Approximately 10% of the plants in the field showed symptoms characterized by yellow-brown necrotic spots on twigs, with leaves and young shoots later turning brown and dry. The main stem blighted and died after 20 days and black acervuli were observed on symptomatic twigs. Seven twig samples were collected from five diseased plants. Tissue fragments were excised from lesions on symptomatic twigs, sterilized by sequential dipping in 75% ethanol for 15 s and in 1% mercuric chloride for 30 s, rinsed in sterile distilled water, and placed onto potato dextrose agar (PDA) (Fang 1998). Petri plates were incubated at 25°C under diffused daylight (12 h/day). Six fungal isolates with similar morphological characteristics were obtained from three diseased plants and purified by single spore techniques. Pure cultures produced white aerial mycelium, turning slightly yellow after 5 to 7 days on PDA with yellowish green pigmentation. The cultures were covered with black acervuli. Conidia presented three median versicolored cells and two hyaline cells. Fifty conidia from one isolate were measured (19.6 to 24.4 × 5.4 to 7.5 µm, avg. 22.7 × 6.3 µm). Apical cells had 2 to 3 appendages (19.3 to 32.3 µm, avg. 25.3 µm) (Maharachchikumbura et al. 2012). The internal transcribed spacer (ITS) rDNA region, and part of the β-tubulin (TUB) and translation elongation factor 1 alpha (tef1) genes were amplified for one isolate (FZXM038-b) with the primers ITS5/ITS4, BT2A/BT2B, and EF1-526F/EF1-1567R, respectively (Maharachchikumbura et al. 2012). Sequences obtained were deposited in GenBank (ITS-KU534877, TUB-KT783666, tef1-KT783668). Alignments were made using CLUSTAL X v.2.0, and a maximum likelihood phylogram was generated by MEGA v.7.0. Isolate FZXM038-b clustered together with Neopestalotiopsis chrysea reference strains MFLUCC 12-0261 and MFLUCC 12-0262 (90% bootstrap). Seven healthy blueberry (Vaccinium ashei Reade) plants and six isolates were used to conduct pathogenicity tests. A 50 μl conidial suspension (105 conidia/ml) of each isolate was deposited on wounds made in three attached twigs per plant. Three equal wounded twigs in the same plant were inoculated with the same amount of sterile water as control. Wounds were wrapped with aseptic cotton wool for 3 days. Each twig was covered with a clear plastic bag and sterile water was sprayed to promote humid conditions. All blueberry plants were incubated at 28°C. After 5 days, brown necrotic spots were observed on inoculated twigs. After 20 days, twigs blighted and died, and black acervuli were observed on dead tissues. No lesions were observed on the control twigs. To complete Koch’s postulates, the inoculated fungus was reisolated from inoculated twigs and identified as described above. N. chrysea was first reported as a saprobe in Hunan and Guangxi provinces of China (Maharachchikumbura et al. 2014). First report of leaf spot on Daphniphyllum macropodum caused by Neopestalotiopsis clavispora in China

Abstract:Daphniphyllum macropodum Miq., an evergreen arbor, is widely cultivated in southern China for its ornamental and medicinal value (Su et al. 2013). In October 2019, a severe leaf spot was observed on D. macropodum in Jinggangshan National Nature Reserve in Ji'an city, Jiangxi, China (114°06'23″E, 26°32'25″N). The plants were about 15 years old, and the disease incidence was estimated to be 15% (4/26 plants). The disease primarily appeared as small black spots on the leaves. At the late stage, the spots enlarged and coalesced into regular or irregular gray necrotic lesions with dark margins. We collected five samples per plant and total 20 samples were collected to isolated the pathogen strains. The margin of the diseased tissues was cut into 5 mm × 5 mm pieces; surface disinfected with 70% ethanol and 2% NaOCl for 30 s and 60 s, respectively; and rinsed thrice with sterile water. Tissues were placed on potato dextrose agar (PDA) and incubated at 25°C in the dark. Pure cultures were obtained by single-spore isolation method, and the representative isolates, JRM3, JRM6, and JRM8 were used for morphological studies and phylogenetic analyses. The colonies of three isolates grown on PDA were white, cottony, and flocculent, contained undulate edges with dense aerial mycelium on the surface at 25 °C. Conidiomata was black conidial masses on PDA. Conidia were 5-celled, clavate to fusiform, smooth, 19.3 to 24.4 long × 6.1 to 8.6 μm wide (n = 50). The 3 median cells were dark brown to olivaceous, the central cell was darker than the other 2 cells, and the basal and apical cells were hyaline. All conidia developed one basal appendage (3.4 to 8.3 μm long; n = 50), and 2 to 3 apical appendages (18 to 32 μm long; n = 50), filiform. The morphological characteristics of the isolates are comparable with those of the genus Neopestalotiopsis (Maharachchikumbura et al. 2014). The internal transcribed spacer (ITS) regions, β-tubulin 2 (TUB2) and translation elongation factor 1-alpha (TEF1-α) were amplified from genomic DNA for the three isolates using primers ITS1/ITS4, T1/Bt-2b, EF1-728F/EF-2 (Maharachchikumbura et al. 2014), respectively. The sequences of the isolates were submitted to GenBank (ITS, OQ372202 to OQ372204; TUB2, OQ390129 to OQ390131; TEF1-α, OQ390126 to OQ390128). A maximum likelihood and Bayesian posterior probability analyses using IQtree v. 1.6.8 and Mr. Bayes v. 3.2.6 with the concatenated sequences placed JRM3, JRM6, and JRM8 in the clade of N. clavispora. Based on the multi-locus phylogeny and morphology, three isolates were identified as N. clavispora. To confirm pathogenicity, eight healthy 10-year-old D. macropodum plants growing in the field were chosen, and 4 leaves per plant were wounded with a sterile needle and inoculated with 10 μL conidial suspension per leaf (106 conidia/ml). Eight plants inoculated with sterile water were used as control. All the inoculated leaves were covered with plastic bags to maintian a humidity environment for 2 days. The leaves inoculated with conidial suspension showed similar symptoms to those observed in the field, whereas control leaves were asymptomatic for 10 days. The same fungus were re-isolated from the lesions, whereas no fungus was isolated from control leaves. N. clavispora was determined as the pathogen of a variety of plant diseases, including Kadsura coccinea (Xie et al. 2018), Dendrobium officinale (Cao et al. 2022), Macadamia integrifolia (Santos et al. 2019). However, this is the first report of N. clavispora infecting D. macropodum in China. This work provided crucial information for epidemiologic studies and appropriate control strategies for this newly emerging disease.
First Report of Phytophthora Nicotianae Causing Blight of Dendrobium Candidum in Zhejiang Province, China

