Abstract:Green nanomaterials have gained much attention due to their potential use as therapeutic agents. The present study investigated the production of silver nanoparticles (AgNPs) from a silver-resistant Bacillus safensis TEN12 strain, which was isolated from metal contaminated soil and taxonomically identified through 16S rRNA gene sequencing. The formation of AgNPs in bacterial culture was confirmed by using UV-vis spectroscopy with an absorption peak at 426.18 nm. Fourier transform infrared (FTIR) spectroscopy confirmed the involvement of capping proteins and alcohols for stabilization of AgNPs. Moreover, X-ray diffraction analysis (XRD), scanning and transmission electron microscopy (SEM and TEM) confirmed the crystalline nature and spherical shape of AgNPs with particle size ranging from 22.77 to 45.98 nm. The energy dispersive X-ray spectroscopy (EDX) revealed that 93.54% silver content is present in the nano-powder. AgNPs showed maximum antibacterial activity (20.35 mm and 19.69 mm inhibition zones) at 20 mu g mL(-1) concentration against Staphylococcus aureus and Escherichia coli, respectively and significantly reduced the pathogen density in broth culture. Furthermore, AgNPs demonstrated significant anticancer effects in the human liver cancer cell line (HepG2) in MTT assay, whereas, no cytotoxic effects were demonstrated by AgNPs on normal cell line (HEK293). The present study suggests that the biogenic AgNPs may substitute chemically synthesized drugs with wider applications as antibacterial and anticancer agents. (C) 2020 The Authors. Antibacterial, antibiofilm, and anticancer activity of silver-nanoparticles synthesized from the cell-filtrate of Streptomyces enissocaesilis

Abstract:Silver nanoparticles (Ag-NPs) have a unique mode of action as antibacterial agents in addition to their anticancer and antioxidant properties. In this study, microbial nanotechnology is employed to synthesize Ag-NPs using the cell filtrate of Streptomyces enissocaesilis BS1. The synthesized Ag-NPs are confirmed by ultraviolet-visible (UV-Vis), Fourier transform infrared (FT-IR), X-ray diffraction (XRD), energy dispersive X-ray spectroscopy (EDX), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Also, the effects of different factors on Ag-NPs synthesis were evaluated to set the optimum synthesis conditions. Also, the antibacterial, antibiofilm, and anticancer activity of Ag-NPs was assessed. The X-ray diffraction (XRD) analysis confirmed the crystalline nature of the sample and validated that the crystal structure under consideration is a face-centered cubic (FCC) pattern. The TEM examination displayed the spherical particles of the Ag-NPs and their average size, which is 32.2 nm. Fourier transform infrared spectroscopy (FTIR) revealed significant changes in functionality after silver nanoparticle dispersion, which could be attributed to the potency of the cell filtrate of Streptomyces enissocaesilis BS1 to act as both a reducing agent and a capping agent. The bioactivity tests showed that our synthesized Ag-NPs exhibited remarkable antibacterial activity against different pathogenic strains. Also, when the preformed biofilms of Pseudomonas aeruginosa ATCC 9027 , Salmonella typhi ATCC 12023 , Escherichia coli ATCC 8739 , and Staphylococcus aureus ATCC 6598 were exposed to Ag NPs 50 mg/ml for 24 hours, the biofilm biomass was reduced by 10.7, 34.6, 34.75, and 39.08%, respectively. Furthermore, the Ag-NPs showed in vitro cancer-specific sensitivity against human breast cancer MCF-7 cell lines and colon cancer cell line Caco-2, and the IC 50 was 0.160 mg/mL and 0.156 mg/mL, respectively. The results of this study prove the ease and efficiency of the synthesis of Ag-NPs using actinomycetes and demonstrate the significant potential of these Ag-NPs as anticancer and antibacterial agents.

