Abstract:Aims: Metformin HCl, Dapagliflozin and Saxagliptin is a new drug combination for the treatment of Diabetes Mellitus which is one of the oldest and lethal diseases of mankind. Aim of the research work was to develop and validate novel, rapid, sensitive, specific, robust stability indicating analytical method for simultaneous estimation of: Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form as fixed dose formulation. Study Design: Method development and validation was performed as recommended in ICH guideline “Validation of analytical procedures: Test and Methodology Q2 (R1)”. Methodology: Method develop with chromatographic parameters as C18 column (250mm×4.6 mm, 5mm particle size), HPLC system with PDA detector and mobile phase contained a mixture of Phosphate Buffer pH 3.5 and Acetonitrile (80:20 v/v) + 1 ml triethylamine per 100 ml mobile phase. The flow rate was set to 1 ml/min with responses measured at 265 nm, injection volume was 20 μl, and run time of 15 mins. Results: The retention time of Metformin HCl, Dapagliflozin and Saxagliptin was 5.8 min, 6.8 mins and 8.4 min respectively with resolution of 3.5 between Metformin HCl and Dapagliflozin and 4.5 between Dapagliflozin and Saxagliptin. Linearity was established in the range of 250-1500 μg/ml for Metformin HCl, 1.25-7.5 μg/ml for Dapagliflozin and Saxagliptin with correlation coefficients more than 0.999. The percentage recoveries were between 98.39-101.66 for Metformin HCl, 99.01-101.77 for Dapagliflozin and 98.88-101.87 for Saxagliptin Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The forced degradation studies were performed by using HCl, NaOH, H2O2, thermal and UV Radiation. The developed method was successfully applied for the quantification and hyphenated instrumental analysis. Conclusion: Significance of developed method is that it can be utilize for routine or unknown sample analysis of assay of Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form developed by various Pharmaceutical Industry.

Abstract:For multicomponent pharmaceutical dosage forms, simultaneous equation method has emerged out as boon with lots of advantages like easy, rapid and sensitive method. In this study, a UV spectroscopic method has been established and authenticated for simultaneous estimation of Sitagliptin phosphate and Empagliflozin in bulk form. Methanol: Water (5:5) has been employed as common solvent for the proposed method. The optimum conditions for analysis of the drug were established. Sitagliptin phosphate and empagliflozin showed maximum absorbance (ʎmax) at 267nm and 276.2nm respectively. The calibration plot was found to be linear in the range of 50-250μg/ml for sitagliptin phosphate and empagliflozin with R2= 0.9957 and 0.9981 respectively. Method was validated according to ICH guidelines for linearity, accuracy, precision, system suitability, robustness, sensitivity and specificity. The proposed method was simple, precise, sensitive, accurate, rapid and suitable for routine quality analysis of sitagliptin phosphate and empagliflozin in bulk.

Experimental Design Approach for Quantitative Expressions of Simultaneous Quantification of Two Binary Formulations Containing Remogliflozin and Gliptins by RP-HPLC

Abstract:The aim of this study was to develop a fast RP-HPLC method for simultaneous measurement of two antidiabetic formulations (vildagliptin + remogliflozin and teneligliptin + remogliflozin) under identical experimental conditions. Using the Box–Behnken approach and response surface design, the interaction and quadratic influence of three variable parameters, acetonitrile %, pH of the mobile phase, and flow rate, on resolution between the peaks were optimized. To forecast the resolution of peaks (2.7 and 6.5) for the three anti-diabetic medications, the design space with desirability function was used to find the optimal chromatographic conditions. Isocratic elution with 58:42 acetonitrile and phosphate buffer (20 mM KH2PO4, pH adjusted to 4.9 with orthophosphoric acid) over a Zorabx C18 HPLC column with a flow rate of 1.2 mL min−1 separated all three analytes in 2.5 min. In addition, the optimized HPLC process was validated using ICH recommendations. The devised HPLC method’s precision and accuracy were proven by the low percent relative standard deviation (0.60–1.65%), good percentage recovery (98.18–101.50%), and low percentage relative errors (0.20–1.82%). The method’s robustness was also proven by slightly varying the five separate parameters. Finally, the accuracy of the proposed HPLC approach was confirmed using a standard addition method for simultaneous determination of vildagliptin + remogliflozin and teneligliptin + remogliflozin from formulations. Furthermore, the findings demonstrated that experimental design can be successfully used to optimize chromatographic conditions with fewer runs. The devised HPLC method for simultaneous quantification of two binary combinations utilizing the same chromatographic conditions is fast, accurate, precise, and easy, and it might be utilized in laboratories for routine quality control investigations on both formulations.