Abstract:Dendrobium candidum is a traditional Chinese medicinal herb, used raw or processed for health-care products in recent years. Due to the high economic value, there is huge demand and over-collecting of this plant, and hence the resources of wild D. candidum are endangered. In order to meet the increasing demands, tissue cultured seedlings are grown on large scale fields by local enterprises. Since 2001, a root and basal stem rot and die-back disease has been observed on D. candidum in Yiwu city, Zhejiang province, China. In the field, 1-year-old affected transplanted seedlings demonstrate typical symptoms of Phytophthora root and basal stem rot. Symptom progression is acropetal and plants die within a few days. Tap roots are completely rotted with black-brown to black sunken lesions frequently extending up the stem. In contrast, 2- to 3-year-old infected plants only have die-back symptoms with almost no root and basal stem rot. Typical black-brown lesions are observed on the tender top parts of a plant, causing top wilt and defoliation. Dendrobium candidum blight has not been observed in the wild. Five isolates of a Phytophthora species from diseased material were initially identified as Phytophthora nicotianae using morphological features (Stamps et al., 1990; Erwin & Ribeiro, 1996). No oospores were observed in the tissue. Mating types were determined by pairing known P. nicotianae tester strains. Oospores were produced following pairing tester strains and all isolates proved to be of the A2 mating type. PCR amplification of genomic DNA using the primers ITS6 and ITS4 generated sequences of approximately 890 bp. The sequences were submitted to the GenBank database (Acc. No. EF140984–88). Sequence analysis showed that the ITS sequences obtained were identical with others obtained from P. nicotianae (Estevez de Jensen et al., 2006), thus confirming the identity of the pathogen. Mycelial plugs of the five Phytophthora isolates taken from agar medium, were inoculated into the basal stem of 2-month-old transplanted seedlings grown in pasteurized potting substance (mainly chipped pine bark) and to the top parts (new shoots) of 2-year-old plants. Plants were grown in the greenhouse at 25°C, where they developed typical disease symptoms after 7 days. Non-inoculated controls did not develop disease symptoms. The pathogenic isolate was re-isolated from all diseased plants. Isolates have been deposited at the Biotechnology Institute, Zhejiang University. This is first record of phytophthora blight of D. candidum in China.

Sw. is a perennial herbaceous plant with high gardening and herbal value. In August 2014, a severe anthracnose disease was observed on C. ensifolium leaves in Changchun (125.24°E, 43.48°N), Jilin Province, China. More than 80% of the plants were symptomatic. In the early stage of disease development, small brown lesions (1 mm in diameter) were visible on infected leaves, which subsequently developed into oval spots and turned light brown in the center. Lesions ranged from 5 to 15 mm in diameter. As lesions enlarged and coalesced, some leaves dried up. When the humidity was high, the surfaces of the lesions were covered with dark brown acervuli. Small pieces of infected leaves were surface-sterilized in 75% ethanol for 60 s, sterilized for 1 min in 1.5% NaClO, rinsed twice with sterilized distilled water, and plated on potato dextrose agar (PDA) in Petri dishes at 25 ± 1°C for 7 days. Ten fungal isolates were transferred from PDA to synthetic nutrient-poor agar (SNA). White colonies were observed in 2 days with smooth margins, and they covered the Petri dishes after 7 days. Perithecia (n = 100) were brown to dark brown, globose to subglobose, and measured 136.24 to 154.5 × 145.42 to 236.68 µm. Asci and ascospores were absent. Conidiophores and setae were formed directly on the media. Conidiophores (n = 100) were cylindrical, septate, branched, and measured 5.35 to 18.28 × 2.43 to 6.56 µm. Conidia (n = 100) were hyaline, aseptate, smooth, cylindrical, contents granular, and were 12.21 to 18.42 µm in length, and 4.40 to 7.90 µm in width. Setae (n = 100) were brown, cylindrical to conical, measuring 40.62 to 160.58 µm in length, and were 1 to 4 septate. Based upon these morphological and cultural characteristics, the causal agent was identified as Colletotrichum cymbidiicola. Five sequences of the internal transcribed spacer (ITS, KX058529), glyceraldehyde-3-phosphate dehydrogenase (GAPDH, KX068532), actin (ACT, KX058531), and beta-tubulin (TUB, KX058530) regions were obtained and deposited in GenBank. BLAST search results revealed that all sequences showed 99% identity with C. cymbidiicola sequences. Pathogenicity of 10 isolates of C. cymbidiicola from C. ensifolium were confirmed by inoculating healthy leaves of C. ensifolium plants with a spore suspension (1 × 106 spores/ml) produced on PDA. Each isolate was inoculated onto five plants and another five healthy plants were sprayed with sterilized water as controls. All plants were enclosed in plastic bags for 48 h in a greenhouse at 28°C and 12 h/day light cycle. After 7 days, inoculated plants showed similar symptoms to those on the original diseased plants, while control plants remained symptomless. C. cymbidiicola was successfully reisolated from symptomatic plants to fulfill Koch’s postulates. C. cymbidiicola causing disease was reported on cymbidium in Australia, New Zealand, Japan (Damm et al. 2012; Hou et al. 2016), and India (Cannon et al. 2012). Abstract:delta-Chymotrypsin has been alkylated by 1-13C- and 2-13C-enriched tosylphenylalanylchloromethane. In the intact inhibitor derivative, signals due to the 1-13C- and 2-13C-enriched carbon atoms have chemical shifts which titrate from 55.10 to 59.50 p.p.m. and from 99.10 to 103.66 p.p.m. respectively with similar pKa values of 8.99 and 8.85 respectively. These signals are assigned to a tetrahedral adduct formed between the hydroxy group of serine-195 and the inhibitor. An additional signal at 58.09 p.p.m. and at 204.85 p.p.m. in the 1-13C- and 2-13C-enzyme-inhibitor derivatives respectively does not titrate when the pH is changed and it is assigned to alkylated methionine-192. On denaturation/autolysis of the 1-13C-enriched enzyme-inhibitor derivative these signals associated with the intact inhibitor derivative are no longer detected, and a new signal, which titrates from 56.28 to 54.84 p.p.m. with a pKa of 5.26, is detected. The titration shift of this signal is assigned to the deprotonation of the imidazolium cation of alkylated histidine-57 in the denatured/autolysed enzyme-inhibitor derivative. Model compounds which form stable hydrates and hemiketals in aqueous solutions have been synthesized. By comparing the 13C titration shifts of these model compounds with those of the 13C enriched trypsin- and delta-chymotrypsin-inhibitor derivatives, we deduce that, in both of the intact enzyme-inhibitor derivatives, the zwitterionic tetrahedral adduct containing the imidazolium cation of histidine-57 and the hemiketal oxyanion predominates at alkaline pH values. It is estimated that in both the trypsin and delta-chymotrypsin-inhibitor derivatives the concentration of this zwitterionic tetrahedral adduct is 10,000-fold greater than it would be in water. We conclude that the pKa of the oxyanion of the hemiketal in the presence of the imidazolium cation of histidine-57 is 7.9 and 8.9 in the trypsin and delta-chymotrypsin-inhibitor derivatives respectively and that the pKa of the imidazolium cation of histidine-57 is greater than 7.9 and greater than 8.9 when the oxyanion is present as its conjugate acid, whereas, when the oxyanion is present, the pKa of the imidazolium cation is greater than 11 in both enzyme-inhibitor derivatives. We discuss how these enzymes preferentially stabilize zwitterionic tetrahedral adducts in the intact enzyme-inhibitor derivatives and how they could stabilize similar tetrahedral intermediates during catalysis. It is suggested that substrate binding could raise the pKa of the imidazolium cation of histidine-57 before tetrahedral-intermediate formation which would explain the enhanced nucleophilicity of the hydroxy group of serine-195.(ABSTRACT TRUNCATED AT 400 WORDS)
First Report of Diaporthe Cercidis and D. Nobilis Causing Leaf Blotch on Populus Davidiana × P. Bolleana in China.

