Abstract:Introduction: Inhalant abuse is an important health issue especially among children and adolescents who often encounter these agents in the home. Research into the neurobiological targets of inhalants has lagged behind that of other drugs such as alcohol and psychostimulants. However, studies from our lab and others have begun to reveal how inhalants such as the organic solvent toluene affect neurons in key addiction related areas of the brain including the ventral tegmental area, nucleus accumbens and medial prefrontal cortex. In the present study, we extend these findings and examine the effect of toluene on electrophysiological responses of pyramidal neurons in the basolateral amygdala BLA, a region important for generating emotional and reward based information needed to guide future behavior. Methods: Whole-cell patch-clamp electrophysiology recordings of BLA pyramidal neurons in rat brain slices were used to assess toluene effects on intrinsic excitability and excitatory glutamatergic synaptic transmission. Results: Acute application of 3 mM but not 0.3 mM toluene produced a small but significant (~20%) increase in current-evoked action potential (AP) firing that reversed following washout of the toluene containing solution. The change in firing during exposure to 3 mM toluene was accompanied by selective changes in AP parameters including reduced latency to first spike, increased AP rise time and decay and a reduction in the fast after-hyperpolization. To examine whether toluene also affects excitatory synaptic signaling, we expressed channelrhodopsin-2 in medial prefrontal cortex neurons and elicited synaptic currents in BLA neurons via light pulses. Toluene (3 mM) reduced light-evoked AMPA-mediated synaptic currents while a lower concentration (0.3 mM) had no effect. The toluene-induced reduction in AMPA-mediated BLA synaptic currents was prevented by the cannabinoid receptor-1 antagonist AM281. Discussion: These findings are the first to demonstrate effects of acute toluene on BLA pyramidal neurons and add to existing findings showing that abused inhalants such as toluene have significant effects on neurons in brain regions involved in natural and drug induced reward.

Abstract:Although volatile organic solvents such as toluene are used for commercial and industrial uses, they are often voluntarily inhaled for their intoxicating and euphoric effects. Research into the effects of inhalants such as toluene on brain function have revealed actions on a variety of ligand-gated and voltage-activated ion channels involved in regulating neuronal excitability. Previous work from this laboratory has also shown that brief exposures to toluene vapor induce changes in the intrinsic excitability and synaptic transmission of neurons within the medial prefrontal cortex and ventral tegmental area that vary depending on projection target. In the present study, we recorded current-evoked spiking of medium spiny neurons (MSNs) in the nucleus accumbens (NAc) core and shell in adolescent rats exposed to an intoxicating concentration of toluene vapor. Compared to air controls, firing of NAc core MSNs in Sprague–Dawley rats was not altered 24 h after exposure to 10,500 ppm toluene vapor while spiking of NAc shell MSNs was enhanced at low current steps but reduced at higher current steps. When the rheobase current was used to putatively identify MSN subtypes, both "D1-like" and "D2-like" MSNs within the NAc shell but not core showed toluene-induced changes in firing. As toluene may itself have altered the rheobase resulting in misclassification of neuron subtype, we conducted additional studies using adolescent D2-Cre rats infused with a Cre-dependent mCherry reporter virus. Following toluene vapor exposure, spiking of NAc shell D2+ MSNs was enhanced at low current steps but inhibited at higher currents as compared to air controls while there were no differences in the firing of NAc shell D2- MSNs. The toluene-induced change in NAc D2+ shell MSN firing was accompanied by alterations in membrane resistance, rheobase, action potential rise time and height with no changes noted in D2- MSNs. Overall, these data add to a growing literature showing that brief exposures to intoxicating concentrations of toluene vapor causes selective alterations in the excitability of neurons within the addiction neurocircuitry that vary depending on sub-region, cell-type and projection target.