Abstract:Three novel intermolecular proton transfer complexes of an anti-diabetic drug linagliptin were synthesized in methanol with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone, chloranilic acid, and ninhydrin as the acceptors. The stoichiometry of the complexes was determined by Job's method of continuous variation. A swift and precise spectrophotometric method was developed and validated for quantitative analysis of linagliptin in its pure form. This method generated linear calibration curves, covering specific concentration ranges of 2.84–28.35, 2.36–21.26, and 9.45–85.06 μg mL −1 for reagents, such as DDQ, chloranilic acid, and ninhydrin respectively. In all instances, the correlation coefficient exceeded 0.997, demonstrating the method's reliability. The Benesi-Hildebrand equation was applied to determine the spectral factors, such as the transition dipole moment, oscillator's strength, molar extinction, ionization potential, resonance energy, and thermodynamic factors, i.e., association constant and Gibb's free energy. The stability of the synthesized charge transfer complexes was confirmed by ensuring that the factors met the approved ranges. Additionally, the synthesized complexes were characterized by FT-IR and thermogravimetric analysis. The environmental friendliness of the newly developed method was assessed using the AGREE tool. A comparison was made between the proposed method and an existing spectrophotometric technique using an analytical eco-scale approach. The proposed method proved effective in analyzing linagliptin in dosage formulations in the presence of potentially interfering excipients confirming its suitability for quality control purposes in pharmaceutical production.

Abstract:Aim: This study aims on the development of a chemometric-assisted spectroscopic method for the analysis of combined dosage form of emtricitabine (EMT), tenofovir alafenamide fumarate (TEN), and dolutegravir sodium (DOL). The use of a multivariate algorithm to analyse spectrophotometric data is a novel approach to estimating drug concentrations in formulations. Materials and Methods: The quantitative estimation of EMT, TEN, and DOL in tablets was carried out using four chemometric approaches: Classical least square (CLS), inverse least square, partial least square, and principal component regression. Thirty-two ternary mixtures of calibration sets and 16 mixtures of validation sets were prepared. The absorbance data matrix was attained by calculating absorbance at 25 different wavelengths in a range of 240–336 nm (Δλ = 4 nm). The chemometric calculations were performed using Matlab2018a and Minitab software. The developed methods were validated. Results: The great accuracy of the current study was justified by the near-perfect recovery values (100%) and low standard deviation. For chemometrics approaches, the root mean square error of calibration (RMSEC), root mean square error of prediction (RMSEP), and root mean square error of cross-validation (RMSECV) outcomes display decent accuracy and precision. Conclusion: The CLS approach yielded the lowest predicted residual error sum of squares, RMSEC, RMSEP, and RMSECV scores. As a result, CLS might be regarded as the best chemometric approach among all techniques utilized. The label claim determined is in excellent accordance with the mean recoveries for EMT, TEN, and DOL. So, it can be used in quality control laboratories.
Response surface methodology combined Box-Behnken design optimized green kinetic spectrophotometric and HPLC methods to quantify angiotensin receptor blocker valsartan in pharmaceutical formulations

