tutor's song will not occur for most individuals. Here, we review the literature as well as novel studies from our own laboratory concerning sensitive periods for auditory memory formation in zebra finches; these behavioral studies indicate that there are developmental constraints on imitative learning in zebra finches.
