Abstract:Chiral C‐Glycosyl amino acids are prevalent structural motifs found in pharmaceuticals and natural products, making them a significant dimension for exploration in modern pharmaceutical and medicinal chemistry. However, the current synthesis methods for these compounds face numerous environmental and practical challenges, highlighting the critical need to establish more sustainable approaches. Herein, we report a simple and powerful strategy for the stereoselective synthesis of C‐glycoamino acids. This method features the design and use of (‐)/(+)‐camphorsultam as an ideal chiral auxiliary, shielding one prochiral face of the glycine α‐carbon efficiently and imposing a high kinetic barrier for the epimerization of thus‐generated chiral α‐carbon that two configuration products of R and S could be achieved independently. It proceeds under mild conditions, shows broad scope, and allows for the direct stereoselective glycosylation without metal and chiral ligand.

Abstract:C -Glycosyl peptides possess excellent metabolic stability and therapeutic properties, and thus play critical roles in biological studies as well as drug discoveries. However, the limited accessibility of C -glycosyl amino acids has significantly hindered the broader research of their structural features and mode of action. Herein, for the first time we disclose a novel visible-light-driven radical conjugate addition of 1,4-dihydropyridine (DHP)-derived glycosyl esters with dehydroalanine derivatives, generating C -glycosyl amino acids and C -glycosyl peptides in good yields with excellent stereoselectivities. Redox-active glycosyl esters, as readily accessible and bench-stable radical precursors, could be easily converted to glycosyl radicals via anomeric C( sp 3 ) - O bond homolysis under mild conditions. Importantly, the generality and practicality of this transformation was fully demonstrated in >40 examples including 2-dexosugars, oligosaccharides, oligopeptides, and complex drug molecules. Given its mild reaction conditions, robust sugar scope, and high anomeric control and diastereoselectivity, the method presented herein could find widespread utility in the preparation of C ( sp 3 )-linked sugar-based peptidomimetics.

Abstract:C(sp(3))-H and C(sp(2))-H glycosylations of structurally complex amino acids and peptides were accomplished through the assistance of triazole peptide-isosteres. The palladium-catalyzed peptide-saccharide conjugation provided modular access to structurally complexC-alkyl glycoamino acids, glycopeptides andC-aryl glycosides, while enabling the assembly of fluorescent-labeled glycoamino acids. The C-H activation approach represents an expedient and efficient strategy for peptide late-stage diversification in a programmable as well as chemo-, regio-, and diastereo-selective fashion.

Abstract:The glycine motif is widely prevalent in bioactive peptides. Thus, the direct and precise modification of glycine derivatives has attracted significant attention over the past few decades. Among various protocols for the modification of glycine derivatives, the visible-light-driven direct α-C(sp3)–H bond functionalization of glycine derivatives has emerged as a powerful tool to achieve this objective, owing to its merits in atom economy, selectivity, reaction simplicity, and sustainability. This review summarizes the recent advancements in visible-light-driven direct α-C(sp3)–H bond functionalization of glycine derivatives. The contents of this review are organized based on the photocatalysts employed and the various reaction modes in the functionalization process. The mechanism, the challenges encountered, and future trends are also discussed, enabling readers to understand the current developmental status in this field.

Abstract:C‐alkyl glycosides play a crucial role in various bioactive compounds. However, the synthesis of C‐alkyl glycosides poses significant challenges, particularly through C(sp3)‐H glycosylation. Here, we report a set of reactions for constructing C‐alkyl glycosides through directing‐group‐mediated functionalization of unactivated γ‐C(sp3)‐H bonds under mild conditions. Those reactions not only achieve high regioselectivity and stereoselectivity in glycosylation, but also exhibit a wide substrate scope. They are compatible with both arene and alkane substrates, as well as natural and unnatural amino acid substrates. Mechanistic studies have shown that the directing‐group 8‐aminoquinoline (AQ) and picolinamide (PA) may affect the chirality of the β‐carbon of L‐valine through a sterically favorable trans‐palladacycle intermediate, resulting in (R) or (S) configuration of glycosylated amino acid, respectively. These reactions are promising to provide a convenient and powerful tool for constructing C‐alkyl glycosides and carbohydrate‐based drugs in the future.

Abstract:The glycosylation of peptides and proteins can significantly impact their intrinsic properties, such as conformation, stability, antigenicity, and immunogenicity. Current methods for preparing N-linked glycopeptides typically rely on amide bond formation, which can be limited by the presence of reactive functional groups like acids and amines. Late-stage functionalization of peptides offers a promising approach to obtaining N-linked glycopeptides. In this study, we demonstrate the preparation of N-linked glycopeptides through a photoredox-catalyzed site-selective Giese addition between N-glycosyl oxamic acid and peptides containing dehydroalanine (Dha) under visible light conditions. Unlike traditional methods that rely on the coupling of aspartic acid and glycosylamine, this approach utilizes the conjugation of N-glycosylated carbamoyl radicals with Dha, facilitating the straightforward modification of complex peptides.

Abstract:S‐Glycosides are considerably more stable toward chemical degradation and enzymatic hydrolysis than O‐glycosides, and thioglyco‐peptides/proteins also demonstrate critical bioactivities in the living system. However, the general and stereoselective synthetic strategies are still highly challenging. Herein, a stereospecific S‐glycosylation approach was developed by palladium‐catalyzed decarboxylative allylation to form α‐ and β‐thioglycosides respectively under mild conditions. A wide range of substrates were tolerated to afford thioglycosides in high yields with excellent diastereoselectivities. This protocol created the facile synthesis of various S‐linked disaccharides and glycopeptides.

