Abstract:Genetic selection has achieved little progress in reducing mastitis incidence. Mastitis traits are problematic due to the lack of sensitivity of the data and reliance on clinical diagnosis, often missing subclinical cases, and/or on monthly somatic cell count (SCC) measurements. The current measure for mastitis is the lactation average of the somatic cells score (LSCS). We studied two datasets: (1) 148 heifers divided into non-intramammary infected, sub-clinically infected and clinical mastitis groups; (2) data from 89,601 heifers from Israeli Holsteins through the same period divided into "udder healthy" (UH) and "non-healthy" (UNH) by a threshold of SCC 120,000 cells/mL in all nine monthly milk recordings. In study 1, non-infected heifers had significantly (p < 0.05) more partum, production days and overall lifetime milk production compared to clinical and sub-clinically infected. In study 2, UH heifers (20.3%) had significantly higher (p < 0.01) lifetime milk, production days, and lactations. Subdividing datasets by sires, the same analyses detected differences in percentages of UH daughters between the sire groups. Lifetime milk production correlated (r = +0.83, p < 0.001) with udder health status. SCC threshold of less than 120,000 cells/mL during all first lactation measurements indicated healthy udder, providing a valuable insight that this dichotomous trait is advantageous for calculating lifetime net-merit index (NM$) over LSCS.

A Novel Approach to the Management of a Progressive Descemet Membrane Tear in a Patient With Keratoglobus and Acute Hydrops

Abstract:Purpose: To report a case of corneal hydrops in a patient with keratoglobus that was managed with endothelial keratoplasty to achieve corneal stability and prevent a limbus-to-limbus tear in Descemet membrane. Methods: A 30-year-old man with keratoglobus presented with corneal hydrops in his left eye resulting from a central vertical tear in Descemet membrane. His other eye had been previously treated with penetrating keratoplasty using a large graft (an 11-mm donor graft to a 10-mm recipient bed) because of a limbus-to-limbus tear in Descemet membrane without resolution of his edema. An attempt to approximate the edges of the Descemet tear in the left eye by an intracameral air injection failed, and the tear continued to progress peripherally. An endothelial keratoplasty button with anchoring sutures was placed over the Descemet tear because of excessive localized edema. Results: One month after insertion of the sutured endothelial keratoplasty button, the edema had resolved, and 1 year later, the tear remains sealed. The patient's visual acuity improved from counting fingers at 1 foot to 20/100. Conclusions: Reconstitution of the posterior corneal surface in keratoglobus-induced hydrops can be achieved with endothelial keratoplasty over the Descemet tear. Preventing progression of a central Descemet tear is essential to bypass the need for a large-diameter penetrating keratoplasty graft and its complications in a young patient with a history of bilateral corneal hydrops.
Genetic evaluation of udder health traits in Spanish Holstein cows

Abstract:The aims of this study were to estimate the genetic parameters of clinical mastitis (CM) and SCS traits, and to compare the performance of genetic evaluations of CM traits using univariate and bivariate analyses (CM-SCS). Data were edited according to the Udder Health Golden Standard harmonization, and then 6 CM traits and 6 SCS traits were considered, as the result of combining 3 lactation classifications (1, 2, ≥3) and 2 milking periods (early, late). The linear mixed animal models included the ratio of period at risk as a covariate, herd-year of calving, month of calving, and lactation-age as fixed effects, and the permanent environmental effect for traits of ≥3 lactations. Prevalence of CM in early lactation was similar regardless the lactation number (5%-6%), and the estimated heritabilities were 0.01. Prevalences in late lactation ranged from 10% to 24% and heritabilities ranged from 0.03 to 0.05. Estimated heritabilities of SCS ranged from 0.06 to 0.16 with univariate analyses. Somatic cell count (therefore its log-transformation SCS) showed a higher probability of correctly identifying healthy cows than infected cows but there was still up to 36% of healthy cows for which CM was not detected by SCS. Genetic correlations between CM-SCS traits ranged from 0.36 to 0.95, and SCS in lactation 3 and later did not add extra information to SCS in the second lactation for predicting CM. Regarding reliabilities of EBVs for CM traits, bivariate CM-SCS analyses led to substantial increases with respect to the single-trait model for sires (7%-12% more in first lactation and 16%-28% more for second lactation). Sire's rank correlations for CM between univariate and bivariate analyses (0.47-0.92) suggest that discarding sires could be more accurate than selecting candidates for sires of dams. We can conclude that SCS in first lactation could be useful to supplement CM data in first and second lactations to improve udder health genetic evaluation.
Genetic aspects of immunoglobulins and cyclophilin A in milk as potential indicators of mastitis resistance in Holstein cows

