Abstract:Environmental pollution sources including waste or metal accumulation, industrial and agricultural activities can be dangerous. Also, contaminated organic waste (COW) with metals especially, copper ions (Cu2+), can cause toxicity to various ecosystem components, enhance the production of reactive oxygen species (ROS) and consequently cause oxidative stress. The biochemical effect of the COW was monitored by assessing the oxidative stress parameters (OSP) using hydrogen peroxides (H2O2), protein carbonyls (PC), lipid peroxides (LP), catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), 2,2 diphenyl-1-picrylhydrazyl (DPPH), total antioxidant ability (TAA); β-esterase (β-EST); and total amount of protein (TAP) levels on the organic waste (kitchen and vegetable wastes); Hermetia illucens larvae; and larval excreta collected from 7-day post-treated kitchen and vegetables wastes with (1:10; g:mL) distilled water (DW) or 100 mg/mL Cu2+. The OSP levels were significantly higher in the experimental samples from Cu2+ groups than in the control one. Besides that, the OSP levels of H. illucens larvae feed on vegetable waste was significantly higher than those feed on kitchen waste except for β-EST, PC, and TPA. The best, cheap and easy parameters of antioxidants to ensure the entomoremediation ability are total antioxidant capacity. Possible impacts of accumulated and Cu2+ contaminated organic waste on H. illucens larvae were discussed. Also, the ability of insects to produce more antioxidants than input or output sources was approved. The potential use of the OSP as a bioindicator method of the bioremediation ability of H. illucens was proposed.

Abstract:The increased human activities and the worldwide population growth are constantly increasing the production of solid wastes. Over the years, waste management has thus become a prominent issue for several companies and municipalities, and several engineering techniques have been developed over the years in order to convert wastes into other solid materials or fuels. Yet, several techniques are important contributors to environmental pollution, and biological-based solutions have thus become progressively very popular. In particular, insect-based conversion of organic wastes represent eco-friendly tools, and the growth and development of insect species such as the black soldier fly have been tested and improved for a large diversity of organic wastes. However, organic wastes, including food wastes, may contain several pollutants such as heavy metals and catechol which could affect the bioconversion efficiency by incurring physiological costs that would be undetectable at the organismal level, i.e. have null to little effects on the life cycle of Hermetia illucens. In this context, assessments of antioxidant capacities can provide a rapid and low-cost evaluation of the capability of insects to handle exposure to heavy metals and catechol. Here, we aimed at measuring the physiological responses of the black soldier fly H. illucens grown on food wastes (kitchen, fruit or vegetable wastes) contaminated by cadmium, iron, lead or catechol. Biomarkers of oxidative stress (concentrations of hydrogen peroxide and protein carbonyls), non-enzymatic total antioxidant capacity (ascorbic acid amounts) and activity of enzymatic antioxidants (activities of superoxide dismutase and polyphenoloxidase) were measured from the gut of the larvae. We found no evidence of deleterious impacts of food waste contamination by catechol or heavy metals on H. illucens. In most experimental treatments, the array of physiological endpoints we measured for evaluating the degree of oxidative stress experienced by the larvae remained similar to controls. Possible physiological effects were reported for cadmium and catechol only, which tended to increase the oxidation of proteins and hydrogen peroxide in the larvae. Finally, our results suggested that the nature of the food waste could equally affect the physiological responses of the insect.
Effects of pyrethroid and organophosphate insecticides on reared black soldier fly larvae (Hermetia illucens)

