Abstract:We developed a liquid crystal (LC) aptamer biosensor for the sensitive detection of amoxicillin (AMX). The AMX aptamer was immobilized onto the surface of a glass slide modified with a mixed self-assembled layer of dimethyloctadecyl [3-(trimethoxysilyl) propyl] ammonium chloride (DMOAP) and (3-aminopropyl) triethoxysilane (APTES). The long alkyl chains of DMOAP maintained the LC molecules in a homeotropic orientation and induced a dark optical appearance under a polarized light microscope (POM). In the presence of AMX, the specific binding of the aptamer and AMX molecules induced a conformational change in the aptamers, leading to the disruption of the homeotropic orientation of LCs, resulting in a bright optical appearance. The developed aptasensor showed high specificity and a low detection limit of 3.5 nM. Moreover, the potential application of the developed aptasensor for the detection of AMX in environmental samples was also demonstrated. Therefore, the proposed aptasensor is a promising platform for simple, rapid, and label-free monitoring of AMX in an actual water environment with high selectivity and sensitivity.

Abstract:O6-Alkylguanine-DNA alkyltransferase was rapidly and irreversibly inactivated by exposure to O6-benzylguanine or the p-chlorobenzyl and p-methylbenzyl analogues. This inactivation was much more rapid than with O6-methylguanine: incubation with 2.5 microM O6-benzylguanine led to more than a 90% loss of activity within 10 min, whereas 0.2 mM O6-methylguanine for 60 min was required for the same reduction. O6-Benzylguanine was highly effective in depleting the alkyltransferase activity of cultured human colon tumor (HT29) cells. Complete loss of activity was produced within 15 min after addition of O6-benzylguanine to the culture medium and a maximal effect was obtained with 5 microM. In contrast, at least 100 microM O6-methylguanine for 4 hr was needed to get a maximal effect, and this reduced the alkyltransferase by only 80%. Pretreatment of HT29 cells with 10 microM O6-benzylguanine for 2 hr led to a dramatic increase in the cytotoxicity produced by the chemotherapeutic agents 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea (CCNU) or 2-chloroethyl(methysulfonyl)methanesulfonate (Clomesone). Administration of O6-benzylguanine to mice at a dose of 10 mg/kg reduced alkyltransferase levels by more than 95% in both liver and kidney. These results indicate that depletion of the alkyltransferase by O6-benzylguanine may be used to investigate the role of the DNA repair protein in carcinogenesis and mutagenesis and that this treatment may be valuable to increase the chemotherapeutic effectiveness of chloroethylating agents.
Aptasensor for ampicillin using gold nanoparticle based dual fluorescence-colorimetric methods

Abstract:A gold nanoparticle based dual fluorescence-colorimetric method was developed as an aptasensor to detect ampicillin using its single-stranded DNA (ssDNA) aptamer, which was discovered by a magnetic bead-based SELEX technique. The selected aptamers, AMP4 (5'-CACGGCATGGTGGGCGTCGTG-3'), AMP17 (5'-GCGGGCGGTTGTATAGCGG-3'), and AMP18 (5'-TTAGTTGGGGTTCAGTTGG-3'), were confirmed to have high sensitivity and specificity to ampicillin (K(d), AMP7 = 9.4 nM, AMP17 = 13.4 nM, and AMP18 = 9.8 nM, respectively). The 5'-fluorescein amidite (FAM)-modified aptamer was used as a dual probe for observing fluorescence differences and color changes simultaneously. The lower limits of detection for this dual method were a 2 ng/mL by fluorescence and a 10 ng/mL by colorimetry for ampicillin in the milk as well as in distilled water. Because these detection limits were below the maximum residue limit of ampicillin, this aptasensor was sensitive enough to detect antibiotics in food products, such as milk and animal tissues. In addition, this dual aptasensor will be a more accurate method for antibiotics in food products as it concurrently uses two detection methods: fluorescence and colorimetry.
Selection of a new aptamer targeting amoxicillin for utilization in a label-free electrochemical biosensor

