Abstract:The title compound, (1R,2R,3R,5S)-3-(3-chlorophenyl)-1-hydroxy-2-methyl-6-phenyl-8-oxa-6-azabicyclo[3.2.1]octan-7-one, was synthesized and characterized by H-1 and C-13 NMR and HRMS spectroscopy. Its absolute molecular configuration was investigated by X-ray crystallography. The crystal structure analysis revealed that the structure exhibits intermolecular hydrogen bonds of the type O-H horizontal ellipsis O. The methyl group and chlorophenyl group are located in the cis configuration. The conformation is stabilized by weak intermolecular C-H... O interactions.

Abstract:(E)-2-(3-Hydroxy-4-methoxybenzylidene)hydrazinecarbothioamide 3 was synthesized by reacting thiosemicarbazide with 2-hydorxy-3-methoxybenzaldehyde in dry ethanol. The structure was elucidated by spectroscopic (FT-IR, 1H NMR, and 13C NMR) and single crystal X-ray diffraction techniques. A detailed analysis of the intermolecular interactions has been performed based on the Hirshfeld surfaces and their associated two-dimensional fingerprint plots. DFT, spectroscopic, and electrochemical DNA-binding analysis confirmed that the compound is reactive to bind with DNA. Viscometric studies suggested that compound 3 has a mixed mode of interaction and intercalated into the DNA base pairs predominantly along with the possibility of electrostatic interactions.Graphical Abstract

Abstract:A novel triazole derivative was synthesized and characterized by various spectroscopic techniques such as FTIR, NMR and single crystal X-ray diffraction study. The title compound crystallizes in the triclinic space group P1 ̄ and exhibits inter-molecular hydrogen bonds of the type C–H...O and C–H...N. Hirshfeld surface analysis shows that the major intermolecular interaction is mainly due to H...H contacts. DFT calculations were performed to study their electronic properties. QTAIM and NCI studies were used to investigate the weak interactions in five triazole derivatives including the title compound.

Abstract:The three component reaction between 5‑chloro-3-methyl-1-phenyl-1H-pyrazole-4- carbaldehyde, thiosemicarbazide and 4-nitrophenacyl bromide furnished (E)-2-(2-((5‑chloro-3-methyl-1-phenyl-1H-pyrazol-4-yl)methylene)hydrazineyl)-4-(4 nitrophenyl)thiazole which was characterized by FT-IR and NMR spectral methods. These spectroscopic methods confirmed the structural framework of the titled compound. The density functional theory (DFT) approach with a 6–311++G(d,p) method was used and electronic and spectroscopic parameters were analyzed. Frontier molecular orbital analysis of the titled compound found a 3.13 eV energy gap with HOMO primarily populated on pyrazole and thiazole rings whereas LUMO populated on benzene carrying nitro group. According to the Non-Linear Optical (NLO) study, the titled compound might be significant in the fabrication of nonlinear optical materials. The results of the in-silico molecular docking study against cytochrome P450 14-sterol demethylase (CYP51) showed that hydrogen bonds, aromatic and hydrophobic interactions, and van Der Waals forces all contributed to strong binding affinity with the ligand. The experimental and computational spectral comparison led assignments of IR and NMR signals. The ADME predictions suggested a good pharmacological profile of the titled compound.

Abstract:In current work, three N-acetyl pyrazoline derivatives ( 1 - 3 ) have been prepared via 1,4-addition reaction of hydrazine on chalcones, followed by cyclization of the adduct in the presence of acetic acid. The structural interpretation of synthesized compounds was accomplished using various characterization techniques like FT-IR, Ultraviolet/visible spectroscopy, 1 HNMR and 13 C NMR. The synthesized compounds were evaluated for antioxidant, Urease inhibitory and Lipoxygenase inhibitory activities and results revealed that compound 2 showed promising antioxidant, Urease inhibitory and Lipoxygenase inhibitory activity with IC 50 values 33.2± 0.45, 32.7 ± 0.11 and 42.2 ±0.77, respectively. Moreover, DNA binding interactions of the N-acetyl pyrazoline derivatives have also been carried out with SS-DNA (Salmon Sperm DNA) using absorption spectroscopy. Furthermore, molecular docking was conducted for a ready comparison with the experimental data and to determine the interactions involved. ADMET profiling of pyrazoline derivatives was also done using Admet SAR. Besides that, quantum chemical calculations using DFT (density functional theory) were carried out to explore optimized geometries, FMOs (Frontier molecular orbitals), NBOs (Nonbonding orbitals) of synthesized compounds and to predict their NLO (Nonlinear optical) properties. The synthesized compounds showed much better NLO behavior than standard Urea along with a strong agreement between computed and empirically recorded spectral data.

