Abstract:Pueraria lobata (Willd.) Ohwi (Gegen) is a nutritious medicine food homology plant that has been recorded in many ancient remedies for diabetes. However, its hypoglycemic active components and related mechanisms have not been fully elucidated. In this study, potential alpha-glucosidase inhibitors in Pueraria lobata were investigated by liquid-liquid refined extraction, high-resolution profiling and their hypoglycemic activity were evaluated. Ten compounds showing alpha-glucosidase inhibitory bioactivity were separated and identified, including two new compounds, 8-geranyl-7, 3', 4' -trihydroxy-isoflavone (4) and 8-geranyl-7, 2', 4'-trihydroxy-isoflavone (6), and eight known compounds. All screened compounds showed potential hypoglycemic activity, in which compounds 4, 6, 7, 8, 9 and 10 showed strong inhibitory activity (IC50 < 8 mu M). Furthermore, molecular docking simulation revealed binding energy and interacting amino acid residues of the screened compounds with alpha-glucosidase, strongly supporting their potential as alpha-glucosidase inhibitors.

Abstract:The conditions for extracting polysaccharides from tea (Camellia sinensis L.) fruit peel (TFPPs) were studied. Three parameters (temperature, time, and liquid/solid ratio) affecting the extraction of TFPP were optimized using response surface methodology (RSM). Under the optimized conditions, the yield of TFPP was predicted to be 4.98%. The physicochemical properties, in vitro antioxidant activities, and inhibitory effects on α-glucosidase of fractionated TFPPs (TFPP-0, TFPP-20, TFPP-40, and TFPP-60) were investigated. We found that the TFPPs were all acid protein-bound heteropolysaccharides, although with different chemical compositions. They had not only remarkable scavenging activity on 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and reducing activity, but also excellent inhibitory potential against α-glucosidase in vitro. Our results suggest that tea fruit peel could be treated as a potential bioresource for the development of polysaccharide antioxidants.

Abstract:Green tea, green tea polyphenols and epigallocatechin gallate (EGCG) are confirmed to have beneficial effects in the treatment of diabetes mellitus, and a possible mechanism can be ascribed to their inhibitory effect against α-amylase and α-glucosidase in the digestive tract. In this paper, we first investigated the combined inhibitory effect of green tea extracts, green tea polyphenols or EGCG with acarbose on α-amylase and α-glucosidase in vitro. Our results indicated that the interaction between green tea extracts (green tea polyphenols or EGCG) and acarbose was complicated. The combination of green tea extracts, green tea polyphenols or EGCG with acarbose had a synergistic effect on α-amylase and α-glucosidase at low concentrations and the combined effect turned out to be antagonistic at high concentrations according to the Combination Index (CI) values. These findings not only provided some significant quantitative values, but also provide some valuable implications for the combined use of acarbose and GTE (GTP or EGCG) in the treatment of diabetes mellitus.

Abstract:Two water-soluble polysaccharide fractions, coded as TFP-1 and TFP-2, were isolated from tea (Camellia sinensis) flower and further characterized using Ubbelohde viscometer, laser light scattering, particle size analyzer and thermo-gravimetric analyzer. Meanwhile, the intrinsic viscosities, weight-average molar mass, gyration radius and hydrodynamic radius for TFP-1 and TFP-2 were shown as follows: [eta] = 0.767 and 0.207 dL/g, M-w = 15.9 x 10(4) and 1.12 x 10(4) g/mol, R-g = 37.2 and 14.1 nm, R-h = 44.8 and 14.4 nm. Moreover, the shape-factor suggested the molecule morphology adopted a branched spherical shape in NaCl solution and the typical thermal behaviors of TFP-1 and TFP-2 were presented in analysis curves. Furthermore, research also presented the inhibitory effects against alpha-glucosidase and alpha-amylase. Moreover. continuous administration of TFP-2 with 75, 150 and 300 mg/kg/b.wt for 3 weeks caused a significant (p < 0.01) decrease in blood glucose levels of alloxan-induced diabetic mice. Abstract:Tea fruit peel is the main byproduct during manufacture of tea seed oil. The great increase in tea seed oil production in recent years brings the challenge of finding application for tea fruit peel. The aims of this study were to obtain tea fruit peel extracts enriched with bioactive compounds by several solvent extraction methods and to evaluate their antioxidant activity and inhibitory potential against α-glucosidase and α-amylase in vitro. Flavonoids and phenolics were accumulated in ethyl acetate, butanol, and chloroform fractions, and these fractions possessed much better antioxidant activities, including scavenging effect on 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH) and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) cation radicals (ABTS+), and reducing activity, compared with those of the 75% ethanol extract and water fraction. Moreover, the ethyl acetate, butanol, and chloroform fractions exhibited excellent inhibitory activity against α-glucosidase, much stronger than that of the positive control (acarbose). These fractions also showed mild inhibition on α-amylase activity.

