Abstract:The rapid advance and growth of the point-of-care diagnosis industry has provided an impetus for the development of novel signal labels for highly sensitive bio-molecule detections. Colloidal quantum dots (QDs) exhibit superior brightness, facile surface functionalization and exceptional photostability, making them the preferred option for these biological applications. However, the significantly reduced fluorescence intensity and limited photochemical stability in complex biological environments have greatly hampered their further use. Herein, with an innovative alloying engineering strategy, the high-quality water-soluble CdSe/CdxZn1−xS QDs with near-unity PL quantum yield and monoexponential PL decay dynamics are obtained. Notably, for the first time, a record-breaking stability at the single QD level in water with nonblinking behavior persisting for an hour is achieved, which approaching those of state-of-the-art hydrophobic QDs. Furthermore, the nanocomposites formulated with these novel alloyed QDs demonstrate a remarkable PL QY of 96 % and provide an ultrasensitive detection for prostate specific antigen on lateral flow immunoassays. These findings presented here shed new light on the design of high-brightness water-soluble QDs in single-molecule level and QDs-based nanocomposites, significantly pushing ahead toward high-sensitivity biomedical detections and diagnosis.

Abstract:Quantum dots (QDs), owning to their unique optical and electronic properties, are increasingly used for bio-imaging/biosensing in medical and energy fields. Biosynthesis of QDs offers several advantages over the conventional chemical synthesis route, such as low cost, environmental-benignity, and no need for further modification of the products. However, information about the regulation and mechanism of synthesis of QDs with great fluorescence properties is still limited. In this work, we prepared cadmium selenium (CdSe) QDs in vivo using Candida utilis WSH02-08. We regulated the fluorescence properties of the bio-QDs through different precursor concentrations, and found that the bio-QDs with high fluorescence intensity and long photostable lifetime were favoured at an elevated Cd content over Se. The synthesis of the high-quality bio-QDs was attributed to the less formation of Se(0) and the increased formation of CdSe and CdS. Furthermore, such synthesized QDs were directly used in live-cell imaging without further modification. These findings are expected to enable a further design of biosynthesized QDs with great fluorescence properties and expand our understanding of the regulation mechanism of nanoparticles fabrication in microorganisms. Facile Synthesis of N-rich Carbon Quantum Dots from Porphyrins As Efficient Probes for Bioimaging and Biosensing in Living Cells.

Abstract:N-rich metal-free and metal-doped carbon quantum dots (CQDs) have been prepared through one-step hydrothermal method using tetraphenylporphyrin or its transition metal (Pd or Pt) complex as precursor. The structures and morphology of the as-prepared nanoparticles were analyzed by X-ray diffraction, high-resolution transmission electron microscopy, X-ray photoelectron spectroscopy, and Fourier transform infrared spectra. Three kinds of nanocomposites show similar structures except for the presence of metal ions in Pd-CQDs and Pt-CQDs indicated by X-ray photoelectron spectroscopy. All of them display bright blue emission upon exposure to ultraviolet irradiation. The CQDs exhibit typical excitation-dependent emission behavior, with the emission quantum yield of 10.1%, 17.8%, and 15.2% for CQDs, Pd-CQDs, and Pt-CQDs, respectively. Moreover, the CQDs, Pd-CQDs, and Pt-CQDs could serve as fluorescent probes for the specific and sensitive detection of Fe3+ ions in aqueous solution. The low cytotoxicity of CQDs is demonstrated by MTT assay against HeLa cells. Therefore, the CQDs can be used as efficient probes for cellular multicolor imaging and fluorescence sensors for the detection of Fe3+ ions due to their low toxicity, excellent biocompatibility, and low detection limits. This work provides a new route to synthesize highly luminescent N-rich metal-free or metal-doped CQDs for multifunctional applications.

Abstract:In the past two decades, Cu-doped inorganic semiconductors with near-infrared (NIR) emitting have garnered stupendous research interest. Nevertheless, the incompatibility between the NIR emitting and high photoluminescence quantum yield (PLQY) of the Cu-doped fluorescent probes restricted the extensive application in biological imaging. Herein, the water soluble Cu+ doped CdS quantum dots (Cu+: CdS QDs) were prepared by using a one step synthesis method in a N-2 atmosphere. The d-dots possessed an ultra small size (similar to 5 nm), a high QY (25.6%), NIR emission (similar to 700 nm) and a low cytotoxicity because of which they were used as fluorescence probes. Besides, the study of the Cu d state and the mechanism of emission proposed the explanation for the fluorescence enhancement compared with previous reports. Moreover, the optimal conditions of dopants, stabilizers, sulfur ion concentration, pH, heating time and reflux temperature were also studied systematically to acquire bestquality Cu-doped nanocrystals. Due to the excellent optical properties and favourable biocompatibility, the Cu+: CdS QDs as fluorescence probes were successfully applied to label living 3T3 cells.

