Abstract:In a series of laboratory experiments, we presented carnivorous Macrobiotus richtersi (Tardigrada, Macrobiotidae) with nematode prey to assess their importance as predator. We investigated consumption rate for (a) different prey densities (10–400 prey individuals), (b) different prey biomasses (22–80 ng), (c) different prey species (Pelodera teres, Rhabditidae, versus Acrobeloides nanus, Cephalobidae) and (d) different environments (2-D agar surface versus 3-D sand fractions of three different textures). M. richtersi consumed up to 4.6 μg nematode prey in 4 h, that is, 43% of the tardigrade’s body mass. Predation rate was positively correlated with prey density. The optimal prey in the present investigation was the biggest prey because it yielded the highest biomass uptake per time. In addition, the size of M. richtersi played an important role in consumption rate. Bacterivorous nematodes reacted differently to attack. Even in a water film on stiff agar where nematode agility was limited, a vigorous undulation reaction of P. teres led to a measurable reduction in consumption rate. A. nanus, in contrast, showed little response to attack. Microcosm experiments with sands of different particle size demonstrated that M. richtersi is able to chase and consume small bacterivorous nematodes in a 3-D soil matrix. However, consumption rate in sand microcosms was significantly reduced compared with pure agar. The sand matrix improved nematode agility and possibly provided small pores as refuge for the nematodes. The lowest consumption rate was observed in fine sand. Effects of predatory tardigrades on nematode numbers in the field are discussed.

Abstract:Behaviors are adaptive traits evolving through natural selection. Crucially, the genetic, molecular, and neural modifications that shape behavioral innovations are poorly understood. Here, we identify specialized adaptations linked to the evolution of aggression in the predatory nematode Pristionchus pacificus. Using machine learning, we identified robust behavioral states associated with aggressive episodes. These depend on modifications to the invertebrate noradrenergic pathway, with octopamine promoting predatory bouts, and tyramine antagonistically suppressing predation. Strikingly, aggression coincides with rewiring of key circuits across nematode evolution. We find additional octopaminergic neurons with morphological adaptations, including neurites extending to teeth-like structures, and expanded receptor expression throughout head sensory neurons gating prey detection. Thus, evolutionary adaptations in noradrenergic circuits facilitated the emergence of aggressive behavioral states associated with complex predatory traits.

Abstract:ABSTRACT Herbivore natural enemies, including predators, parasitoids and parasites, protect plants by regulating herbivore populations. Some parasites can increase their transmission efficiency by manipulating host behavior. Whether natural enemies can manipulate herbivore behavior to increase top-down control, however, is unknown. Here, we investigate if and how the entomopathogenic nematode Heterorhabditis bacteriophora, an important biocontrol agent, modulates the behavior of the western corn rootworm, Diabrotica virgifera virgifera, a major maize pest, and how these behavioral changes affect the capacity of the nematode to control the rootworm. We found that healthy rootworm larvae are attracted to nematode-infected cadavers shortly before the emergence of the next generation of nematodes. Nematode-infected rootworms release distinct volatile bouquets, including butylated hydroxytoluene (BHT), which attracts rootworms to infected cadavers. In a soil setting, BHT attracts rootworms and reduces nematode resistance, resulting in increased infection rates and rootworm mortality as well as increased nematode reproductive success. Five out of seven tested insect species were found to be attracted to nematode-infected conspecifics, suggesting that attraction of healthy hosts to nematode-infected cadavers is widespread. This study reveals a new facet of the biology of entomopathogenic nematodes that increases their capacity to control a major root pest by increasing the probability of host encounters.

