Abstract:l -Tyrosine serves as a common precursor for multiple valuable secondary metabolites. Synthesis of this aromatic amino acid in Bacillus licheniformis occurs via the shikimate pathway, but the underlying mechanisms involving metabolic regulation remain unclear. In this work, improved l -tyrosine accumulation was achieved in B. licheniformis via co-overexpression of aroG fbr and tyrA fbr from Escherichia coli to yield strain 45A12, and the l -tyrosine titer increased to 1005 mg/L with controlled glucose feeding. Quantitative RT-PCR results indicated that aroA , encoding DAHP synthase, and aroK , encoding shikimate kinase, were feedback-repressed by the end product l -tyrosine in the modified strain. Therefore, the native aroK was first expressed with multiple copies to yield strain 45A13, which could accumulate 1201 mg/L l -tyrosine. Compared with strain 45A12, the expression of aroB and aroF in strain 45A13 was upregulated by 21% and 27%, respectively, which may also have resulted in the improvement of l -tyrosine production. Furthermore, supplementation with 5 g/L shikimate enhanced the l -tyrosine titers of 45A12 and 45A13 by 29.1% and 24.0%, respectively. However, the yield of l -tyrosine per unit of shikimate decreased from 0.365 to 0.198 mol/mol after aroK overexpression in strain 45A12, which suggested that the gene product was also involved in uncharacterized pathways. This study provides a good starting point for further modification to achieve industrial-scale production of l -tyrosine using B. licheniformis , a generally recognized as safe workhorse.
Combinatorial metabolic engineering of industrial Gluconobacter oxydans DSM2343 for boosting 5-keto-D-gluconic acid accumulation

Abstract:Background L-(+)-tartaric acid (L-TA) is an important organic acid, which is produced from the cream of tartar or stereospecific hydrolysis of the cis -epoxysuccinate. The former method is limited by the availability of raw material and the latter is dependent on the petrochemical material. Thus, new processes for the economical preparation of L-TA from carbohydrate or renewable resource would be much more attractive. Production of 5-keto-D-gluconate (5-KGA) from glucose by Gluconobacter oxydans is the first step to produce L-TA. The aim of this work is to enhance 5-KGA accumulation using combinatorial metabolic engineering strategies in G. oxydans . The sldAB gene, encoding sorbitol dehydrogenase, was overexpressed in an industrial strain G. oxydans ZJU2 under a carefully selected promoter, P 0169 . To enhance the efficiency of the oxidation by sldAB , the coenzyme pyrroloquinoline quinone (PQQ) and respiratory chain were engineered. Besides, the role in sldAB overexpression, coenzyme and respiratory chain engineering and their subsequent effects on 5-KGA production were investigated. Results An efficient, stable recombinant strain was constructed, whereas the 5-KGA production could be enhanced. By self-overexpressing the sldAB gene in G. oxydans ZJU2 under the constitutive promoter P 0169 , the resulting strain, G. oxydans ZJU3, produced 122.48 ± 0.41 g/L of 5-KGA. Furthermore, through the coenzyme and respiratory chain engineering, the titer and productivity of 5-KGA reached 144.52 ± 2.94 g/L and 2.26 g/(L · h), respectively, in a 15 L fermenter. It could be further improved the 5-KGA titer by 12.10 % through the fed-batch fermentation under the pH shift and dissolved oxygen tension (DOT) control condition, obtained 162 ± 2.12 g/L with the productivity of 2.53 g/(L · h) within 64 h. Conclusions The 5-KGA production could be significantly enhanced with the combinatorial metabolic engineering strategy in Gluconobacter strain, including sldAB overexpression, coenzyme and respiratory chain engineering. Fed-batch fermentation could further enlarge the positive effect and increase the 5-KGA production. All of these demonstrated that the robust recombinant strain can efficiently produce 5-KGA in larger scale to fulfill the industrial production of L-TA from 5-KGA.
Directed evolution and metabolic engineering generate an Escherichia coli cell factory for de novo production of 4-hydroxymandelate

