Abstract:Motile cilia and flagella beat rhythmically on the surface of cells to power the flow of fluid and to enable spermatozoa and unicellular eukaryotes to swim. In humans, defective ciliary motility can lead to male infertility and a congenital disorder called primary ciliary dyskinesia (PCD), in which impaired clearance of mucus by the cilia causes chronic respiratory infections(1). Ciliary movement is generated by the axoneme, a molecular machine consisting of microtubules, ATP-powered dynein motors and regulatory complexes(2). The size and complexity of the axoneme has so far prevented the development of an atomic model, hindering efforts to understand how it functions. Here we capitalize on recent developments in artificial intelligence-enabled structure prediction and cryo-electron microscopy (cryo-EM) to determine the structure of the 96-nm modular repeats of axonemes from the flagella of the alga Chlamydomonas reinhardtii and human respiratory cilia. Our atomic models provide insights into the conservation and specialization of axonemes, the interconnectivity between dyneins and their regulators, and the mechanisms that maintain axonemal periodicity. Correlated conformational changes in mechanoregulatory complexes with their associated axonemal dynein motors provide a mechanism for the long-hypothesized mechanotransduction pathway to regulate ciliary motility. Structures of respiratory-cilia doublet microtubules from four individuals with PCD reveal how the loss of individual docking factors can selectively eradicate periodically repeating structures.
Hydrodynamic Instabilities Provide A Generic Route To Spontaneous Biomimetic Oscillations In Chemomechanically Active Filaments

Abstract:Non-equilibrium processes which convert chemical energy into mechanical motion enable the motility of organisms. Bundles of inextensible filaments driven by energy transduction of molecular motors form essential components of micron-scale motility engines like cilia and flagella. The mimicry of cilia-like motion in recent experiments on synthetic active filaments supports the idea that generic physical mechanisms may be sufficient to generate such motion. Here we show, theoretically, that the competition between the destabilising effect of hydrodynamic interactions induced by force-free and torque-free chemomechanically active flows, and the stabilising effect of nonlinear elasticity, provides a generic route to spontaneous oscillations in active filaments. These oscillations, reminiscent of prokaryotic and eukaryotic flagellar motion, are obtained without having to invoke structural complexity or biochemical regulation. This minimality implies that biomimetic oscillations, previously observed only in complex bundles of active filaments, can be replicated in simple chains of generic chemomechanically active beads.

Abstract:With exquisite precision and reproducibility, cells orchestrate the cooperative action of thousands of nanometer-sized molecular motors to carry out mechanical tasks at much larger length scales, such as cell motility, division and replication. Besides their biological importance, such inherently non-equilibrium processes are an inspiration for developing biomimetic active materials from microscopic components that consume energy to generate continuous motion. Being actively driven, these materials are not constrained by the laws of equilibrium statistical mechanics and can thus exhibit highly sought-after properties such as autonomous motility, internally generated flows and self-organized beating. Starting from extensile microtubule bundles, we hierarchically assemble active analogs of conventional polymer gels, liquid crystals and emulsions. At high enough concentration, microtubules form a percolating active network characterized by internally driven chaotic flows, hydrodynamic instabilities, enhanced transport and fluid mixing. When confined to emulsion droplets, 3D networks spontaneously adsorb onto the droplet surfaces to produce highly active 2D nematic liquid crystals whose streaming flows are controlled by internally generated fractures and self-healing, as well as unbinding and annihilation of oppositely charged disclination defects. The resulting active emulsions exhibit unexpected properties, such as autonomous motility, which are not observed in their passive analogues. Taken together, these observations exemplify how assemblages of animate microscopic objects exhibit collective biomimetic properties that are starkly different from those found in materials assembled from inanimate building blocks, challenging us to develop a theoretical framework that would allow for a systematic engineering of their far-from-equilibrium material properties.
Flagella-like beating of a single microtubule