Abstract:Nematodes are one of the most diverse groups of organisms, found in various environments, and exhibit various feeding behaviours. displays two types of feeding behaviours: predatory and bacterial feeding. Previously, we showed that the neurotransmitter serotonin plays important roles in regulating predatory feeding in . However, the role of other neurotransmitters in predatory feeding remains unclear. Using the CRISPR/Cas9 system, we generated mutants of the dense-core vesicle (DCV)-based neuropeptide secretion system and genes related to seven neurotransmitters. Predatory assays revealed that the vesicular glutamate transporter -EAT-4, glutamic acid decarboxylase -UNC-25 which is involved in GABA synthesis, and the calcium-dependent activator protein for DCV secretion -UNC-31, play roles in predatory feeding behaviour. We assessed the pharyngeal movement necessary for predation as well as locomotion rate in these mutants. While the and mutants decreased predation movement, the mutant showed a reduction only in bacterial events compared to wild type animals. Additionally, and decreased in motor movement, potentially reducing predation efficiency. Together, these results suggest that glutamate, GABA and DCV secretion modulate feeding behaviours in . Our mutant collection of neurotransmitter-related genes will be useful for future analysis of neurobiology and behavioural evolution.

Abstract:Interactions between predator species shape ecological communities and are crucial in the relationship between biodiversity and ecosystem functioning. They contribute to improving the natural biological control. Cyrtorhinus lividipennis Reuter and Tytthus chinensis Stal (Hemiptera: Miridae) are important natural enemies of brown planthopper, Nilaparvata lugens Stal (Hemiptera: Delphacidae), the most destructive rice pest in Asia. Comparative functional responses of both sexes of the two mirids at different development stages were estimated and the direction, symmetry, as well as intensity of intraguild predation (IGP) were studied in the laboratory. Prey preferences of the two mirid species and the influence of alternative prey on functional responses were also evaluated. The mirids exhibited reciprocal IGP in addition to consuming eggs of BPH. All predator-prey combinations exhibited a type II functional response. Females and fourth instar nymphs of C. lividipennis showed greater voracity for eggs of BPH than of T. chinensis. The two mirids were more voracious to intraguild prey than to extraguild prey. Alternative prey released the functional responses of C. lividipennis to both intra-and extraguild prey. In contrast, alternative prey had no effect on the functional response of T. chinensis. RIGP was symmetrical in the absence of extraguild prey. And in the presence of extraguild prey that was common in the field, resulted in different responses by the mirids.

Abstract:All animals live in tight association with complex microbial communities, yet studying the effects of individual bacteria remains challenging. Here, we exploit the system of the bacterial feeding nematode Pristionchus pacificus, where specific interactions can be studied in isolation. We sequenced the genomes of 84 Pristionchus-associated bacteria to investigate how differences in bacterial metabolic potential affect nematode traits. Specifically, we identified the Paerucumarin biosynthesis pathway as a strong candidate for nematode survival. Hypothesizing that bacterial production of coumarin derivatives contributed to increased lethality, we demonstrated nematicidal effects of two related compounds using supplementation assays. To explore metabolic interactions at a broader scale, we generated nematode transcriptomes for 38 bacterial diets and constructed a coexpression network that revealed distinct developmental and environmental signatures. Interspecies association studies resulted in a bipartite network with more than 2,800 interactions which may assist future studies to identify metabolites affecting various biological processes.

Behavior manipulation of mosquitoes by a mermithid nematode

Abstract:We examined manipulation of mosquito behavior by the parasitic mermithid nematode, Strelkovimermis spiculatus. This nematode species typically infects early instar host larvae and emerges after parasitic development to kill last-instar larvae. Parasitized adults, however, have occasionally been reported from field collections. We obtained low rates (1.7-11.5%) of parasitized adults in laboratory exposures only when Culex pipiens pipiens fourth-instar larvae nearing pupation were exposed to infective nematodes. This did not allow an adequate interval for parasitic development in immature host stages. Parasitized adult females in a multiple-choice assay were three times more likely to seek water than a blood source (63.1 vs. 20.5%), whereas uninfected females were twice as likely to seek blood than water (64%3.9 vs. 32.6%). This altered host behavior benefits the parasite by providing the only mechanism for dispersal and colonization of new host habitats while concurrently avoiding risks from the defensive behaviors associated with blood-feeding. Behavioral alternation in Cx. p. pipiens larval hosts was also examined using larvae infected as second instars to allow for a normal duration of parasitic development. As larvae neared pupation and parasite emergence, parasitized larvae became more spatially aggregated than unparasitized larvae. This altered host behavior benefits the parasite by providing a corresponding increase in post-parasite aggregation, which facilitates formation of large mating clusters and concomitantly reproductive success. Parasites derive fitness gains by overriding host autonomy, whereas hosts have zero fitness once parasitism is established, suggesting a coevolutionary response is inoperative and that the behavioral modifications may be adaptive.

