Abstract:Introduction: Environmental conditions play a prime role in the growth and development of plant species, exerting a significant influence on their reproductive capacity. Soybean is sensitive to high temperatures during flowering and seed developmental stages. Little is known about the combined environmental effect of temperature and CO2 on seed yield and quality and its future generation. Methods: A study was conducted to examine the effect of temperature (22/14°C (low), 30/22°C (optimum), and 38/30°C (high)), and CO2 (420 ppm (ambient; aCO2) and 720 ppm (elevated; eCO2)) on seed yield, quality, and transgenerational seedling vigor traits of soybean cultivars (DS25-1 and DS31-243) using Soil-Plant-Atmospheric-Research facility. Results: A significant temperature effect was recorded among yield and quality attributes. At high-temperature, the 100-seed weights of DS25-1 and DS31-243 declined by 40% and 24%, respectively, over the optimum temperature at aCO2. The harvest index of varieties reduced by 70% when exposed to high temperature under both aCO2 and eCO2, compared to the optimum temperature at aCO2. The seed oil (- 2%) and protein (8%) content altered when developed under high temperature under aCO2. Maximum sucrose (7.5%) and stachyose (3.8%) accumulation in seeds were observed when developed under low temperatures and eCO2. When the growing temperature increased from optimum to high, the seed oleic acids increased (63%), while linoleic and linolenic acids decreased (- 28% and - 43%, respectively). Significant temperature and CO2 effects were observed in progenies with the highest maximum seedling emergence (80%), lesser time to 50% emergence (5.5 days), and higher seedling vigor from parents grown at low-temperature treatment under eCO2. Discussion: Exposure of plants to 38/30°C was detrimental to soybean seed yield, and eCO2 levels did not compensate for this yield loss. The high temperature during seed developmental stages altered the chemical composition of the seed, leading to an increased content of monounsaturated fatty acids. The findings suggest that parental stress can significantly impact the development of offspring, indicating that epigenetic regulation or memory repose may be at play.

Abstract:Improvements in genetics, technology, and agricultural intensification have increased soybean yields, however adverse climate conditions may prevent these gains from being fully realized in the future. Higher growing season temperatures reduce soybean yields in key production regions including the Midwest U.S and better understanding of the developmental and physiological mechanisms that constrain soybean yield under high temperature conditions is needed. This study tested the response of two soybean cultivars to four elevated temperature treatments (+1.7, +2.6, +3.6 and +4.8 oC) in the field over three growing seasons and identified threshold temperatures for response and linear vs. nonlinear trait responses to temperature. Yield declined non-linearly to temperature, with decreases apparent when canopy temperature exceeded 20.9 oC for the locally adapted cultivar and 22.7 oC for a cultivar adapted to more southern locations. While stem node number increased with increasing temperature, leaf area index decreased substantially. Pod production, seed size and harvest index significantly decreased with increasing temperature. The seasonal average temperature of even the mildest treatment exceeded the threshold temperatures for yield loss, emphasizing the importance of improving temperature tolerance in soybean germplasm with intensifying climate change.