Abstract:Kinesin motors can induce a buckling instability in a microtubule with a fixed minus end. Here we show that by modifying the surface with a protein-repellent functionalization and using clusters of kinesin motors, the microtubule can exhibit persistent oscillatory motion, resembling the beating of sperm flagella. The observed period is of the order of 1 min. From the experimental images we theoretically determine a distribution of motor forces that explains the observed shapes using a maximum likelihood approach. A good agreement is achieved with a small number of motor clusters acting simultaneously on a microtubule. The tangential forces exerted by a cluster are mostly in the range 0 - 8 pN towards the microtubule minus end, indicating the action of 1 or 2 kinesin motors. The lateral forces are distributed symmetrically and mainly below 10 pN, while the lateral velocity has a strong peak around zero. Unlike well-known models for flapping filaments, kinesins are found to have a strong "pinning" effect on the beating filaments. Our results suggest new strategies to utilize molecular motors in dynamic roles that depend sensitively on the stress built-up in the system.

A minimal physical model captures the shapes of crawling cells

Abstract:Cell motility in higher organisms (eukaryotes) is crucial to biological functions ranging from wound healing to immune response, and also implicated in diseases such as cancer. For cells crawling on hard surfaces, significant insights into motility have been gained from experiments replicating such motion in vitro. Such experiments show that crawling uses a combination of actin treadmilling (polymerization), which pushes the front of a cell forward, and myosin-induced stress (contractility), which retracts the rear. Here we present a simplified physical model of a crawling cell, consisting of a droplet of active polar fluid with contractility throughout, but treadmilling connected to a thin layer near the supporting wall. The model shows a variety of shapes and/or motility regimes, some closely resembling cases seen experimentally. Our work strongly supports the view that cellular motility exploits autonomous physical mechanisms whose operation does not need continuous regulatory effort.

Abstract:Groups of non-processive myosin motors exhibit complex and non-linear behaviors when binding to actin. These operate at larger scales and time frames than an individual motor, indicating the presence of a strong cooperative disposition. Limits in contemporary microscopy prevent verification of motor-filament binding dynamics, whilst mathematical models rely on continuum abstractions in which cooperativity is implicit and individual motor behavior cannot be separated from the behaviour of the whole. Understanding the fundamental interactions driving the emergent behaviour in actomyosin therefore remains an open question. Here we suggest that the diversity of empirically observed in-vitro oscillations can be explained by a minimal Kuramoto-style phase oscillator model of actomyosin, where cooperativity is orchestrated by the actomyosin geometry and mechanical environment. The model mirrors the irregular and regular saw-tooth oscillations present in in-vitro actomyosin and sarcomeric 'SPOC' experiments with only adjustments of the external mechanical environment, and despite the model's simplicity. Actomyosin-like behaviour thus arises as a generic property of the discontinuous mechanical coupling in an incommensurate architecture, rather than specific to molecular motor reaction kinetics. We demonstrate a range of synchronising behaviours arising from the cooperative motor dynamics that, once synchronised, are stable over a large range of external forces. The nature of the synchronisation patterns allow recruitment of rotors as the external force increases, reducing variance in the backbone's velocity. This is a demonstration of morphological control. Due to interest in this behaviour in contemporary robotics, we build a physical experiment, using electric motors to power our oscillators. Using the experiment we verify both the organisational and control properties of the system. This demonstrates non-biological motors can cooperate similarly to biological motors when working within an actomyosin geometry, suggesting that the actomyosin complex may not depend on motor-specific qualities to achieve its biological function. These findings offer novel insights into synchronising networks of oscillators and have potential applications in emulating actomyosin-like behaviors within contemporary robotics using non-biological motors.

