Abstract:Agricultural wastes rich in β-mannan are an important environmental problem in tropical and sub-tropical countries. This research aims at dealing with this and investigates the valorization of mannan-rich copra meal from virgin coconut oil manufacturing into mannan-oligosaccharides (β-MOS) by enzymatic hydrolysis using β-mannanase from Bacillus licheniformis ( Bl Man26B). Lab-scale process, involving pre-treatment and bioconversion steps, were conducted and evaluated. Lyophilized β-MOS was analyzed and its biological activities were assessed. The size of oligosaccharides obtained ranged from dimers to hexamers with 36.7% conversion yields. The prebiotic effects of β-MOS were demonstrated in comparison with commercial inulin and fructo-oligosaccharides (FOS). In vitro toxicity assays of β -MOS on human dermal fibroblasts and monocytes showed no cytotoxic effect. Interestingly, β-MOS at concentrations ranging from 10 to 200 μg/mL also demonstrated potent anti-inflammatory activity against LPS-induced inflammation of human macrophage THP-1 in a dose-dependent manner. However, at high dose, β-MOS could also stimulate inflammation. Therefore, further investigation must be conducted to ensure its efficacy and safe use in the future. These results indicate that β-MOS have the potential to be used as valued-added health-promoting nutraceutical or feed additive after additional in-depth studies. These finding should be applicable for other agricultural wastes rich in mannan as well.

Abstract:Aspergillus quadrilineatus endo-β-mannanase effectively degraded konjac glucomannan (66.09% w/v), copra meal (38.99% w/v) and locust bean galactomannan (20.94% w/v). High performance liquid chromatography (HPLC) analysis of KG hydrolysate indicated its mannooligosaccharides (MOS) content (656.38 mg/g) with high amounts of DP 5 oligosaccharide. Multi-scale characterization of mannan hydrolysate was done using FTIR and 13C NMR which revealed α and β form of galactose or glucose in MOS, respectively. CM and LBG hydrolysates (1 mg/mL) have shown cytotoxic effect and reduced cell viability of Caco-2 cells by 45% and 62%, respectively. MOS DP (1-4) derived from LBG supported better Lactobacilli biofilm formation as compared to KG hydrolysate containing high DP MOS (5-7). Lactobacilli effectively fermented MOS to generate acetate and propionate as main short chain fatty acids. Lactobacilli produced leucine, isoleucine and valine as branched chain amino acids when grown on LBG hydrolysate.

Abstract:sEH (soluble epoxide hydrolase), which is encoded by the EPHX2 gene, regulates the actions of bioactive lipids, EETs (epoxyeicosatrienoic acids). Previously, we found that high-glucose-induced oxidative stress suppressed sEH levels in a hepatocarcinoma cell line (Hep3B) and sEH was decreased in streptozotocin-induced diabetic mice in vivo. In the present study, we investigated the regulatory mechanisms underlying EPHX2 transcriptional suppression under high-glucose conditions. The decrease in sEH was prevented by an Sp1 (specificity protein 1) inhibitor, mithramycin A, and overexpression or knockdown of Sp1 revealed that Sp1 suppressively regulated sEH expression, in contrast with the general role of Sp1 on transcriptional activation. In addition, we found that AP2α (activating protein 2α) promoted EPHX2 transcription. The nuclear transport of Sp1, but not that of AP2α, was increased under high glucose concomitantly with the decrease in sEH. Within the EPHX2 promoter -56/+32, five Sp1-binding sites were identified, and the mutation of each of these sites showed that the first one (SP1_1) was important in both suppression by Sp1 and activation by AP2α. Furthermore, overexpression of Sp1 diminished the binding of AP2α by DNA-affinity precipitation assay and ChIP, suggesting competition between Sp1 and AP2α on the EPHX2 promoter. These findings provide novel insights into the role of Sp1 in transcriptional suppression, which may be applicable to the transcriptional regulation of other genes.

