Abstract:Atypical fibroxanthoma represents a low‐grade sarcoma that usually presents in the elderly population on sun‐damaged areas of the head and neck regions. This neoplasm is characterized on histology by atypical pleomorphic, epithelioid to spindled cells, arranged in a haphazard pattern, set within a background of solar elastosis. In this case report, we present a unique case of the rare variant of clear‐cell atypical fibroxanthoma arising in an unusual place, specifically the lower extremities.

Abstract:The ubiquitous occurrence of microplastics (MPs) in the marine environment is raising concern for interactions with marine organisms. These particles efficiently adsorb persistent organic pollutants from surrounding environment and, due to the small size, they are easily available for ingestion at all trophic levels. Once ingested, MPs can induce mechanical damage, sub-lethal effects, and various cellular responses, further modulated by possible release of adsorbed chemicals or additives. In this study, ecotoxicological effects of MPs and their interactions with benzo(a)pyrene (BaP), chosen as a model compound for polycyclic aromatic hydrocarbons (PAHs) were investigated in Mediterranean mussels, Mytilus galloprovincialis. Organisms were exposed for 4 weeks to 10 mg/L of low-density polyethylene (LDPE) microparticles (2.34 * 107 particles/L, size range 20–25 μm), both virgin and pre-contaminated with BaP (15 μg/g). Organisms were also exposed for comparison to BaP dosed alone at 150 ng/L, corresponding to the amount adsorbed on microplastics. Tissue localization of microplastics was histologically evaluated; chemical analyses and a wide battery of biomarkers covering molecular, biochemical and cellular levels allowed to evaluate BaP bioaccumulation, alterations of immune system, antioxidant defenses, onset of oxidative stress, peroxisomal proliferation, genotoxicity, and neurotoxicity. Obtained data were elaborated within a quantitative weight of evidence (WOE) model which, using weighted criteria, provided synthetic hazard indices, for both chemical and cellular results, before their integration in a combined index. Microplastics were localized in hemolymph, gills, and especially digestive tissues where a potential transfer of BaP from MPs was also observed. Significant alterations were measured on the immune system, while more limited effects occurred on the oxidative status, neurotoxicity, and genotoxicity, with a different susceptibility of analyzed pathways, depending on tissue, time, and typology of exposure. Molecular analyses confirmed the general lack of significant transcriptional variations of antioxidant and stress genes. The overall results suggest that microplastics induce a slight cellular toxicity under short-term (28 days) exposure conditions. However, modulation of immune responses, along with bioaccumulation of BaP, pose the still unexplored risk that these particles, under conditions of more chronic exposure (months to years) or interacting with other stressors, may provoke long-term, subtle effects on organisms' health status.

Abstract:Electrocardiograms (ECGs) are widely used by clinicians to identify the functional status of the heart. Thus, there is considerable interest in automated systems for real-time monitoring of arrhythmia. However, intra- and inter-patient variability as well as the computational limits of real-time monitoring poses significant challenges for practical implementations. The former requires that the classification model be adjusted continuously, and the latter requires a reduction in the number and types of ECG features, and thus, the computational burden, necessary to classify different arrhythmias. We propose the use of adaptive learning to automatically train the classifier on up-to-date ECG data, and employ adaptive feature selection to define unique feature subsets pertinent to different types of arrhythmia. Experimental results show that this hybrid technique outperforms conventional approaches and is therefore a promising new intelligent diagnostic tool.
Microplastics impair extracellular enzymatic activities and organic matter cycling in oligotrophic sandy marine sediments