Abstract:Black soldier fly larvae (Hermetia illucens) are increasingly considered a valuable alternative protein source for animal feed, but exposure to insecticide residues in the rearing substrate may affect performance. Effects of three pyrethroid (cypermethrin, deltamethrin, permethrin) and three organophosphates (pirimiphos‐methyl, chlorpyrifos‐methyl, malathion) were determined. Results suggest significant reductions in yield and survival when exposed to concentrations equal to or below applicable legal limits. Black soldier fly larvae (Hermetia illucens) receive growing interest as a potential alternative animal feed source. These insects may be exposed to insecticide residues in the rearing substrate. This study aimed to investigate the effects of six different pyrethroid and organophosphate insecticides on this insect species' performance. The toxicity of two "model" substances for each of these classes (cypermethrin; pirimiphos‐methyl) was quantified, with and without the synergist piperonyl butoxide (PBO). Critical effect doses corresponding to 10% yield (CED10) for cypermethrin (0.4 mg/kg) and pirimiphos‐methyl (4.8 mg/kg) were determined. The addition of PBO to cypermethrin enhanced its relative potency with a factor 2.6. These data were compared against the relative toxicity of two analogue substances in each class (permethrin, deltamethrin; chlorpyrifos‐methyl, malathion). Results suggest that exposure to concentrations complying with legal limits can cause significant reductions in yield. Exposure to multiple substances at lower concentrations resulted in negative additive and synergistic effects. Of the tested substances, deltamethrin was most toxic, causing 94% yield at 0.5 mg/kg. Analytical results suggested that transfer of tested substances to the larval biomass was substance‐ and concentration‐specific, but appeared to be correlated to reduced yields and the presence of PBO. Transfer of organophosphates was overall low (<2%), but ranged from 8% to 75% for pyrethroids. Due to very low limits in insect biomass (∼0.01 mg/kg), high transfer may result in noncompliance. It is recommended that rearing companies implement lower contractual thresholds, and that policymakers consider adjusting legally allowed maximum residue levels in insect feed.

Abstract:Black soldier fly (BSF), Hermetia illucens (L.) (Diptera: Stratiomyidae), is predominantly reared on organic wastes and other unused complementary substrates. However, BSF may have a buildup of undesired substances in their body. The contamination of undesired substance, e.g., heavy metals, mycotoxins, and pesticides, in BSF mainly occurred during the feeding process in the larval stage. Yet, the pattern of accumulated contaminants in the bodies of BSF larvae (BSFL) is varied distinctively depending on the diets as well as the contaminant types and concentrations. Heavy metals, including cadmium, copper, arsenic, and lead, were reported to have accumulated in BSFL. In most cases, the cadmium, arsenic, and lead concentration in BSFL exceeded the recommended standard for heavy metals occurring in feed and food. Following the results concerning the accumulation of the undesired substance in BSFL's body, they did not affect the biological parameters of BSFL, unless the amounts of heavy metals in their diets are highly exceeding their thresholds. Meanwhile, a study on the fate of pesticides and mycotoxins in BSFL indicates that no bioaccumulation was detected for any of the target substances. In addition, dioxins, PCBs, PAHs, and pharmaceuticals did not accumulate in BSFL in the few existing studies. However, future studies are needed to assess the long-term effects of the aforementioned undesired substances on the demographic traits of BSF and to develop appropriate waste management technology. Since the end products of BSFL that are contaminated pose a threat to both human and animal health, their nutrition and production process must be well managed to create end products with a low contamination level to achieve a closed food cycle of BSF as animal feed.

Abstract:Contamination of food products with mycotoxins such as aflatoxin B1 (AFB1) poses a severe risk to human health. Larvae of the black soldier fly (BSFL), Hermetia illucens (Diptera: Stratiomyidae), can successfully metabolize AFB1 without any negative consequences on their survival or growth. Organic waste streams contaminated with mycotoxins can be upcycled into protein-rich BSFL as an alternative feed for livestock and the left-over feed residue into nutrient-rich crop fertilizers. However, the underlying mechanisms that allow BSFL to metabolize AFB1 are unknown. In this study, five-day-old BSFL were fed with either a control or an AFB1-spiked (20 μg/kg) diet to elucidate the underlying mechanisms. Larval samples were collected at three timepoints (6 h, 24 h and 72 h) and subjected to RNA-Seq analysis to determine gene expression patterns. Provision of an AFB1-spiked diet resulted in an up-regulation of 357 and a down-regulation of 929 unique genes. Upregulated genes include multiple genes involved in AFB1 metabolism in other (insect) species. Downregulated genes were generally involved in the insects' growth, development, and immunity. BSFL possesses a diverse genetic arsenal that encodes for enzymes capable of metabolizing AFB1 without trade-offs on larval survival. In conclusion, the adverse impact of AFB1 exposure on immunity-related processes is observed in the transcriptomic response, and is indicative of a trade-off between detoxification and immune responses.