Abstract:Soybean is an important oilseed crop worldwide; however, it has a high sensitivity to temperature variation, particularly at the vegetative stage to the pod-filling stage. Temperature change affects physiochemical and genetic traits regulating the soybean agronomic yield. In this regard, the current study aimed to comparatively evaluate the effects of varying regimes of day and night temperatures (T1 = 20°C/12°C, T2 = 25°C/17°C, T3 = 30°C/22°C, T4 = 35°C/27°C, and T5 = 40°C/32°C) on physiological (chlorophyll, photosynthesis, stomatal conductance, transpiration, and membrane damage) biochemical (proline and antioxidant enzymes), genetic ( GmDNJ1 , GmDREB1G;1 , GmHSF-34 , GmPYL21 , GmPIF4b , GmPIP1;6 , GmGBP1 , GmHsp90A2 , GmTIP2;6 , and GmEF8 ), and agronomic traits (pods per plant, seeds per plant, pod weight per plant, and seed yield per plant) of soybean cultivars (Swat-84 and NARC-1). The experiment was performed in soil plant atmosphere research (SPAR) units using two factorial arrangements with cultivars as one factor and temperature treatments as another factor. A significant increase in physiological, biochemical, and agronomic traits with increased gene expression was observed in both soybean cultivars at T4 (35°C/27°C) as compared to below and above regimes of temperatures. Additionally, it was established by correlation, principal component analysis (PCA), and heatmap analysis that the nature of soybean cultivars and the type of temperature treatments have a significant impact on the paired association of agronomic and biochemical traits, which in turn affects agronomic productivity. Furthermore, at corresponding temperature regimes, the expression of the genes matched the expression of physiochemical traits. The current study has demonstrated through extensive physiochemical, genetic, and biochemical analyses that the ideal day and night temperature for soybeans is T4 (35°C/27°C), with a small variation having a significant impact on productivity from the vegetative stage to the grain-filling stage.

Twelve soybean genotypes were grown at day/night temperatures of 30/22, 34/24, 38/26 and 42/28 °C with an average temperature of 26, 29, 32 and 35 °C, respectively under green-house conditions. High temperature stress significantly increased duration of flowering and decreased number of flowers and pods formed as compared to ambient temperature. When plants were grown at elevated temperature pollen germination, pollen size and pollen tube length were declined leading to reduced reproductive efficiency which ultimately resulted reduction in seed yield. The average seed yield was maximum (13.2 g/plant) in plants grown under ambient temperature condition. Seed yield was declined by 8, 14, 51 and 65% as the plants were grown at 30/22, 34/24, 38/26 and 42/28 °C as compared to plants grown under ambient temperature conditions, respectively. The genotypes such as NRC 7 and EC 538828 showed less reduction in yield and stable reproductive biology as compared to other genotypes. It is concluded that for heat tolerance in soybean, breeding efforts needs to be focused on improving the reproductive efficiency.
Screening Soybean Genotypes for High-Temperature Tolerance by Maximin-Minimax Method Based on Yield Potential and Loss

Abstract:Temperature rise between 2.6 and 4.8 °C will impact the productivity of soybean at the turn of the twenty-first century. To predict differences in soybean genotypes to high temperatures, twelve soybean genotypes were grown in greenhouses maintained at a mean temperature of 26, 29, 32, and 35 °C, respectively, with one set in natural conditions. The leaf area, total biomass, photosynthesis, Fv/Fm, pollen germination, and reproductive efficiency were significantly high under natural conditions, and a further increase in temperature to 26, 29, 32, and 35 °C resulted in a decline in these parameters. The average seed yield of 12 soybean genotypes was 13.2 g/plant under ambient temperature and there was mild reduction of 8% and 14% when genotypes were grown at 26 and 29 °C, respectively. Severe decline by 51% and 65% in yield was observed at 32 and 35 °C, respectively. The total stress response index in twelve genotypes ranged from −1068 (JS 95-60) to −333 (EC 538828). NRC7 and EC 538828 performed comparatively better than other genotypes. Screening for high-temperature tolerance in soybean is very constrained in breeding programs. This genetic variability among soybean genotypes to elevated temperature reveals that heat tolerance can be improved through plant breeding programs. Additionally, it emphasizes the significance of identifying efficient selection strategies in improving the productivity of soybean in future climate scenarios.