Abstract:CD27 is a member of the tumor necrosis factor receptor superfamily that is expressed primarily on T cells, as well as on subsets of B cells and NK cells. CD70, which is expressed on activated B and T cells, but not on resting lymphocytes, is a ligand for CD27. Cell surface CD27 can be proteolytically cleaved to produce a 32-kDa soluble CD27 (sCD27) molecule. Elevated levels of sCD27 are seen in a number of disease states and malignancies. Although it has been reported that cerebrospinal fluid sCD27 levels were elevated in people who had AIDS dementia, little is known about CD27 expression in HIV disease. To determine if sCD27 levels were elevated in those with HIV infection, and/or in those with AIDS-associated non-Hodgkin's lymphoma (AIDS-NHL), sCD27 levels were measured in HIV-negative and HIV-positive subjects as well as in people who developed AIDS-NHL. Serum sCD27 levels were seen to be elevated in HIV+ subjects. Furthermore, sCD27 levels were particularly elevated in those subjects who went on to develop AIDS-NHL, with serum sCD27 levels in AIDS-NHL subjects being significantly higher than those in HIV+ subjects who did not develop lymphoma. Most AIDS-NHL cell lines and primary AIDS-NHL tumor specimens expressed both CD27 and its ligand, CD70. The proportion of circulating B cells that expressed cell surface CD27 was substantially reduced in those with HIV infection, and B cells from HIV-infected subjects produced decreased levels of sCD27 in culture. Together, these results indicate that CD27/sCD27 expression is abnormal in HIV infection and suggest that this molecule merits further examination as a potential marker for AIDS-NHL.

Abstract:Tea is consumed worldwide and can reduce the risk of diabetes whereas the hypoglycemic compounds in tea remain unclear. Herein, the chemical compositions comparison and inhibition assays indicated that the inhibitory effect of tea against α-amylase was positively related with the enzyme-mediated oxidation degree of tea and black teas containing high contents of theaflavins exhibited the strongest in vitro inhibitory effect. Affinity selection-mass spectrometry-based high-throughput screening revealed that four theaflavins specifically bind to α-amylase. Compared to other compounds, theaflavins showed one order of magnitude higher in vitro inhibitory effects; thus, theaflavins are possibly the dominant α-amylase inhibitors in tea. A series of “α-amylase–theaflavins” interaction studies jointly demonstrated that theaflavins noncompetitively inhibit α-amylase activity by interacting with the amino acid residues of α-amylase and then inducing changes in the secondary structure. Furthermore, theaflavins (10 mg/kg/d) and black tea extracts (100 mg/kg/d) exhibited comparable hypoglycemic effects as acarbose in diabetic mice.

Physicochemical characterization of puerh tea polysaccharides and their antioxidant and α-glycosidase inhibition

Abstract:Puerh tea is one of the most popular (or healthiest) beverages in Southeast Asia, because of its higher antioxidant, hypolipidemic and antidiabetic effects. One of the important active ingredients, tea polysaccharides (TPS) has long been recognized to possess immune modulatory and antidiabetic effects. In the present work, polysaccharides were isolated from puerh tea (PTPS) with aging (mild fermentation) for 1 year (PTPS-1), 3 years (PTPS-3) and 5 years (PTPS-5), respectively. The results obtained indicated that all of these three kinds of PTPS are acid heteropolysaccharides bound with proteins, with differences in molecular weights and in monosaccharide compositions. The longer aging process not only produced a higher yield of PTPS but also had higher contents of proteins and uronic acids which proportionally correlated with their antioxidant and α-glycosidase inhibitory activities (PTPS-5 > PTPS-3 > PTPS-1). The anti-α-glycosidase activities of PTPS-5 and PTPS-3 were found to be three times more and equally potent than that of the antidiabetic drug acarbose. All of these findings firstly indicate that the aging process not only induced changes in chemical compositions and proteins conjugation in PTPS, but also markedly enhanced antioxidant and anti-α-glycosidase activities. PTPS-5 exhibited potential antidiabetic effect and merits further studies.