Abstract:The purpose of this study was to clarify effects of selected oligosaccharides on concentrations of cecal short-chain fatty acids (SCFAs), total large bowel wet weight and wall weight, and cecal microbiota levels in mice. Mice were respectively given gavage of selected fructooligosaccharides (FOS), galactooligosaccharides (GOS), mannanoligosaccharides (MOS), and chitooligosaccharides (COS) [1000 mg/(kg body weight·d)]. Control group was given physiological saline solution. After 14 d treatment, SCFAs and lactate in mice cecum were significantly increased ( P <0.05) by intake of oligosaccharides, especially FOS and GOS. Thus, providing these oligosaccharides as ingredients in nutritional formulas may benefit the gastrointestinal tract.
Bioconversion of spent coffee grounds to prebiotic mannooligosaccharides – an example of biocatalysis in biorefinery

Abstract:Spent coffee ground (SCG), an agro-industrial waste, have a high content of polysaccharides such as mannan, making it ideal for utilisation for the production of nutraceutical oligosaccharides. Recently, there has been growing interest in the production of mannooligosaccharides (MOS) for health promotion in humans and animals. MOS are reported to exhibit various bioactive properties, including prebiotic and antioxidant activity. In this study, SCG was Vivinal pretreated using NaOH, characterized and hydrolysed using a Bacillus sp. derived endo -β-1,4-mannanase, Man26A, for MOS production. Structural analyses using Fourier-transform infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA) were conducted to assess the efficacy of the pretreatment. Lignin removal by the pretreatment from SCG was clearly shown by TGA. FT-IR, on the other hand, showed the presence of α-linked D -galactopyranoside (812 cm −1 ) and β-linked D -mannopyranoside residues (817 cm −1 ) in both SCG samples, signifying the presence of mannan. Hydrolysis of pretreated SCG by Man26A produced mannobiose and mannotriose as the main MOS products. The effect of simulated gastric conditions on the MOS was investigated and showed this product to be suitable for oral administration. Finally, the prebiotic effect of the MOS on the growth of selected beneficial bacteria was investigated in vitro ; showing that it enhanced Lactobacillus bulgaricus , Bacillus subtilis and Streptococcus thermophilus growth. These findings suggest that SCG is a viable source for the production of MOS which can be orally administered as prebiotics for effecting luxuriant growth of probiotic bacteria in the host's digestive tract, leading to a good health status.

Abstract:A growing demand in novel food products for well-being and preventative medicine has attracted global attention on nutraceutical prebiotics. Various plant agro-processes produce large amounts of residual biomass considered “wastes”, which can potentially be used to produce nutraceutical prebiotics, such as manno-oligosaccharides (MOS). MOS can be produced from the degradation of mannan. Mannan has a main backbone consisting of β-1,4-linked mannose residues (which may be interspersed by glucose residues) with galactose substituents. Endo-β-1,4-mannanases cleave the mannan backbone at cleavage sites determined by the substitution pattern and thus give rise to different MOS products. These MOS products serve as prebiotics to stimulate various types of intestinal bacteria and cause them to produce fermentation products in different parts of the gastrointestinal tract which benefit the host. This article reviews recent advances in understanding the exploitation of plant residual biomass via the enzymatic production and characterization of MOS, and the influence of MOS on beneficial gut microbiota and their biological effects (i.e., immune modulation and lipidemic effects) as observed on human and animal health.

Gut microbiota modulation, prebiotic and bioactive characteristics of date pomace polysaccharides extracted by microwave-assisted deep eutectic solvent

Abstract:This study investigated the characteristics of polysaccharides from date pomace using microwave-assisted deep eutectic solvents. The impact on the gut microbiota and probiotics growth was examined in vitro. The study also examined its antioxidant properties, ability to inhibit enzymes linked to diabetes and high blood pressure, impact on cell growth, and physical properties. The isolated MPS had an average molecular weight of 8073.38 kDa and contained mannose, galacturonic acid, galactose, glucose, and fructose in specific proportions. At a concentration of 1000 mg/L, MPS showed strong antioxidant activity, with significant scavenging rates in various tests such as DPPH (57.0 ± 1.05 %) and ABTS (66.4 ± 2.48 %). MPS displayed 77 %, 80 %, and 43 % inhibition for α-amylase, α-glucosidase, and ACE-inhibition, respectively. MPS displayed significant antiproliferative effects, achieving 100 % and 99 % inhibition against Caco-2 and MCF-7 cells at 2500 mg/L, respectively. MPS showed broad-spectrum antibacterial properties against both Gram-positive and Gram-negative foodborne bacteria. Gemmiger formicilis, Blautia species, Collinsella aerofaciens, and Bifidobacterium longum showed strong positive correlations, suggesting increased SCFA production. Network analysis indicated species correlations, with 86 % showing negative correlations with Escherichia and Enterococcus saccharolyticus. MPS was abundant in Firmicutes, Actinobacteria, and Proteobacteria phyla. Date pomace could serve as a dietary fiber source, promoting better health.
Effects of Carboxymethyl Modification on the Acidic Polysaccharides from Calocybe indica: Physicochemical Properties, Antioxidant, Antitumor and Anticoagulant Activities