Abstract:We developed a novel one-pot polyol approach for the synthesis of biocompatible CdSe quantum dots (QDs) using poly(acrylic acid) (PAA) as a capping ligand at 240°C. The morphological and structural characterization confirmed the formation of biocompatible and monodisperse CdSe QDs with several nanometers in size. The encapsulation of CdS thin layers on the surface of CdSe QDs (CdSe/CdS core–shell QDs) was used for passivating the defect emission (650 nm) and enhancing the fluorescent quantum yields up to 30% of band-to-band emission (530–600 nm). Moreover, the PL emission peak of CdSe/CdS core–shell QDs could be tuned from 530 to 600 nm by the size of CdSe core. The as-prepared CdSe/CdS core–shell QDs with small size, well water solubility, good monodispersity, and bright PL emission showed high performance as fluorescent cell labels in vitro. The viability of QDs-labeled 293T cells was evaluated using a 3-(4,5-dimethylthiazol)-2-diphenyltertrazolium bromide (MTT) assay. The results showed the satisfactory (>80%) biocompatibility of as-synthesized PAA-capped QDs at the Cd concentration of 15 μg/ml.

Abstract:Biosynthesis offers opportunities for cost-effective and sustainable production of semiconductor quantum dots (QDs), but is currently restricted by poor controllability on the synthesis process, resulting from limited knowledge on the assembly mechanisms and the lack of effective control strategies. In this work, we provide molecular-level insights into the formation mechanism of biogenic QDs (Bio-QDs) and its connection with the cellular substrate metabolism in Escherichia coli. Strengthening the substrate metabolism for producing more reducing power was found to stimulate the production of several reduced thiol-containing proteins (including glutaredoxin and thioredoxin) that play key roles in Bio-QDs assembly. This effectively diverted the transformation route of the selenium (Se) and cadmium (Cd) metabolic from Cd3(PO4)2 formation to CdS xSe1- x QDs assembly, yielding fine-sized (2.0 ± 0.4 nm), high-quality Bio-QDs with quantum yield (5.2%) and fluorescence lifetime (99.19 ns) far exceeding the existing counterparts. The underlying mechanisms of Bio-QDs crystallization and development were elucidated by density functional theory calculations and molecular dynamics simulation. The resulting Bio-QDs were successfully used for bioimaging of cancer cells and tumor tissue of mice without extra modification. Our work provides fundamental knowledge on the Bio-QDs assembly mechanisms and proposes an effective, facile regulation strategy, which may inspire advances in controlled synthesis and practical applications of Bio-QDs as well as other bionanomaterials.

Abstract:Heavy-metal-free I-III-VI2 quantum dots (QDs) have recently emerged as favorable alternatives to toxic II-VI QDs for optoelectronic and biological applications, but low fluorescence efficiency is a key issue related to the promising nanocrystals. In this work, we demonstrate big enhancement of two-photon fluorescence efficiency and low threshold two-photon pumped amplification of spontaneous emission (ASE) of Zn-processed CuInS2 (CIS) QDs for the first time. The change of two-photon process caused by Zn2+ cation exchange in CIS QDs is systematically investigated. Two-photon ASE in Zn-processed CIS QDs films is achieved with a record of low threshold fluence of 5.7 mu J/cm(2). With high two-photon fluorescence quantum yields and low threshold ASE, the Zn-processed CIS QDs could become a promising candidate for two-photon bioimaging, nonlinear optical and novel QDs laser devices.

Abstract:I-III-VI QDs (CuInS2/ZnS or AgInS2/ZnS core/shell structures) possess low toxicity, and are a logical replacement for cadmium-based QDs for biomedical applications. Our synthesis of I-III-VI QDs is based on thermal decomposition of less toxic precursors and can be easily scaled up for mass production for sustainable and reliable imaging and sensing experiments. Through nonstoichiometric composition adjustment, we synthesized I-III-VI QDs with reliable and controllable optical properties, including high QYs and tunable photoluminescence. We also developed new zwitterionic amphiphiles and applied them to encapsulate I-III-VI QDs to achieve colloidal stability in proteinaceous solutions with wide pH/ionic ranges, low non-specific binding, and easily bio-conjugation. On the basis of these developments, we applied our I-III-VI QDs in cellular imaging to to specifically target human brain tumor cells.

