Abstract:Background: Diabetes mellitus is a metabolic disorder associated with relative or absolute insulin deficiency or resistance, characterized by hyperglycemia. Modern prescriptions such as pioglitazone have better therapeutic potential, but its side effects and financial burden for developing countries have motivated the researchers to find alternative natural drugs to compete hyperglycemia in patients with diabetes. The present study was conducted to explore the therapeutic potential of selected medicinal plants for the treatment of diabetes as an alternative to allopathic medicines. Method: In present study, hydroalcoholic extracts of Curcuma longa, Lavandula stoechas, Aegle marmelos, and Glycyrrhiza glabra and their polyherbal preparation (PHP) as compound drug were investigated for their antihyperglycemic potential in alloxan-induced diabetic mice. The study subjects (mice) were divided into different groups as normal control, diabetic control, pioglitazone treated (standard drug), test groups (plant extract treated 50, 100, and 150 mg/kg body weight), and PHP-treated group. Blood glucose concentration of all the study animals was determined by Glucose strip test. Qualitative phytochemical analysis of all the plant extracts was also performed following standard methods. Result: It was investigated that treatment of alloxan-induced diabetic mice with hydroalcoholic extracts of studied medicinal plants showed significant (P < .05) effects on fasting blood glucose levels (from baseline to normal range) in a manner comparable to that of the reference drug, pioglitazone (1 mg/kg body weight intraperitoneal). The tested plant extracts significantly (P < .05) reduced the glucose concentration in blood of diabetes-induced mice in a dose-dependent manner. Conclusion: It could be concluded that studied medicinal plants have antihyperglycemic activity. The study findings favor the use of traditional herbal medicinal practices for the management of diabetes that might due to the presence of bioactive phytoconstituents in plants. However, larger studies are required to identify, isolate, and characterize the bioactive phytoconstituents responsible for antihyperglycemic activity of studied medicinal plants.

Abstract:A very widespread biologic correlate of the acquisition of cell walls as osmotic controllers was the development (or retention) of proton pumps as the primary agents of active transport. The majority of plant, fungal, and bacterial cells eject a stream of protons, coupled to the capture of photons, the oxidation of pyridine nucleotides, or the hydrolysis of ATP, and they use the resultant transmembrane difference of potential to drive the uptake of organic nutrients (e.g., amino acids, sugars, nucleotides) and critical ions (K+, phosphate, etc.) and the extrusion of catabolic products and superfluous ions. Plasma-membrane proton pumps also contribute essentially to the background conditions for cytoplasmic pH control, just as in wall-less cells plasma-membrane sodium pumps establish the background conditions for osmotic control. Because the physiologic roles of these two species of ion pumps are so similar, it is not surprising, as described in a previous paper at this symposium,' that plasma-membrane proton pumps closely resemble the pumps for alkali metal cations in animal-cell membranes in general physical properties, in reaction chemistry, in secondary structure, and appreciably in primary structure. But against the impressive similarities between these two species of ion pumps, and among several others as well (e.g., plasma membrane and SR calcium pumps, the gastric proton pump, and the bacterial potassium pump), certain differences stand out sharply and probably will turn out to be correspondingly important to our eventual understanding of the molecular mechanism(s) of ion pumping. For economy, I shall consider only four of the differences: stoichiometry, apparent substrate affinity, intrinsic velocity for one particular reaction step, and a preponderant localization of acidic residues near the N-terminus of the polypeptide sequence. All of these have been most clearly defined in the plasma-membrane proton pumps of fungi which, like the alkali cation pumps, are M, 100,000 proteins that become phosphorylated during the reaction cycle and are blocked by orthovanadate ions. This general class of membrane enzymes has been dubbed EI-E2 ATPases2 or P-type A T p a ~ e s . ~

