Abstract:Chromium (Cr) contamination of agricultural soils is a major threat to human and plant health worldwide and causes reductions in plant growth and crop yields. 24-epibrassinolide (EBL) and nitric oxide (NO) have been shown to ameliorate the reductions in growth caused by the stresses induced by heavy metals; however, the interactions between EBL and NO on the alleviation of Cr-induced phytotoxicity have been poorly studied. Hence, this study was undertaken to examine any beneficial effects of EBL (0.01 µM) and NO (100 µM), applied alone or in combination, on the mitigation of stress induced by Cr (100 µM) in soybean seedlings. Although EBL and NO applied alone reduced the toxic effects of Cr, the combined treatment had the greatest effect. Mitigation of Cr intoxication occurred via reduced Cr uptake and translocation and by ameliorating reductions in water contents, light-harvesting pigments, and other photosynthetic parameters. In addition, the two hormones increased the activity of enzymatic and non-enzymatic defense mechanisms increasing the scavenging of reactive oxygen species, thereby reducing membrane damage and electrolyte leakage. Furthermore, the hormones reduced the accumulation of the toxic compound, methylglyoxal, by amplifying activities of glyoxalase I and glyoxalase II. Thus, applications of NO and EBL can significantly mitigate Cr-phytotoxicity when cultivating soybean plants in Cr-contaminated soils. However, further more-in depth studies including field investigations parallel with calculations of cost to profit ratios and yield losses are requested to validate the effectiveness of NO and/or EBL for remediation agents in Cr-contaminated soils with using key biomarkers (i.e., oxidative stress, antioxidant defense, and osmoprotectants) involved in the uptake, accumulation, and attenuation of Cr toxicity tested in our study.
Brassinosteroid Supplementation Alleviates Chromium Toxicity in Soybean (glycine Max L.) Via Reducing Its Translocation

Abstract:Chromium (Cr) phytotoxicity severely inhibits plant growth and development which makes it a prerequisite to developing techniques that prevent Cr accumulation in food chains. However, little is explored related to the protective role of brassinosteroids (BRs) against Cr-induced stress in soybean plants. Herein, the morpho-physiological, biochemical, and molecular responses of soybean cultivars with/without foliar application of BRs under Cr toxicity were intensely investigated. Our outcomes deliberated that BRs application noticeably reduced Cr-induced phytotoxicity by lowering Cr uptake (37.7/43.63%), accumulation (63.92/81.73%), and translocation (26.23/38.14%) in XD-18/HD-19, plant tissues, respectively; besides, improved seed germination ratio, photosynthetic attributes, plant growth, and biomass, as well as prevented nutrient uptake inhibition under Cr stress, especially in HD-19 cultivar. Furthermore, BRs stimulated antioxidative defense systems, both enzymatic and non-enzymatic, the compartmentalization of ion chelation, diminished extra production of reactive oxygen species (ROS), and electrolyte leakage in response to Cr-induced toxicity, specifically in HD-19. In addition, BRs improved Cr stress tolerance in soybean seedlings by regulating the expression of stress-related genes involved in Cr accumulation, and translocation. Inclusively, by considering the above-mentioned biomarkers, foliar spray of BRs might be considered an effective inhibitor of Cr-induced damages in soybean cultivars, even in Cr polluted soil.

Abstract:Current investigation demonstrated that chromium (Cr) toxicity affects adversely on the normal growth of soybean plants. However, the seed priming with nitric oxide (NO; 100 μM), and spermine (Spm; 0.01 Mm) can significantly alleviate the Cr toxicity in soybean plant. Herein, the hydroponic experiment was conducted to observe the individual as well as the interactive behavior of NO, and Spm on the various morpho-physiological and, biochemical parameters in soybean such as plant growth, plant height, seed germination indices, photosynthesis-related indices such as chlorophyll biosynthesis, PS system II, nutrient uptake of soybean seedlings against Cr (VI) toxicity. Our outcomes deliberated that the alone treatment of NO, and Spm cause a significant improvement in seed germination ratio, photosynthetic pigments, and biomass of plants by restricting Cr uptake; while NO + Spm treatment being more effective in the improvement of soybean growth relative to their individual treatment under Cr stress. Relative to alone treatment of NO, and Spm, the combined treatment significantly modulated the antioxidant activities, and lowered the ROS accumulation, and electrolyte leakage. In addition, seed priming with NO, and Spm mitigate the Cr-induced toxicity by reducing Cr uptake and stimulating the antioxidative defense mechanisms. Hence, these findings confirmed the positive defense interplay of the NO and Spm in the modulation of the Cr tolerance in soybean. However, the underlying defense mechanism of these synergetic effects needs to be further explored.
Chromium (VI)-Induced Alterations in Physio-Chemical Parameters, Yield, and Yield Characteristics in Two Cultivars of Mungbean (Vigna radiata L.)

