Abstract:Photosynthetic microorganisms in microalgal-bacterial granular sludge offer advantages in wastewater treatment processes. This study examined the effects of light intensity and salinity on microalgal-bacterial granular sludge formation and microbial changes. Activated sludge was inoculated into three bioreactors and operated in batch treatment mode for 100 days under different light intensities (0, 60, and 120 μmol m −2 s −1 ) and staged increases in salinity concentration (0, 1, 2, and 3%). Results showed that microalgal-bacterial granular sludge was successfully formed within 30 days, and high light exposure increased algal particle stability and inorganic nitrogen removal (63, 66, 71%), while chemical oxygen demand removal (>95%) was similar across groups. High-throughput sequencing results showed that the critical algae were Chlorella and diatoms, while the main bacteria included Paracoccus and Xanthomarina with high extracellular polymeric substance production. This study aims to enhance the comprehension of MBGS processes in saline wastewater treatment under varying light intensities.

Abstract:A new algal-bacterial granular sludge treatment method was used to treat salty eutrophication water. The results indicated that the treatment removed more than 98% of the total inorganic nitrogen and the total phosphorus after a 15 d cultivation period using 2% salinity simulated eutrophication water. For the 4% salinity simulated water, the total phosphorus was not able to be removed and was even higher in the effluent; and the total inorganic nitrogen was only removed 17%. Thus, the algal-bacterial granules were efficient for removing nitrogen and phosphorus in 2% salinity eutrophication water but were not effective for 4% salinity water. High levels of filamentous algae proliferation growing on the surface of the granules was primarily responsible for the good performance in 2% salinity water. However, the lipid accumulation was greatly enhanced (reactor R2 at a 27.6% increase and reactor R4 at a 107% increase) for both granule types due to the algal growth. Thus, treatment of the salty eutrophication water can also greatly increase the added-value of the algal-bacterial granules.

Abstract:Algal-bacterial granular sludge (ABGS) composed of microalgae and aerobic granular sludge, is a sustainable and promising technology for wastewater treatment. However, the formation mechanism of ABGS has not been clearly defined, and the direct formation of ABGS in saline wastewater has rarely been investigated. This study proposed novel insights into the granulation process of ABGS by assembling the algal barrier, which was successfully cultivated directly in saline wastewater. The results concluded that ABGS with the algal barrier maintained a higher biomass (MLSS of 7046 ± 61 mg/L), larger particle sizes (1.21 ± 0.06 mm), and better settleability (SVI30 of 46 ± 1 mL/g), enabling efficient pollutants removal. Soluble microbial products (SMP) were found to be closely related to the emergence of the algal barrier. In addition, under salinity stress, the high production of extracellular polymeric substances (EPS, 133.70 ± 1.40 mg/g VSS), specifically TB-EPS (90.29 ± 1.12 mg/g VSS), maintained a crucial role in the formation of ABGS. Further analysis indicated that biofilm producing bacteria Pseudofulvimonas and filamentous eukaryote Streptophyta were the key players in ABGS formation with the algal barrier. Furthermore, the enhancement of key genes and enzymes involved in nitrogen metabolism, TCA cycle, and polysaccharide metabolism suggested a more robust protective effect provided by the algal barrier. This study is expected to advance the application of simultaneous ABGS formation and pollutant removal in wastewater.

Abstract:The study investigated the response characteristics of algal-bacterial granular sludge (ABGS) under salinity stress (0 % → 2 %). At 1 % salinity, the sludge performance was inhibited, while recovered rapidly, indicating the ABGS exhibited resistance. However, at 2 % salinity, the suppressed performances did not recover until the stress was eliminated. Under salinity stress, the nutrient removal capacity of the system and the composition and chemical characteristics of extracellular polymers substances also changed. Meanwhile, the ABGS formed adaptation to salinity stress in the early coping process. As a result, the effect of the second 2 % salinity on ABGS was significantly weakened. High-throughput sequencing results showed that the microbial community in ABGS shifted under salinity stress, and the halophilic bacteria genera Arcobacter, Denitromonas, Azoarcus, etc. were enriched, which might be the genetic basis of the adaptation.