Abstract:Shanxin yang (Populus davidiana × P. bolleana) is a commercially valuable hybrid poplar that is widely planted in northern China. Efficient genetic transformation and gene-editing systems have been established for this hybrid poplar (Wang et al., 2011; Wang et al., 2020). However, records of fungal diseases on Shanxin yang are very limited. In July 2020, large necrotic lesions were observed on 16 one-year-old Shanxin yang seedlings planted in a greenhouse of Nanjing Forestry University, Nanjing, China. The disease symptoms appeared mostly on the leaves and not on the stems. Symptoms first manifested as differently sized and shaped brown spots, having clearly demarcated margins. As the disease progressed, the spots coalesced, and large lesions were present on the leaves. Severe infections resulted in whole-plant defoliation and eventually plant death. Small leaf samples (5 mm2) cut from lesion margins were surface sterilized with 75% ethanol for 30 s, followed by 1% NaClO for 90 s and then washed three times with sterile distilled water. After drying on sterilized filter paper, the cut tissues were plated on potato dextrose agar (PDA) supplemented with ampicillin (100 μg/mL) and incubated at 25°C in the dark. Three isolates (named as SX-1, SX-2 and SX-3, respectively) were obtained after 5 days. The isolation frequency was low, which might be due to the greenhouse microclimate that was unfavorable for pathogen spread. Mycelial plugs (5 mm) cut from the leading edge of the mycelia were transferred onto fresh PDA and synthetic nutrient-poor agar (SNA) plates to obtain pure cultures. On both media, colonies of the isolates were white on the front and light yellowish on the back, with concentric zonation. Abundant aerial mycelia developed; the hyphae were hyaline, non-septate, and approximately 0.794-2.961 µm in diameter. On the SNA medium, SX-1 and SX-3 produced globose to subglobose, black pycnidia after 18 days under a 12 h photoperiod. The alpha conidia were fusoid, aseptate, hyaline, smooth, and 6.4 ± 1.2 × 2.4 ± 0.6 µm (n = 50) in size. Under the same conditions, SX-2 produced pycnidia after 20 days, and the conidia were 2.8 ± 0.7 × 7.5 ± 1.3 µm. Beta conidia were not observed on any colony. Based on the morphological characteristics, the isolated mycelia resembled Diaporthe spp. (Gomes et al., 2013). To determine the species identity, genomic DNA from each isolate was extracted, and five loci were amplified, namely, part of the internal transcribed spacer (ITS) amplified with primers ITS1/ITS4 (White et al. 1990); part of the translation elongation factor 1-alpha (EF1-α) with primers EF1-728F/EF1-986R (Carbone and Kohn, 1999); part of the calmodulin (CAL) with primers CAL-228F/CAL-737R (Carbone and Kohn, 1999); part of the β-tubulin (β-tub) with primers Bt2a/Bt2b (Glass and Donaldson, 1995), and part of the histone H3 (HIS) with primers CYLH3F/H3-1b (Glass and Donaldson 1995, Crous et al., 2004). The obtained sequences were deposited in GenBank (accession numbers are listed in Table S1). BLAST analyses showed that the all the amplified fragments were highly homologous to Diaporthe spp. (Table S1). Based on concatenated sequences of the amplicons, a phylogenetic tree was constructed by using Maximum-likelihood and Bayesian inference methods. The multi-locus phylogenetic analyses distinguished SX-1 and SX-3 as D. cercidis, and SX-2 as D. nobilis. To complete Koch's postulates, the pathogenicity of SX-1, as well as SX-2, was tested on both detached and attached leaves of one-year-old Shanxin yang seedlings grown under greenhouse conditions. Healthy leaves were pierced with a sterile needle and then inoculated independently with 5-mm mycelial plugs cut from the edge of the 4-day-old colonies of SX-1 and SX-2 colonies. Controls were inoculated with noncolonized PDA plugs. Three replicates were prepared for each isolate. For the in-vitro tests, detached leaves were placed on wet filter paper in parafilm-sealed Petri dishes and cultured at 25 °C in the dark. For the attached leaf assays, the plants were kept in an 85% humidity chamber immediately after inoculation. All the inoculated leaves developed dark or brown necrotic lesions at 5 days after inoculation, whereas the control leaves showed no symptoms. D. cercidis and D. nobilis were separately reisolated from the inoculated leaves. The former was first described by Yang et al. (2018) as occurring on twigs and branches of Cercis chinensis, and very recently, this pathogen was reported to cause leaf blotch on Acer pictum subsp. mono (Wan et al., 2021). The latter infects some fruit trees (Yu et al., 2018; Sun et al., 2019; Ma et al., 2019) and chestnut (Zhang et al., 2018). All of these studies were conducted in China where there is a great diversity of Diaporthe species (Yang et al., 2018). To our knowledge, this is the first report of both D. cercidis and D. nobilis causing leaf blotch on poplar. The identification of these pathogens is essential for understanding the range of their host species and to manage the resulting fungal diseases, which may cause severe economic damage.
A Destructive New Disease of Citrus in China Caused by Cryptosporiopsis Citricarpa Sp. Nov.

Abstract:Defoliation, dieback and mortality of Satsuma mandarin (Citrus unshiu), as well as kumquat (Fortunella margarita), in Chenggu County, Shaanxi Province, China was first noticed in 2006 and caused substantial economic losses to citrus production. The incidence of leaves infected approached 100% in some badly infected orchards. The disease prevailed only in late winter and early spring, where early symptoms were rounded and target-like spots on leaves. The disease was named target spot. Black conidiomata were observed on the upper surface of the leaves. A Cryptosporiopsis species was consistently recovered from the infected leaves, shoots, and branches. Koch's postulates were fulfilled by inoculating the conidial suspension of Cryptosporiopsis sp. onto the leaves of Satsuma mandarin. Phylogenetic analysis based on LSU sequence data indicated that this taxon clustered in Cryptosporiopsis (teleomorph: Neofabraea, Dermateaceae). Phylogenetic analysis based on ITS, SSU, and TUB indicated that the isolates of Cryptosporiopsis sp. constituted a distinct clade. Further study also demonstrated that this taxon was morphologically distinct from other species of Cryptosporiopsis, thus suggesting it might belong to an undescribed species. The name Cryptosporiopsis citricarpa sp. nov. is given to accommodate the fungal pathogen in this study.
First Report of Phyllosticta Capitalensis Causing Black Spot on Radermachera Hainanensis in China