Abstract:Pharmaceutical pollution has received considerable attention because of the harmful effects of pharmaceutical compounds on human health, even in trace amounts. Amoxicillin is one of the frequently used antibiotics that was included in the list of emerging water pollutants. Therefore, a highly selective and rapid technique for amoxicillin detection is required. In this work, a new aptamer was selected for amoxicillin and utilized for the development of a label-free electrochemical aptasensor. Aptamer selection was performed using the systematic evolution of ligands by exponential enrichment. The selected aptamer showed good specificity against other antibiotics, including the structurally related antibiotics: ampicillin and ciprofloxacin. Among the selected aptamers, Amx3 exhibited the lowest dissociation constant value of 112.9 nM. An aptasensor was developed by immobilization of thiolated Amx3 aptamer onto gold screen–printed electrodes via self-assembly, which was characterized using cyclic voltammetry and electrochemical impedance spectroscopy. The detection was realized by monitoring the change in the differential pulse voltammetry peak current in the ferro/ferricyanide redox couple upon binding of the aptasensor to amoxicillin. The aptasensor showed very good sensitivity with an ultralow limit of detection of 0.097 nM. When the aptasensor was tested using actual spiked milk samples, excellent recovery percentages were observed. The label-free electrochemical aptasensor developed herein is a promising tool for the selective and sensitive detection of amoxicillin in environmental samples.

Abstract:Information-Centric Networking (ICN) is a research field that aims to optimize the Future Internet architecture for the distribution of information and content. Content distribution has emerged as a major use of the Internet infrastructure, explaining the relevance of ICN for the Future Internet. For successful deployment of ICN, its information-centric architecture needs to be supported by business models that provide proper economic incentives to network operators and end users. Although caching is a crucial function in ICN networking, we argue that it does not need to be part of ICN business models. Working from this premise, we identify a set of basic interconnection steps that support the growth, merge and interconnection of ICN islands in the IP Internet. The feasibility and ordering of the steps depend on the uptake of applications over ICN but also on further technical developments in ICN networking, such as ICN inter-domain routing.
Highly sensitive and selective detection of amoxicillin using molecularly imprinted ratiometric fluorescent nanosensor based on quantum dots

Abstract:A dual-emission ratiometric fluorescence sensor (CDs@CdTe@MIP) with a self-calibration function was successfully constructed for AMO detection. In the CDs@CdTe@MIP system, non-imprinted polymer-coated CDs and molecule-imprinted polymer-coated CdTe quantum dots were used as the reference signal and response elements, respectively. The added AMO quenched the fluorescence of the CdTe quantum dots, whereas the fluorescence intensity of the CDs remained almost unchanged. The AMO concentration was monitored using the fluorescence intensity ratio (log(I647/I465)0/(I647/I465)) to reduce interference from the testing environment. The sensor with a low detection limit of 0.15 μg/L enabled detection of the AMO concentration within 6 min. The ratiometric fluorescence sensor was used to detect AMO in spiked pork samples; it exhibited a high recovery efficiency and relative standard deviation (RSD) of 97.94-103.70% and 3.77-4.37%, respectively. The proposed highly sensitive and selective platform opens avenues for sensitive, reliable, and rapid determination of pharmaceuticals in the environment and food safety monitoring using ratiometric sensors.

Abstract:The molecularly imprinted electrochemical aptasensor was constructed based on co-deposition of zinc oxide and gold nanoparticles/reduced graphene oxide composite. Aptamer was used as a new kind of functional monomer and the aptamer-amoxicillin complex was formed by hydrogen bond. Then, the complex was fixed on the surface of the modified electrode by Au–S bond. Three-dimensional imprinted polymeric membrane was formed by electropolymerization of dopamine, and the imprinted sites with good specificity and affinity were formed after elution. Combined with the specificity of molecularly imprinted technology and the affinity of aptamer, the selective recognition of amoxicillin can be realized. Under the optimal experimental conditions, the linear range was from 10 −14 to 10 −8 M, and the detection limit was 3.3 × 10 −15 M. The sensor exhibited satisfactory selectivity, repeatability, and stability and was successfully used for 10 −9 M amoxicillin determination in real water and food samples.