Abstract:Chromium (Cr) presently used in various major industries and its residues possess a potent environmental threat. Contamination of soil and water resources due to Cr ions and its toxicity has adversely affected plant growth and crop productivity. Here, deleterious effects of different levels of Cr (VI) treatments i.e., 0, 30, 60, 90, and 120 μM on two mungbean cultivars, Pusa Vishal (PV) and Pusa Ratna (PR), in hydroponic and pot conditions were evaluated. Germination, seedling growth, biomass production, antioxidant enzyme, electrolytic leakage, oxidative stress (hydrogen peroxide and malondialdehyde), and proline content were determined to evaluate the performance of both cultivars under hydroponic conditions for 15 days. The hydroponic results were further compared with the growth and seed yield attributes of both the genotypes in pot experiments performed over 2 years. Seedling growth, biomass production, total chlorophyll (Chl), Chl-a, Chl-b, nitrogen content, plant height, seed protein, and seed yield decreased significantly under the 120 μM Cr stress level. Activities of antioxidant enzymes superoxide dismutase, catalase, ascorbate peroxidase and peroxidase increased in the leaves following Cr exposure at 60-90 μM but declined at 120 μM. Cr-induced reductions in growth and seed yield attributes were more in the sensitive than in the tolerant cultivar. Cr accumulation in the roots, stems, leaves, and seeds increased with an increase in Cr concentrations in the pot conditions. Furthermore, for both cultivars, there were significant negative correlations in morpho-physiological characteristics under high Cr concentrations. Overall results suggest that (PR) is more sensitive to Cr stress (PV) at the seedling stage and in pot conditions. Furthermore, (PV) can be utilized to study the mechanisms of Cr tolerance and in breeding programs to develop Cr-resistant varieties.
Effects of Chromium Stress on the Content of Phytochelatins and the Activities of Key Enzymes of Ascorbate-glutathione Cycle in Brassica Chinensis L.

Abstract:The content of phytochelatins and the activities of key enzymes of Ascorbate-glutathione cycle in varieties of Aikangqing and Lvxiu were studied with different concentrations of Cr3+.The results showed that the contents of phytochelatins and antioxidant increased with the increase of Cr3+ concentration in both varieties.Under high concentration of Cr3+(250 mg.L-1),O 2.contents in leaves of cabbages were significantly increased(P0.05),and the increased amount of Aikangqing was lower than that of Lvxiu.AsA and GSH contents in leaves of Aikangqing were significantly increased compared to contro(lP0.05),while those of in Lvxiu were not significantly increased(P0.05).DHA and GSSG contents in Aikangqing were 1.87 and 1.72 times as much as control and those were 1.98 and 1.94 times as much as control in Lvxiu,respectively.The activities of key enzymes in AsA-GSH increased with the increase of Cr3+ concentration.The content of phytochelatins(PCs),the activity of ascorbte peroxidase(APX),and dehydrateasorbate reductase(DHAR)in Aikangqing were 30.9%,38.9%,45.2% higher than control under the concentration of 250 mg.L-1 Cr3+ treatment,respectively,while those in Lvxiu were 11.5%,30.5%,43.0% higher than control,respectively.In conclusion,Aikangqing had a higher activities of key enzymes of AsA-GSH and phytochelatins under Cr3+ stress,which attribute to its higher Cr3+ tolerance.Our results showed that there were chromium-resistance differences between varieties of cabbages,this provides the theoretical guidance and technical support for people had the choice of low accumulation cabbages varieties.
Chromium (vi) Uptake and Tolerance Potential in Cotton Cultivars: Effect on Their Root Physiology, Ultramorphology, and Oxidative Metabolism