Repeated toluene exposure increases the excitability of layer 5 pyramidal neurons in the prefrontal cortex of adolescent rats

Abstract:Despite serious health effects, volatile industrial products containing toluene are deliberately inhaled for their psychoactive actions, mainly among adolescents and young adults. Chronic toluene inhalation induces multiple alterations at the cellular and behavioral level; however, modifications of neuronal networks associated with the reward system after repeated toluene exposure are not thoroughly characterized. Here we used whole-cell recordings to determine the effects of repeated exposure to toluene (1000, 4000 or 8000 ppm for 30 min, twice a day, for ten days) on the neurophysiological properties of prelimbic layer 5 pyramidal neurons of the medial prefrontal cortex (mPFC) in adolescent male Wistar rats. Neurons from animals repeatedly exposed to toluene showed a concentration-dependent increase in action potential firing discharge. This increase was related to a reduction of the small-conductance calcium-activated potassium current (after-hyperpolarization current, IAHP) that controls the firing frequency of neurons. Likewise, toluene altered the kinetics of the action potential. The hyperexcitability seen in toluene-exposed animals was also associated with an increase in the glutamatergic spontaneous synaptic activity converging on mPFC neurons. In summary, repeated toluene exposure enhances the excitability of prelimbic layer 5 pyramidal neurons of the mPFC in adolescent rats.

Abstract:Neuronal circuits within the prefrontal cortex (PFC) mediate higher cognitive functions and emotional regulation that are disrupted in psychiatric disorders. The PFC undergoes significant maturation during adolescence, a period when cannabis use in humans has been linked to subsequent vulnerability to psychiatric disorders such as addiction and schizophrenia. Here, we investigated in a rat model the effects of adolescent exposure to Δ 9 -tetrahydrocannabinol (THC), a psychoactive component of cannabis, on the morphological architecture and transcriptional profile of layer III pyramidal neurons—using cell type- and layer-specific high-resolution microscopy, laser capture microdissection and next-generation RNA-sequencing. The results confirmed known normal expansions in basal dendritic arborization and dendritic spine pruning during the transition from late adolescence to early adulthood that were accompanied by differential expression of gene networks associated with neurodevelopment in control animals. In contrast, THC exposure disrupted the normal developmental process by inducing premature pruning of dendritic spines and allostatic atrophy of dendritic arborization in early adulthood. Surprisingly, there was minimal overlap of the developmental transcriptomes between THC- and vehicle-exposed rats. THC altered functional gene networks related to cell morphogenesis, dendritic development, and cytoskeleton organization. Marked developmental network disturbances were evident for epigenetic regulators with enhanced co-expression of chromatin- and dendrite-related genes in THC-treated animals. Dysregulated PFC co-expression networks common to both the THC-treated animals and patients with schizophrenia were enriched for cytoskeletal and neurite development. Overall, adolescent THC exposure altered the morphological and transcriptional trajectory of PFC pyramidal neurons, which could enhance vulnerability to psychiatric disorders.

Abstract:Currently, there is a gap in understanding the neurobiological impact early adolescent toluene exposure has on subsequent actions of other drugs. Adolescent (PND 28-32) male Swiss-Webster mice (N = 210) were exposed to 0, 2000, or 4000 ppm of toluene vapor for 30 min/day for 5 days. Immediately following the last toluene exposure (PND 32; n = 15) or after a short delay (PND 35; n = 15), a subset of subjects' brains was collected for monoamine analysis. Remaining mice were assigned to one of two abstinence periods: a short 4-day (PND 36) or long 12-day (PND 44) delay after toluene exposure. Mice were then subjected to a cumulative dose response assessment of either cocaine (0, 2.5, 5, 10, 20 mg/kg; n = 60), ethanol (0, 0.5, 1, 2, 4 g/kg; n = 60), or saline (5 control injections; n = 60). Toluene concentration-dependently increased locomotor activity during exposure. When later challenged, mice exposed previously to toluene were significantly less active after cocaine (10 and 20 mg/kg) compared to air-exposed controls. Animals were also less active at the highest dose of alcohol (4 g/kg) following prior exposure to 4000 ppm when compared to air-exposed controls. Analysis of monoamines and their metabolites using High Pressure Liquid Chromatography (HPLC) within the medial prefrontal cortex (mPFC), nucleus accumbens (NAc), dorsal striatum (dSTR), and ventral tegmental area (VTA) revealed subtle effects on monoamine or metabolite levels following cumulative dosing that varied by drug (cocaine and ethanol) and abstinence duration. Our results suggest that early adolescent toluene exposure produces behavioral desensitization to subsequent cocaine-induced locomotor activity with subtle enhancement of ethanol's depressive effects and less clear impacts on levels of monoamines.