Abstract:A sensitive signal-on photoelectrochemical aptasensor for antibiotic determination was constructed based on the energy level matching between ferrocene and CuInS2. P-type CuInS2 microflower was complexed with reduced graphene oxide (CuInS2/rGO) to get photocathode current with good photoelectric conversion efficiency and stability. Then, hairpin DNA (HP) was covalently bonded to the electrode surface. A triple helix DNA (THMS) was used as a molecular switch. After the specific recognition between target and THMS in homogeneous solution, ferrocene labeled probe (Fc-T2) was released. Finally, Fc-T2 was captured by the HP, which leaded the obvious increase of photocurrent for the energy level matching between ferrocene and CuInS2. The increase of the photocurrent signal was proportional to the concentration of target amoxicillin (AMOX), the linear range was 100 fM-100 nM with detection limit of 19.57 fM. Meanwhile, the method has been successfully applied for milk and lake water samples analysis with satisfactory results.
A reagentless and reusable electrochemical aptamer-based sensor for rapid detection of ampicillin in complex samples

Abstract:We report the design and fabrication of a "signal-on" electrochemical aptamer-based (E-AB) sensor for detection of ampicillin. The signaling of the sensor is based on target binding-induced changes in the conformation and flexibility of the methylene blue-modified aptamer probe. The sensor's response is fast; signal saturation can be reached in ~ 200s. Since all the sensor components are surface-immobilized, it is regenerable and can be reused for at least three times. It has demonstrated good specificity and is capable of differentiating between ampicillin and structurally similar antibiotics such as amoxicillin. More importantly, it is selective enough to be employed directly in complex samples, including serum, saliva, and milk. Although both alternating current voltammetry (ACV) and square wave voltammetry (SWV) are suitable sensor characterization techniques, our results show that ACV is better suited for target analysis. Even under the optimal experimental conditions, the limit of detection of the sensor obtained in ACV (1µM) is significantly lower than that obtained in SWV (30µM).

Abstract:To develop a sensitive and simple ampicillin (AMP) sensor for trace antibiotic residue detection, the influencing factors of the modification effect of nanogold-functionalized nucleic acid sequences (Adenine: A, Thymine: T) were comprehensively analyzed in this study, including the modification method, base length and type. It was found that under the same base concentration, longer chains are more likely to reach saturation than shorter chains; and when the base concentration and length are both the same, A exhibits a higher saturation modification level compared to T. Based on these research findings, a highly sensitive fluorescence aptamer sensor for detecting ampicillin was constructed using the optimized functionalized sequence (ployA6-aptamer) and experimental conditions (6 hours binding time between nucleic acid aptamer and complementary strand, pH 7 working solution, 20 minutes detection time) based on the principle of fluorescence resonance energy transfer. The sensor has a detection range of 0.18 ng ml −1 to 3.11 ng ml −1 for ampicillin, with a detection limit of 0.04 ng ml −1 . It exhibits significant selectivity and achieves an average recovery rate of 98.71% in tap water and 91.83% in milk. This method can be used not only for residual ampicillin detection, but also for highly sensitive detection of various antibiotics and small biological molecules by replacing the aptamer type. It provides a research basis for the design of highly sensitive fluorescence aptamer sensors and further applications of nanogold@DNA composite structures.

Abstract:The global threat of antibiotic resistance has increased the importance of the detection of antibiotics. Conventional methods to detect antibiotics are time-consuming and require expensive specialized equipment. Here, we present a simple and rapid biosensor for detecting ampicillin, a commonly used antibiotic. Our method is based on the fluorescent properties of chitosan-coated Mn-doped ZnS micromaterials combined with the β-lactamase enzyme. The biosensors exhibited the highest sensitivity in a linear working range of 13.1-72.2 pM with a limit of detection of 8.24 pM in deionized water. In addition, due to the biological specificity of β-lactamase, the proposed sensors have demonstrated high selectivity over penicillin, tetracycline, and glucose through the enhancing and quenching effects at wavelengths of 510 nm and 614 nm, respectively. These proposed sensors also showed promising results when tested in various matrices, including tap water, bottled water, and milk. Our work reports for the first time the cost-effective (Mn:ZnS)Chitosan micromaterial was used for ampicillin detection. The results will facilitate the monitoring of antibiotics in clinical and environmental contexts.