Abstract:Background and Objective: Plants of the genus Marrubium are used to cure several infectious diseases like asthma, rheumatoid and diabetes. This study investigated in vivo antihyperglycemic capacity of the methanolic fraction (MeOHE) of two Marrubium species via animal model. Materials and Methods: The plant extracts were investigated both qualitatively and quantitatively to reveal their phytochemical composition. Pharmacologically speaking, Wistar albino rats of either sex (150-180 g) were used to investigate the antidiabetic effect using 25 rats in each of the two used approaches; respectively Oral Glucose Tolerance Test (OGTT) and streptozotocin-induced diabetes approaches. In addition, various biochemical parameters were investigated to confirm the pharmacological quality of our plants. Evaluated for their possible antidiabetic effect during two consecutive weeks for glycemia levels (2 hrs period). Results: Data revealed that MeOHE induced a significant antihyperglycemic effect, in which M. deserti, a slight decrease in the glucose level at (143±0.5 mg/dL) was obtained while a reduction was recorded for M. vulgare (71.5±0.75 mg/dL) with a same observation for triglycerides, cholesterol, glutamic-oxaloacetic transaminase (TGO), glutamic pyruvic transaminase (TGP), creatinine comparing to standard. Moreover, the result showed high levels of phenolic and flavonoïds content. Conclusion: This modest work confirmed once again the real pharmacological capacity of vegetals and their possible implication to cure diabetes and complications related to it underlying the antihyperglycemic activity of these plants.

Abstract:The current study aimed to establish the mechanisms of antidiabetic activity of methanolic extract of Punica granatum leaves (MEPGL) in nicotinamide/streptozotocin-induced type 2 diabetes in rats. Phytochemical screening, HPLC analysis, and acute toxicity study of MEPGL were carried out. Various concentrations of MEPGL (100, 200, 400, and 600 mg/kg) were administered orally to diabetic rats for 45 days on a daily basis. The antidiabetic effect of MEPGL was examined by measuring blood glucose, plasma insulin, and glycated hemoglobin (HbA1c) levels, as well as with an oral glucose tolerance test. The antioxidant effect of MEPGL was determined by analyzing hepatic and renal antioxidant markers, namely superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH), and lipid peroxidation. The other biochemical markers alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), urea, and creatinine, as well as total cholesterol, triglycerides, and high-density lipoprotein (HDL) were also studied. Type 2 diabetes significantly altered these parameters, while oral administration of the MEPGL significantly ameliorated them. Moreover, the pancreatic histopathological changes were attenuated with MEPGL treatment. In a nutshell, oral MEPGL administration in diabetic rats showed antidiabetic activity due to its antioxidant activity, most probably due to the gallic acid, ellagic acid, and apigenin found in MEPGL.

Abstract:We hypothesized that some medicinal herbs and food plants commonly used in the management of diabetes can reduce glucose peaks by inhibiting key carbohydrate hydrolyzing enzymes. To this effect, extracts of Antidesma madagascariense (AM), Erythroxylum macrocarpum (EM), Pittosporum senacia (PS), and Faujasiopsis flexuosa (FF), Momordica charantia (MC), and Ocimum tenuiflorum (OT) were evaluated for α-amylase and α-glucosidase inhibitory effects based on starch-iodine colour changes and PNP-G as substrate, respectively. Only FF and AM extracts/fractions were found to inhibit α-amylase activity significantly ( P < 0.05 ) and coparable to the drug acarbose. Amylase bioassay on isolated mouse plasma confirmed the inhibitory potential of AM and FF extracts with the ethyl acetate fraction of FF being more potent ( P < 0.05 ) than acarbose. Extracts/fractions of AM and MC were found to inhibit significantly ( P < 0.05 ) α-glucosidase activity, with IC 50 comparable to the drug 1-deoxynojirimycin. In vivo studies on glycogen-loaded mice showed significant ( P < 0.05 ) depressive effect on elevation of postprandial blood glucose following ingestion of AM and MC extracts. Our findings tend to provide a possible explanation for the hypoglycemic action of MC fruits and AM leaf extracts as alternative nutritional therapy in the management of diabetes.
Comparison of the Efficiency of Lepidium sativum, Ficus carica, and Punica granatum Methanolic Extracts in Relieving Hyperglycemia and Hyperlipidemia of Streptozotocin-Induced Diabetic Rats