Abstract:Salt could affect the properties of aerobic granular sludge (AGS) and the biological treatment of wastewater. This study aimed to cultivate aerobic granular sludge under different salinity conditions (0,1 % and 3 %). The performance and microbial community profiles of AGS were studied to explain the effect of salinity. The presence of salinity led to more compact structure and smaller particle size of granular sludge. The experimental results indicated that 3 % salinity could seriously affected NH 4 + -N and TP removal efficiency of the reactor, while the impact on COD was negligible. Extracellular polymeric substance (EPS) played a pivotal role in AGS, especially TB-EPS. A large number of EPS producing bacteria existed in granular sludge regardless of salinity. However, there were significant differences in the AGS strains distribution with different salinities. The absolute dominant strain Thauera was observed at 1 % salinity, while the dominant strain Halomonas was prevailed at 3 % salinity.

Abstract:Summary In the present study, the effects of cefquinome, a 4 generation cephalosporin, on clinical, biochemical, haemato‐logical, and blood gas variables were investigated. Five healthy dogs were injected with cefquinome (1 mg/kg body weight, IM, daily) for 14 days. Negative effects of cefquinome on clinical, biochemical, and haematological variables were not observed, but it did change some blood gas variables.

Abstract:In this study, algae growth was evaluated for its effects on the formation of algae–bacterial granular sludge by comparing aerobic sludge granulation with and without algae. The results revealed that algae initially self-aggregated and subsequently provided attachment carriers for microbial aggregation, resulting in improved biomass growth (4.82 g/L) and sludge settleability (32.32 mL/g). Compared with aerobic granular sludge, algae–bacterial granular sludge was formed approximately 14 days earlier and its size was 15% larger (600 μm). The removal efficiency of COD, NH 4 + -N, TN, and TP in the algae–bacterial granular sludge system was enhanced, reaching 97.55%, 98.19%, 84.88%, and 85.19%, respectively. In addition, algae present in algae–bacterial granular sludge facilitated the production of extracellular polymeric substances and triggered alterations in their chemical composition. Analysis of the protein secondary structures of extracellular polymeric substances revealed that the contents of the β-turn helix and aggregated strands in algae–bacterial granular sludge were 5.56% and 7.01% more than those in aerobic granular sludge during granulation. Additionally, algae–bacterial granular sludge had a higher abundance of Gammaproteobacteria (46.09%). Altogether, this study provides a theoretical basis for understanding the formation of ABGS.

Abstract:The algal-bacterial granular sludge (ABGS) system is a prospective wastewater treatment technology, but few studies focused on the effects of different inoculum types on the establishment of the ABGS system under low aeration conditions (step-decrease superficial gas velocity from 1.4 to 0.5 cm/s). Results from this study indicated that compared with other inocula, the ABGS formed by co-inoculating aerobic granular sludge (AGS) and targeted algae ( Chlorella ) exhibited a shorter granulation period (shortened by 15 days), higher total nitrogen (89.4%) and PO 4 3- -P (95.0%) removal efficiencies, and a greater yield of fatty acid methyl esters (FAMEs) (9.04 mg/g MLSS). This was possibly attributed to that the functional bacteria (e.g. Thauera , Gemmobacter and Rhodobacter ) in the inoculated AGS facilitated the ABGS granulation. The inoculated algae promoted their effective enrichment under illumination conditions and enhanced the production of extracellular polymeric substances, thus improving the stability of ABGS. The enriched algae were attached to the outer layer of the granules, which could provide sufficient oxygen for bacterial metabolism, revealing the inherent mechanisms for the good stability of ABGS under low aeration intensity. Overall, the rapid granulation of ABGS can be achieved by inoculating optimal inocula under low aeration conditions, which is convenient and economically feasible, and motivates the application of algal-bacterial consortia.

