Abstract:Drying treatments are an effective method of preserving the beneficial properties of postharvest mushrooms. The effects of natural-air drying (ND), hot-air drying (HD), vacuum-freeze drying (FD), heat pump drying (HPD) and microwave-vacuum drying (MVD) on the microstructure, flavor- and health-related compounds of F. velutipes root were investigated. The results showed that FD had the least impact on the microstructure of F. velutipes root and its original porous fiber structure appeared complete. It also possessed the highest content of volatile compounds. MVD gave the highest contents of umami amino acids, total phenolics and total flavonoids, and its extract exhibited high antioxidant activity. In addition, different drying treatments had significant effect on the chemical components of F. velutipes root, with FD and MVD may be the potent drying strategies for preservation of flavor and nutraceuticals respectively. Therefore, our results provided essential data support for F. velutipes root processing and functional product development.
Degradation of Tremella fuciformis polysaccharide by a combined ultrasound and hydrogen peroxide treatment: Process parameters, structural characteristics, and antioxidant activities

Abstract:The degradation effect of ultrasound (US)/hydrogen peroxide (H2O2) on Tremella fuciformis polysaccharide (TFP) were studied. The main process parameters of degradation were evaluated and the structural changes and antioxidant activities of TFP before and after US/H2O2 were analyzed. The results showed that the degradation effect of US/H2O2 was significantly higher than that of US or H2O2 alone, and the degradation of TFP was dependent on the duration of its exposure to US, the ultrasonic amplitude, and the H2O2 and TFP concentrations. US/H2O2 reduced the molecular weight (from 8.14 × 105 Da to 1.27 × 104 Da) and particle size (from 710 nm to 182 nm) of the TFP within 50 min and narrowed its molecular weight and particle size distribution. High performance liquid chromatography, Fourier-transform infrared spectroscopy, Carbon-13 nuclear magnetic resonance, scanning electron microscopy, atomic force microscopy, and Congo red results indicated that the treatment could break down the polysaccharide chains, hinder the aggregation, and improve the conformation flexibility of the TFP molecules without changing the primary structure and monosaccharide composition of TFP. Additionally, the degraded TFPs with low molecular weight exhibited a higher antioxidant activity than the original TFP. These findings suggest that the US/H2O2 treatment is a simple and effective method to prepare a TFP of low molecular weight and high bioactivity.

Abstract:Various drying methods play important roles in the preservation of foods. However, how the different drying methods affect the quality of some foods is not clear. This paper evaluates the effects of hot air, vacuum, microwave, and microwave vacuum drying techniques on important qualities and volatile compounds of whole shiitake (Lentinus edodes) mushrooms. These four drying methods resulted in a significantly (p<0.05) increase in the content of total free amino acids and the relative content of sulfur compounds of dried products. Microwave vacuum drying helped to maintain larger amounts of taste-active amino acids, and improved nutrient retention and color attributes. Furthermore, the uniform honeycomb network created by microwave vacuum drying along with a less collapsed structure of dried samples can be used to explain the observed high rehydration ratio. Therefore, microwave vacuum drying should be a potential method for obtaining high-quality dried mushrooms.

Effects of ultrasonic-enzymatic-assisted ethanol precipitation method on the physicochemical characteristics, antioxidant and hypoglycemic activities of Tremella fuciformis polysaccharides

Abstract:This investigation involved the extraction of a novel polysaccharide from the spore fermentation broth of Tremella fuciformis using a method that combined ultrasonic and enzymatic assistance with ethanol precipitation. It was then compared with enzymatic and ultrasonic extraction methods. The objective of this research is to offer a reference point for expanding the application of ultrasonic-assisted enzymatic extraction technology in T. fuciformis polysaccharides (TFPs). Based on single-factor experiments, Box-Behnken was used to optimize the extraction conditions of TFPs by ultrasonic-enzymatic-assisted ethanol precipitation extraction. The results revealed an optimal combination of enzymes, with a cellulase-to-papain ratio of 2:1, an enzyme addition of 4000U/100 mL, an enzymolysis temperature of 49 °C, ultrasonicpower at 3 W/mL and an ultrasonictime of 20 min. The extraction rate of TFPs and α- amylase inhibition rates were 23.94 % and 61.44 %, respectively. Comparing the physicochemical properties, structural characterization and in vitro activity of TFPs extracted through different methods, the results showed that ultrasonic treatment significantly influences the apparent morphology of polysaccharide and could enhance its in vitro biological activity. However, different extraction techniques exhibit insubstantial impact on the chemical composition, glycosidic bonds or glycosidic ring configurations within the polysaccharides. Among them, ultrasonic-enzymatic-assisted ethanol precipitation extraction of polysaccharide has the highest extraction rate and the lowest viscosity. It has significant effects on ABTS+ scavenging activity, α- amylase inhibition rate and glucose dialysis retardation index, polysaccharide treated with ultrasonic-enzymatic showed the best performance. These findings suggest that ultrasonic-enzymatic-assisted ethanol precipitation extraction can enhance the activities of TFPs, thereby providing a valuable insight for their future development and application.
Effect of ultrasound combined with chemical pretreatment as an innovative non-thermal technology on the drying process, quality properties and texture of cherry subjected to radio frequency vacuum drying