Abstract:Background: Diabetes mellitus (DM) is a metabolic disorder characterized by high blood glucose levels that occurs either due to insufficient insulin production or mounting resistance to its action. The purpose of this study was to investigate if methanolic extracts of Lepidium sativum seeds, Ficus carica, and Punica granatum leaves had any effect on blood sugar levels in normal and streptozotocin (STZ) diabetic rats, as well as to explore the most effective extract. Method: Healthy male albino rats weighing 185-266 g were divided into nine groups of eight rats each: normal control, diabetic control, diabetic rats with dietary supplements of L. sativum, F. carica, and P. granatum methanolic extracts, and diabetics treated with insulin. All of the rats were fed on ordinary diet with nutritional pellets and were given water ad libitum. To induce diabetes, all animals were administered with STZ intraperitoneal injection at a dose of 60 mg/kg body weight. For five weeks, the crude plant extracts were given orally to various groups of rats at doses of one hundred and two hundred mg/kg body weight. After that, animal groups were sacrificed and blood samples were taken. Results: Phytochemical analysis revealed that the highest amounts of polyphenolic compounds were present in L. sativum seeds and P. granatum leaves, while leaves of F. carica showed the highest amounts of alkaloid and flavonoid content compared to other extracts. Oral administration of F. carica and L. sativum extracts at the dosage of 100 and 200 mg/kg significantly reduced glucose, lipid profile, kidney, and liver enzyme levels. A significant increase in HbAlc levels was also observed with L. sativum extract at a dose of 200 mg/kg compared to diabetic controls. Mellitus rats supplemented with 100 and 200 mg/kg methanolic extracts of P. granatum had higher serum triglycerides and lower serum low-density lipoprotein cholesterol (LDL-C) than normal control rats. F. carica extract is more effective than L. sativum and P. granatum extracts in the prevention and control of type 2 diabetes mellitus (T2DM) and its consequences.

Abstract:Plantago australis Lam. Subsp. hirtella (Kunth) Rahn is a medicinal plant used as a diuretic, anti-inflammatory, antibacterial, throat cancer treatment and for the control of diabetes. P. australis was collected in the state of Morelos, México. The hydroalcoholic extract (HAEPa) of P. australis was obtained by maceration and concentrated in vacuo. Once dry, it was evaluated through an oral glucose tolerance test (OGTT) in normoglycemic mice and in a non-insulin-dependent diabetic mice model. The expression of PPARγ and GLUT-4 mRNA was determined by rt-PCR, and GLUT-4 translocation was confirmed by confocal microscopy. The toxicological studies were conducted in accordance with the guidelines suggested by the OECD, sections 423 and 407, with some modifications. HAEPa significantly decreased glycemia in OGTT curves, as well as in the experimental diabetes model compared to the vehicle group. In vitro tests showed that HAEPa induced an α-glucosidase inhibition and increased PPARγ and GLUT-4 expression in cell culture. The LD50 of HAEPa was greater than 2000 mg/kg, and sub-chronic toxicity studies revealed that 100 mg/kg/day for 28 days did not generate toxicity. Finally, LC-MS analysis led to the identification of verbascoside, caffeic acid and geniposidic acid, and phytochemical approaches allowed for the isolation of ursolic acid, which showed significant PPARγ overexpression and augmented GLUT-4 translocation. In conclusion, HAEPa induced significant antidiabetic action by insulin sensitization through PPARγ/GLUT-4 overexpression.