Microalgal-bacterial granular sludge process outperformed aerobic granular sludge process in municipal wastewater treatment with less carbon dioxide emissions

Abstract:The aerobic granular sludge (AGS) process and microalgal-bacterial granular sludge (MBGS) process were comparably applied for municipal wastewater treatment in sequencing batch reactors with a height to diameter ratio of eight. For morphological appearances, the yellow aerobic granules were strip-shaped (4.0 mm × 0.62 mm) while the green microalgal-bacterial granules were elliptical-shaped (2.0 mm × 0.75 mm). The dominated rod-shaped bacteria (e.g., Acidobacteria and Bacteroidetes) and the slender configuration might be associated with the strip shape of aerobic granules under weak acid conditions. The nutrients removal performances by MBGS process were generally slightly better than AGS process. In addition, nutrients removal mechanisms were identified to elucidate how organics, ammonia, and phosphorus were removed by AGS process and MBGS process, respectively. Mass balance calculation estimated that MBGS process appeared to achieve much less CO2 emission (5.8%) compared with AGS process (44.4%). Overall, it proved that MBGS process, with the merits of potentially low energy cost, limited CO2 emission, and excellent performance, showed more prospects in municipal wastewater treatment than AGS process.

Abstract:Long-term stability in practical applications is one of the crucial issues challenging the sustainable operation of aerobic granular sludge (AGS)-based wastewater treatment systems. Numerous attempts have been made to improve its long-term stability, among which introducing microalgae to bacterial AGS systems seems promising. This study investigated and recorded the whole process of disintegration and re-granulation of the aged algal-bacterial AGS in a 42-day operation. The disintegration of granules was signaled by broken structure and more effluent biomass discharge, resulting in deterioration of phosphorus removal from 86 % on day 1 to 7 % on day 20 and some slight decrease in chlorophyll a content. The broken granules were found to be filamentous flocs or granule fragments, and the latter with coexisting microalgae possibly accounted for the re-granulation and system recovery. Large proportions of fragments could be developed into new granules on day 30 under no additional strategy adoption. And the effluent biomass, chlorophyll a content and phosphorus removal can recover to their initial levels within 34 days' operation. Interestingly, organics and ammonia removals were not affected, which kept around 99 % during the whole test period. Results from this study provide a more in-depth understanding of the stability of algal-bacterial AGS and its long-term operation management.

Effects of salinity and betaine addition on anaerobic granular sludge properties and microbial community succession patterns in organic saline wastewater

Abstract:In this study, the effects of different salinity gradients and addition of compatible solutes on anaerobic treated effluent water qualities, sludge characteristics and microbial communities were investigated. The increase in salinity resulted in a decrease in particle size of the granular sludge, which was concentrated in the range of 0.5-1.0 mm. The content of EPS (extracellular polymeric substances) in the granular sludge gradually increased with increasing salinity and the addition of betaine (a typical compatible solute). Meanwhile, the microbial community structure was significantly affected by salinity, with high salinity reducing the diversity of bacteria. At higher salinity, Patescibacteria and Proteobacteria gradually became the dominant phylum, with relative abundance increasing to 13.53% and 12.16% at 20 g/L salinity. Desulfobacterota and its subordinate Desulfovibrio, which secrete EPS in large quantities, dominated significantly after betaine addition.Their relative abundance reached 13.65% and 7.86% at phylum level and genus level. The effect of these changes on the treated effluent was shown as the average chemical oxygen demand (COD) removal rate decreased from 82.10% to 79.71%, 78.01%, 68.51% and 64.55% when the salinity gradually increased from 2 g/L to 6, 10, 16 and 20 g/L. At the salinity of 20 g/L, average COD removal increased to 71.65% by the addition of 2 mmol/L betaine. The gradient elevated salinity and the exogenous addition of betaine played an important role in achieving stability of the anaerobic system in a highly saline environment, which provided a feasible strategy for anaerobic treatment of organic saline wastewater.
Effects of salinity on glycerol conversion and biological phosphorus removal by aerobic granular sludge

