Abstract:Hydrothermal treatment (HT) is envisaged as a promising technology to treat the lignocellulosic biomass. HT temperature is an important parameter influencing the hydrolysate compositions such as organic compounds and potential inhibitors, and therefore affect the subsequential anaerobic digestion (AD) process. Herein, HT-AD was employed to treat the wheat straw-derived digestate. The HT temperature of 190 °C was proved to be the best performance with a higehst reducing sugar yield (45.05 mg g −1 ) in the hydrolysate and a highest methane yield (120.8 mL g TS −1 ) from the AD of the hydrolysate, which was 42.5% higher than the methane yield in the control without the hydrolysate addition (84.8 mL g TS −1 ). 3-Furaldehyde was the dominant organic in the hydrolysates. The HT temperature of 210 °C led to the presence of AD inhibitory moieties (e.g., phenols and furans) in the hydrolysate, resulting in a low methane yield. Although the treatments with the addition of 100% hydrolysate outperformed those of 50% hydrolysate in the methane yields in the late stage, the latter had higher methane yields in the first stage, suggesting that the additional ratios of hydrolysates should be carefully considered in AD, especially the detrimental effects of inhibitors and adaptability issues of AD consortia. The MiSeq sequencing showed that the hydrolysis/acidogenesis was dominant in the first stage, while methanogenesis became dominant in the late stage with the acetoclastic/hydrogenotrophic methanogens ( Methanosarcina and Methanobacterium ) enriched in the hydrolysate-feeding reactors. These findings demonstrated that a integration of HT-AD was a promising approach for the digestate valorization and to reduce the potential carbon emission from waste treatments.

Abstract:Anaerobic digestion (AD) with thermal hydrolysis (TH) pretreatment is a promising process for excess sludge treatment, while there lacks of the knowledge from full-scale process about the impact of sludge composition and characteristics on microbial community and performance. The sludge physiochemical indices, microbial community and performance data of four full-scale TH-AD plants were characterized, and their relationships was elucidated. The four plants were operated under almost similar total organic loading rate (OLR) but their methanogenesis performance differentiate into two groups, namely superior group (SupG) and the inferior group (InfG). In both groups, TH effectively solubilized particulate organic compounds, meanwhile raised the ammonia nitrogen (NH4+-N) and volatile fatty acid (VFA) concentration. Compared with the SupG, thermal hydrolyzed sludge of InfG had higher level of VFAs, NH4+-N and total chemical oxygen demand (tCOD), which showed higher inhibition effect on microbes, leading to a community with lower diversity, lower abundance of carbohydrate degrading functional guild, higher protein degrading one, and methanogens that adapted to limited substrates, and further declined the methane production rate. Thus, it was recommended that OLR alone was not sufficient for controlling the system in design and operation, the concentration of VFAs, NH4+-N and tCOD should be equally considered. Their higher concentration, together with the higher abundance of Defluviitoga and Proteiniphilum were recommended as indicators for inferior running condition. Our results proposed that microbial communities played a role of bridge between environmental factors and performance, provided implications for engineering ecology and operational regulation for full-scale sludge TH-AD process.

Abstract:Microbially driven anaerobic digestion (AD) processes are of immense interest due to their role in the biovalorization of biowastes into renewable energy resources. The function-versatile microbiome, interspecies syntrophic interactions, and trophic-level metabolic pathways are important microbial components of AD. However, the lack of a comprehensive understanding of the process hampers efforts to improve AD efficiency. This study presents a holistic review of research on the microbial and metabolic "black box" of AD processes. Recent research on microbiology, functional traits, and metabolic pathways in AD, as well as the responses of functional microbiota and metabolic capabilities to optimization strategies are reviewed. The diverse ecophysiological traits and cooperation/competition interactions of the functional guilds and the biomanipulation of microbial ecology to generate valuable products other than methane during AD are outlined. The results show that AD communities prioritize cooperation to improve functional redundancy, and the dominance of specific microbes can be explained by thermodynamics, resource allocation models, and metabolic division of labor during cross-feeding. In addition, the multi-omics approaches used to decipher the ecological principles of AD consortia are summarized in detail. Lastly, future microbial research and engineering applications of AD are proposed. This review presents an in-depth understanding of microbiome-functionality mechanisms of AD and provides critical guidance for the directional and efficient bioconversion of biowastes into methane and other valuable products.
Effect of sludge retention time on microbial succession and assembly in thermal hydrolysis pretreated sludge digesters: Deterministic versus stochastic processes