The objectives of present study were to quantify the effect of high temperature on changes in leaf thickness, number of stomata on adaxial and abaxial leaf surfaces, gas exchange, chlorophyll fluorescence parameters and seed yield in soybean. Twelve soybean genotypes were grown at day/night temperatures of 30/22, 34/24, 38/26 and 42/28 °C with an average temperature of 26, 29, 32 and 35 °C, respectively, under greenhouse conditions. One set was also grown under ambient temperature conditions where crop season average maximum, minimum and mean temperatures were 28.0, 22.4 and 25.2 °C, respectively. Significant negative effect of temperature was observed on specific leaf weight (SLW) and leaf thickness. Rate of photosynthesis, stomatal conductance and water use efficiency declined as the growing temperatures increased; whereas, intercellular CO2 and transpiration rate were increased. With the increase in temperature chlorophyll fluorescence parameters such as Fv/Fm, qP and PhiPSII declined while there was increase in qN. Number of stomata on both abaxial and adaxial surface of leaf increased significantly with increase in temperatures. The rate of photosynthesis, PhiPSII, qP and SPAD values were positively associated with leaf thickness and SLW. This indicated that reduction in photosynthesis and associated parameters appears to be due to structural changes observed at higher temperatures. The average seed yield was maximum (13.2 g/pl) in plants grown under ambient temperature condition and declined by 8, 14, 51 and 65% as the temperature was increased to 30/22, 34/24, 38/26 and 42/28 °C, respectively.

Abstract:Extreme heat events prolong the reproductive period and threaten soybean yield, whereas the specific stage at which individual fruits growth is delayed, and yield/yield components at the node, region, and plant levels under short-term heat stress in the reproductive stage are elusive. In this study, heat treatments (40/30 degrees C) were applied at 0-6 days (HTF0-6), 6-12 days (HTF6-12), 12-18 days (HTF12-18), and 0-12 days (HTF0-12) after the plant's first flower opened, and a control treatment (32/22 degrees C) was performed. The influences of heat stress on fruit development and yield/yield components at the node, region, and plant levels were investigated. As a result, the growth of individual fruits at nodes was delayed by HTF0-6 and HTF0-12, which was primarily caused by the prolongation of flowering to pods with a length of 2 cm. Interestingly, there were no significant differences in yield between the control treatment and the various high-temperature stress treatments at the plant level. Further analysis of the regional yield of soybean showed that the yield in the bottom and top regions of plants played significant roles in compensating for yield loss in the middle region after HTF0-12. Moreover, the delayed growth of individual fruits in the middle region was negatively correlated with yield. Our results indicate that the prolongation of fruit development induced by HTF0-6 and HTF0-12 may adversely affect soybean yield. However, the spatial compensation of plants could help maintain soybean yield under various short-term high temperature stress treatments during the reproductive period, which should be considered when breeding for and selecting heat-tolerant varieties.

Abstract:Ambient temperatures are predicted to rise in the future owing to several reasons associated with global climate changes. These temperature increases can result in heat stress- a severe threat to crop production in most countries. Legumes are well-known for their impact on agricultural sustainability as well as their nutritional and health benefits. Heat stress imposes challenges for legume crops and has deleterious effects on the morphology, physiology, and reproductive growth of plants. High-temperature stress at the time of the reproductive stage is becoming a severe limitation for production of grain legumes as their cultivation expands to warmer environments and temperature variability increases due to climate change. The reproductive period is vital in the life cycle of all plants and is susceptible to high-temperature stress as various metabolic processes are adversely impacted during this phase, which reduces crop yield. Food legumes exposed to high-temperature stress during reproduction show flower abortion, pollen and ovule infertility, impaired fertilization, and reduced seed filling, leading to smaller seeds and poor yields. Through various breeding techniques, heat tolerance in major legumes can be enhanced to improve performance in the field. Omics approaches unravel different mechanisms underlying thermotolerance, which is imperative to understand the processes of molecular responses toward high-temperature stress.