Abstract:To obtain high-quality cherry products, ultrasound (US) combined with five chemical pretreatment techniques were used on cherry prior to radio frequency vacuum drying (RFV), including carboxymethyl cellulose coating (CMC), cellulase (CE), ethanol (EA), isomaltooligosaccharide (IMO), and potassium carbonate + ethyl oleate (PC + AEEO). The effect of different pretreatments (US-CMC, US-CE, US-EA, US-IMO, US-(PC + AEEO)) on the drying characteristics, quality properties, texture, and sensory evaluation of cherries was evaluated. Results showed that the dehydration time and energy consumption were decreased by 4.17 - 20.83 % and 3.22 - 19.34 %, respectively, and the contents of individual sugars, soluble solid, total phenolics (TPC), natural active substances, total flavonoids (TFC), and antioxidant properties (DPPH, ABTS and FRAP) were significantly increased after US combined with five chemical treatments (P < 0.05). Moreover, the pretreatment played important role in improving texture properties and surface color retention in the dried cherries. According to the sensory evaluation analysis, the dehydrated cherries pretreated with US-CMC exhibited the highest overall acceptance, texture, crispness, color, and sweet taste showed lower off-odor, bitter taste and sour taste compared to control and other pretreatments. The findings indicate that US-CMC pretreatment is a promising technique for increasing physicochemical qualities and dehydration rate of samples, which provides a novel strategy to processing of dried cherry.

Abstract:Plant genes usually have multiple potential poly(A) addition signals, and different sites are used for 3' processing of transcripts from a single gene. In contrast, we show here that four barley genes that also have multiple poly(A) addition signals conforming to the plant consensus use only one signal. In each of these genes, the region of cleavage for poly(A) addition is centered on a conserved pentanucleotide. This AGGCA is followed by a conserved sequence homologous to sequences involved in self-cleavage of plant viroid RNA precursors; immediately following, in turn, are four or five nucleotides complementary to the nucleotides immediately preceding AGGCA in each gene. The presence of these conserved sequences and their association with a single region for poly(A) addition in three different gene types (high-pI and low-pI alpha-amylase genes and a thiol protease) that otherwise are not homologous in their 3' untranslated/flanking sequences suggest that they might participate in some common regulatory mechanism shared by these genes.

Abstract:In this study, the effect of an ultrasound (US) pre-treatment on the process of drying Cistanche slices through far-infrared vacuum drying was investigated with various experimental factors, including the US treatment time (25, 35, 45 min), frequency (20, 40, 60 kHz) and power (150, 180, 210 W). The results showed that compared with the samples without US, the material drying time after the US treatment was reduced by 16–36.8%. The effective moisture diffusion coefficients of Cistanche slices under different US conditions ranged from 1.61122 × 10−8 to 2.39274 × 10−8 m2/s, which agreed with food processing ranges. In addition, the phenylethanoid glycoside, iridoid, polysaccharide, total phenol and total flavonoid contents in Cistanche were significantly increased after US pre-treatment. However, the dried products obtained with the 45 min US treatment had greatly damaged internal structures, collapsed and seriously deformed surfaces, and low contents of active ingredients. Overall, the US pre-treatment could significantly improve the drying quality of Cistanche slices.

