Abstract:We have developed a multimodal photoacoustic remote sensing (PARS) microscope combined with swept source optical coherence tomography for in vivo, non-contact retinal imaging. Building on the proven strength of multiwavelength PARS imaging, the system is applied for estimating retinal oxygen saturation in the rat retina. The capability of the technology is demonstrated by imaging both microanatomy and the microvasculature of the retina in vivo. To our knowledge this is the first time a non-contact photoacoustic imaging technique is employed for in vivo oxygen saturation measurement in the retina.

Abstract:Early diagnosis of ocular diseases improves the understanding of pathophysiology and helps with accurate monitoring and effective treatment. Advanced multimodal ocular imaging platforms play a crucial role in the visualization of the ocular components and provide clinicians with a valuable tool for evaluating different eye diseases. Here, for the first time, we present a non-contact, multimodal photoacoustic remote sensing (PARS) microscopy and swept-source optical coherence tomography (SS-OCT) for in-vivo functional and structural imaging of the eye. The system provides complementary imaging contrasts of optical absorption and optical scattering and is used for non-contact, in-vivo imaging of the murine eye. Results of vasculature and structural imaging as well as melanin content in the retinal pigment epithelium (RPE) layer are presented. Multiwavelength PARS microscopy using Stimulated Raman Scattering (SRS) is applied for the first time, to provide non-contact oxygen saturation estimation in the ocular tissue. The reported work may be a major step toward clinical translation of ophthalmic technologies and has the potential to advance the diagnosis and treatment of ocular diseases.

Abstract:Knowledge of the prevalence rates and associated risk markers of vancomycin-resistant enterococci (VRE) colonization among long-term care facility (LTCF) residents could be used to improve screening policies among newly admitted hospital inpatients. In a cross-sectional survey among 1,215 residents of LTCFs in Jerusalem, the VRE carriage rate was 9.6%. Previous hospitalization and antibiotic treatment were associated with elevated VRE colonization rate. In contrast, moderate and severe levels of dependency and prolonged stay, in an LTCF were associated with a decrease in the VRE colonization rate.

Abstract:We present the first label-free, non-contact, in-vivo imaging of the ocular vasculature using photoacoustic remote sensing (PARS) microscopy. Both anterior and posterior segments mouse eye were imaged. Vasculature of iris, sclera and retina tissues were clearly resolved. To best of our knowledge this the first study showing non-contact photoacoustic imaging conducted on in-vivo ocular tissue. We believe that PARS microscopy has the potential to advance the diagnosis and treatment of ocular diseases.

Label-Free Non-Contact Structural and Functional Vascular Imaging using Photon Absorption Remote Sensing

Abstract:Vascular imaging is critical for understanding human health and disease. Most established non-contact and label-free optical techniques capture predominantly structural information about vasculature. However, in many pathologies, functional changes often precede visible morphological changes. This limits the ability of established modalities to prevent negative patient outcomes. In this study, a new in vivo Photon Absorption Remote Sensing (PARS) microscope is proposed. PARS enables the label-free non-contact structural and functional imaging of vascular structures and their microenvironment. PARS aims to capture the dominant light matter interactions surrounding an absorption event including both non-radiative and radiative relaxation. System performance is demonstrated through wide field of view in vivo imaging of vascular contrast in both mouse ear and chicken embryo. Additionally, video rate imaging of chicken embryo capillaries shows first feasibility for blood flow measurement using PARS. This work represents a promising step for vascular imaging techniques where there is significant demand for a method of non-contact label-free functional imaging.

Abstract:Photoacoustic imaging (PAI) is a noninvasive, nonionizing modality based on the differences in light absorption of various biological tissues. PAI utilizes the endogenous contrast characteristics of traditional optical imaging, while benefiting from high spatial resolution of the ultrasound imaging. A PAI system was developed to reconstruct the two-dimensional cross section image and to visualize the cerebrovascular activities of mouse in vivo. The spatial resolution of the PAI system was determined to be 0.110 mm by a two-point-source phantom with the Rayleigh criterion. The potential applications of the system were clearly demonstrated by successfully mapping a traumatic lesion in the mouse brain cerebral cortex, by its ability to monitor physiological changes in the brain due to carotid ligation and drug stimulation, and two-dimensional sliced images of a traumatic mouse brain at different depths were also provided. Our experimental results indicate that PAI has the potential for studying of traumatic brain injury and physiological functions of the brain.

