Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009

Abstract:Diseases, trauma, and injuries are highly prevalent conditions that lead to many critical tissue defects. Tissue engineering has great potentials to develop functional scaffolds that mimic natural tissue structures to improve or replace biological functions. In many kinds of technologies, electrospinning has received widespread attention for its outstanding functions, which is capable of producing nanofibre structures similar to the natural extracellular matrix. Amongst the available biopolymers for electrospinning, poly (caprolactone) (PCL) has shown favorable outcomes for tissue regeneration applications. According to the characteristics of different tissues, PCL can be modified by altering the functional groups or combining with other materials, such as synthetic polymers, natural polymers, and metal materials, to improve its physicochemical, mechanical, and biological properties, making the electrospun scaffolds meet the requirements of different tissue engineering and regenerative medicine. Moreover, efforts have been made to modify nanofibres with several bioactive substances to provide cells with the necessary chemical cues and a more in vivo like environment. In this review, some recent developments in both the design and utility of electrospun PCL-based scaffolds in the fields of bone, cartilage, skin, tendon, ligament, and nerve are highlighted, along with their potential impact on future research and clinical applications.
Electrospun Composite Poly(L-Lactic Acid)/Tricalcium Phosphate Scaffolds Induce Proliferation and Osteogenic Differentiation of Human Adipose-Derived Stem Cells

Abstract:Development of tissue-engineered bone constructs has recently focused on the use of electrospun composite scaffolds seeded with stem cells from various source tissues. In this study, we fabricated electrospun composite scaffolds consisting of beta-tricalcium phosphate (TCP) crystals and poly(L-lactic acid) (PLA) at varying loading levels of TCP (0, 5, 10, 20 wt%) and assessed the composite scaffolds' material properties and ability to induce proliferation and osteogenic differentiation of human adipose-derived stem cells (hASCs) in the presence of osteogenic differentiating medium. The electrospun scaffolds all exhibited a nonwoven structure with an interconnected porous network. With the addition of TCP, the fiber diameter increased with each treatment ranging from 503.39 +/- 20.31 nm for 0 wt% TCP to 1267.36 +/- 59.03 nm for 20 wt% TCP. Tensile properties of the composite scaffolds were assessed and the overall tensile strength of the neat scaffold (0 wt% TCP) was 847 +/- 89.43 kPA; the addition of TCP significantly decreased this value to an average of 350.83 +/- 38.57 kPa. As the electrospun composite scaffolds degraded in vitro, TCP was released into the medium with the largest release occurring within the first 6 days. Human ASCs were able to adhere, proliferate and osteogenically differentiate on all scaffold combinations. DNA content increased in a temporal manner for each scaffold over 18 days in culture although for the day 12 timepoint, the 10 wt% TCP scaffold induced the greatest hASC proliferation. Endogenous alkaline phosphatase activity was enhanced on the composite PLA/TCP scaffolds compared to the PLA control particularly by day 18. It was noted that at the highest TCP loading levels of 10 and 20 wt%, there was a dramatic increase in the amount of cell-mediated mineralization compared to the 5 wt% TCP and the neat PLA scaffold. This work suggests that local environment cues provided by the biochemical nature of the scaffold can accelerate the overall osteogenic differentiation of hASCs and encourage rapid ossification.

Abstract:Electrospinning technology is widely used for preparing biological tissue engineering scaffolds because of its advantages of simple preparation, accurate process parameters, and easy control. Poly(L-lactide) (PLLA) is regarded as a promising biomass-based polymer for use in electrospinning. The incorporation of Fe3O4 nanoparticles (NPs) could improve the osteogenic differentiation and proliferation of cells in the presence or absence of a static magnetic field (SMF). In this work, these two materials were blended together to obtain electrospun samples with better dispersibility and improved magnetic properties. First, composite PLLA and Fe3O4 NP fibers were prepared by means of electrospinning. The influence of electrospinning conditions on the morphology of the composite fibers was then discussed. Changes in magnetic properties and thermal stability resulting from the use of different PLLA/Fe3O4 mass ratios were also considered. Next, the morphology, crystal state, thermodynamic properties, and magnetic properties of the electrospun samples were determined using scanning electron microscopy (SEM), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and vibration sample magnetization (VSM). The results showed that the fibers prepared using PLLA with Mn = 170,000 exhibited good morphology when electrospun at 12 KV. The magnetic properties of PLLA/Fe3O4 composite electrospun fibers increased with the NP content, with the exception of thermal stability. The results of the present study may help to promote the further development of PLLA/Fe3O4 composite materials in the biomedical field.