Abstract:Mastitis is one of the most frequent and costly diseases affecting dairy cattle. Natural antibodies (immunoglobulins) and cyclophilin A (CyPA), the most abundant member of the family of peptidyl prolyl cis/trans isomerases, in milk may serve as indicators of mastitis resistance in dairy cattle. However, genetic information for CyPA is not available, and knowledge on the genetic and nongenetic relationships between these immune-related traits and somatic cell score (SCS) and milk yield in dairy cattle is sparse. Therefore, we aimed to comprehensively evaluate whether immune-related traits consisting of 5 Ig classes (IgG, IgG1, IgG2, IgA, and IgM) and CyPA in the test-day milk of Holstein cows can be used as genetic indicators of mastitis resistance by evaluating the genetic and nongenetic relationships with SCS in milk. The nongenetic factors affecting immune-related traits and the effects of these traits on SCS were evaluated. Furthermore, the genetic parameters of immune-related traits according to health status and genetic relationships under different SCS environments were estimated. All immune-related traits were significantly associated with SCS and directly proportional. Additionally, evaluation using a classification tree revealed that IgA, IgG2, and IgG were associated with SCS levels. Genetic factor analyses indicated that heritability estimates were low for CyPA (0.08) but moderate for IgG (0.37), IgA (0.44), and IgM (0.44), with positive genetic correlations among Ig (0.25-0.96). We also evaluated the differences in milk yield and SCS of cows between the low and high groups according to their sires' estimated breeding value for immune-related traits. In the high group, IgA had a significantly lower SCS in milk at 7 to 30 d compared with that in the low group. Furthermore, the Ig in milk had high positive genetic correlations between healthy and infected conditions (0.82-0.99), suggesting that Ig in milk under healthy conditions could interact with those under infected conditions, owing to the genetic ability based on the level of Ig in milk. Thus, Ig in milk are potential indicators for the genetic selection of mastitis resistance. However, because only the relationship between immune-related traits and SCS was investigated in this study, further study on the relationship between clinical mastitis and Ig in milk is needed before Ig can be used as an indicator of mastitis resistance.
The genetic basis for the selection of dairy goats with enhanced resistance to gastrointestinal nematodes

Abstract:Gastrointestinal nematodes (GIN) severely affect small ruminant production worldwide. Increasing problems of anthelmintic resistance have given strong impetus to the search for alternative strategies to control GIN. Selection of animals with an enhanced resistance to GIN has been shown to be successful in sheep. In goats, the corresponding information is comparatively poor. Therefore, the present study was designed to provide reliable data on heritabilities of and genetic correlations between phenotypic traits linked to GIN and milk yield in two major dairy goat breeds (Alpine and Saanen). In all, 20 herds totalling 1303 goats were enrolled in the study. All herds had (i) a history of gastrointestinal nematode infection, (ii) uniform GIN exposure on pasture and (iii) regular milk recordings. For all goats, individual recordings of faecal egg counts (FEC), FAMACHA© eye score, packed cell volume (PCV) and milk yield were performed twice a year with an anthelmintic treatment in between. The collected phenotypic data were multivariately modelled using animal as a random effect with its covariance structure inferred from the pedigree, enabling estimation of the heritabilities of the respective traits and the genetic correlation between them. The heritabilities of FEC, FAMACHA© and PCV were 0.07, 0.22 and 0.22, respectively. The genetic correlation between FEC and FAMACHA© was close to zero and -0.41 between FEC and PCV. The phenotypic correlation between FEC and milk yield was close to zero, whereas the genetic correlation was 0.49. Our data suggest low heritability of FEC in Saanen and Alpine goats and an unfavourable genetic correlation of FEC with milk yield.