Abstract:Chromium (Cr) is present in our environment as a toxic pollutant, which needs to be removed using phytoremediation technology. In present study, two transgenic cotton cultivars (J208, Z905) and their hybrid line (ZD14) were used to explore their Cr uptake and tolerance potential using multiple biomarkers approach. Four different levels of Cr (CK, 10, 50, and 100 μM) were applied. Cr caused a significant reduction in root/shoot length, number of secondary roots, and root fresh and dry biomasses at 100 μM. Cr accumulated more in roots and was found higher in hybrid line (ZD14) as compared with its parent lines (J208, Z905) at all Cr stress levels (10, 50, and 100 μM). Cr translocation was less than 1 in all cultivars. Ultrastructural studies at 100 μM Cr showed an increase in number of nuclei and vacuoles and presence of Cr dense granules in dead parts of the cell (vacuoles/cell wall). Malondialdehyde (MDA), hydrogen peroxide (H2O2), total soluble proteins, superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), and glutathione reductase (GR) as a whole were upregulated with elevated levels of Cr. Higher Cr uptake by roots, accelerated metabolism, and Cr sequestration in dead parts of the cell indicate that these cotton cultivars can be useful for Cr accumulation and tolerance.
Mitigation of Negative Effects of Chromium (VI) Toxicity in Faba Bean (Vicia faba) Plants through the Supplementation of Kinetin (KN) and Gibberellic Acid (GA3)

Abstract:The present study was carried out to explore the possible role of kinetin and gibberellic acid (GA3) on faba bean under chromium (Cr) stress. Cr treatment negatively affected growth and biomass production, reduced photosynthetic pigments, and inhibited photosynthesis, gas exchange parameters, antioxidant enzymes, and the glyoxylase cycle. Moreover, Cr stress enhanced the production of malondialdehyde (MDA, 216.11%) and hydrogen peroxide (H2O2, 230.16%), electrolyte leakage (EL, 293.30%), and the accumulation of proline and glycine betaine. Exogenous application of kinetin and GA3 increased growth and biomass, improved pigment contents and photosynthesis, as well as up-regulated the antioxidant system by improving the antioxidant enzyme activities and the content of nonenzymatic components, and the glyoxylase cycle. Additionally, kinetin and GA3 application displayed a considerable enhancement in proline (602.61%) and glycine betaine (423.72), which help the plants to maintain water balance under stress. Furthermore, a decline in Cr uptake was also observed due to kinetin and GA3 application. Exogenous application of kinetin and GA3 ameliorated the toxic effects of Cr in faba bean plants, up-shooting the tolerance mechanisms, including osmolyte metabolism and the antioxidant system.

Chromium stress induced oxidative burst in Vigna mungo (L.) Hepper: physio-molecular and antioxidative enzymes regulation in cellular homeostasis

Abstract:Vigna mungo (L.) Hepper commonly known as blackgram is an important legume crop with good quality dietary proteins and vitamins. Low production of blackgram in the chromium rich soil of Odisha is a serious concern against its demand. Chromium (VI) was tested on V. mungo var. B3-8-8 at 100, 150, 200, 250 and 300 µM concentration on growth, anti-oxidative enzymes and chromium content at 15, 30 and 45 d of treatments. Seed germination and growth decreased with increase dose and duration. Cr uptake induced oxidative burst with significant increase of osmolytes was observed in cell at lower doses but failed to adjust homeostasis at higher dose. Increase of GPX and SOD and decrease of CAT was observed as dose dependent. Increased protein content was detected in < 200 µM Cr concentration whereas, significant decrease of protein was noted thereafter. Down regulation of proteins (29.2 kDa and 32.6 kDa) was observed at > 250 µM of Cr. Total Cr uptake was greater in root than in shoot which might be due to poor translocation of heavy metal or detoxification. Thus, blackgram was able to maintain homeostasis at lower concentrations of Cr by activating the cascade of enzymes following cellular detoxification mechanism. Amelioration effect of chromium-tolerant bacteria on growth, physiological properties and chromium mobilization in chickpea (Cicer arietinum) under chromium stress