Abstract:Microplastics (MPs) are ubiquitous and constantly accumulating in the marine environment, especially sediments. Yet, it is not well clarified if and how their carbon backbone could interact with surrounding sediments, eventually impairing key benthic processes. We assessed the effects of a 'pulse' contamination event of MPs on sedimentary organic matter (OM) quantity, quality and extracellular enzymatic activities (EEAs), which are well established descriptors of benthic ecosystem functioning. Marine sediments were exposed for 30 days to environmentally relevant concentrations (∼0.2 % in weight) of naturally weathered particles (size range 70-210 μm) of polyurethane, polyethylene, and a mixture of the most common polymers that are documented to accumulate in marine sediments. Despite the low concentration, contaminated sediments showed significantly different composition of OM, showing a decrease in lipid content and increase in protein. Moreover, we document a significant decrease (over 25 %) in quantity of biopolymeric C already after 15 days of exposure, compared to controls. Contaminated sediments showed lower C degradation rates (up to -40 %) and altered EEAs, with alkaline phosphatase being ∼50 % enhanced and aminopeptidase being reduced over 35 % compared to control treatments. Overall, the effects generated by the mixture of polymers were smaller than those exerted by the same amount of a single polymer. Our results provide insights on how that MPs can significantly alter marine sedimentary biogeochemistry through altered benthic processes, that could cumulatively impair whole benthic trophic webs by enhancing the accumulation and possible longer-term storage of recalcitrant organic C in the seabed.

Abstract:Microplastics (plastic particles < 5 mm; MPs) are ubiquitous in aquatic environments but their potential adverse ecological effects on biota remain poorly understood. This is in part because in typical ecotoxicology tests the toxic effects of MPs were found to be limited. To capture the potential find-scale effects of MPs on freshwater organisms, we employed ultra-performance liquid chromatography-tandem mass spectrometry based untargeted metabolomics to investigate the metabolic impact of polystyrene microbeads microplastics (PS-MPs) of different sizes (0.1, 1, 10, 100 μm) and concentrations (1, 10, 100 mg/L) on a common freshwater microalga, Microcystis aeruginosa , after a 96-h exposure test. The phenotype-based results illustrated that while PS-MPs had no discernible effects on microalgal growth and photosynthesis, both oxidative stress and microcystin production were slightly increased. Metabolomics analysis revealed that the PS-MPs altered the global metabolic profile of the microalga. Specially, PS-MPs of larger size and higher concentration induced a larger number of differentially expressed metabolites. The PS-MPs significantly disturbed metabolisms involved in amino acid synthesis, membrane formation, nitrogen storage, and antioxidant defense of the microalga, consistent with the phenotypic observations. These results suggested several perturbed metabolic pathways, especially arginine-related pathways, as the mechanism. Our study showed that the insights provided by metabolomics-based approaches can enhance assessments of the ecological impacts of MPs on freshwater organisms.

Abstract:Microplastic contamination is a growing marine environmental issue with possible consequences for seafood safety. Filter feeders are the target species for microplastic (MPs) pollution because they filter large quantities of seawater to feed. In the present study, an experimental contamination of Mytilus galloprovincialis was conducted using a mixture of the main types of MPs usually present in the seawater column (53% filaments, 30% fragments, 3% granules) in order to test the purification process as a potential method for removing these contaminants from bivalves intended for human consumption. A set of molecular biomarkers was also evaluated in order to detect any variations in the expression levels of some genes associated with biotransformation and detoxification, DNA repair, cellular response, and the immune system. Abstract:An untargeted study of multiclass contaminants associated with microplastics (MPs) in the East Mediterranean was carried out. Samples were collected in 2020-2021 from the shoreline at 14 different locations, along the Lebanese coast. Attenuated Total Reflectance (ATR) FTIR spectroscopy showed the predominant presence of polyethylene and polypropylene among plastic debris. The non-polar and polar organic compounds sorbed on the MPs were identified and quantified by GC - TOF MS and LC - electrospray MS/MS, respectively. Deconvolution of accurate GC-MS scan data allowed the identification of >130 organic pollutants, 64 of which could be confirmed by matching with the authentic standards, among which a number of previously unreported in targeted GC-MS(MS) methods. In addition to highly toxic, legacy chlorinated pollutants, high levels (average values from 0.8 to 4.0 μg g-1) of some musks, UV filters and UV absorbers were detected. Untargeted LC-MS demonstrated the persistence of several pesticides (i.e., chlorpyrifos) and pharmaceuticals, such as phenacetin and minoxidil, which were quantified. In addition, a study of metals associated with microplastics using ICP-MS confirmed the high potential of microplastics to act as a vector of, among others, toxic metals, such as Cd, Pb, Bi or Hg.