Abstract:The goal of this study is to further develop a multi-modality eye imaging system and evaluate its feasibility of acquiring images of different modalities simultaneously. An integrated multimodality imaging system combining spectral-domain optical coherence tomography (SD-OCT), photoacoustic microscopy (PAM), and fluorescence microscopy (FM) was developed, and its performance for eye imaging was validated on multiple clinically-relevant retinal disease models in vivo in rabbits. OCT imaging allows for visualization of the different anatomic retinal layers with high axial resolution. PAM can be used to image vasculature, angiogenesis, and hemorrhages. The leakage of neovascularization can be verified with FM and fluorescein dye. Simultaneous imaging with OCT, PAM, and FM ensures co-registration of the three modalities without being affected by motion artifacts caused by breathing, body or eye movements, and heartbeat. This simultaneous multi-modality eye imaging system could be a new tool for applications both in ophthalmology and other fields.

Abstract:Photoacoustic microscopy (PAM) is an emerging imaging technology that can non-invasively visualize ocular structures in animal eyes. This report describes an integrated multimodality imaging system that combines PAM, optical coherence tomography (OCT), and fluorescence microscopy (FM) to evaluate angiogenesis in larger animal eyes. High-resolution in vivo imaging was performed in live rabbit eyes with vascular endothelial growth factor (VEGF)-induced retinal neovascularization (RNV). The results demonstrate that our multimodality imaging system can non-invasively visualize RNV in both albino and pigmented rabbits to determine retinal pathology using PAM and OCT and verify the leakage of neovascularization using FM and fluorescein dye. This work presents high-resolution visualization of angiogenesis in rabbits using a multimodality PAM, OCT, and FM system and may represent a major step toward the clinical translation of the technology.

Abstract:Abstract Histological visualizations are critical to clinical disease management and are fundamental to biological understanding. However, current approaches that rely on bright-field microscopy require extensive tissue preparation prior to imaging. These processes are both labor intensive and contribute to creating significant delays in clinical feedback for treatment decisions that can extend to 2–3 weeks for standard paraffin-embedded tissue preparation and interpretation, especially if ancillary testing is needed. Here, we present the first comprehensive study on the broad application of a novel label-free reflection-mode imaging modality known as photoacoustic remote sensing (PARS) for visualizing salient subcellular structures from various common histopathological tissue preparations and for use in unprocessed freshly resected tissues. The PARS modality permits non-contact visualizations of intrinsic endogenous optical absorption contrast to be extracted from thick and opaque biological targets with optical resolution. The technique was examined both as a rapid assessment tool that is capable of managing large samples (> 1 cm 2 ) in under 10 min, and as a high contrast imaging modality capable of extracting specific biological contrast to simulate conventional histological stains such as hematoxylin and eosin (H&E). The capabilities of the proposed method are demonstrated in a variety of human tissue preparations including formalin-fixed paraffin-embedded tissue blocks and unstained slides sectioned from these blocks, including normal and neoplastic human brain, and breast epithelium involved with breast cancer. Similarly, PARS images of human skin prepared by frozen section clearly demonstrated basal cell carcinoma and normal human skin tissue. Finally, we imaged unprocessed murine kidney and achieved histologically relevant subcellular morphology in fresh tissue. This represents a vital step towards an effective real-time clinical microscope that overcomes the limitations of standard histopathologic tissue preparations and enables real-time pathology assessment.

Abstract:Previously, we described a large Dutch family with hereditary vascular retinopathy (HVR), Raynaud's phenomenon and migraine. A locus for HVR was mapped on chromosome 3p21.1-p21.3, but the gene has not yet been identified. The fact that all three disorders share a vascular aetiology prompted us to study whether the HVR haplotype also contributed to Raynaud's phenomenon and migraine in this family. Whereas the parent-child transmission disequilibrium test (TDT) did not reach significance, the sibling TDT revealed that the HVR haplotype harbours a susceptibility factor for Raynaud's phenomenon and migraine. Identification of the HVR gene will improve the understanding of the pathophysiology of HVR, Raynaud's phenomenon and migraine.

