Abstract:The effects of extraction time, temperature, pressure and different concentration of ethanol and their interactions on the yields of amentoflavone, quercetin and ginkgetin extracted from Taxus chinensis by supercritical CO2 were investigated by using a central composite design (CCD). An CCD experimental design with four factors and five levels was used to optimize the extraction parameters. Ultra performance liquid chromatography (UPLC) was used to analyze the content of the tree components in the extracts. Experimental results show that the main effects of factors and their interactions are significant on the yields (p < 0.05). The optimal extraction conditions were established for the three compounds: yield of 4.47 mg/g for amentoflavone at 48 °C, 25 MPa, 2.02 h and 78.5% ethanol, 3.73 mg/g for quercetin at 46 °C, 24 MPa, 2.3 h, 82% ethanol and 3.47 mg/g for ginkgetin at 48 °C, 20 MPa, 2.38 h, 82% ethanol, respectively.

Abstract:An excellent high-efficiency natural deep eutectic solvent (NADES, ChCl-MA) was screened out and integrated with pulse-ultrasonication technique for extracting phenolic compounds from Carya cathayensis Sarg. peels (CCSPs). Single factor experiment combined with response surface methodology (RSM) using Box-Behnken design (BBD) were employed to investigate significant factors and optimize their influence on extraction of phenolic compounds. Significant synergistic effect triggered by ChCl-MA based pulse-ultrasonication over other methods used alone were proved by comparative study concerning a variety of bioactive components and antioxidant activities. The second-order kinetic model was developed and validated (R2 > 0.99) to describe the extraction process and its mechanism; and second-order kinetic extraction rate constant (k), saturation concentration (Cs), and initial extraction rate (h) were calculated. FT-IR, DSC and SEM results further demonstrated synergistic effect and influence during extraction. Overall, this study provided a green and high-efficiency alternative for the recovery of various phenolics compounds from plant source by-products.
Comparative study on optimization of NADES extraction process by dual models and antioxidant activity of optimum extraction from Chuanxiong-Honghua

Abstract:The natural deep eutectic solvent (NADES) was used as the extraction solvent to extract the four ingredients from Chuanxiong-Honghua. NADES-7 composed of l -proline and lactic acid (1:2) has a better extraction effect compared with traditional solvents. The extraction process was optimized using response surface methodology (RSM) and ant colony algorithm-enabled backpropagation neural network (ACO-BPNN), which were compared with each other. ACO-BPNN has a higher R 2 and predictive value than RSM. The optimal process parameters predicted by the ACO-BPNN were determined to be as follows: solvent-to-material ratio, 8 mL/g; extraction time, 67 min; water concentration, 48%; and extraction temperature, 50 °C. The best predicted comprehensive evaluation value was 4.2634, and the mean value obtained from the experiment was 3.8678. Compared with traditional solvents, the antioxidant activity of NADES-7 extract was better, and the mean value of DPPH, FRAP (ferric reducing antioxidant power), and ABTS assays were 0.1555, 0.1132, and 0.0894 mmol Trolox/g dry weight, respectively. Experimental results show that ACO-BPNN can better optimize the extraction process of many different polar compounds in plant combinations.

Abstract:Natural deep eutectic solvents (NaDESs) are environmentally friendly and efficient for the componential extraction of traditional Chinese medicine compared to conventional organic solvents. In this study, NaDES was screened and employed to extract Danshen–Gegen (DG), and the extraction process was optimised by response surface methodology (RSM) and artificial neural networks (ANN) model. Besides, the in vitro security of extracts of DG were evaluated in PC12 cells. As a result, Betaine-Urea (Bet-Ur) was screened as extraction solvent and ANN model was more accurate than RSM model in optimising the extraction parameter. The extraction process optimised by ANN was as follows: 70% NaDES concentration, 80 mg/mL solid to liquid ratio, 67 °C ultrasonic temperature, and 33 min of ultrasonic time. The comprehensive value of extraction yield was 0.7251 ± 0.84%. IC 50 of Bet-Ur, NaDES DG extract and aqueous DG extract were 0.15%, 0.3% and 10% (v/v).