Abstract:In this study, metabolites involved in the free-biomass filtrates for three endophytic actinomycetes of Streptomyces capillispiralis Ca-1, Streptomyces zaomyceticus Oc-5, and Streptomyces pseudogriseolus Acv-11 were used as biocatalysts for green synthesis of silver nanoparticles (Ag-NPs). Characterization of biosynthesized Ag-NPs was accomplished using UV-Vis spectroscopy, X-ray diffraction patterns (XRD), Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM-EDX), transmission electron microscopy (TEM), and particle size analyzer. The biosynthesized Ag-NPs showed maximum surface plasmon resonance (SPR) at 440 for strain Ca-1 and 450 for both strains of OC-5 and Acv-11. Nanoparticle spherical shape was recorded with size ranging from 23.77 to 63.14 nm, 11.32 to 36.72 nm, and 11.70 to 44.73 nm for Ca-1, Oc-5, and Acv-11, respectively. SEM-EDX analysis exhibited the weight percentages of 17.3, 22.3, and 48.7% for Ag-NPs synthesized by strains Ca-1, Oc-5 and Acv-11, respectively. The activities of biosynthesized Ag-NPs were concentration dependent and the obtained results confirmed the efficacy of Ag-NPs as antimicrobial agents against Gram-positive and Gram-negative bacteria as well unicellular and multicellular fungi. The MIC for Gram-positive bacteria, Gram-negative bacteria (E. coli), and eukaryotic microorganisms was 0.25 mM with clear zone ranging from 10.3 to 14.6 mm, while MIC for Pseudomonas aeruginosa was 1.0 mM for Ag-NPs synthesized by strain Ca-1 and 0.25 mM for those synthesized by strains Oc-5 and Acv-11. Moreover, Ag-NPs exhibited antimicrobial activity against four plant pathogenic fungi represented by Alternaria alternata, Fusarium oxysporum, Pythium ultimum, and Aspergillus niger at 2.0, 1.5, 1.0, and 0.5 mM of Ag-NPs with different degree. In vitro assessment of the antioxidant efficacy of biosynthesized Ag-NPs was achieved by scavenging assay of H2O2, reducing power of Fe3+, or total antioxidant assay. The results showed that antioxidant activities of Ag-NPs were concentration dependent with the highest activity at Ag-NP concentration of 2.0 mM. Furthermore, the biosynthesized NPs have prospective bioinsecticidal activity against Culex pipiens and Musca domestica. Green synthesis of NPs could be quite potential for the development of new bioactive compounds used in different biomedical applications.

Abstract:The study of nanoparticles has gained popularity recently, particularly in biomedicine where metallic AgNPs were widely utilized for their therapeutic benefits. This current study focuses on biosynthesis of AgNPs using marine Streptomyces coelicoflavus MTK30, detailing their characterization, in-vitro, in-vivo and in-silico properties. UV-Vis spectrum exhibited a surface plasmon resonance peak at 441.24 nm, while the FT-IR spectrum O-H stretching at 3416.99 cm −1 showed the stabilization of AgNPs. XRD and SEM revealed the clear, crystalline, spherical shaped and their width was recorded as 98.4 nm with a PdI of 0.229 during DLS analysis. At 40 μl/ml concentration, showed antibacterial activity against clinical pathogens and significant scavenging against DPPH and MCF-7 with IC 50 values of 56.50 and 44.83 μg/ml. In-vivo studies using Swiss albino mice proclaimed that at 10 mg concentration is more effective in wound healing. Molecular docking studies elucidated the anticancer interaction with the breast cancer virulence protein (BRCA1 and C-erbB2) as well as various pathogenic bacteria (SpA, Ag43, esp, and aglD). This research demonstrates the feasibility of utilizing marine Streptomyces coelicoflavus MTK30 to produce biogenic AgNPs with significant antimicrobial, antioxidant, anticancer, wound healing and in-silico modelling revealed that well conserved binding region for drug development approaches.