Abstract:The high-performance liquid chromatographic (HPLC) and kinetic spectrophotometric methods were established to compute valsartan (VAL) in pharmaceutical formulations. The spectrophotometric procedures adopted initial rate, fixed time, and equilibrium strategies to assess VAL. The method was based on the carboxylic acid group of the oxidized VAL with a mixture of potassium iodate (KIO3) and potassium iodide (KI) at room temperature, producing a stable, yellow-coloured absorb at 352 nm. The critical parameters were optimized using green process optimization methodology such as Box-Behnken design (BBD) which belongs to response surface methodology (RSM). After the screening, experiments identified them as significant, and then three crucial factors were optimised: KI volume, KIO3 volume, and reaction time against response as absorbance. The HPLC procedure was also optimized based on the desirability function on RSM-BBD. The parameters such as pH, methanol (%), and flow rate (ml/min) were optimized with the best responses: peak area, symmetry, and theoretical plates. The linearity of spectrophotometric and HPLC methods was within the range of 2-24 and 0.25-11.25 µg/ml, respectively. The developed procedures produced excellent accuracy and precision. The design of the experiment (DoE) setting explained and discussed the individual steps and the importance of independent and dependent variables used to develop the model and optimization. The method was validated as per the International Conference on Harmonization (ICH) guidelines. Furthermore, Youden's robustness study was applied with factorial combinations of the preferred analytical parameters and explored their influence with alternative conditions. The analytical Eco-Scale score was calculated and was found a better option as green methods to quantify VAL. The results were reproducible with the analysis completed with biological fluid and wastewater samples.

Abstract:Endotherms maintain constant body temperature through physiological and behavioral adjustments. Behavioral thermoregulation is an important factor influencing energy balance. We exposed the leaf‐eared mouse, Phyllotis darwini, to temperatures corresponding to its natural thermal range and studied two forms of behavioral thermoregulation: diminishing surface to volume ratio by huddling and heat dissipation by increasing physical contact with the substrate (flattening). We predicted that at low ambient temperatures (Ta) huddling would be used as a heat conservation mechanism and at high Ta flattening would be used for heat loss. We simultaneously measured oxygen consumption (V̇o2) and flattening, in response to three independent factors: huddling, Ta, and body mass. Each experiment was a 6‐h V̇o2 trial where five virgin females were measured at constant Ta. We performed this protocol for two body mass groups, small (ca. 40 g) and large (ca. 70 g), in a metabolic chamber. Treatments were groups with and without the ability to huddle at five different Ta, ranging from 5° to 35°C. A significant interaction between all three factors was found. Huddling and flattening were used as strategies for conserving or dissipating heat, respectively, and the shift between both strategies occurred at the lower limit of thermoneutrality. At Ta below thermoneutrality, huddling was a more effective way of reducing metabolic requirements and was more efficient (HE) in small individuals than large individuals. So, by huddling, small individuals save more energy. At high Ta, flattening appeared to be an equally useful mechanism for heat loss, for both large and small animals.
A stability indicating UPLC method development and validation for the simultaneous estimation of nateglinide and metformin hydrochloride in bulk and tablet dosage form