Abstract:4-hydroxymandelate is a high-value aromatic compound used in the medicine, cosmetics, food, and chemical industry. However, existing natural extraction and chemical synthesis methods are costly and lead to environmental pollution. This study employed metabolic engineering and directed evolution strategies for de novo 4-hydroxymandelate biosynthesis. Two key challenges were addressed: insufficient precursor supply and limited activity of crucial enzymes. Through gene overexpression and multi-level gene interference using CRISPRi, An Escherichia coli chassis capable of producing the key precursor 4-hydroxyphenylpyruvate and the titer reached 5.05 mM (0.91 g/L). A mutant clone was obtained, HmaSV152G, which showed a 5.13-fold improvement in the catalytic rate. During fermentation, a high production of 194.87 mM (32.768 g/L) 4-hydroxymandelate was achieved in 76 h with a batch supply of glucose in a 5-L bioreactor. This study demonstrated the great potential of biosensors in protein engineering and provides a reference for large-scale production of other high-value aromatic compounds.
Improved Linalool Production in Saccharomyces Cerevisiae by Combining Directed Evolution of Linalool Synthase and Overexpression of the Complete Mevalonate Pathway

Abstract:With special fragrance and various biological properties, linalool has wide applications in food, pharmaceuticals and cosmetics industries. Microbial biosynthesis has become a promising approach to linalool production, however its efficiency is limited by the limited precursor supply and poor efficiency of linalool synthase. With the goal of enhancing heterologous linalool production in Saccharomyces cerevisiae, protein engineering and metabolic engineering were simultaneously employed to solve these bottlenecks. Linalool-producing yeast was first constructed by introduction of t670McLIS selected out from a collection of linalool synthases. The efficiency of linalool biosynthesis was then improved by strengthening the supply of geranyl pyrophosphate (GPP) as precursor via overexpression of the complete mevalonate (MVA) pathway and a GPP synthase variant. To accelerate the conversion of the accumulated GPP to linalool, t670McLIS was engineered by means of directed evolution after development of a competition-based color-indicated high-throughput screening method. Expression of the t670McLIS variant in the engineered S. cerevisiae with enhanced precursor supply yielded 53.14 mg/L of linalool in biphasic shake-flask culture. This study delivered an efficient linalool-producing yeast cell factory and meanwhile provided efficient strategies for biosynthesis of other valuable monoterpenes.
Enhancing Gastrodin Production in Yarrowia lipolytica by Metabolic Engineering

Abstract:Gastrodin, 4-hydroxybenzyl alcohol-4-O-β-D-glucopyranoside, has been widely used in the treatment of neurogenic and cardiovascular diseases. Currently, gastrodin biosynthesis has been achieved in model microorganisms. However, the production levels are insufficient for industrial applications. In this study, we successfully engineered a Yarrowia lipolytica strain to overproduce gastrodin through metabolic engineering. Initially, the engineered strain expressing the heterologous gastrodin biosynthetic pathway, which comprises chorismate lyase, carboxylic acid reductase, phosphopantetheinyl transferase, endogenous alcohol dehydrogenases, and a UDP-glucose dependent glucosyltransferase, produced 1.05 g/L of gastrodin from glucose in a shaking flask. Then, the production was further enhanced to 6.68 g/L with a productivity of 2.23 g/L/day by over-expressing the key node DAHP synthases of the shikimate pathway and alleviating the native tryptophan and phenylalanine biosynthetic pathways. Finally, the best strain, Gd07, produced 13.22 g/L of gastrodin in a 5-L fermenter. This represents the highest reported production of gastrodin in an engineered microorganism to date, marking the first successful de novo production of gastrodin using Y. lipolytica.

Metabolic engineering of Saccharomyces cerevisiae for high-level production of gastrodin from glucose

Abstract:Background: The natural phenolic glycoside gastrodin is the major bioactive ingredient in the well-known Chinese herb Tianma and is widely used as a neuroprotective medicine in the clinic. Microbial production from sustainable resources is a promising method to replace plant extraction and chemical synthesis which were currently used in industrial gastrodin production. Saccharomyces cerevisiae is considered as an attractive host to produce natural plant products used in the food and pharmaceutical fields. In this work, we intended to explore the potential of S. cerevisiae as the host for high-level production of gastrodin from glucose. Results: Here, we first identified the plant-derived glucosyltransferase AsUGT to convert 4-hydroxybenzyl alcohol to gastrodin with high catalytic efficiency in yeast. Then, we engineered de novo production of gastrodin by overexpressing codon-optimized AsUGTsyn, the carboxylic acid reductase gene CARsyn from Nocardia species, the phosphopantetheinyl transferase gene PPTcg-1syn from Corynebacterium glutamicum, the chorismate pyruvate-lyase gene UbiCsyn from Escherichia coli, and the mutant ARO4K229L. Finally, we achieved an improved product titer by a chromosomal multiple-copy integration strategy and enhancement of metabolic flux toward the aglycon 4-hydroxybenzyl alcohol. The best optimized strain produced 2.1 g/L gastrodin in mineral medium with glucose as the sole carbon source by flask fermentation, which was 175 times higher than that of the original gastrodin-producing strain. Conclusions: The de novo high-level production of gastrodin was first achieved. Instead of chemical synthesis or plants extraction, our work provides an alternative strategy for the industrial production of gastrodin by microbial fermentation from a sustainable resource.
Biomechanical and histomorphometric characterizations of osseointegration during mini-screw healing in rabbit tibiae.