Abstract:Thermal hydrolysis process (THP) assisted anaerobic digestion (AD) has been demonstrated to be an efficient approach to improve biogas production and solids reduction. Given the faster reaction kinetics in the THP-AD system, reduction of sludge retention time (SRT) is possible. However, a comprehensive understanding of the effects of sludge retention time (SRT) on microbial dynamics and community assemblages is still lacking in THP-AD systems. Thus, twelve THP-AD reactors were operated at different SRTs (10-30 d) to fulfill the knowledge gap. Results showed that, although all the bioreactors displayed good performance, shorter SRT reactors (SRT 10 d) took a longer time to reach the stable state. The total biogas production at SRT of 10 d was lower than that at other longer SRTs, attributing to the limited hydrolytic/fermentative capacities of AD microbiomes. Different SRTs resulted in distinct succession patterns of AD microbiomes. THP sludge reduced the microbial diversity in all the bioreactors over time, but longer SRTs maintained higher biodiversity. Null model analysis suggested that THP-AD microbial community assembly was predominately driven by deterministic selection at the tested SRT range, but stochasticity increased with elevated SRTs, likely attributing to the immigrants from the feedstock. Phylogenetic molecular ecological networks (pMENs) analysis revealed more stable network structures at longer SRTs, evidenced by the lower modularity, shorter harmonic geodesic distance, and higher connectivity. The potential keystone taxa under varied SRTs were identified, some of which were hydrolytic/fermentative bacteria (e.g., Peptostreptococcus, Lutispora, Synergistaceae), suggesting that these species related to organic hydrolysis/fermentation even with low-abundance could still play pivotal ecological roles in maintaining the THP-AD microbial community structure and functions. Collectively, this study provides comprehensive and in-depth insights into the mechanisms underlying community assembly in THP-AD reactors, which could aid in diagnosing system stability.

Abstract:Anaerobic digestion (AD) with thermal hydrolysis pretreatment has been attracted widespread attention in recent years due to its high efficiency. However, few studies focus on systematical comparison of the downstream AD processes for thermal hydrolyzed sludge and their corresponding microbial community compositions, especially on those at pilot scale and above. Thus, this study systematically compared the long-term performance and microbial communities of two pilot-scale anaerobic reactors for thermal hydrolyzed sludge digestion under mesophilic and thermophilic conditions. The results presented that mesophilic anaerobic digestion (MAD) showed a better performance of methane production than thermophilic anaerobic digestion (TAD). The hydraulic retention time (HRT) needed to be longer than 12 days in MAD while 20 days in TAD to achieve the relatively high methane production, which could be explained by that the ammonia nitrogen accumulation especially the free ammonia determined in TAD was higher than that in MAD at all HRTs, emerging an inhibition of methane yield in TAD. High-throughput Illumina sequencing results demonstrated a more diverse microbial community in MAD than that in TAD. TAD was mediated by a suite of thermophiles, such as Coprothermobacter and Methanothermobacter, while taxa harbored in MAD mostly belonged to Bacteroidetes and relatively broad types of methanogens. In addition, hydrogenotrophic methanogens were the predominant of archaea communities in both digesters probably due to the relatively high concentration of ammonia nitrogen.

Abstract:Anaerobic digestion is promising for waste activated sludge (WAS) degradation. However, conventional processes were generally stuck with limited hydrolysis and poor pathogen destruction. Hyperthermophilic digestion at 70 °C has drawn attention in overcoming those issues at a relatively low energy requirement and operating difficulties. In order to illuminate its operation characteristics, a single-stage hyperthermophilic digester was controlled at 70 °C and operated continuously to degrade WAS. 88.7 mL/g VSadded of methane yield could be achieved in the hyperthermophilic system, fourfold higher than that in the mesophilic system. Kinetic analysis revealed that hyperthermophilic digestion was advantageous in converting the non-degradable fraction. Consequently, hydrolysis under the hyperthermophilic condition was able to be significantly improved. Above 10 d was necessary for the hyperthermophilic system to gain such a high methane production. In the case of stability, the organic loading of higher than 10.2 g VS/L/d resulted in increasing limitation from methanogenesis and accumulation of propionic, butyric and valeric acids. In addition to the dominant acetoclastic genus Methanothrix for methane production in the hyperthermophilic system, two hydrogenotrophic methanogens Methanospirillum and Methanothermobacter reached 18.84% and 8.31%, respectively. The genus Coprothermobacter, affiliated with the phylum Firmicutes, made more contribution to protein hydrolysis in the hyperthermophilic digester.