Abstract:An important finding is that the ketalization of 2‐bromo-3-(3,5-di‐tert‐butyl‐4-hydroxyphenyl)-1-phenylprop-2-en-1-one in the presence of amine has been presented. The structure of obtained 2-(3,5-di‐tert‐butyl‐4-hydroxyphenyl)-3,3-diethoxy-1-phenylpropan-1-one is fully characterized by IR, MS, 1 H, 13 C NMR spectrometry, as well as X-ray diffractometry. Experimental spectrum values of the molecule were compared with experimental data. Optimized structures of the molecule were obtained on the B3LYP, HF, M062X methods and 6–31++ G (d,p) basis set. Additionally, Hirshfeld surface analyses was performed in order to obtain information about the interactions in the crystal packing.

Abstract:Thiazolidinones, widely utilized as a framework for synthesizing diverse molecules, demonstrate pharmacological activity and contribute to the creation of compounds with various biological effects, including antimicrobial, analgesic, antitumor, anti-inflammatory, anti-HIV, and antitubercular properties. The primary objective of this study was to develop a novel organic heterocyclic compound. Researchers successfully achieved this goal by synthesizing (Z)-2-((4-methoxycyclohexa-2,5‐dien-1-yl) imino)-3-methyl thiazolidin-4-one dihydrate (ZMThD), and the structural features of this newly synthesized compound were determined using single crystal X-ray diffraction (XRD). The compound crystallized in the orthorhombic system's P2 1 2 1 2 1 space group. A notable feature of the structure is the precisely measured dihedral angle of 45.0° between the benzene and thiazolidinone rings, indicating a lack of π-π stacking and confirming the non-planar nature of the structure.To further support and elucidate the experimental findings, density functional theory (DFT) calculations were conducted using the B3LYP functional in conjunction with a 6–311 G (d, p) basis set. The optimized geometry results obtained from these calculations closely mirrored the experimental results. Additionally, the stability of the crystal structure was examined through Hirshfeld surface analysis and 2D fingerprint plots, revealing that H...H interactions (50.7%) and H...C/C...H interactions (24.3%) were the primary contributors to the intermolecular forces stabilizing the crystalline structure. The potent inhibitory activity of the screened compounds was rationalized through in-silico molecular modeling studies against the Insulin-degrading enzyme (IDE) enzyme. The compound exhibited a remarkable docking score of −6.8 Kcal/mole, characterized by hydrogen-bond and hydrophobic interactions.

Abstract:The compound S‐propyl (E)‐2‐(1,1‐dioxido‐2‐propylbenzo[d]isothiazol‐3(2H)‐ylidene)‐1‐propylhydrazine‐1‐carbothioate has been synthesized and characterized by different spectroscopic techniques, and its crystal structure has been determined by single‐crystal X‐ray diffraction. A detailed analysis of the main intermolecular interaction in solid‐state has been performed by using Hirshfeld surface analysis and DFT calculations. The title compound, C17H25N3O3S2, hereafter 1, has been prepared and fully characterized by FTIR, 1H NMR and 13C NMR. Its crystal structure was determined by single‐crystal X‐ray diffraction. The crystal packing is stabilized by weak CH···O and CH···π interactions. The CLP‐Pixel method was used to quantify the energetically significant molecular dimers. The intermolecular contacts were identified and quantified using Hirshfeld surfaces (HS) and the corresponding fingerprint plots. The main contributions to the HS of 1 come from HH, OH/HO and CH/HC contacts, which cover about 93% of the total HS surface. The enrichment ratios showed that the favorable contacts accountable for the crystal packing are consistent with their contributions to the HS. Interaction region indicator (IRI) analysis was used to visualize the location and type of intermolecular contacts, allowing identify the CH···O contacts as van der Waals interactions. To visualize the 3D topology of interactions in the crystal structure, interaction energy values were used to construct energy framework diagrams, which showed that the dispersion energy dominates over other interaction energies, as expected for crystal packing governed by weak interactions. Finally, a combination of MEP surface, QTAIM and NCIplot analysis energetically confirmed the existence of CH···O and O···O dichalcogen interactions.