Abstract:OBJECTIVE To evaluate effects of different healing times on integration of titanium mini-screws and bones under unloaded conditions. MATERIALS AND METHODS Biomechanical stability measurements and histomorphometric observations were performed in a rabbit tibia model after different healing times: 0 (immediate) and 1, 2, 4, and 8 weeks. RESULTS Biomechanical stability and both maximum torque and maximum pullout load increased with healing time but increased significantly only after 4 weeks. Maximum torque and maximum pullout load both significantly correlated with healing time and with each other. Similarly, histomorphometric analyses showed that new bone formation increased with healing time but increased dramatically only after 4 weeks. The data may provide guidance for determining optimal implant plans in clinical practice. CONCLUSION Mini-screw healing is a continuous process, and week 4 is a critical time point in this process.
Gastrodin Production from p -2-Hydroxybenzyl Alcohol Through Biotransformation by Cultured Cells of Aspergillus foetidus and Penicillium cyclopium

Abstract:The objective of this work was to take advantage of the resting cells of suitable fungus as an in vitro model to prepare gastrodin from p -2-hydroxybenzyl alcohol (HBA), which mainly exists in the metabolites of the plant Gastrodia elata Blume. The one-step biotransformation of HBA into gastrodin was examined with the filamentous fungi cells of Aspergillus foetidus and Penicillium cyclopium AS 3.4513 in this study. The fundamental conditions of biotransformation were screened and compared for both fungi. P. cyclopium AS 3.4513 had better gastrodin-producing capability than A. foetidus through one-step bioconversion. The highest yield of gastrodin was 36 mg/L for A. foetidus ZU-G1 and 65 mg/L for P. cyclopium AS 3.4513 under the respective development condition during 6 days of biotransformation. The comparative results show that P. cyclopium AS 3.4513 reveals great potential to form gastrodin using HBA as the precursor. The products catalyzed by the resting cells of P. cyclopium AS 3.4513 were identified through NMR and ESI-MS. Current results can be applied not only to the chemical synthesis processes that may involve the hydroxylation reaction but also to the industrial production. The selected fungus is the potential biocatalyst for HBA glucosylation.
Systematic metabolic engineering of Bacillus licheniformis for hyperproduction of the antioxidant hydroxytyrosol

Abstract:Hydroxytyrosol (HT) is a natural antioxidant that exhibits various health-related properties. In this work, we focused on establishing and optimizing the production of HT in Bacillus licheniformis . Through protein engineering ketoacid decarboxylase, boosting the phosphoenolpyruvate (PEP) supply, releasing feedback inhibition, and blocking competing pathways, HT production increased to 2078 mg L −1 from zero. Remarkably, the tyrosol catabolic pathway was identified and deletion of the alcohol dehydrogenase gene adhA significantly reduced tyrosol degradation. Next, multiple rate-liming factors were identified, including insufficiency of PEP, erythrose-4-phosphate and 4-hydroxyphenylpyruvate supply, and imbalance of KivD V461I and HpaBC expression levels. The carbon flux toward HT was significantly improved by modulating the glucose transport system, fine-tuning the expression levels of kivD V461I and hpaBC , and enhancing the expression of tyrA fbr , tkt , and zwf , which increased the HT titer to 6299 mg L −1 . Finally, the final strain DHT23 produced 9475 mg L −1 HT under fed-batch conditions, with a yield and productivity of 135.4 mg g −1 glucose and 395 mg (L h) −1 , respectively, which represents the highest HT production to date. Taken together, this study establishes B. licheniformis as a framework for HT hyperproduction and paves the way for achieving the industrial production of HT in the future.