Biological and Hypoglycemic Effects of a Polyherbal Extract on Alloxanized Diabetic Rats

Abstract:The current study investigates the antioxidant, antidiabetic, hepatoprotective, and nephroprotective potentials of a polyherbal mixture containing the methanolic extracts of seeds from Nigella sativa, Cicer arietinum, Silybum marianum, and Citrullus colocynthis and the rhizome of Zingiber officinale. The polyherbal extract (PHE) showed significant total phenolic contents (187.17 GAE/g), ferric reducing power (28%), and radical-scavenging activity (86.16%). The PHE also showed a substantial hypoglycemic effect in alloxan-induced diabetic rats by reducing the blood glucose level of the PHE-treated rats (-48.64%) and increasing the insulin level (107.5%) as compared with the diabetic control group. Likewise, an increase in high-density lipoprotein (HDL) contents (22.95%) with an associated decrease in low-density lipoprotein (LDL) levels (-43.93%) was also noted. A significant decrease in serum levels of liver marker enzymes, e.g., SGPT (-36%), SGOT (-31%), and serum ALP (-12%), was also observed as compared with the standard drug-treated group. Based on the findings of the study, it may be suggested that PHE helps ameliorate the severity of diabetes as a herbal remedy and might be employed in nutra-pharmaceuticals, replacing synthetic antidiabetic compounds.
Squamous differentiation in small-cell carcinoma of the parotid gland.

Abstract:Small-cell anaplastic carcinomas comprise 1% to 2% of major salivary gland malignant tumors and demonstrate an aggressive clinical course. The initial classification of salivary small-cell anaplastic carcinoma was based on the ultrastructural identification of membrane-bound dense core granules, confirming neuroendocrine differentiation. These neuroendocrine-type small-cell carcinomas were felt to arise from neuroendocrine stem cells that migrated to the salivary gland from the neural crest. Absent neuroendocrine differentiation by ultrastructural evaluation was felt to signify origin from ductal cells. Immunohistochemical study has revised this concept because many small-cell carcinomas express at least one neuroendocrine marker, even in the absence of ultrastructural evidence of neuroendocrine differentiation. In addition, glandular differentiation both by ultrastructural and light microscopic study has been found in cases showing neuroendocrine differentiation. Ultrastructural evidence for squamous differentiation, such as desmosomes and tonofilaments, has also been recognized. These new findings have led to a revision of the old histogenetic hypothesis. All of these small-cell carcinomas are presumed to arise from a hypothetical ductal stem cell that can undergo neuroendocrine, squamous, or glandular differentiation. We report a small-cell anaplastic carcinoma of the left parotid gland in a 61-year-old man with squamous differentiation identified by light microscopy and confirmed by immunohistochemical expression of predominantly high rather than low molecular weight cytokeratins. This tumor is distinctive because it shows an abrupt transition from small-cell anaplastic carcinoma with neuroendocrine differentiation to well-differentiated squamous differentiation, which was identified readily by light microscopy. Our findings support this new hypothesis of a single multipotential stem cell by demonstrating bidirectional neuroendocrine and squamous differentiation.

Abstract:Abstract In the Algerian desert, individuals often enjoy a soothing cup of herbal tea made from Ephedra alata Decne before bedtime; this aids in their relaxation. Our previous and earlier investigations demonstrated the richness of the shrub in terms of polyphenols and flavonoids and their effective medicinal properties. In the light of that fact, our interest has been aroused to check for other types of metabolites and for the protective ability of the shrub crude extract (SCE) regarding chemically induced edema and subacute toxicity (following a formalin-induced paw edema model and using a carbamate pesticide “pirimicarb” as a toxic agent, in Wistar male rats, respectively). Evidently, the SCE was used as a preventive agent. Swelling of formalin-injected foot was measured, and the anti-edematous ability was expressed as a percent of paw edema. At the end of induced subacute toxicity procedure, many investigations were carried, namely, checking for biochemical several parameters (hepatic, renal), hematological parameters, oxidative stress status (OSS), and histological examining of liver, spleen, and kidney tissues. The results revealed a remarkable anti-edematous effect. Furthermore, the association of the SCE effect has clearly minimized the OSS, the tissue aberrations, and the disturbance of the other metabolic parameters, along with a reduced immunodepression that was provoked under the effect of pirimicarb. In conclusion, pirimicarb has an important deleterious impact on metabolic and immune functions, and the studied shrub has exhibited an amazing protective potential against chemically induced inflammation and toxicity.