Distribution of Bovine Mastitis Pathogens in Quarter Milk Samples from Bavaria, Southern Germany, between 2014 and 2023-A Retrospective Study

Abstract:The objective of this study was to investigate the distribution of mastitis pathogens in quarter milk samples (QMSs) submitted to the laboratory of the Bavarian Animal Health Service (TGD) between 2014 and 2023 in general, in relation to the clinical status of the quarters, and to analyze seasonal differences in the detection risk. Each QMS sent to the TGD during this period was analyzed and tested using the California Mastitis Test (CMT). Depending on the result, QMSs were classified as CMT-negative, subclinical, or clinical if the milk character showed abnormalities. Mastitis pathogens were detected in 19% of the QMSs. Non-aureus staphylococci (NAS) were the most common species isolated from the culture positive samples (30%), followed by Staphylococcus (S.) aureus (19%), Streptococcus (Sc.) uberis (19%), and Sc. dysgalactiae (9%). In culture-positive QMSs from CMT-negative and subclinically affected quarters, the most frequently isolated pathogens were NAS (44% and 27%, respectively), followed by S. aureus (25% and 17%, respectively) and Sc. uberis (8% and 22%, respectively). In QMSs from clinically affected quarters, the most frequently isolated pathogens were Sc. uberis (32%), S. aureus (13%), Sc. dysgalactiae (11%), and Escherichia (E.) coli (11%). The distribution of NAS and Sc. uberis increased throughout the study period, while that of S. aureus decreased. From June to October, QMSs from subclinically affected quarters increased and environmental pathogens, such as Sc. uberis, were detected more frequently. In conclusion, this study highlights the dynamic nature of the distribution of mastitis pathogens, influenced by mastitis status and seasonal factors. Environmental pathogens still play an important role, especially in clinical mastitis and seasonal dependency, with the number of positive samples continuing to increase. It is therefore essential to continue mastitis control measures and to regularly monitor the spread of mastitis pathogens in order to track trends and adapt targeted prevention measures.
Genome-Wide Association Study for Milk Somatic Cell Score in Holstein Friesian Cows in Slovenia

Abstract:Mastitis is a serious challenge for the dairy industry, leading to economic losses and affecting milk quality. The aim of this study is to identify genetic factors associated with mastitis resistance by conducting a genome-wide association study (GWAS) for the somatic cell score (SCS). Phenotypic records of 350 Holstein Friesian cows were obtained from the Slovenian Cattle Recording Scheme Database and consisted of around 1500 lactation data from 2012 to 2023 collected on a single farm in Slovenia. Corresponding genotypic data were also retrieved from the same database and genotyped using the Illumina BovineSNP50 BeadChip (Illumina, Inc., San Diego, CA, USA). For the association study, three SCS parameters were considered, including lactation mean somatic cell score (LM_SCS), maximum SCS value (SCSMAX), and top three mean value of SCS (TOP3). After performing a GWAS using FarmCPU and BLINK models, five significant SNPs associated with the TOP3 trait were found on BTA 14, 15, 22, and 29. The identified SNP markers were closely linked to six known candidate genes (DNASE1L3, SLC36A4, ARMC1, PDE7A, MMP13, CD44). These results indicate potential genetic markers associated with SCS in the Slovenian Holstein Friesian population.