Abstract:BARCKGORUND In order to improve the drying efficiency and reduce the drying energy consumption of pleurotus eryngii, microwave hot-air flow rolling drying (MHARD) coupled with ultrasonic pretreating time (0, 20, 40, and 60 min) was used to investigate the drying profile, thermal characteristics, water migration, microstructure and rehydration dynamics of pleurotus eryngii using differential scanning calorimetry (DSC), LF-NMR and scanning electron microscopy (SEM). RESULTS Results showed that the drying time of pleurotus eryngii was 80, 70, 60 and 50 min, accordingly. Energy consumption was significantly reduced by ultrasonic pretreatment, and moisture effective diffusivity (Deff ) was increased with the increase of ultrasonic pretreating time. DSC curves showed that the drying process was accelerated by ultrasonic pretreatment significantly by enhancing the heat transfer. Meanwhile, SEM pictures showed that the cell was broken and numbers of irregular holes appeared in the ultrasound-pretreated samples. In terms of rehydration dynamics, Page model could well model the rehydration kinetics of dried pleurotus eryngii with R2 > 0.99. CONCLUSION The findings indicate that ultrasonic pretreatment is a promising method for MHARD of pleurotus eryngii as it can enhance drying process, and show potential for industrial application. Abstract:Ginger was dried by microwave infrared dual-field coupled drying (MIDFCD). The composition, structure, physicochemical properties and biological activity of ginger polysaccharides at various stages of MIDFCD were investigated. The MIDFCD significantly impacted the chemical composition, molecular weight (Mw), microstructure, and physicochemical properties of ginger polysaccharides. However, there were no notable differences in functional group composition. The Mw and chemical composition were notably influenced by microwave-infrared exposure and prolonged drying time. The degradation of polysaccharides due to high temperatures in the later stage resulted in further decreases in Mw and alterations in monosaccharide composition. These changes in chemical composition and Mw affected thermal properties, crystallization properties, particle size, rheological properties, antioxidant capacity, and hypoglycemic activity. These findings suggest that MIDFCD enhances the quality and bioactivity of natural polysaccharides. This study offers theoretical support for MIDFCD processing and the value-added utilization of ginger.

Abstract:This work examined the effect of ultrasound and microwave treatments, separate and in combination, on the physicochemical and functional properties of chestnut starch. The results revealed that the ultrasonic-microwave (UM) and microwave-ultrasonic (MU) dually modified samples exhibited more severe surface damage, weaker birefringence, and lower relative crystallinity and gelatinization enthalpy than the native and single-treated starches. The UM samples showed the highest oil absorption capacity, and the MU samples showed the highest water absorption capacity and the best freeze-thaw stability (five cycles) among all samples. The swelling power, peak, trough, final, and breakdown viscosities, and pasting temperature all decreased regardless of single or dual modification. This study provides a reference for potential industrial applications of ultrasound and microwave treatments for the modification of chestnut starch.

Abstract:The acquisition of histologic material is obligatory in order to establish the diagnosis of malignant pleural mesothelioma (MPM). In particular, tissue acquisition in cases of "dry" MPM (focal pleural thickening without pleural effusion or mediastinal lymph node involvement) is usually performed via a thoracoscopic pleural biopsy. In contrast, the techniques for performing echoendoscopic (transbronchial or transesophageal) needle aspiration of pleural lesions have only rarely been reported due to the theoretical limitations of tissue acquisition in such cases. We herein report the first case of "dry" MPM successfully diagnosed via endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) in a 73-year-old man presenting with a pleural mass in the right costovertebral recess, adjacent to the carina. The patient underwent radical resection, and a definitive pathological examination confirmed the diagnosis of epithelioid MPM.
Effects of drying methods on bioactive components of Ganoderma lucidum fermented whole wheat in products & in vitro digestive model

Abstract:The content of bioactive components is the key to determining the quality of Ganoderma lucidum fermented whole wheat (GW) products, and drying is a necessary link in the initial processing of GW, which will affect the bioactivity and quality of GW. This paper was to assess the effect of hot air drying (AD), freeze drying (FD), vacuum drying (VD) and microwave drying (MVD) on the content of bioactive substances and the characteristics of digestion and absorption of GW. The results showed that FD, VD and AD were beneficial to the retention of unstable substances such as adenosine, polysaccharide and triterpenoid active components in GW, and their contents were 3.84-4.66 times, 2.36-2.83 times and 1.15-1.22 times of MVD, respectively. The bioactive substances in GW were released during digestion. The bioavailability of polysaccharides in the MVD group (419.91 %) was significantly higher than that in the FD, VD and AD groups (68.74 %-78.92 %), but their bioaccessibility (5.66 %) was lower than that in the FD, VD and AD groups (33.41 %-49.69 %). Principal component analysis (PCA) showed that VD is more suitable for GW drying due to the comprehensive performance of 3 aspects in terms of active substance retention, bioavailability and sensory quality.
Effects of combined drying techniques and cellulase hydrolysis on the nutritional value and sensory properties of shiitake mushrooms (Lentinus edodes)