Abstract:Natural medicines are now gaining increasing acceptance around the global scope. Due to the complexity of composition, it is imperative to adopt a unique evaluation system, which is not confined to the content of finite marker compounds. This research presents the successful application of pharmacodynamics-based optimization to the ultrasound-assisted extraction of Chansu, a widely used natural medicine with complex bioactive ingredients. The effect of solvent concentration, solvent to solid ratio, extraction time and particle size, on two response indexes (extraction yield of total content of bufalin, cinobufagin and resibufogenin and anticancer activity) were evaluated by response surface methodology (RSM). Two models were built based on two response indexes, and both of them provided accurate predictions. Compared with extraction yield-based optimization, pharmacodynamics-based optimization aligns well with the integrity and complexity of natural medicines. Moreover, the pharmacodynamics-based optimization gives a more economical and efficient optimal condition, which is specifically embodied in the coarser requirement for particle size and the saving of extraction time and solvent. The recommended extraction conditions were methanol concentration of 83.54% (v/v), solvent to solid ratio of 26.23 mL/g, extraction time of 40.43 min and particle size of 61 mesh (0.212-0.25 mm). The present study elaborated the feasibility and implication of pharmacodynamics-based optimization, which provides useful information for further research on the extraction process of natural medicines.

Abstract:This paper deals with the influence of network topological changes on the resistive losses for a significant period. It is based on a previous work [1] where the goal was to reduce power losses on a peak load situation. The second part of the work, presented here, attempts to reduce the energy losses of transmission network by implementing a cyclic search of topology changes and phase shifting adjustments over a week time period. As the whole problem for such a long period is not tractable in a Mixed Integer Non-Linear Programming approach, a piecewise-linear formulation is presented in order to find a convenient relation between active losses and real flows for each branch. Subsequently, a mixed integer linear formulation is developed to find an optimal topology that tends to minimize the active losses. Considered topological configurations concern the splitting or merging of electric buses and the line switching. Once the new topology has been identified, the interactions between real and reactive power are handled to adjust the taps of phase shifting transformers through a Mathematical Program with Equilibrium Constraints (MPEC). A practical example based on several hundreds of observations (snapshots) of the French national grid is presented. Obtained results confirm the interest of the method.

Abstract:Dried roots and rhizomes of Salvia miltiorrhiza Bge. (Danshen in Chinese) are widely used in Chinese herbal medicine. These components from Danshen have significant anti-oxidation properties owing to high levels of tanshinone IIA (TIIA) and salvianolic acid B (Sal B). To make the best use of this natural resource, response surface methodology (RSM) and artificial neural network (ANN) were used for the modeling and optimization of ultrasound-assisted extraction (UAE) of TIIA and Sal B together to determine the antioxidant activity of the extracts obtained from Danshen. In this study, the Box-Behnken design (BBD) was used to improve extraction time (X1), solvent-to-material ratio (X2), extraction temperature (X3), and ethanol concentration (X4) for the optimal combination of the comprehensive yield of TIIA and Sal B (Y1) and the antioxidant activity (Y2). The optimal process parameters were determined to be as follows: extraction time, 73 min; solvent-to-material ratio, 11 mL/g; extraction temperature, 76℃; and ethanol concentration, 80 %. Using these conditions, the predictive optimal combination revealed a comprehensive evaluation value of 16.2281 and an antioxidant activity of 1.1453 mM FeSO4/5 g, while the experimental average values for these parameters were determined to be 16.1826 and 1.1415 mM FeSO4/5 g, respectively. It was clear that the ANN model had higher accuracy in predictive and optimization capabilities, with higher R2 and lower RMSE, MAE, and relative deviations values, than did RSM. Hence, the ANN model proved to be more effective for the analysis and improvement of the extraction process.