Abstract:A total of 31,396 females born from 2010 to 2013 in 43 large-scale Holstein-Friesian herds were phenotyped for calf and cow disease traits using a veterinarian diagnosis key. Calf diseases were general disease status (cGDS), calf diarrhea (cDIA), and calf respiratory disease (cRD) recorded from birth to 2 mo of age. Incidences were 0.48 for cGDS, 0.28 for cRD, and 0.21 for cDIA. Cow disease trait recording focused on the early period directly after calving in first parity, including the interval from 10 d before calving to 200 d in lactation. For cows, at least one entry for the respective disease implied a score = 1 (sick); otherwise, score = 0 (healthy). Corresponding cow diseases were first-lactation general disease status (flGDS), first-lactation diarrhea (flDIA), and first-lactation respiratory disease (flRD). Additional cow disease categories included mastitis (flMAST), claw disorders (flCLAW), female fertility disorders (flFF), and metabolic disorders (flMET). A further cow trait category considered first-lactation test-day production traits from official test-days 1 and 2 after calving. The genotype data set included 41,256 single nucleotide polymorphisms (SNP) from 9,388 females with phenotypes. Linear and generalized linear mixed models with a logit link-function were applied to Gaussian and categorical cow traits, respectively, considering the calf disease as a fixed effect. Most of the calf diseases were not significantly associated with the occurrence of any cow disease. By trend, increasing risks for the occurrence of cow diseases were observed for healthy calves, indicating mechanisms of disease resistance with aging. Also by trend, occurrence of calf diseases was associated with decreasing milk, protein, and fat yields. Univariate linear and threshold animal models were used to estimate heritabilities and breeding values (EBV) for all calf and cow traits. Heritabilities for cGDS and cRD were 0.06 and 0.07 for cDIA. Genetic correlations among all traits were estimated using linear-linear animal models in a series of bivariate runs. The genetic correlation between cDIA and cRD was 0.29. Apart from the genetic correlation between flRD with cGDS (-0.38), EBV correlations and genetic correlations between calf diseases with all cow traits were close to zero. Genome-wide association studies were applied to estimate SNP effects for cRD and cDIA, and for the corresponding traits observed in cows (flRD and flDIA). Different significant SNP markers contributed to cDIA and flDIA, or to cRD and flRD. The average correlation coefficient between cRD and flRD considering SNP effects from all chromosomes was 0.01, and between cDIA and flDIA was -0.04. In conclusion, calf diseases are not appropriate early predictors for cow traits during the early lactation stage in parity 1.

Heritability estimation of dairy cows using optimal spline curves in southern area of China

Abstract:Somatic cell count (SCC) nowadays has been one of the most important indicators for mastitis in dairy cows.In this study,we selected 290 thousand DHI records from 34 150 first-pregnancy dairy cows in 62 herds,which all existed in the south area of China,including Shanghai,Zhejiang and Jiangsu provinces.The random regression model was used in the analyses of genetic parameters,and different orders of linear,quadratic and cubic spline curves and knots from 4 to 6 were fitted and compared.The most ideal spline with the second order and 4 knots was selected based on AIC criteria.The estimated heritability of SCC was 0.13 to 0.51,much higher than the heritability of mastitis itself.Therefore,SCC is a good indicator for selection of dairy cows promoting mastitis resistance breeding process.
Relationships among somatic cell count, California mastitis test, impedance and bacteriological status of milk in goats and sheep in early lactation

Abstract:The objectives of this trial were to evaluate and compare the test characteristics of a number of indirect tests of bacteriological status of the milk from goats and sheep and to assess the affect of varying levels of prevalence of infection on the performance of those tests.The somatic cell count, California mastitis test (CMT) score, electrical impedance and the bacteriological status of 220 and 262 milk samples from the glands of lactating dairy goats and sheep, respectively, were determined. The sensitivity and specificity of indirect tests in predicting the bacteriological status were compared by analyzing the areas under the receiver operating characteristic curve and calculating the likelihood ratio at various cut-off values.Bacteria were isolated from 17.7 and 4.6% of glands from goats and sheep, respectively. Somatic cell count was a better predictor of bacteriological status than either the CMT score or impedance in both goats and sheep. However, knowledge of the CMT score in both sheep and goats and the impedance in goats increases the likelihood of predicting the presence of a bacterial pathogen compared to no testing at all (P<0.05). Knowledge of the age and days postpartum of the animals at the time of testing did not increase the probability that infection status could be predicted. Variation in cut-off levels to predict infection status reported in the literature may be due to variation in the prevalence of infection in the populations studied. Use of likelihood ratio allows comparison of tests over a range of prevalence's as likelihood ratios are independent of prevalence. The prevalence of infection within a herd should be considered when selecting a cut-off value for indirect testing for the presence of bacteria in milk of small ruminants.