Abstract:An acidic polysaccharide fraction was obtained from Calocybe indica (CIP3a) after subjecting it to hot water extraction followed by purification through DEAE-cellulose 52 and Sepaharose 6B column chromatography. The CIP3a was further modified using chloroacetic acid to yield carboxymethylated derivatives (CMCIP3a). The modified polysaccharide was characterized using various spectroscopic methods. In addition, further antioxidant, antitumor and anticoagulant activities were also investigated. The polysaccharides CIP3a and CMCIP3a were heterogeneous in nature and composed of various molar percentages of glucose, arabinose and mannose with molecular weights of 1.456 × 103 and 4.023 × 103 Da, respectively. The NMR and FT-IR data demonstrated that the carboxymethylation on the polysaccharide was successful. In comparison to CIP3a polysaccharides, the modified derivatives had lower sugar and protein contents, and higher levels of uronic acid. The in vitro antioxidant activity showed that CMCIP3a with higher molecular weight displayed an elevated ability in scavenging the DPPH radical, ABTS, superoxide, hydroxyl radical, ferric reducing power, cupric reducing power and erythrocyte hemolysis inhibition with an EC50 value of 2.49, 2.66, 4.10, 1.60, 3.48, 1.41 and 2.30 mg/mL, respectively. The MTT assay results revealed that CMCIP3a displayed a dose-dependent inhibition on five cancer cells (HT29, PC3, HeLa, Jurkat and HepG-2) in the range of 10–320 μg/mL. The APTT, PT and TT were significantly extended by CMCIP3a in relation to dosage, indicating that the anticoagulant effect of CIP was both extrinsic and intrinsic, along with a common coagulation pathway. These findings demonstrated that carboxymethylation might effectively improve the biological potential of the derivatives and offer a theoretical framework for the creation of novel natural antioxidants, low-toxicity antitumor and antithrombotic drugs.

Abstract:Optimized production of Aspergillus niger ATCC 26011 endo-β-mannanase ( ManAn ) on copra meal resulted in 2.46-fold increase (10,028 U/gds). Purified ManAn (47 kDa) showed high affinity towards guar gum (GG) as compared to konjac gum and locust bean gum with K m 2.67, 3.25 and 4.07 mg/mL, respectively. ManAn efficiently hydrolyzed GG and liberated mannooligosaccharides (MOS). Changes occurring in the rheological and compositional aspects of GG studied using Differential scanning calorimetry (DSC), Thermal gravimetric analysis (TGA) and X-ray diffraction (XRD) revealed increased thermal stability and crystallinity of the partially hydrolyzed guar gum (PHGG). Parametric optimization of the time and temperature dependent hydrolysis of GG (1% w/v) with 100 U/mL of ManAn at 60 °C and pH: 5.0 resulted in 12.126 mg/mL of mannotetraose (M4) in 5 min. Enhanced growth of probiotics Lactobacilli and production of short chain fatty acids (SCFA) that inhibited enteropathogens, confirmed the prebiotic potential of PHGG and M4.

Prebiotic efficacy of coconut kernel cake’s soluble crude polysaccharides on growth rates and acidifying property of probiotic lactic acid bacteria in vitro

Abstract:This paper reports the prebiotic efficacy of soluble crude polysaccharides (SCP) from palm kernel cake on the growth rates and acidifying property of probiotic lactic acid bacteria (LAB), Lactobacillus plantarum ATCC 8014 and Lactobacillus rhamnosus ATCC 53103, to investigate into the potential SCP of an agriculture by-product as a novel prebiotic. The highest extraction yield of SCP (2.90%) was obtained using NaOH. The extracted polysaccharides were highly soluble (>97%). The highest percentage of protein (0.63 ± 0.09%) and the total carbohydrate content (55.36 ± 0.33%) were observed in NaOH-extracted SCP (SCPN) and citric acid-extracted SCP (SCPCA). Water-extracted SCP (SCPW) contained the highest percentage of glucose, whereas SCPCA demonstrated the presence of two major monosaccharides, galactose and mannose. Structural analysis of SCP showed peaks in the range of 1000–1077 cm−1, an indication of the presence of polysaccharides and alpha (α)- or beta (β)-glycosidic linkages. The degree of hydrolysis increased with the increase of pH from 5 to 8 following 4 h incubation. SCPW and SCPCA were more resistant (95%) to α-amylase as compared to fructooligosaccharides (FOS). As in the case of FOS (∼98%), SCPW (∼99%) and SCPCA (∼98%) were resistant to artificial human gastric juice and their degree of hydrolysis had negative correlation with the pH. The product was found to markedly stimulate the proliferation of the tested probiotics and acid production. All the above findings are supportive of the fact that polysaccharides extracted from CKC have a vast potential to be exploited as novel prebiotic.