Antidiabetic and Antioxidant Activities of Plumbago zeylanica Roots in Streptozotocin-induced Diabetic Rats

Abstract:Abstract Objective: The concurrent usage of oral hypoglycemic agents produces side effects, and the herbal medicine Plumbago zeylanica L. (PZ) is being studied to reduce these effects. The antioxidant and antidiabetic effects of PZ roots on streptozotocin-nicotinamide-induced diabetic rats were investigated. Materials and Methods: Triple maceration method was used for the development of PZ extracts. Standardization of the plant extract and in vitro study was carried out by the physicochemical study, 1, 1-diphenyl-2-picryl-hydrazyl, and α-amylase methods. Animals were divided into seven groups, and the experiment was designed into two parts. Seven groups were taken normal control, diabetic control, hydroalcoholic extract of PZ at different doses (100–200 mg/kg), and metformin (MET, 25 mg/kg) for 21 days. The coadministration of a low dose of PZ (100 mg/kg) and MET (100 mg/kg) was administered orally. Blood glucose level (BGL) was measured continuously for 3 weeks of overnight-fasted animals. Biochemical estimation was assessed by total protein, catalase, nitric oxide, and reduced glutathione content. Histological features of the β-cells were observed through histopathological study. Results: Hydroalcoholic PZ (HAPZ) showed the highest antioxidant and antidiabetic activities, respectively. Oral administration of HAPZ extract (200 mg/ml) for 21 days diminished the BGL significantly from 298 ± 7.05 to 230 ± 5.69 mg/dL, whereas coadministration of HAPZ (100 mg/kg) with MET (10 mg/kg) had also reduced BGL from 286 ± 4.10 to 231 ± 5.75 mg/dL. The best outcomes were obtained when PZ extract (100 mg/kg) was combined with a low dose of MET (10 mg/kg). Conclusions: We can conclude that PZ might be the alternative to synthetic medicines for the management of diabetic disorders.

Abstract:Diabetes and its complications represent a real major public health problem in the world because of its high rates of morbidity and mortality. Chronic hyperglycemia, oxidative stress, dyslipidemia and inflammation play a major role in the pathophysiology of diabetes and its vascular complications. The objective of this study was to evaluate the effect of aqueous extract of S. zenkeri on chronic hyperglycemia and its complications in a streptozotocin-induced diabetic Wistar rat model. The barks of S. zenkeri were washed, dried and crushed; the powder was dissolved in distilled water (1:10 weight/volume) then macerated and the filtrate obtained was dried in an oven. Subsequently, after quantification of the bioactive compounds (total polyphenols, flavonoids and alkaloids) present in the extract, an in vivo study was conducted in an animal model of streptozotocin-induced hyperglycemia. For this fact, the rats were divided into four groups of five rats as folow: a normoglycaemia group (NC), an untreated hyperglycaemia group (PC), two hyperglycaemia groups including a test group receiving by esophageal gavage, the aqueous extract of the bark of S zenkerii (AESZ) at a dose of 300 mg/kg body weight and a control group receiving metformin at a dose of 20 mg/kg body weight. During the treatment which lasted 21 days, the weights have been taken every two days and the blood sugar levels every week. At the end of the treatment, the rats were killed under light chloroform anesthesia; the plasma, hemolysate, serum and liver homogenate prepared were used to assay the biochemical parameters of oxidative stress (catalase, MDA), lipid profile (Triglycerides, total cholesterol and HDL-cholesterol) and immunological (CRP and NFS). It emerged that the extract limited weight loss and caused a reduction in blood sugar of -26.59% after 21 days of treatment; the extract caused an increase in the activity of erythrocyte catalase and a reduction in the concentration of hepatic MDA, as well as a very marked reduction in inflammatory cells and CRP. The extract also caused a reduction in dyslipidemiawhich was materialized by a reduction in CRR, AC, AIP and an increase in CPI. These results suggest that this extract contains bioactive compounds capable of reducing chronic hyperglycemia while preventing its complications, thus justifying its traditional use in the management of diabetes.
Mitigation of Diabetes Mellitus Using Euphorbia helioscopia Leaf Ethanolic Extract by Modulating GCK, GLUT4, IGF, and G6P Expressions in Streptozotocin‐Induced Diabetic Rats