Shared and Divergent Transcriptomic Regulation in Nucleus Accumbens D1 and D2 Medium Spiny Neurons by Cocaine and Morphine

Abstract:Substance use disorders (SUDs) induce widespread molecular dysregulation in the nucleus accumbens (NAc), a brain region pivotal for coordinating motivation and reward. These molecular changes are thought to support lasting neural and behavioral disturbances that promote drug-seeking in addiction. However, different drug classes exert unique influences on neural circuits, cell types, physiology, and gene expression despite the overlapping symptomatology of SUDs. To better understand common and divergent molecular mechanisms governing SUD pathology, our goal was to survey cell-type-specific restructuring of the NAc transcriptional landscape in after psychostimulant or opioid exposure. We combined fluorescence-activated nuclei sorting and RNA sequencing to profile NAc D1 and D2 medium spiny neurons (MSNs) across cocaine and morphine exposure paradigms, including initial exposure, prolonged withdrawal after repeated exposure, and re-exposure post-withdrawal. Our analyses reveal that D1 MSNs display many convergent transcriptional responses across drug classes during exposure, whereas D2 MSNs manifest mostly divergent responses between cocaine and morphine, with morphine causing more adaptations in this cell type. Utilizing multiscale embedded gene co-expression network analysis (MEGENA), we discerned transcriptional regulatory networks subserving biological functions shared between cocaine and morphine. We observed largely integrative engagement of overlapping gene networks across drug classes in D1 MSNs, but opposite regulation of key D2 networks, highlighting potential therapeutic gene network targets within MSNs. These studies establish a landmark, cell-type-specific atlas of transcriptional regulation induced by cocaine and by morphine that can serve as a foundation for future studies towards mechanistic understanding of SUDs. Our findings, and future work leveraging this dataset, will pave the way for the development of targeted therapeutic interventions, addressing the urgent need for more effective treatments for cocaine use disorder and enhancing the existing strategies for opioid use disorder.
Elevated synthesis of an axonally transported protein correlates with axon outgrowth in normal and injured pyramidal tracts

Abstract:Axons of the adult mammalian CNS typically fail to regenerate after injury. Among the hypotheses to account for this failure is the proposition that certain axonal proteins necessary for axon growth are expressed in much greater abundance in developing than in mature neurons, and that these proteins are not reinduced after injury to mature axons (Skene and Willard, 1981b). In the present experiments, we have found that hamster pyramidal tract neurons synthesize an acidic, 43K protein that is transported into growing axons during the first 2 weeks of postnatal development, and then declines at least an order of magnitude by the fourth postnatal week. The decline in synthesis of the 43K protein coincides with the cessation of pyramidal tract axon elongation. This protein resembles a “growth-associated protein,” GAP- 43, which is induced during regeneration of CNS axons in lower vertebrates. The 43K protein in hamster pyramidal tract neurons is not reinduced after axotomy in adult animals, which correlates with the failure of the injured axons to regenerate. Injury to neonatal pyramidal tract axons does not reverse or delay the decline in 43K protein synthesis. This is consistent with previous findings (Kalil and Reh, 1982) that pyramidal tract axons regrow for only a brief period after neonatal injury. Taken together, these results lend support to the hypothesis that synthesis of GAP-43 is important for axon growth in development and regeneration.
Effects of Indoor Exposure to Low Level Toluene on Neural Network Alterations During Working Memory Encoding