Abstract:To identify surface proteins of eimerian sporozoite, Eimeria tenella sporozoites were labeled with biotin-LC-hydrazide, and analyzed by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and Western blotting. More than 200 sporozoite proteins were recognized with silver staining after 2D-PAGE. Among them, 7 biotin-labeled proteins were detected as surface molecules after Western blotting and reaction with horseradish peroxidase conjugated streptavidin. Their molecular masses ranged from 23 to 117 kilo daltons and their isoelectric points from 4.3 to 6.3. It appears that the biotin-labeling technique can be used to analyze of surface proteins of eimerian parasites.

Abstract:<p>Marine microplastic has become an important environmental issue of global concern due to its wide distribution and harmful impacts. However, there is still insufficient information on the toxicity mechanism of microplastics to marine organisms. In this study, we developed and applied a high-coverage quantitative metabolomics technique to investigate the toxicity mechanisms of the polystyrene microspheres (micro-PS) on marine mussels (<em>Mytilus coruscus</em>). A total of 3599 metabolites were quantified, including 163 positively identified metabolites, 318 high-confident putatively identified metabolites, and 2602 mass-matched metabolites from the hemolymph of mussels. Metabolomics analysis indicated that micro-PS disrupted the amino acid metabolism, particularly phenylalanine metabolism, which may lead to oxidative stress and neurotoxicity. Micro-PS at environmentally relevant concentrations induced oxidative stress and immunotoxicity in mussels. After 7 days of recovery, along with the significant clearance of micro-PS by mussels, both metabolite levels and biochemical indicators generally returned to the same level as the control group. Overall, the results showed that microplastics at environmentally-relevant concentrations can cause toxic effects on mussels but these influences are reversible. We envisage the usages of high-coverage metabolomics for investigating the toxicity of various types of microplastics under many different conditions, including those relevant to the marine environment.</p>

Effects of polymethylmethacrylate nanoplastics on the polychaete Hediste diversicolor: Behavioural, regenerative, and biochemical responses

Abstract:Plastics, particularly microplastics (MPs) and nanoplastics (NPs), have been regarded as pollutants of emerging concern due to their effects on organisms and ecosystems, especially considering marine environments. However, in terms of NPs, there is still a knowledge gap regarding the effects of size and polymer on marine invertebrates, such as benthic organisms. Therefore, this study aimed to understand, regarding behavioural, physiological, and biochemical endpoints (neurotransmission, energy metabolism, antioxidant status, and oxidative damage), the effects of 50 nm waterborne polymethylmethacrylate (PMMA) NPs (0.5 to 500 µg/L) on the marine benthic polychaete Hediste diversicolor, a key species in estuarine and coastal ecosystems. Results demonstrated that worms exposed to PMMA NPs had a shorter burrowing time than control organisms. Nevertheless, worms exposed to PMMA NPs (0.5 and 500 µg/L) decreased cholinesterase activity. Energy metabolism was decreased at 50 and 500 µg/L, and glycogen content decreased at all concentrations of PMMA NPs. Enzymes related to the antioxidant defence system (superoxide dismutase and glutathione peroxidase) displayed increased activities in H. diversicolor specimens exposed to concentrations between 0.5 and 500 µg/L, which led to no damage at the cell membrane and protein levels. In this study, polychaetes also displayed a lower regenerative capacity when exposed to PMMA NPs. Overall, the data obtained in this study emphasize the potential consequences of PMMA NPs to benthic worms, particularly between 0.5 and 50 µg/L, with polychaetes exposed to 50 µg/L being the most impacted by the analysed NPs. However, since sediments are considered to be sinks and sources of plastics, further studies are needed to better understand the impacts of different sizes and polymers on marine organisms, particularly benthic species.
Unveiling microplastic's role in nitrogen cycling: Metagenomic insights from estuarine sediment microcosms