Abstract:β-Tricalcium phosphate(β-TCP) which synthesize by wet method and poly(L-lactic acid)(PLLA)hybrid membrane was fabricated via electrospinning of the PLLA/β-TCP dispersion for the applications in bone tissue regeneration.The structural properties and morphologies of β-TCP nanoparticles were investigated by FT-IR,XRD,TEM and XPS.The results showed that the diameters of β-TCP particles fall in the range from 219 to 328 nm and with good crystal structure.In order to optimize the technics of electrospinning,the effects of process parameters on the fiber morphology are studied by SEM and diameter distributing.PLLA/β-TCP nanofiber with the diameter from 500 to 700 nm can be produced via electrospinning under the optimium condition.The (mechanical) test result showed that PLLA/β-TCP membrane has a strong surface bonding and high(tensile) strength,the mechanical property of PLLA nonwoven has increased,but the mechanical(property) of PLLA/β-TCP nonwoven shows a downward trend,under wet conditions.

Abstract:Abstract Conductive and degradable nanofibrous scaffolds have great potential in promoting cell growth, proliferation, and differentiation under an external electric field. Although the issue of inferior electrical conductivity in body fluids still exists, polyaniline (PANI)-based degradable nanofibers can promote cell adhesion, growth, and proliferation. To investigate whether the effect is caused by the PANI morphology, we selected three inorganic acids as dopants in the process of PANI in situ oxidative polymerization: hydrochloric acid, sulfuric acid, and perchloric acid. The obtained polyaniline/polylactic acid (PANI/PLA) composite nanofibers were characterized via SEM, FTIR, and XPS analysis, and we confirmed that the PLA nanofibers were successfully coated by PANI without any change to the porous structure of the PLA nanofibers. The in vitro mechanical properties and degradability indicated that the oxidation of acid dopants should be considered and that it was likely to have a higher oxidation degradation effect on PLA nanofibers. The contact angle test demonstrated that PANI/PLA composite nanofibers with different surface morphologies have good wettability, implying that they meet the requirements of bone tissue engineering scaffolds. The surface roughness and cell viability demonstrated that different PANI morphologies on the surface can promote cell proliferation. The higher the surface roughness of the PANI, the better the biocompatibility. Consequently, the regulated surface morphology of PANI/PLA composite nanofibers via different acids doping has positive effect on biocompatibility in tissue engineering.

Abstract:Electrospinning has recently emerged as a potential technique for fabricating biomimetic tissue engineering scaffolds. In this study, Poly (1-lactic acid) (PLLA) /Nano-hydroxyapatite (HA) hybrid nanofibers scaffolds were prepared by electrospinning. The relationship between process parameters and fiber diameter has been investigated. The fiber diameter decreased with decreasing polymer concentration and with increasing electrospinning voltage; After 6 weeks of in vitro degradation, the mass, viscosity-average molecular weight of the nanofibers scaffolds and the pH value of the degradation solution were changed, the fibers lost their surface smoothness and a regular rough topology was generated after 32d of degradation, the degradation rates of PLLA/HA hybrid nanofibers were slower than those of pure PLLA fibers; The biocompatibility of the nanofibers scaffold has also been investigated by culturing cells on the nanofibers scaffold, elementary results showed that the cells adhered and proliferated well on the PLLA/HA hybrid nanofibers scaffolds.