Abstract:In this study, chromium (Cr)-tolerant bacteria were test for their efficiency in alleviating Cr stress in Cicer arietinum plants. On the basis of 16S rRNA gene analysis, the isolates were identified belonging to genus Stenotrophomonas maltophilia, Bacillus thuringiensis B. cereus, and B. subtilis. The strains produced a considerable amount of indole-3-acetic acid in a medium supplemented with tryptophan. The strains also showed siderophore production (S2VWR5 and S3VKR17), phosphorus production (S1VKR11, S3VKR2, S3VKR16, and S2VWR5), and potassium solubilization (S3VKR2, S2VWR5, and S3VKR17). Furthermore, the strains were evaluated in pot experiments to assess the growth promotion of C. arietinum in the presence of chromium salts. Bacterization improved higher root and shoot length considerably to 6.25%–60.41% and 11.3%–59.6% over the control. The plants also showed increase in their fresh weight and dry weight in response to inoculation with Cr-tolerant strains. The accumulation of Cr was higher in roots compared to shoots in both control and inoculated plants, indicating phytostabilization of Cr by C. arietinum. However, phytostabilization was found to be improved manifold in inoculated plants. Apart from the plant attributes, the amendment of soil with Cr and Cr-tolerant bacteria significantly increased the content of total chlorophyll and carotenoids, suggesting the inoculant's role in protecting plants from deleterious effects. This work suggests that the combined activity of Cr-tolerant and plant growth–promoting (PGP) properties of the tested strains could be exploited for bioremediation of Cr and to enhance the C. arietinum cultivation in Cr-contaminated soils.

Abstract:Chromium-VI (Cr (VI)) is a toxic metal pollutant to both plants and animals. In this study, we cultured two transgenic cotton cultivars (J208 (herbicide-resistant), Z905 (insect-resistant)) and their hybrid line (ZD14) in hydroponic media using 0, 10, 50, 100 µM Cr/L for 7 days. Leaf-related growth parameters such as leaf length and width considerably declined. Decrease in leaves’ fresh biomass was less in Z905 followed by ZD14 and J208, while the leaves dry biomass was less in Z905 followed by J208 and ZD14, respectively. Tolerance potential of plant height, root dry weight and leaf dry weight of both transgenic cotton cultivars significantly decreased at Cr concentration (100 µM) as compared with their hybrid line. Cr accumulation in roots tissues was higher (> 90%) than leaf tissues. Stress biomarkers (i.e., (Malondialdehyde (MDA), hydrogen peroxide (H2O2), total soluble protein (TSP)) as a whole upregulated in leaf tissues of all experimental cultivars. Differential changes in superoxide dismutase (SOD) activity were observed, while peroxidase (POD), catalase (CAT) and glutathione-reductase (GR) activities almost showed increasing trend in all cultivars under exceeding levels of Cr. Correlation studies, in terms of principal component analysis (PCA), revealed that 75.36% of the total variation was present among all the traits. The leaf-based ultrastructural modifications at 100 µM Cr stress level occurred more in Z905 followed by J208 and ZD14. All these changes revealed differential role of experimental cotton cultivars against Cr stress. Hybrid line (ZD14) performed well in comparison with its parental cultivars (J208, Z905) as a whole. Deeper molecular studies like transcriptomic and metabolomic need to be carried out to explore the genetic facts regarding such differential responses among these cotton cultivars under Cr stress.