Abstract:Thiamethoxam, a second-generation neonicotinoid insecticide, was found to be toxic to nontarget aquatic organisms. The purpose of this study was to investigate the toxicity of thiamethoxam (0.30, 1.25, and 5.00 mg/L) on zebrafish (Danio rerio) livers at the 7th, 14th, 21st, and 28th days. The reactive oxygen species (ROS), superoxide dismutase (SOD), catalase (CAT), glutathione-s-transferase (GST), malondialdehyde (MDA) content, and DNA damage were used to evaluate the toxic effects of thiamethoxam on zebrafish. Compared to control groups, ROS levels were ascended in the exposure period; SOD and CAT activities were dramatically increased during early exposure and then inhibited. GST activity only increased on days 28. MDA content was slightly elevated on days 21 and 28. Additionally, a clear dose-response relationship was found for DNA damage. In conclusion, thiamethoxam could induce oxidative stress and DNA damage on the exposed zebrafish. (C) 2015 Wiley Periodicals, Inc.

Abstract:Black soldier fly (Hermetia illucens) larvae (BSFL) are a promising, sustainable source of nutrients, however, there is limited knowledge regarding the food safety of consuming BSFL. This study determined the safety of consuming BSFL for direct human consumption in terms of microbial, heavy metal and allergen content. Microbial counts were determined using ISO (International Organization for Standardization) methods, heavy metals were determined using inductively coupled plasma mass spectrometry and allergens were determined via Orbitrap mass spectrometry and ELISA (enzyme-linked immunosorbent assay) kits. Feed and killing method influenced the presence of Bacillus cereus (p = 0.011), and only the killing method influenced Escherichia coli (p < 0.00) and total viable count (TVC) (p < 0.00). Blanching resulted in a 3-log reduction in E. coli and a 3.4 log reduction in the TVC counts. Salmonella spp. and Listeria spp. were not detected in the BSFL samples. Heavy metals were detected although they were below maximum legal limits. Cross-reactive allergens, tropomyosin and arginine kinase, were detected in the BSFL samples, although the clinical significance requires research. The feed fed to the BSFL and blanching were found to influence the safety of consuming BSFL, highlighting the importance of incorporating sufficient decontamination steps, such as blanching, to ensure food safety.

Susceptibility and Biochemical Determination of House Fly (Musca domestica L.) to Lambda-cyhalothrin

Abstract:Background and Objective: The house fly, Musca domestica L., has the ability to develop resistance against pyrethroid insecticides in a short exposure time. This study aimed to screen the susceptibility of house fly to lambda-cyhalothrin and determine the activities of metabolic enzymes in insect. Materials and Methods: House fly populations collected from different locations in Sharkia Governorate, Egypt were used. Bioassay experiments were carried out on 3rd larval instars of each population. Activities of metabolic enzymes were determined. Results: Data of bioassay experiments indicated that population collected from Kafr Sakr is 10.6-fold resistant to lambda-cyhalothrin. Other populations collected from San El-Hagar, Abo Kabir, Abo Hammad and Zagazig showed 8, 9.8, 6.9 and 7.5 fold of resistance as compared on LC50 levels. Biochemical assays revealed that San El-Hagar and Kafr Sakr populations have the highest significant activity of esterase, while Zagazig population showed the highest activity in carboxylesterase recording 2.58 fold higher than laboratory strain followed by populations from Kafr Sakr and San El-Hagar. Results of mixed function oxidase were significantly higher in both Zagazig and Abo Hammad populations with 1.3 and 1.3 fold than what recorded in the laboratory strain. Kafr Sakr and Abo Hammad populations showed the highest significant GST activity recording 174.7 and 170 mmol min–1 g–1 b.wt., respectively. Conclusion: The obtained results showed that populations collected from different locations in Sharkia Governorate have low to moderate resistance to lambda-cyhalothrin with the possible role of carboxylesterase. The existing resistance is a warning to carefully use lambda-cyhalothrin in control programs of this pest in the specified locations.