Abstract:Dried shiitake mushrooms offer rich nutritional value and unique sensory properties, prompting further investigation. The effects of different drying techniques (hot air drying (HAD), infrared hot air drying (IRHAD), pulsed vacuum drying (PVD), vacuum freeze drying (VFD), and natural drying (ND)) combined with enzymatic hydrolysis on the release of flavor compounds and nutrients from shiitake mushrooms were explored. The combination of HAD with cellulase hydrolysis yielded notably high levels of umami amino acids (5.4723 ± 0.1501 mg/g) and 5'-nucleotides (4.0536 ± 0.0062 mg/g), and superior volatile flavors. Combined with cellulase hydrolysis, IRHAD achieved the highest level of total sugars (6.57 ± 0.34 mg/mL), VFD resulted in the greatest soluble protein content (153.21 ± 0.23 μg/mL), PVD yielded the highest total phenolics content (93.20 ± 0.41 μg GAE/mL), and ND produced the maximum reducing sugar content (5.79 ± 0.13 mg/mL). This study addresses crucial gap in the post-drying processing of shiitake mushrooms, offering valuable insights for further product development of shiitake mushrooms.
Influence of Ultrasound-Assisted Vacuum Drying on Physicochemical Characteristics, Antioxidant Activity, and α-Glucosidase Inhibition Activity of Flos Sophorae Immaturus

Abstract:Flos Sophorae Immaturus (FSI) contains a large number of bioactive substances with antioxidant and hypoglycaemic activity. However, a feasible drying process plays an important role in the retention of its biological activity. The present work investigated the effects of ultrasound-assisted vacuum drying (UAVD) on FSI samples in terms of drying time, colour, microstructure, and total flavonoid content (TFC). Meanwhile, the antioxidant activity and α-glucosidase inhibition activity were also evaluated. The results show that the drying time of UVAD samples was decreased by 40% compared to that of the single vacuum-dried (VD) samples (600 W for 10 min). The cellular porous structures of FSI tissue were formed by UAVD, which promoted the migration of water from the inside to the outside. Furthermore, samples treated by UAVD exhibited better antioxidant activities and α-glucosidase and α-amylase inhibition capacities, with DPPH (81.86%), ABTS (88.61%), FRAP (83.05%), α-glucosidase inhibition capacity (89%), α-amylase (85%), drying time (3 h), and total aberration (ΔE) (1.63) being the highest characteristic traits. In this condition, the highest levels of total flavonoid content (TFC), rutin, quercetin, kaempferol, isorhamnetin, and genistein were obtained with 266.94, 239.46, 35.56, 8.54, 10.37, and 5.64 mg/g DW, respectively. The results confirm that UAVD is a novel method that significantly reduced the VD time and promoted the release of the bioactive substances of FSI.

Abstract:To examine the processing characteristics and high quality of an improved microwave vacuum drying system, litchi fruits were dried using intermittent microwave volumetric heating while microwave vacuum drying at 2 W/g was carried out for comparison; the intermittent microwave heating profiles were set as (1) 5 min drying-on, 5 min drying-off; (2) 5 min drying-on, 10 min drying-off; and (3) 5 min drying-on, 15 min drying-off. Energy consumption during drying was determined, and physicochemical properties such as moisture content, vitamin C, total phenolics, color, and sensory evaluation of dried products were assessed. In microwave vacuum drying, intermittent microwave volumetric heating was found to be energy-efficient (about 32 KJ/g to 45 KJ/g) and saved at least 31% of energy consumption compared with microwave vacuum drying as well as decreasing product browning. In addition, microwave volumetric heating had no substantial effects on sugar and protein contents, while antioxidants were affected significantly (p ≤ 0.05). Moreover, sensory evaluation showed that intermittent microwave-assisted vacuum drying (IMVD) increased the acceptance of the dried product compared with microwave vacuum drying (MVD).

Abstract:Hot air drying (HAD) is the most commonly used method for citrus peel drying. However, the long time and low efficiency of this process leads to the degradation of product quality and a reduction in antioxidant activity. Therefore, in this study, to improve the drying efficiency and increase the product quality of dried citrus peel, the effects of microwave vacuum drying (MVD) were compared with HAD and vacuum freeze drying (VFD) with respect to their drying kinetics, bioactive compound profile and antioxidant capacity. The results showed that the drying rate decreased with increasing sample load but increased with increasing microwave power. The Page model was the best model for fitting the drying curve of citrus peel during MVD, and the highest rehydration rate (84.5%) was obtained at a microwave vacuum power of 600 W. The microstructure of citrus peel dried by MVD was assessed by scanning electron microscopy, and a loose structure with highly porous material was observed, whereas citrus peel dried by HAD was compact. The total phenolic content (10.48 ± 0.21 mg GAE/g DW), total flavonoid content (6.54 ± 0.41 mg RE/g DW) and oxygen radical absorbance capacity (279.29 ± 11.37 µmol TE/g DW) values obtained for citrus peel dried via MVD were significantly higher than those obtained using HAD (p < 0.05). Several phenolic acid substances and flavonoid profiles were identified and quantified by HPLC-DAD, including coumaric acid, nobiletin, and tangeretin. The nobiletin content observed after MVD was significantly higher than that measured after HAD and VFD. The overall results of this study revealed that MVD at lower microwave powers appears to be a good alternative to the conventional HAD of citrus peel.