Production of ρ-Hydroxyacetophenone by Engineered Escherichia coli Heterologously Expressing 1-(4-Hydroxyphenyl)-Ethanol Dehydrogenase

Abstract:ρ-Hydroxyacetophenone is an important and versatile compound that has been widely used in medicine, cosmetics, new materials, and other fields. At present, there are two ways to obtain ρ-hydroxyacetophenone. One is to extract it from plants, such as Artemisia capillaris Thunb and Cynanchum otophyllum Schneid, and the other is to synthesize it by using chemical methods. Of these two methods, the second is the main one, although it has problems, such as flammable and explosive reagents, difficult separation of by-products, and harsh reaction conditions. To solve these issues, we adopted genetic engineering in this study to construct engineered Escherichia coli containing Hped gene or EbA309 gene. Whole-cell biotransformation was conducted under the same conditions to select the engineered E. coli with the higher activity. Orthogonal tests were conducted to determine the optimal biotransformation condition of the engineered E. coli. The results showed that the optimal condition was as follows: substrate concentration of 40 mmol/l, IPTG concentration of 0.1 mmol/l, an induction temperature of 25°C, and a transformation temperature of 35°C. Under this condition, the effects of transformation time on the ρ-hydroxyacetophenone concentration and cell growth were further studied. We found that as the transformation time extended, the ρ-hydroxyacetophenone concentration showed a gradually increasing trend. However, when the ρ-hydroxyacetophenone concentration increased to 1583.19 ± 44.34 mg/l in 24 h, cell growth was inhibited and then entered a plateau. In this research, we realized the synthesis of ρ-hydroxyacetophenone by biotransformation, and our findings lay a preliminary foundation for further improving and developing this method.
Evaluation of excretory/secretory products of larval Taenia solium as diagnostic antigens for porcine and human cysticercosis

Abstract:Abstract Excretory/secretory antigens (ES) of larval Taenia solium were obtained by maintaining the bladder worms in Medium 199 for 3 days. Analsysis by SDS-PAGE showed that ES antigens consisted of at least 19 polypeptides, with Mrranging from 14–116 kDa. Analytical isoelectric focusing revealed eight bands with acidic pI. An immunocytolocalization study using the peroxidase method demonstrated the presence of ES epitopes on the tegument of the wall of the spiral canals of bladder worms. The specificity of ES antigens was evaluated by EITB, ELISA and FAST-ELISA using antisera against the common parasites of Chinese pigs and man. ES antigens cross-reacted with the antiserum against larval T. hydatigena of pigs. However, these antigens were generally more specific in diagnosing human cysticercosis. Three host-like molecules with molecular masses 43, 58 and 66 kDa were present in the ES products.
High-titer production of aromatic amines in metabolically engineered Escherichia coli

Abstract:ABSTRACT Aromatic amines are widely used in the pharmaceutical industry. Here, we reported the establishment of a bacterial platform for synthesizing three types of aromatic amines, namely, tyramine, dopamine, and phenylethylamine. Firstly, we expressed aromatic amino acid decarboxylase from Enterococcus faecium ( pheDC ) in an Escherichia coli strain with an increased shikimate (SHK) pathway flux toward L-tyrosine or L-phenylalanine synthesis. We found that glycerol served as a better carbon source than glucose, resulting in 940±46 mg/L tyramine from 4% glycerol. Next, the genes of lactate dehydrogenase ( ldhA ), formate acetyltransferase ( pflB ), phosphate acetyltransferase ( pta ), and alcohol dehydrogenase ( adhE ) were deleted to mitigate the fermentation byproduct formation. The tyramine level was further increased to 1.965±0.205 g/L in shake flasks, corresponding to 2.1 times improvement compared with that of the parental strain. By using a similar strategy, we also managed to produce 703±21 mg/L dopamine and 555±50 mg/L phenethylamine. In summary, we have demonstrated that the knockout of ldhA - pflB - pta - adhE is an effective strategy in improving aromatic amine productions, and achieved the highest aromatic amine titers in E. coli under shake flasks reported to date. Key points Aromatic amino acid decarboxylase from E. faecium was used for aromatic amine production; ldhA, pflB, pta together with adhE were deleted to mitigate the fermentation byproduct formation; Our work represented the best aromatic amine titers reported in E. coli under shake flasks.
Teaching effectiveness analysis plan applied to lectures in medical physiology.