Abstract:"Temperature Staging and Biological Phasing" ('TSBP) is an improved two-phase anaerobic digestion (AD) technology. This technology hydrolyzes waste activated sludge (WAS) at 45 degrees C and converts methane at mesophilic temperature (35-38 degrees C), with hydraulic retention times of 3-5 d and 14-17 d, respectively. In this study, the performance and microbial community dynamics of full-scale TSBP-based sludge anaerobic digestion system were studied, and the technology was evaluated by energy balance and ecological benefit analysis. The stable operation for 390 d showed that the cumulative biogas yield was about 349,041 m(3), the maximum biogas yield rate was 563.68 L/kg VS, and the VS degradation rate of organic matters in the sludge was 47.19%. Proteobacteria and Firmicutes were found to be the dominant bacteria in both thermophilic and mesophilic reactors. Methanobacterium and Methanosarcina were the two most abundant methanogenic genera in the AD samples. The aceticlastic methanogenesis was likely the predominant production pathway of methane in AD processes based on metagenomics. The TSBP system operated stably, and the recovered energy could achieve energy self-sufficiency, which provided technical reference for the anaerobic treatment of sludge.
Mesophilic anaerobic digestion of thermally hydrolyzed sludge in anaerobic membrane bioreactor: Long-term performance, microbial community dynamics and membrane fouling mitigation

Abstract:Thermal hydrolysis (TH) was integrated into an anaerobic membrane bioreactor (AnMBR) to stimulate sludge bioconversion and methane production. Sludge solubilization degree and methane productivity increased with TH temperature. The optimal performance was observed at 125 °C, with a 2.56-time increase in soluble chemical oxygen demand (1116.2 ± 62 mg/L). Methane production rate approached 73.2 ± 7.5 mL/Lreactor/d, rising by 2.96 times. Integration of TH effectively mediated membrane fouling, inducing a stable membrane flux of 10.8 ± 0.5 L/m2/h and low transmembrane pressure (TMP, ≤4.7 kPa). Cake layer deposited on the membrane surface accounted for 66.1 ± 1.0% of total resistance, with proteins and polysaccharides as the main contributors to membrane fouling (or increased TMP) (protein: Rp = 0.89, p0.05 = 0.04; polysaccharide: Rp = 0.99, p0.05 = 0.01). 16s rRNA gene analysis demonstrated that utilization of TH could regulate microbial diversity and enhance the proliferation of organics-degrading bacteria (i.e. Bacteroidetes, Firmicutes, etc.) and methane-producing methanogens (i.e. Methanosaeta, Methanobacterium, etc.), which accelerated the biodegradation of organics while alleviating membrane fouling and upgrading methane production. This study demonstrated the great potential of the combined TH-AnMBR system as a new strategy in promoting sludge solubilization, methane bioconversion and membrane fouling mitigation.

Abstract:Hyperthermophilic anaerobic digestion, especially at 70 oC, has drawn wide attention. In order to acquire the inoculum and digestion characteristics, batch acclimation and continuous operation experiments were conducted under hyperthermophilic (70 oC), thermophilic (55 oC) and mesophilic (35 oC) conditions, respectively. Archaea at each temperature was successfully enriched from the sole-source waste activated sludge (WAS). Hyperthermophilic digestion achieved higher archaea diversity, close to the Shannon index 2.23 for the thermophilic digestion, but the population were not improved, at a 16S rRNA genes 5.99×105 copies mL-1. Hydrogenotrophic methanogens, Methanospirillum and Methanothermobacter, dominated in the hyperthermophilic digester, accounting for 27.15%, while the primary phylum Firmicutes was promoted to 36.31%, with the proteolytic genus Coprothermobacter in Firmicutes at 19.50%. Refractory organic fractions were converted more with a higher digestion temperature, which was demonstrated by the fact that the COD/VS increased to 5.8, 5.2 and 4.2 at 70 oC, 55 oC and 35 oC, respectively, at the end of batch acclimation. In addition, the most solubilization for the dominant fraction protein in the WAS occurred at 70 oC as well. Similar hydrolysis ratio, over 10%, and specific hydrolysis rate, around 0.025 g COD (g VSS·d)-1, were achieved at 70 oC and 55 oC. The higher hydrolysis for hyperthermophilic digestion even resulted in a higher methane yield than that for the mesophilic digestion. Nevertheless, contrary to higher hydrolysis, methanogenesis limited hyperthermophilic digestion in WAS degradation, with an ultimate methane yield 71.2 mL g-1 VSadded, despite an almost complete VFA conversion through the continuous operation.