Abstract:The excessive utilization of antibiotics has led to significant water contamination and posed severe threats to human well-being. Consequently, the pressing imperative to identify antibiotics in the environment arises. In this study, we have successfully synthesized a hollow PCN-222 MOF distinguished by its substantial surface area and abundant functional groups, particularly the porphyrin cores. To augment the electrical conductivity of the hollow PCN-222 (HPCN-222), gold (Au) particles were incorporated within the porphyrin core using a fundamental hydrothermal method. This modification facilitated the effective immobilization of aptamer strands through π-π stacking and electrostatic interactions. As a result, the Au@HPCN-222 composite demonstrated exceptional efficacy as a substrate for immobilizing the aptamer (Apt) onto the GCE surface. By employing differential pulse voltammetry (DPV) we successfully achieved the detection of chloramphenicol (CAP) with a remarkably low limit of detection of 0.0138 ng mL −1 and the peak DPV currents at 0.18 V (vs. Ag/AgCl) were used for calibration. Furthermore, this aptasensor exhibited high selectivity and reproducibility.
Low-cost and highly sensitive colorimetric and visual detection of amikacin in milk using melamine functionalized gold nanoparticles

Abstract:This study developed a rapid, low-cost and highly sensitive analytical method using melamine functionalized gold nanoparticles (MA-Au NPs) as a colorimetric sensor to detect amikacin in milk. The MA-Au NPs were synthesized using a self-assembly process and analyzed using absorption spectroscopy, transmission electron microscopy (TEM), and Fourier transform infrared (FT-IR) spectroscopy. The MA-Au NPs exhibited a distinct surface plasmon resonance (SPR) absorption peak at 520 nm and demonstrated excellent spherical dispersion. The reaction parameters were optimized, with a Au NP concentration of 2.0 × 10 −7 mol L −1 , MA concentration of 1.0 × 10 −6 mol L −1 , pH of 4.0, and reaction time of 15 minutes. In the optimization experiment, the effects of unmodified Au NPs and MA-Au NPs as colorimetric sensors for amikacin determination were compared. As a specific amount of amikacin was added to the Au NP solution, the reaction solution showed a visible change from wine red to purple and then to blue gray; a red-shift in the absorption spectrum of the solutions was observed, which could also be analyzed using the A 640 / A 520 value, and the color changes were recorded using smartphones for visual detection through RGB (red–green–blue) data image analysis, which made the detection easy and fast. As a result, two approaches were devised using visible absorption spectra and smartphones. A linear correlation was established between the A 640 / A 520 value and the concentration of amikacin in the range of 4.0 to 16.0 × 10 −8 mol L −1 . The minimal limit of detection (LOD) for absorption spectrum analysis was 3.75 × 10 −9 mol L −1 , whereas for smartphone analysis, it was 6.0 × 10 −8 mol L −1 . The recoveries ranged from 103.09% to 104.51%, while the relative standard deviations (RSDs) ranged from 3.76% to 4.25%. The proposed method was used to detect actual milk samples. The results showed that the method is simple, selective, and practical.