Abstract:Prey must balance the need to avoid predators with the need to feed, a dilemma central to prey refuge theory. Additionally, prey must also assess predatory imminence, or how close predator threats are in space and time. Predatory imminence theory classifies defensive behaviors into three defense modes—pre-encounter, post-encounter, and circa-strike—each corresponding to increasing levels of predatory imminence—suspecting a predator, detecting a predator, and contact with a predatory attack. Although prey often simultaneously face variations in predatory imminence and spatial distribution of predation risks, research on how these factors intersect to influence defensive behaviors has been limited. Integrating these factors into a complex, naturalistic environment could enable comprehensive analysis of multiple defense modes in consistent conditions within the same study, unlike laboratory tests designed to examine only one mode at a time. Here, we combine prey refuge and predatory imminence theories to develop a model system of nematode defensive behaviors, with as prey and as predator. We show that innately exhibits circa-strike behaviors in a foraging environment comprised of a food-rich, high-risk patch and a food-poor, predator-free refuge. However, after extended experience in this environment, acquires post- and pre-encounter behaviors that proactively anticipate threats rather than merely reacting to attacks. We also demonstrate that these defense modes are potentiated by increasingly harmful predators, with only life-threatening predators capable of eliciting all three defense modes. Finally, our model system reveals that SEB-3 receptors and NLP-49 peptides, key to stress response regulation, vary in their impact and interdependence across defense modes. We find that SEB-3 has a greater impact on the highest-imminence defense mode, while NLP-49 peptides have a stronger effect on the lowest-imminence defense mode. Overall, our model system reveals detailed and comprehensive insights into how stress-related molecular signaling affects behavioral responses to threats.

Abstract:Plants defend themselves against herbivores through the production of toxic and deterrent metabolites. Adapted herbivores can tolerate and sometimes sequester these metabolites, allowing them to feed on defended plants and become toxic to their own enemies. Can herbivore natural enemies overcome sequestered plant defense metabolites to prey on adapted herbivores? To address this question, we studied how entomopathogenic nematodes cope with benzoxazinoid defense metabolites that are produced by grasses and sequestered by a specialist maize herbivore, the western corn rootworm. We find that nematodes from US maize fields in regions in which the western corn rootworm was present over the last 50 y are behaviorally and metabolically resistant to sequestered benzoxazinoids and more infective toward the western corn rootworm than nematodes from other parts of the world. Exposure of a benzoxazinoid-susceptible nematode strain to the western corn rootworm for 5 generations results in higher behavioral and metabolic resistance and benzoxazinoid-dependent infectivity toward the western corn rootworm. Thus, herbivores that are exposed to a plant defense sequestering herbivore can evolve both behavioral and metabolic resistance to plant defense metabolites, and these traits are associated with higher infectivity toward a defense sequestering herbivore. We conclude that plant defense metabolites that are transferred through adapted herbivores may result in the evolution of resistance in herbivore natural enemies. Our study also identifies plant defense resistance as a potential target for the improvement of biological control agents.
Predation by nematode-trapping fungus triggers mechanosensory-dependent quiescence in C. elegans

Abstract:Predation can induce behavioral changes in prey, yet the molecular and neuronal mechanisms underlying prey responses remain poorly understood. Here, we investigated how the nematode Caenorhabditis elegans responds to predation by the nematode-trapping fungus, Arthrobotrys oligospora. We found that A. oligospora predation induced quiescence in C. elegans showing rapid cessation of pharyngeal pumping and movement. Calcium imaging revealed that this quiescence was regulated by the activation of sleep-promoting neurons, ALA and RIS. Genetic analyses demonstrated that ALA were essential for pharyngeal pumping inhibition, whereas both ALA and RIS contributed to movement cessation. Transcriptomic analysis in C. elegans showed the upregulation of immune defense genes in response to A. oligospora predation. We demonstrated that mechanosensation was required for pumping inhibition and transcriptomic regulation upon A. oligospora trapping. These findings suggest that physical constraints imposed by fungal traps trigger a stress-induced quiescence and the upregulation of defense genes in C. elegans. We suggest that trapping-induced quiescence might be a predation strategy used by sessile predators to prevail in the evolutionary arms race.