Abstract:To improve and thus strengthen its teaching program, the Department of Physiology at the University of South Alabama voluntarily embarked on a multiphasic self-assessment of its medical teaching program. One phase of the greater assessment plan included an analysis of the teaching methods of each faculty member. To design and implement this phase, the services of a teaching consultant from the College of Education were obtained. The implementation of the objectives as established by the consultant resulted in 1) the development of a systematic and consistent method of evaluating the teaching practices of the faculty through the design of a standard observation instrument for use in analyzing the teaching of each individual; 2) the sampling of the teaching of each of the seven participating faculty members; 3) the collection and critical review of teaching materials used; 4) an analysis of the effectiveness of the faculty; and 5) the submission of a written report of evaluation results. The seven participating faculty members were observed during the delivery of two lectures each presented to the freshman medical class. Based on the analysis of both the lectures and materials used, a written critique of each faculty member was submitted. Lecture strengths and weaknesses, both of individual members and of the whole department were summarized. Finally, the results of a survey taken of the faculty in which the participants were asked to respond to a series of questions regarding the self-assessment program were most favorably accepted by all participating faculty.
Multilevel metabolic engineering of Bacillus licheniformis for de novo biosynthesis of 2-phenylethanol

Abstract:Due to its pleasant rose-like scent, 2-phenylethanol (2-PE) has been widely used in the fields of cosmetics and food. Microbial production of 2-PE offers a natural and sustainable production process. However, the current bioprocesses for de novo production of 2-PE suffer from low titer, yield, and productivity. In this work, a multilevel metabolic engineering strategy was employed for the high-level production of 2-PE. Firstly, the native alcohol dehydrogenase YugJ was identified and characterized for 2-PE production via genome mining and gene function analysis. Subsequently, the redirection of carbon flux into 2-PE biosynthesis by combining optimization of Ehrlich pathway, central metabolic pathway, and phenylpyruvate pathway enabled the production of 2-PE to a titer of 1.81 g/L. Specifically, AroK and AroD were identified as the rate-limiting enzymes of 2-PE production through transcription and metabolite analyses, and overexpression of aroK and aroD efficiently boosted 2-PE synthesis. The precursor competing pathways were blocked by eliminating byproduct formation pathways and modulating the glucose transport system. Under the optimal condition, the engineered strain PE23 produced 6.24 g/L of 2-PE with a yield and productivity of 0.14 g/g glucose and 0.13 g/L/h, respectively, using a complex medium in shake flasks. This work achieves the highest titer, yield, and productivity of 2-PE from glucose via the phenylpyruvate pathway. This study provides a promising platform that might be widely useful for improving the production of aromatic-derived chemicals.

Engineering of 4-hydroxyphenylacetate-3-hydroxylase from Escherichia coli for efficient biosynthesis of piceatannol

Abstract:Piceatannol, a catechol compound, exhibits various health benefits, with great application value in food, medicine, and cosmetics. However, the low natural abundance of piceatannol limits its cost-effective isolation, and chemical synthesis is difficult. Here, we report an efficient biocatalytic system for the production of piceatannol from resveratrol. Structure-based semi-rational engineering was used to generate HpaBCG209F, a novel HpaBC mutant of 4-hydroxyphenylacetate-3-hydroxylase from Escherichia coli. Biocatalytic activity assays revealed that HpaBCG209F exhibits 3.12 times higher activity toward resveratrol than wild-type HpaBC. We also introduced formic acid dehydrogenase (FDH) as an NADH-regeneration system for HpaBCG209F to support piceatannol production. Co-expression of HpaBCG209F and FDH in E. coli BL21(DE3) enabled the generation of 8.23 mM piceatannol from 10.2 mM resveratrol within 9.5 h under optimized conditions, representing an 80.6% molar conversion ratio. This study therefore successfully developed a highly active HpaBC variant for hydroxylating resveratrol to piceatannol, as well as demonstrated a promising biopathway for piceatannol production.

Abstract:Hydroxytyrosol, a valuable plant-derived phenolic compound, is increasingly produced from microbial fermentation. However, the promiscuity of the key enzyme HpaBC, the two-component flavin-dependent monooxygenase from Escherichia coli, often leads to low yields. To address this limitation, we developed a novel strategy utilizing microbial consortia catalysis for hydroxytyrosol production. We designed a biosynthetic pathway using tyrosine as the substrate and selected enzymes and overexpressing glutamate dehydrogenase GdhA to realize the cofactor cycling by coupling reactions catalyzed by the transaminase and the reductase. Additionally, the biosynthetic pathway was divided into two parts and performed by separate E. coli strains. Furthermore, we optimized the inoculation time, strain ratio, and pH to maximize the hydroxytyrosol yield. Glycerol and ascorbic acid were added to the co-culture, resulting in a 92% increase in hydroxytyrosol yield. Using this approach, the production of 9.2 mM hydroxytyrosol was achieved from 10 mM tyrosine. This study presents a practical approach for the microbial production of hydroxytyrosol that can be promoted to produce other value-added compounds.