Abstract:Objective: While high concentrations of toluene are known to affect multiple human organ systems, research concerning the influence of immediate, short-term exposure to toluene indoors and at low concentrations is scarce. Here, we studied effects of indoor toluene exposure on neural network alterations during working memory (WM) encoding. Methods: A total of 23 healthy college students were recruited. All participants were situated in a closed envi-ronmental chamber with a full fresh air system. Each participant was subjected to four exposure experiments with different toluene concentrations (0, 17.5, 35, and 70 ppb, named Group A, B, C and D, respectively), with at least one week between each experiment. WM Behavioral and 19-channel electroencephalogram (EEG) recordings in a pre-set environmental chamber were conducted simultaneously during each toluene exposure experiment. Neural networks relevant to WM encoding were visualized analyzing the obtained data. Results: 1. No significant difference in WM behavioral performance among the four groups was found. However, a significant increase in whole brain neural network functional connectivity was noted, especially in the frontal region. 2. An outflow directional transfer function (DTFoutflow) revealed higher frontal region values among Group D (the 70 ppb group) as compared to Group A, B and C (the0, 17.5 ppb and 35 ppb groups, respectively), although no differences in frontal region DTFinflow values among the four groups were noted. 3. The DTFFZ-F7, DTFFZ-T5, DTFFZ-P4, DTFFZ-P3, DTFFP2-O2, DTFP3-T4, DTFP3-F4, DTFP4-CZ and DTFP4-T4 values of Group D were found to be higher as compared to those of Group A and B. Furthermore, DTFFZ-F7 and DTFP4-T4 values of Group C were higher as compared to those of Group A. The DTFFZ-F7 values of Group D were higher as compared to those of the Group C. Conclusion: Short-term toluene exposure significantly influences neural networks during cognitive processes such as WM encoding, even at low concentration.
EEG Neural Networks Reveal Influence of Indoor Toluene on Working Memory at Low Concentration

Abstract:Toluene is a common domestic air contaminant and a neurotoxic volatile organic compound. However, few previous studies have directly evaluated the health effects of toluene exposure at a low concentration. This study conducted a within-subject experiment to compare working memory during delayed match-to-sample tasks with EEG recording under 0 ppb and 70 ppb toluene exposure. Compared with the the control group (Ctrl group, 0 ppb), power spectral density analysis showed that the alpha and theta bands in the frontal region of the toluene group (Tol group, 70 ppb) increased considerably in both the encoding and maintenance periods (P < 0.01 for theta, P < 0.05 for alpha). The direct transfer function indicated that the functional connectivity of the frontal network in the theta band (P < 0.01), gamma band (P < 0.01) and alpha band (P < 0.01) significantly decreased, while that of the frontoparietal network increased under toluene exposure in the theta (P < 0.01) and alpha bands (P < 0.01) during the maintenance period. Toluene exposure at the ppb level could cause differences in neural oscillation patterns and functional connectivity even in healthy subjects. Abstract:Trans-anethole is an aromatic compound that has been studied for its anti-inflammation, anticonvulsant, antinociceptive, and anticancer effects. A recent report found that trans-anethole exerted neuroprotective effects on the brain via multiple pathways. Since noxious stimuli may both induce neuronal cell injury and affect synaptic functions (e.g., synaptic transmission or plasticity), it is important to understand whether the neuroprotective effect of trans-anethole extends to synaptic plasticity. Here, the effects of trimethyltin (TMT), which is a neurotoxic organotin compound, was investigated using the field recording method on hippocampal slice of mice. The influence of trans-anethole on long-term potentiation (LTP) was also studied for both NMDA receptor-dependent and NMDA receptor–independent cases. The action of trans-anethole on TMT-induced LTP impairment was examined, too. These results revealed that trans-anethole enhances NMDA receptor-dependent and -independent LTP and alleviates TMT-induced LTP impairment. These results suggest that trans-anethole modulates hippocampal LTP induction, prompting us to speculate that it may be helpful for improving cognitive impairment arising from neurodegenerative diseases, including Alzheimer's disease.