Abstract:A novel antagonistic marine Streptomyces sp. Al-Dhabi-87 that was recovered from the Gulf region of Saudi Arabia was used to synthesize silver nanoparticles (NP) from the culture free extract. The produced NP were confirmed by UV-visible spectroscopy (UV-Vis), high-resolution scanning electron microscope (HRSEM), transmission electron microscope (TEM), Fourier-transform infrared spectroscopy (FTIR), Energy Dispersive Spectroscopy (EDAX), and X-ray Powder Diffraction (XRD), and broth micro dilution techniques were employed for the determination of minimum inhibitory concentrations (MIC) values. The synthesized NP was authenticated by alterations in color and wavelength scanning. HRSEM and TEM analysis confirmed that the size of the NP ranged from 10 to 17 nm and that it was spherical in shape. In addition, the FTIR spectrum revealed a variation in the band values from 500 to 3300 cm&minus;1 respectively. Rietveld refinement analysis of the XRD data confirmed the size of the NP, which coincided with the results of the TEM analysis. In addition, the Riveted refinement analysis supported the TEM data. The NP documented significant activity against the wound infection microbial strains, such as Enterococcus faecalis, Staphylococcus epidermidis, and Staphylococcus aureus. Gram negative bacteria, such as Pseudomonas aeruginosa, Klebsiella pneumonia, and Escherichia coli revealed MIC values of 0.039, 0.078, and 0.152 mg/mL, respectively. The promising activity of NP towards extended-spectrum beta-lactamases E.coli, drug resistant Acinetobacter baumannii, and multidrug resistant S. aureus (at 0.018, 0.039, and 0.039 mg/mL, respectively) was advantageous. Overall, NP that were obtained from the novel Streptomyces sp. Al-Dhabi-87, with its promising antimicrobial activity towards the drug resistant pathogens, would be useful for healing infectious diseases.

Abstract:Recent studies have emphasized the application of nanoscience and nanotechnology to disease prevention, diagnosis, and treatment. This study examined the use of marine actinobacteria in the synthesis of silver nanoparticles (AgNPs). UV–Vis spectrophotometry, Fourier infrared spectroscopy, X-ray diffraction, scanning electron microscopy, and energy dispersive X-ray spectroscopy were used to characterize the primary structure, crystalline nature, functional groups, shape, and elemental signal of the synthesized AgNPs. The mean particle diameter of the AgNPs was measured, and the size was found to be between 23 and 27 nm. The antibacterial activity of the AgNPs was evaluated; it showed enhanced inhibitory activity against Escherichia coli , Bacillus subtilis , and Klebsiella pneumoniae . At an AgNP concentration of 40 g/mL, the test pathogen growth was completely inhibited. The larvicidal property of the synthesized AgNPs showed enhanced activity against An. stephensi (LC5017.511 and LC9050.572), A. aegypti (LC50 22.481 and LC90 61.920), and Cx. quinquefasciatus (LC50 29.548 and LC90 73.123). By analyzing the phenotypic data, the 16S rRNA gene sequence, and the phylogenetic data, the probable isolate was also determined to be Streptomyces parvisporogenes KL3. The study results prove that biosynthesized AgNPs can be used as effective antibacterial and larvicidal agents in drug formulation.

Remote estimation of phytoplankton size fractions using the spectral shape of light absorption.

Abstract:Phytoplankton size structure plays an important role in ocean biogeochemical processes. The light absorption spectra of phytoplankton provide a great potential for retrieving phytoplankton size structure because of the strong dependence on the packaging effect caused by phytoplankton cell size and on different pigment compositions related to phytoplankton taxonomy. In this study, we investigated the variability in light absorption spectra of phytoplankton in relation to the size structure. Based on this, a new approach was proposed for estimating phytoplankton size fractions. Our approach use the spectral shape of the normalized phytoplankton absorption coefficient (a(ph)(λ)) through principal component analysis (PCA). Values of a(ph)(λ) were normalized to remove biomass effects, and PCA was conducted to separate the spectral variance of normalized a(ph)(λ) into uncorrelated principal components (PCs). Spectral variations captured by the first four PC modes were used to build relationships with phytoplankton size fractions. The results showed that PCA had powerful ability to capture spectral variations in normalized a(ph)(λ), which were significantly related to phytoplankton size fractions. For both hyperspectral a(ph)(λ) and multiband a(ph)(λ), our approach is applicable. We evaluated our approach using wide in situ data collected from coastal waters and the global ocean, and the results demonstrated a good and robust performance in estimating phytoplankton size fractions in various regions. The model performance was further evaluated by a(ph)(λ) derived from in situ remote sensing reflectance (R(rs)(λ)) with a quasi-analytical algorithm. Using R(rs)(λ) only at six bands, accurate estimations of phytoplankton size fractions were obtained, with R(2) values of 0.85, 0.61, and 0.76, and root mean-square errors of 0.130, 0.126, and 0.112 for micro-, nano-, and picophytoplankton, respectively. Our approach provides practical basis for remote estimation of phytoplankton size structure using a(ph)(λ) derived from satellite observations or rapid field instrument measurements in the future.
Silver Nanoparticles Derived from Probiotic Lactobacillus casei-a Novel Approach for Combating Bacterial Infections and Cancer