Abstract:Radermachera hainanensis Merr. plants are native in south-central and southeast of China. Plants produce large flowers, and are widely cultivated in China as ornamentals. In April 2020, R. hainanensis Merr. plants grown in Cixi Lvpin Garden (30°26'54″N, 121°25'48″E), Zhejiang Province, were found to have many black circular necrotic lesions. In the early infection stage, the lesions appeared in lower leaves as small black circular spots which developed later into large spots (11 to 38 mm diameter) with grey centers and chlorotic edges. Ultimately, the spots spread and merged. Moreover, infected leaves showed premature leaf fall. Disease intensity reached approximately 20% of plants in the affected field (0.5 ha). After effective chemical control, this disease did not spread to other healthy plants in the same garden. To identify the causative pathogen associated with the disease, ten symptomatic leaves were collected from ten different plants. Leaf tissues were cut from the lesion margins and sterilized as follows: surface sterilized with 75% ethanol for 30 seconds and washed three times in sterile distilled water. The leaf tissues were then dipped into 10% sodium hypochlorite for 3-4 minutes, then washed three times in distilled water and dried on a sterile filter paper. After drying, the surface-sterilized leaf discs were cut to small pieces (3×3 mm) and transferred to potato dextrose agar (PDA) plates and incubated at 28°C for 2 to 3 days under 12 h photoperiod. A total of 15 isolates were obtained from the affected leaves, and all the isolates displayed the same colony characteristics. Then, three single-spore isolates were randomly selected (F2, F5 and F8) for further study. The fungal colonies were dark green with a granular surface, and irregular white edges, later turning black. Conidia were one-celled, oval, and narrow at the end with a single apical end, measuring from 7.8 to 11.1 × 4.6 to 5.9 μm (av. 9.5 × 5.2 μm, n=50). These morphological characteristics were consistent with the description of Phyllosticta capitalensis (Wikee et al. 2013; Guarnaccia et al. 2017). The identity of three representative isolates were confirmed by a multilocus approach. The DNA of three isolates were extracted and partial sequences of ribosomal internal transcribed spacer (ITS), actin (ACT), and translation elongation factor 1-alpha (TEF1-α) were amplified and sequenced as previously described (White et al. 1990; O'Donnell et al. 1998; Carbone & Kohn et al. 1999). The three selected isolates shared 100% identical sequence of ITS, ACT and TEF1-α. Then representative isolate F8 was selected for further study. BLAST analysis in GenBank showed that the obtained sequence of ITS (MZ317550) had 99% identity to P. elongata isolate eSX25240811. Other two sequences of ACT (MZ326837) and TEF1-α(MZ326839) showed 99% and 98% identity to P. capitalensis isolate YLWB01, respectively. The phylogenetic trees were constructed by Bootstrap method with 1000 replications using Maximum Likelihood model implemented in the MEGA 7. Results showed that the isolate F8 clustered with P. capitalensis with 100% bootstrap support. Pathogenicity of strain F8 was tested by Koch's postulates. A pathogenicity test was performed in a greenhouse with 80% relative humidity at 28°C. 20 healthy plants were sprayed with a 1×106 conidia ml-1 suspension (three leaves from each individual plants) and another 20 healthy plants were sprayed with sterile distilled water (three leaves from each individual plant) as control. Conidia was obtained from PDA plates after 7 days of incubation in the biochemical incubator at 28°C and concentration was counted in hemacytometer. After 15 days, disease symptoms were observed on all inoculated leaves, whereas the control plants remained asymptomatic. After that, P. capitalensis was re-isolated only from the infected leaves and identified by morphological and sequence analyses. Early identification of P. capitalensis as a causal agent for black spot is crucial to employ effective disease management strategies to control disease in the field. P. capitalensis has been reported on many crops in China (Cheng et al. 2019; Tang et al. 2020; Liao et al. 2020). However, to our knowledge, this is the first report of black spot disease caused by P. capitalensis on Radermachera hainanensis Merr. in China.
First Report of Leaf Blight of Ginkgo biloba L. Caused by Neopestalotiopsis clavispora G.F. Atk. in Sichuan, China

Abstract:Ginkgo (Ginkgo biloba L.), the oldest existing tree species in the world, is an important ornamental and medicinal plant, widely planted in China. In October 2022, a new leaf blight disease was observed in Chengdu city (30°05'to 31°26'N, 102°54'to 104°53'E). Disease incidence averaged 82.5% across five foci. The typical symptomatology begins when leaf margins turn yellow and small black spots appear at the edge of the leaf, chlorotic areas turn brown, dry and deformed. Gradually, the necrotic lesions spreads to the middle of the leaf and eventually the whole leaf falls off. Infected tissues from ten leaves were cut into small pieces (5 × 5 mm); surface sterilized for 30 s in 3% sodium hypochlorite; 60 s in 75% ethanol; rinsed three times in sterile water; placed onto potato dextrose agar (PDA) amended with streptomycin sulfate (50 μg/mL); and incubated at 25°C for 3 to 8 days. A hyphae was removed from the edge of the fungal colony and placed onto potato dextrose agar (PDA) plates. After incubation at 25℃ with a 12-hour light/dark cycle for 8 days, the colony diameter reached 77.5 to 81.5 mm. Colonies grown on PDA were white, cotton, flocculent, undulating on the surface, dense in aerial hyphae and light yellow on the back. Black pycnidia formed superficially, scattered over the PDA, following two weeks of incubation. Pycnidia contained sticky black conidia. The spores were were spindle shaped, with five cells, and four septations measuring 20.9 to 34.8 µm × 6.8 to 8.8 µm (avg. 28.4 × 7.6 µm; n=40). The three median cells were versicolored, typically two dark brown cells and one light brown cell, whereas the basal and apical cells were hyaline. Conidia had a single basal appendage (2.87 to 4.1 µm long; n = 40) and two to three apical appendages (18.3 to 29.1 µm long; n = 40). Based on colony and conidial morphology, the isolate was identified as N. clavispora (Maharachchikumbura et al. 2014). The partial sequence of the internal transcribed spacer (ITS), β-tubulin gene (TUB2), and translation elongation factor subunit 1-a gene (TEF1) were amplified and sequenced using the universal primer pairs ITS1/ITS4(Zhang et al. 2022), BT2A/BT2B (Li Yuan et al. 2022), and EF1-526F/EF1-1567R (Maharachchikumbura et al. 2012), respectively. Sequences of representative isolate LQYX were deposited in GenBank (ITS: OQ152504, TUB: OQ168328, and TEF1: OQ168329). BLAST results indicated that the ITS, TUB, and TEF1-α sequences showed 99 to 100% identity with N. clavispora sequences at NCBI (GenBank MG729689, MG740735, and MG740758). Identification was confirmed by Bayesian inference using Mr. Bayes. Next, inoculations were conducted on leaves of ten G. biloba in the field to verify the pathogenicity of LQYX. Ten healthy leaves of each plant were surface sterilized with 75% ethanol, and the wound was rubbed out on the leaf edge on the sterilized sanding paper. A conidia suspension (1 × 107 ml-1) was sprayed on the leaves, aseptic water was used as the control, and the transparent plastic bag was used to maintain relative humidity. After 14 days (26 ℃, 14 hours light / 10 hours dark), the inoculated leaves had similar symptoms as the original diseased plants, whereas controls were asymptomatic. The N. clavispora was re-isolated from the infected leaves and identified by morphological characteristics and DNA sequence analysis. The pathogenicity test was repeated three times with similar results, confirming Koch's postulates. To our knowledge, this is the first report of leaf blight of G. biloba caused by N. clavispora in China, which has greatly affected the appearance of the city and should be further studied. This report can help identify this disease and further develop effective control measures.
First Report of Pestalotiopsis lushanensis Causing Brown Leaf Spot on Sarcandra glabra in China