Abstract:Synaptic efficacy changes, long-term potentiation (LTP) and depression (LTD), underlie various forms of learning and memory. Synaptic plasticity is generally assessed under prolonged activation, whereas learning can emerge from few or even a single trial. Here, we investigated the existence of rapid responsiveness of synaptic plasticity in response to a few number of spikes, in neocortex in a synaptic Hebbian learning rule, the spike-timing-dependent plasticity (STDP). We investigated the effect of lowering the number of pairings from 100 to 50, and 10 on STDP expression, using whole-cell recordings from pyramidal cells in rodent somatosensory cortical brain slices. We found that a low number of paired stimulations induces LTP at neocortical layer 4-2/3 synapses. Besides the asymmetric Hebbian STDP reported in the neocortex induced by 100 pairings, we observed a symmetric anti-Hebbian LTD for 50 pairings and unveiled a unidirectional Hebbian spike-timing-dependent LTP (tLTP) induced by 10-15 pairings. This tLTP was not mediated by NMDA receptor activation but requires CB1 receptors and transient receptor potential vanilloid type-1 (TRPV1) activated by endocannabinoids (eCBs). eCBs have been widely described as mediating short- and long-term synaptic depression. Here, the eCB-tLTP reported at neocortical synapses could constitute a substrate operating in the online learning of new associative memories or during the initial stages of learning. In addition, these findings should provide useful insight into the mechanisms underlying eCB-plasticity occurring during marijuana intoxication.

Abstract:Dopamine modulates striatal synaptic plasticity, a key substrate for action selection and procedural learning. Thus, characterizing the repertoire of activity-dependent plasticity in striatum and its dependence on dopamine is of crucial importance. We recently unraveled a striatal spike-timing-dependent long-term potentiation (tLTP) mediated by endocannabinoids (eCBs) and induced with few spikes (~5-15). Whether this eCB-tLTP interacts with the dopaminergic system remains to be investigated. Here, we report that eCB-tLTP is impaired in a rodent model of Parkinson's disease and rescued by L-DOPA. Dopamine controls eCB-tLTP via dopamine type-2 receptors (D2R) located presynaptically in cortical terminals. Dopamine-endocannabinoid interactions via D2R are required for the emergence of tLTP in response to few coincident pre- and post-synaptic spikes and control eCB-plasticity by modulating the long-term potentiation (LTP)/depression (LTD) thresholds. While usually considered as a depressing synaptic function, our results show that eCBs in the presence of dopamine constitute a versatile system underlying bidirectional plasticity implicated in basal ganglia pathophysiology.
Alternations in Neural Oscillation Related to Attention Network Reveal Influence of Indoor Toluene on Cognition at Low Concentration.

Abstract:Despite accumulative literature reporting negative impacts of high-concentration toluene, cognitive effects of toluene at low concentration are still unclear. Twenty-two healthy college students were exposed in a closed environmental chamber to investigate the influence of indoor toluene on cognitive performance and brain activity. During each toluene exposure condition (0 ppb, 17.5 ppb, 35 ppb, and 70 ppb), attention network test and electroencephalogram (EEG) recording were synchronously performed after 4-hour toluene exposure. Characteristic neural oscillation patterns in three attention networks were compared between four groups. The statistical analyses indicated that short-term exposure to toluene had no significant impact on behavioral performance of attention network. However, there was a significant increase in the power of theta and alpha band of executive network and orienting network in the whole brain, especially in frontal region when exposed to toluene. Besides, no significant difference was observed in alerting network. The alternations in neural oscillation demonstrated that more effort was required to accomplish the same tasks when exposed to toluene. The present study revealed that short-term exposure to toluene affected brain activity of attention network even at low concentration, which provided a theoretical basis for the development of safer evaluation methods and standards in the future.