Abstract:In the face of rising antibiotic resistance and the need for novel therapeutic approaches against cancer, the present study delves into the various facets of biosynthesized silver nanoparticles (AgNPs) derived from the probiotic strain Lactobacillus casei (AgNPs-LC), assessing their efficacy in combating bacterial infections, disrupting biofilm formation, interfering with quorum sensing mechanisms, and exhibiting anti-cancer properties. The results showed that the AgNPs-LC had a spherical shape with an average size of 15 nm. The biosynthesized AgNPs-LC showed a symmetrical absorption spectrum with a peak at 458 nm with a diameter of 5-20 nm. AgNPs-LC exhibited significant antibacterial activity against Gram-positive and Gram-negative bacteria and inhibited the biofilm formation (> 50% at sub-MIC) and quorum sensing-mediated virulence factors, such as the production of violacein in C. violaceum (> 80% at sub-MIC), pyocyanin in P. aeruginosa (> 70% at sub-MIC), and prodigiosin in S. marcescens (> 80% at sub-MIC). The exopolysaccharides (EPS) were also found to reduce in the presence of AgNPs-LC. Furthermore, the AgNPs-LC showed anti-cancer and anti-metastasis activity via inhibiting cell migration and invasion of human lung cancer (A-549) cells. Overall, the present study brings out the multifaceted therapeutic capabilities of AgNPs-LC which offer exciting prospects for the development of innovative biomedical and pharmaceutical interventions, making AgNPs-LC a versatile and promising candidate for a wide range of applications in healthcare and medicine. However, further research is essential to fully harness their therapeutic potential.
Enhanced Antibacterial, Anti-biofilm and Antioxidant (ROS) Activities of Biomolecules Engineered Silver Nanoparticles Against Clinically Isolated Gram Positive and Gram Negative Microbial Pathogens

Abstract:Biomolecule-coated silver nanoparticles (AgNPs) were synthesized using ten different medicinal plants and studied their antibacterial and anti-biofilm activity against selected clinically isolated microbial pathogens. Though the bioengineer AgNPs showed effective antibacterial activity, the maximum rate of growth inhibition was recorded in liquid medium. A potential anti-biofilm activity was determined using biomolecule-coated AgNPs derived from Centella asiatica, Plumbago zeylanica and Semecarpus anacardium against Pseudomonas aeruginosa and Staphylococcus epidermidis. Results strongly suggest that bioengineered AgNPs at lower doses can be used for the treatment of multidrug-resistant bacterial infections with biofilm formation. Further, the XTT assay exhibited an increased level of reactive oxygen species production in bacterial strains treated with bioengineered AgNPs using 10 plant species. Present results clearly indicate that biomolecule-coated AgNPs could be used as effective nanodrug for treatment of infectious diseases caused by multidrug resistant bacterial strains in the near future.

Green Synthesized Silver Nanoparticles: Antibacterial and Anticancer Activities, Biocompatibility, and Analyses of Surface-Attached Proteins