Optimizing anaerobic digestion: Benefits of mild temperature transition from thermophilic to mesophilic conditions

Abstract:Anaerobic digestion (AD) plays a significant role in renewable energy recovery. Upgrading AD from thermophilic (50-57 °C) to mesophilic (30-38 °C) conditions to enhance process stability and reduce energy input remains challenging due to the high sensitivity of thermophilic microbiomes to temperature fluctuations. Here we compare the effects of two decreasing-temperature modes from 55 to 35 °C on cell viability, microbial dynamics, and interspecies interactions. A sharp transition (ST) is a one-step transition by 20 °C d-1, while a mild transition (MT) is a stepwise transition by 1 °C d-1. We find a greater decrease in methane production with ST (88.8%) compared to MT (38.9%) during the transition period. ST mode overproduced reactive oxygen species by 1.6-fold, increased membrane permeability by 2.2-fold, and downregulated microbial energy metabolism by 25.1%, leading to increased apoptosis of anaerobes by 1.9-fold and release of intracellular substances by 2.9-fold, further constraining methanogenesis. The higher (1.6 vs. 1.1 copies per gyrA) metabolic activity of acetate-dependent methanogenesis implied more efficient methane production in a steady mesophilic, MT-mediated system. Metagenomic binning and network analyses indicated that ST induced dysbiosis in keystone species and greatly enhanced microbial functional redundancy, causing loss of microbial syntrophic interactions and redundant metabolic pathways. In contrast, the greater microbial interconnections (average degrees 44.9 vs. 22.1) in MT at a steady mesophilic state suggested that MT could better maintain necessary system functionality and stability through microbial syntrophy or specialized pathways. Adopting MT to transform thermophilic digesters into mesophilic digesters is feasible and could potentially enhance the further optimization and broader application of practical anaerobic engineering.
One-step acquirement of superior microbial communities from mesophilic digested sludge to upgrade anaerobic digestion

Abstract:Anaerobic digestion is a promising waste-to-energy alternative technology. However, the efficiency upgrading for conventional mesophilic digestion of organic solid waste is always indispensable. Employing hyperthermophilic or thermophilic microbial community is one of the viable upgrading alternatives. Given the unavailability of the superior microbial communities, mesophilic digested sludge was used as inoculum, and instantly controlled at 70 °C and 55 °C for acclimation of hyperthermophilic and thermophilic inocula, respectively. Waste activated sludge was continuously and synchronously fed into two digesters. After one round, thermophilic digester achieved stable biogas production rate at 0.22 L L−1 d−1, with a methane proportion over 60%, whereas fluctuation was observed in the hyperthermophilic digester, and approximately triple time was needed to reach a relatively stable biogas production rate 0.12 L L−1 d−1. Nevertheless, higher hydrolysis ratio 24.4% was observed in the hyperthermophilic digester despite the lower biogas production. Therefore, methanogenesis step limited the whole anaerobic process for the hyperthermophilic digestion, and digestion at 70 °C was appropriate as a pre-fermentation stage to enhanced hydrolysis. The genus Methanothrix proportion in the thermophilic digester gradually decreased, while another acetoclastic genus Methanosarcina ultimately was acclimated to the dominant methanogen. In addition to Methanothrix, hydrogenotrophic archaea became competitive in the hyperthermophilic digester, with Methanothermobacter dominant at 22.6%. The genus Psychrobacter, affiliated to the phylum Proteobacteria could survive better than the others at 70 °C, with a final proportion of 62.5%.