Abstract:Polyaniline (PANI) has indeed received significant attention and extensive research in both academic and electronic industrial fields. Its unique properties, such as conductivity and processability, make it a promising material for various applications. Researchers and industrialists have explored PANI for its potential use in electronic devices, such as sensors, batteries, capacitors, and actuators, as well as in other areas like corrosion protection, supercapacitors, and electromagnetic shielding. The poor compatibility, tendency to aggregate, and poor mechanical properties of nanostructured PANI have hindered its performance. Cellulose nanocrystals (CNCs) were used as a bio-template for covalent grafting PANI onto hydrophilic CNCs (CNC-g-PANI) through in situ oxidative polymerization. The resulting CNC-g-PANI suspension with the sodium alginate (SA) matrix was wet-spun into composite fibers, which were compared to composite fibers made by physical blending of pure PANI and hydrogen-bonded CNC/PANI. The parameters were characterized to investigate the performance of the composite fibers. The covalent grafted CNC-g-PANI suspension maintained stable dispersion throughout the wet spinning process, making it suitable for both fundamental research and industrial processing. Indeed, the development of CNC-g-PANI@SA composite fibers through the covalent grafting of PANI onto cellulose nanocrystals offers a simple and eco-friendly approach. These composite fibers exhibit improved properties, including enhanced tensile strength, electrical conductivity, flexibility, and fatigue resistance. These attributes make them highly suitable for applications in the antistatic textile and electronic industries.

Abstract:In this study, microfiber films were used as scaffolds for the purpose of vascular tissue engineering. The microfiber films were prepared by electrospinning of poly (l-lactide) (PLLA) and polyvinyl pyrrolidone (PVP). PLLA and PVP with different ratios were blended with dichloromethane as a spinning solvent at room temperature. The properties of the composite microfiber films were investigated by differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and contact angle measurement. The SEM images showed that the morphology of the microfiber films was mainly affected by the weight ratios of PLLA/PVP. The DSC results demonstrated that PLLA and PVP mixed uniformly. And the hydrophilicity of the films measured increased along with the decrease of the PLLA/PVP ratio. Vascular smooth muscle cells (VSMCs) were used to test the cytocompatibility. Cell morphology and cell proliferation were measured by SEM, laser scanning confocal microscopy (LSCM) and MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay after 2, 4, 6 days of culture. The results indicated that the cell morphology and proliferation on the composite films were better than that on the pure PLLA film. Furthermore, morphology and proliferation of VSMCs became better with decreasing of the weight ratio of PLLA/PVP. In addition, adhesion of platelet on the films was observed by SEM. The SEM images showed that the number of adhered platelets decreased with increment of PVP content in the films. The electrospinning microfiber composite films of PLLA and PVP would have potential use as the scaffolds for vascular tissue engineering.
Influences of Process Parameters of Near-Field Direct-Writing Melt Electrospinning on Performances of Polycaprolactone/Nano-Hydroxyapatite Scaffolds

Abstract:In this paper, near-field direct-writing melt electrospinning technology was employed to fabricate a polycaprolactone/nano-hydroxyapatite (PCL/nHA) scaffold for future applications in tissue engineering. The influences of different fabrication parameters on the structural characteristics, mechanical properties, and thermal stability of the scaffolds were discussed. It was found that the moving speed of the receiving plate had the most significant effect on the scaffold performance, followed by the receiving distance and spinning voltage. The results also showed that these process parameters affected the fiber diameter, corresponding coefficient of variation, porosity of the composite scaffolds, and mechanical properties of the samples, including the tensile strength and fiber peeling strength. Moreover, the process parameters could influence the thermal degradation performance and melting process. Although the mass loss of the composite scaffolds was not obvious after degradation, the mechanical performance degraded severely. It was concluded that the more appropriate process parameters for preparing PCL/nHA scaffolds were a spinning voltage of −4 kV, receiving distance of 4 mm, moving speed of receiving plate of 5 mm/s, and melt temperature of 130 °C. This study proved that near-field direct-writing melt electrospinning technology is a good method to obtain PCL/nHA composite scaffolds with an excellent mechanical properties and desired morphology for future tissue engineering applications.