Abstract:To investigate the extraction effect of total flavonoids from Crataegus pinnatifida by using ultrasonic-assisted deep eutectic solvents, and optimize the extraction parameters. Firstly, ten kinds of deep eutectic solvents (DESs) composed with different components were prepared for extracting total flavonoids of C. pinnatifida, and the DESs with the highest yield were selected. Then, water content and the proportion of each component contained in DESs were screened on the basis of the single factor experiments. Meanwhile, the response surface methodology (RSM) was employed to optimize extraction temperature, liquid-solid ratio and ultrasonic time, and obtain the extraction parameters. As results, glycerol/choline chloride (molar ratio 3:1) DESs containing 50% water was chosen as the optimal extraction solvent, and the optimum extraction parameters were as follow: Ultrasonic time (21 min), liquid-solid ratio (42 mL/g) and extraction temperature (72 ℃). Under the above extraction condition, the yield of total flavonoids, rutin, vitexin, hyperoside, quercetin in DESs extract were 7.72%, 0.24%, 0.33%, 0.18% and 0.27%, respectively, which were superior to the traditional alcohol extraction method. The DPPH radical scavenging rate of DESs extract at 0.1 mg/mL was 86%. Compared with methanol extract, the antioxidant activity of DESs extract was enhanced. Therefore, ultrasonic-assisted deep eutectic solvents extraction method could effectively improve the yield of total flavonoids from C. pinnatifida, and would provide the scientific evidence for the resource development and utilization of C. pinnatifida.

Abstract:Health foods containing Panax notoginseng extract are developed in recent years. In this work, reflux extraction process was optimized using a design space approach to improve batch-to-batch consistency of P. notoginseng extract. Saponin yields and dry matter yield were identified as the process critical quality attributes (CQAs) of the extraction process. A risk assessment was applied to determine critical process parameters (CPPs), which were ethanol content (EC), amount of ethanol added (AEA), extraction time (ET) and extraction frequency (EF). Box-Behnken designed experiments were carried out to develop models between CPPs and process CQAs. Determination coefficients were higher than 0.90 for all the models. Higher ET, EF, and AEA all result in higher saponin yields and dry matter yield. Dry matter yield decrease as EC increases. Design space was calculated using a Monte-Carlo simulation method with the acceptable probability of 0.85. Normal operation ranges to attain process CQA criteria with a probability more than 94% are recommended as follows: EC of 84.0-86.0%, AEA of 5.0-7.0 mL/g, ET of 7.0-8.0 h, and EF of 2. Verification experiment results showed that operating EC, AEA, EF and ET within design space can attain CQA criteria. Most of the verification experiment results agreed well with prediction results, which means that the developed models are accurate and applicable in a larger scale extraction process. Abstract:Natural deep eutectic solvents (NADESs), composed of natural primary metabolites, are now widely used as green and sustainable extraction solvents of bioactive components. In the present study, various NADESs were prepared to extract multi-components from different preparations of an herbal formula (Chinese name: Jinqi Jiangtang, JQJT) using ultrasound-assisted extraction (UAE). Results showed that most prepared NADESs provided more effective extraction of phenolic acids and alkaloids from JQJT preparations than conventional solvents. Among the tested NADESs, the solvent formed by choline chloride and laevulinic acid was selected to optimize the operational parameters using response surface methodology. The optimized extraction method was successfully applied to extract six major components in four commercial JQJT products, and quantification analysis was performed by the validated high-performance liquid chromatography with ultraviolet detection (HPLC-UV) method. The quantitative results indicated that preparations from different manufacturers showed different chemical profiles. In conclusion, NADESs-based UAE shows considerable potential as an efficient and green method for extraction of multi-bioactive components from commercial herbal preparations.