Abstract:Chromium (Cr) toxicity is hazardous to the seed germination, growth, and development of plants. γ-aminobutyric acid (GABA) is a non-protein amino acid and is involved in stress tolerance in plants. To investigate the effects of GABA in alleviating Cr toxicity, we treated eight-d-old mustard (Brassica juncea L.) seedlings with Cr (0.15 and 0.3 mM K2CrO4, 5 days) alone and in combination with GABA (125 µM) in a semi-hydroponic medium. The roots and shoots of the seedlings accumulated Cr in a dose-dependent manner, which led to an increase in oxidative damage [lipid peroxidation; hydrogen peroxide (H2O2) content; superoxide (O2•−) generation; lipoxygenase (LOX) activity], methylglyoxal (MG) content, and disrupted antioxidant defense and glyoxalase systems. Chromium stress also reduced growth, leaf relative water content (RWC), and chlorophyll (chl) content but increased phytochelatin (PC) and proline (Pro) content. Furthermore, supplementing the Cr-treated seedlings with GABA reduced Cr uptake and upregulated the non-enzymatic antioxidants (ascorbate, AsA; glutathione, GSH) and the activities of the enzymatic antioxidants including ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), glutathione reductase (GR), glutathione peroxidase (GPX), superoxide dismutase (SOD), catalase (CAT), glyoxalase I (Gly I), and glyoxalase II (Gly II), and finally reduced oxidative damage. Adding GABA also increased leaf RWC and chl content, decreased Pro and PC content, and restored plant growth. These findings shed light on the effect of GABA in improving the physiological mechanisms of mustard seedlings in response to Cr stress.

Abstract:Chromium (Cr) as a toxic metal is widely used for commercial purposes and its residues have become a potential environmental threat to both human and plant health. Oilseed rape (Brassica napus L.) is one of the candidate plants that can absorb the considerable quantity of toxic metals from the soil. Here, we used two cultivars of B. napus cvs. ZS 758 (metal-tolerant) and Zheda 622 (metal-susceptible) to investigate the phenological attributes, cell ultrastructure, protein kinases (PKs) and molecular transporters (MTs) under the combined treatments of Cr stress and reduced glutathione (GSH). Seeds of these cultivars were grown in vitro at different treatments i.e., 0, 400 μM Cr, and 400 μM Cr + 1 mM GSH in control growth chamber for 6 days. Results had confirmed that Cr significantly reduced the plant length, stem and root, and fresh biomass such as leaf, stem and root. Cr noticeably caused the damages in leaf mesophyll cells. Exogenous application of GSH significantly recovered both phenological and cell structural damages in two cultivars under Cr stress. For the PKs, transcriptomic data advocated that Cr stress alone significantly increased the gene expressions of BnaA08g16610D, BnaCnng19320D, and BnaA08g00390D over that seen in controls (Ck). These genes encoded both nucleic acid and transition metal ion binding proteins, and protein kinase activity (PKA) and phosphotransferase activities in both cultivars. Similarly, the presence of Cr revealed elite MT genes [BnaA04g26560D, BnaA02g28130D, and BnaA02g01980D (novel)] that were responsible for water transmembrane transporter activity. However, GSH in combination with Cr stress significantly up-regulated the genes for PKs [such as BnaCnng69940D (novel) and BnaC08g49360D] that were related to PKA, signal transduction, and oxidoreductase activities. For MTs, BnaC01g29930D and BnaA07g14320D were responsible for secondary active transmembrane transporter and protein transporter activities that were expressed more in GSH treatment than either Ck or Cr-treated cells. In general, it can be concluded that cultivar ZS 758 is more tolerant toward Cr-induced stress than Zheda 622.