Abstract:The increasing number of multi-drug-resistant bacteria and cancer cases, that are a real threat to humankind, forces research world to develop new weapons to deal with it. Biogenic silver nanoparticles (AgNPs) are considered as a solution to this problem. Biosynthesis of AgNPs is regarded as a green, eco-friendly, low-priced process that provides small and biocompatible nanostructures with antimicrobial and anticancer activities and potential application in medicine. The biocompatibility of these nanoparticles is related to the coating with biomolecules of natural origin. The synthesis of AgNPs from actinobacterial strain was confirmed using UV-Vis spectroscopy while their morphology, crystalline structure, stability, and coating were characterized using, transmission electron microscopy (TEM), X-ray diffraction (XRD), Zeta potential and Fourier transform infrared spectroscopy (FTIR). Antibacterial activity of biogenic AgNPs was evaluated by determination of minimum inhibitory and minimum biocidal concentrations (MIC and MBC) against Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Staphylococcus aureus. The potential mechanism of antibacterial action of AgNPs was determined by measurement of ATP level. Since the use of AgNPs in biomedical applications depend on their safety, the in vitro cytotoxicity of biosynthesized AgNPs on MCF-7 human breast cancer cell line and murine macrophage cell line RAW 264.7 using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay, cell lactate dehydrogenase (LDH) release and measurement of reactive oxygen species (ROS) level were assessed. The nanoparticle protein capping agent that can be involved in reduction of silver ions to AgNPs and their stabilization was identified using LC-MS/MS. Nanoparticles were spherical in shape, small in size (mean 13.2 nm), showed crystalline nature, good stability (-18.7 mV) and presence of capping agents. They exhibited antibacterial activity (MIC of 8-128 μg ml-1, MBC of 64-256 μg ml-1) and significantly decreased ATP levels in bacterial cells after treatment with different concentrations of AgNPs. The in vitro analysis showed that the AgNPs demonstrated dose-dependent cytotoxicity against RAW 264.7 macrophages and MCF-7 breast cancer cells but higher against the latter than the former. Cell viability decrease was found to be 42.2-14.2 and 38.0-15.5% while LDH leakage 14.6-42.7% and 19.0-45.0%, respectively. IC50 values calculated for MTT assay was found to be 16.3 and 12.0 μg ml-1 and for LDH assay 102.3 and 76.2 μg ml-1, respectively. Moreover, MCF-7 cells released a greater amount of ROS than RAW 264.7 macrophages during stimulation with all tested concentrations of AgNPs (1.47-3.13 and 1.02-2.58 fold increase, respectively). The SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) analysis revealed the presence of five protein bands at a molecular weight between 31.7 and 280.9 kDa. These proteins showed the highest homology to hypothetical proteins and porins from E. coli, Delftia sp. and Pseudomonas rhodesiae. Based on obtained results it can be concluded that biogenic AgNPs were capped with proteins and demonstrated potential as antimicrobial and anticancer agent.

Abstract:Hypothesis The emergence of Multiple Antibiotic Resistance (MAR) in ESKAPE ( Enterococcus faecium , Staphylococcus aureus , Klebsiella pneumoniae , Acinetobacter baumannii , Pseudomonas aeruginosa , and Enterobacter spp.) pathogens is a global challenge to public health. The inherent antimicrobial nature of silver nanoparticles (AgNPs) makes them promising antimicrobial candidates against antibiotic-resistant pathogens. This study explores the combination of AgNPs with antibiotics (SACs) to create new antimicrobial agents effective against MAR ESKAPE microorganisms. Methods AgNPs were synthesized using Streptococcus pneumoniae ATCC 49619 and characterized for structure and surface properties. The SACs were tested against ESKAPE microorganisms using growth kinetics and time-kill curve methods. The effect of SACs on bacterial biofilms and the disruption of cell membranes was determined. The in-vitro cytotoxicity effect of the AgNPs was also studied. Findings The synthesized AgNPs (spherical, 7.37±4.55 nm diameter) were antimicrobial against MAR ESKAPE microorganisms. The SACs showed synergy with multiple conventional antibiotics, reducing their antibacterial concentrations up to 32-fold. Growth kinetics and time-kill studies confirmed the growth retardation effect and bactericidal activity of SACs. Mechanistic studies suggested that these biofilm-eradicating SACs probably resulted in the loss of bacterial cell membrane integrity, leading to leakage of the cytoplasmic content. The AgNPs were highly cytotoxic against skin melanoma cells but non-cytotoxic to normal Vero cells.

Abstract:Objectives: This article highlights the biological synthesis of silver nanoparticles (AgNPs) with their characteristic analysis, and it focuses on the application of synthesized NPs against multidrug resistance (MDR) bacteria. A cytotoxicity study was performed to assess the biocompatibility. Methods: Silver nanoparticle (AgNPs) formation was confirmed by different characterization methods such as UV-Vis spectrophotometer, Dynamic light scattering (DLS)- Zeta, Fourier transform infrared (FTIR), and Transmission electron microscope (TEM). The antimicrobial activity of the AgNPs was checked against various bacterial strains of Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), Enterococcus faecalis (E. faecalis), and Klebsiella pneumonia (K. pneumonia) by disc diffusion, minimum inhibition concentration test (MIC), and kinetic studies. The cytotoxicity of NPs against the Vero cell line was studied by cytotoxic assay. Results: The primary analysis of the formation of nanoparticles (NPs) was made by UV-Vis spectrophotometric analysis at 400 nm. At the same time, the efficient capping checked by FTIR shows the presence of a functional group at different wavelengths 3284, 1641,1573,1388,1288, and 1068 cm-1. At the same time, the transmission electron microscopic analysis (TEM) and DLS show that the shape and size of the synthesized NPs possess an average size of around ∼10-30 nm with spherical morphology. Further, the zeta potential confirmed the stability of the NPs. While the yield of NPs formation from silver salt was determined by an online yield calculator with the EDX analysis results. Synthesized NPs showed bactericidal effects against all the selected MDR pathogens with nontoxic effects against mammalian cells. Conclusion: Our findings indicate the remarkable antimicrobial activity of the biologically synthesized AgNPs, which can be an antimicrobial agent against multi-drug-resistant bacteria.