Abstract:A series of natural deep eutectic solvents (NADESs) were prepared with choline chloride, betaine, and a variety of natural organic acids in order to find new environmentally-friendly green solvents to replace the traditional solvents. The NADESs were employed to extract flavonoids from Potentilla fruticosa L. (PFL) with the help of ultrasound. The eutectic solvent diluted with an appropriate amount of water improved the extraction ability of flavonoids due to the decrease of solution viscosity. The microstructure of the raw sample and the samples subjected to ultrasonic bath in different solutions were observed using scanning electron microscope (SEM) to determine the role of the NADESs in the extraction process. The DPPH method and glucose consumption method were used to study the antioxidant and hypoglycemic ability of flavonoid compounds in PFL. Single factor method and response surface methodology (RSM) were designed to analyze the effects of three extraction parameters, including solvent/solid ratio, ultrasonic power, and extraction time, on the extraction yield, antioxidant capacity, and hypoglycemic capacity, and the corresponding second-order polynomial prediction models were established. The optimal extraction conditions for the maximum extraction yield, antioxidant capacity, and hypoglycemic capacity were predicted by RSM, and the reliability of RSM simulation results was verified by a one-off experiment.

Abstract:Deep eutectic solvents (DESs) have been regarded as a substitute extraction solvent for conventional organic reagents due to their low toxicity and unique solubility. In this study, fifteen choline chloride-based DESs were prepared and successfully integrated with microwave-assisted extraction (MAE) technique for extracting Hibiscus manihot L. flowers natural antioxidants. The selected DES exhibited superior extraction performance than those by water and 60% ethanol. After statistical optimization by single-factor experiments, Taguchi orthogonal design and response surface methodology, the maximal extraction yields of ten natural antioxidants reached 0.880, 0.328, 0.176, 0.243, 5.143, 4.098, 4.852, 0.171, 0.509 and 0.303 mg/g, which was 1.42–1.94 folds to those by conventional extraction media and methods. In addition, ten natural antioxidants were enriched and recovered with a satisfactory recovery yield of 88.45%–92.02%, and the regenerative DES could be recycled at least four times. Overall, this study established a green, safe, high-efficiency and sustainable alternative for efficiently extracting bioactive ingredients from plant matrices.

Abstract:In this study, we employed ultrasound-assisted extraction in conjunction with natural deep eutectic solvents (NADES) to investigate an innovative and sustainable approach for the selective extraction of Panax notoginseng (PN) saponins. Fifteen NADES were prepared, of which choline chloride-urea (ChCl-Ur) was selected as the optimal solvent because of the higher solubility of PN saponins. Response surface methodology using Box-Behnken design was adopted to optimize the extraction process. According to the results, NADES with a water content of 45 % (v/v) and a ChCl/Ur ratio of 1:2 (mol/mol) exhibited superior efficacy as a solvent for extracting saponins from PN. The optimal extraction conditions include a liquid–solid ratio of 40 mL/g, an extraction temperature of 47 °C, and an extraction duration of 36 min. The extraction efficiency of NADES for saponins was superior to that of water and comparable to that of 70 % (v/v) methanol. Additionally, we assessed the anti-inflammatory and anticoagulant properties of the resulting extraction solution. The TNF- α and IL-1 β inhibiting test showed that the NADES extract of saponins exhibited a stronger inhibitory effect on the mRNA expression of inflammatory cytokines TNF- α and IL-1 β when compared to an aqueous extract. And a dose-dependent relationship was observed. In the anticoagulation activity assay, the NADES extract of saponins at a concentration of 25 mg/mL significantly prolonged the activated partial thromboplastin time ( p < 0.05). Additionally, concentrations of 12.5 and 25 mg/mL exhibited significant prolongation of the prothrombin time ( p < 0.01). These findings suggest that NADES can be used as a solvent to extract PN saponins in an efficient way, and the specific concentrations of the extract possess potent bioactivities.