Abstract:A colorimetric aptasensor was developed through spectroscopic analysis and LSPR properties of aptamer functionalized AuNRs. Presence of OTC resulted in AuNRs/aptamer assembly and led to different conjugation and spectroscopic changes. The applied detection approach showed efficient and sensitive results for detection of OTC. Oxytetracycline (OTC) as the frequently applied antibiotics present in human and animal diets and its residue may results to various toxic effects. So, there is a crucial need for developing sensitive method for detection of OTC. Aptamer‐based biosensors (aptasensors) are frequently used in recent applied detection strategies with high affinity and specificity. In the present study, a colorimetric aptasensor was developed for detection of oxytetracycline (OTC) based on aggregation of aptamer functionalized AuNRs. The synthesized AuNRs were characterized and functionalized with specific DNA oligonucleotide as the aptamer to generate an optical signal after binding to the OTC antibiotic. The sensitivity of optical changes in spectroscopic analysis was improved under ambient temperature that resulted in aptamer folded structure and subsequent aggregation state of AuNRs. The longitudinal surface plasmon resonance (LSPR) of AuNRs was decreased in the linear range of applied OTC concentration from 0.1 nM to 100 nM with a detection limit of 0.04 nM. The proposed method was facile and showed sensitive and selective results, that would be consider as an alternative and efficient approach toward rapid detection of OTC.

Abstract:A dual-emission ratio fluorescence probe (NL-CDs/Cu–Cy–I) was fabricated via an electrostatic self-assembly method. NL-CDs served as the reference signal ( E m = 433 nm), remaining insensitive to amoxicillin, while Cu–Cy–I acted as the response signal ( E m = 601 nm), exhibiting varying degrees of fluorescence quenching with amoxicillin concentration changes. This established a fluorescence sensing platform leveraging internal filtration and static quenching synergistically, ensuring high specificity. Compared to the single-signal Cu–Cy–I fluorescence probe, the detection range expanded from 0.5–15 μmol L −1 to 0.2–30 μmol L −1 , with the detection limit decreasing from 0.12 μmol L −1 to 0.073 μmol L −1 , demonstrating significant enhancements in both range and limit. The developed ratiometric fluorescent probe was effectively employed for amoxicillin monitoring in environmental water samples, urine samples, and amoxicillin capsules, achieving recovery rates of 96.5–102.9%. This underscores the accuracy and practical applicability of the NL-CDs/Cu–Cy–I ratio fluorescence probe-based fluorescence analysis method for amoxicillin detection.

Abstract:Antibiotic abuse is becoming increasingly serious and the potential for harm to human health and the environment has aroused widespread social concern. Aminoglycoside antibiotics (AGs) are broad-spectrum antibiotics that have been widely used in clinical and animal medicine. Consequently, their residues are commonly found in animal-derived food items and the environment. A simple, rapid, and sensitive detection method for on-site screening and detection of AGs is urgently required. In recent years, with the development of molecular detection technology, nucleic acid aptamers have been successfully used as recognition molecules for the identification and detection of AGs in food and the environment. These aptamers have high affinities, selectivities, and specificities, are inexpensive, and can be produced with small batch-to-batch differences. This paper reviews the applications of aptamers for AG detection in colorimetric, fluorescent, chemiluminescent, surface plasmon resonance, and electrochemical sensors for the analysis in food and environmental samples. This study provides useful references for future research.

Abstract:An electrochemical biosensor was proposed utilizing an improved amplification strategy for the rapid detection of sulfamethazine (as a model target) in aquatic environments. In this competitive immunoassay, cetyltrimethylammonium bromide-capped gold nanoparticles (CTAB-AuNPs) were used as a signal amplifier and electrode matrix and coated with an antigen-antibody (Cag-Ab(1)) specific binding system as a recognition unit for the target compound. In addition, silver nanoparticle labels were functionalized with dendritic fibrous nanosilica (DFNS@AgNPs) and decorated onto chitosan/single walled carbon nanohorn (CS/SWCNH)-modified glass carbon electrodes (GCEs), which improved the electron transfer rate and increased the surface area, enabling more coating antigens to be captured. Under acidic conditions, massive amounts of the Ag+ bound to the surface of the AuNPs dissolved, and consequently, formed Ag+@CTAB-AuNP complexes, which resulted in a distinctly improved peroxidase-like activity and enhanced current response. Furthermore, the destroyed Ab(1)-Ab(2)-DFNS conjugation greatly decreased the impedance, bringing about the amplification of the electrochemical signals. After optimization of the parameters, the proposed approach exhibited excellent performance, including good sensitivity (LOD, 0.0655 ng/mL) and satisfactory accuracy (recoveries, 79.02%-118.39%; CV, 3.18%-9.82%), which indicates the great potential of this strategy for the rapid detection of trace pollutants in the environments.