Abstract:BACKGROUND:Chromium (Cr) being multifarious industrial used element, is considered a potential environmental threat. Cr found to be a prospective water and soil pollutant, and thus it is a current area of concern. Oilseed rape (Brassica napus L.) is well known as a major source of edible oil around the globe. Due to its higher growth, larger biomass and capability to uptake toxic materials B. napus is considered a potential candidate plant against unfavorable conditions. To date, no study has been done that described the Cr and GSH mechanism at RNA-Seq level.RESULTS:Both digital gene expression (DGE) and transcriptome profile analysis (TPA) approaches had opened new insights to uncover the several number of genes related to Cr stress and GSH alleviating mechanism in two leading cultivars (ZS 758 and Zheda 622) of B. napus plants. Data showed that Cr inhibited KEGG pathways i.e. stilbenoid, diarlyheptanoid and gingerol biosynthesis; limonene and pentose degradation and glutathione metabolism in ZS 758; and ribosome and glucosinolate biosynthesis in Zheda-622. On the other hand, vitamin B6, tryptophan, sulfur, nitrogen and fructose and manose metabolisms were induced in ZS 758, and zeatin biosynthesis, linoleic acid metabolism, arginine and proline metabolism, and alanine, asparate and glutamate metabolism pathways in Zheda 622. Cr increased the TFs that were related to hydralase activity, antioxidant activity, catalytic activity phosphatase and pyrophosphatase activity in ZS 758, and vitamin binding and oxidoreductase activity in Zheda 622. Cr also up-regulated the promising proteins related to intracellular membrane bounded organelles, nitrile hyrdatase activity, cytoskeleton protein binding and stress response. It also uncovered, a novel Cr-responsive protein (CL2535.Contig1_All) that was statistically increased as compared to control and GSH treated plants. Exogenously applied GSH successfully not only recovered the changes in metabolic pathways but also induced cysteine and methionine metabolism in ZS 758 and ubiquinone and other terpenoid-quinone biosynthesis pathways in Zheda 622. Furthermore, GSH increased the level of TFs i.e. the gene expression of antioxidant and catalytic activities, iron ion binding and hydrolase activity as compared with Cr. Moreover, results pointed out a novel GSH responsive protein (CL827.Contig3_All) whose expression was found to be significantly increased when compared than Cr stress. Results further delineated that GSH induced TFs such as glutathione disulphide oxidoreducatse and aminoacyl-tRNA ligase activity, and beta glucosidase activity in ZS 758. Similarly in Zheda 622, GSH induced the TFs for instance DNA binding and protein dimerization activity. GSH also highlighted the proteins that were involved in transportation, photosynthesis process, RNA polymerase activity, and against the metal toxicity. These results indicated that cultivar ZS 758 had better metabolism and showed higher tolerance against Cr toxicity.CONCLUSION:The responses of ZS 758 and Zheda 622 differed considerably at both physiological and transcriptional level. Moreover, RNA-Seq method explored the hazardous behavior of Cr as well as GSH up-regulating mechanism by activating plant metabolism, stress responsive genes, TFs and protein encyclopedia.
Amelioration of Chromium-Induced Oxidative Stress by Combined Treatment of Selected Plant-Growth-Promoting Rhizobacteria and Earthworms via Modulating the Expression of Genes Related to Reactive Oxygen Species Metabolism in Brassica juncea

Abstract:Chromium (Cr) toxicity leads to the enhanced production of reactive oxygen species (ROS), which are extremely toxic to the plant and must be minimized to protect the plant from oxidative stress. The potential of plant-growth-promoting rhizobacteria (PGPR) and earthworms in plant growth and development has been extensively studied. The present study was aimed at investigating the effect of two PGPR ( Pseudomonas aeruginosa and Burkholderia gladioli ) along with earthworms ( Eisenia fetida ) on the antioxidant defense system in Brassica juncea seedlings under Cr stress. The Cr toxicity reduced the fresh and dry weights of seedlings, enhanced the levels of superoxide anion (O 2 • – ), hydrogen peroxide (H 2 O 2 ), malondialdehyde (MDA), and electrolyte leakage (EL), which lead to membrane as well as the nuclear damage and reduced cellular viability in B. juncea seedlings. The activities of the antioxidant enzymes, viz ., superoxide dismutase (SOD), guaiacol peroxidase (POD), ascorbate peroxidase (APOX), glutathione peroxidase (GPOX), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) were increased; however, a reduction was observed in the activity of catalase (CAT) in the seedlings under Cr stress. Inoculation of the PGPR and the addition of earthworms enhanced the activities of all other antioxidant enzymes except GPOX, in which a reduction of the activity was observed. For total lipid- and water-soluble antioxidants and the non-enzymatic antioxidants, viz ., ascorbic acid and glutathione, an enhance accumulation was observed upon the inoculation with PGPR and earthworms. The supplementation of PGPR with earthworms (combined treatment) reduced both the reactive oxygen species (ROS) and the MDA content by modulating the defense system of the plant. The histochemical studies also corroborated that the combined application of PGPR and earthworms reduced O 2 • – , H 2 O 2 , lipid peroxidation, and membrane and nuclear damage and improved cell viability. The expression of key antioxidant enzyme genes, viz ., SOD, CAT, POD, APOX, GR, DHAR , and GST showed the upregulation of these genes at post-transcriptional level upon the combined treatment of the PGPR and earthworms, thereby corresponding to the improved plant biomass. However, a reduced expression of RBOH1 gene was noticed in seedlings supplemented under the effect of PGPR and earthworms grown under Cr stress. The results provided sufficient evidence regarding the role of PGPR and earthworms in the amelioration of Cr-induced oxidative stress in B. juncea .