Abstract:In this study, an ultrasonic-assisted natural deep eutectic solvent (NaDES) was used to extract flavonoids from Perilla frutescens (L.) Britt. leaves. Of 10 tested NaDESs, that comprising D-(+)-glucose and glycerol exhibited the best total flavonoid extraction rate. Response surface methodology (RSM) was used for extraction modeling and optimization, and the total flavonoid content reached 87.48 ± 1.61 mg RE/g DW, which was a significant increase of 5.36% compared with that of 80% ethanol extraction. Morphological changes in P. frutescens leaves before and after extraction were analyzed by scanning electron microscopy (SEM), and the mechanism of NaDES formation was studied by Fourier transform infrared (FT-IR) spectroscopy. Furthermore, 10 flavonoids were identified by UPLC-Q-TOF-MS. In addition, the NaDES extract had better biological activity according to five kinds of antioxidant capacity measurements, cyclooxygenase-2 (COX-2) and hyaluronidase (Hyal) inhibition experiments. Moreover, the stability test revealed that the total flavonoid loss rate of the NaDES extract after four weeks was 37.75% lower than that of the ethanol extract. These results indicate that the NaDES can effectively extract flavonoids from P. frutescens leaves and provide a reference for further applications in the food, medicine, health product and cosmetic industries.

Abstract:Mesona chinensis Benth (MCB) is the source of the most commonly consumed herbal beverage in Southeast Asia and China and is thus an economically important agricultural plant. Therefore, optimal extraction and production procedures have significant commercial value. Currently, in terms of green chemistry, researchers are investigating the use of greener solvents and innovative extraction techniques to increase extract yields. This study represents the first investigation of the optimal conditions for ultrasound-assisted deep eutectic solvent (DES) extraction from MCB. The major factors influencing ultrasound-assisted DESs were optimized using the response surface methodcentral-genetic algorithm-back propagation neural networks. This model demonstrated superior predictability and accuracy compared to the RSM model. Various types of DESs were used for the extraction of MCB constituents, with choline chloride-ethylene glycol resulting in the highest yield. The optimal conditions for maximal extraction were the use of choline chloride-ethylene glycol (1:4) as the solvent with a 40% water content, an extraction duration of 60 min at 60°C, and maintaining a leaf-to-solvent ratio of 20 mL/g. Noticeable enhancements in Van der Waals forces and more robust interactions between DESs and the target chemicals were observed relative to those seen with ethanol (70%, v/v) or water. This investigation not only introduced an environmentally friendly approach for highly efficient extraction from MCB but also identified the mechanisms underlying the improved extraction efficacy. These findings have the potential to contribute to the broader utilization of MCB and provide valuable insights into the extraction mechanisms utilizing deep eutectic solvents. PRACTICAL APPLICATION: This work describes an efficient and green ultrasound-assisted deep eutectic solvent (DES) method for Mesona chinensis Benth (MCB) extraction. Molecular dynamics was used to examine the intermolecular interactions between the solvent and the extracted compounds. It is anticipated that green and environmentally friendly solvents, such as DESs, will be used in further research on foods and their bioactive components. With the development of the herbal tea industry, new products made of MCB are becoming increasingly popular, thus gradually making it a research hotspot.

Abstract:In recent years, natural deep eutectic solvents have been favored greatly due to their environment friendly, mild biological toxicity and simple biodegradability. Natural deep eutectic solvents gradually applied for the extracting bioactive compounds from natural products efficiently. In this study, 20 natural deep eutectic solvents were prepared and their physical and chemical properties were tested. The ultrasonic-assisted extraction method was used to extract flavonoids from Trollius ledebouri and high-performance liquid chromatography-ultraviolet was applied to examine two main bioactive flavonoids (orientin and vitexin). Compared with traditional solvents (water and 60% ethanol solution), natural deep eutectic solvents composed of L(-)-proline and levulinic acid (molar ratio 1:2) show a super extraction efficiency. On this basis, the response surface method was used to optimize the extraction temperature, extraction time, water contents, and solid-liquid ratio. As a consequence, the extraction temperature 60 degrees C, extraction time 18 min, water content 14% (v/v), and the solid-liquid ratio 48mL.g(-1) were chosen as the best extraction process. This study shows that natural deep eutectic solvents can effectively extract flavonoids from T. ledebouri, laying a foundation for the further application of natural deep eutectic solvents to extract bioactive compounds from natural products.