Impact of heat stress during seed development on soybean seed metabolome

Abstract:Seed development is a temperature-sensitive process that is much more vulnerable than vegetative tissues to abiotic stresses. High temperatures during soybean (Glycine max) seed development frequently results in seed with poor germination, increased incidence of pathogen infection, and decreased economic value. Climate change is expected to increase the incidence and severity of summer heatwaves, and the impact of heat stress on seed development will become more widespread during the course of the twenty-first century. Global metabolite profiles were contrasted between seed from heat-tolerant and heat-susceptible genotypes produced under 28/22 °C (control), 36/24 °C, and 42/26 °C day/night temperatures. Germination of seeds from the heat-susceptible genotype was strongly reduced (50 %) for the 36/24 °C treatment and completely inhibited for the 42/26 °C. In contrast, germination was unaffected for the heat-tolerant genotype for the 36/24 °C, and while strongly inhibited, some (25 %) seed from the heat-tolerant genotype were able to germinate even for the 42/26 °C treatment, and in general were less impacted by elevated temperatures as compared to a commonly grown high-yielding, but heat-sensitive genotype. A total of 275 seed metabolites were analyzed by three metabolite profiling methods, and genotype-specific differences and temperature specific differences were identified. A diverse set of antioxidant metabolites, including tocopherols, flavonoids, phenylpropanoids, and ascorbate precursors were found to be enriched in seed of the heat tolerant genotype. The generally greater abundance of these metabolites at both temperatures in the tolerant genotype indicates that these compounds are likely responsible, at least in part, for the greater tolerance to high temperatures during seed development.

Abstract:A pot culture experiment was conducted under controlled condition to assess the effect of drought, high temperature and their combined effects during flowering and pod initiation stages in four soybean genotypes. Reponses of genotypes were measured in terms of physiological traits including gas exchange parameters, chlorophyll content, osmotic potential and relative water content. Invariably, drought, high temperature and combination of stresses significantly affected all the physiological parameters studied. Interestingly, differential responses were observed among the genotypes for the physiological parameters which were used to identify the tolerant and susceptible genotypes under drought, high temperature and combination of stresses. In case of photosynthetic rate, high temperature stress showed two-fold reduction, while drought stress displayed more serious effect leading to three-fold reduction in the photosynthetic rate of soybean genotypes. Transpiration rate increased under elevated temperature than control conditions while it decreased in drought and combined stress in all the genotypes. Chlorophyll fluorescence (Fv/Fm) in ADT 1 and CAT 1260 showed least reduction under stress condition indicating the ability to withstand the stress, while susceptible genotypes CAT 2084 showed drastic reduction. Higher stability of chlorophyll under stress was observed in the genotypes ADT 1 followed by CAT 1260 and least in CAT 2084. Relative water content and osmotic potential was found higher in ADT 1 indicating the tolerance to individual and combined stresses. These findings suggest that maintenance of higher transpiration rate and lower leaf temperature under drought, high temperature and combination of stress, as adaptive traits associated with tolerance of ADT 1 and CAT 1260. These identified genotypes can be used directly for the region where the drought and high temperature is predominant or they can be used as parents in breeding program to develop tolerant varieties under combined stresses of drought and high temperature.
Low temperature and excess moisture affect seed germination of soybean (Glycine max L.) under controlled environments