Abstract:Sarcandra glabra is a species of Chloranthaceae family and this family grow in the southern part of China, Japan, and Southeastern Asia (Li et al. 2019). It is a kind of precious Chinese herbal medicine, which occupies an important position in traditional Chinese herbal medicine. It plays an effective role in the treatment of cancer, rheumatism, pneumonia, digestive tract inflammations, traumatic injuries and fractures, anti-virus, anti-bacterial, antioxidant, etc. (Li et al. 2019; Zheng et al. 2003; Zhou et al. 2013). Since June 2020, we discovered a serious leaf disease in the S. glabra planting base of Shibing County (108.12E 27.03N), in Guizhou Province, with an incidence rate of 60% and yield losses of 40%. Initially, the symptoms developed as small specks where spots were purple with a dark brown halo margin, and round or oval. In later stages, the spots gradually expanded and became dry, whole severe leaf loss. To identify the pathogen, we collected the diseased leaves from S. glabra fields in Shibing County. Small tissue pieces from the edges of lesions were disinfected in 75% ethyl alcohol for 30 s and 1% hypochlorite for 1 min, rinsed five times in sterile water, plated on potato dextrose agar (PDA), and incubated at 28°C in lighted incubator for 3 days. Fungal colonies were consistently isolated and transferred to PDA for morphological characterization (Fang et al. 2007). Pathogenicity tests of the novel isolate HGUP CSH-2 were conducted by spraying spore suspensions with a concentration of 1.6×108 conidia/ml on surface-disinfected (70% ethyl alcohol, 30 s) leaves, while sterile distilled water was used as the control. Plants with inoculated leaves (three per treatment) were placed in lighted growth chambers at 28°C for 5 days and watered as needed (Light to dark ratio 1:1, RH=90%). Symptoms on inoculated leaves were similar to those described previously in the field. The same pathogenic fungus was re-isolated from the infected leaves but not from the non-inoculated leaves. Colonies on PDA attaining 70 mm diam after 7 d at 28°C, with pale honey-colored, sparse aerial mycelia on the surface with black, gregarious conidiomata. Conidiogenous cell discrete or integrated, ampulliform, clavate or subcylindrical, hyaline, smooth-walled, wide at base. Conidia fusoid, ellipsoid, straight to slightly curved, 4-septate, slightly constricted at septa, 22.26-27.17×6.9-8.22 µm (av.±SD: 24.68±1.57×7.68±0.38 µm; n=30). According to the colony and conidia characteristics, the isolate was initially identified as Pestalotiopsis spp. (Liu et al. 2017). The pathogen was confirmed by amplification and sequencing of the internal transcribed spacer region (ITS) gene, the translation elongation factor-1 (TEF1) gene and the β-tubulin (TUB2) gene (Liu et al. 2017) using ITS1/ITS4, Bt2a/T1 and EF1-526F/1567R primers, respectively. The sequences of the PCR products were deposited in GenBank with accession numbers MT919215 (ITS), MT939300 (TUB2) and MT939299 (TEF1). BLAST results of the obtained sequences of the ITS, TUB2 and TEF1 genes revealed 97.16% (479/493 nucleotides), 99.56% (675/678 nucleotides) and 99.89% (890/891 nucleotides) homology with those of Pestalotiopsis lushanensis in GenBank (MG726538, KY464157 and KX895223). Maximum Likelihood method was used for phylogenetic analysis. The result showed that HGUP CSH-2 was together with P. lushanensis with a support rate of 100%. According to the morphological characteristics and molecular phylogenetic analysis, the pathogen was identified as P. lushanensis. So far as we know, our research is the first report of brown leaf spot of S. glabra caused by P. lushanensis in China. Thus, identification of P. lushanensis for this disease is important for the advancement of effective prevention and control practises as future perspectives.
Hemorrhagic cystic lesion of the median nerve: An unusual complication of venipuncture

Abstract:Three recent papers describe neural complications of venipuncture in the cubital fossa (Yamada et al., 2008; Stevens et al., 2012; Moore and Stringer, 2012). We illustrate the case of a patient found to have a hemorrhagic cystic mass in the median nerve in the elbow region as visualized on MRI which was determined to be a thrombosed epineural venous pseudoaneurysm (that is, a contained rupture), historically related to a previous venipuncture. A 54-year-old woman presented with a 9-month history of paresthesias in the radial 3 1/2 digits associated with a new mass in the elbow. These findings arose soon after attempted venipunctures in the antecubital fossa and were accompanied by an adjacent hematoma. The patient described herself as a ‘‘hard stick.’’ From her account, the mass, which had fluctuated in size, had now slowly increased in dimensions. Physical examination revealed a firm lesion just above the antecubital crease. Percussion over it produced radiating paresthesias into the palm. Results of neurological testing and electrophysiological studies were normal. MRI, initially interpreted as showing a nerve sheath tumor at the outside facility, revealed a hemorrhagic cystic lesion within the median nerve (Figs. 1A–1D). The elbow had degenerative changes and a joint effusion. An MR arthrogram excluded a joint connection as would be typical of an intraneural ganglion cyst. At operation a longitudinal incision was made centered upon the mass (Fig. 2A) which was located just deep to the brachial vessels. A bluish colored vascular mass was identified within the epineurium of the median nerve (Fig. 2B). An epineurotomy was performed. The lesion was dissected away from neighboring fascicles and exiting motor branches and removed in toto (Figs. 2C and D). Microscopic evaluation revealed a ruptured vein with an adjacent organizing hematoma that was confined by a greatly dilated venous adventitia, indicative of a venous pseudoaneurysm (Figs. 2E and 2F). Reinterpretation of the MRIs showed evidence of a patent vein proximally and distally and a thrombosed, dilated feeding vein contained within the mass (Figs. 1E and 1F) surrounded by subacute hemorrhage and soft tissue edema. Postoperatively, the patient had transient weakness of the flexor pollicis longus which lasted several days. At 4 month follow-up examination, symptoms had resolved and neurologic function, normalized. The radiologic differential diagnosis included an intraneural vascular lesion (intraneural hemangioma), and hemorrhage within an intraneural ganglion cyst (Gurdjian et al., 1965) or a cystic benign or malignant nerve sheath tumor. Initially, we considered the historical descriptions of hematic (Velpeau, 1856), serosanguineous (Ashhurst, 1882), or blood cysts (Bryant, 1888; Albertin, 1904; Speese, 1915) as well as other cases of an arterio-venous cyst (Moore, 1866); or cysts with hemorrhagic components, such as a fibromyxoma (Linell, 1922), glioma (Bertand and Charrier, 1922), and intraneural ganglion cyst (Bertrand, 1837). Unfortunately, based on the original, vague descriptions, it is difficult to know what the underlying pathologies really were. Disappointingly, the actual specimens, some of which were housed in museums, are not currently available for study. These somewhat ambiguous terms still persist in current usage: for example, the term hematic cyst which applies mostly to orbital tumors (n.b., it is defined as a collection of hematogeous debris within a fibrous pseudocaspule), has also been used in conjunction with peripheral nerve pathology (Kastner, 1966; Pagliughi and Vespasiani, 1979). Venipuncture-related injuries to cutaneous and major nerves are not rare and are underreported. While pseudoaneurysms and hematomas are known examples of mass lesions which may extrinsically compress peripheral nerve (s), we are unaware of a similar type of intraneural ‘‘blood’’ cyst occurring in this setting, due to a venipuncture-related thrombosed epineural venous pseudoaneurysm.
First Report of Fusarium Concentricum As a Causal Agent of Fusarium Leaf Blotch on Pecan (carya Illinoinensis) in Southeast China.