Abstract:Marine microplastics (MPs) pollution, with rivers as a major source, leads to MPs accumulation in estuarine sediments, which are also nitrogen cycling hotspots. However, the impact of MPs on nitrogen cycling in estuarine sediments has rarely been documented. In this study, we conducted microcosm experiment to investigate the effects of commonly encountered polyethylene (PE) and polystyrene (PS) MPs, with two MPs concentrations (0.3% and 3% wet sediment weight) based on environmental concentration considerations and dose-response effects, on sediment dissolved oxygen (DO) diffusion capacity and microbial communities using microelectrode system and metagenomic analysis respectively. The results indicated that high concentrations of PE-MPs inhibited DO diffusion during the mid-phase of the experiment, an effect that dissipated in the later stages. Metagenomic analysis revealed that MP treatments reduced the relative abundance of dominant microbial colonies in the sediments. The PCoA results demonstrated that MPs altered the microbial community structure, particularly evident under high concentration PE-MPs treatments. Functional analysis related to the nitrogen cycle suggested that PS-MPs promoted the nitrification, denitrification, and DNRA processes, but inhibited the ANRA process, while PE-MPs had an inhibitory effect on the nitrate reduction process and the ANRA process. Additionally, the high concentration of PE-MPs treatment significantly stimulated the abundance of genus (Bacillus) by 34.1% and genes (lip, pnbA) by 100-187.5% associated with plastic degradation, respectively. Overall, in terms of microbial community structure and the abundance of nitrogen cycling functional genes, PE- and PS- MPs exhibit both similarities and differences in their impact on nitrogen cycling. Our findings highlight the complexity of MP effects on nitrogen cycling in estuarine sediments and high concentrations of PE-MP stimulated plastic-degrading genus and genes.

Abstract:In a series of 148 patients with essential hypertension, the presence of the atherosclerotic C3-F gene and HLA-antigen frequences were investigated, and the results were compared with those in 62 age and sex matched normotensive controls. The frequency of C3-F was significantly higher in patients (p less than 0.03), as was HLA-B15 in patients with a positive family history of hypertension (p less than 0.05). The presence of the C3-F gene was associated with an increased risk (= 10.2) for coronary heart disease, and B15 was associated with an increased risk (= 6.0) for cerebral events in both familial and non familial cases of hypertension. It is suggested that determination of the C3-F gene and HLA-antigens might be a tool in the identification of hypertensive patients at particular high risk for vascular events, irrespective of BP levels. The study support the suggestions that some genetic factors may act via immunological pathways.

Abstract:To assess the potential effects of microplastics (MPs) on gut microbiome, a simple investigation of gut microbial structure is not sufficient, and the function and association of gutmicrobial structurewith host health should also be taken into account. Here, the effects of two particle sizes (2 and 200 mu m) of polystyreneMPs (PS-MPs) on the gut microbiota of medaka were evaluated following oral administration at 0.3 and 3.0 mu g/mg for 28 days. No change in body length and gut histopathology damage were observed. However, the exposure to PS-MPs significantly decreased fish body weight and disrupted the liver anti-oxidative status. The PS-MPs caused a shift in the gut microbial structure of medaka accompanied by changes in community function, including significant environmental stress, increased carbon degradation/fixation activities, and partially modified nitrogen/phosphorus/sulfur metabolic abilities. Furthermore, the PS-MPs exposure disturbed the glycolipid/tyrosine/energy metabolism and the endocrine balance. A potential correlation between the gutmicroecology and host response to PS-MPs exposure was also observed. These results indicated that the PS-MPs may contribute to gut-liver axis disruption, which could be the underlying toxicological mechanisms of PS-MPs exposure. Abstract:Microplastics (MPs) disturb the normal activity of aquatic organisms at different levels, causing physiological stress and altering feeding, growth, and reproduction. Alterations of epigenetic patterns due to exposure to MPs have scarcely been studied in invertebrates. In this study, Mytilus galloprovincialis mussels (N = 61) were intermittently exposed to different concentrations of pure polystyrene microbeads for three weeks. The concentrations used in this research were similar to those currently found in certain polluted environments (E1), as well as higher doses to which mussels could be further exposed (E2 and E3). After exposure period, the global methylation patterns were investigated using Amplified Fragment Length Polymorphism (AFLPs). Significantly lower methylation was found in exposed groups compared to the control group. The level of hypomethylation increased with the concentration of microbeads. Similar results were found from field samples inhabiting two sites differentially MPs-polluted. The implications of this discovery were analysed and discussed, noting the already known effects of MPs on metabolism and cell division. Further studies on this and other sentinel organisms are recommended to understand the response of the aquatic species to the currently increasing MPs pollution.