Abstract:Abstract Background During the past ten years many quantitative trait loci (QTL) affecting mastitis incidence and mastitis related traits like somatic cell score (SCS) were identified in cattle. However, little is known about the molecular architecture of QTL affecting mastitis susceptibility and the underlying physiological mechanisms and genes causing mastitis susceptibility. Here, a genome-wide expression analysis was conducted to analyze molecular mechanisms of mastitis susceptibility that are affected by a specific QTL for SCS on Bos taurus autosome 18 (BTA18). Thereby, some first insights were sought into the genetically determined mechanisms of mammary gland epithelial cells influencing the course of infection. Methods Primary bovine mammary gland epithelial cells (pbMEC) were sampled from the udder parenchyma of cows selected for high and low mastitis susceptibility by applying a marker-assisted selection strategy considering QTL and molecular marker information of a confirmed QTL for SCS in the telomeric region of BTA18. The cells were cultured and subsequently inoculated with heat-inactivated mastitis pathogens Escherichia coli and Staphylococcus aureus , respectively. After 1, 6 and 24 h, the cells were harvested and analyzed using the microarray expression chip technology to identify differences in mRNA expression profiles attributed to genetic predisposition, inoculation and cell culture. Results Comparative analysis of co-expression profiles clearly showed a faster and stronger response after pathogen challenge in pbMEC from less susceptible animals that inherited the favorable QTL allele 'Q' than in pbMEC from more susceptible animals that inherited the unfavorable QTL allele 'q'. Furthermore, the results highlighted RELB as a functional and positional candidate gene and related non-canonical Nf-kappaB signaling as a functional mechanism affected by the QTL. However, in both groups, inoculation resulted in up-regulation of genes associated with the Ingenuity pathways 'dendritic cell maturation' and 'acute phase response signaling', whereas cell culture affected biological processes involved in 'cellular development'. Conclusions The results indicate that the complex expression profiling of pathogen challenged pbMEC sampled from cows inheriting alternative QTL alleles is suitable to study genetically determined molecular mechanisms of mastitis susceptibility in mammary epithelial cells in vitro and to highlight the most likely functional pathways and candidate genes underlying the QTL effect.

Effects of resistance exercise and creatine supplementation on myasthenia gravis: a case study.

Abstract:PURPOSE The purpose of this case study was to determine the effects of 15 wk of resistance exercise and creatine (Cr) supplementation on body composition, training volume, peak strength, and complete blood chemistry in a patient with myasthenia gravis (MG). METHODS The patient was a 26-yr-old man who was taking prednisone and azathioprine for his condition. The patient self-administered 5 g of Cr per day in addition to resistance exercise 3 times per week. Fasting blood samples were obtained and body weight (BW) and fat free mass (FFM; via hydrostatic weighing) were measured before and after training and Cr supplementation. In addition, isokinetic (Cybex II) peak strength for leg extension (LE), leg flexion (LF), and volume load (repetition x mass lifted) for the first and last resistance training session were determined. RESULTS After Cr supplementation and training, the results demonstrated increases in BW (6.8%), FFM (4.3%), upper body volume load (37.0%), lower body volume load (15.0%), and peak strength for LE (37.0%) and LF (12.5%). Moreover, blood chemistry values remained within normal limits for the duration of the 15-wk study. CONCLUSION These data suggest that resistance exercise plus Cr supplementation may promote gains in strength and FFM in patients with MG.

[Relationship between morphological characteristics of the teat duct and prevalence of intramammary infections with Streptococcus agalactiae in dairy cows]