Abstract:The title compound, N′-tert-butyl-N-(3-methoxylbenzoyl)-N-(4-methyl-1,2,3-thiadiazole-5-formylhydrazine (C16H20N4O3S) was prepared from the reaction of 4-methyl-1,2,3-thiadiazole-5-carbonyl chloride with N′-tert-Butyl-3-methoxylbenzohydrazine, and its structure was characterized by 1Hydrogen Nuclear Magnetic Resonance, High-Resolution Mass Spectrometry, IR spectra, and single crystal X-ray diffraction. The crystal of the title compound belongs to monoclinic system, space group P 21/c with cell parameters a = 17.986(2) Å, b = 8.0180(10) Å, c = 12.0190(14) Å, α = 90°, β = 91.160(5)°, γ = 90°, V = 1732.9(4) Å3, Z = 4, D c = 1.335 g/cm3, μ (Mo Ka) = 0.209 mm−1, F (000) = 736, R = 0.0367 and wR = 0.0932. X-ray diffraction analysis indicates that all rings in the title compound are non-planar. The bioassay results indicated that, the title compound had good fungicide activity against Sclerotinia sclerotiorum, certain extent of insecticidal activity against Plutella xylostella L.

Abstract:A novel series of isatin-s-triazine hydrazone derivatives has been synthesized and reported herein. The synthetic methodology involved the reaction of s-triazine hydrazine precursors with isatin derivatives in the presence of CH3COOH as a catalyst and EtOH as solvent to afford the corresponding target products 6a-e in high yields and purities. The characterization data obtained from elemental analysis, FT-IR, NMR (1H- and 13C-) were in full agreement with the expected structures. Furthermore, an X-ray single crystal diffraction study of one of the target s-triazine hydrazone derivatives, 6c confirmed the structure of the desired compounds. It crystallized in the triclinic crystal system and P-1 space group with a = 10.3368(6) Å, b = 11.9804(8) Å, c = 12.7250(5) Å, α = 100.904(4)°, β = 107.959(4)° and γ = 109.638(6)°. The different non-covalent interactions which contributed in the molecular packing of 6c were analyzed using Hirshfeld analysis. The molecular packing of the organic part of the crystal structure showed important O...H (7.1%), C...H (16.4%), C...C (1.6%), H...H (34.8%), N...H (8.0%) and C...N (4.0%) interactions while for the crystal solvent, the O...H (21.3%), H...H (61.2%) and N...H (8.1%) contacts are the most significant. The studied compound 6c is polar and has a net dipole moment of 5.6072 Debye based on DFT study.

Abstract:The title compound, 4-(4-methoxyphenethyl)-5-(p-tolyl)-2,4-dihydro-3H-1,2,4-triazol-3-one (C18H19N3O2), was characterized by single crystal X-ray diffraction. In order to compare the experimental and theoretical compatibility, the DFT and HF modeling technique was also used. When the theoretical and experimental results are compared, it is seen that the geometric parameters from both investigation techniques are quite compatible. X-ray diffraction (XRD) analysis shows that the structure has crystallized in the orthorhombic space group Pna21. The planes of the triazole and benzyl rings in the molecule make dihedral angles of 46.14(1)ο (C1-C6) and 43.89(1)ο (C12-C17). The molecules in the asymmetric unit are linked by intermolecular N–H···O hydrogen bonds, forming a three-dimensional network. Frontier molecular orbital (FMO) analysis was performed to describe intramolecular interactions. For the molecule the energy difference between the lowest unoccupied molecular orbital (LUMO) and highest occupied molecular orbital (HOMO) was calculated as − 4.95 eV. Potential energy surface (PES) analysis was performed by a semi-empirical method to determine the stable states of molecular structure and to compare them with XRD geometry. Abstract:Hydrazonylsulfones such as Bt-NHNHSO2R and their iminotautomers have been studied as optical materials and for their biological potential. In this work, a structural study has been carried out on N-(1,3-benzothiazol-2-yl)-4-(halogenobenzenesulfonyl)-hydrazides (1: X = F, Cl, Br). For (1: X = F), single-crystal X-ray diffraction, Hirshfeld surface analysis, and PIXEL calculations were conducted, while in (1: X = Cl) and (1: X = Br), only single-crystal X-ray diffraction studies were successfully conducted due to the disordering of the solvent. Each compound crystallises with two independent but similar amino tautomers in the asymmetric units: compound (1: X = F) crystallises in the monoclinic P21/c, and the isostructural pair (X: 1 = Cl and Br) crystallises in the tetragonal P-421c space group. In the most stable motif of the supramolecular arrangement, the molecules of the asymmetric unit are connected by classical N–H(hydrazinyl)···N(thiazoyl) hydrogen bonds and several face-to-face, offset π···π interactions. This motif has a very powerful influence on the crystal structure due to its direct links with the other weaker motifs. Other significant intermolecular interactions found in the structure include N–H(hydrazonyl)···O(sulfonate) bonds. Analogous intermolecular interactions were found in similar compounds, leading to the conclusion that those interactions are the most important instabilizing the solid state of hydrazonylsulfones.