Abstract:Abstract Background Nateglinide and metformin HCl are used in combination for the treatment of type 2 diabetes. A simple, sensitive and reliable UPLC method was developed for simultaneous estimation of nateglinide and metformin HCl using Phenomenox C 18 (50*2.1 mm, 3.5 μm) column at ambient temperature as stationary phase in addition to mobile phase containing 75 volumes of ammonium formate buffer (pH = 3) along with 25 volumes of acetonitrile with a flow rate of 0.2 mL/min with UV detection at 260 nm and a run time of 3 min. The developed method was validated as per ICH Q2(R1) guidelines. Results The separation of metformin HCl and nateglinide was done at retention times of 1.014 min and 1.435 min, respectively. The mean % recovery for nateglinide and metformin HCl in the accuracy study was observed to be 99.9% and 99.2%, respectively. LOD and LOQ values were determined considering the S/N ratio and were found to be 0.09 μg/mL and 0.3 μg/mL, respectively, for nateglinide and 0.75 μg/mL and 2.5 μg/mL, respectively, for metformin HCl. The method was found to be precise with % RSD values of 0.58 and 0.45, respectively, for repeatability and intermediate precision of nateglinide and 0.43 and 0.43, respectively, for repeatability and intermediate precision of metformin HCl which were within acceptance criteria. The method was found to be linear in the range of 7.5–45 μg/mL and 62.5–375 μg/mL for nateglinide and metformin HCl, respectively. The regression equations for nateglinide and metformin HCl were found to be y = 17377 x + 6543.4 and y = 18439 x + 43,537, respectively. The method was found to be robust by deliberate changes in the method parameters like flow rate and mobile phase composition. Forced degradation studies were performed as per ICH Q1A(R2) and Q1B guidelines, and peak purity was observed in all types of degradation studies for both the drugs. Conclusion The developed method was found to be satisfactory as it is simple, sensitive, accurate, precise, robust, rapid, economical and yet stability indicating and can be applied successfully in the routine laboratory analysis for the simultaneous estimation of nateglinide and metformin HCl in the bulk and pharmaceutical dosage forms.
A Simple and Sensitive HPTLC Method for Simultaneous Determination of Metformin Hydrochloride and Sitagliptin Phosphate in Tablet Dosage Form

Abstract:A simple, rapid, and precise high-performance thin-layer chromatographic (HPTLC) method for simultaneous estimation of two antidiabetic drugs, metformin hydrochloride and sitagliptin phosphate, in tablet dosage form has been developed and validated. Chromatography was performed on silica gel 60 F254 plates with butanol : water : glacial acetic acid (6 : 2 : 2, v/v/v) as mobile phase. This system gave a good resolution for metformin hydrochloride ( R f value of 0.35 ± 0.01) and sitagliptin phosphate ( R f value of 0.75 ± 0.01). Detection and quantification were carried out at 227 nm. The linear regression data for the calibration plot showed a good relationship with r = 0.9995 and 0.9991 for metformin hydrochloride and sitagliptin phosphate, respectively. The method was validated for precision and recovery. The limits of detection and quantification were 13.05 and 39.56 ng/μL for metformin hydrochloride and 2.65 and 8.03 ng/μL for sitagliptin phosphate, respectively. The amounts of the drugs in the marketed formulation were 99.86% and 98.91% for metformin hydrochloride and sitagliptin phosphate, respectively.

Abstract:Quality by design is applied for the development of various pharmaceutical processes including analytical methods. By applying QbD approach chemometric based analytical method was developed for the estimation of amlodipine besylate and telmisartan by UV-VIS spectrophotometry. Solvent 0.1 N HCl was utilised and 291.2 nm and 365.2 nm was the wavelength for measurement of absorbance. Effect of input variables on spectrum characteristics were studied for selection of critical parameters and developed method was validated as per ICH Q 2 R1 regulatory guidelines. Linearity of both the drugs was ascertained over the conc range 5-40mcg/ml. The accuracy was found 104.46% for TEL and 96.25% for AMD; and the precision study was shown acceptable data as %RSD 2.5416 for TEL and 5.7364 for AMD. The developed method is rigid, robust and efficient for the estimation of AMD and TEL, which are in 1: 8 proportionate in the composition of dosage form. QbD was applied to build rigid robust method through risk assessment at early stage and defining the design space at the later stage.

Abstract:In the present study, two drugs for treating acromegaly are investigated which are either synthetic growth hormone inhibiting hormone (GHIH) or dopamine receptor agonist. Octreotide (OCT) and bromocriptine (BCT) were quantified with a quick, simple, and sensitive spectrophotometric approach in their authentic forms and commercial dosage forms. This approach was based upon formation of ion pair complex between erythrosine B and investigated drugs in an aqueous buffered solution. For OCT determination the higher sensitivity was obtained using ΔA at 525 nm. On the other hand, BCT was estimated using the absorbance at 556 nm. Under optimal conditions for the reaction, construction of calibration curves were performed within concentration range of 0.4-4 µg ml-1 for OCT and 1-9 µg ml-1 for BCT with with low detection limits (0.094 and 0.235 µg ml-1) and low quantitation limits (0.29 and 0.71 µg ml-1) for OCT and BCT respectively. The developed approach was assessed for linearity, accuracy, precision, limits of detection, and limits of quantitation in compliance with ICH validation recommendations. The suggested approaches demonstrated good linearity, as evidenced by determination coefficients (r2) of 0.999 with a slope 0.14 for OCT and 0.9989 with a slope 0.06 for BCT. Additionally, the environmental friendliness of the investigated method was assessed with some of the recent green analytical chemistry metrics.