Abstract:Cold and excess moisture pose a serious threat to soybean production especially during seed germination in short-season environments. In this study, the effects of low temperature and excess moisture stress on seed germination were investigated in 187 soybean accessions originating from 18 countries. The experiment used a combination of three temperature conditions (i.e., 20°C/14°C d/night, 14°C/10°C, 10°C/10°C) and two moisture levels (i.e., normal and excess). The seed germinability traits measured included germination rate, germination index, germination time, germination uniformity, and coefficient of velocity of germination. Overall, germination rate was lowest in the ‘20°C/14°C + excess moisture’ and germination time was longest in the ‘10°C/10°C + excess moisture’. When compared with ‘20°C/14°C + normal moisture’ treatment, germination rate at ‘10°C/10°C + excess moisture’ decreased by 38%; germination time increased by 20 days; seed viability decreased by 83%; germination uniformity decreased by 70%; and germination speed decreased by 73%. Differences in germination rate, germination index and germination velocity under different treatments were affected by temperature, moisture and their interaction. Variation in germination time uniformity was determined by temperature, with no significant effects of moisture conditions and the interaction of temperature and moisture. It was shown that the temperature-excess moisture interaction led to a sharp decrease in seed germination. Two genotypes including PI 603147 and PI 507702 were identified with a GR over 90% at ‘10°C/10°C + excess moisture’. Abstract:Drought and heat stress are important abiotic stresses restricting plant growth, while the two stresses often occur at the same time in nature, little is known about when these stresses occur in combination. Therefore, attempts were made to understand the impact of water stress imposed under different temperature conditions on various physiological parameters such as nitrogen fixation, photosynthesis, chlorophyll fluorescence, seed yield and quality. Cultivar JS 97–52 was grown in pots under polyhouses maintained at day/night temperatures of 30/22, 34/24, 38/26 and 42/28 °C. At each temperature, pots were divided into two sets, one set was unstressed while second was subjected to water stress at reproductive stage (beginning of seed fill). An increase in temperature and water stress caused the reduction in CO2 exchange, which ultimately affected the nitrogen fixation. Nitrogenase activity was significantly declined in the nodules with an increase in temperature. There was a concomitant decline in leghemoglobin, heme-chrome and ureids content in the nodules which ultimately resulted in the reduction of nitrate reductase activity, chlorophyll content and total free amino acid under high temperatures. A decline in nitrogen and carbon fixation ultimately caused a reduction in seed yield and quality. Water stress when imposed at different temperature further aggravated the effects of temperature, and the combination of water stress and high temperature had more detrimental effect. Thus, understanding the mechanisms by which plants acclimate to water deficit combined with high temperature is crucial for identifying stress tolerant soybean cultivars and to pave the way for obtaining cultivars with higher productivity under combined stress conditions.

Abstract:Seed germination is an important stage in crop development that affects plant performance, crop yield, and quality. Many factors influence seed germination, and one of the most important factors is temperature. The closer the temperature is to the optimum, the faster germination will occur. Temperature affects seed germination in various plants and varies depending on the variety. Therefore, the effect of temperature on seed germination is necessary to investigate, also for soybean varieties. Soybean is one of the world's most valuable oil-seed crops. Generally, proper seedling establishment and germination of soybean seeds are crucial processes in the survival and growth cycle of the crop. Thus, a study was done to investigate the influence of temperature and variety on soybean seed germination at different times after sowing. The experiment was carried out at the Institute of Agronomy, Hungarian University of Agriculture and Life Sciences (MATE), Crop Production Laboratory. Seeds of two soybean varieties were subjected to three different temperatures: 15, 25, and 35 °C, and two soybean varieties, Martina and Johanna were tested. There were four germination times based on the days after sowing: Days 3, 5, 7, and 9. This experiment was set up with a completely randomized block design and four replications. According to the findings of this study, the Martina variety showed a better germination rate as well as higher vigor and viability than the Johanna variety in the first 12 days after sowing. At temperatures of 15 °C and 25 °C, both varieties achieved comparable percentages of viability, but they were more vigorous at 25 °C due to better total seedling length. Thus, the information gained from this study will indirectly determine and confirm the proper temperature for the initial growth of the Martina and Johanna varieties.

Abstract:Introduction: Soybean is sensitive to light and temperature. Under the background of global asymmetric climate warming. Methods: The increase of night temperature may have an important impact on soybean yield. In this study, three varieties with different level of protein were planted under 18°C and 28°C night temperatures for investigating the effects of high night temperatures on soybean yield formation and the dynamic changes of non-structural carbohydrates (NSC) during the seed filling period (R5-R7). Results and discussion: The results indicated that high night temperatures resulted in smaller seed size, lower seed weight, and a reduced number of effective pods and seeds per plant, and thus, a significant reduction in yield per plant. Analysis of the seed composition variations showed carbohydrates were more substantially affected by high night temperature than protein and oil. We observed "carbon hunger" caused by high night temperature increased photosynthesis and sucrose accumulation in the leaves during the early stage of high night temperature treatment. With elongated treated time, the excessive carbon consumption led to the decrease of sucrose accumulation in soybean seeds. Transcriptome analysis of leaves after 7 days of treatment showed that the expression of most sucrose synthase and sucrose phosphatase genes decreased significantly under the high night temperature. Which could be another important reason for the decrease of sucrose. These findings provided a theoretical basis for enhancing the tolerance of soybean to high night temperature.