Abstract:The objective of the study was to determine the relationship between morphological findings of the surface of teat duct particularly the level of ceratosis and the prevalence of intramammary infections (IMI). The study was conducted on a commercial dairy herd housing about 3000 lactating dairy cows. We examined 891 quarters in the middle of lactation. Duplicate samples of quarter foremilk were collected monthly. The bacteriological status of quarters was determined according to the recommendations of IDF. At the same time teats were evaluated by clinical examinations. The appearance of teat skin lesions and the status of the teat duct especially the existence of hyperceratosis (HC) was documented. Four classes of teat duct hyperceratosis were defined: without, slight, medium and severe HC. The rate of IMI in different classes of hyperceratosis of teat duct was compared by Chi-square analysis. Prevalences of intramammary infections were determined three times (P1, P2 and P3) during the study period. Prevalence of infection was high for S. aureus (P1: 5.6% vs. P2: 4.5% vs. P3: 4.3%), Sc. agalactiae (P1: 2.7% vs. P2: 2.6% vs. P3: 2.8%) and CNS (P1: 10.7% vs. P2: 8.8% vs. P3: 9.6%). Furthermore we detected IMI caused by other streptococci, yeast, E. coli and mixed infections. A positive correlation between status of HC and prevalence of IMI for Sc. agalactiae was found. At the second and third sampling time the rate of intramammary infection with Sc. agalactiae in quarters with medium HC (P2: 9.21% and P3: 13.73%) differed significantly (p < 0.05) compared to groups without (P2: 1.56% and P3: 1.91%) and slight hyperceratosis (P2: 2.33% and P3: 2.56%). The results of our study indicate a correlation between morphology of teat duct surface, especially regarding to Sc. agalactiae. On one hand HC can cause high intramammary infection rate with Sc. agalactiae. On the other hand it is possible that HC is the consequence of a quarter infection with Sc. agalactiae. Further research is required.
Liability to diseases and their relation to dry matter intake and energy balance in German Holstein and Fleckvieh dairy cows

Abstract:Dairy cow efficiency is increasingly important for future breeding decisions. The efficiency is determined mostly by dry matter intake (DMI). Reducing DMI seems to increase efficiency if milk yield remains the same, but resulting negative energy balance (EB) may cause health problems, especially in early lactation. Objectives of this study were to examine relationships between DMI and liability to diseases. Therefore, cow effects for DMI and EB were correlated with cow effects for 4 disease categories throughout lactation. Disease categories were mastitis, claw and leg diseases, metabolic diseases, and all diseases. In addition, this study presents relative percentages of diseased cows per days in milk (DIM), repeatability, and cow effect correlations for disease categories across DIM. A total of 1,370 German Holstein (GH) and 287 Fleckvieh (FV) primiparous and multiparous dairy cows from 12 dairy research farms in Germany were observed over a period of 2 yr. Farm staff and veterinarians recorded health data. We modeled health and production data with threshold random regression models and linear random regression models. From DIM 2 to 305 average daily DMI was 22.1 kg/d in GH and 20.2 kg/d in FV. Average weekly EB was 2.8 MJ of NEL/d in GH and 0.6 MJ of NEL/d in FV. Most diseases occurred in the first 20 DIM. Multiparous cows were more susceptible to diseases than primiparous cows. Relative percentages of diseased cows were highest for claw and leg diseases, followed by metabolic diseases and mastitis. Repeatability of disease categories and production traits was moderate to high. Cow effect correlations for disease categories were higher for adjacent lactation stages than for more distant lactation stages. Pearson correlation coefficients between cow effects for DMI, as well as EB, and disease categories were estimated from DIM 2 to 305. Almost all correlations were negative in GH, especially in early lactation. In FV, the course of correlations was similar to GH, but correlations were mostly more negative in early lactation. For the first 20 DIM, correlations ranged from -0.31 to 0.00 in GH and from -0.42 to -0.01 in FV. The results illustrate that future breeding for dairy cow efficiency should focus on DMI and EB in early lactation to avoid health problems.