Abstract:Effective organismal behavior responds appropriately to changes in the surrounding environment. Attaining this delicate balance of sensitivity and stability is a hallmark of the animal kingdom. By studying the locomotory behavior of a simple animal (\textit{Trichoplax adhaerens}) without muscles or neurons, here, we demonstrate how monociliated epithelial cells work collectively to give rise to an agile non-neuromuscular organism. Via direct visualization of large ciliary arrays, we report the discovery of sub-second ciliary reorientations under a rotational torque that is mediated by collective tissue mechanics and the adhesion of cilia to the underlying substrate. In a toy model, we show a mapping of this system onto an "active-elastic resonator". This framework explains how perturbations propagate information in this array as linear speed traveling waves in response to mechanical stimulus. Next, we explore the implications of parametric driving in this active-elastic resonator and show that such driving can excite mechanical 'spikes'. These spikes in collective mode amplitudes are consistent with a system driven by parametric amplification and a saturating nonlinearity. We conduct extensive numerical experiments to corroborate these findings within a polarized active-elastic sheet. These results indicate that periodic and stochastic forcing are valuable for increasing the sensitivity of collective ciliary flocking. We support these theoretical predictions via direct experimental observation of linear speed traveling waves which arise from the hybridization of spin and overdamped density waves. We map how these ciliary flocking dynamics result in agile motility via coupling between an amplified resonator and a tuning (Goldstone-like) mode of the system. This sets the stage for how activity and elasticity can self-organize into behavior which benefits the organism as a whole.
Biomimetic Synchronization in biciliated robots

Abstract:Direct mechanical coupling is known to be critical for establishing synchronization among cilia. However, the actual role of the connections is still elusive - partly because controlled experiments in live samples are challenging. Here, we employ an artificial ciliary system to address this issue. Two cilia are formed by chains of self-propelling robots and anchored to a shared base so that they are purely mechanically-coupled. The system mimics biological ciliary beating but allows fine control over the beating dynamics. We find that the artificial cilia exhibit rich motion behaviors, depending on the mechanical coupling scheme. Particularly, their synchronous beating display two distinct modes - analogous to those observed in C. reinhardtii, the biciliated model organism for studying synchronization. Close examination suggests that the system evolves towards the most dissipative mode. Using this guideline in both simulations and experiments, we are able to direct the system into a desired state by altering the modes' respective dissipation. Our results have significant implications in understanding the synchronization of cilia.

Abstract:We explore a generic mechanism whereby a droplet of active matter acquires motility by the spontaneous breakdown of a discrete symmetry. The model we study offers a simple representation of a "cell extract" comprising, e.g., a droplet of actomyosin solution. (Such extracts are used experimentally to model the cytoskeleton.) Actomyosin is an active gel whose polarity describes the mean sense of alignment of actin fibres. In the absence of polymerization and depolymerization processes ('treadmilling'), the gel's dynamics arises solely from the contractile motion of myosin motors; this should be unchanged when polarity is inverted. Our results suggest that motility can arise in the absence of treadmilling, by spontaneous symmetry breaking (SSB) of polarity inversion symmetry. Adapting our model to wall-bound cells in two dimensions, we find that as wall friction is reduced, treadmilling-induced motility falls but SSB-mediated motility rises. The latter might therefore be crucial in three dimensions where frictional forces are likely to be modest. At a supra-cellular level, the same generic mechanism can impart motility to aggregates of non-motile but active bacteria; we show that SSB in this (extensile) case leads generically to rotational as well as translational motion.

Abstract:Eukaryotic cells and intracellular pathogens such as bacteria or viruses utilize the actin polymerization machinery to propel themselves forward. Thereby, the onset of motion and choice of direction may be the result of a spontaneous symmetry-breaking or might be triggered by external signals and preexisting asymmetries, e.g. through a previous septation in bacteria. Although very complex, a key feature of cellular motility is the ability of actin to form dense polymeric networks, whose microstructure is tightly regulated by the cell. These polar actin networks produce the forces necessary for propulsion but may also be at the origin of a spontaneous symmetry-breaking. Understanding the exact role of actin dynamics in cell motility requires multiscale approaches which capture at the same time the polymer network structure and dynamics on the scale of a few nanometers and the macroscopic distribution of elastic stresses on the scale of the whole cell. In this chapter we review a selection of theories on how mechanical material properties and growth processes interact to induce the onset of actin based motion.