In vitro digestibility and prebiotic activities of a bioactive polysaccharide from Moringa oleifera leaves

Abstract:In this study, the digestion and fermentation properties of a bioactive polysaccharide (MOP-2) purified from Moringa oleifera leaves and its impact on the human colonic microbiota were determined using simulated saliva-gastrointestinal digestion and human fecal fermentation models in vitro. The results showed that the simulated saliva and gastric juices had no effect on the average molecular weight (MW) of MOP-2. The MW of MOP-2 slightly decreased from 155.29 to 145.02 kDa during intestinal digestion, and the reducing sugar content increased from 0.159 to 0.234 mg/ml, indicating that MOP-2 was partially degraded during intestinal digestion. During fermentation, MOP-2 was largely used by human fecal inoculums. Notably, MOP-2 could significantly regulate the structure of the microbial community by improving the relative abundances of some beneficial gut microbiota, such as Phascolarctobacterium, Coprococcus, Roseburia, and Bacteroides. Additionally, after fermentation for 48 hr, MOP-2 could significantly improve the production of short-chain fatty acids, especially n-butyric acid, acetic acid, propionic acid, and n-valeric acid. These results suggested that MOP-2 could potentially be a gut microbiota manipulator aimed at promoting gut health. PRACTICAL APPLICATIONS: Gut microbial community is an important part of the human intestinal environment. The health of gut microbiota is closely associated with host heath. This work reported that a polysaccharide (MOP-2) purified from Moringa oleifera leaves could modulate the microbial structure by improving the relative abundances of some beneficial gut microbiota, which provided useful information for the application of MOP-2 as a prebiotic additive in food industry.
Cross-Feeding and Enzymatic Catabolism for Mannan-Oligosaccharide Utilization by the Butyrate-Producing Gut Bacterium Roseburia hominis A2-183

Abstract:β-Mannan is abundant in the human diet and in hemicellulose derived from softwood. Linear or galactose-substituted β-mannan-oligosaccharides (MOS/GMOSs) derived from β-mannan are considered emerging prebiotics that could stimulate health-associated gut microbiota. However, the underlying mechanisms are not yet resolved. Therefore, this study investigated the cross-feeding and metabolic interactions between Bifidobacterium adolescentis ATCC 15703, an acetate producer, and Roseburia hominis A2-183 DSMZ 16839, a butyrate producer, during utilization of MOS/GMOSs. Cocultivation studies suggest that both strains coexist due to differential MOS/GMOS utilization, along with the cross-feeding of acetate from B. adolescentis E194a to R. hominis A2-183. The data suggest that R. hominis A2-183 efficiently utilizes MOS/GMOS in mono- and cocultivation. Notably, we observed the transcriptional upregulation of certain genes within a dedicated MOS/GMOS utilization locus (RhMosUL), and an exo-oligomannosidase (RhMan113A) gene located distally in the R. hominis A2-183 genome. Significantly, biochemical analysis of β-1,4 mannan-oligosaccharide phosphorylase (RhMOP130A), α-galactosidase (RhGal36A), and exo-oligomannosidase (RhMan113A) suggested their potential synergistic role in the initial utilization of MOS/GMOSs. Thus, our results enhance the understanding of MOS/GMOS utilization by potential health-promoting human gut microbiota and highlight the role of cross-feeding and metabolic interactions between two secondary mannan degraders inhabiting the same ecological niche in the gut.