Abstract:Abstract The objective was to evaluate the potential for the use of a thermal imaging system for the identification of subclinical mastitis in dairy cows. Multiparous Holstein dairy cows (n = 131) within 150 days in milk (DIM; range 65 to 155) were enrolled in the study at a commercial dairy farm in central Texas. Milk samples (10 mL) were collected from each quarter separately and evaluated for somatic cell counts (SCC; n*1000), fat (%), protein (%), lactose (%), SNF (%), BHBA (mM/L), acetone (mM/L), and milk urea nitrogen (MUN; mg/dL). A minimum of four thermal images per cow representing all four quarters were captured using a handheld infrared camera (Fluke TiS 75+) from approximately one-meter distance from the cow. All images were taken inside the milking parlor and after the morning milking during February 2023 to minimize the impact of daily ambient temperatures. Skin temperatures from each quarter (maximum, minimum, average) were extracted using Fluke Connect software. A total of 524 quarter-level milk samples and corresponding udder images were obtained, and 10 images were discarded from analyses due to the lack of appropriate resolution and focus. To prevent collinearity, a correlation matrix was developed using PROC CORR in SAS and variables with significant correlations were removed from the model. Subclinical mastitis was defined at the quarter level when SCC > 250,000. Statistical analyses were performed using mixed linear regression models (PROC MIXED) with cow and DIM as random effects in the model, and logistic regression model (PROC LOGISTIC) was fitted to evaluate the probability of the disease based on temperature and milk variables. Maximum and average udder temperatures did not demonstrate a significant relationship with subclinical mastitis status whereas minimum udder temperature was greater in sick quarters (29.6 ± 0.52 vs 27.5 ± 0.45; P = 0.003). Increased minimum udder temperature was also associated with BHBA (P < 0.0001), acetone (P < 0.0004), and MUN (P < 0.0007) concentrations in milk. Cows with subclinical mastitis had decreased BHBA (0.14 ± 0.037 vs 0.28 ± 0.031; P = 0.0036) and acetone (0.017 ± 0.003 vs 0.044 ± 0.003; P < 0.001). Odds of subclinical mastitis were associated with MUN values in milk (OR 0.94; CI 0.89 - 0.98) but not with any udder surface temperature variables. We conclude that an increase in the minimum temperature of the udder could be used as an indicator of subclinical mastitis but studies with larger datasets and using advanced image analysis techniques are warranted to validate these results.

Abstract:Udder health was studied in 300 dairy herds grouped in three categories according to the bulk milk somatic cell count. In all herds, lactating cows were housed in a free-stall barn during the winter. All herds participated in a three or four-weekly milk recording system, had annual production quota between 300,000 and 900,000 kg, and were stocked with cows of the Holstein-Friesian or Dutch Friesian breeds. The incidence of clinical mastitis was not different among herds with a low (< or = 150,000), middle (151,000 to 250,000), or high (251,000 to 400,000 cells/ml) bulk milk somatic cell count. Clinical mastitis caused by Gram-negative pathogens occurred more often in herds with a low bulk milk somatic cell count. Clinical mastitis caused by Staphylococcus aureus, Streptococcus dysgalactiae, or Streptococcus agalactiae occurred more often in herds with a high bulk milk somatic cell count; however, the incidence of clinical Streptococcus uberis mastitis was not different among the three herd categories. The differences in bulk milk somatic cell count between the categories could be explained by the management practices studied. The incidence of clinical E. coli mastitis was mostly related to housing, hygiene, and milking machine. The incidence of clinical Staphylococcus aureus mastitis was associated with factors that were related to the bulk milk somatic cell count and to factors of which it was not clear whether they were a cause or effect of the high incidence of Staphylococcus aureus mastitis. The incidence of clinical Streptococcus dysgalactiae mastitis was related to nutrition, milking technique, and milking machine. The incidence of clinical Streptococcus uberis mastitis was associated with housing, nutrition, and milking machine. Two groups of farmers and herds could be differentiated. The first group was identified as 'Clean and Accurate', and the second group as 'Quick and Dirty'. The relationship between these two groups and bulk milk somatic cell count category was high. However, the relationship between the two groups and the incidence of clinical mastitis was weak.