Abstract:Metformin is considered as type 2 diabetes first line treatment according to American Diabetes Association and European Association. But, in some cases, di- or tri - therapy should be prescribed for glycemic management, prevention of the maximum dose side effects and induced effectiveness. Co-administration of Linagliptin with metformin has many benefits on diabetic patients such as decrease the possibility of hypoglycemia. For the first time, novel and reliable techniques were developed and verified for the concurrent quantification of metformin hydrochloride and linagliptin, while accounting for the existence of metformin toxic impurity 1-cyanoguanidine in their pure and dosage forms. Method (A) utilizes the zero-order spectrophotometric approach to quantitatively determine the concentration of linagliptin. The measurements are performed at a wavelength of 295 nm. The double divisor derivative ratio spectrophotometric method is used in Method (B) to measure the amounts of metformin and cyanoguanidine at 252 nm and 219 nm wavelengths, respectively. The spectrophotometric method (C) for determining metformin and cyanoguanidine at 252 nm and 223 nm, respectively, is based on the single divisor derivative ratio-zero crossing technique. The obtained findings were subjected to statistical comparison with the reported method, revealing no statistically significant differences. The Green Analytical Procedure Index (GAPI) and Analytical GREEnness Metric approach (AGREE) determined that these approaches had a high degree of environmental friendliness. Additionally, the proposed strategy was deemed to be practical according to the Blue Applicability Grade Index (BAGI) assessment tool.

Abstract:This paper describes the systematic development of green and sustainable HPLC method for quantification of antidiabetic drugs teneligliptin hydrobromide hydrate, metformin hydrochloride and pioglitazone HCl in tablet formulation using Box–Behnken design. Box–Behnken design was used to know the influence of identified critical method parameters, volume of acetonitrile, flow rate and column temperature on the retention time of all three drugs and resolution between two drugs. Statistical analysis by Analysis of variance was computed to understand the potential interactions among critical method parameters. Further mathematical model was validated by using statistical and graphical optimization to define the design space. From the three parameters under investigation the study revealed that the response was more influenced by slight change in volume of acetonitrile, demanding its strict control. A mixture of KH 2 PO 4 phosphate buffer (20 mM): acetonitrile: methanol (40:30:30%v/v) was employed as the mobile phase for chromatographic separation using octadecyl silyl column (250 × 4.6 mm, 5 μm). Detection was performed at 236 nm and 0.86 ml/min was set as the flow rate of mobile phase. The linearity was observed in the range of 12–28 μg/ml for Teneligliptin hydrobromide hydrate, 300–700 μg/ml for Metformin hydrochloride and 9–21 μg/ml for Pioglitazone hydrochloride as shown by r 2 ≥ 0.99 for all three drugs. Retention time of Teneligliptin hydrobromide hydrate, Metformin hydrochloride and Pioglitazone hydrochloride was 4.09, 3.01 and 11.44 min, respectively. The % relative standard deviation for accuracy, precision and robustness were all within the specification, less than 2, which indicates that method was validated properly as per guideline. The studies successfully demonstrate the application of Box–Behnken design in the development of accurate and sensitive liquid chromatographic technique with enhanced method performance. Furthermore, greenness of the analytical method was assessed using 12 principles of green analytical chemistry by AGREE, complex GAPI and Analytical eco-scale tool, indicating that the developed method was ecofriendly.

