Abstract:Anthraquinones are bioactive natural products, some of which are active components in medicinal medicines, especially Chinese medicines. These compounds exert actions including purgation, anti-inflammation, immunoregulation, antihyperlipidemia, and anticancer effects. This study aimed to review the pharmacokinetics (PKs) of anthraquinones, which are importantly associated with their pharmacological and toxicological effects. Anthraquinones are absorbed mainly in intestines. The absorption rates of free anthraquinones are faster than those of their conjugated glycosides because of the higher liposolubility. A fluctuation in blood concentration and two absorption peaks of anthraquinones may result from the hepato-intestinal circulation, reabsorption, and transformation. Anthraquinones are widely distributed throughout the body, mainly in blood-flow rich organs and tissues, such as blood, intestines, stomach, liver, lung, kidney, and fat. The metabolic pathways of anthraquinones are hydrolysis, glycuronidation, sulfation, methylation/demethylation, hydroxylation/dehydroxylation, oxidation/reduction (hydrogenation), acetylation and esterification by intestinal flora and liver metabolic enzymes, among which hydrolysis, glycuronidation and sulfation are dominant. Of note, anthraquinones can be transformed into each other. The main excretion routes for anthraquinones are the kidney, recta, and gallbladder. Conclusion: Some anthraquinones and their glycosides, such as aloe-emodin, chrysophanol, emodin, physcion, rhein and sennosides, have attracted the most PK research interest due to their more biological activities and/or detectability. Anthraquinones are mainly absorbed in the intestines and are mostly distributed in blood flow-rich tissues and organs. Transformation into another anthraquinone may increase the blood concentration of the latter, leading to an increased pharmacological and/or toxicological effect. Drug-drug interactions influencing PK may provide insights into drug compatibility theory to enhance or reduce pharmacological/toxicological effects in Chinese medicine formulae and deserve deep investigation.

Abstract:Anthraquinones are polycyclic compounds with an unsaturated diketone structure (quinoid moiety). As important secondary metabolites of plants, anthraquinones play an important role in the response of many biological processes and environmental factors. Anthraquinones are common in the human diet and have a variety of biological activities including anticancer, antibacterial, and antioxidant activities that reduce disease risk. The biological activity of anthraquinones depends on the substitution pattern of their hydroxyl groups on the anthraquinone ring structure. However, there is still a lack of systematic summary on the distribution, classification, and biosynthesis of plant anthraquinones. Therefore, this paper systematically reviews the research progress of the distribution, classification, biosynthesis, and regulation of plant anthraquinones. Additionally, we discuss future opportunities in anthraquinone research, including biotechnology, therapeutic products, and dietary anthraquinones.

Abstract:Anthraquinones are widely distributed in medicinal plants with a variety of pharmacological activities,but the drugability of anthraquinones is largely restricted owning to their hepatotoxicity and nephrotoxicity.In order to ensure the safety and efficacy of anthraquinone-containing Chinese herbal medicines in clinical application,a large number of studies concerning the absorption and metabolism of anthraquinones have been conducted in vitro and in vivo.At present,the single-pass intestinal perfusion model method,Caco-2 cell model method,intestinal bacteria metabolism,liver/intestinal microsomal metabolism and plasma drug concentration analysis are mainly employed for researching the absorption and metabolism of anthraquinones.This article reviews the application of these approaches on the relationship between the chemical structure and transportation,the absorption and metabolism mechanisms of anthraquinones and the factors on the absorption and metabolism of anthraquinones in the past five years.

Abstract:Quassinoids, one kind of triterpenoids with multiple bioactivities such as anti-cancer, anti-malarial, anti-oxidative, anti-microbial, anti-diabetic, anti-viral, and anti-inflammatory effects, have drawn much attention in recent years. Between 2004 and 2018, the structural characteristics and plant sources of 190 quassinoids were reported. Herein, the structure-activity relationships (SARs) of quassinoids along with the anti-cancer mechanisms of four representative quassinoids, eurycomanone, bruceine D, dehydrobruceine B, and brusatol are discussed. This review might be useful for further research and development of quassinoids.

Abstract:Xanthones, as some of the most active components and widely distributed in various herb medicines, have drawn more and more attention in recent years. So far, 168 species of herbal plants belong to 58 genera, 24 families have been reported to contain xanthones. Among them, Calophyllum, Cratoxylum, Cudrania, Garcinia, Gentiana, Hypericum and Swertia genera are plant resources with great development prospect. This paper summarizes the plant resources, bioactivity and the structure-activity relationships (SARs) of xanthones from references published over the last few decades, which may be useful for new drug research and development on xanthones.

Abstract:Abstract: Antioxidant research has recently become a popular topic. Medicinal plants are important sources of novel active compounds. Diarylheptanoids, a typical family of secondary plant metabolites, are of great interest owing to their extensive spectrum of biological activities. They possess a unique 1,7-diphenylmethane structural skeleton. Thus, this review summarizes the natural linear or macrocyclic diarylheptanoids with antioxidant activity in the last two decades. In addition, the relationships between the structural characteristics of natural diarylheptanoids and their antioxidant capacity were also discussed. All the available data highlight the potential of natural diarylheptanoids as novel antioxidants.