Abstract:Self-propulsion and navigation due to the sensing of environmental conditions - such as durotaxis and chemotaxis - are remarkable properties of biological cells that cannot be reproduced by single-component self-propelled particles. We introduce and study "flexocytes", deformable vesicles with enclosed attached self-propelled pushing and pulling filaments that align due to steric and membrane-mediated interactions. Using computer simulations in two dimensions, we show that the membrane deforms under the propulsion forces and forms shapes mimicking motile biological cells, such as keratocytes and neutrophils. When interacting with walls or with interfaces between different substrates, the internal structure of a flexocyte adapts, resulting in a preferred angle of reflection or deflection, respectively. We predict a correlation between motility patterns, shapes, characteristics of the internal forces, and the response to micropatterned substrates and external stimuli. We propose that engineered flexocytes with desired mechanosensitive capabilities enable the construction of soft-matter robots.

Abstract:When tiny soft ferromagnetic particles are placed along a liquid interface and exposed to a vertical magnetic field, the balance between capillary attraction and magnetic repulsion leads to self-organization into well-defined patterns. Here, we demonstrate experimentally that precessing magnetic fields induce metachronal waves on the periphery of these assemblies, similar to the ones observed in ciliates and some arthropods. The outermost layer of particles behaves like an array of cilia or legs whose sequential movement causes a net and controllable locomotion. This bioinspired many-particle swimming strategy is effective even at low Reynolds number, using only spatially uniform fields to generate the waves.

Encoding spatiotemporal asymmetry in artificial cilia with a ctenophore-inspired soft-robotic platform

Abstract:A remarkable variety of organisms use metachronal coordination (i.e., numerous neighboring appendages beating sequentially with a fixed phase lag) to swim or pump fluid. This coordination strategy is used by microorganisms to break symmetry at small scales where viscous effects dominate and flow is time-reversible. Some larger organisms use this swimming strategy at intermediate scales, where viscosity and inertia both play important roles. However, the role of individual propulsor kinematics - especially across hydrodynamic scales - is not well-understood, though the details of propulsor motion can be crucial for the efficient generation of flow. To investigate this behavior, we developed a new soft robotic platform using magnetoactive silicone elastomers to mimic the metachronally coordinated propulsors found in swimming organisms. Furthermore, we present a method to passively encode spatially asymmetric beating patterns in our artificial propulsors. We investigated the kinematics and hydrodynamics of three propulsor types, with varying degrees of asymmetry, using Particle Image Velocimetry and high-speed videography. We find that asymmetric beating patterns can move considerably more fluid relative to symmetric beating at the same frequency and phase lag, and that asymmetry can be passively encoded into propulsors via the interplay between elastic and magnetic torques. Our results demonstrate that nuanced differences in propulsor kinematics can substantially impact fluid pumping performance. Our soft robotic platform also provides an avenue to explore metachronal coordination at the meso-scale, which in turn can inform the design of future bioinspired pumping devices and swimming robots.

Abstract:Spontaneous locomotion is a common feature of most metazoan cells, generally attributed to the properties of actomyosin networks. This force-producing machinery has been studied down to the most minute molecular details, especially in lamellipodium-driven migration. Nevertheless, how actomyosin networks work inside contraction-driven amoeboid cells still lacks unifying principles. Here, using stable motile blebs from HeLa cells as a model amoeboid motile system, we imaged the dynamics of the actin cortex at the single filament level and revealed the co-existence of three distinct rheological phases. We introduce "advected percolation," a process where rigidity percolation and active advection synergize, spatially organizing the actin network's mechanical properties into a minimal and generic locomotion mechanism. Expanding from our observations on simplified systems, we speculate that this model could explain, down to the single actin filament level, how amoeboid cells, such as cancer or immune cells, can propel efficiently through complex 3D environments.
Minimal actuation and control of a soft hydrogel swimmer from flutter instability