Abstract:Melatonin (MT) and reduced glutathione (GSH) roles in mitigating chromium (Cr) toxicity in sweetpotato were explored. Plants, pre-treated with varying MT and GSH doses, were exposed to Cr (40 μM). Cr severely hampered growth by disrupting leaf photosynthesis, root system, and oxidative processes and increased Cr absorption. However, the exogenous application of 1 μM of MT and 2 mM of GSH substantially improved growth parameters by enhancing chlorophyll content, gas exchange (Pn, Tr, Gs, and Ci), and chlorophyll fluorescence (Fv/Fm, ETR, qP, and Y(II)). Furthermore, malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ), superoxide ion (O 2 •− ), electrolyte leakage (EL), and Cr uptake by roots (21.6 and 27.3%) and its translocation to shoots were markedly reduced by MT and GSH application, protecting the cell membrane from oxidative damage of Cr-toxicity. Microscopic analysis demonstrated that MT and GSH maintained chloroplast structure and integrity of mesophyll cells; they also enhanced stomatal length, width, and density, strengthening the photosynthetic system and plant growth and biomass. MT and GSH improved osmo-protectants (proline and soluble sugars), gene expression, and enzymatic and non-enzymatic antioxidant activities, mitigating osmotic stress and strengthening plant defenses under Cr stress. Importantly, the efficiency of GSH pre-treatment in reducing Cr-toxicity surpassed that of MT. The findings indicate that MT and GSH alleviate Cr detrimental effects by enhancing photosynthetic organ stability, component accumulation, and resistance to oxidative stress. This study is a valuable resource for plants confronting Cr stress in contaminated soils, but further field validation and detailed molecular exploration are necessary.

Abstract:Chromium contamination of the soil is a major scientific concern with reference to crop productivity and human health. In recent years, several approaches are being employed in managing metal toxicity in crop plants. Here, we have investigated about potential and probable crosstalk of nitric oxide (NO) and hydrogen peroxide (H2O2) in mitigating hexavalent chromium [Cr(VI)] toxicity in wheat seedlings. Cr(VI) toxicity reduced the fresh mass and overall growth due to accumulation of reactive oxygen species (ROS) and decreased efficiency of AsA-GSH cycle and downregulation of high affinity sulfate transporter. However, exogenous treatment of NO and H2O2 significantly alleviated Cr toxicity. Application of NO and ROS scavengers reversed stress mitigating effects of NO and H2O2, respectively suggesting that endogenous NO and H2O2 are necessary for rendering Cr toxicity tolerance. Furthermore, NO rescued negative effect of diphenylene iodonium (DPI, NADPH oxidase inhibitor) and H2O2 reversed the negative effect of c-PTIO suggesting that they exhibit independent signalling in mitigating Cr stress. Altogether, data indicated that NO and H2O2 rendered mitigation of Cr stress by up-regulating enzymes (activity and relative gene expression) and metabolites of AsA-GSH cycle, high affinity sulfate transporter (relative gene expression) and glutathione biosynthesis which collectively controlled occurrence of oxidative stress.
Genotype-Dependent Effect of Exogenous 24-Epibrassinolide on Chromium-Induced Changes in Ultrastructure and Physicochemical Traits in Tobacco Seedlings