Abstract:Industrial wastewater often has high levels of salt, either due to seawater or e.g. sodium chloride (NaCl) usage in the processing. Previous work indicated that aerobic granular sludge (AGS) is differently affected by seawater or saline water at similar osmotic strength. Here we investigate in more detail the impact of NaCl concentrations and seawater on the granulation and conversion processes for AGS wastewater treatment. Glycerol was used as the carbon source since it is regularly present in industrial wastewaters, and to allow the evaluation of microbial interactions that better reflect real conditions. Long-term experiments were performed to evaluate and compare the effect of salinity on granulation, anaerobic conversions, phosphate removal, and the microbial community. Smooth and stable granules as well as enhanced biological phosphorus removal (EBPR) were achieved up to 20 g/L NaCl or when using seawater. However, at NaCl levels comparable to seawater strength (30 g/L) incomplete anaerobic glycerol uptake and aerobic phosphate uptake were observed, the effluent turbidity increased, and filamentous granules began to appear. The latter is likely due to the direct aerobic growth on the leftover substrate after the anaerobic feeding period. In all reactor conditions, except the reactor with 30 g/L NaCl, Ca. Accumulibacter was the dominant microorganism. In the reactor with 30 g/L NaCl, the relative abundance of Ca. Accumulibacter decreased to ≤1 % and an increase in the genus Zoogloea was observed. Throughout all reactor conditions, Tessaracoccus and Micropruina, both actinobacteria, were present which were likely responsible for the anaerobic conversion of glycerol into volatile fatty acids. None of the glycerol metabolizing proteins were detected in Ca. Accumulibacter which supports previous findings that glycerol can not be directly utilized by Ca. Accumulibacter. The proteome profile of the dominant taxa was analysed and the results are further discussed. The exposure of salt-adapted biomass to hypo-osmotic conditions led to significant trehalose and PO43--P release which can be related to the osmoregulation of the cells. Overall, this study provides insights into the effect of salt on the operation and stability of the EBPR and AGS processes. The findings suggest that maintaining a balanced cation ratio is likely to be more important for the operational stability of EBPR and AGS systems than absolute salt concentrations.

Abstract:The effects of salinity shock on the anaerobic treatment of fracturing wastewater regarding chemical oxygen demand (COD) removal performance, sludge characteristics and microbial community were investigated. Results showed COD removal efficiency decreased from 76.0% to 69.1%, 65.6%, 33.7% and 21.9% with the increase of salinity from 2.5 g/L to 10, 15, 25 and 45 g/L, respectively. The cumulative biogas production decreased by 13.8%-81.1% when salinity increased to 15-85 g/L. The increase of salinity led to the decline in particle size of granular sludge, and the activity of granular sludge, including SMA, coenzyme F420 and dehydrogenase, was inhibited significantly. Flow cytometry indicated the percentage of damaged cells in granular sludge gradually increased with the increase of salinity. Sequence analysis illustrated that microbial community structure in anaerobic digestion reactor was influenced by the salinity, high salinity reduced the diversity of archaea and decreased the abundance of methanogens, especially Methanosaeta.

Abstract:Settleability is vital important for the stability of effluent quality from the widely used activated sludge (AS) process in wastewater treatment plants. In this study, filamentous algae and seawater condition were used to build a microalgae-bacteria (MB) consortia system with the objective of improving the settleability of AS. Filamentous algae were found to flourish more in aeration seawater reactor (R2) than non-aeration (R1). During 90 days, large contents of N, P were firstly released from the biomass, but were absorbed by the algae to relatively low concentrations (N 1.1 mg/L, P 1.4–2.3 mg/L). Meanwhile, the settleability of the two types of MB consortia both significantly increased, but was superiorly enhanced in aeration condition to a low 5-min sludge volume (SV 5 ) (11.5%, 55 d). Filamentous algae Leptolyngbya , as the most dominant part (9.8%) of microbial community in R2, could tightly bind the bacteria cells and extracellular polymeric substances (EPS) to form larger particles, which greatly accelerated the sedimentation. This study is of interest for the potential use of sludge settleability promotion in wastewater treatment plants, which has promising prospects for preventing the sludge bulking or biomass loss in AS systems.

Seawater-based wastewater accelerates development of aerobic granular sludge: A laboratory proof-of-concept

Abstract:This study aimed to develop an aerobic granular sludge process for the efficient treatment of highly saline wastewater and understand the granulation process in a seawater-based multi-ion matrix. Five identical sequencing batch airlift reactors (SBARs) are used to treat synthetic saline sewage with different proportions of real seawater (0%-100%). The results confirm that aerobic granular sludge can be successfully developed with various proportions of seawater up to 100% and show that seawater not only significantly accelerates granulation but also generates stronger granular structures than does freshwater. The increased presence of gel-forming alginate-like exopolysaccharides in the granules explains why a greater proportion of seawater leads to higher density and improves the cohesive strength of the granules. SEM-EDX analysis further revealed substantial presence of both Ca2+ and Mg2+ phosphate in the granule core as well as in the outer layers providing extra bridging forces in addition to alginate-like exopolysaccharides for accelerating the granule formation and maintaining the structure. It is hoped that this work could explore another approach for saline sewage treatment and bring some clues for the mystery of granulation mechanism.
Optimal algae species inoculation strategy for algal-bacterial granular sludge: Sludge characteristics, performance and microbial community