Abstract:Recently, a new formulation containing Mirabegron (MB) and Solifenacin succinate (SFS) has been approved for the management of over active bladder. However, only one analytical method has been reported for the simultaneous determination of both the analytes. Therefore, the current study was design to develop simple UV derivative spectroscopic and rapid RP-HPLC methods for simultaneous determination of MB and SFS. The chromatographic separation of MB and SFS was performed using Phenomenex Kinetex C18 (150mm × 4.5 mm × 5 μm) analytical column. A mixture of Water: Acetonitrile (20:80%v/v) was consider as mobile phase, at a flow rate of 1ml/min and at detector wavelength 225nm. A linear response was observe over the concentration range 2.5-12.5 μg/ml and 0.5-2.5 μg/ml respectively. The first order derivative method was develop by derivatisation of the zero absorption spectra for the first absorption spectra. The Zero crossing point of MB and SFS at 221 nm and 266 nm was obtain respectively. Beer’s law is obey in the concentration range of 7.5-20 μg/ml and 1.5-4 μg/ml for MB and SFS with correlation coefficient (R2) of 0.9984 and 0.9993 respectively. Both the methods were validated in accordance to guidelines for linearity, precision, repeatability, limit of detection (LOD), Limit of Quantification (LOQ), accuracy and robustness. Further, both the methods were validated and compared statistically using Student’s-t-test and employed for the concurrent estimation of MB and SFS in formulations. The proposed methods were simple, accurate, precise, and rapid. Therefore, they can be use for regular quality control of MB and SFS formulations and dissolution studies as well.

Abstract:A reverse phase high performance liquid chromatography method was developed and validated for simultaneous quantitative estimation of ambroxol hydrochloride, guaiphenesin and terbutaline sulphate. The chromatographic separation was achieved using a Phenomenex Luna C18 (2), 100A, 5μ (4.6 X 250mm) column using 0.012(M) sodium salt solution of 1-hexane sulphonic acid:acetonitrile 73: 27 (V/V) adjusted to pH 3 using orthophosphoric acid having isocratic elution of flow rate 1.0mL/min, detection at 220nm and injection volume 20 μL. The retention times of ambroxol hydrochloride, guaiphenesin and terbutaline sulphate were 25, 6.7 and 4.7 min, respectively. The proposed method was found to have linearity in concentration range of 63.40-95.11 μg/mL for ambroxol, 68.00-302.52 μg/mL for guaiphenesin and 4.88-7.20 μg/mL for terbutaline sulphate. Regression coefficient (R2) were 0.9997, 0.9989 and 0.9984 and percentage recovery were 101.67%, 101.68%, and 99.45% for ambroxol, guaiphenesin and terbutaline sulphate, respectively. The method validation showed excellent results for precision, accuracy, linearity and specificity. The proposed method was successfully used for the quantitative analysis of an oral liquid formulation, being developed in our laboratory, containing ambroxol hydrochloride, guaiphenesin and terbutaline sulphate.

Abstract:An accurate derivative spectrophotometric method was developed and validated for the determination of dipeptidyl peptidase inhibitor vildagliptin in the pharmaceutical dosage form. The second derivative of the UV spectra has enabled the estimation of vildagliptin absorbance at 217 nm without any interferences. Linearity, precision, accuracy, detection (LOD), and quantification (LOQ) limits were established for method validation. Calibration curve was linear in the range of 10-60 μg/mL with a regression coefficient of 0.998. The method was validated as per the International Conference on Harmonization (ICH Q2 (R1)). The limit of detection and the limit of quantification were found to be 2.06 μg/mL and 6.25 μg/mL, respectively. Intra and interday precision data illustrated that the method has acceptable reproducibility as the percentage relative standard deviation (RSD) was less than 2 %, which indicates the precision of the method. The recovery was 98.39 % by the standard addition method. The percentage assay of vildagliptin was 98.06 % which showed good applicability. The following results indicate that the procedure is accurate, precise, and reproducible while being simple and less time-consuming. The method was demonstrated to be adequate for routine analysis in quality control.