Abstract:Diabetes mellitus is a metabolic disorder. Synthetic antidiabetics are the commonly used treatment options associated with complications. The objective of this study was to explore the antioxidative and antidiabetic potential of Euphorbia helioscopia whole plant ethanolic extract using in vitro and in vivo models. For that purpose, the antioxidative potential was explored by using 2,2‐diphenyl‐1‐picrylhydrazyl analysis. In vitro antidiabetic potential of the extract was evaluated using amylase inhibitory analysis. In vivo antidiabetic activity of the extract was assessed in diabetic rats using streptozotocin/nicotinamide (60 mg/kg/120 mg/kg) as an inducing agent. Metformin was used as standard. The results indicated the presence of significant quantities of phenolic 82.18 ± 1.28 mgg −1 gallic acid equivalent (GAE) and flavonoid 66.55±1.22 mgg −1 quercetin equivalent (QE) contents in the extract. Quantitation of phytoconstituents exhibited the presence of sinapic acid, myricetin, and quercetin using HPLC analysis. The extract inhibited α‐amylase by 84.71%, and an antiglycemic potential of 50.34% was assessed in the OGTT assay. Biochemical analysis demonstrated a reduction in urea, creatinine, cholesterol, low‐density lipoprotein, and alkaline phosphatase (p < 0.001) as compared to diabetic control rats at the dose of 500 mg/kg. An upregulation in the expressions of glucokinase, glucose transporter 4, peroxisome proliferator‐activated receptor γ, and insulin‐like growth factor was observed in treated rats in contrast to G6P expression, which was downregulated upon treatment. In conclusion, this study provided evidence of the antioxidative and antidiabetic potential of E. helioscopia whole plant ethanolic extract through in vitro and in vivo analysis and emphasized its promising role as a natural alternative.

Abstract:A computer simulation of the steady-state operation of recessed (Whalen-type) polarographic oxygen electrodes has been developed to give the design factors important for performance optimization. The simulation makes use of a specially formulated three-dimensional orthogonal coordinate system with the geometry identical to the actual recessed cathode and gives the oxygen concentration field induced by it in the surrounding medium. Equations are presented which allow one to calculate, for any recessed cathode, the current sensitivity, maximum stirring artifact, measurement error, and time constant. Comparisons with analytically obtained expressions for the corresponding quantities for idealized, spherosymmetric cathodes demonstrate the unique aspects of recessed-cathode performance. For commonly used electrodes, a recess length-to-cathode diameter ratio of greater than 10 is found to give a negligible stirring artifact, a negligible measurement error, and a rapid response.

Abstract:Withania frutescens L. is a wild perennial woody plant used by the local population for diverse therapeutic purposes. This work aims to study for the first time the potential inhibitory effect of this plant hydroethanolic extract on α-amylase and α-glucosidase activities using in vitro methods and its antidiabetic and antihyperglycemic activities using alloxan-induced diabetic mice as a model for experimental diabetes. Two doses were selected for the in vivo study (200 and 400 mg/kg) and glibenclamide, a well-known antidiabetic drug (positive control) in a subacute study (28 days) where the antihyperglycemic activity was also assessed over a period of 12 h on diabetic mice. The continuous treatment of diabetic mice with the extract of Withania frutescens for 4 weeks succeeded to slowly manage their high fasting blood glucose levels (after two weeks), while the antihyperglycemic test result revealed that the extract of this plant did not control hyperglycemia in the short term. No toxicity signs or death were noted for the groups treated with the plant extract, and it shows a protective effect on the liver and kidney. The in vitro assays demonstrated that the inhibition of alpha-amylase and alpha-glucosidase might be one of the mechanisms of action exhibited by the extract of this plant to control and prevent postprandial hyperglycemia. This work indicates that W. frutescens have an important long term antidiabetic effect that can be well established to treat diabetes.