Abstract:The structure of Polygonum multiflorum root,stem,leaf and root tuber and its anthraquinones distribution were studied to provide a reference to study the accumulation regulation of its anthraquinones.The results showed that the primary and secondary structure of the root and stem and the leaf structure is similar to dicotyledon.The root tuber is composed of periderm,secondary phloem,abnormal vascular,secondary xylem,and primary xylem.The abnormal vascular is the kind of amphicribral vascular bundle.Its anthraquinones is mainly distributed in root tubers and the anthraquinones content in its stem,root and leaf is little.

Abstract:Anthraquinones are bioactive natural products, which are often found in medicinal herbs. These compounds exert antioxidant-related pharmacological actions including neuroprotective effects, anti-inflammation, anticancer, hepatoprotective effects and anti-aging, etc. Considering the benefits from their pharmacological use, recently, there was an upsurge in the development and utilization of anthraquinones as reactive oxygen species (ROS) regulators. In this review, a deep discussion was carried out on their antioxidant activities and the structure-activity relationships. The antioxidant mechanisms and the chemistry behind the antioxidant activities of both natural and synthesized compounds were furtherly explored and demonstrated. Due to the specific chemical activity of ROS, antioxidants are essential for human health. Therefore, the development of reagents that regulate the imbalance between ROS formation and elimination should be more extensive and rational, and the exploration of antioxidant mechanisms of anthraquinones may provide new therapeutic tools and ideas for various diseases mediated by ROS.

Abstract:Seven anthraquinones, including two new natural products, hedyanthraquinones A and B (1, 2), were isolated from Hedyotis caudatifolia. The structures were elucidated based on NMR, MS data and physico-chemical properties. Compounds 1 and 2 displayed moderate cytotoxic activities against a panel of cultured tumor cell lines (HL-60, Bcap37, SMMC7721) with IC50 values of 13.71 ∼ 26.29 μM. The result showed that anthraquinones are typical compounds of Hedyotis genus, which could be useful as characteristic markers in chemotaxonomic research and might helpful for explaining the use of H. caudatifolia in Traditional Chinese Medicine.

Abstract:The anthraquinones (AQs) and derivatives are widely distributed in nature, including plants, fungi, and insects, with effects of anti-inflammation and anti-oxidation, antibacterial and antiviral, anti-osteoporosis, anti-tumor, etc. Inflammation, including acute and chronic, is a comprehensive response to foreign pathogens under a variety of physiological and pathological processes. AQs could attenuate symptoms and tissue damages through anti-inflammatory or immuno-modulatory effects. The review aims to provide a scientific summary of AQs on immune responses under different pathological conditions, such as digestive diseases, respiratory diseases, central nervous system diseases, etc. It is hoped that the present paper will provide ideas for future studies of the immuno-regulatory effect of AQs and the therapeutic potential for drug development and clinical use of AQs and derivatives.

Abstract:Xanthone is a collective name for one kind of natural plant secondary metabolites which possess tricyclic aromatic structure and widely distributed in the plants of several family such as Gentianaceae,Guttiferae,Polygalaceae,Moraceae and Leguminosae.The present studies showed that natural xanthones have wide range of pharmacological activities,including anti-tumor,anti-bacterial,anti-viral,hepatoprotective,pypoglycemic,inhibition of xanthine oxidase,etc.The review on pharmacological activities and related molecular mechanisms,structure-activity relationships of titled constituents over the last decade were summarized in this article,by which the prospect of development and utilization of natural xanthones can be prospected.The article provided the reference for the further research and comprehensive utilization of the natural xanthone resources.

Abstract:Nine anthraquinones were isolated from the leaves of Saprosma hainanense. On the basis of spectroscopic data analyses (HRESIMS, 1D- and 2D-NMIR) and that reported in the literature, The structures were established as 7-hydroxy-1,2-dimethoxy-6-methylanthraquinone (1), 1-hydroxy-3,6-dimethoxyanthraquinone (2), 10,10'-bianthrone (3), 9,10-anthraquinone (4), rubiadin (5), ramnacanthol (6), 2-hydroxy-3-methyl-9,10-anthraquinone (7), 3-hydroxy-1,2-dimethoxyanthraquinone (8) and 3-hydroxy-1-methoxy-2-carbomethoxy-9,10-anthraquinone (9). Among them, compounds 1 and 2 were new natural products, and 1 has no spectroscopic data reported until now. All compounds except 9 were isolated from this genus for the first time. Compounds 2 and 9 showed stronger inhibitory activities to the most of the bacteria, especially to the Staphylococcus aureus, with the minimum inhibitory concentration (MIC) values of 0.087 mu g/mL. Compounds 1 and 6 exhibited the higher inhibitory activities against Bacillus subtilis and Staphylococcus aureus than positive control ciprofloxacin.