Abstract:Owing to their high emission intensity and size-based tunability, lead sulfide (PbS) quantum dots (QDs) have been employed in a broad spectrum of biological applications especially for the second near infrared (NIR-II) biological imaging. However, the toxicity of Pb2+ ions remains an unavoidable risk factor during both preparation and biological applications. Here, we have developed an economical and environmentally friendly synthetic route with a recycle setup to prepare whey-protein-capped PbS QDs. Furthermore, the final Pb2+ ions were allowed to react with excess S2- ions and produce nontoxic precipitate in the end, which helped to avoid polluting the environment. After surface modification with protein, both our prepared QDs and the final reaction wastes demonstrated low toxicity in following both in vitro and in vivo experiments. Notably, the conversion efficiency of Pb2+ ions into PbS QDs in this study was almost 3 times than that of a one-time reaction without recycle. In addition, each batch of as prepared PbS QDs featured good stability and bright fluorescence at the NIR-II window in in vivo imaging experiments. Our work shows a novel green strategy for the sustainable synthesis of biocompatible QDs that contain heavy metal, especially for considering the follow-up biological applications.

Abstract:This study proposed a novel approach for the biosynthesis of CdSe quantum dots (QDs) by a recombinant photosynthetic bacteria Rhodopseudomonas palustris with metallothionein expression. The biosynthesis mechanism and physico-chemical properties of the CdSe QDs were analyzed by various techniques, and its potential application was also investigated. The results showed that the biosynthesized CdSe QDs presented green fluorescence with maximum excitation and emission wavelengths at 460 and 530 nm, respectively. The quantum yield of the biosynthesized CdSe QDs was found out to be 3.85%. They were well-dispersed spherical particles with an average size of 5.37 ± 0.06 nm. Their crystal structure was cubic with the d spacing of 3.1 Å and 2.5 Å, corresponded to the planes of (111) and (220), respectively. The biosynthesis process of CdSe QDs took place in the cytoplasm and may involve some proteins, peptides, lipids or other organic substances covering the surface of CdSe QDs through several specific functional groups, such as hydroxyl or sulfhydryl groups to maintain its stability. The purified CdSe QDs had good biocompatibility and great potential for cellular imaging application. It is concluded that the process may provide a more cost-effective and eco-friendly method not only to recover harmful cadmium in the environment but also to synthesize simultaneously a clean product for various industrial applications.

Abstract:ZnS quantum dots (QDs) are among the most promising emerging fluorescent materials for biolabeling. High-quality colloidal ZnS QDs were synthesized via a new facile chemical precipitation method using the mixture of ethylene glycol (EG) and water as the solvent. The phase structure and morphology of the ZnS QDs were characterized by X-ray powder diffraction and high resolution transmission electron microscopy. The synthesized ZnS QDs have a cubic zinc blende structure with monodispered and small particles. ZnS QDs easily dispersed in water to form stable and clear colloids and the strong tunable trap state emissions from 452 to 516 nm were achieved by varying the reaction time. Fabrication of the CdSeTe Alloyed and CdSeTe/ZnS Core-Shell Quantum Dots

Abstract:The near-infrared (NIR)-emitting quantum dots (Qdots) have great potential for the use in biological imaging and diagnostic applications. In our work, a facile method was developed for the preparation of high quality, water-soluble, and NIR-emitting CdSeTe alloyed Qdots (A-Qdots) with L-cysteine (L-cys) as capping agent. By changing the size and the composition of the A-Qdots, the photoluminescent quantum yields (QYs) can reach as high as 53% and the emission color can be tuned between visible and NIR regions. Based on the fluorescence of the A-Qdots selectively quenched in the presence of Cu2+, the NIR-emitting CdSeTe A-Qdots were applied in ultrasensitive Cu2+ sensing. Furthermore, the prepared CdSeTe A-Qdots have been successfully applied for cell imaging, glucose and cholesterol assay, which demonstrates the great potential of the Qdots for biological applications. In order to improve the biocompatibility of the CdSeTe A-Qdots, new water-soluble CdSeTe/ZnS core-shell Qdots (CS-Qdots) with excellent NIR emission were synthesized in aqueous solution. The prepared CS-Qdots not only possessed high QYs but also exhibited excellent photobstability and favorable biocompatibility. Moreover, the CS-Qdots showed high electrogenerated chemiluminescence (ECL) signal. These characteristics showed their potential applications in cell imaging and biosensing with high sensitivity.