Abstract:Dairy farmers using automatic milking are able to manage mastitis successfully with the help of mastitis attention lists. These attention lists are generated with mastitis detection models that make use of sensor data obtained throughout each quarter milking. The models tend to be limited to using the maximum or average value of the sensor data pattern, potentially excluding other valuable information. They often put cows on the lists unnecessarily, and their sensitivity for abnormal milk classification is too low for automated separation. Therefore, we analyzed sensor data patterns within quarter milkings in order to identify potentially predictive variables for abnormal milk and clinical mastitis classification. The data used in this study was obtained at a commercial dairy farm in Germany in September 2002, where a German Simmental herd was milked by a Lely Astronaut system. In total, 3232 quarter milkings from 63 cows were analysed; 94 quarter milkings were defined as milk with abnormal homogeneity and 270 as clinical mastitis. A data flow diagram was developed to systematically describe the steps involved in the transformation of within quarter milking measurements into variables that potentially predict abnormal milk and clinical mastitis. Three types of pattern descriptors were used: level, variability, and shape. In addition to using the absolute value of the pattern descriptor, the descriptors were considered relative to their expected value based on pattern descriptor values from previous milkings and from other quarters within the same cow milking. Using this method, potentially predictive variables were computed for electrical conductivity, the colours red, green and blue, a combination of colour sensors, and milk production. The importance of a variable in predicting abnormal milk and clinical mastitis was evaluated by computing correlation coefficients as well as information gain ratios. The most important variables came from the sensors for electrical conductivity, blue and green. Variables describing the variability and shape of the measurement patterns were as important as mean and maximum values, and should be included in future modelling. Also variables that are based on absolute values should be considered for future modelling. Results suggest that clinical mastitis and abnormal milk classification models may include similar predictive variables, but requirements for these models differ resulting in the need for different models. The schematic approach to developing potentially predictive variables will be helpful when exploring the usefulness of new sensors, researching other approaches to estimate expected values, and studying sensor data patterns in general.

Abstract:Mastitis is one of the most important infectious diseases and one of the diseases that causes the greatest use of antibiotics in dairy cows. Therefore, updated information on the bacteria that cause mastitis and their antibiotic susceptibility properties is important. Here, for the first time in over 10 years, we updated the bacterial findings in clinical mastitis in Swedish dairy cows together with their antibiotic resistance patterns and risk factors for each bacterial species. During the period 2013–2018, samples from clinical mastitis were collected, together with information on the cows and herds of origin. The samples were cultured, and a total of 664 recovered bacterial isolates were subjected to susceptibility testing. Staphylococcus aureus (S. aureus) was the most common pathogen and accounted for 27.8% of diagnoses, followed by Streptococcus dysgalactiae (S. dysgalactiae) (15.8%), Escherichia coli (E. coli) (15.1%), Streptococcus uberis (S. uberis) (11.4%), Trueperella pyogenes (T. pyogenes) (7.7%), non-aureus staphylococci (NAS) (2.8%), Klebsiella spp. (2.7%), Enterococcus spp. (1.3%), and Streptococcus agalactiae (S. agalactiae) (1.2%). Various other bacteria accounted for 2.6%. Staphylococci were, in general, susceptible to most antibiotics, but 2.6% of S. aureus and 30.4% of NAS were resistant to penicillin. No methicillin-resistant staphylococci were found. All S. agalactiae were susceptible to penicillin. Bimodal and trimodal MIC distributions for penicillin in S. dysgalactiae and S. uberis, respectively, indicate acquired reduced susceptibility in some isolates. The mostly unimodal MIC distributions of T. pyogenes indicate that acquired resistance does usually not occur in this species. Among E. coli, 14.7% were resistant to at least one antibiotic, most often ampicillin (8.7%), streptomycin (7.8%), or sulphamethoxazole (6.9%). Klebsiella spp. had low resistance to tetracycline (9.1%) but is considered intrinsically resistant to ampicillin. Pathogen-specific risk factors were investigated using multivariable models. Staphylococcus aureus, S. dysgalactiae, and T. pyogenes were more common, while E. coli was less common in quarters with more than one pathogen. S. aureus and T. pyogenes were mostly seen in early lactation, while E. coli was more common in peak to mid lactation and S. dysgalactiae in early to peak lactation. Trueperella pyogenes and Klebsiella spp. were associated with a previous case of clinical mastitis in the current lactation. Staphylococcus aureus was associated with tie stalls and T. pyogenes with loose housing. All pathogens except E. coli and S. dysgalactiae had a seasonal distribution. In conclusion, the aetiological agents for clinical bovine mastitis have remained relatively stable over the last 10–15 years, S. aureus, S. dysgalactiae, E. coli and S. uberis being the most important. Resistance to penicillin among Gram-positive agents was low, and in general, antibiotic resistance to other compounds was low among both Gram-positive and Gram-negative agents.