Abstract:Musca domestica Linnaeus is a devastating insect pest of medical and veterinary importance with reports of resistance development to commonly used insecticides worldwide. Rearing substrates usually play a crucial role in determining susceptibility to insecticides and control of insect pests. The aim of the present study was to investigate the effect of five rearing substrates of M. domestica on its susceptibility to different insecticides and activities of metabolic enzymes. After 30 generations of rearing, susceptibility of M. domestica to tested insecticides, viz., malathion, pirimiphos-methyl, alpha-cypermethrin, deltamethrin, methomyl, propoxur, spinetoram, and chlorfenapyr had evident differences. Musca domestica reared on hen liver exhibited reduced susceptibility to all insecticides followed by the strain reared on poultry manure. However, M. domestica reared on milk-based diet showed the highest susceptibility to tested insecticides followed by the strain reared on manures of buffalo and horse. In addition, M. domestica reared on different substrates exhibited significant differences ( p < 0.01) in the activities of glutathione S -transferase (GST), cytochrome P450-dependent monooxygenase, and carboxylesterase (CarE). Overall, hen liver and poultry manure strains exhibited higher activities of metabolic enzymes than those of the milk-based diet, buffalo, and horse manure strains. In conclusion, the data of the present study exhibited a significant effect of rearing substrates on the susceptibility to insecticides and activities of metabolic enzymes in M. domestica . These results could be helpful for the sustainable management of M. domestica on different hosts by selecting appropriate insecticides.

Toxicological assessment of sodium benzoate in Drosophila melanogaster

Abstract:Sodium benzoate (SB), the sodium salt of benzoic acid, is a food preservative with wide applications in the food, cosmetic and pharmaceutical industries due to its ability to kill many microorganisms effectively. Experimental evidence however suggests that excessive intake of SB poses detrimental health risks among consumers in the population. The present study investigated the toxic effects of various concentrations of SB using Drosophila melanogaster as a model. Adult wild-type flies of Canton S strain (1- to 3-days old) was orally exposed to SB (0, 0.5, 1.0, 2.0 and 5.0 mg/5 g diet) to evaluate survival rates for 21 days. Thereafter, we evaluated markers of oxidative stress, antioxidant status and behavioral activity in D. melanogaster exposed to SB for seven (7) days. We observed that SB (2.0 and 5.0 mg/5 g diet) decreased the survival of D. melanogaster. Also, SB inhibited glutathione-S-transferase activity and depleted total thiols and nonprotein thiols contents. Moreover, SB (5 mg/5 g diet) increased nitric oxide (nitrite/nitrate) level and reduced flies' emergence rate. Conclusively, findings from this study revealed that exposure to high concentrations of SB reduced survival rate and induced toxicity via the induction of oxidative stress and inhibition of antioxidant enzymes in D. melanogaster.
Assessment of sublethal effects of permethrin on adult life characteristics and resistance dynamics in Aedes albopictus (Diptera: Culicidae)

Abstract:Mosquitoes are regularly exposed to adverse effects of insecticides employed in field during vector control campaigns. Its primary goal is to eliminate the vector population; nevertheless, this practise typically ignores the residual impacts and long-term repercussions on the remaining population. Here, the current study analysed how sublethal exposure of insecticides alter the life qualities, genotypic and biochemical characteristics of mosquitoes. The resistance ratio value in Laboratory Resistant (Lab-R) larvae increased 10 times (0.010 mg/l to 0.108 mg/l) compared to Laboratory Susceptible (LabS) larvae. It also revealed that the surviving mosquitoes had 50% reduction in hatchability but had longer larval and pupal periods (15 days and 2 days), respectively. The survival rates decrease in female by 2 days but increase in male by 7 days which is of concern and necessitates additional study. Moreover, major role of monooxygenase was confirmed behind resistance development which was further supported by piperonyl butoxide assay where reduction in Tolerance Ratio (TR50) by 12-fold occurred and gene expression profile also showed high expression level of CYP6P12 gene. In resistant strain, cuticular thickness increased by 1.23 times and alteration at codon 1532 (ATC to TTC) on VGSC gene leads to mutation I1532F. The data gleaned from our work highlights the threat of sublethal insecticides on vector control techniques and offers ample evidence that the larval selection alters adult life qualities, metabolic properties and transgenerational features which contributes to the damage caused by resistance.
Protective activity of Streptococcus faecalis against pathogenic action of Escherichia coli O55 in gnotobiotic pigs.