Evidence that microplastics at environmentally relevant concentration and size interfere with energy metabolism of microalgal community

Abstract:To address two current issues in evaluating the toxicity of microplastics (MPs) namely, conflicting results due to species specificity and the ecological irrelevance of laboratory data, this study conducted a 10-day exposure experiment using a microalgal community comprising three symbiotic species. The experiment involved virgin and Benzo[a]pyrene-spiked micron-scale fibers and fragments made of polyethylene terephthalate (PET) and polypropylene (PP). The results showed that, from a physiological perspective, environmentally relevant concentrations of micron-scale MPs decreased saccharide accumulation in microalgal cells, as confirmed by ultrastructural observations. MPs may increase cellular energy consumption by obstructing cellular motility, interfering with nutrient uptake, and causing sustained oxidative stress. Additionally, MPs and adsorbed B[a]P induced DNA damage in microalgae, potentially further disrupting cellular energy metabolism. Ecologically, MPs altered the species abundance in microalgal communities, suggesting they could weaken the ecological functions of these communities as producers and affect ecosystem diversity and stability. This study marks a significant advancement from traditional single-species toxicity experiments to community-level assessments, providing essential insights for ecological risk assessment of microplastics and guiding future mechanistic studies utilizing multi-omics analysis.

Abstract:Microplastics (MPs) and nanoplastics (NPs) have become an important global environmental issue due to their widespread contamination in the environment. This review summarizes existing literature on the effects of MPs/NPs on three important biomarkers including enzymatic activity, gene expression, and histopathology in various organisms from 2016 to 2021 and suggests a path forward for future research. Application of enzymatic activity, gene expression, and histopathology biomarkers are increasingly used in experimental toxicology studies of MPs/NPs because of their early signs of environmental stress to organisms. Between 2016 to 2021, 70% of published studies focused on aquatic organisms, compared to terrestrial organisms. Zebrafish were widely used as a model organism to study adverse impacts of MPs/NPs. Polystyrene (PS) were the most important polymer used in experimental toxicology studies of MPs/NPs. Fewer studies focused on the histopathological alterations compared to studies on enzymatic activity and gene expression of different organisms exposed to MPs/NPs. There is a growing need to better understand toxic effects of environmentally relevant concentrations of MPs/NPs on enzymatic activity, gene expression, and histopathology biomarkers of both aquatic and terrestrial organisms.

Assessing the physiological effects of microplastics on cultured mussels in the Mediterranean Sea

Abstract:Microplastics (MPs) pollution has gained attention due to its ecological threats and potential economic impacts. Yet significant knowledge gaps remain in understanding MPs effects on marine organisms' physiology. This study quantifies the physiological impacts of MPs on farmed mussels (Mytilus galloprovincialis) across various locations in the Mediterranean Sea by combining a laboratory experiment with a Dynamic Energy Budget (DEB) model. Mussels' clearance rates (CR) were measured under different conditions of microplastics and suspended sediment. The DEB model, driven by satellite data and an MPs distribution model, was validated with literature growth and CR data, supporting further the data extracted from the conducted experiment. Results indicate that while the physiological impacts are minimal in most areas, important reductions in CR (8-25%) were estimated in regions like the Gulf of Napoli, leading to reduced growth (6-16%) and reduced reproductive output (7-19%). In addition to microplastic concentrations, seasonal and spatial variations of food availability and suspended inorganic matter importantly control the impacts, with mussels in oligotrophic environments (such as the Gulf of Napoli) showing higher vulnerability to MPs compared to those in more eutrophic locations. This study underscores the utility of bioenergetics models, such as DEB, in evaluating the ecological risks of microplastics and suggests their broader application in MPs research.