Abstract:We report here on a one-pot, mild and low cost aqueous-based synthetic route for the preparation of colloidally stable and highly luminescent dual-doped Ag,Ni:ZnCdS/ZnS core/shell quantum dots (QDs). The pure dopant emission of the Ni-doped core/shell QDs was found to be highly affected by the presence of a second dopant ion (Ag+). Results showed that the PL emission intensity increases while its peak position experiences an obvious blue shift with an increase in the content of Ag+ ions. Regarding the optical observations, we provide a simple scheme for absorption-recombination processes of the carriers through impurity centers. To obtain optimum conditions with a better emission characteristic, we also study the effect of different reaction parameters, such as refluxing temperature, the pH of the core and shell solution, molar ratio of the dopant ions (Ni:(Zn+Cd) and Ag:(Zn+Cd)), and concentration of the core and shell precursors. Nonetheless, the most effective parameter is the presence of the ZnS shell in a suitable amount to eliminate surface trap states and enhance their emission intensity. It can also improve the bio-compatibility of the prepared QDs by restricting the Cd2+ toxic ions inside the core of the QDs. The present suggested route also revealed the remarkable optical and chemical stability of the colloidal QDs which establishes them as a decent kind of nano-scale structure for light emitting applications, especially in biological technologies. The suggested process also has the potential to be scaled-up while maintaining the emission characteristics and structural quality necessary for industrial applications in optoelectronic devices.
Quantum Dots with Highly Efficient, Stable, and Multicolor Electrochemiluminescence

Abstract:Outstanding photoluminescence (PL) and electro-luminescence properties of quantum dots (QDs) promise possibilities for them to meet challenging expectations of electrochemiluminescence (ECL), which at present relies on inefficient and spectral-irresolvable emitters based on transition-metal complexes (such as Ru(bpy)(3)(2+)). However, ECL is reported to be extremely sensitive to the surface traps on the QDs likely because of the spatially and temporally separated electrochemical charge injections. Results here reveal that, by engineering the interior inorganic structure (CdSe/CdS/ZnS core/shell/shell structure) and inorganic-organic interface using new synthetic methods, the trap-insensitive QDs with near-unity PL quantum yield and monoexponential PL decay dynamics in water generated narrow band-edge ECL with efficiencies about six orders of magnitude higher than that of the standard Ru(bpy)(3)(2+)). The band-edge and spectrally resolved ECL from CdSe/CdS/ZnS core/shell/shell QDs demonstrated a new readout scheme using electrochemical potential. Excellent ECL performance of QDs uncovered here offer opportunities to realize the full potential of ECL for biomedical detection and diagnosis.
Stable Monodisperse Pb1-xCdxS Quantum Dots for NIR-II Bioimaging by Aqueous Coprecipitation of Bimetallic Clusters

Abstract:Aqueous lead sulfide (PbS) quantum dots (QDs) synthesized by traditional methods are unstable, so that they are usually coated with cadmium sulfide (CdS) to prevent oxidation, which are complicated and not satisfactory for mass production. Here, stable ternary Pb1-xCdxS QDs were synthesized by in situ coprecipitation of Pb4-nCdnO4 bimetallic clusters in an aqueous solution, which possess a uniform size of 4.0 +/- 0.2 nm and the second near-infrared (NIR-II) emission at 1100 nm with photoluminescence quantum yield (PLQY) as high as 17.72 %. Stored at 4 degrees C and in colloidal form, the PLQY of Pb1-xCdxS QDs remained at 90.9 % of the initial value after 15 days, while stored as powder, the spectra did not change after 5 months. The high PLQY and good water compatibility of Pb1-xCdxS QDs provide a good performance in vivo vasculature imaging and lymphatic system imaging at a very low power density (10 mW cm(-2)) in the NIR-II window.
Large scale synthesis of stable tricolor Zn(1-x)Cd(x)Se core/multishell nanocrystals via a facile phosphine-free colloidal method.