Abstract:A robust microbial community is essential for the overall stability and performance of the anaerobic digestion process. In this study, two digesters of a full-scale temperature-phased anaerobic digestion plant treating waste activated sludge were sampled for one year. The acidogenesis reactor (AR) was run at 45 +/- 2 degrees C for six months in Period I and was run at 38 +/- 2 degrees C for six months in Period II. While the methanogenesis reactor (MR) was run at 36 +/- 3 degrees C throughout the year. 16S rRNA amplicon sequencing and GeoChip 5.0 results showed that samples were clearly differentiated by reactors and periods. The elevated temperature in AR during Period I improved the effects of phase separation between the AR and MR. In AR, Fervidobacterium, assigned to Class Thermotogae, had a higher relative abundance of 8.9% in Period I. The abundance of genes involved with carbon degradation was significantly higher in Period I than Period II. In MR, the relative abundance of Methanosarcina increased from 19.8% in Period I to 30.6% in Period II. In addition, the influent characteristics, reactor performance, and operating parameters were determined as the key variables shaping the microbial community, contributing to a total of 76.3% and 69.5% of the variance of the AR and MR, respectively. A Comparative Study of Thermophilic and Mesophilic Anaerobic Co-Digestion of Food Waste and Wheat Straw: Process Stability and Microbial Community Structure Shifts

Abstract:Renewable energy recovery from organic solid waste via anaerobic digestion is a promising way to provide sustainable energy supply and eliminate environmental pollution. However, poor efficiency and operational problems hinder its wide application of anaerobic digestion. The effects of two key parameters, i.e. temperature and substrate characteristics on process stability and microbial community structure were studied using two lab-scale anaerobic reactors under thermophilic and mesophilic conditions. Both the reactors were fed with food waste (FW) and wheat straw (WS). The organic loading rates (OLRs) were maintained at a constant level of 3 kg VS/(m(3).d). Five different FW:WS substrate ratios were utilized in different operational phases. The synergetic effects of co-digestion improved the stability and performance of the reactors. When FW was mono-digested, both reactors were unstable. The mesophilic reactor eventually failed due to volatile fatty acid accumulation. The thermophilic reactor had better performance compared to mesophilic one. The biogas production rate of the thermophilic reactor was 4.9-14.8% higher than that of mesophilic reactor throughout the experiment. The shifts in microbial community structures throughout the experiment in both thermophilic and mesophilic reactors were investigated. With increasing FW proportions, bacteria belonging to the phylum Thermotogae became predominant in the thermophilic reactor, while the phylum Bacteroidetes was predominant in the mesophilic reactor. The genus Methanosarcina was the predominant methanogen in the thermophilic reactor, while the genus Methanothrix remained predominant in the mesophilic reactor. The methanogenesis pathway shifted from acetoclastic to hydrogenotrophic when the mesophilic reactor experienced perturbations. Moreover, the population of lignocellulose-degrading microorganisms in the thermophilic reactor was higher than those in mesophilic reactor, which explained the better performance of the thermophilic reactor. Two-step partial nitrification-anammox process for treating thermal-hydrolysis anaerobic digester effluent: Start-up and microbial characterisation

Abstract:Although the thermal hydrolysis has been proven to be an effective pre-treatment method for anaerobic digestion (AD), the ammonium-rich anaerobic digester effluents are unavoidably produced by thermal hydrolysis pre-treatment AD (THP-AD). In this study, a two-step partial nitrification-anammox (PN-A) process was conducted for the anaerobic digester effluents. By combining control of the solid retention time (SRT), dissolved oxygen (DO) and hydraulic retention time (HRT) (SRT 20 d, DO concentration 0.5 -0.8 mg/L, HRT 11 h), start-up of the PN reactor was achieved with a total nitrogen loading rate of 2.34 kg N/(m(3).d), generating an NH4+-N/NO2--N molar ratio of around 1.0-1.4 in the effluent. The anammox reactor was then successfully started by feeding the effluent of the PN reactor, with a total nitrogen removal rate of 1.25 kg N/(m(3).d). The system became unstable after 100 days of operation, and surface analyses of the anammox sludge particles suggested that the main reason was the adsorption of microsized suspended organic flocs in the influent onto the anammox sludge particles. Further study showed that the microsized suspended organic flocs contained large quantities of bacteria, which can compete with anammox bacteria. In addition, analyses of the microbial community reveal that Nitrosomonas and Candidatus brocadia were the main ammonium-oxidising bacteria and anammox bacteria in this study, respectively. The trends of their relative abundance during the PN-A system process confirmed the successful start-up of the PN-A reactors and the effectiveness of the start-up operations. These findings are expected to provide references for the treatment of anaerobic digester effluents from THP-AD via the biological removal of nitrogen. (C) 2019 Elsevier Ltd. All rights reserved.
Influence of thermal hydrolysis on sludge anaerobic digestion: Release of humic acid promotes electron transport of methanogen