Abstract:Malto-oligosaccharides (MOS) are α-1,4 glycosidic linked linear oligosaccharides of glucose, which have a diverse range of functional applications in the food, pharmaceutical, and other industries. They can be used to modify the physicochemical properties of foods thereby improving their quality attributes, or they can be included as prebiotics to improve their nutritional attributes. The degree of polymerization of MOS can be controlled by using specific enzymes, which means their functionality can be tuned for specific applications. In this article, we review the chemical structure, physicochemical properties, preparation, and functional applications of MOS in the food, health care, and other industries. Besides, we offer an overview for this saccharide from the perspective of prospect functional ingredient, which we feel lacks in the current literature. MOS could be expected to provide a novel promising substitute for functional oligosaccharides.

Abstract:This work aimed to produce mannooligosaccharide-rich carbohydrates from coconut meals for food application by using environmentally friendly processes. Coconut meal from coconut milk manufacturing was defatted by subcritical ethanol extraction at 100 °C before hydrolysis in subcritical water at different temperatures (120−230 °C), followed by ethanol precipitation. The obtained ethanol-insoluble precipitates contained mainly carbohydrates (89.09–98.88%) with small amounts of protein (0–5.33%) and 5-hydroxymethyl-2-furaldehyde (0–13.16 mg/100 g). The molecular weights of the final product were in the range of 0.83–34.70 kDa. The highest yield of the final product was from the treatment at 220 °C (10.76 g/100 g defatted coconut meal), while the treatment at 230 °C possessed the highest content of mannooligosaccharides with a degree of polymerization of 2–6 (46.12%). The obtained carbohydrates showed antioxidant capacities and enhanced in vitro growth of lactobacilli and bifidobacteria. Acetic and propionic acids were the major short-chain fatty acids formed by fermentation.

Abstract:Bacillus subtilis strain AP-1, which produces α-glucosidase with transglucosidase activity, was used to produce a series of long-chain isomaltooligosaccharides (IMOs) with degree of polymerization (DP) ranging from 2 to 14 by direct fermentation of maltose. A total IMOs yield of 36.33 g/L without contabacillusmination from glucose and maltose was achieved at 36 h of cultivation using 50 g/L of maltose, with a yield of 72.7%. IMOs were purified by size exclusion chromatography with a Superdex 30 Increase column. The molecular mass and DP of IMOs were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/MS). Subsequently, linkages in produced oligosaccharides were verified by enzymatic hydrolysis with α-amylase and oligo-α-1,6-glucosidase. These IMOs showed prebiotic properties, namely tolerance to acidic conditions and digestive enzymes of the gastrointestinal tract, stimulation of probiotic bacteria growth to produce short-chain fatty acids and no stimulating effect on pathogenic bacteria growth. Moreover, these IMOs were not toxic to mammalian cells at up to 5 mg/mL, indicating their biocompatibility. Therefore, this research demonstrated a simple and economical method for producing IMOs with DP2-14 without additional operations; moreover, the excellent prebiotic properties of the IMOs offer great prospects for their application in functional foods.

Abstract:Commelina communis L., a functional food and herbal plant in Asia, has been used against obesity, diabetes, and infections for centuries. A growing body of studies has demonstrated that indigestible polysaccharides are significant in obesity management. However, the structures and bioactivities of homogeneous polysaccharides from C. communis remain unclear. This study presented the structural characterization, simulated digestion, and human gut Bacteroides proliferation promotion activity of a novel homogeneous polysaccharide (CCB-3) from C. communis. The results showed that CCB-3 was an arabinoglucuronoxylan, primarily composed of arabinose, galactose, xylose, glucuronic acid (GlcA), and 4-O-methyl GlcA with a molecular weight (Mw) of 58.8 kDa. Following a 6-hour exposure to simulated gastrointestinal fluid, the Mw of CCB-3 remained unchanged, revealing that CCB-3 was an indigestible polysaccharide. Notably, CCB-3 could promote the proliferation of B. thetaiotaomicron, B. ovatus, and B. cellulosilyticus and produce short-chain fatty acids (SCFAs) and 1,2-propanediol. These findings might shed light on the discovery of polysaccharide-based leading compounds from C. communis against obesity.