Electrospun Magnetic Poly(l-Lactide) (PLLA) Nanofibers by Incorporating PLLA-stabilized Fe3O4 Nanoparticles.

Abstract:Magnetic poly(L-lactide) (PLLA)/Fe3O4 composite nanofibers were prepared with the purpose to develop a substrate for bone regeneration. To increase the dispersibility of Fe3O4 nanoparticles (NPs) in the PLLA matrix, a modified chemical co-precipitation method was applied to synthesize Fe3O4 NPs in the presence of PLLA. Trifluoroethanol (TFE) was used as the co-solvent for all the reagents, including Fe(II) and Fe(III) salts, sodium hydroxide, and PLLA. The co-precipitated Fe3O4 NPs were surface-coated with PLLA and demonstrated good dispersibility in a PLLA/TFE solution. The composite nanofiber electrospun from the solution displayed a homogeneous distribution of Fe3O4 NPs along the fibers using various contents of Fe3O4 NPs. X-ray diffractometer (XRD) and vibration sample magnetization (VSM) analysis confirmed that the co-precipitation process had minor adverse effects on the crystal structure and saturation magnetization (Ms) of Fe3O4 NPs. The resulting PLLA/Fe3O4 composite nanofibers showed paramagnetic properties with Ms directly related to the Fe3O4 NP concentration. The cytotoxicity of the magnetic composite nanofibers was determined using in vitro culture of osteoblasts (MC3T3-E1) in extracts and co-culture on nanofibrous matrixes. The PLLA/Fe3O4 composite nanofibers did not show significant cytotoxicity in comparison with pure PLLA nanofibers. On the contrary, they demonstrated enhanced effects on cell attachment and proliferation with Fe3O4 NP incorporation. The results suggested that this modified chemical co-precipitation method might be a universal way to produce magnetic biodegradable polyester substrates containing well-dispersed Fe3O4 NPs. This new strategy opens an opportunity to fabricate various kinds of magnetic polymeric substrates for bone tissue regeneration.
Polyester/Polylactic Acid/Stainless Steel Composite Bone Scaffolds Made by Electrochemical Treatment: Process Design and Property Evaluations

Abstract:This study combines and twists 75D polyester (PET) multi-filaments and polylactic acid (PLA) multi-filaments with twist coefficients of 2, 3, 4, 5, and 6 to form 150D PET/PLA plied yarns. The 0.08-mm-diameter stainless steel (SS) fibers are made into SS braids with a 60-tooth braid gear and a take-up gear with 60, 70, 80, 90, or 100 teeth. PET/PLA plied yarn and SS braids are then combined and electrochemically treated with an electric current of 100, 200, 300, 400, or 500 mA at 60 °C for 24 hours, forming the PET/PLA/SS composite bone scaffolds. PET/PLA/SS composite bone scaffolds are observed by scanning electron microscope (SEM) and energy dispersive spectroscope (EDS), and tested for weight increase rate and biocompatibility. The experiment results show that the optimal twist coefficient for PET/PLA plied yarn is 4 and the optimal tooth number on the take-up gear for SS braids is 80. SEM observation result shows that hydroxyapatite (HA) deposits on the surface of PET/PLA/SS composite bone scaffolds and attaches to the PET/PLA plied yarns. Finally, regardless of electric currents, all PET/PLA/SS composite bone scaffolds possess good biocompatibility.

Innovative electrospun PCL/fibroin/l-dopa scaffolds scaffolds supporting bone tissue regeneration

Abstract:Poly-caprolactone is one of the most promising biocompatible polymers on the market, in particular for temporary devices that are not subjected to high physiological loads. Even if completely resorbable in various biological environments, poly-caprolactione does not play any specific biological role in supporting tissue regeneration and for this reason has a limited range of possible applications. In this preliminary work, for the first time l-dopa and fibroin have been combined with electrospun poly-caprolactone fibers in order to induce bioactive effects and, in particular, stimulate the proliferation, adhesion and osteoconduction of the polymeric fibers. Results showed that addition of low-molecular weight fibroin reduces the mechanical strength of the fibers while promoting the formation of mineralized deposits, when testedin vitrowith KUSA-A1 mesenchymal cells. l-dopa, on the other hand, improved the mechanical properties and stimulated the formation of agglomerates of mineralized deposits containing calcium and phosphorous with high specific volume. The combination of the two substances resulted in good mechanical properties and higher amounts of mineralized deposits formedin vitro.