Abstract:Gold nanoparticles (AuNPs) were extensively employed for in-situ detection sulfadiazine (SDZ) residues, yet current synthesis methods suffer from complex procedures, reagent pollution of the environment, and low particle quality. This study presents a novel synthesis method using discarded longan seed extract as a reducing agent to synthesized high-quality AuNPs, and then can be used for in-situ SDZ detection. Response surface methodology (RSM) was employed to optimize synthesis parameters, which resulted in five optimal combinations that enhanced the flexibility of synthesis. These AuNPs, ranging in size from 18.26 nm to 33.8 nm with zeta potentials from - 29.5 mV to - 14.3 mV, were successfully loaded with functional groups from longan seed extract. In the detection of SDZ, the colorimetric aptasensor demonstrated excellent sensitivity and selectivity over other antibiotics with a limit of detection and quantification at 70.98 ng·mL-1 and 236.59 ng·mL-1 in the concentration range of 200-800 ng·mL-1. Recoveries of spiked SDZ samples ranged from 97.90% to 106.7%, with RSD values below 9.25%. Meanwhile, the aptasensor exhibited exceptional diagnostic efficacy (AUC: 0.976) compared to UV absorption methods in the ROC evaluation. In conclusion, this study highlights the potential of using AuNPs synthesized from longan seed extract coupled with aptamer technology as a straightforward detection method for SDZ in river water, offering promising applications in environmental monitoring.

Abstract:In this study, a gold nanoparticle-based colorimetric aptasensor for Staphylococcus aureus (S. aureus) using tyramine signal amplification (TSA) technology has been developed. First, the biotinylated aptamer specific for S. aureus was immobilized on the surface of the wells of the microtiter plate via biotin–avidin binding. Then, the target bacteria (S. aureus), biotinylated-aptamer–streptavidin-HRP conjugates, biotinylated tyramine, hydrogen peroxide and avidin–catalase were successively introduced into the wells of the microtiter plate. After that, the existing catalase consumed the hydrogen peroxide. Finally, the freshly prepared gold (III) chloride trihydrate was added, the color of the reaction production would be changed and the absorbance at 550nm could be measured with a plate reader. Under optimized conditions, there was a linear relationship between the absorbance at 550nm and the concentration of S. aureus over the range from 10 to 106cfumL−1 (with an R² of 0.9947). The limit of the developed method was determined to be 9cfumL−1.
Highly Specific and Rapid Colorimetric Detection of Tetracycline in Pills and Milk Based on Aptamer-Controlled Aggregation of Silver Nanoparticles

Abstract:Tetracycline (TET) residues exist in our lives and endanger our health. For rapid visual detection of tetracycline, we designed a TET colorimetric aptasensor based on aptamer (Apt) to control the aggregation of silver nanoparticles (AgNPs). Apt was adsorbed on the surface of AgNPs to protect the AgNPs from the influence of salt and maintained a dispersed state. The color of the solution was bright yellow. When TET was present, Apt desorbed from the surface of AgNPs and highly combined with TET to form a rigid structure. AgNPs without Apt protection aggregated under the induction of salt, and their color changed to pink. By comparing the three Apts of TET with different chain lengths, we found that the shortest Apt had the best effect on stabilizing AgNPs. The aptasensor constructed based on this mechanism was not only sensitive and fast. It is also highly specific and does not respond to substances similar to TET (chlortetracycline (CTC) and oxytetracycline (OTC)). Under the best conditions, the linear range was 40–480 nM, and the detection limit was 4.33 nM. This method is used in the detection of actual pills and milk samples, with high recovery rate and small relative error. This proves the potential of the aptasensor in food safety testing.