Abstract:Biosynthesized silver nanoparticles (Ag-NPs) hold great potential for preventing, treating, diagnosing, and controlling diseases. The primary objective of this study is on marine halophilic bacteria ( Marinobacter litoralis CARE-V18) and their extracellular synthesis of Ag-NPs. The marine bacterial synthesized Ag-NPs is characterized using dynamic light scattering (DLS), zeta potential analysis (ZPA), Fourier transform infrared spectroscopy (FTIR), energy dispersive X-ray (EDX), scanning electron microscopy (SEM), and X-ray diffraction (XRD). The antibiofilm and antibacterial activity of the bacterially synthesized Ag-NPs against human bacterial pathogens are tested ( A. baumannii , K. pneumonia , and MRSA). Furthermore, the acute toxicity effects of Ag-NPs in zebrafish embryos (ZFE) ( Danio rerio ) are also examined in this study. The acute toxicity of Ag-NPs at different time points of post-fertilization (3, 12, 24, 48, 72, and 96 h) eggs of zebrafish. Results shows dose-dependent antibacterial and antibiofilm activity, with significant inhibition observed at 100 and 200 μg/ml concentrations against A. baumannii, K. pneumonia , and MRSA. Thelethal concentration (LC 50 ) values determined as 12.5 and 50 μg/ml. The acute toxicity of Ag-NPs treated ZFE are displayed concentration-dependent mortality rates and increased hatching delay.

Abstract:The use of nanoparticles (NPs) as an alternative to the current generation of conventional antibiotics has exploded in the research community in recent years, as evidence of the superiority of NPs over antibiotics in the treatment of pathogens has been steadily presented. However, therapy with NPs may result in the removal of both multidrug-resistant (MDR) pathogens and commensal bacteria due to the broad-spectrum activity of NPs and the non-specificity of target bacteria. Therefore, the fabrication of MDR-pathogen-targeting NPs is necessary. In this study, biogenic silver nanoparticles (Bio-AgNPs) were synthesized using bacterial cell-free supernatant from three communicating gram-negative bacteria. The size, physical features, and morphology of the AgNPs were characterized by dynamic light scattering (an average size of 158–168 nm), X-ray diffraction (co-ordinate patterns), and transmission electron microscopy (spherical structure). The antibacterial activity of the Bio-AgNPs as minimum inhibitory concentration values was obtained between 0.8 and > 6.4 μg mL −1 for bacterial strains. Mechanistic studies of Bio-AgNPs have revealed that biofilm inhibition, protein leakage, hyperproduction of reactive oxygen species, and physical cell damage are plausible mechanisms underlying the activity of Bio-AgNPs against gram-negative pathogens. Overall, the Bio-AgNPs synthesized in this study may bolster the potential use of Bio-AgNPs as a stand-in for traditional antibiotics, and offer potential specificity against bacterial targets.