Abstract:Microwave freeze-drying of foamed blueberry puree assisted by wave-absorbing material was investigated experimentally. Freeze-drying times and dried product qualities were evaluated. Unfoamed and foamed samples were prepared with whey protein isolate as the foaming agent and pectin as the stabilizer. Silicon carbide (SiC) was used as the wave-absorbing pad of the sample in comparison with quartz as the wave-transparent pad. The effect of foaming treatment was first examined. At 30 °C of radiation temperature and 15 Pa of chamber pressure with a quartz pad, the foamed sample saved 39.1% of drying time compared with the unfoamed one. The appropriate increasing temperature could shorten the freeze-drying time. The effect of microwave heating was then explored. With quartz pad at 0.5 W/g of microwave power, 30 °C and 15 Pa, only limited enhancements were attained for the two samples. With SiC pad at 0.25 W/g, 30 °C, and 15 Pa, the microwave freeze-drying time of the foamed sample was decreased by 50.0% and 17.9%, respectively, compared with conventional freeze-drying times of unfoamed and foamed samples. When the power was increased to 0.5 W/g, the drying time was shortened by 56.5% and 28.6%, respectively. There were no significant differences in color, total monomeric anthocyanin (TMA), and total phenolic content (TPC) between microwave and conventional freeze-dried products. TMA and TPC retentions in all products were more than 80% and 75%, respectively. The wave-absorbing material-assisted microwave freeze-drying of foamed material could greatly reduce the drying time and improve the process economy, while maintaining the comparable product quality with the conventional freeze-drying.

Abstract:The effects of ultrasound (US), ultrahigh pressure (UP), cold plasma (CP), and high pressure and pulsed electric fields (PEF) pretreatments were evaluated for drying characteristics, bioactive components, and pectin properties of jujube slices during microwave coupled with pulsed vacuum drying (MPVD). The effects of ultrasound (US), ultrahigh pressure (UP), cold plasma (CP), and high pressure and pulsed electric fields (PEF) pretreatments were evaluated for drying characteristics, bioactive components, and pectin properties of jujube slices during microwave coupled with pulsed vacuum drying (MPVD). US, CP, and PEF reduced drying time by 18.75%, while UP reduced it by 6.25%. Samples treated with US and PEF showed improved color and rehydration (345.11%, 319.24%), with SEM revealing larger pores post‐pretreatment. PEF‐treated jujube had highest phenol (16.95 mg GAE/g DW) and water‐soluble pectin(WSP) (43.71 mg/g AIR), correlating positively with rehydration (r = 0.67). CP had highest flavonoid content (17.01 mg RE/g DW) and US showed highest antioxidant activity (DPPH 80.57%, ABTS 89.65%). Chelate‐soluble pectin (CSP) Sodium carbonate‐soluble pectin (NSP) was positively correlated with hardness (r = 0.71, 0.69). These findings suggest pretreatments improve drying efficiency, nutrient retention, and pectin functionality in jujube products. Practical Applications Drying for jujube (Ziziphus jujuba Mill.) is the most common method of processing and ensure the sale. Three kinds of non‐thermal processing pretreatment method (ultrasound, ultrahigh pressure, and cold plasma) combined with novel drying method, microwave coupled pulsed vacuum drying (MPVD) was studied in the application on the dehydration of jujube slice in this paper. The drying characteristics and pectin properties of winter jujube were analyzed and treated by MPVD compared with freeze drying and hot air drying, for both technological research and processing practice. The drying kinetics, bioactive compounds, antioxidant capacity, sensory attributes, and pectin properties were also used as descriptors in order to promising dehydration method for obtaining high value‐added jujube products. Moreover, the relationship between the cell wall pectin and the phenols, hardness and crispness were analyzed, which give a new insight to study the changing scheme in the nutrients and the texture during the dehydration for fruits and vegetables.