Abstract:Peroral administration of Streptococcus faecalis suppresses the lethal effect of simultaneous or subsequent infection with pathogenic Escherichia coli O55 in gnotobiotic colostrum-free piglets. The presence of S. faecalis in the intestine and mesenteric lymph nodes in artificially diassociated colostrum-free piglets influences neither multiplication of E. coli in the intestine nor its penetration into the organism.

Abstract:We have previously established and reported a novel monoclonal antibody (mAb), U5A2‐13, which recognizes a phenotypically similar population of natural killer (NK)‐like T cells. Using U5A2‐13 mAb, we now describe the functional properties of U5A2‐13+ T cells in both NK1.1‐positive or ‐negative mouse strains. Similar to NK1.1+ T cells, hepatic U5A2‐13+ T cells of C57BL/6 (NK1.1+ strain) mice, but not U5A2‐13– T cells, could be induced to produce large amounts of IL‐4 and IFN‐γ by stimulation with glycolipid α‐galactosylceramide (α‐GalCer) present on dendritic cells (DC) in a dose‐dependent manner. The abundant production of these cytokines from U5A2‐13+ T cells of BALB/c (NK1.1– strain) mice is similar to that noted in C57BL/6 mice. Cytokine production by cultures stimulated with DC of β2‐microglobulin‐deficient mice was significantly less than that of cultures stimulated with DC of intact mice. Overall, U5A2‐13+ T cells recognize α‐GalCer presented by CD1d, indicating that U5A2‐13+ T cells can be used to analyze NK‐like T cell function in various strains of mice.
Evaluation of oxidative stress biomarkers in Aiolopus thalassinus (Orthoptera: Acrididae) collected from areas polluted by the fertilizer industry

Abstract:The waste products of the fertilizer industry such as heavy metals, but especially phosphates and sulphates, are a serious problem that influences the structure and functioning of ecosystems. The levels of Cd, Pb, Zn, Cu, sulphates and phosphates were measured in soil samples from four sites: a control and sites that were 1, 3 and 6 km (sites A-C) away from the Abu-Zaabal Fertilizer Company. Oxidative stress markers (protein carbonyls, lipid peroxides), antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), polyphenoloxidase (PPO) and ascorbate peroxidase (APOX)) were evaluated in the tissues of Aiolopus thalassinus, collected from the corresponding sites. The highest concentrations of Cu and Zn were found in the soil from site A. The level of protein carbonyls in the brain, thoracic muscles and gut of the males and females from sites A, B and C were 11.82, 4.38, 5.97 (males) and 19.04, 16.65, 7.79 (females) times higher, respectively, compared to the individuals from the control site. Lipid peroxides levels in both sexes were significantly correlated with the distance from the source of the contamination. In the brain, thoracic muscles and gut of the males and females collected from site A, the level of lipid peroxides were 15.41, 23.49, 11.50 (males) and 25.36, 11.34, 15.37 (females) times higher compared to the values of the control animals. The activities of SOD, PPO, CAT and APOX were significantly affected by the environmental pollutants. The enzymatic and non-enzymatic oxidative markers in the Aiolopus thalassinus, a common insect species that inhabits various ecosystems, can also be used as a relevant biomarker of the pollution that is caused by the fertilizer industry.