Abstract:Self-recognition is observed abundantly throughout the natural world, regulating diverse biological processes. Although ubiquitous, often little is known of the associated molecular machinery, and so far, organismal self-recognition has never been described in nematodes. We investigated the predatory nematode Pristionchus pacificus and, through interactions with its prey, revealed a self-recognition mechanism acting on the nematode surface, capable of distinguishing self-progeny from closely related strains. We identified the small peptide SELF-1, which is composed of an invariant domain and a hypervariable C terminus, as a key component of self-recognition. Modifications to the hypervariable region, including single–amino acid substitutions, are sufficient to eliminate self-recognition. Thus, the P. pacificus self-recognition system enables this nematode to avoid cannibalism while promoting the killing of competing nematodes.

Predatory interactions between prey affect patch selection by predators

Abstract:Abstract: When predators can use several prey species as food sources, they are known to select prey according to foraging efficiency and food quality. However, interactions between the prey species may also affect prey choice, and this has received limited attention. The effect of one such interaction, intraguild predation between prey, on patch selection by predators was studied here. The predatory mite Neoseiulus californicus preys on young larvae of the western flower thrips Frankliniella occidentalis and on all stages of the two-spotted spider mite Tetranychus urticae. The two prey species co-occur on several plant species, on which they compete for resources, and western flower thrips feed on eggs of the spider mites. A further complicating factor is that the thrips can also feed on the eggs of the predator. We found that performance of the predatory mite was highest on patches with spider mites, intermediate on patches with spider mites plus thrips larvae and lowest on patches with thrips larvae alone. Patch selection and oviposition preference of predators matched performance: predators preferred patches with spider mites over patches with spider mites plus thrips. Patches with thrips only were not significantly more attractive than empty patches. We also investigated the cues involved in patch selection and found that the attractiveness of patches with spider mites was significantly reduced by the presence of cues associated with killed spider mite eggs. This explains the reduced attractiveness of patches with both prey. Our results point at the importance of predatory interactions among prey species for patch selection by predators. Significance statement: Patch selection by predators is known to be affected by factors such as prey quality, the presence of competitors and predators, but little is known on the effects of interactions among prey species present on patch selection. In this paper, we show that patch selection by a predator is affected by such interactions, specifically by the feeding of one prey species on eggs of the other.

Abstract:Herbivore natural enemies protect plants by regulating herbivore populations. Whether they can alter the behavior of their prey to increase predation success is unknown. Coping with antagonistic predation risks: Predator‐dependent unique responses are dominant in Ceriodaphnia cornuta

Abstract:Inducible defenses of prey are evolved under diverse and variable predation risks. However, during the co‐evolution of prey and multiple predators, the responses of prey to antagonistic predation risks, which may put the prey into a dilemma of responding to predators, remain unclear. Based on antagonistic predation pressure from an invertebrate (Chaoborus larvae) and a vertebrate (Rhodeus ocellatus) predator, we studied the responses of multiple traits and transcriptomes of the freshwater crustacean Ceriodaphnia cornuta under multiple predation risks. Chaoborus predation risk altered the expression of genes encoding cuticle proteins and modulated the biosynthesis of steroid hormones, cutin, suberine, and wax, leading to the development of horns and increase in size at the late developmental stage. Meanwhile, fish predation risk primarily triggered genes encoding ribosomes and those involved in unsaturated fatty acid biosynthesis and cysteine and methionine metabolism, resulting in smaller individual size and earlier reproduction. Inducible responses of both transcriptome and individual traits revealed that predator‐dependent unique responses were dominant and the dilemma of antagonistic responses was relatively limited. However, the unique individual traits in response to invertebrate predation could be significantly impaired by vertebrate predation risk, even though the unique responses to different predators were extremely weakly correlated and could be elicited simultaneously. These results indicate that diverse predator‐dependent unique responses are favored by Ceriodaphnia during its co‐evolution with multiple predators. Nonetheless, Ceriodaphnia is not a generalist that can fully adopt all predator‐dependent unique responses simultaneously under multiple predation risks.