Abstract:Thermal hydrolysis pretreatment (THP) has been widely applied to promote anaerobic digestion (AD). Humic acids (HAs) are a group of insoluble organics and often present in the conventional AD of sludge, but are reportedly substantially released during THP. Thus, the influences of the released HAs on AD shall be investigated, in particular on methanogenesis. Our results indicated that the presence of HAs extracted from thermally hydrolyzed sludge improved methanogenesis by 15.8–80.8%. However, HAs harvested at different temperatures presented distinctively different capabilities in enhancing methane production. Seemingly, the promotion efficiency of HAs to methanogenesis did not significantly respond with any individual characteristics of HAs in aspects of molecular weight, electrical activity and electroactive group compositions. However, the efficiency presented an evident correlation with the superimposed effects of the characteristics aforementioned. Therefore, a hypothesis on the mechanism involved in the phenomenon observed above was proposed that only the HAs with low molecular weight, allowing the molecules to penetrate cells could affect methanogenesis, and not all of the electroactive groups of HAs were involved in microbial electron transfer. Moreover, it was found that only the group of quinones (Q1) with side chain of electron withdrawing group played key roles in accelerating methyl acetyl-CoA degradation and promoting methanogenesis. Directional regulation of methanogenesis promotion assisted with HAs was realized by selectively removing or retaining the Q1 groups in HAs, which also evidently sustained the proposed mechanism aforementioned. The findings of this study demonstrate that high temperature was conducive to the release of organics during sludge thermal hydrolysis, whereas the temperature above 160°C led to changes of HAs structure and then subsequently undermined methane production from sludge.

Abstract:Temperature is the critical factor affecting the efficiency and cost of anaerobic digestion (AD). The current work develops a shift-temperature AD (STAD) between 35 °C and 55 °C, intending to optimise microbial community and promote substrate conversion. The experimental results showed that severe inhibition of biogas production occurred when the temperature was firstly increased stepwise from 35 °C to 50 °C, whereas no inhibition was observed at the second warming cycle. When the organic load rate was increased to 6.37 g VS/L/d, the biogas yield of the STAD reached about 400 mL/g VS, nearly double that of the constant-temperature AD (CTAD). STAD promoted the proliferation of Methanosarcina (up to 57.32 %), while severely suppressed hydrogenophilic methanogens. However, when the temperature was shifted to 35 °C, most suppressed species recovered quickly and the excess propionic acid was quickly consumed. Metagenomic analysis showed that STAD also promoted gene enrichment related to pathways metabolism, membrane functions, and methyl-based methanogenesis.
Hydrochar-Promoted Methane Production in Mesophilic and Thermophilic Anaerobic Digestion of Hydrothermal Pre-Treated Sludge

Abstract:Hydrochar produced during the hydrothermal conversion of organic solid waste could enhance the anaerobic digestion (AD) efficiency of hydrothermal pre-treated sludge. However, there was still a lack of systematic research on the effect of hydrochar on improving the methane production and microbial communities of the AD of hydrothermal pre-treated sludge under different temperature conditions. This study explored the effect of hydrochar on methane production from the mesophilic and thermophilic AD of hydrothermal pre-treated sludge and the mechanism of microbial action based on metagenomics analysis. Hydrochar could improve the methane production efficiency of mesophilic and thermophilic AD at different initial concentrations of hydrothermal pre-treated sludge. However, the effect of hydrochar in promoting AD varied under different AD temperatures. Both temperature and hydrochar were crucial factors that could influence the microbial community. Moreover, hydrochar increased the relative abundance of archaea in the AD system, resulting in an increment of 4.99% to 15.30% compared to the control group. Mesophilic reactors exhibit greater microbial diversity. Hydrochar resulted in the significant enrichment of Synergistota in the thermophilic AD system and the enrichment of Firmicutes in the mesophilic AD system, thereby promoting the hydrolysis of proteins and polysaccharides during AD. This study has practical significance for the resource treatment of excess activated sludge.
Progressive Microbial Community Networks with Incremental Organic Loading Rates Underlie Higher Anaerobic Digestion Performance.