Abstract:Major colonic haemorrhage remains a difficult diagnostic and therapeutic problem. We propose that those patients who continue to bleed after resuscitation are best served by immediate laparotomy. High flow antegrade irrigation and intra‐operative colonoscopy can then be used to localize the site of bleeding and allow appropriate excisional surgery rather than blind colonic resection.
Metabolic Engineering of Escherichia coli for High-Level Production of l-Phenylalanine

Abstract:l-Phenylalanine (l-Phe) is widely used in the food and pharmaceutical industries. However, the biosynthesis of l-Phe using Escherichia coli remains challenging due to its lower tolerance to high concentration of l-Phe. In this study, to efficiently synthesize l-Phe, the l-Phe biosynthetic pathway was reconstructed by expressing the heterologous genes aroK1, aroL1, and pheA1, along with the native genes aroA, aroC, and tyrB in the shikimate-producing strain E. coli SA09, resulting in the engineered strain E. coli PHE03. Subsequently, adaptive evolution was conducted on E. coli PHE03 to enhance its tolerance to high concentrations of l-Phe, resulting in the strain E. coli PHE04, which reduced the cell mortality to 36.2% after 48 h of fermentation. To elucidate the potential mechanisms, transcriptional profiling was conducted, revealing MarA, a DNA-binding transcriptional dual regulator, as playing a crucial role in enhancing cell membrane integrity and fluidity for improving cell tolerance to high concentrations of l-Phe. Finally, the titer, yield, and productivity of l-Phe with E. coli PHE05 overexpressing marA were increased to 80.48 g/L, 0.27 g/g glucose, and 1.68 g/L/h in a 5-L fed-batch fermentation, respectively.

Abstract:Hydroxytyrosol is an olive-derived phenolic compound of increasing commercial interest due to its health-promoting properties. In this study, a high-yield hydroxytyrosol-producing Saccharomyces cerevisiae cell factory was established via a comprehensive metabolic engineering scheme. First, de novo biosynthetic pathway of hydroxytyrosol was constructed in yeast by gene screening and overexpression of different phenol hydroxylases, among which paHD (from Pseudomonas aeruginosa) displayed the best catalytic performance. Next, hydroxytyrosol precursor supply was enhanced via a multimodular engineering approach: elimination of tyrosine feedback inhibition through genomic integration of aro4K229L and aro7G141S, construction of an aromatic aldehyde synthase (AAS)-based tyrosine metabolic pathway, and redistribution of metabolic flux between glycolytic pathway and pentose phosphate pathway (PPP) by introducing the exogenous gene Bbxfpkopt. As a result, the titer of hydroxytyrosol was improved by 6.88-fold. Finally, a glucose-responsive dynamic regulation system based on GAL80 deletion was implemented, resulting in the final hydroxytyrosol yields of 308.65 mg/L and 167.98 mg/g cell mass, the highest known from de novo production in S. cerevisiae to date.

Abstract:1,3-Butanediol (1,3-BDO) is an important C4 platform chemical widely used as a solvent in cosmetics and a key intermediate for the synthesis of fragrances, pheromones, and pharmaceuticals. The development of sustainable bioprocesses to produce enantiopure 1,3-BDO from renewable bioresources by fermentation is a promising alternative to conventional chemical routes and has aroused great interest in recent years. Although two metabolic pathways have been previously established for the biosynthesis of (R)-1,3-PDO, the reported titer and yield are too low for cost-competitive production. In this study, we report the combination of different metabolic engineering strategies to improve the production of (R)-1,3-BDO by Escherichia coli, including (1) screening of key pathway enzymes; (2) increasing NADPH supply by cofactor engineering; (3) optimization of fermentation conditions to divert more flux into 1,3-BDO pathway; (4) reduction of byproducts formation by pathway engineering. With these efforts, the best engineered E. coli strain can efficiently produce (R)-1,3-BDO with a yield of 0.6 mol/mol glucose, corresponding to 60% of the theoretical yield.