Abstract:The economically important nut crop pecan (Carya illinoinensis (Wangenh.) K. Koch) is seriously affected by increasing incidence of fungal disease worldwide (Xiao et al 2021). The top leaves of the pecan variety 'Pawnee' in the orchard of Zhejiang A&F University, Zhejiang, China were damaged by massive dark brown plaques in summer to autumn 2021. The causal agent was isolated from leaves with target plaques following the steps: sterilized with 70% alcohol (30 s × 2), rinsed with sterilized water (3 ×) before and after 5% sodium hypochlorite (30 s), excised the plaques, and placed on PDA medium at 28℃ in a dark incubator for 3-d. The mycelium on the edge of each colony was transferred to fresh SNA medium in dark for 2 weeks to induce conidia formation. A few conidia-germinated mycelia were transferredand inoculated on new plates containing fresh PDA medium to obtain the purified cultures. Koch's postulates were applied to validate the pathogenicity of the purified isolates. Non-woundedly healthy leaves (disinfected with 5% sodium hypochlorite) of 'Pawnee' seedlings were inoculated with 5 mm 7-d old purified cultures. Dark-brown spots appeared on the leaves 2 days post inoculation at 25℃. The spots became larger accompanied by partially cracking and slight deformation of inoculated leaves from day 2 to day 4, while the control leaves remained asymptomatic. A re-isolated strain ZJ-6 from these infected leaves was identified as the pathogenic isolate with the same symptom as the previous one. Morphologically, aerial mycelia of the pathogenic isolate ZJ-6 cashmere and white. The reverse of colony orange. The edge of the colony appeared gradually thinner, the aerial mycelia loose and flocky, and the matrix mycelium whitened. Hyphae were septate, translucent with smooth wall and 1.47-7.14 µm in width. Microconidia (n = 20) obovoid to fusoid, mainly with 0-septate, 4.45-7.78×4.79-16.25 µm. Macroconidia (n = 20) sickle, mainly with 3-5 septa, 5.56-10.28×56.67-114.54 µm. Simultaneous of monophialidic and polyphialidic conidiophores. Conidiophore width 1.47-3.68 µm, slightly smaller than vegetative hyphae. The morphological characteristics matched with previous descriptions of Fusarium species (Nirenberg and O'Donnell 1998; Wang et al 2013). The identity of ZJ-6 was confirmed by phylogenetic reconstruction using the concatenated sequences of the ATP citrate lyase (ACL1), Calmodulin (CaM), the internal transcribed spacer (ITS) rDNA region, ribosomal RNA gene (LSU), the largest subunit of DNA-dependent RNA polymerase II (RPB1), partial translation elongation factor-1 alpha (TEF) and β-Tubulin (TUB). To this end, the genomic DNA of ZJ-6 was extracted by the M5 hipermix-MF859 (Mei5 Biotechnology) and submitted to GenBank under the accession numbers OP933646, OP933647, OP925890, OP925889, OP933396, OP933648, and OP933397, respectively. The obtained sequences of ZJ-6 were used for nucleotide BLAST against thetandard databases, respectively, and the strains with sequence identification values above 98% were selected to construct multiple alignment for building a phylogenetic tree. This analyses allowed the identification of ZJ-6 as Fusarium concentricum Nirenberg & O'Donnell, a species with few reports that can cause serious damage to the fruits and branches of other hosts (Hasan et al 2020; Huda-Shakirah et al 2020; Wang et al 2013). This is the first report of pathogenic F. concentricum on pecan in Southeast China that caused no harvest of infected plants.
Interview data versus questionnaire data in the diagnosis of carpal tunnel syndrome in epidemiological studies.

Abstract:AIMS In most epidemiological studies on carpal tunnel syndrome (CTS), the case definition is based on questionnaire data with or without neurophysiological testing. The aim of this study was to test if the use of questionnaire data results in misclassification of cases. METHODS In two studies, involving 940 and 311 participants, respectively, the people indicating CTS symptoms (tingling) in a questionnaire were clinically interviewed. In a subpopulation (n = 404), all went through an interview regardless of their questionnaire answers. RESULTS Only 35-45% of the participants reporting tingling once a week or more in the questionnaire actually had symptoms consistent with CTS when interviewed. The remaining 55-65% had no or infrequent symptoms or symptoms because of other disorders. Few potential CTS cases were missed. The positive predictive values were 0.48 (95% CI = 0.30-0.66) and 0.52 (95% CI = 0.38-0.67) on the right and left hands, respectively. The sensitivities, specificities and negative predictive values ranged from 0.87 to 1.00. CONCLUSIONS Questionnaire information overestimates the prevalence of CTS symptoms. However, asking about tingling in a questionnaire is a simple and sensitive first step to detect potential CTS cases, but symptoms should be confirmed by interview.

Abstract:To develop a system for gene replacement in Nocardia corallina, a protocol for electroporation was optimized by systematic alterations of growth conditions, field strength, time constant and the electroporation buffer. Transformation efficiencies of 0.5 x 10(6) - 3 x 10(6) transformants/microgram plasmid DNA were obtained routinely. The gene encoding the polyhydroxyalkanoate (PHA) synthase I of N. corallina was cloned and interrupted by insertion of a kanamycin-resistance gene. The resulting plasmid was introduced into N. corallina by electroporation to inactivate the wild-type gene by homologous recombination. Kanamycin-resistant clones were screened by Southern hybridization for the absence of the wild-type gene and analyzed for PHA accumulation.
First Report of Colletotrichum Plurivorum Causing Anthracnose on Pecan (carya Illinoinensis) in China

Abstract:Pecan (Carya illinoinensis) is an economically important nut crop worldwide (Xiao et al 2021). Anthracnose symptoms were found on pecan fruits and leaves in plantations in Anhui and Jiangsu provinces, China in August 2019. Irregular, dark brown or black spotted lesions firstly appeared on the surface and inside of fruits, and spread to all leaves. The symptoms resulted in 30% to 50% leaf drop and nearly a half of fruit decay in almost all trees of the susceptible cv. Wichita. The causal agent were isolated from fruits with target symptoms following the steps: surface disinfected with 75% ethanol (2×, 30 s), rinsed with sterile deionized water (3×), ~ 0.5 cm small fragments of the fruits excised and plated on potato dextrose agar (PDA) medium and incubated at 28 °C in dark for 3-d. Mycelium of each colony was picked and incubated on fresh PDA at 25 °C with a 12-hour light/dark cycle for 6-d to induce conidia formation. One 5-mm hyphal plug produced from each single spore isolate was transferred onto fresh PDA to obtain the pure cultures. Koch's postulates was employed for pathogenicity determination of the isolates. Non-wounded healthy leaves from seedlings of the disease susceptible cv. Pawnee were disinfected with 1% NaClO and inoculated with 5-mm 5-d hyphal of each isolate at 25 ℃. Tiny lesion spots were visible on the leaves after 2 days post inoculation (DPI) with isolate W-6 (the only pathogenic one among all isolates), and expanded over time until to the leaves withered, while the control leaves and leaves inoculated with other isolates remained asymptomatic. The pathogenicity of W-6 were confirmed using leaves and fruits of living Pawnee trees growing in Linglong Mountain Plantation, Lin'an, Hangzhou, China (119⁰38'51″E, 30⁰12'39″N, elevation: 119m). Three experimental replicates were conducted separately with three bio-replicates for all pathogenetic testing. The same symptoms were observed on both detached leaves and leaves and fruits of living trees.. The colony of W-6 have round cottony mycelium with complete edges and showed the fastest growth rate 3 - 4 DPI. After 7 DPI, white aerial mycelium turned yellowish brown and formed Acervulus in the mycelium. Conidia (n=50) one-celled, 12.0 - 20.0 μm × 3.5 to 6.0 μm width. Hyaline cylindrical with slightly rounded ends and two or three large guttulate at the centre. Most Acervulus dark brown and slightly irregular in shape, 12.70 × 18.79 μm (n=10). Setae were dark brown in color with average length around 34.10 μm (n=10). These characteristics matched previous descriptions of Colletotrichum orchidearum species complex, including C. plurivorum (Damm et al 2019). The identity of W-6 was confirmed by multi-locus phylogenetic analysis using the internal transcribed spacer (ITS) rDNA region and partial sequences of the conserved genes glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), beta-tubulin 2 (TUB2), and chitin synthase (CHS). The sequences of W-6 were used for Basic Local Alignment Search Tool (BLAST) against NCBI GenBank and the sequences with 100% identity to that of W-6 were achieved, respectively. The concatenated sequences of the ACT-CHS-GAPDH-ITS-TUB2 was used for building a phylogenetic tree. The molecular analyses allowed the identification of the pathogen as C. plurivorum. It was known that 9 of the 11 Colletotrichum species causing pecan anthrax worldwide were reported in southern China (Brenneman 1989; Oh et al 2021). This is the first report of C. plurivorum as causal agent of pecan in China.