Abstract:Background: The increase in nosocomial multidrug resistance and biofilm-forming bacterial infections led to the search for new alternative antimicrobial strategies other than traditional antibiotics. Silver nanoparticles [AgNP] could be a viable treatment due to their wide range of functions, rapid lethality, and minimal resistance potential. The primary aim of this study is to prepare silver nanoparticles and explore their antibacterial activity against biofilms. background: The number of deaths and diseases is expected to increase significantly due to nosocomial multidrug resistance and biofilm-forming bacterial infections. The World Health Organization supports researchers in identifying an alternative antimicrobial that works differently than traditional antibiotics. Silver nanoparticles could be a viable treatment because of their broad spectrum of activity, fast killing power, and low resistance value. Methods: AgNPs with specific physicochemical properties such as size, shape, and surface chemistry were prepared using a chemical reduction technique, and then characterized by DLS, SEM, and FTIR. The activity of AgNPs was tested alone and in combination with some antibiotics against MDR Gram-negative and Gram-positive planktonic bacterial cells and their biofilms. Finally, mammalian cell cytotoxicity and hemolytic activity were tested using VERO and human erythrocytes. Results: The findings of this study illustrate the success of the chemical reduction method in preparing AgNPs. Results showed that AgNPs have MIC values against planktonic organisms ranging from 0.0625 to 0.125 mg/mL, with the greatest potency against gram-negative bacteria. It also effectively destroyed biofilm-forming cells, with minimal biofilm eradication concentrations [MBEC] ranging from 0.125 to 0.25 mg/ml. AgNPs also had lower toxicity profiles for the MTT test when compared to hemolysis to erythrocytes. Synergistic effect was found between AgNPs and certain antibiotics, where the MIC was dramatically reduced, down to less than 0.00195 mg/ml in some cases. Conclusion: The present findings encourage the development of alternative therapies with high efficacy and low toxicity.

Biosynthesis of silver nanoparticle using Bacillus licheniformis culture-supernatant for combating pathogenic biofilms

Abstract:Bacterial biofilms pose a significant threat to healthcare due to their recalcitrance to antibiotics and disinfectants. This study explores the anti-biofilm potential of Bacillus licheniformis cell-free culture supernatant (CFS) and its derived silver nanoparticles (bSNPs) against Staphylococcus aureus and Pseudomonas aeruginosa. The CFS exhibited potent anti-biofilm activity against both bacterial species, even at low concentrations, while devoid of significant bactericidal effects, mitigating resistance risks. Characterization studies revealed the non-proteinaceous nature and thermal stability of the CFS's anti-biofilm agent, suggesting a robust and heat-resistant structure. Green synthesis of bSNPs from CFS resulted in nanoparticles with significant anti-biofilm properties, particularly against P. aeruginosa, indicating differences in susceptibility between the bacterial species. Epifluorescence microscopy confirmed bSNPs' ability to inhibit and partially disrupt biofilm formation without inducing cellular lysis. The study highlights the potential of B. licheniformis CFS and bSNPs as promising biofilm control agents, offering insights into their mechanisms of action and broad-spectrum efficacy. Further research elucidating the underlying molecular mechanisms and identifying specific bioactive compounds is warranted for the translation of these findings into clinically relevant applications for combating biofilm-associated infections.
Biosynthesized silver nanoparticles for inhibition of antibacterial resistance and biofilm formation of methicillin-resistant coagulase negative Staphylococci

Abstract:The ability of a natural stabilizing and reducing agent on the synthesis of silver nanoparticles (Ag NPs) was explored using a rapid and single-pot biological reduction method using Nocardiopsis sp. GRG1 (KT235640) biomass. The UV-visible spectral analysis of Ag NPs was found to show a maximum absorption peak located at a wavelength position of ∼422 nm for initial conformation. The major peaks in the XRD pattern were found to be in excellent agreement with the standard values of metallic Ag NPs. No other peaks of impurity phases were observed. The morphology of Ag NPs was confirmed through TEM observation, demonstrating that the particle size distribution of Ag NPs entrenched in spherical particles is in a range between 20 and 50 nm. AFM analysis further supported the nanosized morphology of the synthesized Ag NPs and allowed quantifying the Ag NPs surface roughness. The synthesized Ag NPs showed significant antibacterial and antibiofilm activity against biofilm positive methicillin-resistant coagulase negative Staphylococci (MR-CoNS), which were isolated from urinary tract infection as determined by spectroscopic methods in the concentration range of 5-60 µg/ml. The inhibition of biofilm formation with coloring stain was morphologically imaged by confocal laser scanning microscopy (CLSM). Morphological alteration of treated bacteria was observed by SEM analysis. The results clearly indicate that these biologically synthesized Ag NPs could provide a safer alternative to conventional antibiofilm agents against uropathogen of MR-CoNS.
Exploring the Potential of Halotolerant Actinomycetes from Rann of Kutch, India: A Study on the Synthesis, Characterization, and Biomedical Applications of Silver Nanoparticles