Abstract:Tetramethylenedisulfotetramine (tetramine or TETS), a potent convulsant, triggers abnormal electrical spike activity (ESA) and synchronous Ca2+ oscillation (SCO) patterns in cultured neuronal networks by blocking gamma-aminobutyric acid (GABAA) receptors. Murine hippocampal neuronal/glial cocultures develop extensive dendritic connectivity between glutamatergic and GABAergic inputs and display two distinct SCO patterns when imaged with the Ca2+ indicator Fluo-4: Low amplitude SCO events (LASE) and High amplitude SCO events (HASE) that are dependent on TTX-sensitive network electrical spike activity (ESA). Acute TETS (3.0 µM) increased overall network SCO amplitude and decreased SCO frequency by stabilizing HASE and suppressing LASE while increasing ESA. In multielectrode arrays, TETS also increased burst frequency and synchronicity. In the presence of TETS (3.0 µM), the clinically used anticonvulsive perampanel (0.1-3.0 µM), a noncompetitive AMPAR antagonist, suppressed all SCO activity, whereas the GABAA receptor potentiator midazolam (1.0-30 µM), the current standard of care, reciprocally suppressed HASE and stabilized LASE. The neuroactive steroid (NAS) allopregnanolone (0.1-3.0 µM) normalized TETS-triggered patterns by selectively suppressing HASE and increasing LASE, a pharmacological pattern distinct from its epimeric form eltanolone, ganaxolone, alphaxolone, and XJ-42, which significantly potentiated TETS-triggered HASE in a biphasic manner. Cortisol failed to mitigate TETS-triggered patterns and at >1 µM augmented them. Combinations of allopregnanolone and midazolam were significantly more effective at normalizing TETS-triggered SCO patterns, ESA patterns, and more potently enhanced GABA-activated Cl- current, than either drug alone.
Alterations in Functional Connectivity of Executive Control Network Reveal Influence of Indoor Toluene on Cognition at Low Concentration

Abstract:Background: The mechanism for indoor toluene on cognition at low concentrations remains unknown. Objective: The present study combines attention network testing (ANT) and electroencephalography (EEG) to investigate the brain network mechanisms associated with indoor low concentrations of toluene exposure. Methods: 22 healthy volunteers were exposed for 4 h to four conditions of toluene (0 ppb, 17.5 ppb, 35 ppb, and 70 ppb). Upon exposure to toluene, electroencephalographic activity was recorded and attention network tasks were performed. In this study, the direction transfer function (DTF) has been used to evaluate human brain functional connectivity among neural EEG signals. Results: We have evaluated the results of twenty-two subjects who completed all experiments. No significant difference was found between the four toluene conditions in behavioral performance. The DTFs indicated that the frontal cortex is the main brain region of the executive control network. In the 70 ppb condition, theta band DTF values were significantly higher than those in the 0 ppb and 17.5 ppb conditions. Theta band DTF values from the frontal region to other brain regions (DTFs_outflow) in 70 ppb condition were higher than in 0 ppb condition. Conclusion: Theta band functional connectivity in the frontal region was significantly enhanced after exposure to toluene at low concentrations. The alternations in neural connectivity demonstrated that more effort was required to accomplish the same tasks when exposed to toluene. These results enable us to understand the early effects of indoor pollutants on cognition and the brain network mechanism.
Lung-injury depresses glutamatergic synaptic transmission in the nucleus tractus solitarii via discrete age-dependent mechanisms in neonatal rats