Abstract:Tannase (E.C. 3.1.1.20) is hypothesized to be involved in the metabolism of gallates and gallic acid (GA) in pu-erh tea fermentation. In this work, we measured tannase in Aspergillus niger fermented tea leaves and confirmed the production of fungal tannase during pu-erh tea fermentation. A decrease in catechin and theaflavin gallates and a significant increase in GA content and the relative peak areas of ethyl gallate, procyanidin A2, procyanidin B2, procyanidin B3, catechin-catechin-catechin, epiafzelechin, and epicatechin-epiafzelechin [variable importance in the projection (VIP) > 1.0, p < 0.05, and fold change (FC) > 1.5] were observed using high performance liquid chromatography (HPLC) and metabolomics analysis of tea leaves fermented or hydrolyzed by tannase. In vitro assays showed that hydrolysis by tannase or polymerization of catechins increased the antioxidant activity of tea leaves. In summary, we identified a metabolic pathway for gallates and their derivatives in tea leaves hydrolyzed by tannase as well as associated changes in gallate and GA concentrations caused by fungal tannase during pu-erh tea fermentation.
Study on the Hypoglycemic Mechanism of Tea Polysaccharides and Tea Polyphenols

Abstract:The investigation on the hypoglycemic mechanism of tea polysaccharides and tea polyphenol in diabetic SD rats induced by alloxan was conducted.Methods:After feeding tea polysaccharides or polyphenols for 3 weeks,the changes of the level of blood glucose,insulin and the activities of amylase,sucrase and maltase in small intestine were inspected in diabetic rats.Results about that the suppressive effect of high level of blood glucose induced by alloxan was observed in the groups feeded with tea polysaccharides or tea polyphenols obviously in the experimental period.Compared with control group,the level of blood insulin in the group feeded with tea polysaccharides was increased significantly (P0.05),the activity of sucrase and maltase were decreased significantly.Almost the same change without significance was observed in the group feeded with tea polyphenols while compared with the control group.Investigation indicated that the tea polysaccharides and tea polyphenols have remarkable suppressive hyperglycemic effect on diabetic rats induced by alloxan.These results suggested that functionary mechanisms of the suppressive hyperglycemic effect of tea polysaccharides may be induced by controlling the activities of sucrase and maltase.
Identification and Isolation of α-Glucosidase Inhibitors from Siraitia grosvenorii Roots Using Bio-Affinity Ultrafiltration and Comprehensive Chromatography

Abstract:The discovery of bioactive compounds from medicinal plants has played a crucial role in drug discovery. In this study, a simple and efficient method utilizing affinity-based ultrafiltration (UF) coupled with high-performance liquid chromatography (HPLC) was developed for the rapid screening and targeted separation of α-glucosidase inhibitors from Siraitia grosvenorii roots. First, an active fraction of S. grosvenorii roots (SGR2) was prepared, and 17 potential α-glucosidase inhibitors were identified based on UF-HPLC analysis. Second, guided by UF-HPLC, a combination of MCI gel CHP-20P column chromatography, high-speed counter-current countercurrent chromatography, and preparative HPLC were conducted to isolate the compounds producing active peaks. Sixteen compounds were successfully isolated from SGR2, including two lignans and fourteen cucurbitane-type triterpenoids. The structures of the novel compounds (4, 6, 7, 8, 9, and 11) were elucidated using spectroscopic methods, including one- and two-dimensional nuclear magnetic resonance spectroscopy and high-resolution electrospray ionization mass spectrometry. Finally, the α-glucosidase inhibitory activities of the isolated compounds were verified via enzyme inhibition assays and molecular docking analysis, all of which were found to exhibit certain inhibitory activity. Compound 14 exhibited the strongest inhibitory activity, with an IC50 value of 430.13 ± 13.33 μM, which was superior to that of acarbose (1332.50 ± 58.53 μM). The relationships between the structures of the compounds and their inhibitory activities were also investigated. Molecular docking showed that the highly active inhibitors interacted with α-glucosidase through hydrogen bonds and hydrophobic interactions. Our results demonstrate the beneficial effects of S. grosvenorii roots and their constituents on α-glucosidase inhibition.

Abstract:Recent research has demonstrated the positive impact of herbal tea consumption on postprandial blood glucose regulation. This study conducts a comparative analysis of aqueous and ethanol extractions on four herbal teas (Mallotus, Cyclocarya, Rubus, and Vine) to assess their phytochemical profiles and functional attributes. Phytochemical contents, antioxidant activities, α-glucosidase inhibitory activities, and chemical compositions are investigated via colorimetric analyses and UPLC-Q-Orbitrap HRMS/MS, respectively. Results indicate that Vine, among the teas studied, exhibits the most pronounced glucose-regulating effects under both extraction methods. While ethanol extractions yield higher phytochemical content overall, the compositions vary. Conversely, aqueous extracts demonstrate unexpectedly potent antioxidant activities and comparable α-glucosidase inhibitory activities to ethanol extracts. Phytochemical contents correlate positively with antioxidant activities and α-glucosidase inhibitory activities. However, antioxidant activities exhibit a weak positive correlation with α-glucosidase inhibitory activities. These findings provide evidence that aqueous extracts from herbal teas contain valuable phytochemical compositions beneficial for antioxidants and individuals with hyperglycemia, suggesting their potential as functional ingredients to enhance the nutritional value of herbal food products.