Social predation by a nudibranch mollusc

Abstract:Social predation is a common strategy used by predators to subdue and consume prey. Animals that use this strategy have many ways of finding each other, organizing behaviors and consuming prey. There is wide variation in the extent to which these behaviors are coordinated and the stability of individual roles. This study characterizes social predation by the nudibranch mollusc, Berghia stephanieae, which is a specialist predator that eats only the sea anemone, Exaiptasia diaphana. A combination of experimental and modeling approaches showed that B. stephanieae does predate upon E. diaphana in groups. The extent of social feeding was not altered by length of food deprivation, suggesting that animals are not shifting strategies based on internal state. It was unclear what cues the individual Berghia used to find each other; choice assays testing whether they followed slime trails, were attracted to injured anemones, or preferred conspecifics feeding did not reveal any cues. Individuals did not exhibit stable roles, such as leader or follower, rather the population exhibited fission-fusion dynamics with temporary roles during predation. Thus, the Berghia provides an example of a specialist predator of dangerous prey that loosely organizes social feeding, which persists across hunger states and uses temporary individual roles; however, the cues that it uses for aggregation are unknown.

Abstract:Behavioral characteristics such as aggressiveness and predation preference of the phytoseiid predatory species, Amblyseius swirskii, Neoseiulus californicus and Phytoseiulus persimilis (Acari: Phytoseiidae) were investigated when applied together. We quantified the chance and time of a successful attack to a heterospecific larva as aggressiveness of adult females. In order to the determine the predation preference of adult females equal combination of con/heterospecific larvae were presented to as prey in absence of their main prey ( Tetranychus urticae Koch) for 24 h. Experiments were carried out in growth chamber at 25±1 oC, 65±5% RH and 16:8 (L: D) hour photoperiod regimens. The results showed that adult females of N. californicus needed 39.65±7.46 min. to attack and kill the larva of P. persimilis which was remarkably less than that recorded (227.80±28.38 min.) for adult females of P. persimilis to attack and kill N. californicus larva. N. californicus was estimated more aggressive than P. persimilis . The mean estimated attack time of A. swirskii and P. persimilis was estimated 18.55±2.89 and 201.70±25.42 min., respectively. A. swirskii was considered more aggressive than P. persimilis against heterospecific larvae. The predation preference index (Manly´s β ) were indicated that N. californicus was able to recognize con/heterospecific larva, resulted more preference for heterospecific larvae ( P. persimilis ) in absence of T. urticae , while adult P. persimilis showed no preference (between con and heterospecific larva). Manly´s β , for A. swirskii was 0.706 and 0.294, for P. persimilis 0.369 and 0.630 on hetero and conspecific larvae, respectively. Results of this study showed that, A. swirskii and N. californicus are general predators were able to roecgnize con/heterospecific larva and preferred to feed on heterospecific larvae and get benefit from intraguild predation.

Insights into the importance of nematode prey for chironomid larvae

Abstract:The potential predatory ability of Chironomus riparius, well-known detritus-feeding chironomid larvae, on nematodes was investigated in a series of laboratory experiments. We analyzed the consumption rate (i) for different prey densities of adult and juvenile Caenorhabditis elegans (25–1200 and 25–2400 prey individuals, respectively), (ii) for different substrates (detritus, sand, and gravel), and (iii) with a natural nematode community as prey. This is the first study to demonstrate a clear predator–prey interaction between chironomids and nematodes. The number of ingested C. elegans increased with increasing prey density. Within 4 h, C. riparius consumed up to 0.77 mg of nematode prey (557 adult individuals), which corresponded to 92.5% of the chironomid’s biomass, and as many as 763 juvenile C. elegans (0.0226 mg; 3.2% of chironomid biomass). Significantly more nematodes were devoured in fine sediment (sand and detritus) than in gravel. Within a natural assemblage, C. riparius consumed 41% of the total nematodes, preferentially those 1- to 2-mm in length; thus, for example, only the percentage of the most common species, Trischistoma monohystera, was significantly reduced. Gut analyses of the chironomids confirmed the ingestion of nematodes. In general, this study provides a fundamental approach to our understanding of the benthic food web.