Abstract:ABSTRACT Although biotic interactions among members of microbial communities have been conceived to be crucial for community assembly, it remains unclear how changes in environmental conditions affect microbial interaction and consequently system performance. Here, we adopted a random matrix theory-based network analysis to explore microbial interactions in triplicate anaerobic digestion (AD) systems, which is widely applied for organic pollutant treatments. The digesters were operated with incremental organic loading rates (OLRs) from 1.0 g volatile solids (VS)/liter/day to 1.3 g VS/liter/day and then to 1.5 g VS/liter/day, which increased VS removal and methane production proportionally. Higher resource availability led to networks with higher connectivity and shorter harmonic geodesic distance, suggestive of more intense microbial interactions and quicker responses to environmental changes. Strikingly, a number of topological properties of microbial network showed significant (P < 0.05) correlation with AD performance (i.e., methane production, biogas production, and VS removal). When controlling for environmental parameters (e.g., total ammonia, pH, and the VS load), node connectivity, especially that of the methanogenic archaeal network, still correlated with AD performance. Last, we identified the Methanothermus, Methanobacterium, Chlorobium, and Haloarcula taxa and an unclassified Thaumarchaeota taxon as keystone nodes of the network. IMPORTANCE AD is a biological process widely used for effective waste treatment throughout the world. Biotic interactions among microbes are critical to the assembly and functioning of the microbial community, but the response of microbial interactions to environmental changes and their influence on AD performance are still poorly understood. Using well-replicated time series data of 16S rRNA gene amplicons and functional gene arrays, we constructed random matrix theory-based association networks to characterize potential microbial interactions with incremental OLRs. We demonstrated striking linkage between network topological features of methanogenic archaea and AD functioning independent of environmental parameters. As the intricate balance of multiple microbial functional groups is responsible for methane production, our results suggest that microbial interaction may be an important, previously unrecognized mechanism in determining AD performance.
Responses of anaerobic digestion of food waste to coupling effects of inoculum origins, organic loads and pH control under high load: Process performance and microbial characteristics

Abstract:This study investigated responses of anaerobic digestion (AD) of food waste (FW) with different inocula to varying organic loads and to pH control under high load in terms of process performance and microbial characteristics. Without pH control, digester inoculated by thickened sludge obtained high methane yield of 547.8 +/- 27.8 mL/g VS under organic load of 7.5 g VS/L but was inhibited by volatile fatty acids (VFAs) under higher loads (15 and 30 g VS/L). However, digesters inoculated by anaerobic sludge obtained high methane yields of 575.9 +/- 34.2, 569.3 +/- 24.8 and 531.9 +/- 26.2 mL/g VS under organic loads of 7.5, 15 and 30 g VS/L and VFAs inhibition only appeared under extremely high load of 45 g VS/L. Digesters under VFA inhibition with high load were significantly enhanced by controlling single ecological factor pH at 6.5, 7.0 and 7.5, as indicated by shorter lag phases, higher peak values of methane production rate, greater methane yields and fast VFAs degradation. Maximum methane recovery was obtained with pH control at 7.5 under high load. VFA inhibition was accompanied by the degeneration of ecological functions of Syntrophomonadaceae and unidentified Bacteroidales and the dominant growth of unidentified Clostridiales. Under high load and pH control, high stability was strongly associated with obvious growth of Methanosarcina, which enriched methanogenic pathways thus improved system robustness and tolerance to VFAs. Moreover, pH control stimulated the growth of syntrophic Bacteria Syntrophomonadaceae while maintaining the high activity of hydrogenotrophic methanogens therefore sustained efficient syntrophic communities of Bacteria and methanogens and avoided over accumulation of VFAs. pH control promoted adaptive selection of methanogens, leading to obvious decline of archaeal community diversity. This study provided practical guidance on digester configurations of high-load AD of FW and expanded the understanding of responses to coupling effects of inoculum origins, organic loads and pH control under high load concerning process performance and microbial community dynamics.