Abstract:Transition periods (TPs) are brief stages in CNS development where neural circuits can exhibit heightened vulnerability to pathologic conditions such as injury or infection. This susceptibility is due in part to specialized mechanisms of synaptic plasticity, which may become activated by inflammatory mediators released under pathologic conditions. Thus, we hypothesized that the immune response to lung injury (LI) mediated synaptic changes through plasticity-like mechanisms that depended on whether LI occurred just before or after a TP. We studied the impact of LI on brainstem 2nd-order viscerosensory neurons located in the nucleus tractus solitarii (nTS) during a TP for respiratory control spanning (postnatal day (P) 11-15). We injured the lungs of Sprague-Dawley rats by intratracheal instillation of Bleomycin (or saline) just before (P9-11) or after (P17-19) the TP. A week later, we prepared horizontal slices of the medulla and recorded spontaneous and evoked excitatory postsynaptic currents (sEPSCs/eEPSCs) in vitro from neurons in the nTS that received monosynaptic glutamatergic input from the tractus solitarii (TS). In rats injured before the TP (pre-TP), neurons exhibited blunted sEPSCs and TS-eEPSCs compared to controls. The decreased TS-eEPSCs were mediated by differences in postsynaptic α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic-acid receptors (AMPAR). Specifically, compared to controls, LI rats had more Ca2+-impermeable AMPARs (CI-AMPARs) as indicated by: 1) the absence of current-rectification, 2) decreased sensitivity to polyamine, 1-Naphthyl-acetyl-spermine-trihydrochloride (NASPM) and 3) augmented immunoreactive staining for the CI-AMPAR GluA2. Thus, pre-TP-LI acts postsynaptically to blunt glutamatergic transmission. The neuroimmune response to pre-TP-LI included microglia hyper-ramification throughout the nTS. Daily intraperitoneal administration of minocycline, an inhibitor of microglial/macrophage function prevented hyper-ramification and abolished the pre-TP-LI evoked synaptic changes. In contrast, rat-pups injured after the TP (post-TP) exhibited microglia hypo-ramification in the nTS and had increased sEPSC amplitudes/frequencies, and decreased TS-eEPSC amplitudes compared to controls. These synaptic changes were not associated with changes in CI-AMPARs, and instead involved greater TS-evoked use-dependent depression (reduced paired pulse ratio), which is a hallmark of presynaptic plasticity. Thus we conclude that LI regulates the efficacy of TS → nTS synapses through discrete plasticity-like mechanisms that are immune-mediated and depend on whether the injury occurs before or after the TP for respiratory control.
2-Heptanone reduces inhibitory control of the amygdala over the prelimbic region in rats

Abstract:Basolateral amygdala (BLA) nuclei and their reciprocal connections with prelimbic (PL) and infralimbic (IL) regions of the medial prefrontal cortex (mPFC) are involved in the regulation of fear. 2-Heptanone is released in urine in stressed rats, and the olfactory detection of this odor produces immediate avoidance and alarm reactions and modifies neuronal activity in limbic connections in non-stressed rats. If 2-heptanone acts as a danger signal, then long-lasting actions would be expected. The aim of the present study was to investigate whether the forced inhalation of 2-heptanone modifies the response capacity of the BLA-mPFC circuit in the long term (48 h). Single-unit extracellular recordings were obtained from the PL and IL during electrical stimulation of the BLA (square-wave pulses; 1 ms, 20 µA, 0.3 Hz, 110 stimuli over a total duration of 360 s) in three groups of Wistar rats: control group (no sensory stimulation), unpredictable auditory stimulation group, and 2-heptanone stimulation group. A brief-latency (1 ms), short-duration (5 ms) paucisynaptic response followed BLA stimulation and was unaffected by any sensorial stimulation. The paucisynaptic response was followed by a mostly inhibitory and long-lasting (>750 ms) afterdischarge in the control and auditory stimulation groups. In the 2-heptanone group, the inhibitory afterdischarge shifted to an excitatory afterdischarge after ∼250 ms in the PL and after ∼500 ms in the IL. Importantly, the rats that were included in this study were born in local housing facilities. Thus, these animals were never in contact with predators and instead in contact with only conspecifics. These results indicate that the forced inhalation of 2-heptanone is able to modify BLA-mPFC responsivity in the long term. 2-Heptanone decreases inhibitory control of the amygdala over mPFC activity. Disinhibition of the mPFC may lead to the adaptive expression of defensive behaviors, even in animals that are not in the presence of predators.
GABAA positive modulator and NMDA antagonist-like discriminative stimulus effects of isoflurane vapor in mice