Abstract:The house fly Musca domestica , the medical pest is a vector of many pathogenic organisms, that had showed insecticide resistance against popular active chemical ingredients, which represents a global issue that hinders successful insect control. This study aimed at investigating the effect of carbon quantum dot nanoparticles (CQDNPs) on some biochemical and cellular parameters in cells of the house fly larvae. In this study, serial concentrations of CQDNPs were tested against the house fly larvae. For each concentration, four replicates of third instar larvae of Musca domestica (twenty-five larvae for each one) were involved. Control groups were regarded by adding plain dechlorinated water to the larval medium. The LC 50 of CQDNPs on the house fly larvae was determined after 24 hours of incubation. Levels of total protein and albumin were inspected along with the levels of antioxidant enzymes regarding superoxide dismutase, catalase and glutathione reductase and enzymes of oxidative stress regarding lipid peroxidase and nitric oxide. Moreover, comet assay was performed in tissues of the treated larvae. Results revealed that the LC 50 (3.19 mg/ml) showed significant decrease in the total protein and albumin levels. Further, significant decrease in the activities of the antioxidant enzymes was implied along with significant increase of cellular damage enzymes in tissues of the treated larvae. Eventually, DNA damage was detected implying the larvicidal potency of carbon quantum dots against the house fly larvae. To conclude, CQDNPs exhibit insecticidal potency against the house fly larvae regarding cellular and molecular aspects.

Abstract:Organophosphorus pesticides form a diverse group of chemicals, having a wide range of physicochemical properties with crucial toxicological actions and endpoints. These are extensively used to control pests of different food (fruits, vegetables, tea, etc.) and non-food (tobacco, cotton, etc.) crops. Malathion is an important widely used organophosphorus pesticide but its hepatotoxic effects on fish are not well studied. Therefore, the current study was designed to investigate the hepatotoxic effects of Malathion on rohu (Labeo rohita) fish in a semi-static system using different parameters. The LC50 of Malathion was found to be 5 µg/L for rohu for 96 h through Probit analysis and was used for further toxicity testing. To find the hepatotoxic effects of Malathion, changes in different biochemical indices including protein contents, Lipid Peroxidation (LPO), activities of four protein metabolic enzymes [Aspartate Aminotransferase (AAT), Lactate Dehydrogenase (LDH), Alanine Aminotransferase (AlAT), and Glutamate Dehydrogenase (GDH)], seven antioxidant enzymes [Catalase (CAT), Superoxide Dismutase (SOD), Peroxidase (POD), Glutathione (GSH), Glutathione Reductase (GR), Glutathione-s-transferase (GST), and Glutathione Peroxidase (GSH-Px)], DNA damage [in term of comet tail length, tail moment, DNA percentage in tail, and olive tail moment], reactive oxygen species (ROS), and Histopathological alterations were assayed. Malathion exposure led to a time-reliant significant (P < 0.05) decrease in protein contents and a significant (P < 0.05) increase in ROS, LPO, enzymatic activities, and DNA damage. The histopathological examination of the liver showed different changes including hepatic necrosis, fatty infiltration, hemorrhage vacuolation, glycogen vacuolation, congestion, and cellular swelling. The current study clearly revealed Malathion as a potent hepatotoxic pesticide; therefore the injudicious, indiscriminate and extensive use of Malathion should be prohibited or at least reduced and strictly monitored.

Abstract:The black soldier fly (BSF), Hermetia illucens L. (Diptera: Stratiomyidae), has a great bioconversion potential and ability to develop on diverse substrates. Although the use of catering waste and food by-products containing meat and fish would reduce the footprint of the insect sector, to date, in Europe, their use is still facing legal obstacles for insects as food and feed. Since a major request from the EU insect sector is to diversify the spectrum of allowed substrates, and considering that variations in diet composition could influence insect immune responses, we evaluated the impact of different catering wastes on BSF prepupae immunity. Insects were reared on five diets: one based on feed for laying hens and four based on catering waste containing (i) fruits and vegetables; (ii) fruits, vegetables and bread; (iii) fruit, vegetables, bread and dairy products; (iv) fruits, vegetables, bread, meat and fish. The gene expression of two antimicrobial peptides (AMPs), one defensin and one cecropin, was assessed. Moreover, the hemolymph inhibitory activity against Escherichia coli DH5α and Micrococcus yunnanensis HI55 was evaluated using diffusion assays in solid media. The up-regulation of both AMPs' encoding genes was observed in insects fed a bread-added and dairy product-added diet. All hemolymph samples showed inhibitory activity against both bacteria, affecting the colony size and number. The obtained results show how catering waste positively influences the H. illucens immune system. The possibility of modulating AMP expression levels through the diet opens up new perspectives in the management of insect health in mass rearings.