Abstract:A series of novel Hydroxylated 9,10-anthraquinone derivatives were designed and synthesized as potential antitumor agents starting from the natural product Emodin. The structures of the compounds were identified by NMR, ESI-MS and elemental analysis. Their antitumor activities against KLE cell line were tested by MTT assay. Some of them were found to exhibit a cytotoxicity profile more potent than Emodin on KLE cell line.

Abstract:Hedyotis caudatifolia Merr. et Metcalf. (HC), a folk medicine in Yao nationalities areas in China, was used to investigate the chemical constituents. Through silica gel and Sephadex LH-20 column chromatography, nine compounds were isolated and purified. By physical and chemical properties, IR, MS (EI-MS, high resolution EI-MS), 1D NMR (1H NMR, 13C NMR) and 2D NMR (HSQC, 1H–1H COSY, HMBC), their structures were identified as β-sitosterol (1), stigmasterol (2), scopolin (3), 2-hydroxy-1,7,8-trimethoxyanthracene-9,10-dione (4), oleanolic acid (5), ursolic acid (6), methyl barbinervate (7), β-daucosterol (8) and p-Hydroxybenzoic acid (9). These compounds were isolated from HC for the first time, and 4 a new anthraquinone whose biological activities are worth to be investigated in future. These compounds may contribute to the HC’s pharmacological effects on treating diseases, and may be used as candidates for control index in establishing the quality control standard of HC.

Abstract:A new anthraquinone (4) and three known anthraquinones (1–3) were isolated from Galium verum L. Their structures were identified as 1,3-dihydroxy-2-methylanthraquinone (1), physcion (2), 2-hydroxy-1,3-dimethoxyanthraquinone (3), 2,5-dihydroxy-1,3-dimethoxyanthraquinone (4) by means of chemical and spectroscopic analysis. Compound 2 was isolated from the genus Galium for the first time. In addition, compound 4 was assayed for antimicrobial activity in vitro.

Abstract:The marine fungus Pannicillium sp.was isolated and purified from mangrove Kandelia Candel.T with solid phase extraction device.An anthraquinone was then isolated from the zymotic fluid of Pannicillium sp.with various chromatographic techniques,such as reversed phase silica gel(Lichrospher RP-18,RP-8) and Sephadex LH-20 column chromatography.Through a series of 1H NMR,13C NMR,COSY,HMQC,HMBC and ESI MS experiments,its structure was confirmed to be 5-methoxyl-11,12,18,19-tetra-hydroxyl-anthraquinone(1).

Abstract:Anthracycline antibiotics now were used extensively as clinic medicines, but new anthracyclines with novel bioactivities are still urgently needed because of the drug-resistant problem. During our screening for lead compounds from marine fungi from the South China Sea, three anthraquinones (1-3) were found to be produced by the marine-derived endophytic fungus isolate 1850 from a leaf of Kandelia candel from an estuarine mangrove in Hong Kong. Their structures were elucidated as averufin (1), nidurufin (2) and versicolorin C (3) by analysis of spectroscopic data. These three anthraquinones had also been obtained from another marine fungus (isolate 2526). The results that the same metabolites were produced by two different fungal strains derived from two different species mangrove plants shows the presence of some close connections between the two fungal strains and the two species mangrove plants.

Abstract:Anthraquinones are phenolic compounds known best for their laxative activity. Rheum australe D. Don (Indian rhubarb, Polygonaceae) is a prominent medicinal plant with anthraquinones as the bioactive compounds, among a few others. The online literature search was carried out to collect data on the phytochemistry and pharmacological activity of anthraquinones, particularly those isolated from R. australe. Anthraquinones are commonly found in the genera of Rheum, Senna, Aloe, Frangula, and Rubia, which can be quantitatively and qualitatively determined using both conventional and advanced analytical methods. The anthraquinones of R. australe were found in free and glycosidic forms, which were best extracted by the microwave-assisted extraction method. Various chromatographic techniques were commonly conducted to isolate the pure compounds. In addition to its laxative activity, anthraquinones of R. australe also showed potential antibacterial, cytotoxic, and antioxidant properties.

Abstract:Objective: To summarize the advances in research on pharmacological activity of xanthones in recent years. Methods: The related literature at home and abroad was collected and summarized. Results: Xanthones poses versatile pharmacological activities : antimalaria action ; liver protection ; therapy of urgent and chronic hepatitis ; antioxidation ; antitubercle ; antidepression ; therapeutic heal illness; disease of liver and gall; hematic illness; protect the myocardium against the damages induced by ischemia-reperfusion. Conclusion:Xanthones have versatile pharmacological activities, so they are the leading compounds from nature product which can developped in a wide space. However, there are still some deficiencies during the research on pharmacological activities of xanthones.