Abstract:RationaleSeveral neurotransmitter systems have been hypothesized to be involved in the in vivo effects of volatile anesthetics. Drug discrimination may represent a novel procedure to explore the neurochemical systems underlying the sub-anesthetic behavioral effects of these compounds.ObjectivesThe purpose of the present study was to examine the contribution of GABAA and NMDA receptors to the discriminative stimulus effects of a behaviorally active sub-anesthetic concentration of isoflurane vapor.MethodsSixteen B6SJLF1/J mice were trained to discriminate 10 min of exposure to 6,000 ppm isoflurane vapor from air. Substitution tests were conducted with volatile anesthetics, abused vapors, GABAA positive modulators, NMDA antagonists, and nitrous oxide.ResultsThe volatile anesthetics, enflurane and halothane as well as the abused vapors toluene and 1,1,1-trichloroethane fully substituted for isoflurane. The GABAA positive modulators, pentobarbital, midazolam, and zaleplon but not the direct GABAA agonist, muscimol, produced high levels of partial substitution for isoflurane. The anticonvulsant, valproic acid fully substituted for isoflurane but a second, tiagabine, did not substitute. The competitive NMDA antagonist, CGS-19755, fully and the non-competitive NMDA antagonist, dizocilpine, partially substituted for isoflurane. The glycine-site NMDA antagonist, L-701,324 did not substitute for isoflurane. Gamma-hydroxybutric acid and nitrous oxide gas also failed to substitute for isoflurane.ConclusionsThe discriminative stimulus effects of sub-anesthetic concentrations of isoflurane vapor are shared by other vapor anesthetics and abused inhalants. The discriminative stimulus effects of isoflurane vapor appear to be mediated by both positive allosteric modulation of GABAA receptors as well as antagonism of NMDA receptors.

Abstract:▪ Background and Purpose Methamphetamine (METH) use disorder has risen dramatically over the past decade, and there are currently no FDA‐approved medications due, in part, to gaps in our understanding of the pharmacological mechanisms related to METH action in the brain. Experimental Approach Here, we investigated whether transient receptor potential ankyrin 1 (TRPA1) mediates each of several METH abuse‐related behaviours in rodents: self‐administration, drug‐primed reinstatement, acquisition of conditioned place preference, and hyperlocomotion. Additionally, METH‐induced molecular (i.e., neurotransmitter and protein) changes in the brain were compared between wild‐type and TRPA1 knock‐out mice. Finally, the relationship between TRPA1 and the dopamine transporter was investigated through immunoprecipitation and dopamine reuptake assays. Key Results TRPA1 antagonism blunted METH self‐administration and drug‐primed reinstatement of METH‐seeking behaviour. Further, development of METH‐induced conditioned place preference and hyperlocomotion were inhibited by TRPA1 antagonist treatment, effects that were not observed in TRPA1 knock‐out mice. Similarly, molecular studies revealed METH‐induced increases in dopamine levels and expression of dopamine system‐related proteins in wild‐type, but not in TRPA1 knock‐out mice. Furthermore, pharmacological blockade of TRPA1 receptors reduced the interaction between TRPA1 and the dopamine transporter, thereby increasing dopamine reuptake activity by the transporter. Conclusion and Implications This study demonstrates that TRPA1 is involved in the abuse‐related behavioural effects of METH, potentially through its modulatory role in METH‐induced activation of dopaminergic neurotransmission. Taken together, these data suggest that TRPA1 may be a novel therapeutic target for treating METH use disorder.

Maturation of renal organic acid transport: substrate stimulation by penicillin and p-aminohippurate (PAH).

Abstract:Maturation of the renal transport system for - aminohippurate (PAH) in rabbits was measured by an in vitro technique. The ability of renal cortical slices to accumulate PAH increased rapidly after two weeks of age, showed a peak at four weeks and then declined to adult levels. Twice daily administration of PAH (200 mg/kg/day i.p.) for seven days to one-week-old rabbits significantly increased PAH uptake by renal cortical slices. Treatment of nursing rats with 200 or 400 mg/kg of PAH daily for three or seven days significantly enhanced PAH uptake in vitro but had no effect on N-methylnicotinamide uptake. Penicillin administration to nursing rats also produced a significant increase in PAH transport, but did not enhance N-methylnicotinamide transport. The data support the hypothesis that treatment of young animals with organic anions leads to selective stimulation of the mechanisms responsible for renal organic acid transport.