Abstract:Here we report a new "green" method to synthesize Zn(1-x)Cd(x)Se (x = 0-1) and stable red-green-blue tricolor Zn(1-x)Cd(x)Se core/shell nanocrystals using only low cost, phosphine-free and environmentally friendly reagents. The first excitonic absorption peak and photoluminescence (PL) position of the Zn(1-x)Cd(x)Se nanocrystals (the value of x is in the range 0.005-0.2) can be fixed to any position in the range 456-540 nm. There is no red or blue shift in the entire reaction process. Three similar sizes of alloyed Zn(1-x)Cd(x)Se nanocrystals with blue, green, and yellow emissions were successfully selected as cores to synthesize high quality blue, green, and red core/shell nanocrystal emitters. For the synthesis of core/shell nanocrystals with a high quantum yield (QY) and stability, the selection of shell materials has been proven to be very important. Therefore, alternative protocols have been used to optimize thick shell growth. ZnSe/ZnSe(x)S(1-x) and CdS/Zn(1-x)Cd(x)S have been found as an excellent middle multishell to overcoat between the alloyed Zn(1-x)Cd(x)Se core and ZnS outshell. The QYs of the as-synthesized core/shell alloyed Zn(1-x)Cd(x)Se nanocrystals can reach 40-75%. The Cd content is reduced to less than 0.1% for Zn(1 -x)Cd(x)Se core/shell nanocrystals with emissions in the range 456-540 nm. More than 15 g of high quality Zn(1-x)Cd(x)Se core/shell nanocrystals were prepared successfully in a large scale, one-pot reaction. Importantly, the emissions of such thick multishell nanocrystals are not susceptible to ligand loss and stability in various physiological conditions.

Abstract:We demonstrate a simple and efficient biosynthesis method to prepare easily harvested biocompatible cadmium telluride (CdTe) quantum dots (QDs) with tunable fluorescence emission using yeast cells. Ultraviolet-visible (UV-vis) spectroscopy, photoluminescence (PL) spectroscopy, X-ray diffraction (XRD), and transmission electron microscopy (TEM) confirm that the CdTe QDs are formed via an extracellular growth and subsequent endocytosis pathway and have size-tunable optical properties with fluorescence emission from 490 to 560 nm and a cubic zinc blende structure with good crystallinity. In particular, the CdTe QDs with uniform size (2-3.6 nm) are protein-capped, which makes them highly soluble in water, and in situ bio-imaging in yeast cells indicates that the biosynthesized QDs have good biocompatibility. This work provides an economic and environmentally friendly approach to synthesize highly fluorescent biocompatible CdTe QDs for bio-imaging and bio-labeling applications.
Synthesis of Colloidal Quantum Dots with an Ultranarrow Photoluminescence Peak

Abstract:Localization of exciton wavefunctions away from the inorganicorganic interface is proposed for synthesizing colloidal spheroidal quantum dots (QDs) with an ultranarrow photoluminescence (PL) peak width. This strategy is demonstrated by synthesizing uniform-alloy CdxZn1-xSe and CdxZn1-xSe/ZnSe/ZnS core/shell QDs. For blue-emitting QDs, the ensemble PL full width at halfmaximum (fwhm) reaches 10.2 nm and the corresponding single-dot PL fwhm is 5.2 nm. The ensemble and single-dot PL fwhm values for green-emitting QDs are 16.3 and 9.7 nm, respectively. These record-low PL fwhm values for spheroidal QDs are close to the requirements for ideal displays. To control the composition homogeneity, the synthetic scheme is separated into four consecutive steps, namely, nucleation of monodisperse CdSe core QDs, epitaxial growth of thickness-controlled CdSe/ZnSe core/shell QDs with a low degree of spontaneous alloying, Cu-catalyzed alloying for uniform-alloy QDs, and additional epitaxy of outer ZnSe and ZnS shells onto uniform-alloy CdxZn1-xSe QDs. Among all metal ions tested, copper ions are identified to be the best catalysts for forming uniform-alloy CdxZn1-xSe QDs, which can enter a QD rapidly to initiate alloying and effectively exit a QD after the alloying process under the experimentally feasible conditions.

Abstract:This study details a novel method for the extracellular microbial synthesis of cadmium sulfide (CdS) quantum dots (QDs) by the white rot fungus Phanerochaete chrysosporium. P. chrysosporium was incubated in a solution containing cadmium nitrate tetrahydrate, which became yellow from 12h onwards, indicating the formation of CdS nanocrystals. The purified solution showed a maximum absorbance peak between 296 and 298 nm due to CdS particles in the quantum size regime. The fluorescence emission at 458 nm showed the blue fluorescence of the nanoparticles. X-ray analysis of the nanoparticles confirmed the production of CdS with a face-centered cubic (fcc) crystal structure. The average grain size of the nanoparticles was approximately 2.56 nm, as determined from the full width at half-maximum (FWHM) measurement of the most intense peak using Scherer's equation. Transmission electron microscopic analysis showed the nanoparticles to be of a uniform size with good crystallinity. The changes to the functional groups on the biomass surface were investigated through Fourier transform infrared spectroscopy. Furthermore, the secretion of cysteine and proteins was found to play an important role in the formation and stabilization of CdS QDs. In conclusion, our study outlines a chemical process for the molecular synthesis of CdS nanoparticles.