Abstract:Background: As photoacoustic (PA) techniques progress towards clinical adoption, providing a high-speed live feedback becomes a high priority. To keep up with the instantaneous optical feedback of conventional light microscopes, PA imaging would need to provide a high-resolution video-rate live feed to the user. However, conventional PA microscopy typically trades resolution, sensitivity and imaging speed when optically scanning due to the difficult opto-acoustic confocal geometry. Here, we employ photoacoustic remote sensing (PARS), an all-optical technique that relies on optical confocal geometry, to provide a high-resolution live display in a reflection-mode PA architecture. Methods: Employing a conventional x-y galvanometer scanner and a 600 KHz pulse repetition rate laser we implement a system capable of acquiring 2.5 frames per second in 2D. To complement this fast scanning optical system, we implement a computationally inexpensive image reconstruction method that is able to render the frames with minimal overhead, providing a live display. Results: Employing the proposed method, we demonstrate a live feedback with frame rates as high as 2.5 Hz in 2D and also report the first results of 3D imaging with a non-contact label-free reflection-mode technique. The method is validated with phantom studies and in-vivo imaging. Employing a repetition rate of 600 KHz, a live feed of carbon fibers is realized with a C-scan rate of 2.5 Hz. The imaging resolution was measured to be 1.2 µm, the highest reported for a real-time reflection-mode architecture. The mean and peak SNR were measured to be 44 and 62 dB respectively in-vivo. 3D visualizations of carbon fiber phantoms and mouse ear microvasculature structure are also demonstrated. Conclusions: In summary, we present a method that has a small computational overhead for image rendering, resulting in a live display capable of real-time frame rates. We also report the first 3D imaging with a non-contact label-free reflection-mode PA technique. The all-optical confocal geometry required by PARS is significantly easier to implement and maintain than the opto-acoustic geometry of conventional PA microscopy techniques. This results in a system capable of high resolution and sensitivity, imaging at real-time rates. The authors believe this work represents a vital step towards a clinical high-resolution reflection-mode video-rate PA imaging system.

Abstract:Spectral-domain optical coherence tomography (SD-OCT) offers depth-resolved imaging of optical scattering contrast but is limited in sensitivity to optical absorption. Dual-modality imaging combined with the noncontact absorption contrast of photoacoustic remote sensing (PARS) microscopy can augment SD-OCT applications with specific molecular and functional contrasts in an all-optical, fiber-based platform. To develop a fiber-based multimodal PARS and SD-OCT imaging system, which efficiently uses a common 1050-nm light source for SD-OCT and PARS interrogation. PARS microscopy has predominantly utilized a 1310-nm interrogation light source to date. Hence, a recent dual-modality PARS and 1050-nm SD-OCT imaging system required three distinct wavelengths including a 532-nm PARS excitation, necessitating a free-space optical architecture with discrete subsystems. Here, we validate the first use of a 1050-nm interrogation wavelength for PARS. This enables the transition to fiber-based interferometry as is standard in modern SD-OCT systems, though infeasible with inclusion of an additional 1310-nm wavelength. PARS interrogation functionality is integrated using a broadband optical circulator. Dual-modality imaging is demonstrated in carbon fiber phantoms and a mouse ear in vivo. SD-OCT provided a 4.5-μm lateral resolution, 8.8-μm axial resolution in air, and >101 dB of sensitivity, and PARS contributed 532-nm optical absorption contrast with a 47-dB SNR, and lateral and axial resolutions of 2.4 and 35 μm, respectively. Total interrogation power was reduced from 90% to 58% of the ANSI limit compared to a previous three-wavelength approach. Adapting PARS to use the 1050-nm SD-OCT light source for interrogation enabled implementation of a fiber-based dual-modality system configuration, with image quality maintained. This will facilitate development of potential applications demanding handheld, catheter-based, or endoscopic form factors.

Abstract:Observation and characterization of any changes in anatomical structures of ocular components remain as a conventional technique for diagnosis, staging, therapeutic treatments, and post-treatment monitoring of any ophthalmic disorders. The existing technologies fail to provide imaging of all of the various components of the eye simultaneously at one scanning time, i.e., one can recover vital patho-physiological information (structure and bio-molecular content) of the different ocular tissue sections only one after another. This article addresses the longstanding technological challenge by use of an emerging imaging modality [photoacoustic imaging (PAI)] in which we integrated a synthetic aperture reconstruction technique (SAFT). Experimental results-with experiments being conducted in excised tissues (goat eye)-demonstrated that we can simultaneously image the entire structure of the eye (∼2.5 cm) depicting clearly the distinctive ocular structures (cornea, aqueous humor, iris, pupil, eye lens, vitreous humor, and retina). This study uniquely opens an avenue for promising ophthalmic (clinical) applications of high clinical impact.