Abstract:The algal-bacterial granular sludge (ABGS) system is emerging as a promising technology for future wastewater treatment. This study assessed the impact of different algae species inoculation on granulation, performance, and microbial communities within ABGS systems. The experimental setup included single-species inoculations ( Chlorella sp. (R1), Scenedesmus sp. (R2), and Desmodesmus sp. (R3)) and a mixed-species inoculation strategy (R4). Results revealed that R4 achieved the fastest completed granulation process (15 days) with the largest average granule diameter (772.93 μm) and highest physical strength (2.24 ± 0.26%) in the end of the experiment. The relative abundance of extracellular polymeric substances secreting bacteria of R4 maintained high level in whole operation time. Algae assimilation capacity and the abundance of functional bacteria can also influence removal performance. In mature stage, only the average effluent total nitrogen (3.15 ± 2.87 mg/L), total phosphorus (0.37 ± 0.27 mg/L), chemical oxygen demand (25.25 ± 2.98 mg/L) concentration in R4 was lower than that of Grade I discharge standard of municipal wastewater treatment plants in China. The best inorganic carbon utilization and lipid production ability were observed in R4 and R3, respectively. The choice of inoculated algae species was identified as a key factor for bacterial community dynamics. Overall, above results demonstrated that mixed algae species inoculation can be selected as the optimal algae inoculation strategy due to its excellent granulation, performance, and acceptable carbon utilization and lipid production.

Abstract:This study investigated the biological treatment of high-salinity wastewater using aerobic granular sludge (AGS). The removal performance, AGS characteristics, and microbial community were examined under salinity conditions ranging from 0.5 to 3.0 wt.%. When the salinity was increased, the removal efficiency of the chemical oxygen demand did not change substantially. The mean removal efficiency of NH4+-N ranged from 43.7 to 72.6% during the operation period. As for the concentration of extracellular polymeric substances (EPS), the polysaccharide/protein (PS/PN) ratio increased from 0.37 to 0.57. The concentrations of the mixed liquor suspended solid (MLSS) and mixed liquor volatile suspended solid (MLVSS) were 4,560 and 3,170 mg/L, respectively, in the end phase, and the MLVSS/MLSS ratio decreased from 0.81 to 0.69. The sludge volume index (SVI30) decreased linearly from 88 to 58 mL/g. In the microbial community analysis, Nitrosomonas species accounted for 22.9% of the total bacteria in the initial phase, whereas Halomonas accounted for 43.2% in the end phase. Hence, the gradually increased salinity appears to facilitate the dominance of halophilic microorganisms in the long-term operation of an AGS-based biological treatment system and the stable retention of AGS.

Abstract:Microalgal–bacterial granular sludge (MBGS) process has become a focal point in treating municipal wastewater. However, it remains elusive whether the emerging process can be applied for the treatment of aquaculture wastewater, which contains considerable concentrations of nitrate and nitrite. This study evaluated the feasibility of MBGS process for aquaculture wastewater treatment. Result showed that the MBGS process was competent to remove respective 64.8%, 84.9%, 70.8%, 50.0% and 84.2% of chemical oxygen demand, ammonia–nitrogen, nitrate–nitrogen, nitrite–nitrogen and phosphate–phosphorus under non-aerated conditions within 8 h. The dominant microalgae and bacteria were identified to be Coelastrella and Rhodobacteraceae, respectively. Further metagenomics analysis implied that microbial assimilation was the main contributor in organics, nitrogen and phosphorus removal. Specifically, considerable nitrate and nitrite removals were also obtained with the synergy between microalgae and bacteria. Consequently, this work demonstrated that the MBGS process showed a prospect of becoming an environmentally friendly and efficient alternative in aquaculture wastewater treatment.