Abstract:This study investigated the biological activities, prebiotic potentials, modulating gut microbiota, and rheological properties of polysaccharides derived from date seeds via microwave-assisted deep eutectic solvent systems. Averaged molecular weight (246.5 kDa) and a monosaccharide profile (galacturonic acid: glucose: mannose: fructose: galactose), classifying MPS as a heteropolysaccharide. MPS, at concentrations of 125-1000 µg/mL, demonstrates increasing free radical scavenging activities (DPPH, ABTS, MC, SOD, SORS, and LO), potent antioxidant potential (FRAP: 51.2-538.3 µg/mL; TAC: 28.3-683.4 µg/mL; RP: 18.5-171.2 µg/mL), and dose-dependent antimicrobial activity against common foodborne pathogens. Partially-purified MPS exhibits inhibition against α-glucosidase (79.6 %), α-amylase (85.1 %), and ACE (68.4 %), along with 80 % and 46 % inhibition against Caco-2 and MCF-7 cancer cells, respectively. Results indicate that MPS fosters the growth of beneficial fecal microbiota, including Proteobacteria, Firmicutes, and Actinobacteria, supporting microbes responsible for major SCFAs (acetic, propionic, and butyric acids) production, such as Ruminococcus and Blautia.

Abstract:The purpose of this study was to identify and determine the physical and structural characterization of the water-soluble galactomannan extracted from endosperm splits of Chinese S. cannabina seeds. The Sesbania galactomannan (SP) was extracted and purified using a novel method with a high yield (40.3 ± 7.2%). The molecular structure of SP was determined by monosaccharide composition, FTIR and NMR spectroscopy. The structural data showed that SP was galactomannan which composed by a β-(1/4)-linked mannose backbone with galactopyranosyl residues attached through α-(1/6) linkages. The constant mannose/galactose (M/G) ratio and average molecular weight (Mw) of SP was 1.6:1 and 2.16 × 105 g/mol, respectively. The physical results revealed that SP had many branches on the backbone and existed as a random coil state in aqueous solution. SP was a good biopolymer which had smooth and clearer surface with homogeneous composition, and had some degree of crystallinity and prebiotic activity. As a consequence, SP could be a potential prebiotic and was expected to be suitable for applications in food, pharmaceutical, biomedical or cosmetic industries as a promising new biomaterial.

Abstract:Pectin recovered from mango peel biomass can be used as a potential source for pectic oligosaccharide hydrolysate with excellent probiotic growth-enhancing performance and prebiotic potentials. Consequently, the objectives of the current study were to optimise the enzyme hydrolysis treatment of mango peel pectin (MPP) and to evaluate the pectic oligosaccharide effects of Lactobacillus reuteri DSM 17938 and Bifidobacterium animalis TISTR 2195. Mango of "chok anan" variety was chosen due to its excessive volume of biomass in processing and high pectin content. The optimal treatment for mango peel pectic oligosaccharide (MPOS) valorisation was 24 h of fermentation with 0.3% (v/v) pectinase. This condition provided small oligosaccharides with the molecular weight of 643 Da that demonstrated the highest score of prebiotic activity for both of B. animalis TISTR 2195 (7.76) and L. reuteri DSM 17938 (6.87). The major sugar compositions of the oligosaccharide were fructose (24.41% (w/w)) and glucose (19.52% (w/w)). For the simulation of prebiotic fermentation, B. animalis TISTR 2195 showed higher proliferation in 4% (w/v) of MPOS supplemented (8.92 log CFU/mL) than that of L. reuteri (8.53 CFU/mL) at 72 h of the fermentation time. The main short chain fatty acids (SCFAs) derived from MPOS were acetic acid and propionic acid. The highest value of total SCFA was achieved from the 4% (w/v) MPOS supplementation for both of B. animalis (68.57 mM) and L. reuteri (69.15 mM). The result of this study therefore conclusively advises that MPOS is a novel pectic oligosaccharide resource providing the opportunity for the sustainable development approach through utilising by-products from the fruit industry.

Abstract:In this study, three yeast α-mannans (LZ-MPS, MC-MPS, and G-MPS) were extracted from different sources of Kluyveromyces marxianus. The total sugar content of the three α-mannans ranged from 91.13–97.10%, whereas no proteins were detected. A structural arrangement was proposed using ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, and one-dimensional and two-dimensional nuclear magnetic resonance. The main chain of the three yeast α-mannans was formed by a →6)-α-D-Manp-(1→ unit, which was slightly different from the repeating unit of the branch structure. The prebiotic potential of LZ-MPS, MC-MPS, and G-MPS was assessed using in vitro fermentation with pure and faecal cultures. The three yeast α-mannans could be utilised as substrates for the growth of Lactobacillus and Lactococcus strains. In addition, the three yeast α-mannans markedly regulated the intestinal microbiota composition by increasing the relative abundances of Bacteroides, Parabacteroides, and Phascolarctobacterium and decreasing the abundance of pathogenic bacteria.