Abstract:ABSTRACT. In this study, five different thiourea derivatives were synthesized from aryl amines according to the reported method. 1-Benzoyl-3-(4-methoxyphenyl)thiourea (2) was confirmed with single crystal XRD analysis while 1-benzoyl-3-phenylthiourea (1), 1-benzoyl-3-(4-hydroxyphenyl)thiourea (3), 1-benzoyl-3-(2-nitrophenyl) thiourea (4) and 1-benzoyl-3-p-tolylthiourea (5) were elucidated with FTIR and NMR techniques. The geometry optimization of the targeted molecules was accomplished with density functional theory applying B3LYP function. The experimental (XRD) and calculated (DFT) bond angles and bond lengths were compared. The frontier molecular orbitals and molecular electrostatic potential were computed to determine the charge density distribution and possible sites for electrophilic and nucleophilic reactions of the crystalline compound. The synthesized compounds were evaluated as an anti-radical scavenger and enzyme (esterases and protease) inhibitor using in-vitro models. The results confirmed that the synthesized molecules have good anti-oxidant property while a moderate enzyme inhibiting activity. Docking study was conducted with acetylcholine and butyrylcholine esterase which suggested that molecules under study have a potential to inhibit these esterases and protease enzymes. On the basis of in-vitro studies, it is concluded that compound 2 is most active against all tested assays. Abstract:A novel (E)-1-(4-methylbenzylidene)-4-(3-isopropylphenyl) thiosemicarbazone was synthesized in a one-pot four-step synthetic route. Fourier transform infrared spectroscopy (FTIR), 1H and 13C nuclear magnetic resonances (NMR), single-crystal X-ray diffraction, and UV-visible absorption spectroscopy were utilized to confirm the successful preparation of the title compound. Single-crystal data indicated that the intramolecular hydrogen bond N(3)-H(3)···N(1) and intermolecular hydrogen bond N(2)-H(2)···S(1) (1 − x, 1 − y, 1 − z) existed in the crystal structure and packing of the title compound. Besides the covalent interaction, the non-covalent weak intramolecular hydrogen bond N(3)-H(3)···N(1) discussed by atoms in molecules (AIM) theory also functioned in maintaining the title compound's crystal structure. The strong intermolecular hydrogen bond N(2)-H(2)···S(1) (1 − x, 1 − y, 1 − z) discussed by Hirshfeld surface analysis played a major role in maintaining the title compound's crystal packing. The local maximum and minimum electrostatic potential of the title compound was predicted by electrostatic potential (ESP) analysis. The UV-visible spectra and HOMO-LUMO analysis revealed that the title compound has a low ΔEHOMO–LUMO energy gap (3.86 eV), which implied its high chemical reactivity due to the easy occurrence of charge transfer interactions within the molecule. Molecular docking and in vitro antifungal assays evidenced that its antifungal activity is comparable to the reported pyrimethanil, indicating its usage as a potential candidate for future antifungal drugs.

Abstract:A single crystal of new 2-substitued quinazoline derivative has been successfully synthesized through a simple and novel process based on a two-step reaction involving cyclisation, oxidation and hydrasination. The structure of the newly synthesized product was fully characterized by spectroscopic analysis NMR ( 1 H & 13 C), IR and HRMS. This intriguing compound crystalizes crystallises in a monoclinic system with P21/c space group. Notably, the stability of crystal packing was assured via hydrogen bonds, π- stacking and van der Waals interactions. An intermolecular interaction examination of the crystal structure was performed using Hirshfeld surface analysis and two-dimensional fingerprint plots. In addition, the electrostatic surface potential was calculated using density functional theory. In addition, the 3-amino-2-(2-thienyl)-4(3H)-quinazolinone, was applied to selected bacterial strains, and its antibacterial activity was assessed. In addition, a molecular docking study has been realised.

Abstract:The diethyl 3-(4-substitutedbenzoyl)indolizine-1,2-dicarboxylates have been synthesized by reacting the electron-deficient diethyl but-2-ynedioate with 1-(2-(4-substitutedphenyl)-2-oxoethyl)pyridin-1-ium bromide, the intermediates obtained from the reaction of pyridine and 4-substituted phenacyl bromides. The synthesized compounds were investigated by FT-IR, NMR ( 1 H and 13 C), LC-MS, and elemental analysis spectroscopic methods along with PXRD. The molecular structures were determined using the single crystal X-ray diffraction technique which revealed that 5a and 5c crystallize in triclinic space group P 1 ̄ . The crystal structures of 5a and 5c preferred the C-H ··· O hydrogen-bonded dimeric ring motifs, resembling those observed in the earlier reported 5b . Furthermore, various computational analyzes such as Hirshfeld surface, Energy framework, and MEP analysis predicted the interaction site and inter-molecular interactions in these molecules. The FMOs analysis was carried out to obtain global reactivity parameters for the synthesized compounds ( 5a-c ). The NCI and QTAIM studies revealed the nature as well as qualitative and quantitative description of intra and inter-molecular hydrogen bonding interactions present in these molecules and their crystal packing.