Abstract:Background Concerns on microplastics (MPs) in food are increasing because of our increased awareness of daily exposure and our knowledge gap on their potential adverse health effects. When particles are ingested, macrophages play an important role in scavenging them, potentially leading to an unwanted immune response. To elucidate the adverse effects of MPs on human health, insights in the immunotoxicity of MPs are essential. Objectives To assess the effect of environmentally collected ocean and land weathered MP particles on the immunological response of macrophages using a state-of-the art in vitro immunotoxicity assay specifically designed for measuring particle toxicity. Methods Environmentally-weathered macroplastic samples were collected from the North Pacific Subtropical Gyre and from the French coastal environment. Macroplastics were identified using (micro)Raman-spectrometry, FT-IR and Py-GC-MS and cryo-milled to obtain size-fractionated samples up to 300 μm. Physiochemical MP properties were characterized using phase contrast microscopy, gel-permeation chromatography, nuclear magnetic resonance, and differential scanning colorimetry. Macrophages (differentiated THP-1 cells) were exposed to particles (<300 μm) for 48 h before assessment of cell viability and cytokine release. Using both the physiochemical particle properties and biological data, we performed multi-dimensional data analysis to explore relationships between particle properties and immunotoxicological effects. Results We investigated land-derived polyethylene, polypropylene, polystyrene, and polyethylene terephthalate, water-derived polypropylene macroplastics, and virgin polyethylene fibers and nylon MPs. The different plastic polymeric compositions and MP size classes induced distinct cytokine responses. Macrophages had the largest response to polyethylene terephthalate-particle exposure, including a dose-related increase in IL-1β, IL-8, and TNF-α secretion. Smaller MPs induced cytokine production at lower concentrations. Additionally, a relationship between both physical and chemical particle properties and the inflammatory response of macrophages was found. Discussion This research shows that MP exposure could lead to an inflammatory response in vitro , depending on MP material and size. Whether this implies a risk to human health needs to be further explored.
Multigenerational exposure of microplastics on the microbiota of E. affinis (copepod): a comparative study between biodegradable and nonbiodegradable microplastics

Abstract:The accumulation of plastic debris around the world, especially in marine environments, has been well documented during the past decades. Recent studies have found that inorganic surfaces of microplastics (MPs) can be used by microorganisms as living substrates and form an ecosystem named "plastisphere." Some microorganisms present in MPs are capable of producing polymer-degrading enzymes. In addition, MPs can also serve as vectors and carry microorganisms (including potential pathogens) into higher trophic levels through their ingestion by animals. In this study, impacts on copepod microbiota during chronic exposure to MPs were investigated by exposing copepods to a classic single-use polymer (low-density polyethylene (LDPE)) and a biodegradable polymer (polybutylene adipate terephthalate (PBAT)). Copepods were exposed to "virgin" and "weathered" MPs during four generations at an environmentally relevant concentration of 300 μg/L, followed by one "detoxification" generation without MP exposition. Impacts of MP exposure on copepod microbiota were investigated using 16S rRNA gene high-throughput sequencing. The result of nonmetric multidimensional scaling (NMDS) analysis showed that copepods (with or without MP exposure) carried distinguishable microbiota as compared with the microbiota of water and microalgae used for maintaining copepods. According to the results of permutational analysis of variance (PERMANOVA), the microbiota of MP-exposed (both PBAT and LDPE) copepods was significantly different from the microbiota of unexposed copepods during generations one to four. After "detoxification," however, no significant difference in microbiota composition was observed among all generation five copepods. Altogether, impacts on copepod microbiota of MP exposure for multiple generations were observed, despite plastic origin (biodegradable or not) and aging conditions. Furthermore, copepod microbiota seemed to return to their original structure as soon as the MP exposure stopped.