Abstract:Despite abundant research that has been carried out on the potential health benefits of pu-erh tea, no consensus till now, has been reached on which constituent is mainly responsible for its bioactivity. In this work, the aqueous extract (AE) and its fraction enriched with active constituents were prepared from pu-erh tea, and their bioactivities were investigated. It was found that tea polyphenols (TP), tea polysaccharides (TPS), caffeine (Caf), protein (Pro), amino acids (AA) were accumulated in several fractions after solvent extraction despite not being separated completely. Meanwhile, 95% ethanol precipitate (EP) and ethyl acetate fraction (EF) possessed remarkable antioxidant activity and potent inhibitory effects against α-glycosidase in vitro. Furthermore, all the extracts (50 mg/kg BW) showed a significant (p<0.05) effect on postprandial hyperglycemia in diabetic mice as compared with a model group, and the suppression of EP was not significantly (p>0.05) different from that of acarbose at the same dosage (50 mg/kg BW), which indicate that these fractions could be developed as potential anti-diabetic agents.

Abstract:alpha-Glucosidase and protein tyrosine phosphatase 1B (PTP-1B) signaling pathway dual regulators decrease postprandial blood glucose levels and improve insulin sensitivity, making it a new treatment strategy for type 2 diabetes. This study examined in vitro antidiabetic activities of 8-C-ascorbyl-(-)-epigallocatechin (AE), found in oolong tea. AE inhibited alpha-glucosidase (IC50 = 142.8 NM) with activity higher than that of acarbose (IC50 = 250.2 NM). AE significantly promoted glucose-consumption and activated the insulin signaling pathway through enhancing the protein levels of p-GSK38 and p-Akt and inhibiting the expression of PTP-1B, along with slightly inhibitory activity against PTP-1B. Docking analysis showed AE inhibited alpha-glucosidase activity via binding to the catalytic site through hydrogen bonds and Pi-Pi interactions, as well as a good shape match to the active pocket. In addition, AE could relieve oxidative damage and possessed good antioxidant capacity. Taken together, the results of this study indicate that AE exhibits antidiabetic activity in vitro, making it a potential functional food ingredient and drug candidate for management of type 2 diabetes.

Abstract:Natural α-glucosidase inhibitors from plant-based foods such as catechins offer an attractive strategy for their potential anti-diabetic effects. In this study, infusions of three different tea types (green, white, and oolong) were investigated for their total phenolic (TPC) and catechins (EGCG, ECG, EGC, and EC) content, and for their α-glucosidase inhibitory activities. We observed that the level of TPC in white tea was significantly higher compared to oolong and green tea, which suggests higher content of EGCG and ECG catechins in fresh young leaves. Our findings showed that the higher content of such catechins in the infusion of white tea well correlated with a strong inhibition of α-glucosidase, and such inhibition was demonstrated to be more effective than the FDA-approved drug acarbose. Then, we computationally explored the molecular requirements for enzyme inhibition, especially for the most active catechins EGCG and ECG, as well as their disposition/stability within the active site. Abstract:Cyclocarya paliurus , as an important edible and medicinal product, has shown a good prospect in the prevention of diabetes mellitus (DM). However, it is unclear which active compounds derived from C. paliurus play a significant role in inhibiting α-glucosidase activity. After being enriched by D-101 macroporous resin, five eluent fractions with different polarity were obtained and their inhibitory activities on α-glucosidase were evaluated by an enzyme inhibition assay in vitro . The result showed that 70% ethanol fraction of C. paliurus leaves exhibited remarkable α-glucosidase inhibitory activity with the IC 50 value of 17.81 μg/mL. The 70% ethanol fraction was incubated with α-glucosidase and then active compounds would form enzyme-inhibitor complexes. The complexes could be separated from inactive components by the interception ability of ultrafiltration membrane under centrifugation. A total of 36 active compounds were screened from C. paliurus leaves and the chemical structures were further characterized by UPLC-QTOF-MS/MS. Furthermore, molecular docking was performed to investigate possible inhibitory mechanisms between active compounds and α-glucosidase. The docking result showed that cyclocarioside I, pterocaryoside B, arjunolic acid, cyclocarioside Z5, cypaliuruside D and cyclocarioside N could be embedded well into the active pocket of α-glucosidase, and had significant affinity interactions with critical amino acid residues by forming hydrogen bonds, hydrophobic interactions and van der waals, and affinity energies ranged from -9.3 to -6.7 kJ/mol. The results indicated that the developed method is rapid and effective for high throughput screening of potential α-glucosidase inhibitors from complex mixtures. Moreover, C. paliurus exhibited a remarkable inhibitory activity on α-glucosidase, making it a promising candidate for the prevention of DM.