Abstract:In this paper, we reported the development of an efficient and highly productive synthetic protocol for preparing new 2,5-diamino-1,3,4-thiadiazole derivatives functionalized with N -(2,2,2-trichloroethyl)carboxamide group. The method of their preparation was based on the reaction of oxidative dehydrosulfurization of N -(2,2,2-trichloro-1-(2-(phenylcarbamothioyl)hydrazine-1-carbothioamido)ethyl)carboxamides and N -(1-(2-carbamothioylhydrazine-1-carbothioamido)-2,2,2-trichloroethyl)benzamide under the action of a mixture of iodine and triethylamine in DMF. This reaction took place at room temperature for two hours and allowed obtaining the target products with a 69-75% yield. Using the developed protocol, we obtained ten new derivatives of 1,3,4-thiadiazole. Their structure was proven by IR, 1 H NMR, 13 C NMR, 1 H- 1 H COSY, 1 H- 13 C HSQC, 1 H- 13 C HMBC spectroscopy, and LC/HRMS spectrometry.

Abstract:The current study focuses on the synthesis of a new organic compound namely, ethyl 2-benzoyl-3,3-bis(methylthio)acrylate, whose structural characterization was performed by single-crystal X-ray analysis. The central portion of the title molecule is planar. The benzoyl and one methylthio group are perpendicular to this plane while the ester and the other methylthio group lie close to the plane. In the crystal, C–H···O hydrogen bonds form chains along the b -axis direction which pack with normal Van der Waals interactions. A Hirshfeld surface (HS) analysis showed H⋅⋅⋅H contacts account for 48.3% of the intermolecular contacts in the crystal packing. The energy framework analysis shows that the contribution of dispersion forces in crystal packing is dominant among other energies. The density functional theory (DFT) study using the 6–31 + G(d,p) basis set provides further insight into reactivity and electronic and optical properties in the gas phase. The HOMO–LUMO gap for the title compound is small which indicates its soft nature and high reactivity. Natural population analysis (NPA) reveals excellent charge transfer and donor–acceptor properties in the compound. The computed global reactivity descriptors also indicate the electronic stability and reactivity of the structure. Molecular electrostatic potential (MESP) map suggested electron-rich sites around the carbonyl (C = O) and oxygen atoms while the corner carbon atoms of the benzene ring are suitable for nucleophilic attack. Furthermore, the significant dipole moment ( μ o ) and polarizability ( α o ) suggest its high polarizability and optical properties. The measured static first hyperpolarizability ( β o ) value of 266.74 au indicates its nonlinear optical (NLO) properties.

Abstract:This work reports the convenient approach for the synthesis of thiazole based thiourea derivatives (1-21) from 2-bromo-1-(4-fluorophenyl)thiazole-1-one and phenyl isothiocyanates. The scope and diversity were achieved from readily available phenyl isothiocyanates. This protocol involves an oxidative C-S bond formation. Moreover, hybrid thiazole based thiourea scaffolds (1-21) according to literature known protocol were screened in vitro for anticancer Potential against breast cancer, antiglycation and antioxidant inhibitory profile. All newly developed scaffolds were showed moderate to good inhibitory potentials ranging from 0.10 ± 0.01 μM to 11.40 ± 0.20 μM, 64.20 ± 0.40 μM to 385.10 ± 1.70 μM and 8.90 ± 0.20 μM to 39.20 ± 0.50 μM against anticancer, antiglycation and antioxidant respectively. Among the series, compounds 12 (IC 50 = 0.10 ± 0.01 μM), 10 (IC 50 = 64.20 ± 0.40 μM) and 12 (IC 50 = 8.90 ± 0.20 μM) with flouro substitution at phenyl ring of thiourea were identified to be the most potent among the series having excellent anticancer, antiglycation and antioxidant potential. The structure of all the newly synthetics scaffolds were confirmed by using different types of spectroscopic techniques such as HREI-MS, 1 H- and 13 C-NMR spectroscopy. To find structure-activity relationship, molecular docking studies were carried out to understand the binding mode of active inhibitors with active site of enzymes and results supported the experimental data.