Abstract:This study was designed to investigate the hypoglycemic and hypolipidemic effects of plantago psyllium seeds added to diet in two different doses (0.5% and 1.0% ww) on strepotozotocin (STZ) induced diabetic rats and/or hyperlipidemic rats. Eighty male adult albino rats were used in this study, divided into 10 groups 8 rats each. G (1): served as healthy control, G (2) was diabetic control. G (3) was hyperlipidemic control. G (4) was diabetic fed on high fat high cholesterol diet (HFHC). Gs: (5-6) diabetic rats fed on diet supplemented with plantago psyllium seeds (0.5% &1% ww).Gs (7-8) hyperlipidemic rats fed on HFHC diet supplemented with plantago psyllium seeds (0.5% & 1% ww).Gs (9-10) diabetic rats fed on HFHC diet supplemented with psyllium seeds (0.5% & 1% ww). After 6 weeks, rats were sacrificed under diethyl ether anesthesia. Serum and tissue samples were collected to perform the biochemical analysis. Serum glucose, insulin, insulin resistance (HOMA-IR) and beta cell function index (HOMA-β) were measured. Moreover, Serum lipids profile including total lipid (TL), phospholipids (PL),total cholesterol (TC),triacylglycerols (TAGs),low density lipoprotein cholesterol (LDL-C),very low density lipoprotein cholesterol (VLDL-C), as well as LDL-C/ HDL-C, TC/HDL-C, TAGs/HDL-C ratios and atherogenic index (AI) were measured. Also, antioxidant status was evaluated by measuring total antioxidants capacity (TAC) level, glutathione peroxidase (GPx), glucose 6 phosphate dehydrogenase (G6PD) activities, malondialdehyde (MDA) and oxidative stress index (OSI). The results were supported with microscopic examination of pancreatic tissues. The results of this study revealed that administration of plantago psyllium seeds improve glucose homeostasis by decreasing blood glucose, increasing β-cell function, increasing serum insulin level and insulin sensitivity in STZ-induced diabetic rats. Also, plantago psyllium seeds ameliorate insulin resistance in hyperlipidemic groups by decreasing insulin level. The result of this study approved that psyllium seeds improve dyslipidemia relevant to HFHC diet and diabetes mainly through decreasing TL, PL, TC, TAGs, LDL-C, VLDL-C and increasing HDL-C. Furthermore, plantago psyllium seeds showed an important cardio protective effect through decreasing AI , TC/HDL-C,TAGs/HDL-C and LDL-C/ HDL-C ratios as a risk for coronary heart disease (CHD).The study revealed that plantago psyllium seeds has a significant antioxidant activity and can be used to protect tissue from oxidative stress as indicated in microscopic examination of pancreatic tissue showed amelioration in the appearance of cells .The result showed a significant decrease in serum MDA and OSI and a significant increase in TAC level ,GPx and G6PDactivities .The protective effect of plantago psyllium seeds is due to their antioxidant potential which represented by their rich content of dietary fibers, flavonoids, poly unsaturated fatty acids, and some polyphenols that appears to protect against reactive oxygen species (ROS).