Abstract:Germination is a key event in plant life cycle and imbibition temperature is an important factor for the major reorganization pro- cesses in the germinating seeds. Sub-physiological temperature at this stage affects virtually all aspects of cellular function including protein folding, kinetic parameters of membrane fluidity, protein assembly, and general metabolic processes. In this article we will review the re- search which has been carried out to decipher the basic mechanisms of low temperature (LT) stress with special emphasis on germination. With the better understanding of LT tolerance in some plant species we will also discuss how these attributes can be transferred in the im- portant food crops to attain better germination stamina at sub-physiological temperatures. At germination stage, the cellular machinery in the embryo is not at its proper place and is going through reorganisation and any environmental severity has devastating effects on the fragile plant. But LT resistant species have evolved the ability to acclimatise, with the remodelling of cell and tissue structures and the reprogram- ming of metabolism and gene expression. Metabolic networks are redirected towards the synthesis of cryoprotectant molecules, which in association with other proteins bring about physical and biochemical restructuring of cell membranes through changes in the lipid composi- tion and induction of other non-enzymatic proteins that alter the freezing point of water. Genetic engineering of crops for enhanced seed germination performance will certainly help to achieve optimum agricultural yields..

Abstract:With global warming, the impact of high‐temperature stress on crop production is one of the major issues facing agriculture. Combining a series of field and controlled environment experiments, the current study aimed at understanding causes of yield loss due to heat stress in lima bean (Phaseolus lunatus L.) in order to guide breeding for heat tolerance. A growth‐stage analysis of multi‐year field trials suggested that yields were reduced by sensitivity to heat during floral transition. High nighttime temperatures during this period were also associated with delayed pod set and subsequent harvest. These effects were validated in controlled environments, with additional tests revealing how nighttime heat stress generally reduces seeds per pod but can also reduce the number of pods set. Using an intra‐plant dual temperature treatment design for vegetative and reproductive organs, it was determined that effects on pollen viability and release are primary factors underlying heat stress‐associated yield losses in lima bean. Elevated nighttime temperatures did not reduce aboveground biomass, indicating the physiological basis for temperature sensitivity was not driven by a photosynthate deficit, but instead could be attributed to changes in source‐sink dynamics. Still, the basis for sensitivity to heat differed among genotypes, opening new targets for a multi‐mechanistic breeding approach for heat tolerance. This article is protected by copyright. Abstract:Elevated temperature and water deficit are the major abiotic factors restricting plant growth. While in nature these two stresses often occur at the same time; little is known about their combined effect on plants. Therefore, the main objective of the current study was to observe the effect of these two stresses on phenology, dry matter and seed yield in soybean. Two soybean genotypes JS 97-52 and EC 538828 were grown under green-house conditions which were maintained at different day/night temperatures of 30/22, 34/24, 38/26 and 42/28 °C with an average temperature of 26, 29, 32 and 35 °C, respectively. At each temperature, pots were divided into three sets, one set was unstressed while second and third set were subjected to water stress at vegetative and reproductive stage, respectively. As compared to 30/22 °C increase in temperature to 34/24 °C caused a marginal decline in leaf area, seed weight, total biomass, pods/pl, seeds/pl, harvest index, seeds/pod and 100 seed weight. The decline was of higher magnitude at 38/26 and 42/28 °C. Water stress imposed at two growth stages also significantly affected dry matter and yield. The highest average seed yield (10.9 g/pl) was observed at 30/22 °C, which was significantly reduced by 19, 42 and 64% at 34/24, 38/24 and 42/28 °C, respectively. Similarly, compared to unstressed plants (11.3 g/pl) there was 28 and 74% reduction in yield in plants stressed at vegetative and reproductive stage. Thus, both temperature and water stress affected the growth and yield but the effect was more severe when water stress was imposed at higher temperatures. JS 97-52 was more affected by temperature and water stress as compared to EC 538828. Though drought is the only abiotic factor that is known to affect the water status of plants, but the severity of the effect is highly dependent on prevailing temperature.