Preparation of Polypyrrole-Embedded Electrospun Poly(lactic Acid) Nanofibrous Scaffolds for Nerve Tissue Engineering.

Abstract:Polypyrrole (PPy) is a biocompatible polymer with good conductivity. Studies combining PPy with electrospinning have been reported; however, the associated decrease in PPy conductivity has not yet been resolved. We embedded PPy into poly(lactic acid) (PLA) nanofibers via electrospinning and fabricated a PLA/PPy nanofibrous scaffold containing 15% PPy with sustained conductivity and aligned topography. There was good biocompatibility between the scaffold and human umbilical cord mesenchymal stem cells as well as Schwann cells. Additionally, the direction of cell elongation on the scaffold was parallel to the direction of fibers. Our findings suggest that the aligned PLA/PPy nanofibrous scaffold is a promising biomaterial for peripheral nerve regeneration.

Abstract:BACKGROUND/AIMS Carbon dioxide (CO2) insufflation has been used in endoscopic retrograde cholangiopancreatography (ERCP). However, its usefulness and safety are controversial in elderly patients. Our aim was to assess the safety and usefulness of CO2 insufflation during ERCP in elderly patients. METHODOLOGY Between April 2010 and June 2011, a total of 60 patients 75 years old or older, who underwent ERCP, were randomized into the CO2 group (n = 30) and the air group (n = 30). Main outcomes were determined by assessing abdominal symptoms according to the Wong-Baker FACES Pain Rating Scale, calculating the volume of residual gas retention within the intestines on abdominal X-ray quantitatively and observing the cardiopulmonary states. RESULTS 30 patients in the CO2 group and 30 patients in the air group were analyzed. Abdominal distension (P < 0.01), discomfort (P < 0.01) and nausea (P < 0.01) at 2 hours after ERCP were significantly reduced in the CO2 group. The gas volume scores immediately after ERCP (P < 0.01) and at 2 hours (P < 0.01) were significantly lower in the CO2 group. CONCLUSIONS CO2 insufflation instead of air insufflation is safe and useful for the prevention of post-ERCP abdominal symptoms in elderly patients.
Electrospun fibrous scaffold of hydroxyapatite/poly (ε-caprolactone) for bone regeneration

Abstract:Development of fibrous scaffold of hydroxyapatite/biopolymer nanocomposite offers great potential in the field of bone regeneration and tissue engineering. Hydroxyapatite (HA)/poly (ε-caprolactone) (PCL) fibrous scaffolds were successfully prepared by electrospinning dopes containing HA and PCL in this work. It was found that pre-treating HA with γ-glycioxypropyltrimethoxysilane (A-187) was effective in improving HA dispersion both in solutions and in a PCL matrix. Mechanical properties of the scaffolds were greatly enhanced by the filling of A187-HA. The bioactivity of PCL was remarkably improved by the addition of HA and A187-HA. Fibroblasts and osteoblasts were seeded on scaffolds to evaluate the effect of A-187 on biocompatibility of HA/PCL composites. Based on this study, good dispersion of HA in PCL matrix was granted by pretreatment of HA with A-187 and A187-HA/PCL fibrous scaffolds were obtained by electrospinning. These results demonstrated that the scaffolds may possess improved mechanical performance and good bioactivity due to A187-HA incorporation.
Biomineralized poly (l-lactic-co-glycolic acid)-tussah silk fibroin nanofiber fabric with hierarchical architecture as a scaffold for bone tissue engineering