Abstract:Mimusops zeyheri Sond. And Aloe marlothii A.Berger are used traditionally in South Africa to manage many diseases, including diabetes mellitus. The mechanism through which these extracts exert blood glucose lowering is not well understood or reported. This study was aimed at assessing M. zeyheri and A. marlothii plant extracts for their potential to exhibit antidiabetic activity and their associated mechanisms. We evaluated the action of both extracts on major genes involved in the insulin signalling pathways in skeletal muscle cells. The in vitro cytotoxic effects of M. zeyheri and A. marlothii extracts were evaluated using the MTT assay and glucose uptake was evaluated using a glucose oxidase assay. The amount of translocated GLUT-4 was determined using the flow cytometry. Conventional PCR was used to determine the expression of GLUT-1 and GLUT-4 and RT-qPCR. IRS-1 total protein and Phospho-Akt were determined using ELISA. Plant extracts stimulated glucose absorption by skeletal muscle cells. M. zeyheri extract increased glucose absorption in muscle cells after 1 and 3 h of incubation. A 2-fold increase in translocated GLUT-4 was noted with M. zeyheri. The mRNA levels of GLUT-4 and GLUT-1 remained uniform in all treatments, while IRS-1, PI3K, Akt1, Akt2, and PPAR-γ were downregulated by both extracts. The expression of GLUT-4 was significantly increased by the action of insulin and M. zeyheri extract at 500 μg/mL. This study validates the traditional use of aqueous extracts of A. marlothii and M. zeyheri as hypoglycaemic plants and raises the assertion that the selected plant extracts utilise the IRS-1/PI3K/Akt pathway.

Abstract:BACKGROUND: Carbohydrate digestive enzymes play a major role in the management of the postprandial hyperglycemia. A chronic hyperglycemia can lead to serious health problems due to excessive production of several reactive oxygen species. Therefore, the inhibition of carbohydrate digestive enzyme and the use of antioxidant natural product can be an important strategy to control the glycaemia level and prevent against the complication of diabetes.AIM: The study aims to perform a phytochemical analysis, antioxidant activity, inhibitory effect on α -amylase, α -glucosidase (in vitro and in vivo) and the intestinal glucose absorption in Wistar rats of Artemisia campestris aqueous extract (AcAE) and hydro-ethanolic extract (AcEE).RESULTS: The test of total phenolic content, show that the AcAE has the highest quantity of polyphenol (44.65 ± 0.54 μ g GAE/mg extract) compared to the AcEE (31.7 ± 0.53 μ g GAE/mg extract) significantly. The amount of flavonoid and condensed tannins content in AcAE is 24.41 ± 3.57 μ g QrE/mg extract, 14.31 ± 5.26 μ g CE/mg respectively. The AcAE has also exhibit a great antioxidant activity in DPPH-scavenging and Ferric reducing antioxidant power assay (FRAP) compared to AcEE with an IC 50 = 0.355 ± 0.057 mg/mL and IC 50 = 0.269 ± 0.025 mg/mL. However, in a β -carotene bleaching assay the AcEE has the highest effect with an IC 50 = 0.319 ± 0.097 mg/mL. The both extract of Artemisia campestris L. (250 mg/kg) decreased postprandial hyperglycemia in the normal and alloxane diabetic rats in a very significant manner after starch or sucrose administration as an α -amylase and α -glucosidase substrate respectively. This result is confirmed in vitro by a remarkable inhibitory effect on α -amylase digestive enzymes by an IC 50 = 1.259 ± 0.128 mg/mL and IC 50 = 0.602 ± 0.072 mg/mL receptively for AcAE and AcEE. For the α -glucosidase enzyme, the both extracts significantly inhibit α -glucosidase activity compared to the control and they are almost similar to each other. Using a jejunum perfusion technique (in situ), Artemisia campestris L. decrease the intestinal D-glucose absorption activity significantly compared to the control and comparable to the Phlorizin used as a positive control by an amount of glucose absorbed equal a 6.53 ± 0.57, 5.34 ± 0.64 and 4.71 ± 0.24 mg/10 cm/h, for AcAE, AcEE and Phlorizin respectively.CONCLUSIONS: These results showed that the Artemisia campestris L. has highest phenolic content, antioxidant activity and demonstrated a postprandial anti-hyperglycemic effect via the inhibiting of the carbohydrate digestive enzyme ( α -amylase and α -glucosidase) and the intestinal glucose absorption.