Abstract:The leaves of Rubus suavissimus (also called "Chinese sweet tea") is used not only as a beverage tea and food additive but also as a folk herbal medicine for the treatment of diabetes mellitus. To systematically investigate its material foundation of efficacy for treating diabetes, herein, a hyphenated strategy by off-line coupling high-speed countercurrent chromatography, ultrafiltration HPLC-UV-MS and prep-HPLC was developed. And thus, α-glucosidase inhibitors from Rubus suavissimus leaves was comprehensively profiled, purified and characterized. As a result, twenty-six compounds were identified as potential α-glucosidase inhibitors and favorably isolated by prep-HPLC. Their structures were identified via UV, MS, and 1H-NMR. Notably, fourteen compounds, including protocatechuic acid (1), myketin (3), epicatechin (4), vanillic acid (6), apigenin (11), catechin (12), ferulic acid (15), luteolin (16), 3, 3′-di-O-methylellagic acid (17), chlorogenic acid (19), 3, 3′-di-O-methylellagic acid-4′-O-β-D-glucoside (20), cinnamic acid (21), syringate (24) and ethylbrevifolin-carboxylate (25), were reported in leaves of Rubus suavissimus for the first time. In addition, α-glucosidase inhibitory activities of compounds 1–26 were evaluated, and eighteen compounds exhibited stronger α-glucosidase inhibitory activities than acarbose (IC50 value at 214.24 ± 3.48 μg/mL, positive control). The results indicated that the proposed method is a highly efficient strategy for comprehensive profiling and purification of bioactive target compounds, including both major and minor components, from natural products.

Abstract:The effects of tea water extracts (TWE), crude tea polysaccharides (CTP), and a tea polysaccharide fraction (TPF) were tested on hyperglycemic diabetic mice. Results indicated that TWE, CTP, and TPF could significantly decrease fasting blood glucose (FBG) and glucosylated serum protein (GSP) in alloxan-induced diabetic mice compared to the control group. In vitro antioxidant activities of TWE, CTP, and TPF for scavenging hydroxyl radicals and superoxide radicals decreased with the degree of purification and were lowest for TPF. High-performance gel permeation chromatography (HPGPC) and component analysis revealed the molecular mass distribution and constituents of TWE, CTP, and TPF, indicating that a 100-120 kDa fraction contained the hypoglycemic activity. This fraction was essentially composed of polysaccharides (approximately 90%) with substantial amounts of arabinogalactan proteins. The second-derivative IR spectra of TWE, CTP, and TPF with peak intensity around 1075 and 1045 cm(-1), which characterize galactopyranose in the backbone and arabinofuranose units in side branches, respectively, further substantiated the importance of the arabinogalactan proteins. Taken together, the results indicate that a soluble tea polysaccharide is the major hypoglycemic factor in tea and that this polysaccharide may be developed to a potential natural hypoglycemic functional ingredient.

Abstract:Peptides are considered promising sources of nutraceuticals. In this study, a mixture of peptides was prepared from Paeonia ostii 'Feng Dan' seed meal protein by continuous enzymolysis. Successive separation and purification procedures, including ultrafiltration and reversed-phase high-performance liquid chromatography (RP-HPLC), were performed, and six novel peptides were identified by liquid chromatography-electrospray ionization source-mass spectrometry/mass spectrometry (LC-ESI-MS/MS). In an in vitro antidiabetic activity test, Tyr-Phe-Phe-Met exhibited stronger α-glucosidase inhibitory activity (48.17 ± 3.34% at 1 mg/mL) than the other peptides. Docking studies of this peptide into the active site of α-glucosidase showed that the formation of hydrogen bonds could be critical for the enzymatic trapping of inhibitory peptides. Furthermore, two novel peptides, Phe-Phe-Phe-Met (IC50 = 245.46 ± 44.01 µM) and Tyr-Tyr-Phe-Met (IC50 = 306.71 ± 48.17 µM), with improved α-glucosidase inhibitory activity, were designed based on molecular docking. Therefore, the seed meal of Paeonia ostii could be considered a functional food ingredient for the management of hyperglycemia, and three novel peptides were identified as α-glucosidase inhibitors.