Abstract:A metal-free, visible-light driven C-H ketoalkylation of glycine derivatives and peptides with cycloalkyl hydroperoxides is presented. This protocol provides a straightforward route to unnatural amino acids bearing a distal carbonyl group with moderate to good yields. Preliminary mechanistic studies revealed that both a SET event and radical chain propagation were involved in this transformation.

Abstract:The presence of a pI 9.0 cationic peroxidase isozyme from milk stage pericarp of six susceptible and five resistant inbreds was correlated significantly with previously reported field data on percentage infection by Aspergillus flavus in the inbreds and their hybrids. The isozyme was constitutively expressed in some additional maize tissues and lines examined, and frequently induced by mechanical damage, heat shock, Fusarium proliferatum, and/or Bacillus subtilis in other lines tested. Native/IEF two-dimensional electrophoresis identified the isozyme as the previously genetically identified px5. A cDNA clone expressed in black Mexican sweet (BMS) maize cell cultures produced the pI 9.0 isozyme. In addition to potential use in marker-assisted breeding, enhanced expression of this cationic peroxidase through breeding or genetic engineering may lead to enhanced disease or insect resistance.

Abstract:AbstractWe have developed a highly efficient and practical approach for palladium‐catalyzed trifluoroacetate‐promoted N‐quinolylcarboxamide‐directed glycosylation of inert β‐C(sp3)−H bonds of N‐phthaloyl α‐amino acids with glycals under mild conditions. For the first time, C(sp3)−H activation for glycosylation was achieved to build C‐alkyl glycosides. This method facilitates the synthesis of various β‐substituted C‐alkyl glycoamino acids and offers a tool for glycopeptide synthesis.

Abstract:Photochemical glycosylation has attracted considerable attention in carbohydrate chemistry. However, to the best of our knowledge, visible-light-promoted glycosylation via photoactive glycosyl donor has not been reported. In the study, we report a photosensitizer-free visible-light-mediated glycosylation approach using a photoactive 2-glycosyloxy tropone as the donor. This glycosylation reaction proceeds at ambient temperature to give a wide range of O -glycosides or oligosaccharides with yields up to 99%. This method is further applied in the stereoselective preparation of various functional glycosyl phosphates/phosphosaccharides, the construction of N -glycosides/nucleosides, and the late-stage glycosylation of natural products or pharmaceuticals on gram scales, and the iterative synthesis of hexasaccharide. The protocol features uncomplicated conditions, operational simplicity, wide substrate scope (58 examples), excellent compatibility with functional groups, scalability of products (7 examples), and high yields. It provides an efficient glycosylation method for accessing O / N -glycosides and glycans.

Abstract:Carbohydrates play pivotal roles in an array of essential biologic processes and are consequently involved in many diseases. To meet the needs of glycobiology research, chemical enzymatic and non-enzymatic methods have been developed to generate glycoconjugates with well-defined structures. Herein, harnessing the unique properties of C6-oxidized glycans, we report a straightforward and robust strategy for site- and stereo-selective glyco-modification of biomolecules with N-terminal tryptophan by a carbohydrate-promoted Pictet-Spengler reaction, which is not adapted to typical aldehyde substrates under biocompatible conditions. This method reliably delivers highly homogeneous glycoconjugates with stable linkages, which has great potential for functional modulation of peptides and proteins in glycobiology research. Moreover, this reaction can be performed at the glyco-sites of glycopeptides, glycoproteins and living cell surfaces in a site-specific manner. Control experiments indicated that the protected α-O atom of aldehyde donors and free N-H bond of the tryptamine motif are crucial for this reaction. Mechanistic investigations demonstrated that the reaction exhibited a first-order dependence on both tryptophan and glycan, and deprotonation/rearomatization of the pentahydro-β-carbolinium ion intermediate might be the rate-determining step.

Abstract:Glycoamino acids and glycoproteins, especially C‐glycoamino acids and C‐glycoproteins, attract tremendous interests in the realm of biology and chemistry due to their wide occurrence in natural products, excellent pharmacodynamic properties and dynamic stability. Compared with the traditional construction of the amino acid or the glycosyl moiety de novo, post‐modification of amino acids and peptides through direct glycosylation reactions constitute a powerful synthetic approach to attaining C‐glycoamino acids and C‐glycopeptides rapidly on account of the high step‐economy, broad functional group tolerance, and conservation of the chirality of the amino acid and glycosyl moieties. In this review, the recent developments in the synthesis of C‐glycoamino acids and C‐glycopeptides via the post‐modification of amino acids and peptides are summarized. Additionally, the important and representative examples, synthetic applications, and crucial mechanism investigations of these glycosylation reactions are also highlighted.

Abstract:A room temperature, visible-light-promoted and redox neutral direct C-H amination of glycine and peptides has been firstly accomplished by using N-acyloxyphthalimide or -succinimide as nitrogen-radical precursor. The present strategy provides ways to introduce functionalities such as N-acyloxyphthalimide or -succinimide specifically to terminal glycine segment of peptides. Herein, mild conditions and high functional-group tolerance allow the preparation of non-natural α-amino acids and modification of corresponding peptides in this way.

Abstract:We report a simple oxidative Csp3‐H functionalization reaction of glycine derivatives by visible‐light photoredox catalysis. A wide range of glycine derivatives readily undergo the oxidative cyclization readily to afford various 2‐substituted benzoxazoles. Importantly, this photocatalytic intramolecular dehydrogenative coupling reaction allows for the C‐H functionalization of glycine derivatives involving short peptides under mild conditions, which may have value in preparing peptide‐derived pharmacologically active molecules.