Abstract:In bone tissue engineering, the fabrication of a scaffold with a hierarchical architecture, excellent mechanical properties, and good biocompatibility remains a challenge. Here, a solution of polylactic acid (PLA) and Tussah silk fibroin (TSF) was electrospun into nanofiber yarns and woven into multilayer fabrics. Then, composite scaffolds were obtained by mineralization in simulated body fluid (SBF) using the multilayer fabrics as a template. The structure and related properties of the composite scaffolds were characterized using different techniques. PLA/TSF (mass ratio, 9:1) nanofiber yarns with uniform diameters of 72±9μm were obtained by conjugated electrospinning; the presence of 10wt% TSF accelerated the nucleation and growth of hydroxyapatite on the surface of the composite scaffolds in SBF. Furthermore, the compressive mechanical properties of the PLA/TSF multilayer nanofiber fabrics were improved after mineralization; the compressive modulus and stress of the mineralized composite scaffolds were 32.8 and 3.0 times higher than that of the composite scaffolds without mineralization, respectively. Interestingly, these values were higher than those of scaffolds containing random nanofibers. Biological assay results showed that the mineralization and multilayer fabric structure of the composite nanofiber scaffolds significantly increased cell adhesion and proliferation and enhanced the mesenchymal stem cell differentiation toward osteoblasts. Our results indicated that the mineralized nanofiber scaffolds with multilayer fabrics possessed excellent cytocompatibility and good osteogenic activity, making them versatile biocompatible scaffolds for bone tissue engineering.

Abstract:The solution electrospinning process (SEP) is a cost-effective technique in which a wide range of polymeric materials can be electrospun. Electrospun materials can also be easily modified during the solution preparation process (prior SEP). Based on this, the aim of the current work is the fabrication and nanomodification of scaffolds using SEP, and the investigation of their porosity and physical and mechanical properties. In this study, polylactic acid (PLA) was selected for scaffold fabrication, and further modified with multi-walled carbon nanotubes (MWCNTs) and hydroxyapatite (HAP) nanoparticles. After fabrication, porosity calculation and physical and mechanical characterization for all scaffold types were conducted. More precisely, the morphology of the fibers (in terms of fiber diameter), the surface properties (in terms of contact angle) and the mechanical properties under the tensile mode of the fabricated scaffolds have been investigated and further compared against pristine PLA scaffolds (without nanofillers). Finally, the scaffold with the optimal properties was proposed as the candidate material for potential future cell culturing.
Fabrication of a PLA/PVA-BIO-HA Polymeric Membrane by the Electrospinning Technique

Abstract:In the present work, the fabrication of a membrane composed of polylactic acid (PLA), polyvinyl alcohol (PVA), and Biological Hydroxyapatite (BIO-HA) is reported using the coaxial electrospinning technique. The membrane fabrication process involved mixing a solution of PLA and trichloromethane (TCM) with a second solution of PVA, isopropyl alcohol (IPA), distilled water, and BIO-HA at 110 °C. Subsequently, the electrospinning process was carried out using a voltage of 25 kV for 30 min on a rotating drum collector at 1000 rpm. The membrane was characterized through Scanning Electron Microscopy (SEM), Energy-Dispersive X-ray spectroscopy (EDS), Fourier-Transform Infrared spectroscopy (FTIR), Thermogravimetric Analysis (TGA), and Differential Scanning Calorimetry (DSC). The morphological results revealed the presence of randomly arranged fibers with an average diameter of 290 ± 9 nm and interfiber spacing ranging from 200 to 700 nm, which are characteristics conducive to cell proliferation. Additionally, FTIR studies confirmed the presence of BIO-HA and the constituent elements of the polymers in the composite membrane. The polymeric membrane in contact with human mesenchymal stem cells was characterized as showing significant differences in its behavior at 6, 24, and 72 h post-contact. These studies indicate that the membrane provides physical support as a scaffold due to its suitable morphology for cell adhesion and proliferation, attributable to the electrospinning conditions as well as the polymers contained in BIO-HA. Membrane toxicity was confirmed through a cytotoxicity study using fluorescence microscopy, which showed that the membrane provided a favorable environment for cell proliferation. These results suggest that exposure to BIO-HA enhances its potential application in bone and joint tissue regeneration.