Abstract:Natural rubber is an important industrial raw material and an economically important perennial in China. In recent years, A new leaf fall disease, caused by Neopestalotiopsis aotearoa Maharachch., K.D. Hyde & Crous, has occurred in Indonesia, Malaysia, Thailand, Sri Lanka, and other major rubber planting countries. In May and July of 2020, this disease was first found on 2-year-old rubber seedlings in two plantations located in Ledong and Baisha counties in Hainan Province, China. In the two plantations of approximately 32 ha, 15% of the rubber seedlings had the disease and the defoliation was more than 20%. The infected leaves turned yellow and watery, and dark brown and nearly round lesions of 1-2 mm in diameter were formed on the leaves. When the humidity was high, the center of the lesion was grey-white, and the lesions had many small black dots, black margins and surrounded by yellow halos. When the disease was severe, leaves fell off. To identify the pathogen, leaf tissues were collected from lesion margins after leaf samples were surface-sterilized in 75% ethanol, rinsed with sterile water for three times, and air dried. The leaf tissues were plated on potato dextrose agar (PDA) and incubated at 28°C for seven days. Fungal cultures with similar morphology were isolated from 90% of tested samples and two isolates (HNPeHNLD2001 and HNPeHNLD2002) were used in pathogenicity and molecular tests. Rubber leaves (clone PR107) were inoculated with conidial suspension (106 conidia/ml), and inoculated with PDA were used as the control, Each treatment had 3 leaves, and each leaf was inoculated with 3 spots and incubated at 28oC under high moisture conditions. Five days later, leaves inoculated with conidial suspension showed black leaf spots resembling the disease in the field, whereas the control leaves remained symptomless. The fungal cultures isolated from the inoculated tissues, had identical morphology compared with the initial isolates. Colonies on PDA were 55-60 mm in diameter after seven days at 28°C, with undulate edges, pale brown, thick mycelia on the surface with black, gregarious conidiomata; and the reverse side was similar in color. Black conidia were produced after eight days of culture on PDA. Conidia were fusoid, ellipsoid, straight to slightly curved, 4-septate, ranged from 18.35 to 27.12 μm (mean 22.34 μm) × 4.11 to 7.03 μm (mean 5.41 μm). The basal cells were conic with a truncate base, hyaline, rugose and thin-walled, 4.35 to 6.33 μm long (mean 4.72 μm). Three median cells were doliform, 12.53 to 18.97 μm long (mean 15.26 μm), hyaline, cylindrical to subcylindrical, thin- and smooth-walled, with 2-3 tubular apical appendages, arising from the apical crest, unbranched, filiform, 14.7 to 25.3 μm long (mean 19.94 μm). The basal appendages were singlar, tubular, unbranched, centric, 3.13 to 7.13 μm long (mean 5.48 μm). Morphological characteristics of the isolates were similar to the descriptions of N. aotearoa (Maharachchikumbura et al. 2014). The rDNA internal transcribed spacer (ITS) region, translation elongation factor 1-αgenes (TEF), and beta-tubulin (TUB2) gene were amplified using the primer pairs ITS1/ITS4, EF1-728F/EF1-986R and T1/Bt-2b (Pornsuriya et al. 2020), respectively. The sequences of these genes were deposited in GenBank (ITS Accession Nos.: MT764947 and MT764948; TUB2: MT796262 and MT796263; TEF: MT800516 and MT800517). According to the latest classification of Neoprostalotiopsis spp. (Maharachchikumbura et al. 2014) and multilocus phylogeny, isolates HNPeHNLD2001 and HNPeHNLD2002 were clustered in the same branch with N. aotearoa. Thus, the pathogen was identified as N. aotearoa, which is different from N. cubana and N. formicarum reported in Thailand (Pornsuriya et al. 2020; Thaochan et al. 2020). The Neopestalotiopsis leaf spotdisease of rubber tree (H. brasiliensis) was one of the most serious and destructive leaf diseases in major rubber planting countries in Asia. ( Tajuddin et al. 2020) The present study of leaf fall disease on rubber tree caused byN. aotearoa is the first report in China. The finding provides the basic pathogen information for further monitoring the disease and its control.

First Report of Blight Caused by Rhizoctonia solani AG4-HGI on Pinellia ternata in Guizhou, China

Abstract:Pinellia ternata (Thumb.) has been used for over 1000 years as a traditional Chinese herbal medicine (Ying et al. 2007) and is widely cultivated in Guizhou Province, China. It is cultivated over an area of 2000 hectares, and is of great value to underdeveloped regions. In April 2020, blight was observed in a field of P. ternatain Bijie County, Guizhou Province, China (27°30'N, 105°28'E). Around 20 hectares of P. ternata were surveyed and the disease incidence ranged from 10 to 12%. The disease symptoms included light brown lesions formed on the stems near the soil line. The color of the lesions became darker, and the stems became constricted around the lesions and broke, associated with the leaf blight. To identify the causal agent of this blight, 22 diseased plants (about 30 d-old） were collected, the margins of the infected parts were cut into small pieces (5 mm) and surface disinfested with 1% NaOCl for 10 min, 75% ethanol for 30 s, and rinsed three times in sterile distilled water. The pieces were blotted dry with sterile filter paper and placed on potato dextrose agar (PDA, Hopebio, China), incubated at 28℃ in darkness until fungal hyphae growth was visible. Sixteen cultures with different morphologies were recovered from the samples. When representative isolates of each culture type were inoculated onto plants, one produced similar blight symptoms. The representative isolate was called CD-1. The colony color was first white but turned light brown after grown on PDA for 6-7 d, and produced dark brown sclerotia. The hyphae were branched at right angles, with a slight constriction at the base of the branches and a septum near the junction where the branch separates from the main hyphae. Hyphal cells were stained with 0.5% Safranin O and 3% KOH and were observed to be multinucleate. These morphological features indicated that CD-1 likely is R. solani (Sneh et al. 1991). When paired with tester strains AG1 and AG4(provided by Dr. Genhua Yang, Yunnan Agricultural University). CD-1 showed anastomosis with isolate of AG4 (Fenille et al. 2002). Genomic DNA was extracted from the isolate (Thangaraj et al. 2018) using a fungal genomic DNA extraction kit (Tiangen, China). The internal transcribed spacer (ITS) regions were amplified using the primers ITS1/ITS4 (White et al. 1990). A 535 bp fragment was amplified that showed 99% coverage and 99.4% identity with an isolate of R. solani AG4-HGI (GenBank: HG934417). The gene sequence was deposited in GenBank as accession #OL518945. Pathogenicity tests were performed using 30 d-old plants planted in sterilized soil in pots. Cut mycelial discs (diameter 6 mm) from 3-day-old PDA cultures and placed beside stems of 21 healthy plants. Nine plants treated with agar plugs were control samples. Inoculated plants were maintained at 24 ± 5℃ in a green house and watered every two days with sterilized water. Typical blight symptoms developed on the inoculated plants at d 3-5 post inoculation, whereas the control plants remained healthy. The experiments were repeated three times, and the isolates was re-isolated from the inoculated plants and identified as R. solaniAG4 by morphological features and molecular method. R. solani has been reported to cause blight of many plants such as coffee (Ren et al. 2018) and sesame (Cochran et al. 2018). To the best of our knowledge, this is the first report of R. solani AG4-HGI causing disease on P. ternate, both in China and worldwide. This finding suggests that this pathogen may cause a threat to cultivation and production of P. terenata.
First report of Neopestalotiopsis rosae causing leaf blight on Shatangju in southern China