Abstract:Micro-organisms propel themselves in viscous environments by the periodic, nonreciprocal beating of slender appendages known as flagella. Active materials have been widely exploited to mimic this form of locomotion. However, the realization of such coordinated beating in biomimetic flagella requires complex actuation modulated in space and time. We prove through experiments on polyelectrolyte hydrogel samples that directed undulatory locomotion of a soft robotic swimmer can be achieved by untethered actuation from a uniform and static electric field. A minimal mathematical model is sufficient to reproduce, and thus explain, the observed behavior. The periodic beating of the swimming hydrogel robot emerges from flutter instability thanks to the interplay between its active and passive reconfigurations in the viscous environment. Interestingly, the flutter-driven soft robot exhibits a form of electrotaxis whereby its swimming trajectory can be controlled by simply reorienting the electric field. Our findings trace the route for the embodiment of mechanical intelligence in soft robotic systems by the exploitation of flutter instability to achieve complex functional responses to simple stimuli. While the experimental study is conducted on millimeter-scale hydrogel swimmers, the design principle we introduce requires simple geometry and is hence amenable for miniaturization via micro-fabrication techniques. We believe it may also be transferred to a wider class of soft active materials.

Abstract:This research describes an initial exploration of the phenomenon of caregivers' learning of their child's detention, an area of exploration that has little previous attention in the literature. This initial exploration was rooted in a desire to gain an understanding of the caregiver's experience with both their family member (adjudicated juvenile) and the criminal justice system. A qualitative, phenomenological approach was used in this study to gain rich understanding into the lived experiences shared by twelve caregivers of youth being adjudicated in the juvenile justice system. Two overarching themes emerged from the data: 1) the system, with communication, fairness, and substitute parental provider as notable subthemes, and 2) the family, with family strength and relationship to support needs as a notable subtheme. Gaining insight into this critical point of family crisis has implications for intervention frameworks addressing family stability and support for long-term outcomes.

Abstract:Nonlinear biomolecular interactions on membranes drive membrane remodeling crucial for biological processes including chemotaxis, cytokinesis, and endocytosis. The complexity of biomolecular interactions, their redundancy, and the importance of spatiotemporal context in membrane organization impede understanding of the physical principles governing membrane mechanics. Developing a minimal in vitro system that mimics molecular signaling and membrane remodeling while maintaining physiological fidelity poses a major challenge. Inspired by chemotaxis, we reconstructed chemically regulated actin polymerization inside vesicles, guiding membrane self-organization. An external, undirected chemical input induced directed actin polymerization and membrane deformation uncorrelated with upstream biochemical cues, suggesting symmetry breaking. A biophysical model incorporating actin dynamics and membrane mechanics proposes that uneven actin distributions cause nonlinear membrane deformations, consistent with experimental findings. This protocellular system illuminates the interplay between actin dynamics and membrane shape during symmetry breaking, offering insights into chemotaxis and other cell biological processes.

Abstract:Cytoskeletal networks, which are essentially motor-filament assemblies, play a major role in many developmental processes involving structural remodeling and shape changes. These are achieved by nonequilibrium self-organization processes that generate functional patterns and drive intracellular transport. We construct a minimal physical model that incorporates the coupling between nonlinear elastic responses of individual filaments and force-dependent motor action. By performing stochastic simulations we show that the interplay of motor processes, described as driving anti-correlated motion of the network vertices, and the network connectivity, which determines the percolation character of the structure, can indeed capture the dynamical and structural cooperativity which gives rise to diverse patterns observed experimentally. The buckling instability of individual filaments is found to play a key role in localizing collapse events due to local force imbalance. Motor-driven buckling-induced node aggregation provides a dynamic mechanism that stabilizes the two dimensional patterns below the apparent static percolation limit. Coordinated motor action is also shown to suppress random thermal noise on large time scales, the two dimensional configuration that the system starts with thus remaining planar during the structural development. By carrying out similar simulations on a three dimensional anchored network, we find that the myosin-driven isotropic contraction of a well-connected actin network, when combined with mechanical anchoring that confers directionality to the collective motion, may represent a novel mechanism of intracellular transport, as revealed by chromosome translocation in the starfish oocyte.