Abstract:Greenhouse hydroponic experiments were carried out using three different heavy metal accumulation tobacco genotypes to evaluate how different genotypes responded to chromium (Cr) toxicity in the presence of 24-epibrassinolide (EBR; a biologically active brassinosteroid). The results showed that Cr stress caused a marked reduction in plant biomass, chlorophyll content, chlorophyll fluorescence, and photosynthesis parameters but induced malondialdehyde accumulation and ultrastructure damage, with 2010-38 (L) less affected. Foliar application of 24-epibrassinolide (0.1 μM) on Cr-stressed plants greatly alleviated Cr-induced inhibition of growth and photosynthesis, oxidative stress and ultrastructure damage, decreased Cr accumulation in different parts of leaves and roots, with the exception of the upper and lower of leaves of genotype L, and maintained ion homeostasis. Regarding genotypes, L was more tolerant than M and H, as it absorbed less Cr and also performed better in all of the studied parameters. These findings suggest a potential role for 24-epibrassinolide in Cr stress alleviation and the utilization of elite genetic resources in future breeding programs to develop low Cr accumulation genotypes. These results advocate a positive role for 24-epibrassinolide in reducing pollutant residues from health point of view.
Chromium toxicity induced oxidative damage in two rice cultivars and its mitigation through external supplementation of brassinosteroids and spermine

Abstract:The chromium (Cr) induced phytotoxicity avowed the scientific community to develop stress mitigation strategies to restrain the Cr accumulation inside the food chain. Whereas, brassinosteroids (BRs), and spermine (SPM) are well-known growth-promoting phytohormones, which enhance the plants health, and resilient the toxic effects under stress conditions. Until now, their interactive role against Cr-mitigation is poorly known. Hence, we conducted the hydroponic experiment to perceive the behavior of seed primed with BRs, or/and SPM treatment against Cr disclosure in two different rice cultivars (CY927; sensitive, YLY689; tolerant). Our findings delineated that BRs (0.01 μM), or/and SPM (0.01 mM) remarkably alleviated Cr-induced phytotoxicity by improving the seed germination ratio, chlorophyll pigments, PSII system, total soluble sugar, and minimizing the MDA contents level, ROS extra generation, and electrolyte leakage through restricting the Cr accretion in roots, and shoots of both rice cultivars under Cr stress. Additionally, the BRs, or/and SPM modulated the antioxidant enzyme, and non-enzyme activities to reduce the Cr-induced cellular oxidative damage as well as maintained the ionic hemostasis in both rice cultivars, especially in YLY689. Concisely, enhanced the plants biomass and growth. Overall, our outcomes revealed that BRs and SPM interact positively to alleviate the Cr-induced damages in rice seedlings on the above-mentioned indices, and combine treatment is much more efficient than solely. Moreover, the effect of BRs, or/and SPM was more obvious in YLY689 than CY927 to hamper the oxidative stress, and boost the antioxidant capacity.

Abstract:Chromium (Cr) pollution affects plant growth and biochemical processes, so, the relative uptake of glycine, nitrate, and ammonium by pak choi (Brassica chinensis) seedlings in treatments with 0mgL-1 and 10mgL-1 Cr (VI) were detected by substrate-specific 15N-labelling in a sterile environment. The short-term uptake of 15N-labelled sources and 15N-enriched amino acids were detected by gas chromatography mass spectrometry to explore the mechanism by which Cr stress affects glycine uptake and metabolism, which showing that Cr stress hindered the uptake of ammonium and glycine but increased significantly the uptake of nitrate. Cr stress did not decrease the active or passive uptake of glycine, but it inhibited the conversion of glycine to serine in pak choi roots, indicating that the metabolism of glycine to serine in roots, rather than the root uptake, was the limiting step in glycine contribution to total N uptake in pak choi. Since Cr affects the relative uptake of different N sources, a feasible way to reduce Cr-induced stress is application of selective fertilization, in particular nitrate, in pak choi cultivation on Cr-polluted soil.