Abstract:This study aimed to assess the effect of Hermetia illucens meal (HM) dietary inclusion on meagre oxidative status. Thus, fish were fed a fishmeal-based diet (CTR diet) and three other diets with increasing levels of HM inclusion, namely 10%, 20%, and 30% (diets HM10, HM20, and HM30, respectively). At the end of the trial, hepatic and intestine superoxide dismutase, catalase, and glucose-6-phosphate dehydrogenase activities and malondialdehyde concentration were unaffected by the diet composition. Liver glutathione peroxidase activity was higher in the fish fed the HM20 diet than in the fish fed the CTR and HM30 diets, and glutathione reductase activity linearly increased with the dietary HM level. The hepatic total glutathione and reduced glutathione contents were significantly lower in fish fed the HM20 diet than in fish fed the CTR and HM10 diets. In the intestine, the oxidized glutathione (GSSG) content and oxidative stress index linearly increased with the increase in dietary HM level, with the GSSG content of fish fed the HM20 diet being significantly higher than of fish fed the CTR diet. In conclusion, 30% HM might be included in meagre diets without negatively affecting hepatic and intestine oxidative status.

Abstract:Abstract Malathion, an extensively used organophosphorus pesticide, poses a high potential risk of toxicity to humans and the environment. Shewanella (S.) oneidensis MR-1 has been proposed as a strain with excellent bioremediation capabilities, capable of efficiently removing a wide range of hard-to-degrade pollutants. However, the physiological and biochemical response of S. oneidensis MR-1 to malathion is unknown. Therefore, this study aimed to examine how S. oneidensis MR-1 responds physiologically and biochemically to malathion while also investigating the biodegradation properties of the pesticide. The results showed that the 7-day degradation rates of S. oneidensis MR-1 were 84.1, 91.6, and 94.0% at malathion concentrations of 10, 20, and 30 mg/L, respectively. As the concentration of malathion increased, superoxide dismutase and catalase activities were inhibited, leading to a significant rise in malondialdehyde content. This outcome can be attributed to the excessive production of reactive oxygen species (ROS) triggered by malathion stress. In addition, ROS production stimulates the secretion of soluble polysaccharides, which alleviates oxidative stress caused by malathion. Malathion-induced oxidative damage further exacerbated the changes in the cellular properties of S. oneidensis MR-1. During the initial stages of degradation, the cell density and total intracellular protein increased significantly with increasing malathion exposure. This can be attributed to the remarkable resistance of S. oneidensis MR-1 to malathion. Based on scanning electron microscopy observations, continuous exposure to contaminants led to a reduction in biomass and protein content, resulting in reduced cell activity and ultimately leading to cell rupture. In addition, this was accompanied by a decrease in Na + /K + - ATPase and Ca 2+ /Mg 2+ -ATPase levels, suggesting that malathion-mediated oxidative stress interfered with energy metabolism in S. oneidensis MR-1. The findings of this study provide new insights into the environmental risks associated with organophosphorus pesticides, specifically malathion, and their potential for bioremediation. Abstract:The authors describe two patients with bilateral subdural effusion and cerebral displacement associated with spontaneous intracranial hypotension (SIH) and discuss the possible pathophysiological origins of these abnormalities. The signs seen on magnetic resonance imaging in both cases, such as tonsillar descent, subdural effusion, meningeal enhancement, downward displacement of the optic chiasm, and crowding of this structure and the hypothalamus between the pituitary gland and brain, can help to establish the diagnosis of SIH. Therapy with a lumbar epidural blood patch resulted in the rapid resolution of all symptoms and most morphological abnormalities. The authors propose diagnostic and management strategies based on their own experiences and the reported cases of SIH in the medical literature.