Abstract:Heat stress is increasingly a problem in global agriculture production, both in increasing occurrences and extended durations. Understanding the molecular mechanisms of the soybean heat stress response is essential for breeding heat tolerant soybeans. Plant associated microbiomes are known to mitigate adverse effects from abiotic stress. Soybean heat stress studies have primarily focused on response to short periods of stress, and how soybean responds on a transcriptional level to a soil microbiome is poorly understood. We hypothesize a soil microbiome may help soybean survive long-term heat stress exposure. We used RNA-seq to measure the transcriptional responses in four soybean exposed to two temperature regimes and grown in two soil microbiome conditions. We identified unique responses to temperature based on the soil microbiome conditions and to the different genotypes, with fewer changes across genotypes in response to a soil microbiome. Our findings provide insights on the interaction of soil microbiome with heat stress response in soybean and identify gene targets to further study the soybean heat stress tolerance with applications to develop improved varieties.

Abstract:Many of the world’s important crops such as soybean are now widely distributed, beyond their original zones of natural selection and chilling temperature is one of the major abiotic environmental factors which limit their growth and yield. Various molecular mechanisms, underlying chilling stress and plant responses are yet to be discovered. Chilling temperatures impart maximum damage to the soybean seeds, sown in wet soils, during imbibition, causing poor germination rate, reduced seedling emergence, decreased seedling vigor, and ultimately severe loss in yield. Here we report the changes in proteome of soybean seeds exposed to low temperature (4°C) during imbibition in a known chilling-resistant soybean cultivar Z22, using two-dimensional gel electrophoresis (2-DE) and Matrix-Assisted Laser Ionization Time of Flight Mass Spectrometry (MALDI-TOF/MS). Total 40 protein spots were differentially expressed in response to low temperature, in which, 25 protein spots were up-regulated and fifteen protein spots were down-regulated. According to the search in primary databases, these proteins were a part of many metabolic pathways including, cell defense, energy, protein synthesis, cell growth/division, storage, transcription and transport. To investigate the change in mRNA levels during imbibition at 4°C, quantitative RT-PCR of nine up-regulated protein genes was carried out, which clearly demonstrated an increase in the transcript levels of five genes, whereas no change was observed for other four genes. Results of this study provide some invaluable insights about the response of soybean seeds to low temperature imbibition which include; (1) alcohol dehydrogenase I and RAB21 might contribute in decreasing the effect of anoxia, resulting from water up-take during imbibition, (2) stress-related proteins such as LEA and GST24 probably played a pivotal role in confronting low temperature stress and (3) expression of some crucial enzymes (malate dehydrogenase and phosphoenolpyruvate carboxylase) involved in TCA cycle was enhanced, which might be beneficial for seeds, in stress conditions during germination.

Abstract:Summary Flowering time is one of important agronomic traits determining the crop yield and affected by high temperature. When facing high ambient temperature, plants often initiate early flowering as an adaptive strategy to escape the stress and ensure successful reproduction. However, here we find opposing ways in the short‐day crop soybean to respond to different levels of high temperatures, in which flowering accelerates when temperature changes from 25°C to 30°C, but delays when temperature reaches 35°C under short day. phyA‐E1, possibly photoperiodic pathway, is crucial for 35°C mediated late flowering, however does not contribute to promoting flowering at 30°C. 30°C‐induced upregulation of FT2a and FT5a leads to early flowering, independent of E1. Therefore, distinct responsive mechanisms are adopted by soybean when facing different levels of high temperatures for successful flowering and reproduction.