Abstract:Optical-resolution photoacoustic microscopy (OR-PAM) is used for in vivo imaging of a variety of albino and pigmented eyes taking advantages of requiring no exogenous dye, performing high-resolution imaging, and achieving morphologic and functional imaging at the same time. However, to accurately diagnose the ophthalmic disease in the OR-PAM images, vascular enhancement algorithms are necessary for extracting vessels and quantifying them correctly. Vascular enhancement algorithms developed for other imaging technologies, are not suitable to be used for OR-PAM, because of the underlying differences in the physics of the formation of images. In this study, a new vascular enhancement algorithm called photoacoustic imaging vasculature enhancement filter (PAIVEF) is proposed, which not only enhances vasculature including micro-vessels signals, suppresses noise signals effectively, but also achieves highly sensitive and accurate enhancement of the vasculature within a large depth range in and out of the system's depth of focus (DOF). Using the PAIVEF, the morphologic and functional 3D images of the whole rat's ocular anterior vasculature segment was displayed simultaneously for a depth range of ∼0.6 mm, which was ∼7 times of the system's DOF. This study paves the way for the application of OR-PAM technology in ophthalmic disease research.

Abstract:Significance: The development of ultralow energy photoacoustic microscopy (PAM) on the clinically relevant pigmented rabbit eye model paves a road toward translation of the emerging PAM technology in ophthalmology clinics. Aim: Since the eye is particularly vulnerable to laser damage, we aim to develop an ultralow energy PAM system to significantly improve the laser safety of PAM by increasing the sensitivity of the system and reducing the incident laser energy for imaging. Approach: A multichannel data acquisition circuit with two-stage signal amplification was specially designed, which, in combination with the application of 3 by 3 median filter in the spatial domain, significantly improved the signal-to-noise ratio of the PAM system. The safety of this system was validated by histopathology, fluorescein angiography, and fundus photography. Results: Experiments performed on pigmented rabbits demonstrated that, when using this ultralow energy PAM system, satisfactory image quality can be achieved in the eye with an incident laser fluence that is only 1% of the American National Standards Institute safety limit. Fundus photography, fluorescein angiography, and histopathology were performed after the imaging procedure, and no retinal or ocular damage was observed. Conclusions: The proposed ultralow energy PAM system has excellent safety and holds potential to be developed into a clinical tool for ocular imaging.

Abstract:In this paper, we design a multimodal visible/near-infrared photoacoustic microscopy and optical coherence tomography (VIS/NIR-PAM-OCT) system for imaging both retina and retinal pigment epithelium (RPE)/choroid complex layer. F127 and DSPE-PEG-cRGD encapsulated IR-1048 nanoparticles (FINPs) exhibiting absorption peak up to 1,064 nm were utilized as contrast agents to enhance NIR-PAM for in vivo imaging of fundus tissues. The fundus structure and vessels are clearly visualized by the multimodal imaging, and their parameters were quantitatively analyzed. NIR-PAM and OCT imaging of fundus were time-serially monitored over 60 min following the intravenous injection of FINPs into rats. The results indicated a 134 % increase in image signals in PAM at 1 min, along with an 8.23 % intensity enhancement in OCT. Moreover, laser-induced choroidal neovascularization (CNV) was specifically detected and accurately quantified using VIS/NIR-PAM-OCT. Lastly, FINPs demonstrated excellent biocompatibility in hematology analysis and pathology testing.

Abstract:Although optical absorption is strongly associated with the physiological status of biological tissue, existing high-resolution optical imaging modalities, including confocal microscopy1,2, two-photon microscopy3,4 and optical coherence tomography5, do not sense optical absorption directly. Furthermore, optical scattering prevents these methods from imaging deeper than ∼1 mm below the tissue surface. Here we report functional photoacoustic microscopy (fPAM), which provides multiwavelength imaging of optical absorption and permits high spatial resolution beyond this depth limit with a ratio of maximum imaging depth to depth resolution greater than 100. Reflection mode, rather than orthogonal or transmission mode, is adopted because it is applicable to more anatomical sites than the others. fPAM is demonstrated with in vivo imaging of angiogenesis, melanoma, hemoglobin oxygen saturation (sO2) of single vessels in animals and total hemoglobin concentration in humans.

Abstract:Photoacoustic Remote Sensing (PARS) microscopy is an emerging label-free optical absorption imaging modality. PARS operates by capturing nanosecond-scale optical perturbations generated by photoacoustic pressures. These time-domain (TD) modulations are usually projected by amplitude to determine absorption magnitude. However, significant information on the target's material properties is contained within the TD signals. This work proposes a novel clustering method to learn TD features which relate to underlying biomolecule characteristics. This technique identifies features related to constituent biomolecules, enabling single-acquisition virtual tissue labelling. Colorized visualizations of tissue are produced, highlighting specific tissue components. This is demonstrated on freshly resected murine brain tissue, clearly discerning structures including myelinated and unmyelinated neurons (white and gray matter) and nuclear structures.