Abstract:A tremendous increase in the green synthesis of metallic nanoparticles has been noticed in the last decades, which is due to their unique properties at the nano dimension. The present research work deals with synthesis mediated by the actinomycete Streptomyces tendae of silver nanoparticles (AgNPs), isolated from Little and Greater Rann of Kutch, India. The confirmation of the formation of AgNPs by the actinomycetes was carried out by using a UV-Vis spectrophotometer where an absorbance peak was obtained at 420 nm. The X-ray diffraction pattern demonstrated five characteristic diffraction peaks indexed at the lattice plane (111), (200), (231), (222), and (220). Fourier transform infrared showed typical bands at 531 to 1635, 2111, and 3328 cm−1. Scanning electron microscopy shows that the spherical-shaped AgNPs particles have diameters in the range of 40 to 90 nm. The particle size analysis displayed the mean particle size of AgNPs in aqueous medium, which was about 55 nm (±27 nm), bearing a negative charge on their surfaces. The potential of the S. tendae-mediated synthesized AgNPs was evaluated for their antimicrobial, anti-methicillin-resistant Staphylococcus aureus (MRSA), anti-biofilm, and anti-oxidant activity. The maximum inhibitory effect was observed against Pseudomonas aeruginosa at (8 μg/mL), followed by Escherichia coli and Aspergillus niger at (32 μg/mL), and against Candida albicans (64 μg/mL), whereas Bacillus subtilis (128 μg/mL) and Staphylococcus aureus (256 μg/mL) were much less sensitive to AgNPs. The biosynthesized AgNPs displayed activity against MRSA, and the free radical scavenging activity was observed with an increase in the dosage of AgNPs from 25 to 200 μg/mL. AgNPs in combination with ampicillin displayed inhibition of the development of biofilm in Pseudomonas aeruginosa and Streptococcus pneumoniae at 98% and 83%, respectively. AgNPs were also successfully coated on the surface of cotton to prepare antimicrobial surgical cotton, which demonstrated inhibitory action against Bacillus subtilis (15 mm) and Escherichia coli (12 mm). The present research integrates microbiology, nanotechnology, and biomedical science to formulate environmentally friendly antimicrobial materials using halotolerant actinomycetes, evolving green nanotechnology in the biomedical field. Moreover, this study broadens the understanding of halotolerant actinomycetes and their potential and opens possibilities for formulating new antimicrobial products and therapies.

Abstract:This study describes phycocompounds of the non-N2-fixing filamentous cyanobacterium Lyngbya sp., which has potential bio-reducing and stabilizing heavy metal-accumulating properties for synthesizing silver nanoparticles (AgNPs), whose formation was confirmed by the colour change of the Lyngbya sp.-AgNP solution from pale green to deep brown. The reduction of 'Lyngbya sp.-AgNPs', called Lsp-AgNPs, was proved by UV-visible photo-spectrometry analysis with an obtained peak value at 426 nm. Lsp-AgNPs were characterised by analytical techniques, XRD, FESEM, DLS and FTIR. The XRD analysis with 5-70 theta was obtained at 2ϴ angles ranging from 38.79º with intensity, indicating the crystal structure of Lsp-AgNPs. The FESEM analysis indicated the area size at 20-50 µm; in the DLS analysis, the peak at 400 d nm indicated the size and distribution of Lsp-AgNPs. In FTIR analysis, the peaks were obtained at wavenumbers 3338, 1639, and 542 cm-1, which indicated the presence of N-H, -OH and C=O functional groups in Lsp-AgNPs. Those had in vitro antibacterial activities against Gram-negative Escherichia coli (MTCC 443) and Pseudomonas aeruginosa (MTCC 1688) and Gram-positive Staphylococcus aureus (MTCC 7443) bacterial strains with zone of inhibitions (ZOI) of 16, 12 and 14 mm, respectively, with comparing the antibiotic gentamycin as a positive control, as was monitored with agar-well diffusion method. Furthermore, the MIC value was 50 mg/ml, and MBC values of 65 mg/ml of Lsp-AgNPs were effective against those bacteria. Thus, Lsp-AgNPs had potential antibacterial activities against MDR pathogenic S. aureus, E. coli and P. aeruginosa. In conclusion, MDR pathogenic bacteria could be controlled as prodrugs in the future.