Abstract:How bacterial feeding fauna affects colonization and survival of bacteria in soil is not well understood, which constrains the applicability of bacterial inoculants in agriculture. This study aimed to unravel how food quality of bacteria and bacterial feeders with different feeding habits (the selective feeding flagellate Cercomonas longicauda versus the non-selective feeding nematode Caenorhabditis elegans) influence the abundance of two bacteria that compete for resources in simple model communities. Microcosms consisted of either one gfp-tagged bacterial strain (Pseudomonas fluorescens DSM50090 or one of two biocontrol strains P. fluorescens CHA0 or Pseudomonas sp. DSS73) or combinations of two bacterial strains. DSM50090 is a suitable food bacterium, DSS73 is of intermediate food quality, and CHA0 is inedible to the bacterial feeders. Bacterial and protozoan cell numbers were measured by flow cytometry. In the presence of flagellates, CHA0 increased its abundance as compared to the other biocontrol strain DSS73 or to DSM50090, which were both eaten by the flagellates. In contrast, the number of CHA0 declined as compared to DSS73 when the model community was subjected to nematode predation pressure. Hence, the results suggested that the outcome of competition among bacteria depended on their ability to cope with the prevailing bacterial predator.

Abstract:Understanding the mechanisms underlying the success of biological invasions is essential to employ effective prediction and management strategies. Escape from natural enemies in invaded regions (enemy release hypothesis, ERH) and increased competitive ability are hallmarks of invasive species; however, these two processes are rarely studied within the same context. Here, we examined the effect of enemy release on the competition outcomes of a successful invasive insect pest in North America, the African fig fly (Zaprionus indianus). Parasitoid wasps such as Leptopilina heterotoma that parasitize drosophilid larvae may seek out established species with known host suitability over a novel species, so we hypothesized Z. indianus may have low susceptibility to parasitoids, giving them a competitive advantage over co-occurring drosophilids. We tested this hypothesis by comparing the adult emergence rates from Z. indianus larvae reared alone or in competition with Drosophila hydei or D. simulans larvae in the presence and absence of parasitoid wasps under low and high larval densities. At low larval densities, Z. indianus emerged at equal rates to D. hydei but outcompeted D. simulans, and these outcomes were not affected by parasitoids. However, at high densities, the addition of parasitoids shifted competition outcomes in favor of Z. indianus, suggesting enemy release provides a competitive advantage under some circumstances. These results indicate that the strength of enemy release in Z. indianus is widely dependent on contextual factors such as density and competitor species. This study emphasizes how a community approach to testing the ERH is vital as the overall interpretation of the presence and strength of enemy release differed between intraspecific and interspecific experiments. Further investigation of how these results apply to field environments could offer insight into how Z. indianus alters ecosystems and how productive biological control may limit the spread of Z. indianus.

Abstract:Nematode-trapping fungi develop complex trapping devices to capture and consume nematodes. The dynamics of these organisms is especially important given the pathogenicity of nematodes and, consequently, the potential application of nematode-trapping fungi as biocontrol agents. Furthermore, both the nematodes and nematode-trapping fungi can be easily grown in laboratories, making them a unique manipulatable predator-prey system to study their coevolution. Several different aspects of these fungi have been studied, such as their genetics and the different factors triggering trap formation. In this review, we use the nematode-trapping fungus Arthrobotrys oligospora (which forms adhesive nets) as a model to describe the trapping process. We divide this process into several stages; namely attraction, recognition, trap formation, adhesion, penetration, and digestion. We summarize the latest findings in the field and current knowledge on the interactions between nematodes and nematode-trapping fungi, representing both sides of the predator-prey interaction.