Abstract:Shatangju (Citrus reticulata Blanco cv. Shatangju) belongs to genus Citrus and was cultivated extensively in southern China. In April 2022, a leaf blight-like symptom (firstly brown spots appeared on infecting leaves, then these brown spots extended, finally the whole leaves displayed blight-like symptom) was observed on 5%~10% of Shatangju seedlings (around five hundreds in total) in an orchard located in Wuhan city, Hubei, China. Diseased leaves from three seedlings were collected and cut into pieces (0.2 to 0.5 cm). These pieces were surface-sterilized using 75% ethanol for 3 min, rinsed with sterile distilled water for several times, then placed on potato dextrose agar (PDA) and incubated at 26°C with 12-h light/dark cycle. Over 20 pieces plated, wherein 30% were identified as Colletotrichum fructicola, 60% as Neopestalotiopsis spp., and 10% developed saprophytes. C. fructicola was a known pathogen on citrus, thus Neopestalotiopsis spp. was further investigated. Eight single-conidium colonies of the Neopestalotiopsis spp. were obtained, wherein STJ-8 was chosen as a representative for further study. The average growth rate of STJ-8 was 15.1±0.5 mm/day (n=5). Fungal colonies produced white cottony mycelium with abundant black acervuli distributed in concentric rings 6-8 days after planting, which ranged from 342.3 to 710.5 μm in diameter (n=100). Conidia were fusoid, five cells, four septa with average dimensions of 25.36×5.47 μm (n=100). Basal and apical cells were hyaline, wherein three middle cells were brown with darker septa. The apical cell was cylindrical with two to three transparent accessory filaments (13.7 to 30.5 µm in length, n=80). Basal cell was conic with an appendage (4.1 to 8.8 µm in length, n=40). Partial sequences of internal transcribed spacer (ITS), translation elongation factor 1-alpha (TEF-1α), and β-tubulin (TUB2) were amplified with reported primers (White et al. 1990; Lee et al. 2006; Maharachchikumbura et al. 2014), sequenced, and submitted to GenBank (accession nos. ITS: OP236541; TEF-1α: OP250124; TUB2:OP263094). BLASTn results showed 100% identity with the corresponding sequences of Neopestalotiopsis rosae. A multilocus phylogenetic analysis showed STJ-8 was closest to N. rosae. Thus, STJ-8 was identified as N. rosae. Pathogenicity tests were performed on one-year-old Shatangju seedlings and detached primary leaves by inoculating needle-wounded leaves with seven days old 5-mm mycelial plugs/acervuli (about 5000 spores) of STJ-8. Control seedlings/leaves were inoculated with 5-mm PDA plugs/sterile water drops. All inoculated detached leaves were cultured at same the place with STJ-8 cultured, while inoculated seedlings were put in a growth chamber at 26°C under a 16-h light/dark cycle (60% humidity). Symptoms developed on all inoculated leaves (except healthy control) 2 and 4 days post-inoculation by mycelial plugs and acervuli, respectively. N. rosae was re-isolated from the inoculated leaves, confirming Koch's postulates. N. rosae has been reported to cause diseases on various plants worldwide (Rebollar-Alviter et al. 2020; Xavier et al. 2021; Lawrence et al. 2022). In China, N. rosae has been reported to cause leaf spot/blight on pecan and strawberry (Wu et al. 2021; Gao et al. 2022), which caused great loss on these crops. To our knowledge, this is the first report of N. rosae causing leaf disease on citrus. Our study is important for developing control strategies against N. rosae in future.
First Report of Medical Tree Peony Root Rot Caused by Fusarium solani in Tongling, China

Abstract:Tree peony bark, a main component of Chinese traditional medicine used for alleviating fever and dissipating blood stasis, is mainly produced in Tongling, China. Recently, tree peony cultivation in this area was seriously affected by root rot, with approximately 20 to 30% disease incidence each year. The disease severely affects yield and quality of tree peony bark. During the past 2 years, we collected 56 diseased tree peony plants from Mudan and Fenghuang townships in Tongling. We found reddish brown to dark brown root rot in mature roots, especially on those with injuries. Plant samples collected were disinfected with 2% sodium hypochlorite and isolations were conducted on potato sucrose agar (PSA). Eleven isolates were obtained and all had white fluffy aerial hypha on PSA. Two types of conidia were produced; the larger, reaphook-shaped ones had three to five septa and the smaller, ellipse-shaped ones had one or no septum. The reaphook-shaped conidia were 20.15 to 37.21 × 3.98 to 5.27 μm and the ellipse-shaped conidia were 6.02 to 15.52 × 2.21 to 5.33 μm in size. Chlamydospores were produced, with two to five arranged together. Biological characteristics of the fungi indicated that the optimum temperature for the mycelial growth on PSA was 25 to 30°C and the optimum pH range was 5.5 to 7.0. The above morphological characteristics point the fungal isolates to be Fusarium solani. To confirm pathogenicity, 30 healthy 1-year-old tree peony seedling plants were grown in pots (25 cm in diameter) with sterilized soil and a conidial suspension from one isolate (FH-1, 5 × 105 conidia/ml) was used for soil inoculation. Inoculated seedlings were maintained at 28°C in a greenhouse with a 12-h photoperiod of fluorescent light. Seedlings inoculated with distilled water were used as controls. After 3 weeks, the roots were collected and rinsed with tap water. Dark brown lesions were observed in the inoculated mature roots but not in the control roots. To confirm the identity of the pathogen, F. solani strains were reisolated from the lesions and total genomic DNA was extracted with the cetyltriethylammnonium bromide method from the mycelia of the reisolated strains (1). PCR was performed using the fungal universal primers ITS4 (5'-TCCTCCGCTTATTGATATGC-3') and ITS5 (5'-GGAAGTAAAAGTCGTAACAAGG-3') to amplify a DNA fragment of approximately 590 bp. The purified PCR products were sequenced (Invitrogen Co., Shanghai, China) and shared 100% sequence identity with each other. A comparison of the sequence (JQ658429.1) by the Clustal_W program (2) with those uploaded in GenBank confirmed with the fungus F. solani (100% sequence similarity to isolate S-0900 from the Great Plains of the United States; EU029589.1). To our knowledge, this is the first report of F. solani causing medical tree peony root rot in China.