Abstract:Objective To study the best extraction process of Qiming granules.Methods Using the content of puerarin,flavonoids and total nitrogenand as an assessing index to determine the optimum extraction conditions.Content of ethanol,amount of ethanol,extracting temperature and extracting time were selected by orthogonal design and the analysis of variance.Results The optimum conditions for the extraction of puerarin,flavonoids and total nitrogen were 65% ethanol,which was 8 times of the amount of herb material,refluxed 2 hours at 90℃.Conclusion The optimal extraction process is steady and reliable.

Abstract:Variable selection methods can simplify the modelling process and improve the accuracy of models constructed for rapid monitoring using near-infrared (NIR) spectroscopy. In this study, NIR spectroscopy was applied in combination with chemometrics to determine the icariin, salvianolic acid B, and puerarin content, as well as the relative density through the concentration process of a Yangxinshi tablet (YXST) water extraction solution. Partial least squares with several variable selection algorithms were used for the modelling. Variable selection methods, including synergy interval partial least squares (Si-PLS), a genetic algorithm, competitive adaptive reweighted sampling (CARS), and their combination, were comparatively applied to calibrate the regression model. The performances of the models obtained were systematically evaluated according to the correlation coefficients of prediction (Rp), the relative standard error of prediction (RSEP), and the residual predictive deviation (RPD). For icariin, salvianolic acid B, and puerarin, the Si-PLS model showed optimal results. The Rp, RSEP, and RPD of Si-PLS were 0.9905, 7.84%, and 7.38 for icariin; 0.9781, 9.48%, and 4.80 for salvianolic acid B; and 0.9668, 12.81%, and 3.92 for puerarin, respectively. For the relative density, the CARS-Si-PLS acquired the best prediction results. The Rp, RSEP, and RPD of CARS-Si-PLS were 0.9725, 0.47%, and 4.31 for the relative density, respectively. This study demonstrated that variable selection methods can simplify the modelling process and improve the accuracy of the models. In addition, the methods developed in this study are suitable for a quality monitoring of the concentration process of YXST water extraction solution, and they provide reference for real-time monitoring of the concentration processes used in the preparation of other types of traditional Chinese medicine.
Comprehensive Evaluation of Deep Eutectic Solvents in Extraction of Bioactive Natural Products

Abstract:Deep eutectic solvents (DESs) are emerging as green and sustainable solvents for efficient extraction of bioactive compounds or drugs. This work aimed to comprehensively evaluate the potential and effectiveness of DESs for extraction of different types of natural compounds from biomass. Five Chinese herbal medicines including Berberidis Radix, Epimedii Folium, Notoginseng Radix et Rhizoma, Rhei Rhizoma et Radix, and Salviae Miltiorrhizae Radix et Rhizoma were selected to assess the efficiency of DESs on extraction of alkaloids, flavonoids, saponins, anthraquinones, and phenolic acids, respectively. Totally 43 types of choline chloride-, betaine-, and l-proline-based DESs with different polarity, viscosity, composition, and solubilization abilities were tailored to test their extraction efficiency, and the operation conditions were statistically optimized using response surface methodology to produce the most efficient process. In this work, DES solvents were first introduced to extract alkaloids and anthraquinones. The results indicated that most prepared DESs proved to be efficient solvents for extraction of alkaloids, but lower extractability for anthraquinones. The extraction capacity of DES may be correlated with their physical–chemical properties, including H-bonding interactions, polarity, viscosity, and pH. This study demonstrated that DESs were suitable green extraction solvents for selectively and efficiently extracting bioactive compounds from biomaterials.
